Monosomic alien addition lines (MAALs) are useful for assigning linkage groups to chromosomes. We examined whether the chromosomal rearrangements following the introduction of a single onion (Allium cepa) chromosome into the Allium fistulosum genome were produced by homeologous crossing over or by a nonreciprocal conversion event. Among the monosomic lines available, 17 were studied by fluorescent genomic in situ hybridisation, using total A. cepa genomic DNA as the probe and total A. fistulosum genomic DNA as the competitor. In this way, rearrangements such as chromosomal translocations between A. cepa and A. fistulosum were identified as terminal regions consisting of tandem DNA repeats. Homeologous crossing over between the two closely related genomes occurred in 4 of the 17 lines, suggesting that such events are not rare. On the basis of a detailed molecular cytogenetic characterisation, we identified true monosomic alien addition lines for A. cepa chromosomes 3, 4, 5, 7, and 8 that can reliably be used in genetic studies.  相似文献

We used a complete set of Allium fistulosum - shallot (A. cepa Aggregatum group) monosomic addition lines (FF+1A - FF+8A) to identify shallot chromosomes affecting the production of sugars. In the alien addition lines grown over two years in an experimental field at Yamaguchi University (34 degrees N, 131 degrees E), shallot chromosomes 2A and 8A altered sugar contents in leaf-bunching onion (A. fistulosum). Except for FF+2A, every monosomic addition accumulated non-reducing sugars in winter leaf blades. FF+8A caused an increase in the amounts of non-reducing sugars in the winter. FF+2A hardly produced non-reducing sugar throughout the two-year study. These results indicated that genes related to non-reducing sugar metabolism are located on the 2A and 8A chromosomes. The results of regression analyses using 2002 data on A. fistulosum and the monosomic addition set revealed a correlation (r = 0.63 +/- 0.07; mean +/- SE., n = 9) between reducing sugar and monosaccharide (Glc+Fru) contents but no correlation between non-reducing sugar and sucrose contents. This result indicates the existence of other polysaccharides (e.g., scorodose) as non-reducing sugars in the leaf blade.  相似文献

Onion and shallot (Allium cepa L.) exhibit wide variation in bulb fructan content, and the Frc locus on chromosome 8 conditions much of this variation. To understand the biochemical basis of Frc, we conducted biochemical and genetic analyses of Allium fistulosum (FF)-shallot (A. cepa Aggregatum group) alien monosomic addition lines (AALs; FF+1A-FF+8A) and onion mapping populations. Sucrose and fructan levels in leaves of FF+2A were significantly lower than in FF throughout the year, and the springtime activity of acid invertase was also lower. FF+8A showed significantly higher winter sucrose accumulation and sucrose phosphate synthase (SPS) activity. Inbred high fructan (Frc_) lines from the 'W202Ax Texas Grano 438' onion population exhibited significantly higher sucrose levels prior to bulbing than low fructan (frcfrc) lines. Sucrose synthase (SuSy) activity in these lines was correlated with leaf hexose content but not with Frc phenotype. Markers for additional candidate genes for sucrose metabolism were obtained by cloning a major SPS expressed in onion leaf and exhaustively mining onion expressed sequence tag resources. SPS and SuSy loci were assigned to chromosome 8 and 6, respectively, using AALs and linkage mapping. Further loci were assigned, using AALs, to chromosomes 1 (sucrose phosphate phosphatase), 2 (SuSy and three invertases) and 8 (neutral invertase). The concordance between chromosome 8 localization of SPS and elevated leaf sucrose levels conditioned by high fructan alleles at the Frc locus in bulb onion or alien monosomic additions of chromosome 8 in A. fistulosum suggest that the Frc locus may condition variation in SPS activity.  相似文献

