SYNOPSIS. Cnidarians posses polytrophic feeding capacities including:1) endocytosis ofpaniculate food, 2) absorption of dissolvedorganic material, 3) utilization of assimilates from zooxanthellae.Heteroxeniafuscescens seems to be specialized to fuel metabolismonly by epidermal uptake of organic compounds and by materialoriginating from the cytosymbionts. Therefore, these soft coralsare appropriate organisms to study the contribution of bothsubstrates to nutrition quantitatively. By the uptake of only12 amino acids and glucose, experimentally determined, approximately80% of the energy demand can be covered, the contribution ofalgal products not included. Autoradiographic studies show thatabsorption occurs through epidermal membranes and that compoundsare transported through the mesogloea in both directions atlow rate. Furthermore, adaptations can be shown which intensifyabsorption of organic compounds and which reduce the loss ofenergy-rich substances from the coelenteron. For example, materialoriginating from zooxanthellae which would wash out into thesea due to the polyfunctionality of the coelenteron, is partlyretained by pharyngial tissue and the epidermis. Time-series 18O and 13C records from cohabiting massive coralPorites australiensis and giant clamTridacna gigas from the Great Barrier Reed of Australia, and from calcareous green algae in a core through modernHalimeda bioherm accreting in the eastern Java Sea, provide insights into the complex links between environmental factors and stable isotopes imprinted in these reef skeletal materials. The aragonitic coral and giant clam offer 20 years and 15 years of growth history, respectively. The giant clam yields mean 18O and 13C values of-0.5±0.5 and 2.2±0.2 (n=67), which agree well with the predicted equilibrium values. The coral yields mean 18O and 13C values of-5.6±0.5 and-1.8±0.7 (n=84), offering a striking example of kinetic and metabolic fractionation effects. Although both the coral and giant clam harbor symbionts and were exposed to a uniform ambient environment during their growth histories, their distinct isotopic compositions demonstrate dissimilar calcification pathways. The 18O records contain periodicities corresponding to the alternating annual density bands revealed by X-radiography and optical transmitted light. Attenuation of the 18O seasonal amplitudes occurring in the giant clam record 8 years after skeletal growth commenced is attributed to a changeover from fast to slow growth rates. Extreme seasonal 18O amplitudes of up to 2.2 discerned in both the coral and giant clam records exceed the equivalent seasonal temperature contrast in the reef environment, and are caused by the combined effect of rainfall and evaporation during the monsoon and dry seasons, respectively. Thus in addition of being useful temperature recorders, reef skeletal material of sufficient longevity, such asPorites andTridacna, may also indicate rainfall variations. Changing growth rates, determined from the annual growth bands, may exert a primary control on the coral 13C record which shows a remarkable negative shift of 1.7 over its growth history, by comparison with only 0.15 negative shift in the contemporaneous giant clam record. Use of coral 13C records as proxies of fossil fuel CO2 uptake by the ocean must be regarded with caution. The 18O and 13C records fromHalimeda are remarkably uniform over 1000 years of bioherm accretion history (18O=-1.7±0.2; 13C=3.9±0.1,n=28), in spite of variable Mg-calcite cements present in the utricles. Most of the cement infilling is probably syndepositional, and both theHalimeda aragonite and the Mg-calcite cements containign 12.3 mole % MgCO3 are deposited in isotopic equilibrium. Therefore, in favorable circumstances these algal skeletal remains may act as the shallow water analogs of benthic foraminifera in deep sea sediments in recording ambient sea water isotopic composition and temperature.  相似文献

