Summary The structure of anther tissues has been studied during microsporogenesis in male-sterile and -fertile onions. Three types of abnormal tapetal behaviour have been observed within the single line II/3ms containing the cms-S cytoplasm: type 1, the premature breakdown of the tapetum at the tetrad stage, type 2, the hypertrophy of the tapetum after the diad stage followed by its premature autolysis and, type 3, in which the tapetum remains in good condition but for an abnormally long period of time. Tapetal autolysis proceeds in the same manner in both male-steriles and -fertiles with only the stage at which it occurs differing between the types of plants. Mitochondria were prominent in the tapetal tissue of all onion types throughout all stages of microsporogenesis and were still visible during the last stages of tapetal autolysis. In a detailed study of type 2 behaviour, no differences in mitochondrial volumes were found until the tapetum hypertrophied.  相似文献

The effects of lead and copper on the arrangement of microtubule (MT) cytoskeleton in root tip cells of Allium sativum L. were investigated. Batch cultures of garlic were carried out under defined conditions in the presence 10−4 M Pb/Cu of various duration treatments. With tubulin immunolabelling and transmission electron microscopy (TEM), we found four different types of MT structures depending on the cell cycle stage: the interphase array, preprophase band, mitotic spindle and phragmoplast were typical for the control cells. Pb/Cu affected the mechanisms controlling the organization of MT cytoskeleton, and induces the following aberrations in interphase and mitotic cells. (1) Pb/Cu induced the formation of atypical MT arrays in the cortical cytoplasm of the interphase cells, consisting of skewed, wavy MT bundles, MT fragments and ring-like tubulin aggregations. (2) Pb/Cu disordered the chromosome movements carried out by the mitotic spindle. The outcome was chromosome aberrations, for example, chromosome bridges and chromosome stickiness, as well as inhibition of cells from entering mitosis. (3) Depending on the time of exposure, MTs disintegrated into shorter fragments or they completely disappeared, indicating MT depolymerization. (4) Different metals had different effects on MT organization. MTs were more sensitive to the pressure of Cu ions than Pb. Moreover, TEM observations showed that the MTs were relatively short and in some places wavy when exposed to 10−4 M Pb/Cu solutions for 1–2 h. In many sections MTs were no longer visible with increasing duration of treatment (>4 h). Based on these results, we suggested that MT cytoskeleton is primarily responsible for Pb/Cu-associated toxicity and tolerance in plants.  相似文献

Summary The DNA of the organellar genomes of Allium cepa has been examined to detect restriction fragment length polymorphisms. Differences can be shown between both the chloroplastal and mitochondrial genomes of the N and cms-S cytoplasms in their restriction fragment profiles. Southern blot analysis of the mtDNA profiles using probes containing defined mitochondrial genes also detected polymorphisms. No differences can be shown between the organellar genomes of the N and cms-T onions by either of these techniques. These data indicate different origins for the two sterility-conferring cytoplasms, suggesting autoplasmic and alloplasmic origins for the cms-T and cms-S cytoplasms, respectively. No evidence of the presence of virus-like particles was found in any of the cytoplasms.  相似文献

Abstract

Karyological and chemical variations in Allium sativum L. — Results about a research on the karyotypes of three Italian strains of Allium sativum are reported. The lack of sexual reproduction in Allium sativum favours the formation of strains regarded as clones; each of them represents an isolated genetic system. The anatomy and the chemical composition of the essence was studied to improve the understanding of the variations in the different strains comprised in this species. Two of the clones examined (Tuscan and Sicilian) belong to U type and one (Piedmontese) to the H type, according to Hrubý and Konvi?ka's classification based on the morpho-physiological characters. In this study evidence shows that: a) The three clones do not present anatomical differences. b) The morpho-physiological characters are strictly correlated with the chemical composition of the essence; in fact the gas-chromatographic study of the two U type clone essences shows the same qualitative composition, each chemical component being present more or less in the same percentage. The essence of Piedmontese clone, is qualitatively and quantitatively different from the others. c) The number and the general morphology of the chromosomes of the three root tip karyotypes agree with the literature, nevertheless the karyotypes sometimes show differences in the two satellited pairs of chromosomes. The Tuscan karyotype (U type) is comparable with the OU type of Konvi?ka and Levan. The Piedmontese karyotype (H type) is comparable with the OH karyotype of the same authors; instead the Sicilian karyotype is very different from all karyotypes in literature and from the Tuscan karyotype belonging to the same U type because of the lack of the pseudosatellite in a partner of pair no. 6. Research evidences that, for the time being, it is impossible to use only chromosome morphology to establish the clone types: nevertheless their chemical characteristics seem to correspond to their morpho-physiological features.  相似文献