We analyzed Japanese bunching onion (Allium fistulosum L.) - shallot (Allium cepa L. Aggregatum group) alien chromosome addition lines in order to assign the genes involved in the flavonoid biosynthesis pathway to chromosomes of the shallot. Two complete sets of alien monosomic additions (2n = 2x + 1 = 17) were used for determining the chromosomal locations of several partial sequences of candidate genes, CHS, CHI, F3H, DFR, and ANS via analyses of PCR-based markers. The results of DNA marker analyses showed that the CHS-A, CHS-B, CHI, F3H, DFR, and ANS genes should be assigned to chromosomes 2A, 4A, 3A, 3A, 7A, and 4A, respectively. HPLC analyses of 14 A. fistulosum - shallot multiple alien additions (2n = 2x + 2 - 2x + 7 = 18 - 23) were conducted to identify the anthocyanin compounds produced in the scaly leaves. A direct comparison between the genomic constitution and the anthocyanin compositions of the multiple additions revealed that a 3GT gene for glucosylation of anthocyanidin was located on 4A. Thus, we were able to assign all structural genes involved in flavonoid biosynthesis influencing bulb color to individual chromosomes of A. cepa.  相似文献

First and second backcrosses of amphidiploid hybrids (2n = 4x = 32, genomes AAFF) between shallot (Allium cepa Aggregatum group) and A. fistulosum were conducted to produce A. cepa - A. fistulosum alien addition lines. When shallot (A. cepa Aggregatum group) was used as a pollinator, the amphidiploids and allotriploids set germinable BC(1) and BC(2) seeds, respectively. The 237 BC(1) plants mainly consisted of 170 allotriploids (2n = 3x = 24, AAF) and 42 hypo-allotriploids possessing 23 chromosomes, i.e., single-alien deletions (2n = 3x-1 = 23, AAF-nF). The single-alien deletions in the BC(1) progeny showed dwarfing characteristics and were discriminated from the allotriploids (2n = 24) and hyper-allotriploids (2n = 25) by means of flow cytometric analysis. The chromosome numbers of 46 BC(2) seedlings varied from 16 to 24. Eight monosomic additions (2n = 2x+1 = 17, AA+nF) and 20 single-alien deletions were found in these BC(2) seedlings. Consequently, six kinds of A. cepa - A. fistulosum alien chromosome additions possessing different chromosome numbers (2n = 17, 18, 20, 21, 22, 23) were recognized in the BC(1) and BC(2) populations. A total of 79 aneuploids, including 62 single-alien deletions, were analyzed by a chromosome 6F-specific isozyme marker (Got-2) in order to recognize its existence in their chromosome complements. This analysis revealed that two out of 62 single-alien deletions did not possess 6F. One (AAF-6F) out of the possible eight single-alien deletions could be identified at first. The present study is a first step toward the development of a useful tool, such as a complete set of eight different single-alien deletions, for the rapid chromosomal assignment of genes and genetic markers in A. fistulosum.  相似文献

