The tribe Hyoscyameae (Solanaceae) is restricted to Eurasia and includes the genera Archihyoscyamus, Anisodus, Atropa, Atropanthe, Hyoscyamus, Physochlaina, Przewalskia and Scopolia. Even though the monophyly of Hyoscyameae is strongly supported, the relationships of the taxa within the tribe remain unclear. Chloroplast markers have been widely used to elucidate plant relationships at low taxonomic levels. Identification of variable chloroplast intergenic regions has been developed based on comparative genomics of chloroplast genomes, but these regions have a narrow phylogenetic utility. In this study, we present the chloroplast genome sequence of Hyoscyamus niger and make comparisons to other solanaceous plastid genomes in terms of gene order, gene and intron content, editing sites, origins of replication, repeats, and hypothetical open reading frames. We developed and sequenced three variable plastid markers from eight species to elucidate relationships within the tribe Hyoscyameae. The presence of a horizontally transferred intron in the mitochondrial cox1 gene of some species of the tribe is considered here a likely synapomorphy uniting five genera of the Hyoscyameae. Alternatively, the cox1 intron could be a homoplasious character acquired twice within the tribe. A homoplasious inversion in the intergenic plastid spacer trnC-psbM was recognized as a source of bias and removed from the data set used in the phylogenetic analyses. Almost 12 kb of plastid sequence data were not sufficient to completely resolve relationships among genera of Hyoscyameae but some clades were identified. Two alternative hypotheses of the evolution of the genera within the tribe are proposed.  相似文献

Cytological studies were carried out on 14 taxa belonging to Amitostigma , Chusua , Galearis , Habenaria , Hemipilia , Hemipiliopsis , Herminium , Peristylus and Ponerochis , collected mostly from the south-eastern part of the Hengduan Mountain Region, south-west China. Cytological data on 11 of the taxa are reported for the first time. The interphase nuclei were either of the simple chromocentre type or intermediate between simple and complex chromocentre types. The nuclear morphology of Hemipiliopsis at interphase supports the conclusion that it is related more closely to Chusua and Ponerochis than to Habenaria . At the whole tribe level, however, the results did not indicate a clear correlation between morphological features of the interphase nuclei and phylogeny. The somatic chromosome numbers were 2 n  = 42 in ten species and 2 n  = 32, 38, 40, 64 and 72 in four species. The chromosome counts of 2 n  = 32 and 64 in Habenaria aitchsonii are rare in the genus. It is proposed that the repeated change of chromosome number from x  = 7 to x  = 8 has played an important role in the evolution of the tribe Orchideae. 利用扫描电镜研究了茄科 (Solanaceae)天仙子族 (Hyoscyameae)中国特有属马尿泡属 (PrzewalskiaMaxim .)马尿泡 (PrzewalskiatanguticaMaxim .)和天仙子属 (HyoscyamusL .)天仙子 (HyoscyamusnigerL .)的花器官发生和发育 ,研究结果表明 :马尿泡和天仙子花器官的发生和发育具有以下 3个共同特征 :1)符合Hofmeister规律 ,即新器官的发生首先出现在花顶已经存在的器官之间 ;2 )花冠的发育模式符合茄科植物所具有的“后合瓣”(“latesympetaly”)现象 ,即花瓣单独发生但后来又通过它们基部分生组织的融合而连合起来 ;3)花被五基数且花器官原基发生顺序为向心发育。但是它们的花萼原基具有不同的发生方式。天仙子花萼裂片原基的发生方式为环状发生 ;马尿泡花萼裂片原基的发生方式为螺旋状发生 ,但 5个花萼裂片原基在都出现后就连成了一个环。马尿泡是介于天仙子属和山莨菪属之间的类群 ,它比天仙子属原始但较山莨菪属进化。  相似文献

The chromosome numbers of 27 populations of Buddleja , comprising 14 species, were counted. The basic chromosome number of all species was x  = 19, confirming previous reports. Different ploidy levels (2 n  = 38, 76, 114, 228) were observed in these taxa, representing diploids, tetraploids, hexaploids, and dodecaploids, respectively. The chromosome numbers of B. yunnanensis , B. brachystachya , and B. macrostachya are reported for the first time. The tetraploid 2 n  = 76 is a new ploidy level for B. myriantha . Particular attention was given to B. macrostachya , because of the variation in morphology and ploidy level between isolated populations of this species. Two types of interphase nuclei were recognized: the complex chromocentre type in B. macrostachya and the simple chromocentre type in the other species. Biogeographically, most of the polyploidy in the Asiatic species occurs in the Sino-Himalayan region. It seems to be associated with the uplift of the Himalayan Mountains, the orogeny of this region playing an important role in the evolution of polyploidy in these taxa.  © 2007 The Linnean Society of London. Karyological studies were carried out on ten populations comprising six species of Soroseris in the tribe Lactuceae (Asteraceae), all endemic to the alpine Sino-Himalayan region and poorly known cytogenetically. The single species of Stebbinsia (one population) and two species of Syncalathium (three populations) were also examined for the first time. The basic chromosome number for the three genera is x  = 8. Stebbinsia and most species of Soroseris are diploids with 2 n  = 2 x  = 16 = 14m + 2sm and have a karyotype asymmetry type 1A. Three species of Soroseris are tetraploid (2 n  = 4 x  = 32 = 28m + 4sm). A karyotype of 2 n  = 2 x  = 16 = 14m(2SAT) + 2sm with type 1A asymmetry was found in Syncalathium kawaguchii , and of 2 n  = 2 x  = 16 = 6m + 10sm with type 2A asymmetry for two populations of Syncalathium souliei . The relationships between Soroseris and the other two genera are discussed. Floral organogenesis and development of Przewalskia tangutica Maxim.endemic to China and Hyoscyamus niger L., which belong to the tribe Hyoscyameae (Solanaceae), were studied using scanning electron microscope. They have three common characters of floral organ initiation and development: 1) initiation of the floral organs in the two species follows Hofmeister’s rule; 2) the mode of corolla tube development belongs to the “late sympetaly” type; 3) primordia of the floral appendages initiated in a pentamerous pattern and acropetal order. But initiation of the calyx lobe primordia showed different modes in these two species. The calyx lobe primordia of H. niger have simultaneously whorled initiation, while those of P. tangutica have helical initiation, but the five calyx lobe primordia form a ring after all five calyx lobe primordia occur. The systematic significance of the present results in the genera Hyoscyamus and Przewalskia is discussed in this paper.  相似文献

Chromosome numbers and morphology in 22 populations belonging to 11 taxa of Aconitum subgenus Lycoctonum (Ranunculaceae) from China were studied. Some taxa were diploid, with 2 n  = 16, but four species and two varieties were found to be tetraploid, with 2 n  = 32. They are concentrated in the Hengduan Mountains region in south-west China, indicating that polyploidy could have played an important role in the speciation of the subgenus in this region, one of the areas of the world with a high concentration of endemic species. The karyotypes of ten species of Holcoglossum (Orchidaceae), a highly endangered and diversified genus from China, were investigated to study the infrageneric relationships, biogeography, and speciation patterns in the Hengduan Mountains. The karyotype formulae of the studied species are as follows: 2 n  = 38 = 20m + 18sm in H. subulifolium , 2 n  = 38 = 22m + 16sm in H. amesianum , 2 n  = 38 = 26m + 12sm (6 SAT) in H. lingulatum , 2 n  = 38 = 26m + 12sm in H. wangii , 2 n  = 38 = 10m + 28sm in H. kimballianum , 2 n  = 38 = 14m + 22sm + 2st in H. flavescens , 2 n  = 38 = 24m + 12sm + 2st in H. rupestre , 2 n  = 38 = 14m + 20sm + 4st in H. sinicum , 2 n  = 38 = 16m + 14sm + 8st in H. weixiense , and 2 n  = 76 in H. tsii . The karyotypes of two tropical species, H. amesianum and H. subulifolium , are the most primitive in the genus, whereas those of four temperate alpine species, H. sinicum , H. rupestre , H. weixiense , and H. flavescens , are more advanced. H. tsii is a tetraploid and H. rupestr e may be one of its ancestors. The low frequency of polyploidy in Holcoglossum in the Hengduan Mountains region supports the conclusion that chromosome stasis during rapid speciation is common there.  © 2007 The Linnean Society of London. Twenty-two chromosome counts of 19 taxa (21 populations) in the tribe Anthemideae and one member (one population) of the tribe Inuleae of the family Asteraceae are reported. The Anthemideae studied belong to the subtribes Artemisiinae (14 Artemisia taxa, and one species each of the genera Dendranthema , Filifolium and Neopallasia ) and Tanacetinae (one species each of the genera Lepidolopha and Tanacetopsis ). From the Inuleae, we studied one Inula species. Five counts are new reports (including two at generic level), six are not consistent with previous counts and the remainder are confirmations of very limited (one to four records) previous data. Most of populations of Anthemideae studied have the basic chromosome number x  = 9, with ploidy levels ranging from 2 x to 10 x . Dysploidy is also present, with two x  = 8 diploid taxa. The karyomorphology of 11 species of the genus Incarvillea Juss. is reported. The chromosome numbers of all species studied are 2 n  = 22. The interphase nuclei and prophase chromosomes were found to be of the simple chromocentre type and the interstitial type, respectively. The asymmetry of the karyotype of I. arguta (two populations) in subgenus Amphicome is type 2A. The karyotypes of Incarvillea s inensis var. sinensis , I.  s inensis var. przewalskii, and I. olgae in subgenus Incarvillea are of asymmetry type 3A. The remaining nine species and one variety in subgenus Pteroscleris are also of asymmetry 3A. Data on three species and one variety studied are first reports. This study indicates that karyotype variation at the diploid level appears to be the predominant feature of chromosome evolution in the genus Incarvillea . According to this study of karyomorphology, morphological characteristics and geographical distribution, it seems that the three subgenera should be regarded as three independent genera. 横断山区是中国柴胡属Bupleurum植物的分布中心。本文对横断山区6个种2变种进行了染色体记数报道,其中4个种2变种是首次报道。对横断山区的10个种4个变种、中国北方(河北和黑龙江)的3个种的nrDNA ITS进行测序,同时从GenBank里面下载同属的来自非洲和地中海西部的16个nrDNA ITS序列数据,结合染色体数目变化结果,初步探讨了横断山区柴胡属植物的系统发育。结果表明横断山区可能是现代柴胡属植物的频度中心和多样分布中心之一。它们的祖先种可能是非洲北部的木本柴胡属植物B.fruticosum,或者是地中海西部的柴胡属植物,推测是通过中东和高加索扩散而形成的,其中与非洲南部特有种B.mundtii的亲缘关系也较近;染色体基数演化趋势是:8是较原始基数,6和7是次生基数,其染色体异基数变异和多倍化可能是物种形成、进化以及向外扩散的主要方式;在ITS系统发育树中,中国柴胡属植物染色体基数为8的种类聚为一支,染色体基数为6和7的种类聚为了一支,不支持舒璞等(1998)关于中国柴胡属的属下分类系统。结合已有的形态学、细胞学、孢粉学证据和ITS系统发育树,建议窄竹叶柴胡B.marginatum var.stenophyllum独立成种。  相似文献

This study encompasses 25 chromosome counts of 18 species in the subtribe Artemisiinae (tribe Anthemideae) of the family Asteraceae, from Mongolia. Most (15 species) belong to Artemisia , the largest genus of the subtribe, whereas the others come from two genera very closely related to it: Ajania (two species) and Neopallasia (one species). Eleven counts are new reports, three are not consistent with previous reports and the remainder confirm scanty earlier information. The majority of the species have x  = 9 as their basic chromosome number, but there are some taxa with x  = 8. Ploidy levels range from 2 x to 6 x . A karyomorphological study of two species in Asteropyrum (Ranunculaceae) and five in presumably related genera (one each in Caltha , Coptis , and Dichocarpum and two in Thalictrum ) reveals close similarities between Asteropyrum and Caltha , but obvious differences between Asteropyrum and Coptis , Dichocarpum , and Thalictrum , in some karyological features, such as the structure of the interphase nuclei, chromosome size, basic chromosome number, and condensation behaviour of chromosomes during the mitotic prophase. The chromosomes of Asteropyrum are categorized as being of the R-type and the basic chromosome number is confirmed as x  = 8, indicating a possible close affinity of Asteropyrum to members in the subfamily Helleboroideae, in particular to Caltha and