The lactate dehydrogenase activity in reactions of lactate oxidation and synthesis was studied in subfractions of the chicken brain, heart and liver at the embryonal, early postembryonal and adult stages of development after thyroxine administration. It has been shown that during embryogenesis thyroxine predominantly enhanced the rate of lactate oxidation in the mitochondrial tissues. A marked increase in the lactate synthesis was found in cytoplasm of the adult chicken tissues. Specificity of enzyme activity alterations was detected in the chicken brain during ontogenesis after thyroxine administration.  相似文献

We studied properties of lactate dehydrogenase isozymes expressed at different stages of Danio rerio ontogenesis. H4-LDH and a minor fraction H3M1 are expressed during embryonic development. The muscle isozyme (M4) appears after the beginning of muscle contractions in the embryo. H4 and M4 isozymes isolated from the heart and skeletal muscle of the adult fish, respectively, show significant differences in terms of Km, activation energy (AE), and inactivation temperature. H4-LDH isozymes isolated from unfertilized eggs, the skeletal muscle of larvae, and the heart of the adult fish differ in Km and activation energy, as well as in inactivation temperature. We propose that these differences may be associated with a ligand interacting with the H4 isozyme at different steps of ontogenesis.  相似文献

Activities of the digestive enzymes (lactase, maltase, saccharose, di-, tri- and aminopeptidases, alkaline and acid phosphatase) in the gastrointestinal tract, the kidney and the liver in one-day-old and adult rats as well as in 10-day-old rats after injections of hydrocortisone (H) or thyroxine (T4) were determined. On the basis of the results obtained it is summarized that (1) high levels of the digestive enzyme activities are observed in the nondigestive organs in immature and adult rats; (2) H induces the enzyme activities in the small intestine more than in other organs; T4 does not influence the activities of the most of enzymes studied both in the digestive and in the nondigestive organs; (3) high activities of a number of enzymes in the colon in one-day-old rats implies its involvement in the digestion at early stages of the ontogenesis.  相似文献

Adaptive specific features of energy metabolism in fish ontogenesis   总被引:1,自引:0,他引:1  
A review of data on the pattern of change of the intensity of oxygen consumption during early ontogenesis of different fish species (rainbow trout, loach, zebrafish, carp, and grass carp) is provided. It has a similar pattern: this index increases in the period of embryonic and larval development and, after passing of larvae to an active feeding, it begins to gradually decline. This dynamics is determined by specific features of an increase in the rate of oxygen uptake and body weight in the course of early stages of fish ontogenesis. For determining optimal temperature conditions of development, a method of total (for a definite stage of development) oxygen uptake was suggested, which makes it possible to determine minimal energy expenditures necessary for the process of a particular stage of embryogenesis to take place. Analysis of temperature dependence of kinetic properties of enzymes with reference to the Michaelis constant (Km) for lactate dehydrogenase demonstrated that minimal Km, corresponding to maximal enzyme-substrate affinity, for embryos of different fish species differs in correspondence with differences in temperature conditions of development of these species in nature. For embryos of one species developing at changing temperature conditions (salmonids), this index changes in accordance with a temperature drift in nature.  相似文献

The evolution of endothermy is one of the most significant events in vertebrate evolution. Adult mammals and birds are delineated from their early ontogenetic stages, as well as from other vertebrates, by high resting metabolic rates and consequent internal heat production. We used the embryonic development of a bird (Gallus gallus) as a model to investigate the metabolic transition between ectothermy and endothermy. Increases in aerobic capacity occur at two functional levels that are regulated independently from each other: (i) upregulation of gene expression; and (ii) significant increases in the catalytic activity of the main oxidative control enzymes. Anaerobic capacity, measured as lactate dehydrogenase activity, is extremely high during early development, but diminishes at the same time as aerobic capacity increases. Changes in lactate dehydrogenase activity are independent from its gene expression. The regulatory mechanisms that lead to endothermic metabolic capacity are similar to those of ectotherms in their response to environmental change. We suggest that the phylogenetic occurrence of endothermy is restricted by its limited selective advantages rather than by evolutionary innovation.  相似文献

The effect of various activity regimes on metabolism of pigeon pectoralis was examined by measurement of blood lactate following exercise, total lactate dehydrogenase activity of pectoral muscle, and proportions of specific isoenzymes of pectoral muscle lactate dehydrogenase. Sprint-trained birds had the highest pectoral muscle lactate dehydrogenase activity (1409 IU · g−1 wet tissue), while endurance-trained birds had the highest peak lactate levels (287 mg · dl−1, extrapolated from decay curves) and fastest half-time of the lactate response (4.8 min) following exercise, but the lowest lactate dehydrogenase activity (115 IU · g−1 wet tissue). Immobilization of one wing for 3 weeks following endurance training produced a marked increase in lactate dehydrogenase activity of the immobilized muscle, compared to that in the contralateral pectoralis and endurance-trained muscle. Aerobic forms of the lactate dehydrogenase enzyme (that favor conversion of lactate to pyruvate) predominated in pectoral muscle of endurance-trained birds, while cage-confined birds exhibited primarily the anaerobic isoenzymes. These results demonstrate that conversion of pectoral muscle lactate dehydrogenase isoenzymes, total lactate dehydrogenase activity, and half-time of lactate response after exercise is dependent on activity regime in pigeons. In this respect, pigeon pectoral muscle responds to training and disuse in a manner similar to that of mammalian skeletal muscle. Accepted: 10 September 1996  相似文献

