Cyanamide is an allelochemical produced by hairy vetch (Vicia villosa Roth.). Its phyotoxic effect on plant growth was examined on roots of onion (Allium cepa L.) bulbs. Water solution of cyanamide (2-10 mM) restricted growth of onion roots in a dose-dependent manner. Treatment of onion roots with cyanamide resulted in a decrease in root growth rate accompanied by a decrease in accumulation of fresh and dry weight. The inhibitory effect of cyanamide was reversed by its removal from the environment, but full recovery was observed only for tissue treated with this chemical at low concentration (2-6 mM). Cytological observations of root tip cells suggest that disturbances in cell division may explain the strong cyanamide allelopathic activity. Moreover, in cyanamide-treated onion the following changes were detected: reduction of mitotic cells, inhibition of proliferation of meristematic cells and cell cycle, and modifications of cytoskeleton arrangement.  相似文献

The aim of this study was to evaluate municipal sludge (MS) for its toxic potential by Allium cepa and also to understand the effect of vermicomposting on the reduction of toxicity, if any. Municipal sludge (MS) and vermicomposted sludge (VS) were evaluated. Elemental analysis of MS showed the presence of heavy metals. Morphological studies of A. cepa roots indicated coiled and wavy roots on exposure to MS but no root abnormality was reported in VS. Under genotoxic studies, inhibition in mitotic index was concentration dependent and the control values of 11.76 gradually reduced to 5.40 at 10% MS leachate whereas mitotic index was increased to 9.48 at 10% VS leachate. Exposure of leachate induced chromosomal aberrations, micronucleus formation and binucleate cells in a dose dependent manner. However, mitotic aberrations were observed significant at 10% MS leachate but they were insignificant at 10% VS leachate. The wet and dry weight of roots, root elongation and chlorophyll contents were reduced as the concentration of leachate increased but VS leachate did not produce considerable reduction. The wet and dry weight of A. cepa roots were 20.312 g and 3.250 g respectively and they were reduced to 10.82 g and 1.68 g respectively at 10% MS leachate but VS leachate showed an increase to 18.127 g and 2.53 g respectively. Total chlorophyll in control, 10% MS leachate and 10% VS leachate were 0.245 g, 0.162 g and 0.214 g respectively. It could be concluded that the MS was toxic to a remarkable extent but vermicomposting of sludge might be beneficial for bioremediation and recommended before land filling.  相似文献

The length of cells of the pericycle, endodermis and middlecortex not actively involved in lateral root primordia (LRP)development was measured in primary roots of Allium cepa, Pisumsativum and Daucus carota. The presence of two cell populationsin the pericycle was demonstrated in all three species. In Alliumcepa and Pisum sativum, pericyclic cells located opposite xylempoles were significantly shorter than cells lying opposite phloempoles. In both species, LRP originated opposite xylem poles.Our results, furthermore, strongly suggest that in regions ofthe root far from the apical meristem, numerous pericyclic cellsundergo transverse division both previous to and during LRPinitiation, decreasing in mean length throughout this period.In Daucus carota, LRP begin to form in pericyclic cells locatednext to the phloem poles, such cells were significantly shorterthan those opposite xylem poles, even in areas of the primaryroot located close to the root tip. Cells also appear to dividetransversely in regions far from the root tip in this species,leading to a conspicuous drop in the mean length of those cellslocated in portions of the pericycle destined to give rise toLRP. Two different cell populations can also be distinguishedin the endodermis of Allium cepa and Pisum sativum, althoughobservations were less conclusive in Daucus carota. Fly ash is a by-product of coal-fired electricity generation plants. The prevalent practice of disposal is as slurry of ash and water to open lands or ash ponds located near power plants and this has lain to waste thousands of hectares all over the world. Wind and leaching are often the causes of off-site contamination from fly ash dumpsites. Vetiver (Vetiveria zizanioides) grown on fly ash for three months showed massive, mesh-like growth of roots which could have a phytostabilizing effect. The plant achieved this without any damage to its nuclear DNA as shown by comet assay done on the root nuclei, which implies the long-term survival of the plant on the remediation site. Also, when Vetiver is used for phytoremediation of coal fly ash, its shoots can be safely grazed by animals as very little of heavy metals in fly ash were found to be translocated to the shoots. These features make planting of Vetiver a practical and environmentally compatible method for restoration of fly ash dumpsites. Lack of DNA damage in Vetiver has been compared to that in a sensitive plant i.e. Allium cepa. Our results suggested that apart from traditional end-points viz. growth parameters like root length, shoot length and dry weight, comet assay could also be included in a battery of tests for initial, rapid and effective selection of plants for restoration and phytoremediation of polluted sites.  相似文献

