荞头(Alliumchinense)中三倍体广养和四倍体长沙养,于8月底取鳞茎播种于本校实验地,9月抽苔现蕾期至10月花谢期,分别取花药和雌蕊,以醋酸洋红压片确定花药发育阶段。每样取鲜重50mg以上,测过氧化物酶活性(据张志良.植物生理学实验指导.北京:高等教育出版社,1990.154),3次重复取平均值。脯氨酸提取(据Kern等.CropScience,1972,12:835)后用日立835-50型氨基酸分析仪测定。另以平行样品测定含水量,获得干重。结果(表1)表明:1.不同品种荞头及花器官不同部位过氧化物酶在同一小孢子发育阶段的活性不同。花药中过氧…  相似文献

多花黑麦草幼苗于3 叶期转移至1/2Hoagland 溶液中培养,同时加入PEG6000 用以模拟5种强度的渗透胁迫。植物叶内过氧化物酶活性、游离脯氨酸含量和质膜相对透性均对渗透胁迫强度的增加作出了反应,但反应的敏感性依次降低。从时间进程上看,过氧化物酶活性升降在前,游离脯氨酸积累继之,嗣后出现质膜相对透性增高。每个测定日内叶片中游离脯氨酸含量与质膜相对透性呈显著至极显著正相关。  相似文献

人为控制环境下,对玉米幼苗叶片中电解质外渗率、MDA含量以及保护酶活性的昼夜变化同步进行了测定。结果表明:叶片电解质外渗率的昼夜变化呈现双峰曲线,分别在光照4 h和光照后黑暗4 h达到峰值;叶片中MDA含量表现为单峰的波形曲线,黑暗开始的4 h内最高;随着光照时间的延长,SOD酶、POD酶和CAT酶活性均表现出不同程度的降低,但均在16:00(光照8 h)至20:00(光照12 h)之间达到一昼夜内的最低点。随着光照后黑暗时间的延长,各保护酶活性均增加,但增加的速率不同,至次日8:00所有保护酶活性均接近光照前水平。光照和黑暗条件下,各保护酶活性差异极显著。保护酶活性的昼夜变化与叶片中的可溶性蛋白质含量并无显著的相关关系。  相似文献

外源精胺(Spm)降低了离体小麦叶片衰老时MDA的含量,且降低程度与精胺的浓度成正比。0.2mmol/L的精胺提高了小麦幼苗体内的超氧化物歧化酶(SOD),过氧化氢酶(CAT),过氧化物酶(POD)及抗坏血酸过氧化物酶(ASP)的活性。体内及体外试验表明精胺既可诱导SOD与POD的合成,又可直接作用于酶分子上以提高酶的活性;精胺对CAT合成仅能诱导,对已有酶活性无调节作用;精胺对ASP的合成无影响,却能促进已有酶的活性。  相似文献

热激诱导的玉米幼苗耐热性及其与脯氨酸的关系   总被引:2,自引:1,他引:1  
研究了热激对玉米幼苗耐热性的效应及其与脯氨酸的关系。结果表明,培养2.5 d的玉米幼苗经过42℃热激4 h并于26.5℃下恢复4 h后,提高了玉米幼苗在48℃下的存活率,并且热激及其后的恢复过程中都表现出脯氨酸的积累。不同浓度的外源脯氨酸预处理也可提高玉米幼苗内源脯氨酸的水平和抗氧化酶抗坏血酸过氧化物酶(APX)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽还原酶(GR)、过氧化物酶(GPX)的活性,从而提高玉米幼苗在高温胁迫下的存活率。这些结果暗示热激过程中脯氨酸的积累所诱发的抗氧化酶活性的增强可能是热激诱导的玉米幼苗耐热性形成的生理基础之一。  相似文献