Two complete sets of Allium fistulosum L.– A. cepa monosomic addition lines (2n=2x+1=17) together with an AFLP linkage map based on a cross between A. cepa and A. roylei Stearn were used to re-evaluate the eight A. cepa linkage groups identified in the mapping study. The linkage groups could be assigned to individual, physical chromosomes. The low level of molecular homology between A. cepa and A. fistulosum enabled the identification of 186 amplified fragment length polymorphisms (AFLP™ markers) present in A. cepa and not in A. fistulosum with ten different primer combinations. With the monosomic addition lines the distribution of the markers over the eight chromosomes of A. cepa could be determined. Of these 186 AFLP markers 51 were absent in A. roylei and consequently used as markers in the mapping study (A. cepa ×A. roylei cross). Therefore, these 51 AFLP markers could be used to assign the eight A. cepa linkage groups identified in the mapping study to physical chromosomes. Seven isozyme and three CAPS markers were also included. Two of the linkage groups had to be split because they included two sets of markers corresponding to different chromosomes. A total of 20 (approx. 10%) of the A. cepa-specific AFLP markers were amplified in more than one type of the monosomic addition lines, suggesting unlinked duplications. The co-dominant isozyme and CAPS markers were used to identify the correspondence of linkage groupsoriginating from A. cepa or from A. roylei. With the aim of establishing a complete monosomic alien tomato chromosome addition series in a potato background, the backcross progenies derived from repeated crossing of potato (+) tomato fusion hybrids to potato were screened through RFLP and GISH analyses. Because of the availability from our previous work of seven of the possible 12 tomato monosomic additions, selected from BC2 populations, attention was paid to those alien additions that were missing. Thus, since the alien additions were already available for tomato chromosomes 1, 2, 4, 6, 8, 10 and 12, efforts were made to select for chromosomes 3, 5, 7, 9 and 11 by screening specific BC3 populations. In all, 105 plants from four BC3 populations were screened through a combination of RFLP and GISH analyses in order to complete the series. Among the newly selected alien addition lines, five were monosomic additions for all the remaining chromosomes and one was a disomic addition for chromosome 11. When using conventional cytogenetics the selection of monosomic alien additions is highly laborious. All the tomato chromosomes showed a variable rate of transmission. Chromosome 6 was transmitted at 29.6% and 81.5% frequency in populations 2705 and 2701 respectively. The present study showed that molecular markers and molecular cytogenetics applied in this study were most efficient and rapid because a pre-selection for the desired genotypes was possible by screening a population with chromosome-specific markers for the presence of one tomato chromosome at a time. We have constructed a linkage map of bunching onion (Allium fistulosum L., 2n = 16) using an F(2) population of 225 plants. The map consists of 17 linkage groups with 212 bunching onion SSR markers and 42 bulb onion (A. cepa L.) SSR, InDel, CAPS or dCAPS markers, covering 2,069 cM. This is the first report of a linkage map mainly based on SSR markers in the genus Allium. With the 103 anchor markers [81 bunching onion SSRs, 11 bulb onion SSRs and 11 bulb onion non-SSRs (1 InDel, 9 CAPSs and 1 dCAPS)] whose chromosome assignments were identified in A. cepa and/or A. fistulosum, via the use of several kinds of Allium alien addition lines, 16 of the 17 linkage groups were connected to the 8 basic chromosomes of A. cepa.  相似文献

We measured the antioxidant contents and antioxidative activities in eight Allium fistulosum–shallot monosomic addition lines (MAL; FF+1A–FF+8A). The high antioxidative activity lines (FF+2A and FF+6A) showed high polyphenol accumulation. These additional chromosomes (2A and 6A) would therefore have anonymous genes related to the upregulation of polyphenol production, the antioxidative activities consequently being increased in these MALs.  相似文献

In Allium wakegi, which is an allodiploid species between Allium cepa and Allium fistulosum, each genome can be clearly distinguished using genomic in situ hybridization (GISH). Genomic DNA of A. cepa and A. fistulosum is differentiated both qualitatively and quantitatively. We wanted to isolate nucleotide sequences that give genome-specific signals on A. cepa chromosomes in GISH experiments in A. wakegi. We isolated 23 clones that show GISH-like signal patterns in fluorescence in situ hybridization (FISH) and analyzed their distribution in the A. cepa- and A. fistulosum-derived genomes of A. wakegi. There was considerable variation in the abundance and distribution of these cloned sequences on the chromosomes of the two species. The degree of A. cepa specificity varied among the clones. Twenty-two of the clones showed an even distribution over most chromosome arms with some clustering in the pericentromeric regions, but one clone showed very distinct terminal signals on some chromosomes. Whereas these sequences are not specific for A. cepa, changes in bases in nucleotide sequences and in their amount result in genome-specific characteristics in GISH experiments.  相似文献

To produce alien monosomic addition lines (AMALs) of Allium cepa (genomes CC, 2n = 2x = 16) carrying extrachromosomes from Allium roylei (RR, 2n = 2x = 16), reciprocal backcrossing of allotriploids (2n = 24, CCR) with diploids (2n = 16, CC) and selfing of a single allotriploid were carried out. The chromosome numbers in the BC2F1 and BC1F2 progenies ranged from 16 to 32. Forty-eight plants were recorded to possess 2n = 17 among a total of 169 plants in observation. Through the analyses of isozymes, expressed sequence tag (EST) markers, and karyotypes, all eight possible types of A. cepaA. roylei monosomic addition lines (CC+1R–CC+8R) could be identified. Seven types of representative AMALs (without CC+2R) were used for the GISH analysis of somatic chromosomes. Except for CC+6R, all AMALs showed an entire (unrecombined) extrachromosome from A. roylei in the integral diploid background of A. cepa. A single recombination between A. cepa and A. roylei was observed on the extrachromosome in the remaining type. All alloplasmic AMALs possessing A. roylei cytoplasm showed high or complete pollen sterility. Only the autoplasmic CC+4R with A. cepa cytoplasm possessed relatively high pollen fertility. The bulbs of CC+4R displayed the distinct ovoid shape that discriminates them from spherical or oval ones in other AMALs. Downy mildew screening in the field showed higher resistance in A. roylei, a hypo-allotriploid (CCR-nR, 2n = 23), and an allotriploid (CCR, 2n = 24). Meanwhile, no complete resistance was found in some AMALs examined. This was the first trial toward the establishment of a complete set of A. cepaA. roylei monosomic additions.  相似文献

