The partial sterility of hybrids has been a major barrier for utilization of the strong heterosis expressed in hybrids between Oryza sativa ssp. indica and O. sativa ssp. japonica. Wide-compatibility varieties, comprising a special class of germplasm, are able to produce fertile hybrids when crossed to both indica and japonica varieties. However, all the work on wide compatibility and majority of studies on indica/japonica hybrid sterility reported so far were based only on spikelet fertility; thus, it is not known to what extent male and female gamete abortions influence hybrid sterility. In this study, we investigated pollen fertility, embryo sac fertility, and spikelet fertility in an F1 population of 202 true hybrid plants derived from a three-way cross (02428/Nanjing 11//Balilla). A partial regression analysis showed that the pollen and embryo sac fertility contributed almost equally to spikelet fertility. QTL analysis based on a linkage map of 191 polymorphic marker loci identified two QTLs for pollen fertility, one QTL for embryo sac fertility, and three QTLs for spikelet fertility. The S5 locus, previously identified as a locus for wide compatibility by spikelet fertility analysis, is a major locus for embryo sac fertility, and a QTL on chromosome 5 had a major effect on pollen fertility. These two loci coincided with the two major QTLs for spikelet fertility. The study also detected a QTL on chromosome 8, showing a large effect on spikelet fertility but no effect on either pollen or embryo sac fertility. Very little interaction among the QTLs was detected. The implications of the findings in rice breeding programs are discussed.  相似文献

The development of the unisexual male and female flowers of Zea mays from bisexual initials in both tassels and ears has been reinvestigated with SEM and TEM. The early stages of spikelet branch primordia, spikelet initiation, and early flower development are similar in both flowers, though differences in rates of growth of glumes, lemmas, and palea were detected. In both tassel and ear flowers, a pair of stamens arises opposite the lemmas and a third stamen initiates later at right angles to the first pair but from a point on the meristem below its insertion. Gynoecia develop on both tassel and ear flowers first as a ridge which overgrows the apical meristem giving rise to the stylar canal and the elongate silk. Male flowers arise in the tassel through selective vacuolation and abortion of the cells of the early gynoecium. The single female flower in each ear spikelet arises through the vacuolation and abortion of stamens in the upper flower and the repression of growth of and the eventual regression of the lower flower in each spikelet. The significance of these selective organ abortions for practical applications is discussed.  相似文献

A compromise classification of the genus Zea, reflecting both phylogeny and practical needs, recognizes six taxa, as follows: Section Luxuriantes : Zea perennis. Zea diploperennis, Zea luxurians. Section Zea : Zea mays ssp. mexicana (Neo-volcanic Plateau), Zea mays ssp. parviglumis Iltis & Doebley ssp. n. var. parviglumis (Rio Balsas drainage, Pacific slope from Guerrero to Jalisco), Zea mexicana ssp. parviglumis var. huehuetenangensis Iltis & Doebley var. n. (Pacific slope, western Guatemala, Prov. Huehuetenango), Zea mays ssp. mays. The new subspecies is distinguished by smaller spikelets and rachis joints, the varieties by different habitats, blooming dates and their genetic behavior in relation to cultivated Zea mays. The pattern of expression of two genes coding for proteins rich in proline, HyPRP (hybrid proline-rich protein) and HRGP (hydroxyproline-rich glycoprotein), has been studied in maize (Zea mays) embryos by RNA analysis and in situ hybridization. mRNA accumulation is high during the first 20 d after pollination, and disappears in the maturation stages of embryogenesis. The two genes are also expressed during the development of the pistillate spikelet and during the first stages of embryo development in adjacent but different tissues. HyPRP mRNA accumulates mainly in the scutellum and HRGP mRNA mainly in the embryo axis and the suspensor. The two genes appear to be under the control of different regulatory pathways during embryogenesis. We show that HyPRP is repressed by abscisic acid and stress treatments, with the exception of cold treatment. In contrast, HRGP is affected positively by specific stress treatments.  相似文献

The enzymatic maceration method was used to isolate an intact embryo sac ofCrinum asiaticum and its component cells. Best results were obtained when using enzyme solutions that contained pectinase hemicellulase, cellulase and pectolyase. Aseptic ovules were incubated in the enzyme solution for 1.5 hr at 25 C. This allowed the isolation of embryo sacs to yield up to 20% of the amount present. An isolated embryo sac usually consists of an egg cell, synergids, antipodals and a central cell. Some embryo sacs can be digested as gametophytic protoplast. The size, shape and position of the isolated embryo sac seemingly possessed similarities with those of the fixed embryo sac in the ovary. An isolated embryo sac can be in a living state when the result of the fluorochromatic reaction (FCR) and protoplasmic streaming is positive. When cultured in proper media, 68% of the isolated gametophytic protoplasts were observed to have sustained their positive FCR for more than 1 month.  相似文献