The hypothesis was tested that the avoidance of acid subsoil by the velvet bean Mucuna pruriens is based on a mechanism acting on the whole root system rather than on individual roots. In a split-root experiment with circulating nutrient solution the growth of plants with Al-containing (+/+) or Al-free (0/0) solution on both sides of the root system was compared with that of plants which had a choice (0/+). Two levels of Al (110 and 185 μ M ) were tested at two levels of Ca (50 and 1250 μ M ). In the 185 μ M Al treatment the concentration of monomeric Al varied between 53 μ M , directly after refreshing the solution, and 5 μ M at harvest time.
An external Al concentration of 110 μ M had no effect on shoot and root dry weight, while 185 μ M Al applied to both sides of the root system (+/+) increased root dry weight and reduced shoot dry weight and shoot/root ratio, compared with the 0/0 control. Application of 185 μ M Al to half of the roots, ied to a significant shift in root growth in favour of the control side; this response is described here as Al avoidance. On the basis of total root length, root dry weight and root surface area, the ratio of 0/+ roots was 3.1, 2.8 and 2.4, respectively.
Al avoidance at 185 μ M Al was confirmed in another experiment, in which root response was measured to a local P source, supplied in a third compartment containing only KH2PO4. A significant increase in root length and dry weight in this compartment was observed, when other roots of the same plant were growing in the presence of Al. This result indicates that Al avoidance by Mucuna roots is related to P preference.  相似文献

The role played by lateral roots and root hairs in promoting plant anchorage, and specifically resistance to vertical uprooting forces has been determined experimentally. Two species were studied, Allium cepa (onion) which has a particularly simple root system and two mutants of Arabidopsis thaliana, one without root hairs (rhd 2-1) and another with reduced lateral root branching (axr 4-2). Maximum strength of individual onion roots within a plant increased with plant age. In uprooting tests on onion seedlings, resistance to uprooting could be resolved into a series of events associated with the breakage of individual roots. Peak pulling resistance was explained in a regression model by a combination of a measure of plant size and the extent to which the uprooting resistance of individual roots was additive. This additive effect is termed root co-operation. A simple model is presented to demonstrate the role played by root co-operation in uprooting resistance. In similar uprooting tests on Arabidopsis thaliana, the mutant axr 4-2, with very restricted lateral development, showed a 14% reduction in peak pulling resistance when compared with the wild-type plants of similar shoot dry weight. The uprooting force trace of axr 4-2 was different to that of the wild type, and the main axis was a more significant contributor to anchorage than in the wild type. By contrast, the root hair-deficient mutant rhd 2-1 showed no difference in peak pulling resistance compared with the wild type, suggesting that root hairs do not normally play a role in uprooting resistance. The results show that lateral roots play an important role in anchorage, and that co-operation between roots may be the most significant factor.  相似文献

The effects of cadmium chloride concentration on root, bulb and shoot growth of garlic (Allium sativum L.), and the uptake and accumulation of Cd2+ by garlic roots, bulbs and shoots were investigated. The range of cadmium chloride (CdCl2 x 2.5H2O) concentrations was 10(-6) - 10(-2) M. Cadmium stimulated root length at lower concentrations (10(-6) - 10(-5) M) significantly (P < 0.005) during the entire treatment period. The seedlings exposed to 10(-3) - 10(-2) M Cd exhibited substantial growth reduction (P < 0.005), but did not develop chlorosis. Garlic has considerable ability to remove Cd from solutions and accumulate it. The Cd content in roots of garlic increased with increasing solution concentration of Cd2+. The roots in plants exposed to 10(-2) M Cd accumulated a large amount of Cd. approximately 1,826 times the control. The Cd contents in roots of plants treated with 10(-3), 10(-4), 10(-5) and 10(-6) M Cd were approximately 114, 59, 24 and 4 times the control, respectively. However, the plants transported only a small amount of Cd to their bulbs and shoots and concentrations in these tissues were low.  相似文献

