During the mating season the female mallards produce sex pheromones, diesters of 3-hydroxy fatty acids, in their uropygial glands. Subcellular fractionation by sucrose and Nycodenz density gradient centrifugations and electron microscopic examination of the fractions showed that diesters of 3-hydroxy acids and the enzymes that catalyze the formation and esterification of the 3-hydroxy fatty acids are located in the catalase-containing fractions, probably peroxisomes, whereas monoester synthesizing activities are located in the endoplasmic reticulum. Fatty acyl-CoA reductase that would provide fatty alcohol needed for the synthesis of monoester and diester waxes was found both in the peroxisomal and endoplasmic reticulum fraction. Upon daily intramuscular injection of estradiol into the females in the nonmating season, the short chain monoester waxes of the uropygial glands were replaced by long chain monoester waxes, and subsequently the monoester waxes were replaced by diester waxes. Injection of thyroxine with estradiol hastened the induction of the compositional changes including diester synthesis. Similar changes, including the synthesis of the female pheromones, were induced in the uropygial glands by the hormone treatment of males that do not normally produce diesters at any time during their life cycle. The structure and composition of the diesters induced by hormone treatment of both males and females were identical to those of the female pheromones produced during their mating season. Electron microscopic examination of diaminobenzidine-treated glands showed that peroxisomes proliferated in the gland of the females in the mating season and in the estradiol-treated males that produce the diesters.  相似文献

Wax esters constitute the major products of the uropygial glands of male mallard ducks (Anas platyrhynchos). Combined gas-liquid chromatography and mass spectrometry of the acyl and alcohol portions of the wax ester revealed dramatic seasonal variation. Of the total of nearly 100 compounds detected in the acyl portion, more than 90 were identified as n-, monomethyl-, dimethyl-, and trimethyl-C6 to C20 acids. The major components were 2-methyl- and 4-methyl C6 acids which constituted nearly 75% of the total acids during most of the year. As the birds went through the post-nuptial molt into eclipse, these short chain acids decreased to only 4% of the total acids. Fatty acids longer than C12 became the major components during the summer months when the birds were in eclipse and by fall the characteristic shorter branched acids reappeared as the dominant components. Fatty alcohols did not undergo such dramatic changes. This is the first report of biochemical changes that occur during eclipse.  相似文献

The uropygial gland secretion of some grebes (Podicipediformes) had been shown to contain saturated and unsaturated aliphatic hydrocarbons and monoester waxes. While ester waxes are common constituents of preen gland secretions, nonisoprenoid hydrocarbons have not been detected hitherto. The wax constituents are very complex, belonging to several multibranched homologous series, including unusual acids with ethyl branches. The waxes were identified by gas-liquid chromatography-mass spectrometry and equivalent chain length comparisons. A method for the prediction of equivalent chain length values of unknown methyl esters is offered. The results are discussed from a chemotaxonomic viewpoint.  相似文献

Cell-free extracts from the uropygial gland of goose catalyzed the incorporation of malonyl-CoA and methylmalonyl-CoA into n- and multi-branched fatty acids, respectively, with NADPH as the preferred reductant. Methylmalonyl-CoA was shown to be incorporated almost exclusively into the acyl portion of wax esters by the cell-free extract while malonyl-CoA was incorporated into polar lipids and both the acyl and alcohol portions of the wax. The optimal pH for the synthesis of both n- and multibranched acids was 6.0. Apparent Km and Vmax for malonyl-CoA were 2 times 10- minus-4 M and 250 nmol per min per mg, respectively, while the Km and Vmax for methylmalonyl-CoA were 7.7 times 10- minus-4 M and 0.8 nmol per min per mg, respectively with 105,000g supernatant; but partial purification resulted in a tenfold decrease in Km values. The partially purified synthetase preparation catalyzed the formation of n-C16 acid (80%) and n-C18 acid (20%) from acetyl-CoA and malonyl-CoA. With the same synthetase preparation and the appropriate primer methylmalonyl-CoA was converted into 2,4,6,8-tetramethyldecanoic acid and 2,4,6,8-tetramethylundecanoic acid which were identified by radio gas-liquid chromatography and combined gas chromatography-mass spectrometry. Experiments with an equimolecular mixture of acetyl-CoA and propionyl-CoA showed that the synthetase preferred acetyl-CoA as a primer. Since malonyl-CoA is known to be rapidly decarboxylated in the gland, acetyl-CoA and methylmalonyl-CoA are expected to be the major primer and elongating agent, respectively, available in the gland and therefore 2,4,6,8-tetramethyldecanoic acid should be the major product. Combined gas-liquid chromatography and mass spectrometry demonstrated that this acid was in fact the major acid of the gland.  相似文献