Summary. In Lavatera thuringiaca, kariokinesis and simultaneous cytokinesis during the meiotic division of microsporogenesis follow a procedure similar to that which takes place in the majority of members of the class Angiospermae. However, chondriokinesis occurs in a unique way found only in species from the family Malvaceae. Chondriokinesis in such species is well documented, but the relationship between the tubulin cytoskeleton and rearrangement of cell organelles during meiosis in L. thuringiaca has not been precisely defined so far. In this study, the microtubular cytoskeleton was investigated in dividing microsporocytes of L. thuringiaca by immunofluorescence. The meiotic stages and positions of cell organelles were identified by staining with 4′,6-diamidino-2-phenylindole. We observed that, during prophase I and II, changes in microtubular cytoskeleton configurations have unique features, which have not been described for other plant species. At the end of prophase I, organelles (mostly plastids and mitochondria) form a compact envelope around the nucleus, and the subsequent phases of kariokinesis take place within this arrangement. At this point of cell division, microtubules surround the organelle envelope and separate it from the peripheral cytoplasm, which is devoid of plastids and mitochondria. In telophase I, two newly formed nuclei are tightly surrounded by the cell organelle envelopes, and these are separated by the phragmoplast. Later, when the phragmoplast disappears, cell organelles still surround the nuclei but also move a little, starting to occupy the place of the disappearing phragmoplast. After the breakup of tetrads, the radial microtubule system is well developed, and cell organelles can still be observed as a dense envelope around the nuclei. At a very late stage of sporoderm development, the radial microtubule system disappears, and cell organelles become gradually scattered in the cytoplasm of the microspores. Using colchicines, specific inhibitors of microtubule formation, we investigated the relationship between the tubulin cytoskeleton and the distribution of cell organelles. Our analysis demonstrates that impairment of microtubule organization, which constitutes only a single component of the cytoskeleton, is enough to disturb typical chondriokinesis in L. thuringiaca. This indicates that microtubules (independent of microfilaments) are responsible for the reorganization of cell organelles during meiotic division. Correspondence: D.  Interspecific hybrids between Allium cepa and A. ampeloprasum have been generated as a first step for the introduction of S-cytoplasm from onion into leek. Pre-zygotic barriers of crossability were observed after the arrival of pollen tubes at the end of the style when entering the cavity. Nevertheless, micropyle penetration of pollen tubes and the formation of hybrid embryos were also observed. After accomplishing in vitro culture of ovaries and ovules successively, triploid hybrid plants with 24 chromosomes were obtained. Their hybrid nature was confirmed by RAPD analysis, genomic in situ hybridization, and morphological analysis. Allium sativum germplasm collected from different parts of India was subjected to critical analysis of 14 easily noticeable morphological traits. Variability encountered in quantitative as well as qualitative, bulb and plant traits was documented. While analysing this variability, an attempt was made to highlight the genotype-environment interaction, its impact on the expression of morphological traits in this apomict and evaluate the different collections for intraspecific phylogeny.  相似文献

 Mitochondrial (mt) DNA variation for six petaloid cytoplasmic male-sterile (CMS) and three fertile maintainer lines of carrot was assessed to establish genetic relationships. Total DNA was digested with restriction enzymes and probed with six homologous mtDNA cosmid probes. The six CMS accessions derived from wild carrot, four from Guelph, Ontario, one from Orleans, Massachusetts, and one from Madison, Wisconsin, were more closely related with each other (F=0.91) than with fertile maintainer lines derived from cultivated germplasm (F=0.62). The fertile maintainer lines were likewise found to be more similar to each other (F=0.78) than to the sterile lines. Three sterile lines, originating from wild carrot populations within 1 km of each other in Guelph, Ontario, were most closely related (F=0.96). The high degree of similarity among the six petaloid CMS lines which originated from individual wild carrot plants, some from geographically diverse regions, suggests that the cytoplasm responsible for this trait was imported to, or else evolved, only once in North America.  Results are reported on the production and characterization of somatic hybrids between Allium ampeloprasum and A. cepa. Both symmetric and asymmetric protoplast fusions were carried out using a polyethylene-based mass fusion protocol. Asymmetric fusions were performed using gamma ray-treated donor protoplasts of A. cepa and iodoacetamide-treated A. ampeloprasum protoplasts. However, the use of gamma irradiation to eliminate or inactivate the donor DNA of A. cepa proved to be detrimental to the development of fusion calli, and thus it was not possible to obtain hybrids from asymmetric fusions. The symmetric fusions yielded a high number of hybrid calli and regenerated plants. The analysis of the nuclear DNA composition using interspecific variation of rDNA revealed that most of the regenerated plants were hybrids. Flow cytometric analysis of nuclear DNA showed that these hybrid plants contained a lower DNA content than the sum of the DNA amounts of the parental species, suggesting that they were aneuploid. A shortage of chromosomes in the hybrids was confirmed by genomic in situ hybridization. Chromosome counts in metaphase cells of six hybrids revealed that these plants lacked 2–7 leek chromosomes. One hybrid showed also the loss of onion chromosomes. The hybrids had an intermediate phenotype in leaf morphology. The application of these somatic hybrids in breeding is discussed. 1. Alliin lyase (EC 4.4.1.4) was purified up to sevenfold from garlic-bulb homogenates. The enzyme was unstable to storage at -10 degrees , particularly in dilute concentrations, but the addition of glycerol (final concentration 10%, v/v) stabilized the activity completely for at least 30 days. 2. The purified enzyme had an optimum pH for activity at 6.5. The addition of pyridoxal phosphate stimulated the reaction rate and the stimulation became more marked as the purification proceeded. 3. Hydroxylamine (10mum) and cysteine (0.5mm) inhibited the enzyme activity by more than 80%. Spectral studies indicated that cysteine reacted with pyridoxal phosphate bound to the protein. 4. The K(m) values for S-methyl-, S-ethyl-, S-propyl-, S-butyl- and S-allyl-l-cysteine sulphoxides were determined. With S-allyl-l-cysteine sulphoxide the K(m) was 6mm and the V(max.) was greater than those with the other substrates tested. 5. The thioether analogues of the substrates were competitive inhibitors for the lyase reaction. The K(i) decreased with increasing chain length of the alkyl substituent. With S-ethyl-l-cysteine sulphoxide as substrate the K(i) was 33, 8 and 5mm respectively for S-methyl-, S-ethyl- and S-propyl-l-cysteine. 6. The addition of EDTA or Mg(2+), Mn(2+), Co(2+) or Fe(2+) stimulated the reaction rate. Other bivalent cations either had no effect or gave a strong inhibition. In the presence of EDTA no further increase of activity was observed with added Mg(2+).  相似文献