The ontogeny of tassels and ears in two annual Mexican teosintes, Zea mays subsp. mexicana and Z. mays subsp. parviglumis, was examined using scanning electron microscopy and light microscopy. Ear development in these annual teosintes follows a pattern previously described as leading to the bisexual mixed inflorescence in Z. diploperennis. Common bud primordia are initiated in the axils of distichously arranged bracts along the ear axis. These common primordia bifurcate to form paired sessile and pedicellate spikelet primordia. Development of pedicellate spikelets is arrested leaving the sessile spikelets, along with the adjoining rachis segment, to form solitary grains enclosed within cupulate fruitcases. Development of the central tassel spike is similar to that previously described in the Z. diploperennis tassel, except that the first formed axillary bud primordia form precocious tassel branches. The origin of these tassel branches suggests a possible mechanism for the transition from a distichous spike, characteristic of teosinte, to a polystichous spike, typical of maize.  相似文献

Summary Various developmental phases can be distinguished in the definition of the archesporium and the early life of the embryo, takingZea mays (maize) as a model within the family Gramineae, and other families where pertinent: (1) the isolation of the megasporocyte and the functional spore derived from it; (2) the maturation of the specialized walls of the embryo sac, and their reinforcement by ensheathments derived from the contiguous nucellar cells during a sequence of phased genetic ablation; (3) the differentiation of the synergids, the associated flange, and the filiform apparatuses; (4) the blocking of the pollen tube pathway by secondary secretions in the micropylar region and the coagulation of the pollen tube cytoplasm within the filiform apparatuses during the process of fertilization; and finally (5) the development of a compound cutinized envelope of four fused layers (six where the outer integument is also involved) after fertilization. For the nascent haploid generation, the period of maximum vulnerability in respect to both pathogen invasion and the transition from diplophase control occurs during these phases. It is concluded that many of the protective features form a prophylactic shield and are key components of the angiosperms in general, which may have contributed to their evolutionary success as a group. Other physiological or biochemical adaptations or barriers may also supplement the mainly structural features described here.  相似文献

One day after pollination, embryo sacs of Zea mays, containing zygotes and a few endosperm nuclei, were partially isolated and grown in culture. The zygotes underwent normal embryo development, and 40% germinated on a modified Murashige and Skoog medium with 0.1 mg/liter 6-benzylaminopurine. After the appearance of a root and a shoot, individual plantlets were transferred to soil and grown to sexual maturity. The techniques for partial mechanical isolation and successful growth to maturity of such young embryos provide the tools necessary for the recovery of plants from in vitro-fertilìzed embryo sacs.  相似文献

The molecular and genetic control of inflorescence and flower development has been studied in great detail in model dicotyledonous plants such as Arabidopsis and Antirrhinum . In contrast, little is known about these important developmental steps in monocotyledonous species. Here we report the analysis of the Zea mays mutant branched silkless1–2 (bd1–2) , allelic to bd1 , which we have used as a tool to study the transition from spikelet to floret development in maize. Floret development is blocked in the female inflorescence (the ear) of bd1–2 plants, whereas florets develop almost normally in the male inflorescence (the tassel). Detailed phenotypic analyses indicate that in bd1–2 mutants ear inflorescence formation initiates normally, however, the spikelet meristems do not proceed to form floret meristems. The ear spikelets, at anthesis, contain various numbers of spikelet-like meristems and glume-like structures. Furthermore, growth of branches from the base of the ear is often observed. Expression analyses show that the floral-specific MADS box genes Zea mays AGAMOUS1 ( ZAG1 ), ZAG2 and Zea mays MADS 2 ( ZMM2 ) are not expressed in ear florets in bd1–2 mutants, whereas their expression in tassel florets is similar to that of wild type. Taken together, these data indicate that the development from spikelet to floret meristem is differentially controlled in the ear and tassel in the monoecious grass species Zea mays , and that BRANCHED SILKLESS plays an important role in regulating the transition from spikelet meristem to floral meristem during the development of the female inflorescence of maize.  相似文献

