Clostridium putrefaciens grew well in most media used routinely for culturing anaerobes, but produced spores only on lactose-egg yolk agar. The D80° was 8–14 min, z value was 4°–6° and D γ, 0.16 Mrad. The inhibitory interactions of pH, NaCl, NaNO2 and incubation temperature are described.  相似文献

Abstract: Solubilization of rat striatal membranes with sodium cholate, followed by reconstitution into phospholipid vesicles, leads to a 6.5-fold increase in the agonist high-affinity binding sites of the D1 dopamine receptor. These high-affinity binding sites display differential sensitivity toward temperature. When reconstituted receptors were preincubated for 1 h at 0–4°C (on ice) or at 22°C (room temperature) followed by radioligand binding assays with dopamine, neither the high-affinity values of the receptor for dopamine nor the percent receptors in the high-affinity state (31–39%) were changed from control reconstituted receptors, which were not subject to any preincubations. At 30°C, there was a partial loss in the number of high-affinity D1 receptors with only 25% of the total receptor population in the high-affinity state; there was no change in the affinity values of the high-affinity binding sites. At 37°C, there was a 40% loss in total number of D1 receptor binding sites. All the high-affinity binding sites were lost and the remaining 60% of binding activity represented the low-affinity binding state of the receptor. These results indicate that the high-affinity binding sites of the reconstituted D1 dopamine receptors are uniquely sensitive to higher temperatures.  相似文献

1. Hyalella montezuma is endemic to Montezuma Well, Arizona, and is exposed to minimal diel and seasonal temperature fluctuations in the pelagic zone (21 ± 4 °C). Juvenile H . montezuma feed in the pelagic zone during the day and migrate into the littoral vegetation at night, while adults remain primarily in the littoral vegetation.
2. Oxygen consumption ( V O2) of adult and juvenile H . montezuma was measured at 20, 25 and 30 °C. The V O2 of both adult and juvenile H . montezuma increased with temperature. However, the V O2 of juveniles was significantly greater than that of adults at all temperatures, with greatest divergence at 30 °C where mean juvenile V O2 (6.31 μl mg–1 dry weight (DW) h–1) was almost twice that of adults (3.60 μl mg–1 DW h–1).
3. Survivorship of juveniles was significantly lower (54%) at 30 °C than at 27.5 °C (95%) after 4 h, whereas adults showed at least a 93% survivorship at both temperatures.
4. Our data suggest that temperature may have been the proximate cue that elicited the diel horizontal migration of juvenile H . montezuma in Montezuma Well, with the behaviour maintained and enhanced by intensive invertebrate predation in the pelagic and littoral zones.  相似文献

Objectives: To explore proliferation of glomerular mesangial cells (GMC) and secretion of extracelluar matrix (fibronectin induced by sublytic C5b-9 complexes), and then ascertain the role of phosphatidylinositol 3-kinase (PI3-k)/Akt signal pathway in these processes, by using small hairpin RNAs.
Material and methods: The expression of cyclin D2, 3H-thymidine into DNA and production of fibronectin including thrombospondin-1 and transforming growth factor-β1 in the GMCs stimulated by sublytic C5b-9 or transfected with expression vectors of PI3-k and Akt short hairpin RNA or LY294002 (PI3-k inhibitor) were measured by Real-time quantitative polymerase chain reaction (PCR), Western blot, enzyme-linked immunosorbent assay (ELISA) and 3H-thymidine incorporation (3H-TdR), respectively.
Results: The expression of cyclin D2, 3H-thymidine into DNA and fibronectin in the GMCs stimulated by sublytic C5b-9 could all be increased, and the elevations of these parameters mentioned above were also markedly reduced in the GMCs transfected with vectors of PI3-k and Akt short hairpin RNA or LY294002, respectively.
Conclusions: These data indicate that sublytic C5b-9 can promote proliferation of GMCs and secretion of fibronectin as well as synthesis of thrombospondin-1 and transforming growth factor-β1. The PI3-k/Akt signal pathway in these reactions, mediated by sublytic C5b-9 complexes, may play at least a partial role.  相似文献