The involvement of cell-wall polymer synthesis in auxin-mediated elongation of coleoptile segments from Zea mays L. was investigated with particular regard to the growth-limiting outer epidermis. There was no effect of indole acetic acid (IAA) on the incorporation of labeled glucose into the major polysaccharide wall fractions (cellulose, hemicellulose) within the first 2 h of IAA-induced growth. 2,6-Dichlorobenzonitrile inhibited cellulose synthesis strongly but had no effect on IAA-induced segment elongation even after a pretreatment period of 24 h, indicating that the growth response is independent of the apposition of new cellulose microfibrils at the epidermal cell wall. The incorporation of labeled leucine into total and cell-wall protein of the epidermis was promoted by IAA during the first 30 min of IAA-induced growth. Inhibition of IAA-induced growth by protein and RNA-synthesis inhibitors (cycloheximide, cordycepin) was accompanied by an inhibition of leucine incorporation into the epidermal cell wall during the first 30 min of induced growth but had no effect on the concomitant incorporation of monosaccharide precursors into the cellulose or hemicellulose fractions of this wall. It is concluded that at least one of the epidermal cell-wall proteins fulfills the criteria for a growth-limiting protein induced by IAA at the onset of the growth response. In contrast, the synthesis of the polysaccharide wall fractions cellulose and hemicellulose, as well as their transport and integration into the growing epidermal wall, appears to be independent of growth-limiting protein and these processes are therefore no part of the mechanism of growth control by IAA.Abbreviations CHI cycloheximide - COR cordycepin - DCB 2,6-dichlorobenzonitrile - GLP growth-limiting protein(s) - IAA indole-3-acetic acid  相似文献

The hemoglobin of the sea snakeMicrocephalophis gracilis was purified and the primary structure of the and chains determined. This is the first sea snake hemoglobin structure characterized, and apparently also the first complete structure of any snake hemoglobin (an chain of a viper was known), allowing judgments of reptilian variants. Variations between the sea snake form and other reptilian forms are large (52–65 differences for the chains), of similar order as those between the sea snake and avian (56–65 differences) or human (58 differences) forms. Functionally, 19 residues at / contact areas and 7 at heme contacts are exchanged in relation to the human and chains. Four positions of the sea snake hemoglobin contain residues thus far unique to this form. However, all replacements appear compatible with conserved overall functional properties.  相似文献

Coral planulae settle, then metamorphose and form polyps. This study examined the morphological process of metamorphosis from planulae into primary polyps in the scleractinian corals Acropora nobilis and Acropora microphthalma, using the cnidarian neuropeptide Hym-248. These two species release eggs that do not contain Symbiodinium. The mode of acquisition of freshly isolated Symbiodinium (zooxanthellae) (FIZ) by the non-symbiotic polyp was also examined. Non-Hym-248 treated swimming Acropora planulae did not develop blastopore, mesenteries or coelenteron until the induction of metamorphosis 16 days after fertilization. The oral pore was formed by invagination of the epidermal layer after formation of the coelenteron in metamorphosing polyps. At 3 days after settlement and metamorphosis, primary polyps exposed to FIZ established symbioses with the Symbiodinium. Two–four days after exposure to FIZ, the distribution of Symbiodinium was limited to the gastrodermis of the pharynx and basal part of the polyps. Eight–ten days after exposure to FIZ, Symbiodinium were present in gastrodermal cells throughout the polyps.  相似文献