SO_2对玉米幼苗POD、Pro和MDA的影响   总被引:2,自引:0,他引:2  
本文采用室内水培及密闭箱静态熏气方法,研究了4个不同浓度(0mg/m3,10mg/m3,30mg/m3和90mg/m3)SO2对晋单51和农大108两个品种玉米幼苗过氧化物酶(POD)、脯氨酸(Pro)和丙二醛(MDA)的影响。研究结果表明,随着SO2浓度的升高,两种玉米幼苗的POD活性呈下降的趋势,且当SO2浓度为10mg/m3、30mg/m3和90mg/m3时POD活性分别降至对照的22.6%、18.9%和12.1%;而结果发现Pro含量随SO2浓度升高逐渐升高,且当SO2浓度与POD处理相同时脯氨酸含量分别上升至对照的195.0%、235.4%和496.5%,SO2浓度为90mg/m3时Pro含量最高,且不同浓度处理时农大108Pro含量均比晋单51高;然而,MDA含量随SO2浓度的升高呈先降低后逐渐升高的趋势,在90mg/m3处升至最高值为对照的310.6%,仍然是农大108MDA含量比晋单51高。因此研究显示低浓度SO2能使玉米幼苗适应胁迫,但高浓度SO2暴露会影响植物生长发育。  相似文献

以金沙江干热河谷主要树种坡柳、银合欢和山毛豆幼苗为材料,通过盆栽苗自然干旱胁迫,同时以浇水处理为对照,探讨了干旱胁迫下三树种幼苗叶片的相对含水量、质膜透性以及脯氨酸含量的变化情况。结果表明,干旱胁迫后三树种叶片的相对含水量、质膜相对透性以及脯氨酸含量都发生了变化,只是变化的幅度和进程不同。三项生理指标的综合分析及结合其自然表现,坡柳具有较强的抗旱能力,其次为银合欢,最后是山毛豆。  相似文献

在有K无Na、有Na无K、无K无Na三种培养液中培养番茄和玉米幼苗,试验表明,在有Na无K液的培养下,番茄和玉米幼苗叶片游离脯氨酸含量比无K无Na溶液培养的低;可溶性糖含量和叶绿素a/b比值比无K无Na溶液培养的高;Na可以提高过氧化物酶活性。因此,在一定程度上Na可代替K对番茄、玉米幼苗的生长起促进作用。  相似文献

The effects of putrescine and ethephon on peroxidase (POD; EC 1.11.1.7), polyphenol oxidase (PPO; EC 1.14.18.1), catalase (CAT; EC 1.11.1.6) activities and proline content in spinach leaves under saline stress were investigated. In control conditions, putrescine increased PPO and CAT activities and proline content, but decreased POD activity. Ethephon increased these three enzyme activities but did not affect proline content. In saline conditions, putrescine increased POD and CAT activities and proline content, while it decreased PPO activity. Ethephon increased both PPO and CAT activities and proline content, but decreased POD activity. Putrescine and ethephon have opposite effects on the enzyme activities and proline accumulation because they acts as antagonists.  相似文献

在华北平原潮土上进行田间试验,研究施氮量对夏玉米根际和非根际土壤酶活性及氮含量的影响.结果表明: 不同施氮水平下土壤酶活性均表现出一致的季节性变化趋势.与不施氮处理相比,施氮处理能显著提高根际和非根际土壤硝态氮含量及根际土壤铵态氮含量,显著增强土壤几丁质酶、β-葡萄糖苷酶、纤维素酶和木聚糖酶活性.在整个玉米生育期内,非根际土壤硝态氮含量显著高于根际土壤;非根际土壤铵态氮含量在灌浆期显著高于根际土壤,但在苗期和完熟期则显著低于根际土壤;根际土壤几丁质酶、β-葡萄糖苷酶、纤维素酶和木聚糖酶活性都显著高于非根际土壤.施氮对土壤有机质含量没有显著影响;在0~180 kg·hm-2施氮范围内,施氮可明显增加土壤全氮含量,但当施氮量超过180 kg·hm-2时,土壤全氮含量则明显下降.综上,适量施氮能显著增强土壤酶活性,提高土壤全氮含量,改善土壤生化性质.  相似文献

以祁连圆柏和圆柏为材料,研究了圆柏属植物的抗寒性与解剖结构变化,以及淀粉与可溶性糖粉转化的关系.结果表明,祁连圆柏和圆柏叶片的上下表皮细胞外覆盖一层角质膜,叶肉细胞间隙大,形成发达的通气组织.在生长季节,二者叶绿体内均积累了很多淀粉粒,在冷冻休眠季节淀粉粒消失或变小,同时叶片可溶性糖分含量增加,祁连圆柏的可溶性糖分增幅高于圆柏;而休眠季节圆柏叶绿体受到低温伤害,脂质球数目增多,出现脂质小滴,部分叶绿体变形,祁连圆柏类囊体的结构受到影响较小,脂质球数量少,叶绿体形状没有发生明显的变化.因此,祁连圆柏和圆柏抗寒性与发达的通气组织和淀粉粒生长季节的积累有关,在低温胁迫下祁连圆柏叶绿体表现出高的稳定性,通过可溶性糖分的调节力强,耐寒性增强.  相似文献

