The mating system of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), is based on leks. In field cage choice experiments we examined lek site selection as influenced by the presence of fruit and the presence of conspecific flies. Males were allowed to choose between artificial trees containing fruit and trees without fruit. Males showed a significant preference for the tree baited with fruit. To determine which fruit-related stimuli were important, males were allowed to choose between visual and/or olfactory fruit-derived stimuli. Males preferred to lek on trees presenting both stimuli. To determine the influence of conspecifics on lek site selection, males were allowed to choose between trees containing male and/or female conspecific flies. The presence of pheromone-emitting males was preferred. In addition, both in the field and on artificial trees, we examined the effect of leaf size on lek site selection. In the field, males preferred to lek on leaves of a particular size. Moreover, leaf integrity was important, as males tended to select undamaged leaves as lek sites. In field cage experiments, males consistently chose to lek on trees containing the largest leaves. We conclude that the choice of lek site is influenced by the presence of fruit and of other lekking males, while leaf size and integrity determine male location within a lek. 相似文献

Recent evidence suggests that the nutritional state of male Mediterranean fruit flies, Ceratitis capitata (Wied.) (medfly), is an important influence on various components of their reproductive biology, including mating success. The objective of the present study was to examine experimentally the effect of temporary starvation on the mating success of wild male C. capitata. Males were maintained on protein–sugar or sugar-only diets, and for each diet we compared the mating success of continuously fed males versus males starved for 18 or 24 h immediately before testing. In trials conducted on field-caged, host trees, males starved for 24 h obtained only about half as many matings as fed males for both diets. However, when the starvation period was 18 h, starved males reared on the protein–sugar diet mated significantly less frequently than fed males, whereas starved males reared on sugar mated as often as fed males. Measurements of male pheromone calling and female attraction revealed that reduced mating success likely reflected the decreased signaling activity of starved males. 相似文献

The induction of several biochemical indicators of larval tissue histolysis in the Medfly, Ceratitis capitata (Diptera: Tephritidae) (Wiedemann) was studied. Using synchronized third instar larvae, we have determined the time of occurence of gut evacuation (12 h before puparium formation, bpf), disappearance of digestive enzymes (10 h bpf), and jumping from the food (8 h bpf). We can also correlate these events temporally with other early landmarks of metamorphosis.The decrease in protein content between 0 hours and 144 hours after puparium formation (0–144 h apf) corresponds to a sharp increase of total acid proteolytic activity measured in vitro from 0 to 44 h apf. This activity appears to be lysosomal, judging by the activation of other lysosomal markers, such as acid phosphatase and -glycosidases. The maximum proteolytic activity occurs during the pre-pupa to pupa transition, i.e. during morphogenesis from the cryptocephalic to the phanerocephalic pupa. The results of endopeptidase inhibitor assays indicate that in dipterans, members of the aspartic and cystein proteinase families are responsible for the degradation of larval tissues. 相似文献

We examined the intra-tree foraging behavior of individually-released, wild-population Mediterranean fruit flies (medflies), Ceratitis capitata (Wiedemann), on field-caged host trees bearing each of three different densities (0, 3, or 12 per tree) of non-infested host fruit (kumquat) or each of two levels of fruit quality (12 non-infested fruit or 12 fruit infested with eggs and covered with host marking pheromone). With increasing density of non-infested fruit, medflies tended to remain longer in trees, visit more fruit before leaving, oviposit more often, accept a proportionately smaller number of fruit visited, and emigrate sooner after the last egg was laid (i.e. have a shorter Giving-Up-Time). Medflies spent much less time, oviposited much less often, and exhibited a longer Giving-Up-Time on trees harboring pheromone-marked fruit than non-infested fruit. Variation in temperature within the range at which experiments were conducted (25–36°C) had little detectable influence on foraging behavior. We compare our findings with published findings on the intra-tree foraging behavior of another tephritid fly, Rhagoletis pomonella (Walsh), and with current foraging behavior theory. We discuss implications of our findings with respect to medfly management strategies, particularly fruit stripping in eradication programs and use of synthetic marking pheromone for control.
Résumé Nous avons étudié le comportement de prospection dans un arbre, de femelles d'une population sauvage de C. capitata, libérées individuellement à l'intérieur de cages contenant des Eriobotrya japonica (kumquat), portant chacun 3 densités différentes de fruits no contaminés (0, 3, 12 par arbre) et chacun 2 niveaux de qualité de fruits: 12 fruits non infestés ou 12 fruits contaminés par des oeufs et recouverts de phéromone de marquage de l'hôte. C. capitata avait terndance à rester plus longtemps dans les arbres, à visiter plus de fruits avant le quitter, à pondre plus souvent, à accepter proportionnellement un nombre plus réduit de fruits déjà visités, à émigrer plus tôt après la ponte du dernier oeuf (c'est-à-dire à présenter un temps d'abandon plus bref), quand la densité des fruits non contaminés augmentait. C. capitata a dépensé beaucoup moins de temps, pondu beaucoup moins souvent, et présenté un temps d'abandon plus long sur les arbres portant des fruits marqués par la phéromone que sur ceux ayant des fruits non contaminés. Les variations de température dans la gamme de cells où les observations ont eu lieu (23–36°C) n'ont eu qu'une faible influence décelable sur le comportement de prospection. Nous avons comparé nos résultats avec ceux publiés sur la prospection à l'intérieur de l'arbre par une autre téphritide (Rhagoletis pomonella) et avec la théorie dominante sur le comportement de prospection. Nous discutons les conséquences de nos résultats sur les stratégies de lutte contre C. capitata, en particulier l'élimination des fruits dans les plans d'erradication et l'utilisation de phéromone synthétique de marquage.相似文献

