The role of environmental factors in seasonal succession of six clones of marine diatoms was examined by testing effects of various combinations of temperature (5°, 10°, 15°, 20°C) and photoperiod (10, 12, 14, 16 h) on time to germination of resting stages and subsequent vegetative cell division rates. Resting stages were induced in batch cultures using a low nutrient medium and a cold, dark period. A three-factor ANOVA showed that species, temperature, daylength, and their interactions signficantly (P =0.0001) injuenced germination time and division rate. Resting stages of Chaetoceros similis Cleve and Odontella aurita (Lyngbye) Agardh were most successful in producing populations of cells on warm, long days (20°C/16 h). Chaetoceros didymus Ehrenberg and C. cinctus Gran were most successful under more moderate conditions (15°C/14 h). Resting stages of Ditylum bright-wellii (West) Grunow germinated and cells divided rapidly under all conditions. No consistent pattern was evident with Thalassiosira rotula Meunier. We compared results of the laboratory study with environmental conditions prevailing during the presence of these species in a British Columbia fjord. Favorable growth conditions determined for C. didymus, D. brightwellii, and T. rotula were similar to conditions present when these species were commonly found in the fjord.  相似文献

The effect of five constant temperatures (16, 22, 26, 31 and 36°C) on biological (survival and duration of developmental stages, fecundity and longevity of females, sex-ratio) and demographic parameters (R o , G, r m and λ) of the two main phytophagous mites that attack cassava in Africa,Mononychellus progresivus Doreste andOligonychus gossypii (Zacher), was studied in the laboratory. Experiments were performed simultaneously on the two mite species reared on the same cassava variety (1M20) under controlled conditions: 70±10% r.h. and 12L∶12D. The lower thermal threshold was 13°C forM. progresivus and 11°C forO. gossypii. Both species developed in the range from 22 to 36°C. The shortest development time was obtained at 31°C for both species; it was 7.2 days forM. progresivus and 8.2 days forO. gossypii. Maximum fecundity of both species was recorded at 26°C with 42.1 eggs forM. progresivus and 36.3 eggs forO. gossypii The highest intrinsic rate of increase (r m ) was obtained at 31°C for both species with 0.289 and 0.214 forM. progresivus andO. gossypii, respectively.  相似文献

The influence of environmental parameters on mycelial linear growth ofPleurotus ostreatus, P. eryngii, P. pulmonarius, Agrocybe aegerita, Lentinula edodes, Volvariella volvacea andAuricularia auricula-judae was determined in two different nutrient media in a wide range of temperature, forming the basis for the assessment of their temperature optimaV. volvacea grew faster at 35°C,P. eryngii at 25°C,P. ostreatus andP. pulmonarius at 30°C,A. aegerita at 25 or 30°C andA. auricula-judae at 20 or 25°C depending on the nutrient medium used andL. edodes at 20 or 30°C depending on the strain examined. The mycelium extension rates were evaluated on seven mushroom cultivation substrates: wheat straw, cotton gin-trash, peanut shells, poplar sawdust, oak sawdust, corn cobs and olive press-cake. The mycelium extension rates (linear growth and colonization rates) were determined by the ‘race-tube’ technique, and were found to be the highest on cotton gin-trash, peanut shells and poplar sawdust forPleurotus spp. andA. aegerita. Wheat straw, peanut shells and particularly cotton gin-trash supported fast growth ofV. volvacea, whereas wheat straw was the most suitable substrate forL. edodes andA. auricula-judae. Supplemented oak sawdust and olive press-cake were poor substrates for most species examined, white almost all strains performed adequately on corn cobs.  相似文献

The development, reproduction and longevity ofCybocephalus micans Reitter andC. nigriceps nigriceps (Sahlberg) were studied under controlled laboratory conditions. The data obtained explain the distribution of the 2 predators in different climatic regions of Israel. Under constant temperatures ranging between 16° and 36° C, the duration of development of the egg. larva and pupa of the 2 species became shorter as the temperature increased. For each developmental stage, at each temperature tested, the mean duration of development ofC. n. nigriceps was higher than that ofC. micans. The average time needed for completion of a generation at 28° C was about 39 days forC. micans, as compared with 56 days forC. n. nigriceps. Progeny production of both cybocephalid species was higher and adult longevity was longer at 28° C than at 32° C.  相似文献