In spite of the importance of chemoreception and chemical signals in the social organization of lizards, there are only a few studies examining the chemical composition of secretions of lizards used for scent marking. The secretion of the femoral glands of male Iberian rock lizards (Lacerta monticola cyreni) contains 44 lipophilic compounds, including several steroids (mainly cholesterol), and n-C6 to n-C22 carboxylic acids, and minor components such as esters of carboxylic acids, alcohols, squalene, and one lactone. These compounds were identified on the basis of mass spectra, obtained by GC-MS. Most lipids were detected in all individuals, although relative proportions of each chemical show a high interindividual variability. This variability might be related to the characteristics or physical and health condition of males and might be the basis of female choice based on chemical cues observed in this lizard species.  相似文献

Recent studies have indicated that avian social behavior is influenced by olfactory cues. During the reproductive season a change in the chemical composition of uropygial gland secretion has been reported in some species and the hypothesis that olfactory signals may be produced by this gland has been proposed. To examine this hypothesis we performed two behavioral experiments to determine whether a female’s uropygial gland produce chemical signals that stimulate mating behaviors in domestic chickens. In Experiment 1 the role of the female’s uropygial gland in male mating behavior was examined by removing and examining the female’s uropygial gland. The frequency of mounts and copulations of intact male birds with sham-operated female birds was significantly higher than with uropygial glandectomized female birds. With respect to the number of waltzing that is one of the courtship displays intact males showed no significant difference between sham-operated female birds and uropygial glandectomized female birds. In Experiment 2 the relationship between male olfaction and the female’s uropygial gland was investigated using olfactory bulbectomized male birds. The number of mounts and copulations of sham-operated male birds with sham-operated female bird was significantly higher than with uropygial glandectomized female birds. In contrast olfactory bulbectomized male birds showed no significant differences in the number of mounts and copulations between sham-operated female birds and uropygial glandectomized female birds. These results indicate that intact and sham-operated male birds prefer to mate with female birds with the uropygial gland. The number of courtship waltzing of sham-operated male birds showed no significant difference. However olfactory bulbectomized male birds significantly courted to uropygial glandectomized female birds. Summarizing our results show that while anosmic males did not have any preference both intact and sham-operated male birds chose to mate with female birds having an intact uropygial gland suggesting that mate preference involves in male olfaction and that the female’s uropygial gland acts as a source of social odor cues in domestic chickens.  相似文献

The uropygial gland secretion of penguins (Sphenisciformes) is a complex mixture of monoester waxes. The wax fatty acids of Pygoscelis and Eudyptes are predominantly 3-methyl- and 3, x-dimethyl-substituted, whereas 2- and 4-monomethyl-, or 2, x-as well as 4, x-dimethyl-substituted acids predominate in the case of Spheniscus. The wax alcohols are very similar within all three species and are mainly composed of mono- and dimethyl-substituted compounds with branches predominantly in the middle or near the methyl end of the carbon chain. From these data, it is concluded that, chemotaxonomically, the penguins are closely related to the tubenoses (Procellariiformes).  相似文献

