The role of reactive oxygen species (ROS) in copper (Cu) toxicity to two freshwater green algal species, Pseudokirchneriella subcapitata (Korshikov) Hindák and Chlorella vulgaris Beij., was assessed to gain a better mechanistic understanding of this toxicity. Cu‐induced formation of ROS was investigated in the two algal species and linked to short‐term effects on photosynthetic activity and to long‐term effects on cell growth. A light‐ and time‐dependent increase in ROS concentrations was observed upon exposure to environmentally relevant Cu concentrations of 50 and 250 nM and was comparable in both algal species. However, effects of 250 nM Cu on photosynthesis were different, leading to a 12% reduction in photosynthetic activity in P. subcapitata, but not in C. vulgaris. These results indicate that differences in species‐specific sensitivities measured as photosynthetic activity were not caused by differences in the cellular ROS content of the algae, but probably by different species‐specific ROS defense systems. To investigate the role of ROS in Cu‐mediated inhibition of photosynthesis, the ROS scavenger Ntert‐butyl‐α‐phenylnitrone (BPN) was used, resulting in a reduction of Cu‐induced ROS production up to control level and a complete restoration of photosynthetic activity of Cu‐exposed P. subcapitata. This finding implied that ROS play a primary role in Cu toxicity to algae. Furthermore, we observed a time‐dependent ROS release process across the plasma membrane. More than 90% of total ROS were determined to be extracellular in P. subcapitata, indicating an efficient method of cellular protection against oxidative stress.  相似文献

Currently there is a surge of interest in exploiting toxicogenomics to screen the toxicity of chemicals, enabling rapid and accurate categorisation into classes of defined mode-of-action (MOA), and prioritising chemicals for further testing. Direct infusion FT-ICR mass spectrometry-based metabolomics can provide a sensitive and unbiased analysis of metabolites in only 15 mins and therefore has considerable potential for chemical screening. The water flea, Daphnia magna, is an OECD test species and is utilised internationally for toxicity testing. However, no metabolomics studies of this species have been reported. Here we optimised and evaluated the effectiveness of FT-ICR mass spectrometry metabolomics for toxicity testing in D. magna. We confirmed that high-quality mass spectra can be recorded from as few as 30 neonates (<24 h old; 224 μg dry mass) or a single adult daphnid (301 μg dry mass). An OECD 24 h acute toxicity test was conducted with neonates at copper concentrations of 0, 5, 10, 25, 50 μg l−1. A total of 5447 unique peaks were detected reproducibly, of which 4768 were assigned at least one empirical formula and 1017 were putatively identified based upon accurate mass measurements. Significant copper-induced changes to the daphnid metabolome, consistent with the documented MOA of copper, were detected thereby validating the approach. In addition, N-acetylspermidine was putatively identified as a novel biomarker of copper toxicity. Collectively, our results highlight the excellent sensitivity, reproducibility and mass accuracy of FT-ICR mass spectrometry, and provide strong evidence for its applicability to high-throughput screening of chemical toxicity in D. magna. The study was carried out to assess the effect of both short-and long-term copper exposure on five cladoceran species differing in body size and habitat, i.e. Daphnia magna, D. pulex, D. galeata, Bosmina longirostris, and Chydorus sphaericus. The species-specific 48 h EC50 values for fed neonates were used to determine the chronic exposure levels (52% and 65% of the EC50 values). The experiment was run at two food levels. Long-term copper exposure retarded growth in each of the species studied. However, the copper treatments did not affect the clutch sizes of the experimental animals. At a low food level, copper exposure increased mortality in every species studied. The intrinsic rate of increase, r, was reduced in the copper-exposed populations. The sensitivity to copper was higher in the small lake species, compared to D. magna and D. pulex that are commonly used in standard toxicity tests. Of the species studied, B. longirostris had the highest sensitivity to both acute and chronic copper stress. The different sensitivities of the species studied are discussed in an ecological context.  相似文献