为探讨多花地宝兰(Geodorum recurvum)胚胎发育的系统分类学意义,采用石蜡制片法对多花地宝兰胚囊和胚的发育进行解剖学观察。结果表明,在开花前,多花地宝兰胚珠原基发育缓慢,开花授粉后胚珠原基快速发育成"树状二杈分枝结构",随后在"分枝结构"末端形成孢原细胞,开始胚囊发育。多花地宝兰的胚囊发育属于单孢蓼型胚囊,胚珠具有双层珠被。孢原细胞形成后,经过细胞膨大延长发育形成胚囊母细胞,胚囊母细胞经过减数分裂形成线性四分体,在珠孔端形成1个功能大孢子,功能大孢子经过3次有丝分裂形成8核胚囊。多花地宝兰的胚发育具有藜型和紫苑型两种方式。双受精完成后,多花地宝兰合子进行一次橫裂后形成基细胞和顶细胞;基细胞经过多次分裂形成细胞团,细胞团中的细胞向不同方向膨大延长形成多个胚柄细胞;顶细胞有两种分裂方式,一种是横裂形成藜型胚,一种是纵裂形成紫苑型胚。因此,推测多花地宝兰在兰科植物系统分类学上属于较为原始种。  相似文献

Summary Wide hybridization is a useful tool in plant breeding, but little is known about its possible range. For the cereals, wheat, barley and rye, this was tested with 15 different species of the Poaceae and Panicoideae. Embryo formation could be obtained with Agropyron repens, Alopecurus agrestis, Dactylis glomerata, Festuca glauca, Hordeum bulbosum, Lolium perenne, Pennisetum americanum, and Zea mays. As well, haploid as diploid embryos occurred. New embryo culture techniques should enable these embryos to grow to plants.  相似文献

本文对普通栽培稻不同品种类型间杂种小穗败育的细胞学基础及雌性败育的过程进行了研究,结果表明:(1)引起杂种小穗败育的原因有胚囊败育、花粉败育、开花时花药不开裂和雌雄异熟。其中胚囊败育而丧失受精能力是引起低结实率的最重要的因素,开花时花药不开裂和雌雄异熟在一定程度上形成了雌雄性细胞时间和空间的隔离屏障。(2)杂种植株的所有大孢子母细胞都能进行正常的减数分裂,形成四个大孢子,败育主要发生在靠近合点端的功能大孢子分化形成胚囊的早期,有的功能大孢子在进行第一次有丝分裂前便萎缩解体,多数走向败育的功能大孢子能完成一次或二次有丝分裂,形成二核或四核败育胚囊。败育的共同特征是无液泡的分化,细胞质少或退化,在败育胚囊残迹部位,解体的珠心细胞和萎缩的胚囊残渍混杂垛叠。已受精的杂种子房没有观察到胚及胚乳发育的异常。籼粳杂种胚囊败育频率较高。  相似文献

Seeds of the longcell mutant in maize (Zea mays L) have a defective-kernel phenotype: the embryo aborts at the early coleoptilar stage and the endosperm is reduced in size. Mutant embryos have severe alterations in morphogenesis. They have a suspensor-, an embryo axis- and a scutellum-like structure, but the shoot apical meristem (SAM) is not formed. Scanning electron microscopy showed that most of the cells in longcell embryos are tubular and abnormally enlarged. The level of expression of several genes involved in basic metabolism is not severely affected during early and mid embryogenesis, but storage molecule accumulation is reduced. Genes which in normal conditions are only expressed after germination, are expressed during kernel development in the longcell seeds. Mutant embryos undergo cell death in late embryogenesis. Nuclei in dying embryos are TUNEL positive, and different genes coding for hydrolytic enzymes are up-regulated. The expression of genes related to oxidative stress is also altered in longcell embryos. These results lead us to suggest that the longcell mutant may be cytokinesis-defective.  相似文献

Carbon-14, photosynthetically fixed in leaves of Zea mays L. and translocated to developing kernels, passed through specialized basal endosperm cells prior to movement into the starchy endosperm and embryo. Removal of the cap from the primary root of Zea mays activatescell division in the quiescent centre. Regeneration of a normal structure in the new cap isachieved by progressive changes in the structures of the cellorganelles, apparently in response to the position that thecells containing them occupy within the growing cellular ensembleat the root apex. Isozyme patterns of esterase and malate dehydrogenase were analyzed at different stages of in vitro culture of immature embryos and glumes of Zea mays L. viz. explant, callus formation, root formation and shoot formation. Significant changes in isoenzyme patterns of esterase and MDH were observed besides the appearance of specific and new isozymes. Specific fast migrating isozymes were noted in differentiating calli of embryo and glume calli which were absent at other stages suggesting a possible association of these isozyme patterns with in vitro differentiation.  相似文献
