Hordeum murinum L. is a species complex composed of related taxa, including the subspecies glaucum, murinum and leporinum. However, the phylogenetic relationships between the different taxa and their cytotypes, and the origin of the polyploid forms, remain points of controversy. The present work reports a comparative karyotype analysis of seven accessions of the H. murinum complex representing all subspecies and cytotypes. The karyotypes were determined by examining the distribution of the repetitive Triticeae DNA sequences pTa71, pTa794, pSc119.2, pAs1 and pHch950, the simple sequence repeats (SSRs) (AG)10, (AAC)5, (AAG)5, (ACT)5, (ATC)5, and (CCCTAAA)3 via in situ hybridization. The chromosomes of the three subgenomes involved in the polyploids were identified. All tetraploids of all subspecies shared the same two subgenomes (thus suggesting them to in fact belong to the same taxon), the result of hybridization between two diploid ancestors. One of the subgenomes present in all tetraploids of all subspecies was found to be very similar (though not identical) to the chromosome complement of the diploid glaucum. The hexaploid form of leporinum came about through a cross between a tetraploid and a third diploid form. Exclusively bivalent associations among homologous chromosomes were observed when analyzing pollen mother cells of tetraploid taxa. In conclusion, the present results identify all the individual chromosomes within the H. murinum complex, reveal its genome structure and phylogeny, and explain the appearance of the different cytotypes. Three cryptic species are proposed according to ploidy level that may deserve full taxonomic recognition.  相似文献

Genomic in situ hybridization (GISH) was used for a chromosomal composition study of the later generations of interspecific hybrids between A. cepa L. and A. fistulosum L., which are relatively resistant to downy mildew (peronosporosis). GISH revealed that F2 hybrids, which did not produce seeds, were triploids (2n = 3x = 24) with 24 chromosomes and possessed in their complements 16 chromosomes of A. fistulosum L. and eight chromosomes of A. cepa L. or eight chromosomes of A. fistulosum L. and 16 chromosomes of A. cepa L. The advanced F5 hybrid, which produced few seeds, was amphidiploid with 32 chromosomes. BC1F5 hybrid was triploid with eight chromosomes of A. fistulosum L. and 16 chromosomes of A. cepa L., which did not produce seeds. BC2 (BC1F5) plant was amphidiploid that possessed 4 recombinant chromosomes and produced few seeds. GISH results point to 2n-gametes formation in macro- and microsporogenesis of the hybrids. The mechanism of 2n-gametes formation and the possibility of apomixes events in the backcrossing progeny are discussed.  相似文献

Stachys recta L. is a very polymorphous species in which numerous subspecies were recognised. S. recta L. subsp. serpentini (Fiori) Arrigoni is a typical endemism growing on serpentine soils in northern Apennines and particularly in Tuscany (Italy). In order to contribute to a better knowledge of this plant and to its differentiation with respect to S. recta L. subsp. recta, the micromorphological characters (non-glandular and glandular trichomes) and the essential oil composition of the two subspecies were investigated. Micromorphological characters were studied using scanning and transmission electron microscopy, while light microscopy was used for histochemical observations. Essential oil analysis was carried out by gas chromatography and mass spectrometry.In the two examined taxa, the morphology and distribution of glandular and non-glandular trichomes, and the different essential oil composition, may be considered distinctive characters at subspecies level. This is consistent with the taxonomic classification considering S. recta subsp. serpentini a subordinate taxon of S. recta.  相似文献

Recombinational and mutational analyses were performed in an attempt to elucidate the nature of the genic differences and the ancestral relationship between the indica and japonica subspecies of Oryza sativa L. by taking representative varieties from each group. The absence of cytological aberrations at meiosis in indica×japonica hybrids suggest that genetic differences are predominantly genic rather than chromosomal. The pattern of variation induced in the key characters, and the frequency and spectra of mutations obtained in each group, failed to support the hypothesis that indica has given rise to japonica, and suggested that the two subspecies have originated from a common ancestor.  相似文献