We examine how interspecific competition and two types of size-selective predation affect population density, variability and persistence in laboratory cultures of two species of Daphnia, D. magna and D. longispina. When both species were analysed together, and for D. longispina alone, there were weak negative relationships between mean population density and population variability. Interspecific competition resulted in lower population densities and higher population variability. Extinct populations had lower densities and were also more variable than persisting ones. There was still an effect of population variability on extinction probability after the effect of density on population variability had been accounted for. Hence, the effects of population density and variability on population persistence were partly independent of each other. The effects of size-selective predation on population persistence were more species-specific and not directly related to density or variability. Since the effects of species interactions on persistence were large, we suggest that it is likely that population vulnerability analyses not incorporating effects of interspecific interactions are often misleading.  相似文献

SUMMARY 1. Surprisingly few data compare the apparent responses of diatoms and macroinvertebrates to metals in streams. We examined variation in metals, diatoms and macroinvertebrates between 51 streams in metal‐mining areas of Wales and Cornwall, U.K., using a survey design with multiple reference and polluted sites. 2. To quantify variations in metals between sites, we calculated cumulative criterion unit (CCU) scores, a recently defined measure of total stream metal concentration and toxicity, to account for additive effects of each metal relative to putative toxic thresholds. We compared assemblage responses among epilithic diatoms and macroinvertebrates to CCU scores or individual metal concentrations using correlation and detrended correspondence analysis (DCA). 3. Macroinvertebrate diversity, richness and total abundance declined and evenness increased with increasing copper concentrations. Trends with CCU scores were significant but less pronounced. Some individual macroinvertebrate taxa varied significantly in abundance with CCU scores, copper or zinc, but overall assemblage composition correlated only with manganese, pH and nitrate. 4. Among diatoms, pH and conductivity explained the major variations in assemblage composition, and neither diversity, richness nor evenness varied with metal concentration. Nevertheless, the single strongest predictor of diatom assemblages on ordination axis 2 was the CCU score. The abundances of some macroinvertebrate taxa, particularly grazers, also explained significant variations in diatom assemblages that were linked to both metals and acid–base status. 5. Diatom species apparently tolerant of high metal concentrations included Psammothidium helveticum, Eunotia subarcuatoides, Pinnularia subcapitata and Sellaphora seminulum. Of these, P. helveticum, E. subarcuatoides and P. subcapitata were abundant at lower pH than S.seminulum and might indicate metal enrichment over different pH ranges. Sensitive species included Fragilaria capucina var. rumpens, Achnanthes oblongella and Tabellaria flocculosa. 6. We conclude that macroinvertebrates at these sites reflected metal pollution most strongly through variations in diversity while effects on diatoms were best reflected by changes in assemblage composition. We suggest that, with further refinement, CCU scores might be useful in evaluating the possible effects of metal pollution on benthic organisms in European rivers.  相似文献

Abstract. The Mediterranean Basin harbours paleo‐endemic species with a highly restricted and fragmented distribution. Many of them might also be of the remnant type, for which the regional dynamics depends on the persistence of extant populations. Therefore, a key issue for the long‐term persistence of these species is to assess the variability and effects of ecological factors determining plant performance. We investigated the spatio‐temporal variability in plant traits and ecological factors of Ramonda myconi, a preglacial relict species with remnant dynamics, in 5 populations over 4–7 yr. Ecological factors contributing to fecundity showed a high degree of between‐year variability. Pre‐dispersal fruit predation had a minor influence on total reproductive output, and most of the variability was found among individuals within populations and years. Spatio‐temporal variability in growth and survival was rather low but significant, whereas recruitment showed important between‐population variability. Among‐year variability in fecundity and growth was related to climatic fluctuations on a regional scale, notably rainfall and temperature in a particular period, while the spatial variability in survival and recruitment was explained by within‐population (patch) habitat quality. Although R. myconi is able to withstand repeated periods of drought, water availability seems to be the most important factor affecting plant performance in all the study populations. These findings suggest that the long‐term persistence of species showing remnant population dynamics in habitats under the influence of Mediterranean climate might be threatened by increased aridity as a result of climate change.  相似文献