DNA fingerprinting with three repetitive DNA sequences (OPX2, PB6-4 and Sat-121) was carried out on a set of 10 monosomic additions of Beta procumbens and 75 anonymous B. patellaris-derived monosomic additions in B. vulgaris, for characterization of the alien chromosomes at the DNA level. The probes are Procumbentes-specific and distributed over all chromosomes. Morphological characteristics were also used for the classification of B. patellaris monosomic addition families and for comparison with the morphology of the addition families of B. procumbens.DNA fingerprinting revealed unique patterns for almost all individual addition chromosomes of B. procumbens. However, it was concluded that chromosomes 1 and 6 of B. procumbens may be identical with the only difference that the chromosome referred to as 6 carries a susceptible allele for beet cyst nematode (BCN) resistance. In contrast, it was concluded that the two addition types with chromosome 2 are carrying different chromosomes of B. procumbens, so that one of them was renumbered to become the new chromosome 6.DNA fingerprinting of 75 anonymous B. patellaris-derived monosomic additions facilitated the identification and characterization of the alien chromosomes and the grouping of these additions into nine different groups. Several of these groups could be divided in two sub-groups on the basis of small differences in banding patterns. The results of the DNA fingerprinting led to the conclusion that B. patellaris most likely is an allotetraploid. It was also deduced that the BCN gene(s) in this species are homozygous and located on chromosome 1, while the pair of homoeologous chromosomes does not carry such BCN gene(s). Because of the allotetraploid nature of B. patellaris, preferential association occurs between the two homologous chromosomes containing the allele(s) for BCN resistance. Each group of B. patellaris addition families united by DNA fingerprinting had comparable morphological characteristics. Some of these morphological traits appeared to be chromosome-specific and were very useful for primary classification of the addition families. However, the present study showed that these morphological traits are not adequate for the identification of all alien chromosomes without the aid of additional markers. Because of similarities observed between molecular characteristics or the effects on plant morphology of several chromosomes of B. procumbens and B. patellaris it was concluded that B. procumbens could have been involved in the evolutionary history of B. patellaris.  相似文献

A non-radioactive digoxigenin-labelled DNA method was used successfully to identify RFLP markers in 54 Triticum aestivum cv Chinese Spring — Agropyron cristatum (2n=28, genome PPPP) P-genome monosomic addition lines. Southern analysis using a set of 14 DNA probes identifying each homoeologous chromosome arm, combined with two restriction enzymes HindIII and EcoRI, indicated that six A. cristatum chromosomes (1P, 2P, 3P, 4P, 5P and 6P) and five A. cristatum chromosome arms (2PS, 2PL, 5PL, 6PS and 6PL) have been individually added to the wheat genome. The added chromosomes of three lines were Agropyron translocated chromosomes. It was also found that two addition plants possessed an Agropyron-wheat translocation. These results showed that RFLP analysis using the set of assigned wheat probes was a powerful tool in detecting and establishing homoeology of alien A. cristatum chromosomes, or arms, added to wheat, as well as in screening the alien addition material. The creation of the monosomic addition lines should be useful for the transfer of disease-resistance genes from A. cristatum to wheat.  相似文献