The present paper deals with cytotaxonomy ofChelidonium majus L. s. 1. taxa and their hybrids. Based on results of hybridization experiments, cytology and reproductive isolation, a new combination,Ch. asiaticum (Hara) Krahulcová, is proposed. In nature, closely related species may hybridize while still retaining their distinctive identities. Chromosomal regions that experience reduced recombination in hybrids, such as within inversions, have been hypothesized to contribute to the maintenance of species integrity. Here, we examine genomic sequences from closely related fruit fly taxa of the Drosophila pseudoobscura subgroup to reconstruct their evolutionary histories and past patterns of genic exchange. Partial genomic assemblies were generated from two subspecies of Drosophila pseudoobscura (D. ps.) and an outgroup species, D. miranda. These new assemblies were compared to available assemblies of D. ps. pseudoobscura and D. persimilis, two species with overlapping ranges in western North America. Within inverted regions, nucleotide divergence among each pair of the three species is comparable, whereas divergence between D. ps. pseudoobscura and D. persimilis in non-inverted regions is much lower and closer to levels of intraspecific variation. Using molecular markers flanking each of the major chromosomal inversions, we identify strong crossover suppression in F1 hybrids extending over 2 megabase pairs (Mbp) beyond the inversion breakpoints. These regions of crossover suppression also exhibit the high nucleotide divergence associated with inverted regions. Finally, by comparison to a geographically isolated subspecies, D. ps. bogotana, our results suggest that autosomal gene exchange between the North American species, D. ps. pseudoobscura and D. persimilis, occurred since the split of the subspecies, likely within the last 200,000 years. We conclude that chromosomal rearrangements have been vital to the ongoing persistence of these species despite recent hybridization. Our study serves as a proof-of-principle on how whole genome sequencing can be applied to formulate and test hypotheses about species formation in lesser-known non-model systems.  相似文献

Summary Uptake and assimilation of nitrate by two subspecies ofHypochaeris radicata L. were investigated under laboratory conditions as well as in the field.H. radicata ssp.radicata grows on relatively nitrogen-richer soils thanH. radicata ssp.ericetorum. Attempts were made to relate nitrate uptake and nitrate assimilation in the two subspecies to their different distribution in the field.No differences between the two subspecies with respect to nitrate uptake and nitrate assimilation were observed under laboratory conditions. In plants from the field intact tissue nitrate reductase was higher in ssp.radicata than in ssp.ericetorum. Five early growth characters were examined in six races of Arabidopsis thaliana (L.) Heynh, their reciprocal F1 hybrids (1974) and F1 by tester hybrids, using a seventh race as a paternal tester. Three of the five characters were also examined at two nutrient levels in reciprocal F1 hybrids (1972) of all seven races. Analyses of F1 and F1 by tester hybrids revealed significant maternal effects in all characters examined in F1 hybrids (1972) and in root length and plant weight of F1 (1974) and F1 by tester hybrids. Significant reciprocal effects were found for plant weight in F1 by tester hybrids and for seed weight, percentage of germination and root length in F1 (1974) and F1 by tester hybrids. The presence of significant maternal and/or reciprocal components in both F1 (1974) and F1 by tester diallels suggests that differences in maternal cytoplasm rather than maternal genotype per se were responsible for much of the variation resulting from these non-direct genetic effects.  相似文献

Summary The present paper is part of a series of papers comparing two ecologically distinct subspecies ofHypochaeris radicata L.Hypochaeris radicata ssp.radicata grows in more fertile soils thanH. radicata ssp.ericetorum.The dry-weight yield of plants grown from seeds was measured after a 25 days culture period on varying levels of nitrate. Roots and shoots were analyzed for total organic nitrogen and free nitrate. Dry-weight yield showed an almost identical response over the whole range of nitrate concentrations used. The chemical analysis did not reveal differences between the subspecies that could explain the distribution in the field. In a second experiment the growth rate of the two subspecies was measured under optimal nitrate supply during a two month period. Both subspecies exhibited initially the same relative growth rate but one month after sowing the growth rate of both subspecies declined, with a sharper decline in ssp.ericetorum resulting in an 82 per cent difference in dry-weight between both subspecies two months after sowing. Some pronounced formative differences became evident. Shoot to root ratio of ssp.ericetorum showed little response to increasing nitrate concentrations whereas ssp.radicata was clearly responsive in this respect.The contribution of the tap root in total plant dry-weight was relatively higher in ssp.Ericetorum. The decline in growth rate and the formative differences can explain the absence of ssp.ericetorum from nutrient rich soils. when grown on ammonium ssp.radicata had a slightly but not significantly higher yield than ssp.ericetorum. The difference was even greater at low pH. It is concluded that both subspecies are adapted to ammonium nutrition at pH 4.5 as well as at pH 6 and that the absence of ssp.radicata from acidic soils cannot be explained by the low pH. The possibility that both subspecies have different abilities to reduce their growth when nitrate resources become limiting, and yet remain capable for further growth and reproduction is discussed.Grassland species research group, publ. no.16.  相似文献