The objectives of this study were to evaluate the sensitivity of two bacterial tests commonly used in metal toxicity screening — the Vibrio fischeri bioluminescence inhibition test and the Pseudomonas putida growth inhibition test — in comparison to the standard acute Daphnia magna test, and to estimate applicability of the selected methods to the toxicity testing of environmental samples. The D. magna acute test proved to be more sensitive to cadmium (Cd), zinc (Zn) and manganese (Mn) than the two bacterial assays, whereas P. putida seems to be the most sensitive species to lead (Pb). Manganese appears to be slightly toxic to D. magna and non-toxic to the two selected bacteria. This leads to the conclusion that even in regions with high background concentrations, manganese would not act as a confounding factor. Low sensitivity of V. fischeri to heavy metals questions its applicability as the first screening method in assessing various environmental samples. Therefore, it is not advisable to replace D. magna with bacterial species for metal screening tests. P. putida, V. fischeri and/or other bacterial tests should rather be applied in a complex battery of ecotoxicological tests, as their tolerance to heavy metals can unravel other potentially present toxic substances and mixtures, undetectable by metal-sensitive species.  相似文献

The taxonomic problem of cryptic species has long been recognized. Hemiboea subcapitata C. B. Clarke is a widespread and morphologically diverse species including two varieties, H. subcapitata var. subcapitata C. B. Clarke and H. subcapitata var. guangdongensis (Z. Y. Li) Z. Y. Li. However, genetic distance and molecular phylogenetic analyses based on nuclear ITS and four plastid DNA sequences (atpB‐rbcL, matK, rbcL, rpS16 intron) revealed that H. subcapitata var. guangdongensis is sister to H. subacaulis, and separated from H. subcapitata var. subcapitata, suggesting that it should be raised to the rank of an independent species as H. guangdongensis (Z. Y. Li) X. Q. Li & X. G. Xiang, comb. & stat. nov. Morphologically, the new species can be distinguished from H. subacaulis by its calyx 5‐sect from base, segments narrowly oblong‐lanceolate (versus calyx 5‐sect from base or 2‐lipped and adaxial lip 2‐lobed from below middle, segments ovate), peduncle glabrous (versus pubescent), and vermiform sclereids dispersed in leaf mesophyll (versus without sclereids).  相似文献

Effects of dispersal and the presence of predators on diversity, assembly and functioning of bacterial communities are well studied in isolation. In reality, however, dispersal and trophic interactions act simultaneously and can therefore have combined effects, which are poorly investigated. We performed an experiment with aquatic metacommunities consisting of three environmentally different patches and manipulated dispersal rates among them as well as the presence or absence of the keystone species Daphnia magna. Daphnia magna reduced both local and regional diversity, whereas dispersal increased local diversity but decreased beta‐diversity having no net effect on regional diversity. Dispersal modified the assembly mechanisms of bacterial communities by increasing the degree of determinism. Additionally, the combination of the D. magna and dispersal increased the importance of deterministic processes, presumably because predator‐tolerant taxa were spread in the metacommunity via dispersal. Moreover, the presence of D. magna affected community composition, increased community respiration rates but did not affect bacterial production or abundance, whereas dispersal slightly increased bacterial production. In conclusion, our study suggests that predation by a keystone species such as D. magna and dispersal additively influence bacterial diversity, assembly processes and ecosystem functioning.  相似文献