its allies. Asteropyrum peltatum and Asteropyrum cavaleriei are shown to be more or less differentiated from each other in gross morphology and palynology, and somewhat independent in geographical distribution, albeit with intermediate forms occurring where their distribution zones overlap. In Myrtaceae (Myrteae), the diploid chromosome number 2 n  = 2 x  = 22 is the most common, although variations of ploidy level occur, with some triploid (2 n  = 3 x  = 33) and tetraploid (2 n  = 4 x  = 44) records. Karyotype details in this group are scarce because the chromosomes are small (< 2 μm). In this work, we carried out a karyotypic analysis of 15 species of Myrtaceae grouped in different subtribes and genera. Measurements of chromosome length (long arm, L ; short arm, S ) were taken and several karyotypic parameters were calculated for each species. Despite the abundance of woody legumes in Brazilian Amazonian rain forests, there are few chromosome counts on the native species of this important region. The present work presents such data for 13 species of Caesalpinioideae (the genera Bauhinia , Caesalpinia , Cassia , Chamaecrista and Senna ) and Papilionioideae ( Bowdichia , Centrosema and Dioclea ) collected from 17 natural populations. Our report represents the first chromosome counts for the genera Bowdichia and Dioclea and for four of the studied species. Observations are made on chromosome morphology, size, condensing behaviour and interphase nucleus structure. Banding with fluorochromes carried out for the first time in Caesalpinioideae revealed discrete CMA+/DAPI terminal (GC-rich) bands on 2–4 chromosome pairs of most species analysed, with a few species presenting discrete CMA/DAPI+ (AT-rich) bands. Significant differences in chromosome size, morphology and condensing behaviour were observed among members of the controversial tribe Cassieae ( Cassia , Chamaecrista and Senna ), revealing the tribe to be a heterogeneous group from the karyological point of view.  © 2004 The Linnean Society of London, Botanical Journal of the Linnean Society , 2004, 144 , 181–191.  相似文献

Cytological studies were carried out for the first time on five populations of Trigonobalanus doichangensis in China and Thailand. In all populations, the pattern of interphase nuclei was of the simple chromocentre type, the mitotic prophases were of the proximal interstitial type and chromosome numbers were 2 n  = 2 x  = 14. Two B chromosomes were commonly observed at prophase and prophase–metaphase, but rarely at metaphase. Karyotype variation among the populations at the diploid level was limited, but there were some distinguishing cytological characters. Based on the comparison of all the available data on cytology, taxonomy, phytogeography and molecular systematics related to the genus Trigonobalanus , we recommend that the three species of Trigonobalanus comprise the subfamily Trigonobalanoideae and that the genus should not be segregated into three monotypic genera.  © 2007 The Linnean Society of London, Botanical Journal of the Linnean Society , 2007, 154 , 321–330.  相似文献

There are 20 Bellevalia species in Turkey, half of which are endemic. Chromosome numbers are known for 15 species. A chromosome survey of 145 Bellevalia individuals showed that the karyotype is remarkably stable. All are based on x  = 4. The majority are diploid with 2 n  = 8, but there is also a polyploid series of 2 n  = 16, 24 and 32. Aneuploidy occurs only at the octoploid level. Eleven individuals had metacentric B chromosomes, one had acrocentric Bs and one had telocentric Bs. 首次报道了唇形科Lamiaceae独一味属Lamiophlomis和五种糙苏属Phlomis植物的染色体数目和核型。它们的间期核均属球状前染色体型,有丝分裂前期染色体均为中间型。中期染色体核型公式如下:独一味L.rotata,2n=2x=22=18m 4sm;糙苏P.umbrosa,2n=2x=22=22m;裂萼糙苏P.ruptilis,2n=2x=22=22m;假秦艽P.betonicoides,2n=2x=22=22m;黑花糙苏P.melanantha,2n=2x=22=22m(2sat);糙毛糙苏P.strigosa,2n=6x=66=60m 6sm;染色体基数均为x=11。这表明独一味与糙苏属的糙苏组sect.Phlomoides植物具有相同的染色体基数,反映出两者较为密切的系统演化关系,推断独一味很可能是糙苏属的一个种。  相似文献