The transport activities of two primary ATP-dependent organic-anion transporters in the tonoplast of isolated barley (Hordeum vulgare L. cv. Klaxon) vacuoles have been characterised with N-ethylmaleimide glutathione (NEM-SG) and taurocholate as substrates. The transporters showed different sensitivities to organic anions and a variety of transport inhibitors and drugs. The vacuolar uptake of NEM-SG was inhibited by carbonylcyanide 4-trifluoromethoxyphenylhydrazone, 4,4-diisothiocyanatostilbene-2,2-disulfonic acid (DIDS), S-(2,4-dinitrophenyl)glutathione, alkyl-S-glutathione derivatives and taurocholate but stimulated by probenecid. The uptake of taurocholate was inhibited by vinblastine, DIDS and probenecid. Both transporters were unaffected by verapamil. The kinetic properties of the transporters indicate a general preference for amphiphilic anions with some substrate overlap. These characteristics of the transporters are similar to those displayed by the multidrug resistance protein of mammalian drug-resistant cells. The characteristics of valine uptake by isolated microcolonies of Galaxea fascicularis (Linnaeus 1758) were studied under various conditions including light, dark and feeding. The results demonstrated the presence of: (1) a linear component which might represent either a diffusional transport or a low-affinity carrier-mediated transport (apparent carrier affinity >250 mol·l–1), and (2) a high-affinity active carrier-mediated transport (apparent carrier affinity about 5 mol·l-1). The latter is mediated by two different systems: (i) a Na+-dependent carrier, stimulated by light and operative in both fed and unfed polyps, and (ii) a Na+-independent carrier, light insensitive and present only in unfed polyps. Competition experiments with other amino acids show that the Na+-dependent carrier is highly specific for neutral amino acids, as indicated by the high inhibition constants of basic and acidic amino acids. Differences in phosphate metabolism of symbiotic and aposymbiotic Condylactus suggest that the host animal makes available quantities of phosphate to support growth of zooxanthellae. Nitrite may serve as a nitrogen source for symbionts as indicated by host removal of nitrite from sea water.The presence of zooxanthellae is responsible for removal of phosphate from sea water in the dark whereas there is excretion during light periods. There is a greater uptake of nitrite from sea water in the light compared with the dark in symbiotic animals.Since nitrate is removed from sea water by aposymbiotic animals, the presence of nitrate reducing bacteria is proposed.  相似文献

Summary Tradescantia virginiana L. plants were cultivated under contrasting conditions of temperature, humidity, light quality and intensity, and nutrient status in order to investigate the effect of growth conditions on the water relations parameters of the leaf epidermal cells. Turgor pressure (P), volumetric elastic modulus (), half-time of water potential equilibration (T 1/2), hydraulic conductivity (L p ) were measured with the miniaturized pressure probe in single cells of the upper and lower epidermis of leaves. Turgor differed (range: 0.1 bar to 7.2 bar) between treatments with lowest values under warm and humid conditions and additional supply of fertilizer, and highest values under conditions of low air humidity and low nutrient supply. The volumetric elastic modulus changed by 2 orders of magnitude (range: 3.0 bar to 350 bar, 158 cells), but was only affected by the treatments, in as much as it was dependent on turgor. The turgor dependence of , measured on intact leaves of T. virginiana, was similar to that for cells of the isolated (peeled) lower epidermis, where as a function of turgor was linear over the whole range of turgors. This result has implications for the discussion of pressure/volume curves as measured by the pressure bomb where changes in bulk leaf are frequently discussed as adaptations to certain treatments. The measurements of the hydraulic conductivity indicate that this parameter varies between treatments (range of means: 2.4×10-6 cm s-1 bar-1 to 13.4×10-6 cm s-1 bar-1). There was a negative correlation for L p in cells of intact leaves as a function of turgor which was altered by the growing conditions. However, a correlation with turgor could not be found for cells from isolated epidermis or cells from a uniform population of plants. The large variation in L p from cell to cell observed in the present and in previous studies was accounted for in a study of 100 cells from a uniform population of plants by the propagation of measurement errors in calculating L p . Summary The epidermal tissues of marine mussels can accumulate amino acids from surrounding sea water. In the present study, gill tissue isolated from the California coastal mussel,Mytilus californianus, was used in conjunction with intact, actively pumping mussels to study epidermal transport processes. There appeared to be at least four pathways for this uptake: i) a -neutral pathway which transports taurine; ii) an -acidic pathway specific for substrates such as aspartate; iii) an -neutral pathway having a general specificity for this class of compound, but which also accepts the basic amino acid, lysine; and iv) a second -neutral pathway, also of broad specificity, capable of accepting the imino acid, proline, as a substrate. Replacement of Na in sea water with choline reduced uptake of leucine, taurine, aspartate, and proline by more than 95%, and reduced lysine uptake by 75%, suggesting that Na-independent pathways play no significant role in epidermal transport in the gill. Isolated gill tissue was used to estimate the maximum transport capacities (J max's) of the pathways, which ranged from approximately 5 to 25 mol/(g·hr). Apparent Michaelis constants (K t *'s) of the epidermal transporters were estimated using a convection-diffusion model introduced previously (Wright and Secomb, Am J Physiol 247:R346–R355, 1984). TheseK t *'s ranged from 1 to 5 M. Summary The total and available forms of manganese in high organic matter content soils are reported. Most of the manganese is retained in an unavailable form either by organic matterper se or by some components of organic matter and, as a result, the available forms are extremely low and fall below the critical limits. Retention studies indicate that, exchange processes in the soil are more responsible for the retention of added manganese than redox processes. Organic matter as such does not quantitatively represent the component or components that influence the manganese retentive capacity of the soil. There is an indication to suggest that, if most of the total manganese is in organic form then fixation of added manganese various inversely with total manganese content.  相似文献

