目的:探讨Egr-1基因沉默对人肺腺癌A549细胞放射敏感性的影响。方法:选用A549细胞株作为研究对象,将其分成A、B、C三组,即空白对照组(只加入RPMI-1640培养)、阴性对照组(加入LV3-NC-sh RNA)、实验组(加入EGR1-homo-2294-sh RNA),采用慢病毒介导的sh RNA干扰技术使实验组细胞Egr-1基因沉默表达。利用荧光显微镜、自动化荧光定量细胞成像分析系统分析sh RNA转染,利用FQ-PCR分析Egr-1表达,再利用细胞克隆形成实验检测细胞放射敏感性参数的差异。结果:慢病毒介导的sh RNA成功转染阴性对照组、实验组细胞;空白对照组与阴性对照组Egr-1表达无差异(P0.05),实验组与空白对照组、阴性对照组比较Egr-1均受到明显抑制(P0.05);克隆形成实验中细胞放射敏感性参数D0、SF2实验组细胞与空白对照组、阴性对照组比较均存在明显差异(P0.05)。结论:Egr-1基因沉默使A549细胞放射敏感性降低,Egr-1表达可能与肿瘤细胞对放射的敏感性有关。  相似文献

目的:克隆人颗粒溶素和穿孔素基因并构建真核表达载体pGNLY-2A-PFP,观察其在人肺癌A549细胞中的表达情况。方法:体外分离培养外周血单个核细胞并抽提总RNA,RT-PCR扩增人颗粒溶素和穿孔素的基因片段,并将其分别插入pMD18-T进行测序,鉴定正确后构建pIRES-GNLY、pIRES-PFP及pGNLY-2A-PFP并转染人肺癌A549细胞,RT-PCR和间接免疫荧光检测目的蛋白表达,流式细胞Annexin V/PI检测转染后细胞凋亡情况。结果:成功获取了人颗粒溶素和穿孔素cDNA,构建了真核表达载体pIRES-GNLY、pIRES-PFP、pGNLY-2A-PFP,转染A549细胞后检测出了目的蛋白的表达,pGNLY-2A-PFP转染组细胞死亡率高于其他对照组(P<0.05),死亡细胞以凋亡为主。结论:人颗粒溶素和穿孔素基因可以在人肺癌A549细胞中表达,二者共表达能够促进细胞凋亡,这将有助于颗粒溶素和穿孔素在肿瘤治疗中应用的后续研究。  相似文献

探讨IL-8对肺腺癌A549细胞迁移的影响及其可能机制。用MTT法选择了合适的IL-8使用浓度。分别用划痕试验及Transwell试验证明了IL-8可以促进肺腺癌A549细胞的迁移。Westernblot结果表明:(1)IL-8可以促进MMP-2蛋白的表达,而对MMP-9的表达无明显影响;(2)IL-8可促进JNK/SAPK磷酸化蛋白的表达;(3)抑制剂(SP600125)可以阻断IL-8对MMP-2蛋白表达的影响。划痕试验从反面验证了低表达的MMP-2可以抑制A549细胞的迁移。表明IL-8可通过JNK/SAPK信号通路调控MMP-2蛋白的表达,进一步促进肺腺癌A549细胞的迁移。  相似文献

