In a culture of Alocasia indica (2n=28), one plant was found to be desynaptic. At pachytene, pairing apparently was complete, but at diakinesis and metaphase I on the average only 2.36 and 1.53 bivalents respectively were found. Desynapsis is of the medium-strong type according to Prakken. Later meiotic stages were irregular and pollen sterility was 95%. The origin must have been spontaneous mutation.  相似文献

An efficient, simple micropropagation method was developed for Alocasia amazonica using corms in semisolid and liquid cultures. Explants were cultured onto Murashige and Skoog (MS) medium (Murashige and Skoog, Physiol. Plant. 15:473–497, 1962) supplemented with different cytokinins (Benzyladenine [BA, 2.22–13.32 μM], kinetin [2.32–13.95 μM], Thidiazuron [TDZ, 0.45–4.54 μM]) and cytokinin in combination with auxins [naphthalene acetic acid (NAA, 0.54–5.37 μM)/indole acetic acid (IAA, 0.57–5.71 μM)/indole butyric acid (IBA, 0.49–4.9 μM)]. All supplementary-induced shoot proliferation and the optimal results was on the medium supplemented with 2.27 μM TDZ, which induced 5.1 shoots per explant. Among the different concentrations of sucrose (0–120 g l−1) tested for shoot proliferation, 30 g l−1 was found suitable for corm cultures of Alocasia amazonica. The optimal shoot proliferation and biomass values were with the plantlets grown at 30 μmol m−2 s−1 photosynthetic photon flux (PPF) and 25°C. Liquid cultures found suitable for shoot proliferation and biomass accumulation was compared to semisolid cultures. Comparative studies of bioreactor systems [continuous immersion (with or without net) and temporary immersion in liquid media using ebb and flood] revealed that shoot multiplication and growth were greatest with the raft bioreactor system. Plantlets (cormlets) from the bioreactor were hydroponically cultured for 30 days, and 100% of plants were acclimatized successfully. The simple efficient method of production of plantlets (cormlets) is useful for large-scale multiplication of this important ornamental plant. An erratum to this article can be found at  相似文献

Plantlets of Alocasia amazonica regenerated under a photon flux density (PFD) of 15 or 30 μmol m−2 s−1 showed better growth and development than those grown under higher PFDs. While chlorophyll a and chlorophyll b decreased, the number of stomata increased with increasing PFD. Photoperiods also affected plantlet growth and stomatal development. Highest growth was observed for the short photoperiod (8/16 h) and for equinoctial (12/12 h) light and dark periods. Very few stomata developed in the leaves of plantlets grown under a short photoperiod (8/16 h) and the number of stomata increased with increasing light period. In conclusion, both light intensity and photoperiod independently affect growth of A. amazonica and development of stomata, depending on the intensity and duration of light treatment.  相似文献

High concentrations of heavy metals and organic pollutants in municipal sewage sludge are key factors limiting its use in agriculture. The objectives of this study were to decrease the heavy metal and polycyclic aromatic hydrocarbon concentrations in sewage sludge by phytotreatment and to determine, in a field experiment, whether co-planting is more effective than using a mono-crop of Sedum alfredii. Four treatments were used in the plot experiment: no sludge, no plants, S. alfredii and co-planting S. alfredii and Alocasia marorrhiza. The results showed that co-planting produced tubers and shoots of A. marorrhiza that were suitable as a safe animal feed and good organic K fertilizer, respectively. Co-planting was more effective than mono-planting at reducing concentrations of total Zn and diethylenetriaminepentaacetic acid (DTPA)-extractable Zn, Cd, and Cu in the sludge. Co-planting decreased the concentrations of DTPA-extractable heavy metals and benzo[a]pyrene (B[a]P) in the sludge significantly compared with the unplanted sludge. Decreases of 87, 75, 85, 31, and 64% were obtained for B[a]P and DTPA-extractable Zn, Cd, Cu, and Pb, respectively, compared with the fresh sludge. These results indicate that co-planting can reduce significantly the environmental risks associated with heavy metals and B[a]P in sewage sludge for further disposal.  相似文献

Callus induction from petiole explants has been achieved in Alocasia micholitziana `Green Velvet'. The highest percentage (71%) of explants inducing callus was obtained on MS medium supplemented with 0.5 M 2,4-D and 0.5 M kinetin in the dark after 4 months of culture. Shoots were regenerated at the highest frequency of 33.3% under light condition when 0.5 M BA was added to MS medium with the average of 7.8±2.3 shoots per callus explant. The callus-derived shoots rooted on hormone free MS medium and within 4 weeks the plantlets were ready for acclimatization. The regenerated plants appeared morphologically similar to mother plants.  相似文献