We have purified a novel alliinase (EC 4.4.1.4) from roots of onion (Allium cepa L.). Two isoforms with alliinase activity (I and II) were separated by concanavalin A-Sepharose and had molecular masses of 52.7 (I) and 50.5 (II) kD on sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and 51 (I) and 57.5 (II) kD by gel filtration fast-protein liquid chromatography. Isoform I had an isoelectric point of 9.3, while isoform II had isoelectric points of 7.6, 7.9, 8.1, and 8.3. The isoforms differed in their glycosylation. Both contained xylose/fucose containing complex-type N-linked glycans, and isoform II also contained terminal mannose structures. Both isoforms had activity with S-alk(en)yl-L-cysteine sulfoxides. Unlike other allium alliinases, A. cepa root isoforms had cystine lyase activity. We cloned a gene from A. cepa root cDNA and show that it codes for A. cepa root alliinase protein. Homology to other reported allium alliinase genes is 50%. The gene coded for a protein of mass 51.2 kD, with two regions of deduced amino acid sequence identical to a 25- and a 40-amino acid region, as determined experimentally. The A. cepa root alliinase cDNA was expressed mainly in A. cepa roots. The structure and function of the alliinase gene family is discussed.  相似文献

Greenhouse experiments were conducted to evaluate the effect of Nod factor Nod Bj-V (C18:1, MeFuc) of Badyrhizobium japonicum on the growth of soybean and corn. Three-day-old seedlings of soybean and corn were grown in hydroponic solutions containing four concentrations (0, 10(-7), 10(-9) or 10(-11) M) of Nod factor. After 7 d of treatment, Nod factor enhanced soybean and corn biomass. Nod factor elicited profound effects on root growth resulting in 34-44% longer roots in soybean. More detailed analyses of the roots, using a scanner based image analysis system, revealed that Nod factor increased the total length, projected area and surface area of the roots and decreased the diameter of soybean roots, while it increased the total length of corn roots. Stem injection of soybean plants with 10(-7) M Nod factor resulted in increased dry matter accumulation. These results suggest that Nod factor, besides mediating early stages of nodulation, has more general plant growth-promoting effects.  相似文献

Inorganic polyphosphate (poly P) has been considered to be a translocatable form of phosphate (Pi) in arbuscular mycorrhizal fungi (AMF). Here we examined time-course changes in poly P content during the AMF colonization process. Onion (Allium cepa) plants were cultured with or without inoculation with Gigaspora margarita for 2-8 wk with periodic sampling. Poly P in the extracts, purified through gel filtration, was quantified by the reverse reaction of polyphosphate kinase. The length of poly P in mycorrhizal roots appeared to be shorter than in extraradical hyphae or in spores of the AMF, indicating that AMF depolymerize poly P before providing Pi to the host. The poly P content increased as colonization proceeded, and was highly correlated with the weight of the colonized roots. These results support the model that AMF supply Pi to the host through the poly P pool, and that the poly P content of a mycorrhizal root can be a good indicator of the Pi-supplying activity of AMF.  相似文献

Oxidative stress-mediated genotoxicity of wastewaters taken from two different cities, Saharanpur (SWW) and Aligarh (AWW), were compared with a battery of short-term assays namely the Allium cepa genotoxicity test, the plasmid-nicking assay, and the Ames fluctuation test. Both test-water samples - when used undiluted - increased the frequency of chromosomal abnormalities and/or micronuclei and alterations in the mitotic index of root cells of Allium cepa. Bridges and fragmentation of the chromosome were the predominant effects of the Saharanpur water sample while the Aligarh sample induced mainly chromosome fragmentation. Single- and double-strand breaks were also observed in plasmid DNA treated with these test wastewaters. The plasmid-nicking assay performed on SWW resulted in linearization of plasmid DNA when 18μl was tested (in a total reaction volume of 20μl). However, with the same amount of AWW, all three forms of plasmid, viz. supercoiled, open circular and linear were observed. Supplementation with specific scavengers of reactive oxygen species (ROS) caused a significant decline in mutagenicity of test-water samples in all the tests, pointing at oxidative stress as the mediator of the observed genotoxicity. The role of heavy metals in the AWW-induced oxidative stress and that of phenolics in SWW cannot be ruled out.  相似文献