目的:探讨苦参碱对大鼠乳腺癌细胞荷瘤大鼠生长及其炎性因子与免疫功能的影响。方法:雌性Wistar大鼠60只,随机分为对照组(n=10),乳腺癌细胞荷瘤大鼠建模组(n=50)。建模大鼠再随机分为5组(n=10):模型组、苦参碱低(50 mg/kg)、中(100 mg/kg)、高(200 mg/kg)剂量组和香菇多糖(200 mg/kg)组。除对照组外,每只大鼠右腋皮下接种0.4 ml Walker 256乳腺癌细胞悬液(5×107 cells/ml)。按10 ml/kg的体积腹腔注射给药,2次/日,连续14 d。14 d后麻醉大鼠,腹主动脉取血,取瘤组织称取质量,计算抑瘤率;检测大鼠外周血IL-2、IFN-γ、IL-6、IL-10、TGF-β、CD3+、CD4+、CD8+、IgG、IgM、IgA水平。结果:苦参碱低、中、高剂量组和香菇多糖组大鼠瘤块平均质量分别为(4.99±0.93)g、(4.52±0.92)g、(4.22±1.18)g、(4.52±0.92)g,与模型组(6.62±1.20)g比较均明显降低(P<0.01)。苦参碱各剂量组与香菇多糖组比较,瘤块平均质量无明显差异(P>0.05)。苦参碱低、中、高剂量组和香菇多糖组抑瘤率分别为24.6%、31.7%、36.3%和27.9%,组间无统计学差异。模型组大鼠IL-2、IFN-γ、CD8+较对照组明显降低(P<0.01);而IL-6、IL-10、TGF-β、CD3+、CD4+、IgG、IgM、IgA较对照组明显升高(P<0.01)。苦参碱低、中、高剂量组和香菇多糖组IL-2、IFN-γ、CD8+较模型组显著升高(P<0.01,P<0.05);而IL-6、IL-10、TGF-β、CD3+、CD4+、IgG、IgM、IgA较模型组明显降低(P<0.01,P<0.05)。与香菇多糖组比较,苦参碱低、中剂量组IgM、IgA明显升高(P<0.01,P<0.05);高剂量组IL-2和IFN-γ、IgA水平明显升高(P<0.01,P<0.05);低剂量组IFN-γ水平明显降低(P<0.05);高剂量组IL-10、CD4+水平明显降低(P<0.01,P<0.05)。结论:苦参碱具有明显的抑制肿瘤生长的作用,其抑瘤机制与其提高细胞和体液免疫、减轻炎症反应有关。  相似文献

目的:探讨菊花总黄酮对小儿RSV感染A549细胞诱导RANTES及MCP-1释放作用影响。方法:实验分为细胞对照组,病毒对照组,菊花总黄酮组和病毒唑组。在Hep-2细胞和A549细胞分别加入菊花总黄酮和病毒唑的含药维持液,测定上述两种药物的最大无毒浓度;RSV病毒感染Hep-2细胞,观察药物对RSV的病毒抑制作用;RSV感染A549细胞,ELISA法测细胞趋化因子RANTES及MCP-1含量。结果:菊花总黄酮50%有效率优于病毒唑组,差异有统计学意义(P0.05);RANTES及MCP-1释放抑制作用比较中,菊花总黄酮组RANTES、MCP-1明显降低,优于病毒唑组,差异有统计学意义(P0.05)。结论:菊花总黄酮能够抑制RSV病毒活性,明显降低A549细胞释放RANTES、MCP-1,缓解患儿的呼吸道症状,对临床具有指导意义,值得临床推广。  相似文献

采用慢病毒载体质粒PLJM1将NapsinA基因转染到人肺腺癌细胞——A549细胞中,获得稳定表达Napsin A蛋白的特性并鉴定,通过转化生长因子-β1刺激A549细胞发生上皮-间质转化,体外构建上皮-间质转化模型并鉴定。MTT法检测转基因前后A549细胞在上皮-间质转化过程中生长速率的变化;流式细胞术检测其细胞周期的改变,最后予Western blot检测黏着斑激酶的表达情况,探讨Napsin A基因对A549细胞在上皮-间质转化过程中增殖的影响及其机制。结果表明转染后的A549细胞表达Napsin A蛋白明显增加(P<0.01);A549细胞发生上皮-间质转化后细胞E钙蛋白表达下调(P<0.01),Ⅰ型胶原表达上调(P<0.01);转基因细胞在体外上皮-间质转化模型中增殖速度减慢(P<0.05),且细胞周期被阻滞在G_1期(P<0.01),其表达整合素信号传导通路的基础分子——黏着斑激酶的量显著下降(P<0.01)。提示Napsin A基因可以抑制A549细胞在上皮-间质转化过程中的进一步增殖,其机制可能与抑制整合素信号传导通路有关。  相似文献