A whole-plant carbon balance model incorporating a light acclimation response was developed for Alocasia macrorrhiza based on empirical data and the current understanding of light acclimation in this species. The model was used to predict the relative growth rate (RGR) for plants that acclimated to photon flux density (PFD) by changing their leaf type, and for plants that produced only sun or shade leaves regardless of PFD. The predicted RGR was substantially higher for plants with shade leaves than for those with sun leaves at low PFD. However, the predicted RGR was not higher, and in fact was slightly lower, for plants with sun leaves than for those with shade leaves at high PFD. The decreased leaf area ratios (LARs) of the plants with sun leaves counteracted their higher photosynthetic capacities per unit leaf area (Amax). The model was manipulated by changing parameters to examine the sensitivity of RGR to variation in single factors. Overall, RGR was most sensitive to LAR and showed relatively little sensitivity to variation in Amax or maintenance respiration. Similarly, RGR was relatively insensitive to increases in leaf life-span beyond those observed. Respiration affected RGR only at low PFD, whereas Amax was moderately important only at high PFD.  相似文献

  • 1 Two closely related species of Colocasiomyia alocasiae (Okada) and C.xenalocasiae (Okada) (Diptera, Drosophilidae) breed in inflorescences of Alocasia odora C. Koch (Araceae), a hermaphroditic understorey clonal herb.
  • 2 The two drosophilid species form a synhospitalic pair in Okinawa with alocasiae breeding in the upper half of the inflorescence and xenalocasiae breeding in the lower half.
  • 3 C.alocasiae also has the following combination of life history traits: small body size, many eggs, and early reproductive maturity. In contrast, xenalocasiae can be described as having larger body size, fewer eggs, and delayed reproductive maturity.
  • 4 Resource partitioning between the two species on the same host may be affected by these life history traits which are associated with their larval habitats.

Summary Photosynthetic capacities and respiration rates of Alocasia macrorrhiza leaves were measured for 4 weeks following reciprocal transfers between high (20% of full sun) and low (1% of full sun) light environments. Photosynthetic capacities and respiration rates of mature, high-light leaves were 1.7 and 4.5 times those of low-light leaves, respectively. Following transfer, respiration rates adjusted within 1 week to those characteristic of plants grown in the new environment. By contrast, photosynthetic capacities either did not adjust or changed only slowly following transfer. Most of the difference in respiration between high- and low-light leaves was related to the carbohydrate status as determined by the daily PFD and little was directly related to the maintenance costs of the photosynthetic apparatus. Leaf construction cost was directly proportional to maximum photosynthetic capacity. Consequently, although daily carbon gain per unit leaf area was the same for low-light and high to low-light transferred plants within a week after transfer, the carbon return per unit of carbon investment in the leaves remained lower in the high to low transfer plants throughout the 4 week measurement period. Conversely, in high-light, the low leaf construction cost of the low to high-light transferred plants resulted in carbon gain per unit investment just as high as that of the high-light plants.  相似文献

Trypsin inhibitors from three edible aroids of the family Araceae, viz. taro (Colocasia esculenta) var. esculenta) giant taro (Alocasia macrorrhiza) and giant swamp taro (Cyrtosperma chamissonis) obtained from the Pacific region, were isolated by affinity chromatography and purified by gel filtration. The Mr of these inhibitors, as determined by gel filtration were 35 000–38 000, but were ca 20 000 by SDS gradient PAGE. A time course of heating in SDS showed a ready dissociation of the native protein into subunits of equal size. Further experiments showed that there were no disulfide bonds between these subunits. A single N-terminal sequence was found for each inhibitor showing that the two subunits had similar primary structure. Each of the N-terminal sequences showed homology with that of soybean trypsin inhibitor. To our knowledge, this finding follows only one other example of a Kunitz family inhibitor being located in a monocotyledonous, rather than dicotyledonous, plant species, and indicates that the ancestral gene from which Kunitz family inhibitors originate predates the evolutionary divergence of flowering plants into monocotyledons and dicotyledons.  相似文献

When leaves of Alocasia macrorrhiza that had been preconditioned in 10 micromoles photons per square meter per second for at least 2 hours were suddenly exposed to 500 micromoles photons per square meter per second, there was an almost instantaneous increase in assimilation rate. After this initial increase, there was a secondary increase over the next minute. This secondary increase was more pronounced in high CO2 (1400 microbars), where assimilation rate was assumed to be limited by the rate of regeneration of ribulose 1,5-bisphosphate (RuBP). It was absent in low CO2 (75 microbars), where RuBP carboxylase/oxygenase (Rubisco) was assumed to be limiting. It was therefore concluded that it represented an increase in the capacity to regenerate RuBP. This fast-inducing component not only gained full induction rapidly, but also lost it rapidly in low photon flux density (PFD) with a half time of 150 to 200 seconds. It was concluded that in environments with fluctuating PFD, this fast-inducing component is an important factor in determining a leaf's potential for photosynthetic carbon gain. It is especially important during brief periods (<30 seconds) of high PFD that follow moderately long periods (1 to 10 minutes) of low PFD.  相似文献