Abstract

The effects of no. 20 diesel oil exposure, 2,4-dichlorophenol (2,4-DCP) exposure and combined exposure on the antioxidant defences of Daphnia magna have been studied systematically for the first time. Daphnia magna was exposed for 1 day or 10 days to several concentrations of 0, 0.005, 0.01, 0.05, 0.1, 0.5 or 1.0 mg L?1 solutions. Antioxidant defences consisting of the levels of reduced glutathione (GSH) and glutathione disulfide (GSSG) and activities of superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), selenium dependent glutathione peroxidase (Se-GPx) and glutathione S-transferase (GST) of daphnids were determined to evaluate their protective roles and to analyse the occurrence of oxidative stress. The possible antioxidant defence mechanisms are discussed. Furthermore, GST can be a potential biomarker and an early-warning index for the pollutants in waters in that GST responded sensitively to 1 day and 10 days of exposure to diesel oil and 2,4-DCP and 10 days of combined exposure. Crossover comparisons showed an antagonistic action about the no-observed-effect concentration (NOEC) against Daphnia magna, which needs further studies.  相似文献

Chiral fungicide prothioconazole has a wide range of antifungal spectrum; however, little research has been conducted to evaluate prothioconazole on an enantiomeric level. Five target pathogens and three common aquatic organisms were tested for the enantioselective bioactivity and toxicity of prothioconazole in this work. The antifungal activity of the enantiomers against wheat phytoalexin, rice blast fungus, exserohilum turcicum, Alternaria triticina, and Fusarium avenaceum was determined, and it was found that (?)‐prothioconazole were 85 to 2768 times more active than (+)‐prothioconazole toward these target organisms. In order to reflect the risk to aquatic ecosystem, the acute toxicity of the enantiomers to Daphnia magna, Chlorella pyrenoidosa, and Lemna minor L. was assessed. It was observed that the toxicity of (?)‐prothioconazole to D. magna was 2.2 times higher than (+)‐prothioconazole, but it was lower to C. pyrenoidosa and L. minor L. The toxicities of (+)‐enantiomer and (?)‐enantiomer to D. magna and C. pyrenoidosa were synergy, indicating that the racemate had higher threat to the organisms. It could be concluded that the effects of prothioconazole on target organisms and the acute toxicity to nontarget species were enantioselective with (?)‐enantiomer possessing higher efficiency and lower toxicity. Such enantiomeric differences should be taken into consideration when assessing the performance of prothioconazole.  相似文献

Short-term toxicity tests with Daphnia magna are reasonably reproducible. The sensitivity of Daphnia pulex, Daphnia cucullata and Daphnia magna proved to be about the same in short-term tests with 15 different chemical compounds.Reproduction toxicity experiments with Daphnia magna can easily be carried out within three weeks. At least duplicate experiments are necessary to arrive at an approximate no-effect level.  相似文献

Diaphanosoma brachyurum (Cladocera: Sididae) is a common limnetic species in summer-temperate and tropical water bodies. Few studies have investigated the sensitivity of D. brachyurum to toxic chemicals despite this species often being dominant in natural lakes and ponds. We performed acute toxicity tests of three heavy metals, copper (Cu), zinc (Zn), and cadmium (Cd), to D. brachyurum. For D. brachyurum, the lethal concentration (LC)50 values of Cu (24-h LC50 = 16.4 μg/L, 48-h LC50 = 10.4 μg/L) and Zn (24-h LC50 = 253.4 μg/L, 48-h LC50 = 174.1 μg/L) were lower than those for D. magna, one of the most used test organisms for toxic chemicals. On the other hand, for D. brachyurum the 24-h LC50 of Cd (166.4 μg/L) was much greater than that for D. magna, and the 48-h LC50 of Cd (69.8 μg/L) was comparable. Our results indicate that D. brachyurum may be more strongly influenced by Zn and Cu than is D. magna. It is likely that the summer plankton community in which Diaphanosoma species is dominant is more sensitive to heavy metals than a community in which Daphnia species are dominant.  相似文献