The activity of the enzymes of glycolysis (phosphofructokinase, aldolase, pyruvate kinase, lactate dehydrogenase) and hexose monophosphate shunt (glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase) was determined in the eye tissues of the rabbit at different stages of ontogenesis. The activity of these enzymes in the retina was shown to be higher than in other eye tissues. In the uveal tract (iris, ciliary bodies, uvea) the activity of glycolytic enzymes changes with the age. The greatest changes in the activity of enzymes were found during the period of the opening of eyelids. The activity of the enzymes of hexose monophosphate shunt in the eye tissues increases with the age. The relative activity of dehydrogenases of the hexose monophosphate shunt after the establishment of visual function is, however, not high and does not exceed that of phosphofructokinase and pyruvate kinase in the eye tissues of the rabbit.  相似文献

The paper considers the problem of peculiarities of maturation of the stellate ganglion nerve elements in mammals of different species. This process differs in precocious and altricial animals. It has been shown that in spite of some individual peculiarities, the neurons, fibers, and conducting pathways in altricial animals are not, on the whole, completely formed morphologically and functionally. In the course of postnatal ontogenesis, not only an increase of cell sizes and development of dendrite tree, but also reorganization of nerve connections with target organ occur. The postnatal ontogenesis is also accompanied by an increase of the excitation transmission rate along the fibers and by their myelination. The asymmetry of the right and left stellate ganglia (SG) by their sizes and functional peculiarities, which exists in adult animals appears as soon as at early stages of postnatal development. The neural elements of precocious animals are changed to a lesser extent in postnatal ontogenesis and are, in many aspects, similar to those of adult organisms as early as at birth.  相似文献

The development of activity of lactate dehydrogenase was investigated by histochemical methods under the light and electron microscopes and also by biochemical methods. The studies were carried out in the areas of the nigro-striatal pathway of 7 foetal groups, two days-old piglets, three weeks old piglets and adult pigs. It was demonstrated that the activity of LDH appears early in the prenatal ontogenesis. High activity of the enzyme appears in the studied areas of the brain already in foetuses about 60 days old (100 mm) afterwards it rises and the highest activity is observed in about 86 days old foetuses. It then decreases just before birth. After birth in two days and three weeks old piglets the LDH activity decreases further. In adults a high activity of the examined enzyme was confirmed.  相似文献

Studies were conducted to analyze the effect of the thyroid hormone on ventricular myosin during ontogenesis of mice, rats and rabbits. Hypothyroidism was induced in mice and rats by administering propylthiouracyl in drinking water. Rabbits were made hyperthyroid by chronic administration of thyroxine. The change in the thyroid state of rats and rabbits influenced young and adult animals differently depending on whether V1 or V3 was the major ventricular isomyosin form present. Measurements of Ca2+-ATPase activity of myosins from young and old control animals and from animals with changed thyroid state showed that hypothyroidism in rats is associated with a greater decrease of myosin ATPase in young rats which contain V1 isomyosin only, when compared with old rats which contain a preponderance of V3 isomyosin and less of the V1 form. In rabbits, ATPase activity of ventricular myosin was more elevated after thyroxine administration in adult rabbits, which contain V3 isomyosin only, than in young rabbits in which myosin consists of V1 and V3 isomyosins. Ventricular myosins of young and adult mice did not differ in their ATPase activity and the treatment of mice with propylthiouracyl had only slight effect on myosin ATPase. It can be concluded based on these results that the hypothesis concerning hypothyroidism inducing transformation of V1 into V3 isomyosin does not hold generally.  相似文献

It was shown that the glial fibrillary acidic protein (GFAP) content in developing (fetal) human brain is sharply increased. The expression of GFAP was observed already on the 7th-8th week after gestation, the GFAP concentration being less than 0.05% in comparison with adult brain. GFAP can be immunohistochemically detected in radial glial cells. At early stages of development the presence of antigenic determinants of 68 kDa and 100 kDa polypeptides interacting with monoclonal antibodies alongside with native GFAP (51 kDa) and its low molecular weight forms was demonstrated. These antigenic determinants cannot be detected at later stages of development and are absent in adult brain. The data obtained testify to changes in the gene expression of intermediate filament proteins at early stages of human brain ontogenesis.  相似文献

Study of peculiarities of formation of preference in use of one of forepaws at learning of the manipulatory food-procuring skill has been carried out at different stages of early ontogenesis of white rats. It was established that as early as at the 3-week age the rat pups were already able to learn getting food from a narrow test-tube; however, no expressed preference in using one of the extremities was revealed. After repeated testing of adult rats at the age of 4 months the number of ambidexters was significantly higher than in the control animal group. At learning of the 30-day old rat pups of this manipulatory skill, in the majority of the rat pups, no significant lateralization also was found in using the forepaw. At repeated testing of adult rats the amount of ambidexters did not differ from that in the control group of animals. The obtained results indicate the existence of critical periods in formation of lateral preference in performance of the manipulatory food-procuring skill. There also was shown effect of learning in early ontogenesis of the capability for lateralization of this skill in the adult state in rats.  相似文献