Fitting a double negative exponential function to N mineralization data can be used to characterize two organic nitrogen pools; an easily decomposable (Ndpm) and a resistant one (Nrpm). The relevance of those two calculated N mineralization pools was investigated by adding easily decomposable organic material to soils. Soil amended with crop residues of sugar-beet or bean was mixed with an equal amount of coarse sand, incubated at 35 °C and leached at specific time-intervals. Upon leaching, NH4 + and NO3 - were measured in the extracts. A double negative exponential function was fitted to the data and two organic N pools were defined. Fitting a double negative exponential function to N mineralization data to characterize an active and resistant organic N pool was sometimes impossible; the N mineralization data did not always resemble a negative exponential function. Additionally, the size of the two pools calculated were not constant with time and were often meaningless; the Nrpm pool was greater than the soil organic N content, the size of the Nrpm pool was smaller than the Ndpm pool or one of the N pools was negative. Relevant values for both Nrpm and Ndpm which were consistent with incubation time were only obtained when excessive amounts of organic material, normally not dealt with in the field, were applied.  相似文献

Summary Effects of increasing rates of lime and phosphorus addition on concentrations of available nutrients in soil and on P, Al and Mn uptake by two pasture legumes, lotus (Lotus pedunculatus Cav.) and white clover (Trifolium repens L.), were studied in a pot experiment using a highly leached acid (pH 4.2) soil.Liming resulted in an increase in exchangeable Ca and thus in percentage base saturation, with concomitant decreases in levels of exchangeable Al, Fe and Mn. The relationship between exchangeable Ca and Al was linear and negative with a gradient of almost unity. Liming had no consistent effect on measured CEC values. Increasing lime rates significantly reduced concentrations of Mg, K and Na in saturation paste extracts but had no effect on exchangeable Mg, K and Na levels.With increasing lime additions, available phosphate indicesviz water soluble, resin-, Morgan-and Williams-extractable all decreased significantly, Truog-extractable was unaffected, while Brayextractable generally increased. Fractionation revealed that lime additions caused a decrease in easily soluble, Fe-bound and to a lesser extent Ca-bound phosphate fractions, had no effect on reductant soluble phosphate, but resulted in an increase in the Al-bound fraction. P uptake and yield of both legumes increased with lime and P additions.Correlations between available phosphate indices and yield of both legumes were weak or nonsignificant. However, high, significant positive correlation coefficients were found between available phosphate and plant uptake of P. Indices of available Al and Mn were not generally significantly correlated with plant uptake of Al or Mn but significant negative correlations were found between available Al and Mn and yield of both species.  相似文献