Lindernia procumbens and L. dubia are common annual weeds in flooded rice fields of Japan. Two subspecies of L. dubia, subsp. major and subsp. dubia, are usually recognized in Japan but they are both regarded as synonyms of L. dubia elsewhere. In a cluster analysis based on AFLP, most L. dubia subsp. major formed a separate cluster from L. dubia subsp. dubia although 11% of haplotypes classified using AFLP were not coincident with classification using the shape of leaf bases, which is the commonly used identification trait. Artificial F1 plants between L. procumbens and L. dubia subsp. major, and between L. procumbens and L. dubia subsp. dubia did not produce seed. Forty percent of capsules produced by F1 plants from these two subspecies were slimmer and 80% pollen were sterile in slimmer capsules. However, seed number of most F1 capsules was not different from that of self-fertilized plants, suggesting that there was no complete reproductive isolation between the subspecies. Natural hybridization of these subspecies may have occurred but we are not aware of it because F1 plants are rare and F2 plants are indistinguishable from these subspecies.  相似文献

A dimorphic pattern of macrocephalic (wide, short) and stenocephalic (narrow, long) body shapes is observed in snail-feeding carabid beetles globally. The former exhibits high performance in crushing snail shells with powerful jaws, whereas the latter specializes in eating snails'' soft body directly by inserting the head into the shell. In the snail-feeding species Damaster blaptoides, the subspecies D. b. capito has a wide, short forebody, and D. b. fortunei has a narrow, long forebody. They exhibit distinct morphologies despite their geographic and phylogenetic proximity. To examine the genetic basis of the morphological differences between these two subspecies, we conducted quantitative genetic analyses by crossing these subspecies and producing F1 and backcross hybrids. The hybrids had body shapes intermediate between the parental subspecies. The variation between wide, short and narrow, long forebodies was based on negative genetic correlations between width and length of the head and thorax. Between one and eight genetic factors were involved in the morphological differences between subspecies. We suggest that the morphological integration of forebody parts in a small number of loci has facilitated the marked morphological diversification between subspecies of D. blaptoides.  相似文献

Summary The presence and activity of -aminolevulinic acid dehydratase (ALA-D, E.C.4.2.1.24) inZea mays L. was studied. Seedlings of several inbred lines and their hybrids were employed as enzyme source, and enzyme activity and the kinetic parameters (Vmax and KM) determined. The wide variations of the enzyme activity and kinetic parameters found in the tested lines support the hypothesis that ALA-D varies in concentration and isoenzyme distribution. Enzyme activity was greater in some hybrids than in their parent lines. As the same hybrids correspond to varieties appreciated for their high productivity and good morphological characteristics it is suggested that ALA-D activity levels could be adopted as an auxiliary parameter for establishing heterosis.  相似文献