Acclimation of respiration to the light environments is important for a plant’s carbon balance. Respiratory rates of mature leaves of Alocasia odora, a typical shade‐tolerant species, were measured during the night for 14 d after reciprocal transfers between high‐ (330 µ mol m?2 s?1) and low‐light (20 µ mol m?2 s?1) environments. Following the transfer, both the rate of CO2 efflux and that of O2 uptake of A. odora leaves adjusted to the new light environments. The O2‐uptake rates changed more slowly than the CO2‐efflux rates under the new environments. Leaf mass per area also changed after the transfer. We analysed whether substrate availability or ATP‐consumption rates influence the respiratory acclimation. Since the addition of sucrose to leaf segments did not influence the O2‐uptake rates, the change of respiratory substrate availability was not responsible for the respiratory acclimation. The addition of an uncoupler induced increases in the O2‐uptake rates, and the degree of enhancement significantly decreased after the transfer from low to high irradiance. Thus, the change in ATP‐consumption rates was responsible for the changes in respiratory rates in the plants transferred from low to high light. Potential rates of O2 uptake, as measured in the presence of both the substrate and the uncoupler, changed after the transfer, and strongly correlated with the O2‐uptake rates, irrespective of the directions of transfer (r = 0·961). There was a strong correlation between maximal activities of NAD‐isocitrate dehydrogenase and the potential rates of O2 uptake (r = 0·933), but a weaker correlation between those of cytochrome c oxidase and the potential rates (r = 0·689). These data indicate that the changes of light environments altered the respiratory rates via the change of the respiratory ATP demand, and that the altered rates of respiration will induce the changes of the respiratory capacities.  相似文献

Treatment of human serum with ammonium sulfate fraction (0-50%) of Alocasia macrorhiza tuber extract resulted in precipitation at neutral pH. The precipitate was dissolved at pH 10.5 and chromatographed on Sephadex G-100 column. Two protein peaks were resolved. While the first peak represented alpha2-macroglobulin and haptoglobin, the second peak accounted for specific Alocasia protein. Incidentally the Alocasia protein was shown to be responsible for selective and specific precipitation of alpha2-macroglobulin and haptoglobin from serum. Thus the plant protein in its pure form or in crude stage could be used for the rapid isolation of two of the prominent alpha2-globulins.  相似文献

Two taxonomically undescribed Colocasiomyia species were discovered from inflorescences of Alocasia macrorrhizos in Kota Kinabalu City, Sabah, Borneo, Malaysia. The aims of this study were to investigate the reproductive ecology of the flies and the plant, ascertain the importance of the flies as pollinators and examine the intimate association between flowering events and life history of the flies. We conducted sampling, observations and field pollination experiments. The flies were attracted by the odour of female‐phase inflorescences in the early morning on the first day of anthesis. They fed, mated and oviposited in the inflorescences for 1 day. On the second day, the flies, covered with pollen grains, left the male‐phase inflorescences for the next female‐phase inflorescences. The immature forms of both fly species hatched, developed and pupated within the infructescences without damaging the fruits, and developed adults emerged when the mature infructescences dehisced. The flowering events and fly behaviours were well synchronized. In field pollination experiments, inflorescences bagged with a fine mesh (insect exclusion) produced almost no fruits, whereas those bagged with a coarse mesh (bee exclusion) produced as many fruits as the open‐pollinated controls. These results indicate that these flies are the most efficient and specialised pollinators for their host, A. macrorrhizos. These flies, in return, depend on A. macrorrhizos for food and habitat through most of their life cycle. This study provides a deeper insight into the less recognised, highly intimate pollination mutualism between Araceae plants and Colocasiomyia flies.  相似文献

Chloroplast movements in a rainforest understory plant Alocasia macrorrhiza (L.) G. Don are striking, creating changes in leaf transmittance that are visible to the naked eye. We have characterized the light requirements for these changes and the resulting changes in light penetration to different cell layers within the leaf and through the entire thickness of the leaf. Plants were grown either in a relatively constant, growth-chamber environment or in a variable, greenhouse environment. Irradiance-response curves for chloroplast movement were the same for both groups of plants, saturating at about 1 000 μmol m−2 s−1, though only the greenhouse-grown plants normally encountered light sufficient to drive the movement. Chloroplast movement caused changes in whole-leaf transmittance on the order of a few percent across the entire visible spectrum. Transmittance changes were larger within the leaf, especially directly under the palisade layer. Chloroplast movement could be manipulated experimentally by removing blue wavelengths from the spectrum of incident light or by treating with cytochalasin D.  相似文献