目的:探讨组蛋白去乙酰化酶抑制剂(HDIs)trichostatinA(TSA)对非小细胞肺癌(NSCLC)A549细胞放疗敏感性的影响.方法:以5Gy γ-射线照射细胞,或同时以TSA处理细胞.采用MTT法检测细胞存活率,Armexin V-PI染色检测细胞凋亡,流式细胞仪检测caspase-3活性.结果:SGy γ-射线可轻度降低细胞存活率,仅有少量细胞发生凋亡,但同时以TSA和γ-射线处理细胞,细胞存活率明显下降,凋亡细胞显著增多,且明显提高活化的caspase-3水平.结论:TSA通过促进caspase-3激活增强A549细胞对γ-射线的敏感性.  相似文献

目的探讨西咪替丁对轮状病毒性肠炎患儿炎性因子水平的影响。方法选取2015年3月~2017年3月我院收治的轮状病毒性肠炎患儿104例,随机分为治疗组和对照组各52例,对照组在常规治疗基础上加用利巴韦林治疗,治疗组在常规治疗基础上加用利巴韦林联合西咪替丁治疗,比较两组临床疗效和炎性因子改善情况。结果治疗组的临床总有效率为94.44%,明显高于对照组的85.19%,两组比较差异有统计学意义(P0.05)。两组患儿治疗后炎性因子水平均明显降低,与治疗前比较差异有显著统计学意义(P0.05);而且治疗后两组患儿的炎性因子水平比较,治疗组明显低于对照组,差异有统计学意义(P0.05)。结论西咪替丁可明显降低轮状病毒性肠炎患儿炎性因子水平,改善患儿临床症状,提高临床治疗效果,值得临床推广。  相似文献

目的:探讨基于RNA干扰CMTM7对肺腺癌A549细胞凋亡的影响。方法:选择肺腺癌A549细胞株分为si RNA组、阴性对照组与空白对照组,在对数生长的A549细胞中转染RNA干扰CMTM7载体、脂质体空载体和不进行转染,观察A549细胞的生长、凋亡与细胞周期状况。结果:MTT实验显示si RNA转染组的抑制率明显高于阴性对照组和空白对照组(P0.05),阴性对照组与空白对照组的对比无明显差异(P0.05)。流式细胞术实验表明si RNA转染组的细胞凋亡率明显高于阴性对照组和空白对照组(P0.05),阴性对照组与空白对照组的对比无明显差异(P0.05)。流式细胞术实验表明si RNA转染组的G0/G1期细胞数目较阴性对照组和空白对照组增多明显(P0.05),同时si RNA转染组的S、G2/M期细胞数目较阴性对照组和空白对照组明显减少(P0.05),阴性对照组和空白对照组对比差异无统计学意义(P0.05)。结论:RNA干扰CMTM7能够促进肺腺癌A549细胞凋亡,抑制肿瘤细胞的生长,其作用机制可能通过干扰细胞周期而实现。  相似文献

观察褪黑素以及褪黑素联合顺铂是否可以抑制A549细胞增殖并促进细胞凋亡,探讨ERK1/2MAPK信号通路在这一过程中的作用,为肺腺癌的研究提供实验依据。用不同浓度的褪黑素、不同浓度的顺铂、褪黑素联合顺铂刺激体外培养的肺腺癌细胞系A549细胞,MTT法检测各组细胞的活性,运用Real-time RT-PCR检测凋亡相关基因Bcl-2、Bax、p53的表达水平,Western-blot检测ERK1/2MAPK的活性变化。结果:1)单纯使用褪黑素和顺铂均可抑制A549细胞增殖,褪黑素联合顺铂时,抑制作用更加显著,一定浓度的褪黑素可降低顺铂的使用浓度;2)未经处理的A549细胞Bcl-2/Bax很高,p53基因表达较低,经褪黑素以及褪黑素联合顺铂刺激后A549细胞中Bcl-2/Bax明显下降,p53基因表达明显升高,并有浓度依赖;3)经褪黑素和顺铂刺激后A549细胞中磷酸化ERK1/2MAPK水平明显降低。褪黑素联合顺铂可抑制A549细胞增殖并且诱导凋亡相关基因表达,褪黑素和顺铂对A549细胞的抑制作用可能与ERK1/2MAPK信号通路有关。  相似文献