BACKGROUND AND AIMS: Allowing for dispersal limitation, a species' geographic distribution should reflect its environmental requirements. Comparisons among closely related species should reveal adaptive differentiation in species characteristics that are consistent with their differences in geographic distribution. This expectation was tested by comparing characteristics of seedlings of spruce species in relation to environmental factors representative of their current natural ranges. METHODS: Seedlings were grown from a total of 34 populations representing eight North American spruce (Picea) species in a controlled environment chamber for 140 d. Traits related to the potential of seedling establishment, including tolerance to stress events (high temperature, desiccation) were evaluated. Correlations were sought between these characteristics and modal values of latitude, aridity and continentality in the geographic range of each species. KEY RESULTS: Many seedling traits changed significantly in response to stress events, but only the response of chlorophyll concentration differed significantly among species. Components of seedling growth were good correlates of species distribution. Seedling relative growth rate (RGR) and specific leaf area (SLA) were positively correlated with latitude, and leaf weight ratio (LWR) negatively correlated with aridity. Seed mass was negatively correlated with latitude. CONCLUSIONS: Relationships found between seedling traits and geographical variation in environmental conditions suggest that factors such as temperature regime, water availability and perhaps litter depth affect species range in North American spruces. Seedling characteristics appear to be elements in a reasonably distinct environmental niche for each spruce species at the continental scale.  相似文献

采用相同浓度(25μgNO2·m^-3)不同处理时间(1、3、5、7d)和相同处理时间(2h)不同浓度(40、60、80、100μgNO2·m^-3)两种方法进行熏气处理,研究了汽车尾气对五角槭、山荆子、山梨和茶条槭苗木叶液pH值、相对电导率、MDA含量、叶绿素含量、SOD和POD活性及ASA含量的影响.结果表明.随着熏气时间的延长和熏气浓度的增加,4个树种苗木的叶液pH值、叶绿素含量和ASA含量均逐渐降低,相对电导率、MDA含量、SOD和POD活性逐渐上升,不同树种苗木叶片组织的各项指标间存在较大差异,相同浓度处理7d后.山梨苗木叶片组织的pH降幅最大,山荆子次之,茶条槭最小;叶绿素含量的下降顺序为山梨>山荆子>茶条槭>五角槭;ASA含量的下降幅度以五角槭和茶条槭较小,山荆子和山梨较大,随着处理时间的延长.五角槭苗木叶片组织的相对电导率和MDA含量上升幅度最大.分别比对照升高了68.1%和52.50%,山荆子次之,茶条槭最小.随着处理浓度的增加,山荆子苗木的相对电导率和MDA上升幅度最大,分别比对照升高99.8%和52.5%,山梨次之,五角槭和茶条槭较小.4个树种苗木的自由基清除系统受到明显影响.SOD、POD活性呈上升趋势,茶条槭除ASA变幅稍高于五角槭外.各项生理指标的变化明显低于其它3个树种,表现出相对较强的抗性,山荆子的各项生理指标变化幅度相对较大,且其SOD和POD在最高浓度处理时开始遭到破坏.表现出相对较弱的抗性。  相似文献

Effects of cadmium (Cd(2+)) on photosynthetic and antioxidant activities of maize (Zea mays L.) cultivars (3223 and 32D99) were investigated. Fourteen-day-old cultivar seedlings were exposed to different Cd concentrations [0, 0.3, 0.6 and 0.9mM Cd(NO(3))(2).4H(2)O] for 8 days. The results of chlorophyll fluorescence indicated that different levels of Cd affected photochemical efficiency in 3223 much more than that in 32D99. In parallel, the level of Cd at 0.9mM caused oxidative damage but did not indicate cessation of PSII activity of the cultivars; plant death was not observed at highly toxic Cd levels. Additionally, the increase in Cd concentration caused loss of chlorophylls and carotenoid and membrane damage in both cultivars, but greater membrane damage was observed in 3223 than in 32D99. Depending on Cd accumulation, a significant reduction in dry biomass was observed in both cultivars at all Cd concentrations. The accumulation of Cd was higher in roots than in leaves for both cultivars. Nevertheless, cultivar 3223 transferred more Cd from roots to leaves than did 32D99. On the other hand, our results suggest that there were similar responses in SOD, APX and GR activities with increasing Cd concentrations for both cultivars. However, POD activity significantly increased at highly toxic Cd levels in 32D99. This result may be regarded as an indication of better tolerance of the Z. mays L. cultivar 32D99 to Cd contamination.  相似文献