The sensitivity of neonates of four Daphnia species to zinc was tested in relation to their mean body size. These mean sizes of these four Daphnia spp were: D. magna, 0.813 ± 0.055 mm, D.␣pulicaria, 0.745 ± 0.063 mm, D. pulex, 0.645 ± 0.044 mm and D. galeata, 0.611 ± 0.058 mm. A positive relationship between EC50 (24, 48) values and neonates size was found. The smaller the size of the daphnid the higher was the sensitivity to heavy metal toxicity. For all tested species did the EC50 values decrease with time; the decrease was most marked for D. magna and the least for D. galeata. The EC50 values of D. magna were higher than would be expected on basis of its body size.  相似文献

To test inter- and intraspecific variability in the responsiveness to elevated CO2, 9–14 different genotypes of each of 12 perennial species from fertile permanent grassland were grown in Lolium perenne swards under ambient (35 Pa) and elevated (60 Pa) atmospheric partial pressure of CO2 (pCO2) for 3 years in a free air carbon dioxide enrichment (FACE) experiment. The plant species were grouped according to their functional types: grasses (L. perenne, L. multiflorum, Arrhenatherum elatius, Dactylis glomerata, Festuca pratensis, Holcus lanatus, Trisetum flavescens), non-legume dicots (Rumex obtusifolius, R. acetosa, Ranunculus friesianus), and legumes (Trifolium repens, T. pratense). Yield (above a cutting height of 4.5 cm) was measured three times per year. The results were as follow. (1) There were highly significant differences in the responsiveness to elevated pCO2 between the three functional types; legumes showed the strongest and grasses the weakest yield increase at elevated pCO2. (2) There were differences in the temporal development of responsiveness to elevated pCO2 among the functional types. The responsiveness of the legumes declined from the first to the second year, while the responsiveness of the non-legume dicots increased over the 3 years. During the growing season, the grasses and the non-legume dicots showed the strongest response to elevated pCO2 during reproductive growth in the spring. (3) There were no significant genotypic differences in responsiveness to elevated pCO2. Our results suggest that, due to interspecific differences in the responsiveness to elevated pCO2, the species proportion within fertile temperate grassland may change if the increase in pCO2 continues. Due to the temporal differences in the responsiveness to elevated pCO2 among species, complex effects of elevated pCO2 on competitive interactions in mixed swards must be expected. The existence of genotypic variability in the responsiveness to elevated pCO2, on which selection could act, was not found under our experimental conditions. We investigate the uncertainties associated with modeling the potential health effects on piscivorous animals of mercury released to the atmosphere. The multimedia modeling system combines an atmospheric fate and transport model, an aquatic cycling model, and a terrestrial food web model. First, the modeling system is used to calculate point values of the animals' hazard quotients (i.e., measures of toxic dose). Next, we use a simplified version of the modeling system to conduct a probabilistic analysis for the Great Lakes region that takes into account input uncertainty, variability, and uncertainty and variability combined. The use of two different software packages for the combined uncertainty/variability analysis led to similar results except for high values (>90th percentile) where some differences were evident. A sensitivity study was performed on the combined uncertainty and variability analysis. Regional variability caused more than 70% of the variance in the results, with the fish bioaccumulation factor accounting for the majority of the variability. The major sources of uncertainty were the speciation of the mercury emissions, the lake pH, and the sediment burial rate.  相似文献