1. The hypothesis that a 3 °C elevation in temperature and doubled CO2 concentration would have no effect on the synchronization of winter moth egg hatch with budburst in oak was tested by comparing the separate and interactive effects of ambient and elevated (+ 3 °C) temperature and ambient and elevated (doubled to 340 p.p.m.) CO2 in eight experimental Solardomes. In addition, an outdoor control was compared with the ambient temperature/CO2 treatment combination.
2. Elevated temperature accelerated darkening (preceding egg hatch by about 5–10 days) and hatching of eggs developing off the trees; elevated CO2 had no effect. The same effects were observed in eggs developing on the trees.
3. Within treatments, date of egg hatch was the same on trees with early or late budburst.
4. Egg darkening and budburst were closely synchronized at both ambient and elevated temperatures.
5. Both eggs and trees required fewer cumulative heat units (day degrees > 4 °C), for hatching and budburst, respectively, at ambient than elevated temperatures. The requirements in the outdoor control treatment were similar to those in the ambient Solardome treatment.
6. Egg hatch between 10 and 25 °C, on a temperature gradient in the laboratory, required a constant number of heat units; fewer were required below 10 °C.
7. Elevated temperatures, in the Solardomes and the field, delayed adult emergence from the pupae.
8. The results suggest that a general increase in temperature with climatic change would not affect the closeness of the synchronization between egg hatch of winter moth and budburst of oak.  相似文献

SUMMARY. 1. Nemurella pictetii Klapæplek took 2 years to complete its life cycle in both the laboratory and a small stream in the English Lake District.
2. Hatching time (days after oviposition for 10%. 50% and 90% of the eggs to hatch) and hatching period (days between dates for 10% and 90% hatched) decreased with increasing water temperature in the laboratory, and the relationships were well described by a power-law. Estimates of the mean time for 50% hatching in the stream varied between 16 and 31 days after oviposition. depending on temperature.
3. Larval instars numbered fifteen for males and seventeen for females with a constant ratio of 1.18 between successive instars (conformed with Dyar's rule). Larval growth was exponential at four constant temperatures in the laboratory; mean instantaneous growth rates were 0.40±0.01% day−1 at 5.9°C, 0.43±0.01% day−1 at 8.2°C, 0.46±0.01% day−1 at 12. 1°C. 0.56±0.02%day−1 at 19.8°C. No larvae survived after instar XI at 19.8°C.
4. Larval growth was exponential in the stream and was scarcely affected by variations in water temperature (range 4.2 -14.0°C); mean growth rates for three year-classes were 0.41±0.02, 0.43±0.08, 0.54±0.05% day−1. Their similarity to laboratory growth rates under optimum conditions suggests that the availability of resources, such as food and space, was not restricting growth in the stream.  相似文献

Abstract— The soluble proteins released from the synaptic vesicles of rat cerebral cortex were studied. One fraction (D4) of these proteins was released in parallel with release of acetylcholine when synaptic vesicles were incubated at 37°C for 10 min in isotonic medium. Another fraction (Dj) was liberated from synaptic vesicles when their membranes were ruptured by mild treatment under hyposmotic conditions and freeze-thawing after release of D1 fraction. Fractions D1 and D2 contained 12 and 9 per cent, respectively, of the total protein in the synaptic vesicles. Some properties of these fractions were investigated by zone electrophoresis and ultracentrifugation, and by measuring their binding capacities for [14C]acetylcholine and various enzyme activities related to acetylcholine metabolism.  相似文献

SUMMARY. Information on temperature (T°C) and time from fertilization to 50% hatch ( D days) for five species of salmonid fishes has been used to assess several mathematical models relating D and T . No single equation gave the best fit to all five data sets. The power law with temperature correction (α), log101 D = log10 a + b log10 ( T - α) and the quadratic, log10 D = log10 a + bT + b 1 T 2 (where a, b, b 1, and α are constants), each accounted for over 97 % of the variance of D and were good fits to the observed data points for all five species. There was little difference between the predictions obtained from these two equations within the range of observed temperatures. Therefore, the simpler power-law model is preferred. However, there were substantial within-species differences between values of D predicted from extrapolations of the two models from 2 or 3°C down to 0°C. When more data for low temperatures become available it will be possible to make a more objective choice of model.  相似文献