Significant inter-populational differences in propensity to attempt boring into (accept) various types of fruit for oviposition were found among Ceratitis capitata females from two wild sources and one laboratory source. Evidence suggests that (a) fruit size had a strong influence whereas fruit taxonomic status had little influence on the acceptance pattern of each population, and (b) at least a portion of the inter-populational variation had a genetic basis.
Zusammenfassung Bei zwei Wildherkünften und einem Laboratoriumsstamm von Ceratitis capitata wurden Weibchen auf ihre Neigung zur Probebohrung in Früchte vor der Eiablage untersucht; dabei ergaben sich gesicherte Unterschiede zwischen den Populationen. Using a genetic sexing line based on pupal colour dimorphism in Ceratitis capitata Wied., field cage experiments were carried out with a wild population in Italy. The experiments were designed to monitor the competitiveness of males of this line in comparison with wild males and to assess the effect of removal of females prior to release on the proportion of wild females mated by the released males.The results showed that the genetic sexing strain competed well with the wild population but there was a large degree of intra-strain mating. Removal of females prior to release radically changed the mating pattern from intra to inter-strain mating. This change greatly increases the efficiency of the sterile insect method of control.It was concluded that as well as the obvious economic advantages of genetic sexing, significant biological advantages would also be associated with the integration of such a strain in a sterile insect control programme.
Résumé Des expériences en cage en champ ont été réalisées en Italie avec des populations naturelles de Ceratitis capitata en utilisant une lignée dont le sexage était basé sur le dimorphisme de la couleur des pupes. Les expériences ont été conçues pour contrôler la compétitivité des mâles de cette lignée face aux mâles sauvages, et d'évaluer l'effet du retrait des femelles avant le lâcher sur le pourcentage de femelles sauvages accouplées avec des mâles lâchés.Les expériences montrent que la compétition ne défavorise pas la lignée génétiquement sexable mais révèlent une fréquence élevée d'accouplement intralignée.Le retrait des femelles avant le lâcher modifie radicalement l'orientation des accouplements d'intra vers interlignées. Ce changement accroît fortement l'efficacité de la technique d'utilisation des mâles stériles dans la lutte biologique. On en a conclu que non seulement des intérêts économiques évidents mais aussi des avantages biologiques significatifs seraient retirés de l'intégration de telles lignées sexables génétiquement dans les programmes de lutte avec des mâles stériles.相似文献

Male Mediterranean fruit flies, Ceratitis capitata (Wiedemann), are attracted to the synthetic compound trimedlure. Despite the common use of trimedlure in control programs, the underlying basis of male attraction remains unknown. In a series of laboratory trials, we compared the mating success of (treated) males exposed to trimedlure and (control) males not given access to trimedlure. When tested immediately after exposure, treated males mated more frequently than control males. This mating advantage was short-lived, however, and males tested as little as 24 h after exposure had no advantage over control males. When tested immediately after exposure, treated males exhibited higher levels of pheromone calling than control males. Consequently, more females were attracted to treated males than to control males in field tests. 相似文献