Two pennate diatoms, Amphora coffeaeformis and Nitzschia ovalis, were used to evaluate potential responses to the future CO2 and temperature increases with respect to cell-specific growth rate, elemental composition, size, population growth rate, and carrying capacity. Diatoms were subjected to four different treatments over a 2 week period (approximately 4 generations): a control (28°C and present-day CO2, ~400 ppm), high CO2 (28°C with high CO2, ~750 ppm), high temperature (31°C and present-day CO2, ~400 ppm), and greenhouse-effect treatment (31°C with high CO2, ~750 ppm). The results indicated that both the cell-specific growth rates and the carrying capacity of A. coffeaeformis decreased at the higher temperature treatment, whereas N. ovalis did not differ among all treatments. No significant difference was found in either species’ elemental cell composition, but higher C:N and C:P ratios were observed for A. coffeaeformis and N. ovalis, respectively, in high CO2 and greenhouse-effect treatments. Smaller cell sizes were observed for both species under the greenhouse-effect treatment, a phenomenon that could alter benthic food webs in the future.  相似文献

Chiasma frequency variation has been studied in 12 populations ofCepaea hortensis and the results compared with those previously obtained forC. nemoralis from the same populations. Variation in chiasma frequency is effectively restricted to a single large bivalent. In both species there are significant differences between populations in chiasma frequency. Although mean population mean chiasma frequencies are similar inC. hortensis andC. nemoralis, C. hortensis is less variable thanC. nemoralis. This can be seen, to a greater or lesser extent, at several levels-(i) within cells, (ii) between cells, (iii) between snails, and (iv) between populations-and is most noticeable when interstitial chiasmata are considered separately from terminal chiasmata. Data from 6 populations suggest that chiasma frequency remains stable between successive years. It is suggested that, since sympatric populations were studied, the most likely explanation for differences in chiasma frequency statistics between the two species is thatC. hortensis andC. nemoralis are responding differently to similar selective pressures.  相似文献

Long-term in vitro culture of Plasmodium berghei was established using the Petri dish candle jar method of Trager and Jensen (1976). Cultures were established at 22, 27 and 37°C. As optimal growth was observed at 27°C, subsequent cultivation was carried out at this temperature. RPMI 1640 medium was modified by incorporating additional glucose (1 mg ml−1) and bactopeptone (1 mg ml−1) in the medium. This medium was found suitable for maintenance of mouse erythrocytes in vitro. P. berghei cultures were maintained using candle jars and this modified RPMI 1640 medium for 45 weeks.  相似文献

The development of the eggs and larvae and minute tubercles on the skin surface ofParacheilognathus himantegus larvae were observed. The egg began to hatch approximately 68 hours after insemination and the larvae reached the free-swimming stage 23 days after hatching at water temperature of 22±1°C. The larval development and minute tubercles on the skin surface of this species were similar to those ofAcheilognathus lanceolata, A. limbata, A. signifer andTanakia tanago. However, the shape of the ripe eggs ofP. himantegus differed from those of the four species. As regards the shape of eggs, there was a common characteristic amongP. himantegus, Rhodeus uyekii andA. limbata from Korea. As regards larval development,P. himantegus had two characters also found inRhodeus. These facts seem to suggest thatP. himantegus is closely related toA. lanceolata, A. limbata, A. signifer andT. tanago but is more specialized than these four species, except forA. limbata from Korea.  相似文献

The effect of growth temperature, medium composition and pH were examined in shake-flask-scale studies to determine the optimum conditions for cellulase production in mixed cultures of Trichoderma reesei Rut C30 and Aspergillus phoenicis. The optimum temperature and pH were 27°C and pH 4.6. Decreases in medium complexity and cost were achieved through reductions in the concentration of Ca, Mg, and K salts. Ten litre fermentations were implemented to study the kinetics of substrate utilization and product formation. The pH at which the fermentation was controlled appeared to be the critical parameter for batch growth of mixed cultures of Trichoderma reesei Rut C30 and Aspergillus phoenicis.  相似文献