采用扫描电镜和气相色谱/质谱联用技术研究了角蜡Ceroplastes ceriferus (Fabricius) 和日本龟蜡蚧C. japonicus Green蜡泌物的超微结构及化学成分。结果表明,这2种蜡蚧分泌蜡质和形成蜡壳的过程基本相似。在1、2龄期分泌的蜡质为“干蜡”,蜡壳为星芒状,虫体周缘的蜡芒均为2大节,与其2个龄期发育相对应,每一个大节又分为若干小节。同时,虫体背面中央蜡质堆积成帽状,也分为均匀的多层。由此说明泌蜡过程具有节律性。虫体周缘与蜡芒对应的突起区上分布着密集的刻点状腺孔,每一个腺孔分泌1根蜡丝,这在以往玻片标本中是观察不到的。雌性第3龄幼虫和成虫期,虫体分泌“湿蜡”,形成龟背状蜡壳,泌蜡腺孔主要为三格腺和四格腺。在肛突区发现了密集的泌蜡腺孔,排列为纵条纹状。从角蜡蚧蜡泌物甲酯化处理样品中检测到14个组分,从直接测试 (未经甲酯化处理) 样品中检测到14个组分;而从日本龟蜡蚧则分别检测到10个组分和25个组分。它们的主要成分是一系列高级的长链饱和与不饱和的烃、脂肪酸、脂肪醇、酯类、醛类以及杂环、多环或大环状化合物。对它们可能的生物生态学功能进行了讨论。  相似文献

Hydrocarbon and wax ester components of cuticular lipids of the braconid parasitoid Habrobracon hebetor Say reared at 25 degrees C on larvae of a pyralid moth have been identified by GC-MS and analyzed with respect to adult age, mating status, and diet. The hydrocarbons range in carbon number from C(21) to C(45) and consist of a homologous series of n-alkanes, 11-, 13-, and 15-methyl alkanes, 13,17-dimethyl alkanes, and Z-5, Z-7, and Z-9-alkenes. The wax esters found in the cuticular lipid fraction are a series of homologous compounds with the acid portion being short chain, unbranched, even carbon number acids from C(8) to C(20) (predominately C(8) to C(16)). The alcohol portions of the esters are secondary alcohols with carbon number from C(22) to C(25) (predominately C(23) and C(25)) with the hydroxyl function located at C(6), C(7), C(8), and C(9). Gender, age, and nutritional states were significant factors for variation in several of the individual esters, but mating status did not affect wax ester composition. Ontogenetic examinations indicated that prepupal, and early pupal cuticular lipids contain only hydrocarbons. Low levels of wax esters are detectable in late stage pupae, and somewhat greater quantities of wax esters are present on newly eclosed adults. When pharate adults emerge from the cocoon, however, their cuticular lipids consist of approximately equal amounts of hydrocarbons and wax esters, and 6d post emergence from the cocoon, wax esters are the predominant lipid component.  相似文献

Pathogens such as bacteria, fungi and malaria and related haemosporidians provoke negative effects on the fitness of their hosts. Animals have developed a range of defensive mechanisms to resist or eliminate these parasitic infections and their negative fitness costs. The uropygial gland secretion has been proposed to act as defensive barrier of skin and plumage in the fight against bacteria and fungi, and may prevent birds from acquiring haemosporidian infections. Thus, the secretion of uropygial glands of birds may favour survival of individuals. However, whether uropygial gland secretion influence survival remains unknown. Here we explore if the size of the uropygial gland and malaria infection influence survival of house martins Delichon urbica. We showed, for the first time, that the volume of the uropygial gland positively predicted survival prospects of malaria infected house martins. Malaria infected birds had the lowest probability of survival, with the effect of gland size on survival prospects depending on infection: infected house martins with larger uropygial glands were better able to survive to the next breeding season, while infected birds with small uropygial glands were not. These results highlight the importance of uropygial gland secretion in the life history of wild birds.  相似文献

