3-Deoxy-d-arabino-heptulosonate 7-phosphate synthase, the first enzyme of the shikimate pathway, was purified to electrophoretic homogeneity from tubers of Solanum tuberosum L. cv Superior. The enzyme is a dimer with a native molecular weight of 110,000. The enzyme appears to be hysteretic. The enzyme activity is stimulated by Mn2+ and l-tryptophan. Chromatofocusing resolved two forms of the enzyme with isoelectric points of 7.8 and 8.4, respectively. The enzyme closely resembles an analogous activity previously isolated from roots of Daucus carota (JA Suzich, JFD Dean, KM Herrmann 1985 Plant Physiol 79: 765-770). 相似文献

One-node potato (Solanum tuberosum L. cv. Katahdin)cuttings were used to study early anatomical changes associatedwith tuberization. Starch deposition and the percentage frequencyof cells in mitosis increased in the medullary region of thebud within 1 d after cutting, whereas increases in average cellsize were not detected until 4 d after cutting. Age-dependent changes in the concn (nmol g d. wt1) andrelative proportion of polar lipid fatty acids in potato tubertissue were characterized over a 32-month storage period. Astubers advanced in age from 2 to 14 months, a decrease (approximately22%) in concn of both saturated (16:0, 18:0) and unsaturated(18:2, 18:3) fatty acids was evident. This decrease was followedby an exponential increase through 32 months. Electrolyte leakagestudies with excised tissue revealed age-dependent differencesin membrane permeability. The relationship between maximum electrolyteleakage (% total) and tissue age was described by a cubic polynomial.Leakage declined approximately 4% as tissues aged from 2 to7 months; however, from 7 to 24 months, leakage increased 21%.These trends were followed by partial restoration of membranefunction in 24 to 32-month-old tissues, as indicated by attenuatedleakage. Similarly, the trend in double bond index (DBI) withage was defined by a cubic polynomial; however, the minima andmaxima were the opposite of those for electrolyte leakage vs.age. The regression of DBI upon leakage was inverse and linear(r = 0·97, P < 0.01). Furthermore, the effectof decreasing temperature from 5 to 2 °C on tissuepermeability (after equilibration with the bathing medium) wasalso dependent upon DBI. The intensity of “tuberization stimulus” in potato shoots (Solanum tuberosum L.) can be assessed from cuttings containing one or more leaves. Cuttings maintained in a mist chamber under long days will form tubers from underground buds if prior to taking the cutting the leaves received sufficient exposure to photoperiods less than the critical photoperiod. The greatest tendency to tuberize was found in cuttings that consisted of a single, fully expanded leaf and its subtended bud. Grafts showed that genetical differences in critical photoperiod resided in properties of the leaf. Short days before cutting tended to shift growth from above ground buds of two-node cuttings to below ground buds, even if the number of short days was insufficient for tuber induction. As few as 6 short days reduced growth of shoots at the upper bud and increased underground growth of shoots and stolons. 相似文献

The time course of sulfate uptake in relation to respiration is shown for aging slices of potato, Solanum tuberosum L. cv. Russet. For comparative purposes, a time course for phosphate uptake is also shown. Malonic acid depressed both the respiratory rise and the sulfate accumulating ability of aging discs. A low dose of ultraviolet radiation given at the onset of aging reduced sulfate uptake by one-half without lowering respiration. We hypothesize that sulfate uptake is controlled by at least two mechanisms: one associated with oxidative metabolism and the other mediated by protein synthesis. 相似文献

为探究马铃薯HXK家族的功能,本研究利用生物信息学方法,对马铃薯HXK基因家族成员进行鉴定,并采用实时荧光定量PCR对该基因在不同模拟逆境胁迫下的表达进行分析。研究利用同源比对方法从马铃薯基因组中鉴定获得到6个马铃薯HXK基因,分别命名为StHXK1-StHXK6。该家族基因分别分布于马铃薯的6条染色体上,其编码蛋白长度为497~533 aa,等电点为5.36~6.11,分子质量在53 736.38~58 184.71 Da,均为亲水蛋白。亚细胞定位预测表明,StHXK家族成员主要在叶绿体中表达。该家族成员二级结构均以α-螺旋和不规则卷曲为主,三级结构高度相似。顺势元件分析发现马铃薯HXK基因主要包含光响应元件、激素响应元件、胁迫响应元件及生长发育相关元件。qRT- PCR结果表明,StHXK基因在ABA诱导下均上调表达显著,50 μmol/L ABA胁迫下,StHXK1和StHXK2在处理24 h相对表达量较高,分别为对照的286.4倍和152.02倍,200 mmol/L NaCl处理下,除StHXK1和StHXK4基因相对表达量在处理时间2 h呈上调表达,其余StHXK基因均呈不同程度的下调表达。在10% PEG胁迫处理下,StHXK基因相对表达量呈不同程度的上调表达。综上所述,StHXK基因对ABA、NaCl和PEG胁迫处理均存在不同程度的响应。 相似文献