Esterase D (ESD) is a nonspecific esterase widely distributed in various organisms. ESD plays an important role in regulating cholesterol efflux, inhibiting viral replication and lung cancer growth. MT2A (metallothionein 2A) is the most important isoform of metallothionein (MTs) in human and high expression of MT2A in tumors represents poor prognosis and metastatic behavior. However, there are no reports about the molecular mechanism of ESD in the regulation of tumor metastasis. In this study, we found for the first time that activation ESD promoted its interaction with MT2A and decreased the protein level of MT2A, which resulting in the concentration of free zinc ions up-regulated, and inhibited the migration of A549 lung cancer cells in vitro.  相似文献

Bcl-2 small hairpin RNAs enhance radiation-induced apoptosis in A549 cells   总被引:2,自引:0,他引:2  
Bcl-2, a prominent member of the family of proteins, is responsible for dys-regulation of apoptosis and resistance to chemotherapy and radiotherapy. This study investigated whether small hairpin RNA (shRNA) targeting Bcl-2 could render A549 cells more susceptible to gamma radiation-induced apoptosis. Recombinant Bcl-2 shRNAs expression vector were transfected into A549 cells with Lipofectamine 2000. Transfected cells were screened in 800 mg/ml G418 screening medium, and after stable transfection, silencing was examined. Expression of the Bcl-2 protein was assayed using Western blot in A549 cells. Inhibition of cell growth was assessed by a MTT assay. Apoptosis was determined by morphological observation and flow cytometry. Expression levels of Bcl-2 protein from A549 cells decreased after stable transfection with Bcl-2 shRNAs. No differences in Bcl-2 protein levels between control shRNA group and untreated cells were noted. After stable transfection with Bcl-2 shRNAs the viability of cells was less than after stable transfection with those with control shRNAs and untransfected A549, respectively (P<0.05). Control shRNA had no significant effect on growth of cells. Radiation significantly inhibited the growth of cells stably transfected with Bcl-2 shRNA (P<0.05). No difference in survival between the cells with control shRNA and untransfected cells was noted. Using Giemsa staining, cells stably transfected with Bcl-2 shRNA combined with radiation at 48 h displayed changes of apoptosis. After treatment with radiation apoptotic rates of the A549 cells stably transfected with Bcl-2 shRNA significantly increased (P<0.05), compared with the cells with control shRNA and untransfected cells. shRNAs against the Bcl-2 mRNA increases radiation-induced apoptosis in A549 cells.  相似文献

目的: 探讨厚朴酚与吉非替尼协同影响非小细胞肺癌A549细胞的作用。方法: 以浓度为6.25~500 μmol/L厚朴酚、0.625~100 μmol/L吉非替尼分别处理A549细胞24 h,CCK-8实验检测细胞活力 (n=3),选24 h及100 μmol/L厚朴酚与5 μmol/L吉非替尼作后续处理(n=3);采用对照组、厚朴酚组、吉非替尼组和厚朴酚+吉非替尼组的析因分析设计;克隆形成检测细胞增殖;蛋白印迹测蛋白表达;流式细胞术检测细胞凋亡及分选CD44+和CD133+细胞。结果: 与对照组比,厚朴酚和吉非替尼组的克隆形成率显著降低(P<0.05);凋亡率显著升高(P< 0.05);CD44+和CD133+细胞数量显著减少(P<0.05);Ki67和PCNA及干细胞标记蛋白SOX2和OCT4表达显著下调(P<0.05);Bax/Bcl-2表达比例显著上调(P<0.05)。与厚朴酚组或吉非替尼组比较,厚朴酚+吉非替尼组进一步促进了上述改变(P<0.05),且凋亡率、Bax/Bcl-2、SOX2和OCT4等指标都存在厚朴酚和吉非替尼的交互作用(P< 0.05)。结论: 厚朴酚与吉非替尼促进A549细胞凋亡和抑制其干细胞样特性,且联合用药效果优于单一给药。二者对A549细胞的抑癌作用有交互影响。  相似文献