Lipid constitutents of diapausing pupae of the tobacco hornworm, Manduca sexta (L), were identified by thin layer and gas-liquid chromatography, IR spectroscopy, and gas-liquid chromatography-mass spectrometry. Surface wax was a mixture of long chain polar compounds: oxoalcohol esters, oxoaldehydes, primary alcohol esters, and oxoalcohols, as listed in descending order of abundance. The distribution of the alcohols and aldehydes was C28 (75-85%), C27 (5%), and C26 (10-15%). The C26 compounds were largely 11-oxo isomers, but the C28 compounds consisted of similar amounts of 11- and 12-oxo isomers. The identities of the oxoaldehydes were confirmed by selective and complete NaBH4 reductions to yield oxoalcohols and diols, respectively. Mass spectral interpretations were verified with mass spectra of the oxoaldehyde, oxoalcohol, and diol synthesized from 12-hydroxystearic acid. Reduction of the total lipids with NaBH4 and hydrolysis of the product with ethanolic KOH gave oxoalcohols (85%), primary alcohols (8%), and oxoacids (5%); 30-40% of the oxoalcohols were derived from NaBH4-reduced oxoaldehydes, 5-10% were from free oxoalcohols, and 50% were from esters. Primary alcohols only existed as esters. Large quantities of fatty oxoalcohols relative to fatty oxoacids in the saponified mixture suggested the presence of esters other than those composed of long chain acids and alcohols. Oxoacids appeared to be mainly oxidation products of the oxoaldehydes.  相似文献

The uropygial glands of birds serve multiple functions, and there is great interspecific variability in the composition and properties of their secretions. A special case is the secretion in the hoopoes Upupa epops, and green woodhoopoes Phoeniculus purpureus, which, contrary to the commonly white and odourless secretions, are dark with pungent odour. Recently, bacteria have been isolated from glands of both woodhoopoes and hoopoes and here we test the hypothesis that bacteria are responsible of some of the special properties of glands and secretions of this group of birds. We explore natural seasonal changes and intersexual differences in the properties of hoopoe glands and secretions, check the natural occurrence of bacteria within secretions, and analyse the effect of experimental injection of antibiotics on uropygial gland properties. Male glands underwent no seasonal changes, and their secretions were invariably white and odourless, very similar to female glands outside the breeding season. However, in comparison to the uropygial gland of non‐breeding females, those of incubating females showed a marked increase in size and volume of secretion produced, which became dark and pungent. All these parameters increased until the hatching date and returned to values similar to those in the prelaying phase towards the end of the nestling period. Nestling glands produced secretions similar to those of females in colour and odour. Gland size of both females and nestlings predicted the amount of secretion produced. Microscopic techniques confirmed the presence of bacteria at high density and in active division in all dark secretions examined. The antibiotic treatment significantly reduced the load of enterococci in nestling glands, did not affect size of glands, but diminished the volume of secretion, which was lighter in colour than that of control nestlings. In nesting females, the experimental injection of antibiotic affected some measurements of gland size and secretion colour. Because the experiment did not affect general health estimates (immunocompetence, body condition or growing) of nestlings, our results suggest that some of the special properties of hoopoe glands are mediated by the presence of symbiotic bacteria.  相似文献

Although chemoreception plays an important role in social organization of many lizards, only a few studies have examined the chemicals found in secretions used for intraspecific communication. We report the composition of the secretion of the femoral glands of males of the spiny-footed lizard (Acanthodactylus erythrurus). On the basis of mass spectra, obtained by GC/MS, we identified 45 lipophilic compounds, including several alcohols ranging from 10 to 29 carbon atoms (mainly hexacosanol and tetracosanol), steroids (mainly cholesterol and dehydrocholesterol), n-C9 to n-C20 carboxylic acids, esters of carboxylic acids, and minor components such as lactones, ketones, squalene and a-tocopherol. Some of these compounds are reported for the first time in lizards. Adult and subadult males differed in the composition of secretions, with C9 to C15 carboxylic acids being more abundant in younger than in older lizards, whereas C16 to C20 carboxylic acids were more abundant in older lizards. Also, older lizards had significant lower proportions of cholesterol and campesterol but higher proportions of dehydrocholesterol.  相似文献

The compositions of the uropygial gland secretions of the long-eared owl, eagle owl, and barn owl have been determined. The waxes of the first two owls, which are closely related, are composed of 2-alkyl-substituted fatty acids and n- or monomethyl-branched alcohols with even-numbered branching positions. In addition, some dimethyl-substituted alkanols were observed. In contrast to these waxes, the secretion of the barn owl is composed of 3-methyl- and 3,5-, 3,7-, 3,9-, 3,11-, 3,13-, and 3,15-dimethyl-branched fatty acids and n- as well as monomethyl-substituted alkanols branched at positions 2, 3, and 4. The mass spectra of esters of 2-alkyl-substituted fatty acids are discussed.  相似文献