A hobo-related sequence, Cchobo, with high similarity to the Drosophila melanogaster HFL1 and hobo108 elements was isolated from the medfly. Thirteen PCR-derived clones, which share 97.9–100% DNA identity, were sequenced, seven of which do not show frame-shift or stop codon mutations in their conceptual translations. The consensus sequence has 99.7% DNA identity with the D. melanogaster hobo element HFL1. In a phylogenetic analysis with other hobo-related elements, Cchobo clusters with the HFL1 and hobo108 elements from D. melanogaster and hobo-related elements from D. simulans, D. mauritiana and Mamestra brassicae. These elements may have undergone horizontal transfer in the recent past. The genomic distribution of Cchobo was studied by FISH to mitotic and polytene chromosomes, which revealed that Cchobo is distributed within both the heterochromatin and euchromatin. Intra- and interstrain polymorphisms were detected both at euchromatic and heterochromatic sites. These findings suggest that active copies of the element may be present in the medfly genome. 相似文献

A proteinaceous trypsin inhibitor was purified from Crotalaria pallida seeds by ammonium sulfate precipitation, affinity chromatography on immobilized trypsin-Sepharose and TCA precipitation. The trypsin inhibitor, named CpaTI, had M(r) of 32.5 kDa as determined by SDS-PAGE and was composed of two subunits with 27.7 and 5.6 kDa linked by disulfide bridges. CpaTI was stable at 50 degrees C and lost 40% of activity at 100 degrees C. CpaTI was also stable from pH 2 to 12 at 37 degrees C. CpaTI weakly inhibited chymotrypsin and elastase and its inhibition of papain, a cysteine proteinase, were indicative of its bi-functionality. CpaTI inhibited, in different degrees, digestive enzymes from Spodoptera frugiperda, Alabama argillacea, Plodiainterpunctella, Anthonomus grandis and Zabrotes subfasciatus guts. In vitro and in vivo susceptibility of Callosobruchus maculatus and Ceratitis capitata to CpaTI was evaluated. C. maculatus and C. capitata enzymes were strongly susceptible, 74.4+/-15.8% and 100.0+/-7.3%, respectively, to CpaTI. When CpaTI was added to artificial diets and offered to both insect larvae, the results showed that C. maculatus was more susceptible to CpaTI with an LD(50) of 3.0 and ED(50) of 2.17%. C. capitata larvae were more resistant to CpaTI, in disagreement with the in vitro effects. The larvae were more affected at lower concentrations, causing 27% mortality and 44.4% mass decrease. The Sterile Insect Technique (SIT) is used in many regions worldwide to manage wild populations of the Mediterranean fruit fly (`medfly'), an important pest species. This technique relies on released sterile males outcompeting their wild counterparts in fertilizing ova of wild females. Numerous studies have investigated the ability of sterile males to secure copulations, an essential step toward overall success. Here we progress further along the mating sequence by studying reproductive barriers that may remain ahead of sterile males that manage to secure copulations in field cage experiments and whether ability to pass these barriers is influenced by a male's age, diet and size, or the size of his mate. Amongst those virgin males that succeeded in copulating, both the number of sperm stored by mates and the chances of having any sperm stored at all decreased with age. Sperm tended to be stored asymmetrically between the females' two spermathecae, and this tendency was more apparent when few sperm were stored. In accord with effects of male age on number of sperm stored, sperm of older males were stored more asymmetrically than that of young males. We found no evidence that male size, male diet or female size influenced copula duration, number of sperm stored or allocation of sperm between the female's two spermathecae. The decline in number of sperm stored as males aged was not accompanied by age-dependent changes in copula duration, indicating that copula duration and insemination success are not deterministically linked. We discuss these results in light of their relevance to SIT and the medfly mating system. 相似文献

Summary Using the alcohol dehydrogenase (ADH) locus a genetic sexing system is being developed in the Mediterranean fruit fly Ceratitis capitata based on the sensitivity of ADH null mutations to environmental ethanol. A series of null mutants have been induced at this locus, however, none proved viable as homozygotes. One of these null mutants was translocated to the male determining chromosome and this line can be used for genetic sexing. When larvae from this line were reared on larval medium containing various concentrations of allyl alcohol, 97% of the emerging adults were males; in the absence of the allyl alcohol the sex ratio in the line is distorted in favour of the females. It is proposed that the higher ADH activity of the females (homozygous positive) in comparison with the males (heterozygous null) is responsible for their lower survival in larval medium containing allyl alcohol. ADH converts the allyl alcohol to the lethal ketone. The possible use of this line to sex large populations of medflies for use in sterile insect release programmes is discussed. 相似文献