In the present study, we applied Chromosome Aberration (CA) and Micronucleus (MN) tests to Allium cepa root cells, in order to evaluate the water quality of Guaecá river. This river, located in the city of S?o Sebasti?o, SP, Brazil, had been affected by an oil pipeline leak. Chemical analyses of Total Petroleum Hydrocarbons (TPHs) and Polycyclic Aromatic Hydrocarbons (PAHs) were also carried out in water samples, collected in July 2005 (dry season) and February 2006 (rainy season) in 4 different river sites. The largest CA and MN incidence in the meristematic cells of A. cepa was observed after exposure to water sample collected during the dry season, at the spring of the river, where the oil leak has arisen. The F(1) cells from roots exposed to such sample (non-merismatic region) were also analyzed for the incidence of MN, showing a larger frequency of irregularities, indicating a possible development of CA into MN. Lastly, our study reveals a direct correlation between water chemical analyses (contamination by TPHs and PAHs) and both genotoxic and mutagenic effects observed in exposed A. cepa cells.  相似文献

Root development was studied in winter wheat ( Triticum aestivum L. cv Starke II) grown at 5,10, 15 and 20°C in nutrient solutions with phosphate concentrations of 10, 100 or 1000 μM . The plants were grown for 38 days (5 and 10°C), 19 days (15°C) or 14 days (20°C). At the end of the cultivation period the phosphate influx in the roots was determined with 32P-phosphate. Root development (lateral and seminal roof length and number) was monitored throughout the cultivation period on the same individuals by repeated (approximately every second day) photocopying of the roots for measurements with digitizer and appropriate software. The 5°C treatment yielded no laterals, and the seminals were only slightly affected by the different phosphate treatments. The 10 μM phosphate treatment gave high root:shoot dry weight ratio, high average lateral root length and high specific root length [m root (g root fresh weight)-1]. The 1000 μM phosphate treatment yielded the highest number of laterals per m seminal root, and usually also the highest absolute numbers. Phosphate influx decreased with increased P status of the roots. It is argued that phosphate influx is dependent on factors such as P status, root geometry and relative root extension rate.  相似文献

Acidification of natural waters and soils leads to the release of aluminium, causing damage to living organisms. In the present experiments, growing roots of Allium cepa L. were exposed to water solution of AICl3 in concentrations from 10-5 to 10-1 M . After a few days in concentrations around 10-3 M . the roots showed macroscopical signs of being affected; they became bent and their terminal portions tended to loosen and drop off. Microscopically, the affected zone of the roots revealed a new cellular phenomenon: Nucleolar material was extruded from the nuclei into the cytoplasm forming elongated, rod-like bodies, which eventually divided into two, one distinct body at each pole of the cell. The derivation of these bodies from the nucleoli was further strengthened by the observation that they absorbed light green differentially Apparently, aluminium has a specific effect on the nucleoli of certain root cells.  相似文献

Monosomic alien addition lines (MAALs) are useful for assigning linkage groups to chromosomes. We examined whether the chromosomal rearrangements following the introduction of a single onion (Allium cepa) chromosome into the Allium fistulosum genome were produced by homeologous crossing over or by a nonreciprocal conversion event. Among the monosomic lines available, 17 were studied by fluorescent genomic in situ hybridisation, using total A. cepa genomic DNA as the probe and total A. fistulosum genomic DNA as the competitor. In this way, rearrangements such as chromosomal translocations between A. cepa and A. fistulosum were identified as terminal regions consisting of tandem DNA repeats. Homeologous crossing over between the two closely related genomes occurred in 4 of the 17 lines, suggesting that such events are not rare. On the basis of a detailed molecular cytogenetic characterisation, we identified true monosomic alien addition lines for A. cepa chromosomes 3, 4, 5, 7, and 8 that can reliably be used in genetic studies.  相似文献