Age at sexual maturity among male polar bears (Ursus maritimus) and timing of the mating season were studied by microscopic and macroscopic analysis of the reproductive organs from 165 male and 92 female Greenland polar bears. Males became sexually mature in the spring following their fifth birthday. Testes sampled during the spring reached 95% of their asymptotic length at 5.8 years. Some spermatozoa were found in 2- to 4-year-old bears during the spring, but only in low concentrations in a mixture of other cell types. The first corpus luteum was found on 1 April and most ovulations were estimated to occur before mid-May which indicate that the peak of the mating season begins in late March and ends in May. These findings are somewhat different from a similar study from Svalbard, where the mating season is estimated to peak later and male polar bears are found to become mature earlier. We hypothesize that these differences occur because adolescent males have their peak testicular activity after the main breeding period and therefore appear to be fully sexually mature during summer and not during spring.  相似文献

采用扫描电镜、红外光谱、气相色谱/质谱联用技术研究了柑橘真棉蚧Eupulvinariacitricola(Kuwana)(半翅目:蚧科)蜡泌物的超微形态、红外光谱和卵囊蜡质的化学成分特征。结果发现:该蚧雌虫背面腺体分泌湿蜡,在体表面形成薄的半透明蜡壳,背面蜡壳分为6个小区,表面由湿蜡凝结成片状和蜡块状构造。雌成虫产卵期由腹面多格腺分泌丝状蜡结成卵囊。雄若虫也分泌湿蜡,雄蛹的蜡茧薄,表面有浅的凹凸花纹。用雌成虫卵囊、背面蜡壳和雄茧的蜡质作红外光谱分析,它们的谱图具有共同的基本特征,吸收峰群分为5个区域:2900cm-1附近的3个强吸收峰组成的峰群是>CH2和—CH3的特征吸收峰;1800cm-1~1500cm-1之间的吸收峰群是羰基(>C=O)和C=C键的伸缩振动;1500cm-1~1400cm-1区域是>CH2和—CH3基团的吸收峰;1400cm-1~1000cm-1之间为饱和碳链—C—C—C—骨架振动。730cm-1附近为环状化合物的基团吸收峰群。由此确定它们的组成都是长碳链结构的脂肪酸、脂肪醇、烃类、酯类或带有环状结构的化合物。其主要区别在第2、3、4区吸收峰的形式、强度和数量,反映出基团的种类和数量不同。雌成虫卵囊蜡质甲酯化后GC/MS检测出7个组分(表1),全部为长链脂肪酸。未经甲酯化的蜡质检测出4类16个组分(表2),可分为长链烃、酸、醇、酯。第一类为饱和长链烃,5个组分,占总组分的38.38%;第二类是长链脂肪酸,包括直链饱和酸和不饱和酸共6个组分,占31.59%;第三类是直链饱和醇,占2.87%;第四类是酯类化合物,4个组分,占总组分的27.16%。  相似文献