Summary The viability of a series of recombination suppressor (RS) strains in Ceratitis capitata, all previously found to contain a reciprocal autosomal translocation, was assessed for egg hatchability and adult emergence in both the homozygous and heterozygous state. Except in T 30C, which contains a Y-autosome translocation in addition to the A-A translocation, egg hatch was significantly reduced in all heterozygous translocation strains, and ranged from 42.4% to 58.5% in seeded eggs compared to a control value of 82.8%. Adult emergence from hatched eggs was affected to a lesser extent, but with a range of 59.5% to 84.2%, compared to the control value of 83.1%, remained significantly reduced in 4 of the 6 translocation strains, as well as in the male line of T 30C. In the homozygous configuration all strains, except T 19 and T 109, showed a significant reduction in egg hatchability, whereas adult emergence was not adversely affected. A significant reduction in the egg hatchability of the translocation heterozygotes compared to that of the homozygotes was observed in 5 of the 7 strains, the observed reduction in T 55/109 being non-significant while that of T 30C was significantly increased. The behaviour of translocations as recombination suppressors and their suitability for inclusion in breeding schemes for the isolation of induced recessive mutations is discussed.This work forms part of a Joint FAO/IAEA research programme on the development of genetic sexing mechanisms for the Mediterranean fruit fly, Ceratitis capitata (Wied.) 相似文献

Summary Polytene chromosomes of three genetic sexing strains of Ceratitis capitata were analyzed. The genetic sexing mechanism is based on a pupal color dimorphism (white-brown) and is the result of a reciprocal translocation between the Y chromosome and the autosome bearing the w locus (white pupal case). The analyzed polytene chromosomes were derived from two different pupal tissues, the orbital bristle and fat body cells. The Y chromosome is visible in both tissues, while the autosomes present a different banding pattern. Based on these features, the autosome breakpoints in the three Y; autosome translocations were mapped, and the homology of the translocated autosome in both tissues was established. In addition, the location of the break-points was compared to the stability of these three strains. 相似文献

Wild Mediterranean fruit fly females, Ceratitis capitata (Wiedemann), from an essentially monophagous population on the island of Hawaii were exposed to natural mock orange (Murraya paniculata) or sweet orange (Citrus sinensis) host fruit hung from branches of potted trees for 3-day periods in field enclosures. Subsequently, when flies were released individually onto potted trees harboring one or the other (or a mixture) of these fruit types, a higher proportion visited the type of fruit with which they were familiar (and visitors found familiar fruit faster) compared with the fruit type with which they were unfamiliar. Moreover, fruit-finding flies of this monophagous population attempted oviposition exclusively in the familiar fruit type, and thus appeared to be just as capable of learning to accept fruit for oviposition as wild flies from a previously-tested polyphagous population on the island of Maui. Additional tests were conducted in which flies were exposed to natural or colored-wax-covered mock oranges or sweet oranges and tested for response to colored-wax-covered natural or artificial fruit. Results suggested that fruit size was the principal character learned and used in finding mock orange or sweet orange fruit, while fruit color and odor appeared to be of little or no importance in this regard.
Résumé Des femelles sauvages d'une population essentiellement monophage de C. capitata Wiedemann, provenant de l'île de Hawaï, ont été mises en présence pendant des périodes de 3 jours dans des enceintes dans la nature à des fruits de Murraya paniculata et de Citrus sinensis suspendus à des branches d'arbres empotés. Quand les mouches ont été libérées individuellement sur les arbres empotés portant l'un ou l'autre de ces fruits (ou leur mélange), une plus forte proportion a visité le fruit avec lequel elles étaient familiarisées (et l'ont trouvé plus vite) que le fruit avec lequel elles ne n'étaient pas. De plus, les femelles découvrant des fruits de cette population monophage ne tentèrent de pondre que dans le type de fruit avec lequel elle étaient familiarisées. Elles se montrèrent aussi capables que les mouches d'une population polyphage de l'île de Maui d'apprendre à accepter de nouveaux fruits pour pondre. Des expériences complémentaires ont été réalisées dans lesquelles les mouches étaient mises en présence de fruits de M. paniculata ou de C. sinensis naturels ou couverts de cire colorée ou encore de fruits artificiels. Studies were conducted to determine attraction and feeding propensity of Mediterranean fruit fly, Ceratitis capitata (Wiedemann), to different protein bait mixtures with and without the insecticides malathion, spinosad, and phloxine B. Protein baits were more attractive to females than to males. Protein-starved females responded more than protein-fed females. The type of protein (USB® yeast hydrolysate enzymatic, Mazoferm®E802, Nu-Lure®Insect Bait, or Provesta® 621 autolyzed yeast extract) in the bait had a major influence on C. capitata attraction, which was strongest to fresh Provesta. Aged baits (four day-old) were not as attractive as fresh baits. In feeding propensity studies, highest response was observed for USB protein. On the basis of attraction and feeding responses Provesta (attraction and feeding) and USB (feeding) outperformed the standard Nu-Lure. Protein-starved flies were much more likely to feed on protein compared to protein-fed flies. For protein-starved flies, a mixture of Provesta and malathion repelled fruit flies, compared to a mixture of Provesta and spinosad or phloxine B. This was not the case with protein–fed flies. The wasp Fopius arisanus (Sonan), one of C. capitata's primary natural enemies in Hawaii, would not consume protein baits. Our studies suggest that spinosad or phloxine B, with low contact toxicity, mixed with protein baits offers a more environmentally friendly choice for control of C. capitata and conservation of F. arisanus, whereby the nontarget effects of broad spectrum contact poisons such as malathion can be avoided. Presumably, due to greater selectivity with spinosad and phloxine B bait treatments, the host would be killed, but not the natural enemy. 相似文献