DNA reassociation kinetics were used to determine inter- and intraspecific variation in genome organization and complexities in species ofGracilaria andGracilariopsis. Results indicate the presence of three second order components corresponding to fast, intermediate and slow fractions. Repeated sequences varied from 13–95%. Three geographic populations ofGracilaria tikvahiae were similar with 13–27% repeated sequences.Gracilaria sp. cultivars G-1 and G-6 with 35% and 95% repeated sequences, respectively, were distinct from each other andG. tikvahiae. No pattern of genome organization and complexity was found which permitted a distinction betweenGracilaria andGracilariopsis. Comparison of the percent of unique and repetitive sequences (U/R) indicated a wide range of ratios, withGracilaria tikvahiae populations having the highest values (2.7–7.3) andGracilaria sp. cultivar G-6,G. blodgettii andGracilariopsis lemanieformis the lowest (0.05–1.80). Unique component complexities varied one order of magnitude, from 108 forGracilaria takvahiae to 107 forGracilaria sp. cultivar G-6,G. blodgettii andGracilariopsis lemanieformis. Information for genome size, organization and complexity is used to develop a nuclear genome profile forGracilaria blodgettii andGracilariopsis lemanieformis which are characterized by commercial grade agars having high gel strengths (> 700 g cm?2) and elevated melting temperatures (99 °C).  相似文献

Aerobic biodegradation of propylene glycol by soil bacteria   总被引:1,自引:0,他引:1  
Propylene glycol (PG) is a main component of aircraft deicing fluids and its extensive use in Northern airports is a source of soil and groundwater contamination. Bacterial consortia able to grow on PG as sole carbon and energy source were selected from soil samples taken along the runways of Oslo Airport Gardermoen site (Norway). DGGE analysis of enrichment cultures showed that PG-degrading populations were mainly composed by Pseudomonas species, although Bacteroidetes were found, as well. Nineteen bacterial strains, able to grow on PG as sole carbon and energy source, were isolated and identified as different Pseudomonas species. Maximum specific growth rate of mixed cultures in the absence of nutrient limitation was 0.014 h?1 at 4 °C. Substrate C:N:P molar ratios calculated on the basis of measured growth yields are in good agreement with the suggested values for biostimulation reported in literature. Therefore, the addition of nutrients is suggested as a suitable technique to sustain PG aerobic degradation at the maximum rate by autochthonous microorganisms of unsaturated soil profile.  相似文献

Factors affecting the micropropagation of Veratrum californicum, a slow-growing species that is a potentially valuable source of cyclopamine, were investigated. Sterile cultures were initiated on modified Murashige and Skoog medium, and clones from individual donor plants were assigned to experimental conditions when approximately 100 shoots of each clone were available. The effects of temperature, light quality, and plant growth regulators on multiplication and survival were assessed. Four clones from which large greenhouse populations were obtained were selected for in-depth analysis. When shoots were cultured at 10°C and 16°C, multiplication ratios consistently >1 were observed from three of four clones and two of four clones, respectively, during the five-subculture cycles. None of the clones stably increased when cultured at 24°C, and plants from this treatment did not survive acclimatization in the greenhouse. Only one clone showed increased multiplication ratios in response to plant growth regulator treatments, with maximum multiplication when shoots were cultured with 9 μM benzyladenine and 0.5 μM naphthaleneacetic acid. Light quality in the laboratory did not affect multiplication ratio but did affect subsequent greenhouse survival. The size of plants derived from culture was most often equivalent (65% of 1,271) to 3-yr-old seed-derived plants. Although the growth of clones during acclimatization differed, plants derived from cultures incubated at 16°C had the best rates of overall greenhouse survival. Temperature and light treatments in vitro critical to long-term plant survival were demonstrated and will assist the establishment of a mass propagation system for V. californicum.  相似文献