The ontogenetic and seasonal development of wax composition and cuticular transpiration of sun and shade leaves of ivy (Hedera helix L.) was analysed by investigating leaves varying in age between 4 and 202 d. It was discovered that the total amount of solvent-extractable wax was composed of two distinct fractions, separable by column chromatography: (i) a less polar or apolar monomeric wax fraction consisting of the typical linear, long-chain aliphatics usually described as cuticular wax components and (ii) a polar, oligomeric wax fraction consisting of primary alcohols and acids mostly esterified to C12-, C14- and C16-ω-hydroxyfatty acids. The apolar wax fraction, which could be analysed directly by gas chromatography coupled with mass spectrometry (GC-MS), exhibited pronounced seasonal changes in composition. Wax amounts in the apolar fraction reached a maximum after about 30 d and gradually decreased again during the remaining period of the season investigated. In contrast, the polar wax fraction, which was analysable by GC-MS only after transesterification, rapidly increased early in the season, reaching a plateau after 40 d, and then remained constant during the rest of the season. Thus, total amounts of solvent-extractable cuticular waxes, which can be determined gravimetrically, will only be detected by GC-MS after fractionation and transesterification, a methodological approach rarely applied in the past in cuticular wax analysis. Additionally, investigation of the cutin polymer matrix after depolymerisation through transesterification, revealed that only those primary alcohols and acids forming an essential part of the apolar and the polar wax fractions were esterified during the investigated season and incorporated in increasing amounts into the cutin polymer matrix (matrix-bound wax fraction). Thus, it can be concluded that a complete analysis of cuticular wax of ivy and its seasonal development can only be achieved if all the relevant fractions (i) the less polar or apolar, (ii) the polar and (iii) the wax fraction bound to the cutin polymer matrix are investigated. Cuticular transpiration rapidly decreased within the first 30 d and essentially remained constant during the rest of the season. Thus, changes in cuticular water permeability were closely correlated with the most prominent changes in wax amounts and composition occurring during the first 30 d of ontogenetic leaf development. However, during the remainder of the year, up to 202 d, cuticular transport properties remained constant, although significant quantitative and qualitative changes in cuticular wax composition continued to occur. Thus, our study clearly demonstrated that there will be no simple relationship between chemical composition of cuticular waxes and transport properties of isolated ivy leaf cuticles. Long-chain aldehydes, alcohols, hydrocarbons and wax esters were major components of the external lipids of adult Aleyrodes singularis. In exuviae, acetate esters replaced the hydrocarbons as a major component. The major long-chain alcohol and aldehyde from adults were C32 and were essentially the exclusive components of the wax particles. The major alcohol from exuviae was C26 and the aldehydes were C26, C28, C30 and C32. The major acetate esters were C28 and C30 in both adults and exuviae. There were wax esters of similar carbon number in adults and exuviae although the exuviae had a greater amount of wax esters with unsaturated fatty acids. The fatty acid and alcohol composition of the wax esters differed markedly between adults and exuviae. Wax esters of adults had similar amounts of C16, C18, C20, C22 and C24 fatty acids while those from exuviae contained largely C16 and C18. The major alcohol in the wax esters of adults was C22 and those of exuviae were C26 and C28. The distribution of fatty acids and alcohols among wax esters of varying chain length also differed between adults and exuviae: in adults C22 was the major fatty acid found in the dominant wax ester, C44 and the C22 alcohol was the major alcohol and found in wax esters C42 and C44. In exuviae C16 and C18 were the major fatty acids found in most wax esters and a C28 alcohol was the major alcohol found in wax esters C44 and C46, the two dominant wax esters in exuviae. It was clear that the difference in chemistry of the wax esters between the adults and exuviae is not evident unless the acid and alcohol moieties are characterized.  相似文献

Diesters of 3-hydroxy C8, C10, and C12 acids, the female mallard duck pheromones, were found as the major products of the uropygial glands only during the breeding season. The 3-hydroxy acids were identified by mass spectrometry of the trimethylsilyl ethers of the methyl esters and of the diols derived from LiAlH4 reduction of the hydroxy acids. A cell-free extract from the gland catalyzed conversion of dodecanoic acid to 3-hydroxydodecanoic acid which was identified by radio thin-layer and radio gas chromatographic analysis of the enzymic products as methyl-3-acetoxydodecanoate and as diacetate of the diol generated by LiAlH4 reduction of the enzymic product. The enzymic introduction of the hydroxyl group at C-3 was catalyzed mainly by a 50,000g pellet prepared from a 1000g supernatant obtained from the cell-free extract. This reaction required ATP, CoA, and O2, and the CoA ester of the acid was more efficiently converted than the free acid to the 3-hydroxy acid. KCN at 1 mM and 50% CO did not inhibit the reaction. 3H from 3H2O was incorporated into 3-hydroxydodecanoic acid during the enzymic synthesis of this acid from dodecanoic acid. Mass spectrometry of the 3-hydroxy acid generated by the particulate fraction in the presence of H2 18O showed that 18O was incorporated as expected from hydration of a delta 2 double bond. From the above results it is tentatively concluded that peroxisomal acyl-CoA oxidase converts the acyl-CoA to the 2-enoyl-CoA which is hydrated to generate the 3-hydroxy acid.  相似文献