This study reports on the factors involved in regulating the composition and structure of bacterial communities epiphytic on intertidal macroalgae, exploring their temporal variability and the role of copper pollution. Culture-independent, molecular approaches were chosen for this purpose and three host species were used as models: the ephemeral Ulva spp. (Chlorophyceae) and Scytosiphon lomentaria (Phaeophyceae) and the long-living Lessonia nigrescens (Phaeophyceae). The algae were collected from two coastal areas in Northern Chile, where the main contrast was the concentration of copper in the seawater column resulting from copper-mine waste disposals. We found a clear and strong effect in the structure of the bacterial communities associated with the algal species serving as host. The structure of the bacterial communities also varied through time. The effect of copper on the structure of the epiphytic bacterial communities was significant in Ulva spp., but not on L. nigrescens. The use of 16S rRNA gene library analysis to compare bacterial communities in Ulva revealed that they were composed of five phyla and six classes, with approximately 35 bacterial species, dominated by members of Bacteroidetes (Cytophaga-Flavobacteria-Bacteroides) and α-Proteobacteria, in both non-polluted and polluted sites. Less common groups, such as the Verrucomicrobiae, were exclusively found in polluted sites. This work shows that the structure of bacterial communities epiphytic on macroalgae is hierarchically determined by algal species > temporal changes > copper levels.  相似文献

We conducted an outdoor container experiment to test the hypothesis that the genetic composition of resident populations influences the establishment success of immigrant species. We manipulated the genetic compositions (source populations) of populations of the water flea Daphnia magna, a strong competitor in pond and shallow lake zooplankton communities, and monitored the establishment success of immigrant cladoceran species of a regional species pool. We show that establishment success is affected by the source population of the resident D. magna as well as by the presence/absence of macrophytes and the presence/absence of fish in the containers. Our results provide evidence that the genetic composition of resident populations can impact community assembly and metacommunity dynamics, and that community genetics can influence ecosystem functioning.  相似文献

The production and release of chemical compounds by invasive plants can affect competitors and native species overall, destabilizing ecological interactions and harming ecosystem functioning. Hedychium coronarium is an invasive macrophyte common on Brazilian riparian areas that produces a wide variety of allelochemicals, but little is known about their effect on aquatic species. Here, we identified the major chemical compounds of the aqueous extract of H. coronarium rhizomes and assessed its toxicity, evaluating the growth inhibition of one alga (Raphidocelis subcapitata) and one macrophyte (Lemna minor), and the lethality of cladoceran (Ceriodaphnia silvestrii and Daphnia similis) and Chironomidae larvae (Chironomus sancticaroli). The majoritarian compounds of H. coronarium rhizomes were Coronarin D and Coronarin D Ethyl Ether. The aqueous extract was toxic for all tested species. We observed growth inhibition in R. subcapitata, as well as reduction in biomass in L. minor. Chironomus sancticaroli and cladoceran were the most sensible species. The aqueous extract of H. coronarium rhizomes was toxic on tested conditions, suggesting that the rhizome compounds may interfere on aquatic organisms and in the dynamic of trophic webs of aquatic ecosystems on invaded areas.

Objective: The purpose of the study was to present smoothed percentiles for body weight and height, waist circumference, and body mass index (BMI) in Cypriot children and to compare their BMI 85th and 95th percentiles with those of children in other countries. Research Methods and Procedures: The study was a cross‐sectional study, including a representative sample of 2472 healthy children (49.1% boys) in Cyprus ages 6 to 17 years, who were evaluated during the 1999–2000 school year. Body weight and height and waist circumference were measured using standard procedures. BMI was calculated as weight in kilograms per height in square meters. Smoothed, sex‐specific percentiles for these variables were calculated using polynomial regression models. Crude weight, height, waist, and BMI percentile values are presented in sex‐specific tables and smoothed percentile curves are presented in charts. The 85th and 95th percentiles for BMI were compared with measurements from other countries, because of the concern of the upper limits of BMI in respect to the evaluation of obesity. Results: The 85th and 95th BMI percentile values are higher in Cypriot boys than in Swedish and Iranian boys through all ages and in girls ages 6 to 15 years, whereas after the age of 15 years, both Swedish and Iranian girls’ percentiles are equalized with their Cypriot peers. Discussion: Weight, height, waist circumference, and BMI values and charts are presented for the first time for Cypriot children and adolescents. Much concern should be addressed to the observation that for the majority of the Cypriot sample, the upper BMI limits are higher than the peers of developing and developed countries.  相似文献