The growth of 22 strains of Azolla pinnata R. Br., 3 strains of A. filiculoides Lam. and one strain each of A. mexicana Presl and A. caroliniana Willd. was tested separately in liquid culture media kept in controlled, artificial light (30 klux) growth cabinets. Three temperature levels were used: 33°C (37/29°C day/night), 29°C (33/25°C) and 22°C (26/18°C)/ Photoperiod was 12 h a day.For most A. pinnata strains (except three) and an A. mexicana strain the maximum weekly relative growth rate was higher at 33°C than at 22°C, but not for A. filiculoides and A. caroliniana. The highest value of maximum relative growth rate corresponded to 1.9 doubling days and in most strains this occurred in the first week. As the plants grew, the growth rate slowed down more severely at higher temperatures. The maximum biomass was higher at 22°C than at 33°C in all strains. At 22°C, it took 30–50 days to attain maximum biomass and the highest value was 14 g N m?2 or 320 g dry m?2 by A. caroliniana, followed by 12 g N m?2 or 290 g dry wt. m?2 by one strain of A. filiculoides. At 29°C, the maximum biomass was attained in 20–35 days. The highest value was 6.3 g N m?2 or 154 g dry wt. m?2 by A. caroliniana. At 33°C, most A. pinnata strains gave a maximum biomass of less than 4 g N m?2 after 13–23 days, while some strains grew up to 30 days, resulting in a higher maximum biomass. The highest maximum biomass at 33°C was 5.5 g N m?2 or 140 g m?2 dry wt. by A. pinnata from Cheng Mai while the maximum biomass of A. filiculoides and A. caroliniana was much less. Azolla filiculoides requires lower temperature than other species for its growth. Azolla pinnata has the best tolerance to high temperatures among the four species. Azolla mexicana could not be discriminated from A. pinnata in its response to temperature. Azolla caroliniana may keep an intermediate position between A. filiculoides and A. pinnata in temperature response.The formation of ammonia in the medium was examined and it occurred mostly under stationary growth conditions, but, at 33°C, some strains of A. pinnata and A. mexicana released or formed ammonia at 0.3–0.8 μg N ml?1 per week during their initial exponential growth stage.  相似文献

The collembolan Proisotoma minuta (Tullberg) was successfully maintained in a laboratory culture that could be used to produce large numbers of P. minuta of standard size and physiological condition. The general life history of P. minuta was studied using a substrate comprising a mixture of plaster of Paris and charcoal with baker's yeast as food. The mean time taken for eggs to hatch following oviposition was 10.6 ± 1.3 days at 17.5°C and 6 ± 0.5 days at 23°C. Conditions in the culture were very important for the production of large insect populations. Growth of the cultures was limited by the increasing population size and was probably affected by contamination of the habitat by the deposition of excretory materials. Proisotoma minuta from a population that had ceased egg production started oviposition after individuals were transferred to a fresh plaster of Paris and charcoal habitat.  相似文献

To test the effect of freezing on soil biota, isolated from the shortgrass prairie of northeastern Colorado, a series of experiments were performed using gnotobiotic soil microcosms.Pseudomonas paucimobilis was used to examine the effects of freezing on bacteria of different growth stages. Secondly, the effect of multiple freeze-thaw cycles was tested on an assemblage of bacterial species. Lastly, the effect of freezing on predator-prey interactions was studied usingP. paucimobilis and an amoebal predator,Acanthamoeba polyphaga. A temperature of ?9°C was not detrimental toP. paucimobilis at any growth stage. A single severe freeze-thaw cycle (?27°C to 23°C) resulted in 40–60% mortality ofP. paucimobilis and the mixed bacteria, although additional freezing events did not reduce the populations further. Multiple freeze-thaw cycles (?9°C to 23°C) gave 40–60% mortality ofP. paucimobilis and the mixed bacteria. Predator-prey population cycles were possibly desynchronized by freeze-thaw events.  相似文献

Two species of cold-temperate algae from the North Atlantic Ocean,Polyides rotundus andFurcellaria lumbricalis, were tested for growth and survival over a temperature range of −5 to 30 °C. In comparisons of eastern and western isolates, bothF. lumbricalis, a North Atlantic endemic, andP. rotundus, a species having related populations in the North Pacific, were quite homogeneous.F. lumbricalis tolerated −5 to 25°C and grew well from 0 to 25°C, with optimal growth at 10–15 °C.P. rotundus tolerated −5 to 27°C, grew well from 5 to 25°C, and had a broad optimal range of 10–25°C. Both species tolerated 3 months in darkness at 0°C. In neither case could any geographic boundary be explained in terms of lethal seasonal temperatures, suggesting that these species are restricted in distribution by strict thermal and/or daylength requirements for reproduction. The hypothesis that northern species are more homogeneous than southern taxa in terms of thermal tolerance was supported. A second hypothesis, that disjunct cold-temperate species should be more variable than pan-Arctic species, was not supported.  相似文献