Morphological variation within and between the taxa of the Altissimae (Vernonia altissima, V. ovalifolia, andV. flaccidifolia) and Giganteae (V. gigantea) was investigated. The data for these studies were obtained from population samples, herbarium specimens, and greenhouse-grown transplants. The taxa were hybridized in all possible combinations, and F2 progeny were produced fromV. altissima XV. ovalifolia F2’s. Fertility in the hybrids was estimated by examination of chromosome pairing at meiosis and by pollen stainability. It is concluded thatVernonia altissima should be regarded as a synonym ofV. gigantea and thatV. ovalifolia should be reduced to subspecific rank asV. gigantea ssp.ovalifolia. Anthyllis vulneraria L. (Fabaceae) s. lato includes many cryptic taxa, ranging from 25 to 60 subspecies according to different authors. The delimitation of intraspecific taxa of A. vulneraria s. lato has always been complicated and inconsistent. Different data sets (multivariate analyses of morphological variation, allozymes, chloroplast SSRs and ITS) have not resolved the existing problem with distinguishing some subspecies. We used the amplified fragment length polymorphism (AFLP) analysis to describe the differentiation in this species complex and to characterize variation on a geographic scale. Some correlation was found between genetic variability and geographic distribution (western-eastern directional variation), but AFLP data analysis did not reveal clear intraspecific structure of the seven analysed taxa. 23 species, one subspecies and three varieties have been described or named for the pan-tropical genus Lasianthus Jack (Rubiaceae) in continental Africa. In the present revision thirteen species, five subspecies and one variety are recognized; additionally two hybrids are proposed. Of the previously named taxa, the subspecies is raised to species rank whereas two species are degraded to subspecies and variety status, respectively. Five species are synonymised including two varieties, and four are left with uncertain status, including one variety. The distribution of the genus in Africa consists of two distinct elements: one component is found in the Guineo-Congolian regional centre of endemism and one in the eastern part of Afiomontane Archipelago-like regional centre of endemism. A centre of species diversity is found in Eastern Arc Mts., where six endemic species occur together with L. laxinervis and the more widespread L. kilimandscharicus. Four of the species are strict endemics of the Uluguru Mts. In addition to the presentation of the new section: Lasianthus Sectio Membranacei, sect. nov., this revision contain two hybrids from Uluguru Mts.: L. macrocalyx × pedunculatus and L. cereifizorus × pedunculatus, and following new taxa and new combinations: L. africanus ssp. biokoensis, subsp. nov.; L. africanus ssp. mayumbensis, comb. et stat. nov.; L. kilimandscharicus ssp. glabrescens, ssp. nov.; L. kilimandscharicus ssp. glabrescens var. xanthospermus, comb. et stat. nov.; L. kilimandscharicus ssp. hirsutus, ssp. nov.; L. laxinervis, stat. nov.; L. pedunculatus ssp. angustisepalus, ssp. nov.  相似文献

Inter-Simple Sequence Repeat (ISSR) molecular markers and morphological analysis were used in order to characterize wild populations and cultivated forms of orphan crop species Capparis spinosa L. in a Mediterranean island complex. Nineteen wild populations belonging to two different subspecies, C. spinosa subsp. spinosa and subsp. rupestris, were sampled in different environments in Sicily and the surrounding islets Lampedusa, Pantelleria and Salina. Different biotypes cultivated in Pantelleria and Salina were analysed. Six ISSR primers were selected for genetic characterization, and all clear and reproducible bands were scored and analysed. Among the 47 ISSR bands obtained, 97.5% were polymorphic. Results of AMOVA and STRUCTURE analysis suggested a clear genetic distinctness between subspecies at the regional level and suggested the existence of two taxonomic groups among wild populations, with different ecological preferences and distinctive morphological characters. Cultivated forms showed genetic affinity to subsp. rupestris. ISSR analysis not only provided specific molecular markers to discriminate the taxa, but also proved useful in supporting the hypothesis of a hybrid origin of the intermediate phenotypes found in overlapping distribution areas. The identified molecular markers provided a basic tool for the DNA fingerprinting of wild and commercial capers in the Mediterranean region and nearby territory.  相似文献

More than 50 collections of five species forming the Achillea nobilis group were analysed for their leaf flavonoid complement. Major accumulation trends were found to be C-glycosylflavones and flavonol 3-O-glycosides. The most common pattern consisted of the C-8-glycosylfiavones (vitexin and orientin), the C-6-glycosylflavone (isoörientin) together with minor amounts of di-C-glycosylapigenins and quercetin 3-O-glycosides. Additionally, C-6-glycosylflavones (isovitexin) and their 7-methyl ethers swertisin and swertiajaponin were sporadically accumulated, characterizing particularly two subspecies of A. nobilis. Whereas C-glycosylflavone dominated profiles were typical of most species, two taxa exhibited a flavonol dominated profile (A. ligustica; A. virescens p.p.). Regarding the infraspecific and interpopulational variations of flavonoid accumulation trends, their systematic and ecological significance is briefly discussed.  相似文献