Essential Arginyl Residue at the Active Site of Pyrophosphate:Fructose 6-Phosphate 1-Phosphotransferase from Potato (Solanum tuberosum) Tuber 下载免费PDF全文
The aim of this work was to test the proposal that the active site of pyrophosphate:fructose 6-phosphate 1-phosphotransferase (PFP) contains an essential arginyl residue. Enzyme activity was inhibited equally in the glycolytic and gluconeogenic directions by arginine-modifying reagents. The second-order rate constants for 2,3-butanedione and phenylglyoxal were 13.1 [plus or minus] 0.45 and 55.3 [plus or minus] 1.3 M-1 min-1, respectively. The corresponding values for the kinetic order of inactivation by these modifying reagents were 0.84 [plus or minus] 0.049 for 2,3-butanedione and 0.89 [plus or minus] 0.052 for phenylglyoxal. The substrates, fructose 6-phosphate and pyrophosphate, and a range of substrate analogs protected the enzyme from inactivation by 2,3-butanedione. These data suggest that modification of no more than one arginyl residue at, or close to, the active site is required to inhibit the enzyme. This result supports the proposal that the active site of PFP in plants is equivalent to that of the bacterial ATP-phosphofructokinase (S.M. Carlisle, S.D. Blakeley, S.M. Hemmingsen, S.J. Potato shoots were propagated in vitro by placing nodes fromsprouted tubers on Murashige and Skoog type medium without hormones.The vigour of growth and the rate of node production increasedwith both day-length and temperature over the ranges 824h and 1525 °C respectively. Propagation rates ofup to x 10 per month were obtained. In vitro plantlets spontaneouslyformed roots either in agar or liquid cultures. Plantlets leftin the culture jars for 34 months eventually senescedand formed small tubers in 16 and 24 h day-lengths. Cell division and cell expansion during early stages of tuberdevelopment were studied using developing axillary buds on single-leafcuttings from potato (Solanum tuberosum L.). Cuttings takenfrom plants induced to form tubers, by short day (SD) treatment,were compared with cuttings from non-induced (long day, LD)plants. In the apical zone of the buds, cell division occurredfrom the first day after cutting, in both LD and SD cuttings.The planes of these divisions were transverse, associated withelongation of the buds. At day 5, a new orientation of celldivision was observed in the subapical zone of SD cuttings only.These divisions were longitudinal, associated with radial growth.Cell expansion occurred in both SD and LD cuttings, and wasnot uniquely related to the onset of tuber formation. The effects of a number of manipulative treatments together with the application of the growth hormones IAA and GA on leaf expansion in the potato, variety Majestic, were investigated. It was found that removal of the apex and its surrounding non-expanded leaves resulted in an increase in both the rate and the extent of expansion of Ihe remaining upper leaves on the system. Application of IAA and GA to decapitated shoots showed that the rate of leaf expansion was decreased by IAA both alone and in combination with GA, whereas it was increased by GA treatment. Further investigations indicated that the response of a leaf to GA treatment was determined by the area and position of the leaf al the time of treatment and also by the number of expanding leaves on the system. It is postulated that this is due, at least in part, to competition for some factor or factors, which could be nutrients, within the system. 相似文献

Kinetin-induced tuberization of isolated stolons was investigated with regard to accumulation of labelled kinetin, starch, protein and nucleic acid synthesis. Using kinetin-8-14C, it was found that more labelled meterial appeared at the locus of tyber formation that in other parts of the stolon. Substantial accumulation was evident before visible signs of tuber formation. The basal portion of the stolon also accumulated substantial amounts of labelled material. Kinetin-treated stolons showed extensive starch accumulation which was not evident in gibberellic acid-treated or untreated stolons. Starch accumulation occurred before any visible sign of tuber formation. There was no marked differences in the ability of the apical 0.5 cm of kinetin-treated and untrated stolons to incorporate 14 C uridine into RNA and 14C leucine into TCA precipitable protein. From these results it was concluded that kinetin-induced tuber formation may not involve the synthesis of new proteins. It is suggested that kinetin may be regulating tuber formation by suppressing starch hydrolase activity and stimulating starch synthetase activity whereas gibberellic acid inhibits tuber formation by promoting starch hydrolase activity or by suppressing starch synthesizing enzymes. 相似文献

Suberin, a cell specific, wall-associated biopolymer, is formed during normal plant growth and development as well as in response to stress conditions such as wounding. It is characterized by the deposition of both a poly(phenolic) domain (SPPD) in the cell wall and a poly(aliphatic) domain (SPAD) thought to be deposited between the cell wall and plasma membrane. Although the monomeric components that comprise the SPPD and SPAD are well known, the biosynthesis and deposition of suberin is poorly understood. Using wound healing potato tubers as a model system, we have tracked the flux of carbon into the aliphatic monomers of the SPAD in a time course fashion. From these analyses, we demonstrate that newly formed fatty acids undergo one of two main metabolic fates during wound-induced suberization: (1) desaturation followed by oxidation to form the 18:1 ω-hydroxy and dioic acids characteristic of potato suberin, and (2) elongation to very long chain fatty acids (C20 to C28), associated with reduction to 1-alkanols, decarboxylation to n-alkanes and minor amounts of hydroxylation. The partitioning of carbon between these two metabolic fates illustrates metabolic regulation during wound healing, and provides insight into the organization of fatty acid metabolism.Key Words: suberin, potato, Solanum tuberosum, carbon flux analysis, abiotic stress 相似文献

Tuberization response of single-node leaf cuttings from induced potato plants (Solanum tuberosum L.) was reversed when pretreated with 5 millimolar ethyleneglycol-bis-(beta-aminoethyl ether)N,N'-tetraacetic acid (EGTA) + 50 micromolar calcium ionophore (A23187) and resumed when transferred to a CaCl(2)-containing medium. Tuberization was inhibited by LaCl(3), chlorpromazine, and trifluoperazine at 5 to 10 micromolar. These results suggest a role for calcium in the tuberization process. 相似文献