The relationship between specific rate of respiration (respiration rate per unit root dry weight) and concentration of reduced nitrogen was examined for maize ( Zea mays L.) roots. Plants with 2 primary nodal root axes were grown for 8 days in a split-root hydroponic system in which NO-3 was supplied to both axes at 1.0 mol m−3, to one axis at 1.0 mol m−3 and the other axis at 0.0 mol m−3 or to both axes at 0.0 mol m−3 Respiration rates and root characteristics were measured at 2-day intervals. Specific rate of respiration was positively correlated in a nonlinear relationship with concentration of reduced nitrogen. The lowest specific rates of respiration occurred when neither axis received exogenous NO−3 and the concentration of reduced nitrogen in the axes was less than 9 mg g−1. The greatest rates occurred in axes that were actively absorbing NO−3 and contained more than 35 mg g−1 of reduced nitrogen. At 23 mg g−1 of reduced nitrogen, below which initiation of lateral branches was decreased by 30–50%. specific rate of respiration was 17% greater for roots actively absorbing NO−3 than for roots not absorbing NO−3 Increases in specific rate of respiration associated with concentrations of reduced nitrogen greater than 23 mg g−1 were concluded to be attributable primarily to proliferation of lateral branches.  相似文献

The association of enzyme activities in developing kernels with specific storage product accumulation at maturity was analyzed in different parts of Zea mays inbred OH43 kernels. Maize kernels were harvested at 20 and 55 days post-pollination and dissected into basal region, pericarp, embryo, lower endosperm, middle endosperm and upper endosperm. Mature (55 days pos(-pollination) kernel parts were analyzed for starch, total protein, zein and oil content. Immature (20 days post-pollination) kernel parts were assayed for activities of 15 enzymes of sugar and amino acid metabolism. Statistical analyses of the data suggested that glucokinase (EC 2.7.1.2) fructokinase (EC 2.7.1.4) and phosphofructokinase (EC 2.7.1.1 11) activities were primarily associated with oil accumulation, whereas ADP'-glueose pyrophosphorylasc (EC 2.7.7.27) and sucrose synthase (EC 2.4.1.13) activities were associated with starch accumulation. The results suggest that oil biosynthesis utilizes inveitase-mediated sucrose degradation in a pathway not requiring pyrophosphatc. whereas starch biosynthesis utilizes a sucrose synthase-mediated pathway of sucrose degradation in a pathway requiring pyrophosphatc. Additional groups of enzyme activities were associated with each oilier but not with any specific storage product and appeared to be associated with general metabolic activity.  相似文献

Two controlled experiments were conducted to evaluate the potential for vascular plants to germinate and establish in milled peatlands and to assess whether easily measured plant traits can be used to predict their probable success. Study species included twenty species of perennial herbs, shrubs and trees occurring frequently in undisturbed bogs or abandoned milled bogs in Québec, Canada. First, a glasshouse experiment was performed to test the effect of burial under peat on germination and seedling emergence. Second, a growth chamber experiment was conducted to measure relative growth rate and other growth parameters of seedlings between 1 and 3 weeks of age. In the burial experiment, seedling emergence decreased exponentially with peat depth for most species examined. The slope of the exponential decline varied between species and was strongly correlated to seed mass. Seeds less than 0.1 mg in mass were most sensitive to burial. In the seedling growth experiments, Betula species had the highest absolute and relative growth rates, which may help to explain their prevalence in milled bogs. Relative growth rate (RGR) was not correlated with seed mass, however it was strongly correlated with leaf area ratio (LAR) and especially specific leaf area (SLA) of seedlings, except for species with seed mass less than 0.01 mg. Screening of species for seed mass and SLA should help predict their germination and establishment success or failure in milled peatlands and allow more directed interventions to favour the establishment of desirable species in milled bogs.