The work presents data on reproductive and protective behavior of non-copulative female crickets Gryllus argentinus Sauss. at the beginning of adult ontogenesis. There is studied character of interaction of females with "virtual" males (the method of "trackball". The forms of female motor reactions are described and characterized in the model of male intraspecies signals--the call and aggressive ones. The character of development of female acoustic behavior is described by a change in the motor reaction forms in the course of adult ontogenesis. It is shown that at once after moulting the reproductive behavior on adults has not yet been formed, but is maturing gradually for several days. The passive protective behavior was established to be the most expressed at the initial stages of adult ontogenesis.  相似文献

Total protein content in blood serum and different lymphoid organs, such as bursa, spleen and thymus was investigated in chickens at two different circadian stages (0800 or 1600 hrs; early or late photophase) following administration of either saline or hormones (thyroxine or hydrocortisone or epinephrine). The results suggest that the lymphoid organs may respond differently to the exogenous administration of different hormones depending on the time of their administration.  相似文献

Functional relationship between wave form of complex spike (CS) and depression time of simple spike (SS) in discharge of cerebellar Purkinje cells was studied after their activation with afferent climbing fiber at different terms of postnatal ontogenesis in norm and after treatment with harmaline. The experiments were carried out on three age groups of Wistar rats: rat pups (2 weeks), the adult (4–6 months), and the old animals (22–26 months). It was established that the SS duration in norm was approximately equal in rat pups, adult, and old animals, whereas it markedly decreased form the young to the old animals during the SS depression in the Purkinje cell discharge. Frequency of small action potential (lAP) and their number in the Purkinje cell discharge were approximately equal in young rat pups and adult animals, while in old animals these parameters were higher, on average, by 30%. After administration of harmaline, all CS parameters in rat pups and old animals increased in parallel with the depression time elongation. In adult rats, harmaline did not produce statistically significant changes of the mean values of CS parameters, but an increase of the simple spike depression time was observed. The obtained results allow concluding that the SS wave form and the simple spike depression time in norm are functionally coupled and change with age. The effect of harmaline on the CS wave forms as well as on interrelation of the CS duration and the CS depression time in the Purkinje cell discharge was more pronounced at the early and the late stages of Wistar rat postnatal ontogenesis.  相似文献

This study demonstrates that under subtropical environmental conditions the circulating levels of triiodothyronine (T 3 ) and thyroxine (T 4 ) in adult male roseringed parakeets undergo parallel changes according to a diurnal cycle with a trough in the early morning and a peak in the evening. Chronic afternoon administration of melatonin (25 µg/ 100 g body wt./ day for 30 consecutive days) abolishes time-bound changes in the concentrations of T 3 by suppressing its evening peak, but does not abolish the diurnal rhythm of T 4 . Exogenous melatonin, however, causes a significant increase in the evening value of serum T 4 and a decrease in its value at midnight resulting in phase-shifting of the diurnal fluctuations of T 4 compared to that in the control birds. It appears that exogenous melatonin can alter the diurnal profiles of serum thyroid hormones in a wild psittacine bird.  相似文献

The role of sex steroids in the programming of the level of serum corticosteroid binding globulin (CBG) of male and female rats has been studied at different stages of ontogenesis. It was shown that castration of adult males lead to the increase of the level of CBG, but not to the elimination of sex differences. Gonadectomy of males up to 28th day of postnatal life results in complete feminization of the CBG content in these animals at the age of 10-12 weeks. The castration after 35th day of life does not prevent the formation of the male phenotype of CBG content. The results of administration of testosterone-propionate (TP) to castrated males at different periods of ontogenesis suggests that the sensitivity to irreversible negative action of androgens appears after 28th day of life and disappears after the puberty. It was concluded that short period of ontogenesis from 29th to 35th days of life is critical for the realization of the irreversible masculinization of CBG level upon the influence of androgens in the physiological conditions. It was found that injections of both synthetic estrogens diethylstilbestrol or TP in the sensitive period of ontogenesis lead to the expression of male phenotype of CBG level in a similar fashion.  相似文献

Chicken liver mitochondria were isolated in relatively pure form as indicated by electron microscopy and marker enzyme assay. The rate of respiration, respiratory control index and ADP/O ratios with several different substrates indicated that chicken liver mitochondria are more uncoupled than rat liver mitochondria. Chickens have ten-fold higher malate concentrations in liver than do rats, 2-oxoglutarate was also more abundant in chicken livers. Fasted birds had a five-fold increase in beta-hydroxybutyrate as compared with fed birds; whereas malate and lactate concentrations decreased. Fasted birds had increased levels of isocitrate dehydrogenase (NADP dependent) and lactate dehydrogenase in the cytosol, and increased malate dehydrogenase (NAD dependent), isocitrate dehydrogenase (NADP dependent) and malic enzyme activities in the mitochondria.  相似文献