Alocasia comprises over 113 species of rainforest understorey plants in Southeast Asia, the Malesian region, and Australia. Several species, including giant taro, Alocasia macrorrhizos, and Chinese taro, Alocasia cucullata, are important food plants or ornamentals. We investigated the biogeography of this genus using plastid and nuclear DNA sequences (5200 nucleotides) from 78 accessions representing 71 species, plus 25 species representing 16 genera of the Pistia clade to which Alocasia belongs. Divergence times were inferred under strict and relaxed clock models, and ancestral areas with Bayesian and maximum likelihood approaches. Alocasia is monophyletic and sister to Colocasiagigantea from the SE Asian mainland, whereas the type species of Colocasia groups with Steudnera and Remusatia, requiring taxonomic realignments. Nuclear and plastid trees show topological conflict, with the nuclear tree reflecting morphological similarities, the plastid tree species' geographic proximity, suggesting chloroplast capture. The ancestor of Alocasia diverged from its mainland sister group c. 24 million years ago, and Borneo then played a central role in the expansion of Alocasia: 11-13 of 18-19 inferred dispersal events originated on Borneo. The Philippines were reached from Borneo 4-5 times in the Late Miocene and Early Pliocene, and the Asian mainland 6-7 times in the Pliocene. Domesticated giant taro originated on the Philippines, Chinese taro on the Asian mainland.  相似文献

The present authors have shown previously that both respiration rates and in vivo activities of the alternative oxidase (AOX) of leaves of Alocasia odora, a shade species, are lower than those in sun species, thereby optimizing energy production under limited light conditions (Noguchi et al., Australian Journal of Plant Physiology 28, 27–35, 2001). In the present study, mitochondria isolated from A. odora leaves were examined in order to investigate the biochemical basis for the differences in respiratory parameters. Alocasia odora and spinach plants were cultivated under both high and low light intensities, mitochondria were isolated from their leaves, and their respiratory properties compared. Mitochondrial content of leaf extracts from the two species was estimated using fumarase activities and antibody detection of porin (the voltage-dependent anion channel of the outer mitochondrial membrane). On a mitochondrial protein basis, spinach leaves showed higher capacities of the cytochrome pathway and cytochrome c oxidase (COX) than A. odora leaves. However, on a mitochondrial protein basis, A. odora showed higher capacities of AOX, which had a high affinity for ubiquinone when activated by pyruvate. Alocasia odora also had larger amounts of mitochondrial protein per leaf dry weight, even under severely shaded conditions, than spinach. Lower growth light intensity led to lower activities of most pathways and proteins tested in both species, especially glycine-dependent oxygen uptake. In the low light environment, most of the AOX protein in A. odora leaves was in its inactive, oxidized dimer form, but was converted to its reduced active form when plants were grown under high light. This shift may prevent over-reduction of the respiratory chain under photo-oxidative conditions.  相似文献

Alocasia (Alocasia macrorrhiza [L.] G. Don) and soybean (Glycine max [L.]) were grown under high or low photon flux density (PFD) conditions to achieve a range of photosynthetic capacities and light-adaptation modes. The CO2 assimilation rate and in vivo linear electron transport rate (Jf) were determined over a range of PFDs and under saturating 1-s-duration lightflecks applied at a range of frequencies. At the same mean PFD, the assimilation rate and the Jf were lower under the lightfleck regimes than under constant light. The activation state of two, key enzymes of the photosynthetic carbon reduction cycle pathway, ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and fructose-1,6-bisphosphatase, and the photosynthetic induction states (ISs) were also found to be lower under flashing as compared to continuous PFD. Under all conditions, the IS measured 120 s after an increase in PFD to constant and saturating values was highly correlated with the Rubisco activation state and stomatal conductances established in the light regime before the increase. Both the fructose-1,6-bisphosphatase and Rubisco activities established in a particular light regime were highly correlated with the mean Jf in that regime. The relationships between enzyme activation state and Jf and between IS and enzyme activation state were similar in soybean and Alocasia and were not affected either by growth-light regime, and hence photosynthetic capacity, or by flashing versus constant PFD. The common relationship between the linear Jf and the activation state of key enzymes suggests that electron transport may be the determinant of the signal regulating IS, at least to the extent that the IS is controlled by the activation state of key stromal enzymes.  相似文献