Fopius arisanus (Sonan) and Diachasmimorpha tryoni (Cameron) are two important solitary endoparasitoids of tephritid fruit flies. The former species attacks host eggs while the latter attacks host larvae, and both species emerge as adults from the host puparium. This study investigated intrinsic competition between these two parasitoids, as well as aspects of intraspecific competition within each species in the Mediterranean fruit fly, Ceratitis capitata (Wiedemann). Parasitization by F. arisanus resulted in direct mortality of host eggs and prolonged development of host eggs and larvae. Superparasitism by F. arisanus was uncommon when mean parasitism per host patch was <50%, but increased with rising rates of parasitism. Superparasitism by D. tryoni was more common. In superparasitized hosts, supernumerary individuals of F. arisanus were killed through physiological suppression, while supernumerary larvae of D. tryoni were killed mainly through physical attack. In multiparasitized hosts, dissections showed that 81.6% of D. tryoni eggs in the presence of F. arisanus larvae died within 3 days, indicating physiological inhibition of egg hatch. Rearing results further showed that F. arisanus won almost all competitions against D. tryoni. The ratio of D. tryoni stings to ovipositions was lower in hosts not previously parasitized by F. arisanus than in parasitized hosts, suggesting that D. tryoni can discriminate against parasitized hosts. The mechanism that F. arisanus employs to eliminate D. tryoni is similar to that it uses against all other larval fruit fly parasitoids so far reported. The results are discussed in relation to the competitive superiority of early acting species in fruit fly parasitoids, and to a possible competitive-mediated mechanism underlying host shift by D. tryoni to attack non-target flies following the successful introduction of F. arisanus in Hawaii. 相似文献

The effect of temperatureon conidial germination, mycelial growth, andsusceptibility of adults of three tephritidfruit flies, Ceratitis capitata(Wiedemann), C. fasciventris (Bezzi) andC. cosyra (Walker) to six isolatesof Metarhizium anisopliae were studied inthe laboratory. There were significantdifferences among the isolates in the effect oftemperature on both germination and growth.Over 80% of conidia germinated at 20, 25 and30°C, while between 26 and 67% conidiagerminated at 35°C and less than 10% at15°C within 24 hours. Radial growth was slowat 15°C and 35°C with all of theisolates. The optimum temperature forgermination and mycelial growth was 25°C. Mortality caused by the six fungal isolatesagainst the three fruit fly species varied withtemperature, isolate, and fruit fly species.Fungal isolates were more effective at 25, 30and 35°C than at 20°C. The LT90values decreased with increasing temperature upto the optimum temperature of 30°C. Therewere significant differences in susceptibilitybetween fly species to fungal infection at allthe temperatures tested. 相似文献

Significant differences in propensity to attempt oviposition into various types of fruit were found among Mediterranean fruit flies, Ceratitis capitata, from the same wild population. Evidence presented suggests repeated exposure to one host fruit type conditions females to that host, and influences female response to other hosts in future encounters. Reversibility of conditioning was also demonstrated. Learning to refuse a novel host type is indicated as being more important than learning to accept a familiar host type in medfly conditioning to hosts.
Résumé Dans la même population sauvage de C. capitata, on a observé des différences significatives dans la tendance à essayer de pondre dans divers types de fruits. Les faits rapportés suggèrent qu'un apprentissage pendant la ponte sur un type de fruits conditionne la femelle à cet hôte, et influe sur les réponses de la femelle à d'autres hôtes lors de rencontres ultérieures.相似文献