The ontogeny of carrots (Daucus carota cv. ''Spartan Premium'') grown under greenhouse conditions in pots of organic soil infected with Meloidogyne hapla was influenced detrimentally as early as 4 days after seeding, as determined through analysis of plant surface area, dry weight, fresh weight, net assimilation rate, relative growth rate, and leaf-area ratio. Only 58% of the diseased carrots were suitable for fresh market, compared with 97% of those grown in nematode-free soil. Growth and development of the shoot system (height, surface area, dry weight, and fresh weight) were retarded by M. hapla as early as 12 days after seeding. During the first 12 days after seeding, root dry weight was greater for diseased plants than for controls. Root growth and development (surface area, dry weight, and fresh weight) associated with this nematode, however, were retarded as early as 16 days after seeding. M. hapla caused a delay in the occurrence of 2nd-, 4th-, and 5th-order roots, and an increase in the occurrence of 6th-order roots in infected plants. Parasitized plants had 44% fewer roots (primary through 6th-order) and 50% less total root length.  相似文献

Plasmodesmatal frequencies (PFs) were analysed in Allium cepa L. roots with a mature exodermis (100 mm from the tip). For all interfaces within the root, the numbers of plasmodesmata (PD) microm(-2) wall surface (Fw) were calculated from measurements of 60 walls on ultrathin sections. For tissues ranging from the epidermis up to the stelar parenchyma, the frequencies were also expressed as total PD numbers mm(-1) root length (Fn), which is most instructive for considering the radial transport of ions and photosynthates (because the tissues were arranged in concentric cylinders). The Fn values were constantly high at the interfaces of exodermis-central cortex, central cortex-endodermis and endodermis-pericycle (4.05x10(5), 5.13x10(5), and 5.64x10(5), respectively). If the plasmodesmata are functional, a considerable symplastic transport pathway exists between the exodermis and pericycle. Two interfaces had especially low PFs: epidermis-exodermis (Fn=8.96x10(4)) and pericycle-stelar parenchyma (Fn=6.44x10(4)). This suggests that there is significant membrane transport across the interface of epidermis-exodermis (through short cells) and direct transfer of ions from pericycle to protoxylem vessels. In the phloem, the highest PF was detected at the metaphloem sieve element-companion cell interface (Fw=0.42), and all other interfaces had much lower PFs (around 0.10). In the pericycle, the radial walls had a high PF (Fw=0.75), a feature that could permit lateral circulation of solutes, thus facilitating ion (inward) and photosynthate (outward) delivery.  相似文献

The present study investigated the effect of ferulic acid (FA; 0–1000 µM) on early growth, and rhizogenesis in mung bean (Vigna radiata) hypocotyls and associated biochemical changes. FA severely affected the radicle elongation and number of secondary roots after 72 h. The root and shoot length, number and length of secondary roots, and seedling dry weight of one-week-old seedlings of mung bean were decreased by 64%. The rooting potential (percent rooting, number and length of adventitious roots) of mung bean hypocotyls under in vitro conditions was significantly inhibited in response to 1–100 µM FA. At 1000 µM there was complete cessation of rooting. FA caused a reduction in the contents of water-soluble proteins and endogenous total phenolics, whereas the activities of proteases, peroxidases, and polyphenol peroxidases increased. The study concludes that FA inhibits root growth and development, and in vitro rooting process in mung bean by interfering with biochemical processes that are crucial for root formation.  相似文献

The pathway of Ca(2+) movement from the soil solution into the stele of the root is not known with certainty despite a considerable body of literature on the subject. Does this ion cross an intact, mature exodermis and endodermis? If so, is its movement through these layers primarily apoplastic or symplastic? These questions were addressed using onion (Allium cepa) adventitious roots lacking laterals. Radioactive Ca(2+) applied to the root tip was not transported to the remainder of the plant, indicating that this ion cannot be supplied to the shoot through this region where the exodermis and endodermis are immature. A more mature zone, in which the endodermal Casparian band was present, delivered 2.67 nmol of Ca(2+) mm(-1) treated root length d(-1) to the transpiration stream, demonstrating that the ion had moved through an intact endodermis. Farther from the root tip, a third zone in which Casparian bands were present in the exodermis as well as the endodermis delivered 0.87 nmol Ca(2+) mm(-1) root length d(-1) to the transpiration stream, proving that the ion had moved through an unbroken exodermis. Compartmental elution analyses indicated that Ca(2+) had not diffused through the Casparian bands of the exodermis, and inhibitor studies using La(3+) and vanadate (VO(4)(3-)) pointed to a major involvement of the symplast in the radial transport of Ca(2+) through the endodermis. It was concluded that in onion roots, the radial movement of Ca(2+) through the exodermis and endodermis is primarily symplastic.  相似文献