Four new Proteus O-specific polysaccharides were isolated by mild acid degradation from the lipopolysaccharides of P. penneri 28 (1), P. vulgaris O44 (2), P. mirabilis G1 (O3) (3), and P. myxofaciens (4), and their structures were elucidated using NMR spectroscopy and chemical methods. They were found to contain non-carbohydrate organic acids, including ether-linked lactic acid and amide-linked amino acids, and the following structures of the repeating units were established: 3)--L-QuipNAc-(13)--D-GlcpNAc-(16)--D-GlcpNAc-(1 (S)-Lac-(2–3) (1) 4)--D-GlcpA-(13)--D-GalpNAc-(14)--D-Glcp-(13)--D-Galp-(14)--D-GalpNAc-(1 L-Ala-(2–6) (2) 3)--D-GalpNAc-(16)--D-GalpNAc-(14)--D-GlcpA-(1 L-Lys-(2–6)--D-GalpA-(14) (3) 4)--D-GlcpA-(16)--D-GalpNAc-(16)--D-GlcpNAc-(13)--D-GlcpNAc-(1 (R)-aLys-(2–6) (4) where (S)-Lac and (R)-aLys stand for (S)-1-carboxyethyl (residue of lactic acid) and N-[(R)-1-carboxyethyl]-L-lysine (alaninolysine), respectively. Ecological conditions were assessed in the zone of river and sea water mixing in the northern Caspian Sea regions exposed to high anthropogenic pressure. We studied the regions of the Volga River foredelta, the water areas adjoining ancient hydrological transects, and the sea regions between the transects. The water areas evaluated by analyzing biochemical degradation of organic compounds (proteinase and amylase activities) in the water and bottom sediments as well as hydrochemical indices of water (salt composition and the contents of dissolved oxygen and biogenic compounds). Under conditions of the active growth of phytoplankton and higher aquatic plants soon after the highest wave of the Volga flood, we studied the distribution of hydrolytic enzyme activities involved in the global production/destruction cycle of high-molecular organic compounds (proteins and polysaccharides). The data obtained with the trypsin–azocasein and -amylase–modified starch tests indicates pollution of water and surface sediments in the lower reaches of Volga and the inflow sea regions.  相似文献

Summary The cytoskeleton of primary tissue-culture cells from the epidermis of Xenopus laevis tadpoles was investigated by phase-contrast, immunofluorescence, and electron microscopy. The connection between the arrangement of different types of filaments and the mechanical properties of the epidermis is discussed. The bilayered epidermis attains stability from thick bundles of tonofilaments interconnecting the basal desmosomes. Twisting of tonofilaments around each other can explain the occurrence of elastic filamentous curls forming a meshwork braced between rows of small desmosomes in the apical region of the epidermis. Actin is arranged as a diffuse meshwork and sometimes forms bundles intermingling with tonofilament bundles. Surface membranes and rows of small desmosomes are delineated by actin and contain -actinin. Actin raises the tension for rounding and spreading of cells. Microtubules stabilize already well-developed lamellae.  相似文献