1 In a glasshouse experiment we studied the effect of raised CO2 concentration (720 p.p.m.) on CH4 emission at natural boreal peat temperatures using intact cores of boreal peat with living vascular plants and Sphagnum mosses. After the end of the growing season half of the cores were kept unnaturally warm (17–20 °C). The potential for CH4 production and oxidation was measured at the end of the emission experiment.
2 The vascular cores ('Sedge') consisted of a moss layer with sedges, and the moss cores (' Sphagnum ') of Sphagnum mosses (some sedge seedlings were removed by cutting). Methane efflux was 6–12 times higher from the Sedge cores than from the Sphagnum cores. The release of CH 4 from Sedge cores increased with increasing temperature of the peat and decreased with decreasing temperature. Methane efflux from Sphagnum cores was quite stable independent of the peat temperatures.
3 In both Sedge and Sphagnum samples, CO2 treatment doubled the potential CH4 production but had no effect on the potential CH4 oxidation. A raised concentration of CO2 increased CH4 efflux weakly and only at the highest peat temperatures (17–20 °C).
4 The results suggest that in cool regions, such as boreal wetlands, temperature would restrict decomposition of the extra substrates probably derived from enhanced primary production of mire vegetation under raised CO2 concentrations, and would thus retard any consequent increase in CH4 emission.  相似文献

1. The cold hardiness of the alpine cockroach Celatoblatta quinquemaculata was investigated. This species is found at 1360 m a.s.l. beneath schist slabs on the Rock and Pillar Range (Central Otago, New Zealand). Cockroaches were collected monthly from January to December 1996, and their LT50 and supercooling points determined.
2. Celatoblatta quinquemaculata was freezing tolerant throughout the year, with a lower lethal temperature in winter of – 8.9 °C. Celatoblatta quinquemaculata was also found frozen under rocks in the field when the under-rock temperature was below – 3 °C, and could survive being frozen at – 5 °C for 4 days in the laboratory.
3. There was a marked decrease in LT50 temperature from – 5.5 °C in April to – 7.5 °C in May. This coincides with decreasing temperatures from summer through autumn to winter, during which temperatures beneath snow-covered rocks may reach – 7.3 °C.
4. Supercooling points fluctuated during the year, with an increase from – 4.2 °C in autumn to – 3.4 °C in winter. Supercooling point was highest in spring, and changes in supercooling point do not appear to be related to changes in LT50.
5. Recordings of environmental temperatures from the Rock and Pillar Range suggest that cockroaches may undergo up to twenty-three freeze–thaw cycles in the coldest month of the year, and that they may remain frozen for periods of up to 21 h. Maximum cooling rates recorded in the field (0.01 °C min–1) were 100-fold slower than laboratory cooling rates, so survival estimates from laboratory experiments may be underestimates.  相似文献

At 14° C, standard metabolic rate (75·1 mg O2 h−1 kg−1), routine metabolic rate (108.8 mg O2 h−1 kg−1), active metabolic rate ( c . 380 mg O2 h−1 kg−1), critical swimming speed (Ucrit 1·7 BL s−1), heart rate 47 min−1), dorsal aortic pressure (3·2 kPa) and ventilation frequency (63 min−1) for triploid brown trout Salmo trutta were within the ranges reported for diploid brown trout and other salmonids at the same temperature. During prolonged swimming ( c . 80% U crit), cardiac output increased by 2·3-fold due to increases in heart rate (1·8-fold) and stroke volume (1·2-fold). At 18° C, although standard and routine metabolic rates, as well as resting heart rate and ventilation frequency increased significantly, active metabolic rate and certain cardiorespiratory variables during exercise did not differ from those values for fish acclimated to 14° C. As a result, factorial metabolic scope was reduced (2·93-fold at 18° C v . 5·13-fold at 14° C). Therefore, it is concluded that cardiorespiratory performance in triploid brown trout was not unusual at 18° C, but that reduced factorial metabolic scope may be a contributing factor to the mortality observed in triploid brown trout at temperatures near 18° C.  相似文献

Abstract: K+-evoked acetyl[3H]choline ([3H]ACh) release was inhibited in a concentration-dependent manner by apomorphine and the D2 agonist quinpirole in striatal slices prepared from euthyroid and hypothyroid rats. However, there was a significant increase in the maximum inhibition observed with both agonists in the hypothyroid compared with the euthyroid group, which paralleled the increased D2 agonist sensitivity reported for stereotyped behavior. The D2 antagonist raclopride decreased, and the D, antagonist SCH 23390 increased, the inhibition of [3H]ACh release by apomorphine, confirming an inhibitory role for D2 receptors and an opposing role for D1 receptors. Because there is no difference in D1 or D2 receptor concentration between the euthyroid and hypothyroid groups, it is suggested that thyroid hormone modulation of D2 receptor sensitivity affects a receptor-mediated event. Following intrastriatal injection of pertussis toxin (PTX), apomorphine no longer inhibited [3H]ACh release. In fact, increased [3H]- ACh release was observed, an effect reduced by SCH 23390, providing evidence that D1 receptors enhance [3H]- ACh release, and confirming that a PTX-sensitive G protein mediates the D2 response. As it has been reported that thyroid hormones modulate G protein expression, this mechanism may underlie their effect on dopamine agonist- mediated inhibition of ACh.  相似文献