Summary A new simple, efficient and rapid in vitro method for mass clonal propagation of leek (Allium ampeloprasum L.) plants, using small (5 mm) flower stalk (peduncle) explants, was established. Adventitious shoots were produced from single subepidermal cells. A wide variation in the percentage of regenerating explants and number of regenerated shoots per explant between individual plants within one cultivar was observed. The concentration of the growth regulators 6-benzylaminopurine and -naphthalene-acetic acid influenced the percentage of regenerating explants and the average number of regenerated shoots per explant. A combination of 10 mg.l–1 6-benzylaminopurine and 10 mg.l–1 -naphthalene-acetic acid, resulted in a maximum percentage of regenerating explants and a high average number of regenerated shoots per explant. The percentage of regenerating explants and the average number of regenerated shoots per explant decreased with increasing flower stalk length (age). The basal explants gave both the highest percentage of regenerating explants and average number of regenerated shoots per explant. An average of 300 shoots per flower stalk was obtained for all plants, making this new in vitro method a powerful tool in hybrid leek breeding.  相似文献

采用聚丙烯酰胺凝胶电泳(PAGE)分离了中国大蒜3个生态型(低温反应敏感型、低温反应中间型和低温反应迟钝型)中18个较典型的品种的酯酶同工酶,并用排序分析法对18个品种的亲缘关系进行了分析,将18个品种分为3个变种群:1.“苏联”蒜变种群(var.Russia),2.吉木萨尔白皮蒜变种群(var.:Jimusaer。),3.中国内陆大蒜变种群(var.China)。其中中国内陆大蒜变种群又可分为5个品种群:①关中蒜品种群,②西北蒜品种群,③西南蒜品种群,④云贵蒜品种群,⑤华东蒜品种群。实验结果初步证实,大蒜生态型不能完全等同于基因型,酯酶同工酶的变化可能更能说明大蒜的亲缘进化关系。  相似文献

Dormancy in garlic cv. Rosado Paraguayo was studied during plantontogeny in relation to endogenous growth regulators and bulb-forming.The differentiation of seed cloves began coincidently with abulb-forming rate of 0.47 and a lack of gibberellin and moderateinhibitor activities. This hormone levels remained at harvest,when the seed cloves, which had completed their final size,were unable to sprout. Summary Two protocols for clonal propagation of kurrat (Allium ampeloprasum var.kurrat) using explants from the basal plates of mature plants are described. In direct formation, explants were cultured in Murashige and Skoog (MS) medium and supplemented with either benzyladenine at 0.0 or 4.4 μM, or supplemented with 7.0 μM benzyladenine and 0.1 μM naphthaleneacetic acid. Shoots appeared after 4 wk of culture. In the two-step procedure, explants were cultured first on MS medium supplemented with 1.4 μM 2,4-dichlorophenoxyacetic acid and 1.4 μM kinetin, and incubated in the dark for 4 wk. They were then transferred to MS medium supplemented with 4.4 μM benzyladenine for shoot formation. All shoots were rooted on MS medium containing 5 g·liter−1 activated charcoal. Normal viable plants were successfully established in soil.  相似文献