The extrachromosome 5A of shallot (Allium cepa L., genomes AA) has an important role in flavonoid biosynthesis in the scaly leaf of Allium fistulosum–shallot monosomic addition lines (FF+nA). This study deals with the production and biochemical characterisation of A. fistulosum–shallot multiple alien addition lines carrying at least 5A to determine the chromosomal locations of genes for quercetin formation. The multiple alien additions were selected from the crossing between allotriploid FFA (♀) and A. fistulosum (♂). The 113 plants obtained from this cross were analysed by a chromosome 5A-specific PGI isozyme marker of shallot. Thirty plants were preliminarily selected for an alien addition carrying 5A. The chromosome numbers of the 30 plants varied from 18 to 23. The other extrachromosomes in 19 plants were completely identified by using seven other chromosome markers of shallot. High-performance liquid chromatography analyses of the 19 multiple additions were conducted to identify the flavonoid compounds produced in the scaly leaves. Direct comparisons between the chromosomal constitution and the flavonoid contents of the multiple alien additions revealed that a flavonoid 3′-hydroxylase (F3′H) gene for the synthesis of quercetin from kaempferol was located on 7A and that an anonymous gene involved in the glucosidation of quercetin was on 3A or 4A. As a result of supplemental SCAR analyses by using genomic DNAs from two complete sets of A. fistulosum–shallot monosomic additions, we have assigned F3′H to 7A and flavonol synthase to 4A.  相似文献

We compared meiotic pairing and recombination between tomato ( Lycopersicon esculentum) and homoeologous Solanum lycopersicoides chromosomes in monosomic additions (MAs) and substitution lines (SLs), each representing a single chromosome of the nightshade in a tomato background. Three configurations of each alien chromosome and its two tomato homoeologues were detected by genomic in situ hybridization in MA-7, -8, and -10 at diakinesis/metaphase-I: 1 trivalent (III), 1 bivalent + 1 univalent (II+I), and 3 univalents (3I). The II+I category was by far the most common, and the univalent was from S. lycopersicoides 91-99.5% of the time, indicating a high degree of preferential (homologous) pairing. In the corresponding substitution lines, association of homoeologous chromosomes was much higher (up to 90% of the cells), presumably due to the absence of homologous partners. However, SL-10 showed a surprisingly high frequency of univalents (about 73%). Genome-wide analysis of chromosome pairing revealed a decrease in the average chiasma frequency for both monosomic additions and substitution lines. Recombination between tomato and the nightshade was restricted in all cases, the reduction being more severe in each monosomic addition than in the corresponding substitution line. Recombination rates in the substitutions were less than those observed for the same chromosomes in the first backcross generation. Chromosomes 8 and 10 showed the highest and the lowest rates of homoeologous recombination, respectively. No recombination was detected between markers on the long arm of chromosome 10, presumably due to the presence of a paracentric inversion differentiating the two genomes in this region. The frequency of homoeologous pairing at diakinesis/metaphase-I was significantly higher than the rate of homoeologous recombination detected in the progeny, suggesting a strong selection against recombinant products in meiotic or post-meiotic stages.  相似文献

Alien chromosome transmission through the female germ line as well as meiosis in pollen mother cells were studied in disomic and ditelosomic alien chromosome additions of beet. Beta vulgaris, carrying an extra pair of chromosomes or telosomes of B. procumbens or B. patellaris, respectively. The alien chromosomes carried genes for resistance to the beet cyst nematode, Heterodera schachtii, and screening for this resistance was used to select plants with or without the alien chromosomes. A great variation for alien chromosome transmission was recorded and plants carrying two extra alien chromosomes were recovered in the backcross progenies of the disomic or ditelosomic additions. However, in these progenies the average frequencies of plants without alien chromosomes (86%) did not clearly differ from that in similar progenies of the original monosomic or monotelosomic chromosome additions, indicating that doubling the number of the alien chromosome did not enlarge their transmission to the next generation. The alien chromosomes fully paired at pachytene and desynapsed again before diakinesis, indicating decreased chiasma formation. At second metaphase nearly 60% of the cells had one extra chromosome, and the remaining cells carried two or no extra chromosomes in about equal proportions. The tetrads looked fully normal. The expected relation between the average number of alien chromosomes in the germ cells and in the plants of the progenies did not show up, indicating a strong selection favouring the female gametes without alien chromosomes.  相似文献