The function of the epidermis in auxinmediated elongation growth of maize (Zea mays L.) coleoptile segments was investigated. The following results were obtained: i) In the intact organ, there is a strong tissue tension produced by the expanding force of the inner tissues which is balanced by the contracting force of the outer epidermal wall. The compression imposed by the stretched outer epidermal wall upon the inner tissues gives rise to a wall-pressure difference which can be transformed into a water-potential difference between inner tissues and external medium (water) by removal of the outer epidermal wall. ii) Peeled segments fail to respond to auxin with normal growth. The plastic extensibility of the inner-tissue cell walls (measured with a constant-load extensiometer using living segments) is not influenced by auxin (or abscisic acid) in peeled or nonpeeled segments. It is concluded that auxin induces (and abscisic acid inhibits) elongation of the intact segment by increasing (decreasing) the extensibility specifically in the outer epidermal wall. In addition, tissue tension (and therewith the pressure acting on the outer epidermal wall) is maintained at a constant level over several hours of auxin-mediated growth, indicating that the inner cells also contribute actively to organ elongation. However, this contribution does not involve an increase of cell-wall extensibility, but a continuous shifting of the potential extension threshold (i.e., the length to which the inner tissues would extend by water uptake after peeling) ahead of the actual segment length. Thus, steady growth involves the coordinated action of wall loosening in the epidermis and regeneration of tissue tension by the inner tissues. iii) Electron micrographs show the accumulation of striking osmiophilic material (particles of approx. 0.3 m diameter) specifically at the plasma membrane/cell-wall interface of the outer epidermal wall of auxin-treated segments. iv) Peeled segments fail to respond to auxin with proton excretion. This is in contrast to fusicoccin-induced proton excretion and growth which can also be readily demonstrated in the absence of the epidermis. However, peeled and nonpeeled segments show the same sensitivity to protons with regard to the induction of acid-mediated in-vivo elongation and cell-wall extensibility. The observed threshold at pH 4.5–5.0 is too low to be compatible with a second messenger function of protons also in the growth response of the inner tissues. Organ growth is described in terms of a physical model which takes into account tissue tension and extensibility of the outer epidermal wall as the decisive growth parameters. The carotenoids in the molluscsClanculus cruciatus, Patella coerulea, Mytilus galloprovincialis, Sepia officinalis andLoligo vulgaris from the Adriatic sea were investigated. Their presence was determined by means of columnar and thin-layer chromatography. Perpetual cultures ofCaulobacter crescentus were maintained in a chemostat and provided with media that provided glucose, glutamate, ammonium chloride, and Na/K phosphate as sources, respectively, of C, of C and N, of N and of P. All cultures were maintained at the same relative flow rate, f/V, so that reproductive rate was constant. As the CNP ratio was shifted from a balanced ratio of 100 g C9–10 g N1 g P, the cells modulated their rates of uptake, reducing the rates of uptake of excess element-sources and accelerating the uptake of potential sources of the limiting element. The change in N-limited cells was the greatest, resulting in a nearly 150-fold increase in rate of uptake of amino acids. Poly--hydroxybutyrate or polyphosphate was stored whenever C or P, respectively, was not the limiting element. Direct measurement of cell and stalk surface areas on purified peptidoglycan sacculi of P-limited cells revealed that the surface of both swarmer and stalked cells, as well as the stalk surface, participated in the accelerated rate of phosphate uptake. Swarmers from N-limited populations were the only cells that exhibited chemotactic responsiveness-to methionine, NH4Cl and glutamate, but not to glucose or phosphate-sufficient to be detectable in a microcapillary assay. In populations that were C-limited and provided with ammonium ions as the principal source of N, morphogenesis and reproduction deteriorated and a steady state could not be maintained. Generally, the responses ofC. crescentus were appropriate to manage unbalanced nutrient supplies, but this oligoheterotrophic organism did not tolerate excesses of inorganic nutrients when limited for its sources of C, of energy, and of ammonium acceptor.  相似文献

In a study of photosynthetic energy storage efficiency (ES), the adaxial surface of the leaves of Vallisneria americana exhibited the highest ES values (22%) of the four aquatic plants examined. V. americana leaves have a dispersed structure and it was possible to measure the energy storage properties of the epidermal cells independently of the rest of the leaf. The abaxial epidermis had a higher value of ES at zero light fluence than the adaxial epidermis but ES in the abaxial epidermis declined much more rapidly with light fluence. Thus the abaxial epidermis is more suited to lower light fluences than the adaxial epidermis. ES declined as the pH rose from 4.0 to 8.0 at a constant dissolved inorganic carbon concentration. This paralleled the change from carbon dioxide to bicarbonate and suggests that these leaves utilise CO2 more efficiently than bicarbonate. ES increased by about 50% at pH 8.0 as leaf sections further from the leaf tip were examined which demonstrates that the older epidermal cells are less well able to use bicarbonate. Exposure to 30 min of a saturating light fluence caused the epidermal chloroplasts to move from the periclinal walls to the anticlinal walls. This decreased the photothermal signal by increasing the thermal diffusion distance and lowering the light fluence due to greater chloroplast shading. The latter effect increased ES.