The duration of the immature stages of Cacopsylla pyri L. was studied under field conditions by artificially infesting pear branches on several dates during the year. The duration of the egg stage decreased from winter to summer, as the season progresses and temperature rises, and slightly increased in September. It ranged from 27.4 to 6.7 days. The same trend was observed in the duration of the first three larval stages (L1−3) which ranged from 18.8 to 10.3 days. For eggs deposited during the period February–August the duration of the last two larval stages (L4−5) ranged from 17.5 to 12.1 days. However, the duration of L4−5 developed from eggs deposited in September and which give rise to winter-form adults were the longest observed. The rate of egg development was related to average ambient temperature with a highly significant linear relationship. This relationship indicates that the egg stage requires a constant number of 158.9 (SD = 5.0) of day-degrees above an average temperature of 2.31°C to complete development. The rate of development both of L1−3 and L4−5 were related to average ambient temperature with curvilinear relationships. These relationships indicate a proportional increase in the developmental rate as temperature rises between 10–22°C. At the higher average temperatures that occurred in the summer experiments (24–27°C) the acceleration of development of L1−3 is reduced and the developmental rate of L4−5 decreases. The developmental rates of L4−5 developed from eggs deposited in September did not follow the established relationship with temperature and they were lower than those in the other periods of the year with the same average temperature.  相似文献

Pinfish Lagodon rhomboides acclimation rates were determined by modelling changes in critical thermal minimum ( T crit min, ° C) estimates at set intervals following a temperature decrease of 3–4° C. The results showed that pinfish gained a total of 3·7° C of cold tolerance over a range of acclimation temperatures ( T acc, ° C) from (23–12° C), that cold tolerance increased with exposure time to the reduced temperature at all T acc, but that the rate of cold tolerance accruement (mean 0·14° C day−1) was independent of T acc. A highly significant ( P < 0·001) multivariate predictive model was generated that described the acclimation rates and thermal tolerance of pinfish exposed to reduction in water temperature: log10 T crit min= 0·41597 − 0·01704 T acc+ 0·04320 T plunge− 0·08376[log10 ( t + 1)], where T plunge is plunge temperature (° C) and t is the time (days). A comparison of the present data, with acclimation rate data for other species, suggests that factors such as latitude or geographic range may play a more important role than ambient temperature in determining cold acclimation rates in fishes.  相似文献

Abstract: The effects of D1 and D2 dopamine ligands on protein kinase C (PKC) activity were examined in synaptoneurosomes. Incubation with D1 agonists (SKF 38393, fenodopam), in the presence of calcium, decreased the soluble and increased the particulate PKC activity. These effects were reversed by SCH 23390, which by itself had the opposite effect of increasing the soluble and decreasing the particulate PKC activity. In contrast, incubation with the D2 agonists [LY 171555, (+)-3-(3-hydroxyphenyl)- N - n -propylpiperidine, RU 24213] increased the soluble and decreased the particulate PKC activity. These effects were reversed by sulpiride. (−)-3-(3-Hydroxyphenyl)- N - n -propylpiperidine had a D2 antagonist profile. Apomorphine showed a biphasic dose-response change; i.e., it decreased particulate PKC activity at the D2 receptor at low concentrations (0.1 µ M ) and increased it at the D1 receptor at higher concentrations (10 µ M ). Pretreatment with tetrodotoxin or omission of calcium in the incubation medium did not alter the responses of the D2 agonists, but it reversed the changes in PKC activity induced by the D1 agonists and converted the biphasic response of apomorphine to a monophasic inhibition. These results indicate that (1) D1 and D2 dopamine receptors are negatively coupled to PKC and (2) the increase in particulate PKC activity seen with the D1 drugs in the presence of calcium is mediated indirectly via a transneuronal effect.  相似文献