Ostreopsis ovata is a benthic dinoflagellate that produces palytoxin and ovatoxins. Blooms of O. ovata causing human health problems and mortality of benthic fauna have been reported from many tropical and temperate marine waters. In the present study we examined the combined effects of temperature and different nutrient conditions on the biochemical composition, growth, toxicity and carbohydrate production of an O. ovata strain originating from the Tyrrhenian Sea. O. ovata cultures with N:P ratios of 1.6, 16 and 160 (N deficient, NP sufficient and P deficient, respectively) were grown at 20 °C and 30 °C. Biomass accumulation, growth rates, cell volumes, biochemical composition, cell toxicity and carbohydrate production in each treatment were studied. Results indicated that under nutrient sufficiency O. ovata biomass accumulation increased significantly compared to N and P deficiency and also that N limitation severely affected growth. The highest growth rates were recorded at 30 °C. Cellular contents and the atomic ratios of C, N and P were higher in the cells grown at 20 °C than in those grown at 30 °C. O. ovata cell volumes increased at 20 °C. N deficiency significantly increased cell toxicity. Toxicity per cell was higher at 20 °C, but per carbon was highest at 30 °C. The highest carbohydrate production was found in conditions of N deficiency and at the lower temperature.Our study suggests that temperature increases due to global warming and nutrient enrichment of coastal waters stimulate the proliferation of O. ovata, particularly for the strains that have become adapted to warm temperate waters.  相似文献

Plasmodium yoelii infected reticulocytes were cultured for 72 h at either 37 or 20°C in MEM (Eagle's modification) medium containing, in addition, glucose, para-aminobenzoic acid and 5% foetal calf serum, buffered at pH 7.3 with sodium bicarbonate/ HEPES and maintained under 10% CO2 in air. Red blood cell numbers were more stable at 20°C than at 37°C. Culture at both temperatures resulted in an increase in parasitaemia of the reticulocyte population over the initial 36 h at 37°C and for at least 72 h at 20°C. The effects of different temperatures appeared to be related to the continued presence of target cells. Parasites were not detected after 72 h culture at 37°C, but persisted for up to 120 h at 20°C. Increasing parasitaemia at both temperatures was associated with changes in the numbers of some parasite development types. Early falls in schizont numbers were associated with an increase in the numbers of ring forms. Trophozoite numbers tended to remain constant throughout the culture period. Viability of parasites cultured for 36 h was confirmed by their infectivity to CBA mice. In addition, parasites progressively incorporated H3-leucine into TCA-precipitable material over the initial 36 h of culture.  相似文献

Age specific fecundity of two parasitoids,P. yaseeni andT. leucaenae, of the leucaena psyllidH. cubana, were studied under laboratory conditions. At 25 °C,P. yaseeni had a greater fecundity (R0=192.9)_thanT. leucaenae (R0=71.2);T. leucaenae however had a lower sex ratio (about 99 % females) thanP. yaseeni (about 50 % females). Innate capacity for increase (rm=0.236) ofT. leucaenae was higher thanP. yaseeni (rm=0.188). Developmental rates of the parasitoids were examined at constant and fluctuating temperatures and equations of the rate of development against temperature were calculated. At 25 °C, mean generation times were 28.0 and 18.1 days forP. yaeseeni andT. leucaenae respectively. At temperatures of 21.5, 25, and 30 °C total development times (egg to adult) were 28.5, 21.9, and 14.7 days inP. yaseeni and 19.2, 12.6, and 9.5 days inT. leucaenae respectively. The level of parasitism was low and pupal mortality was high at the lower temperature of 21.5 °C for both parasitoids. Both parasitoids showed poor survivorship at 100 % RH,P. yaseeni survived particularly well (32 days) at a temperature of 21.5 °C and 44 or 76 % RH. P. yaseeni allocated about 58 % females to first instar psyllid nymphs but only 12 % females to second instars. About 99 % of allT. leucaenae births were females. Significantly largerT. leucaenae females emerged from fifth instar parasitized nymphs than third or fourth instars.  相似文献