Summary Nine different monosomic additions in Beta vulgaris from Beta webbiana were characterized through morphological characters and isozyme markers. The effect of the alien chromosome on the morphology of the recipient species is chromosome specific, and nine morphotypes could be distinguished. The added chromosome caused a growth reduction in the recipient plants. Eleven isozyme systems were used as marker systems. A 6PGDH band was found as a marker for chromosome 7, which contains a resistance gene for the beet cyst nematode in monosomic additions from Beta procumbens and Beta webbiana. A difference in the 6PGDH zymogram pattern between the two species with respect to this chromosome has been noted.  相似文献

The establishment of a C-banded karyotype of hexaploid oat (Avena spp., 2n = 6x = 42) has facilitated the cytological characterization of a monosomic series in 'Kanota', an A. byzantina (C. Koch) cultivar. The 'Kanota' series of monosomics analyzed in this study consists of only 12 of the 21 different chromosome-deficient lines possible plus potential translocated segments of two or three additional chromosomes. These findings were confirmed by RFLP mapping data from studies in which oat probes were assigned to syntenic groups using the 'Kanota' set of monosomic lines. Among the remaining nine monosomic lines analyzed, eight are missing chromosomes represented in the set of 12 unique lines and one line, monosomic K13, is missing a chromosome from the unique set of 12 that possesses a cytologically detectable translocation. This same translocation, involving chromosomes 7C and 14, is found in 5 of the 21 'Kanota' monosomics. The incompleteness of the set of 'Kanota' monosomics might be due to (i) difficulty in identifying individual oat chromosomes without C-banding, (ii) plant genotypic and phenotypic variability in the original source population of the 'Kanota' monosomics, and (or) (iii) a high frequency of monosomic shifts in progency of the original 'Kanota' monosomic lines.  相似文献

The average number of aberrations per aberrant cell was concluded to carry out information on chromosome instability peculiarities induced by different mutagens as it was shown in our previous work. The purpose of the current study was to present comparative analysis of intercellular distribution of number of aberrations and their theoretical approximations. Distribution of numbers of aberrations per cell in Allium cepa L. and Allium fistulosum L. root tip cells induced by different mutagenic factors (gamma-irradiation, thiotepa, formaldehyde and seed aging) have been studied. The results were approximated to theoretical Poisson, geometric and negative binomial distributions. The intercellular distribution of aberrations did not correspond to any of the used theoretical distributions when A. cepa seeds were gamma-irradiated. There was some, but not regular, accordance with theoretical distributions when chemical mutagens thiotepa in A. cepa and formaldehyde in A. fistulosum and seed aging in both species were evaluated. During seed aging frequency of aberrant cells increased more quickly in A. fistulosum in comparison with A. cepa.  相似文献

All 10 chromosomes of maize (Zea mays, 2n = 2x = 20) were recovered as single additions to the haploid complement of oat (Avena sativa, 2n = 6x = 42) among F(1) plants generated from crosses involving three different lines of maize to eight different lines of oat. In vitro rescue culture of more than 4,300 immature F(1) embryos resulted in a germination frequency of 11% with recovery of 379 F(1) plantlets (8.7%) of moderately vigorous growth. Some F(1) plants were sectored with distinct chromosome constitutions among tillers of the same plant and also between root and shoot cells. Meiotic restitution facilitated development of un-reduced gametes in the F(1). Self-pollination of these partially fertile F(1) plants resulted in disomic additions (2n = 6x + 2 = 44) for maize chromosomes 1, 2, 3, 4, 6, 7, and 9. Maize chromosome 8 was recovered as a monosomic addition (2n = 6x + 1 = 43). Monosomic additions for maize chromosomes 5 and 10 to a haploid complement of oat (n = 3x + 1 = 22) were recovered several times among the F(1) plants. Although partially fertile, these chromosome 5 and 10 addition plants have not yet transmitted the added maize chromosome to F(2) offspring. We discuss the development and general utility of this set of oat-maize addition lines as a novel tool for maize genomics and genetics.  相似文献