Provitamin D2 (ergosterol), vitamins D2 and D3 reduced the calcium-mediated peroxidase secretion in three types of sugarbeet cells in suspension cultures. Vitamin D2 was the most effective in habituated non-organogenic cells which were the less sensitive to calcium; provitamin D2 was the most effective in habituated organogenic cells, while normal non-organogenic cells were equally sensitive to the three types of vitamin D. The calcium ionophore A23187 slightly restricted peroxidase release in all cases, except in the habituated organogenic cells in the presence of calcium where it exerted a promotive effect. The inhibiting effect of vitamin D2 was not counteracted by the ionophore except in this habituated organogenic cell line.  相似文献

SUMMARY. 1. It was assumed that the average of twenty-four hourly temperature readings is the'true'daily mean. For a small stream in upper Tees-dale, northern England, estimates of the daily mean were obtained from: (a) The mean of temperature readings taken at regular but less frequent intervals than hourly (twelve, six and three times per day), (b) The daily mid-point (=1/2(maximum + minimum)). (c)Spot temperatures at the two times in the day when the daily temperature trace usually crosses the daily mean value. These estimates were then compared with the'true'mean.
2. The accuracy of the estimate of daily mean decreased with reduced frequency of reading but, even at only three readings per day, was always within ±0.5°C of the true value. Consolidation of the daily estimates gave estimates of monthly and annual means within ±0.1°C of the true values.
3. Nearly 96% of estimates of the daily mean based on the daily midpoint were within ±0.5°C of the true value. The estimates showed a small but statistically significant positive bias.
4. Over 80% of estimates of the daily mean based on the'first mean crossing'were within ±0.5°C of the true value and no bias could be shown.
5. Estimation of daily means from temperature at the time of the 'second mean crossing'was relatively complicated and gave less satisfactory results than did the first mean crossing.
6. The daily mid-point and the temperature at the time of the first mean crossing both gave a good general indication of daily mean temperatures. When consolidated to monthly or annual means the estimates were within ±0.25°C and ±0.10°C, respectively, of the true values.  相似文献

Abstract: Primary cultures of rat ventral mesencephalon were used to elucidate the role of chronic stimulation of dopamine (DA) D2 autoreceptors in the development of fetal dopaminergic neurons in vitro. Cultured dopaminergic neurons, as visualized by tyrosine hydroxylase immunocytochemistry, became more differentiated in the course of cultivation time and exhibited specific high-affinity uptake for [3H]DA. In rat striatal tissue, activation of D2 receptors has been shown to inhibit the release of DA. Previously accumulated [3H]DA was released from the cultures upon depolarization in a Ca2+-dependent manner. K+-evoked [3H]DA release could be inhibited by the selective D2 receptor agonists LY 171555 and N0437 in a concentration-dependent manner. The inhibitory effects of LY 171555 and N0437 were antagonized by the selective DA D2 receptor antagonist sulpiride. These observations are indicative for the expression of functional D2 receptors in the cultures. Daily treatment of these cultures for 7 days with LY 171555 or sulpiride did not lead to any change in protein content, the number of tyrosine hydroxylase-immunoreactive neurons, or the uptake capacity for [3H]DA. Our data demonstrate that chronic stimulation of DA D2 receptors does not impair survival or differentiation of cultured fetal dopaminergic neurons.  相似文献

Abstract. The physiologically active form of vitamin D3, 1,25-dihydroxy-vitamin D3, (1,25(OH)2, D3), induces differentiation of several types of myeloid leukaemia cells. The acquisition of monocyte-like phenotype is accompanied by slower progression through the cell cycle, and G1, block has been reported to be the basis of this effect. It is shown here that human promyelocytic leukaemia HL60 cells treated with analogues of vitamin D3, which are potent inducers of monocytic differentiation, have an additional cell cycle block. Exposure to 10-7m 1,25(OH)2, D3, or 1,25-(OH)2,-16-ene-D3 resulted in monocytic differentiation and the expected G1, block evident at approximately 48 h in a rapidly differentiating variant of HL60 cells (HL60-G), and at 96 h in the more slowly differentiating HL60-240 cells. In addition, a G2,+M block was noted at approximately 72 h in HL60-G and HL60-240 cells. Exposure to vitamin D3, analogues also markedly increased the number of dikaryons, suggesting that cytokinesis was impaired more than karyokinesis. Treatment with a third analogue 25-hydroxy-16,23-diene-D3, produced little differentiation and had minimal effects on the cell cycle parameters. These findings indicate that vitamin D3, analogues regulate cell proliferation by control of the transition of G1, and G2,+M phases, reminiscent of the cdc2/CDK2 type of cell cycle control.  相似文献