We isolated a stem cell subpopulation from human lung cancer A549 cells using FACS/Hoechst 33342. This side population (SP), which comprised 24% of the total cell population, totally disappeared after treatment with the selective ABCG 2 inhibitor fumitremorgin C. In a repopulation study, isolated SP and non-SP cells were each able to generate a heterogeneous population of SP and non-SP cells, but this repopulation occurred more rapidly in SP cells than non-SP. An MTT assay and cell cycle distribution analysis reveal a similar profile between SP and non-SP groups. However, in the presence of doxorubicin (DOX) and methotrexate (MTX), SP cells showed significantly lower Annexin V staining when compared to non-SP cells. Taken together, these results demonstrate that SP cells have an active regeneration capacity and high anti-apoptotic activity compared with non-SP cells. Furthermore, our GeneChip® data revealed a heightened mRNA expression of ABCG2 and ABCC2 in SP cells. Overall these data explain why the SP of A549 has a unique ability to resist DOX and MTX treatments. Therefore, we suggest that the expression of the ABCG2 transporter plays an important role in the multidrug resistance phenotype of A549 SP cells.  相似文献

Proteins destined for degradation by the ubiquitin-proteasome system are labelled with a 76-amino acid peptide, ubiquitin, through a series of conjugation steps by the E1, E2 and E3 enzymes respectively. Ubiquitin carboxy-terminal hydrolase 37 (UCH37) belongs to the UCH proteases family that deubiquitinates ubiquitin-protein conjugates in the ubiquitin-proteasome system. However, it is few reports about the relationship between UCH37 and apoptosis. In order to clarify the role of UCH37 on apoptosis, the A549 cells were chosen for this study. We transfected UCH37 siRNA and pcDNA3.1-UCH37 plasmid into A549 cells, respectively. Using MTT assay, Western blot, Hoechst 33342 staining assay and flow cytometry, we found that silencing of UCH37 in A549 cells induced apoptosis. The ratio of Bax/Bcl-2 was higher in silencing of UCH37 than that in control group after silencing of UCH37 in A549 cells. Meanwhile, experiments with the A549 cell line disclose that silencing of UCH37 could induce efficiently A549 cell apoptosis through activation of caspase-9 and caspase-3. On the other hand, over-expression of UCH37 led to the opposite effect. Hence, UCH37 might play an important role in apoptotic through altering Bax/Bcl-2 ratio and enzymatic activities of caspase-9 and caspase-3.  相似文献

Epithelia in the human airways, from the nasal aperture to the alveoli, are covered in a protective film of fluid containing a number of antimicrobial proteins. Defensins are single-chain, strongly cationic peptides and are one of the most extensively studied classes of antimicrobial peptides. Moxifloxacin (MXF) is a fluoroquinolone that acts against both Gram positive and Gram negative bacteria. In this study, we evaluated the effects of HBD2, MXF and the association MXF/HBD2 on some cytokines and on the ICAM-1 expression in LPS-stimulated A549 cells. Our results suggest that by lowering the epithelial cell-derived IL-1β, IL-6, IL-8 and ICAM-1 expression, the MXF/HBD2 association interferes with the multifunctional cytokine network evolving during inflammatory processes of the respiratory tract; this anti-inflammatory potential could be of great value in the treatment of inflammatory disorders.  相似文献

Recent studies found that peroxiredoxin-I (Prx-I) is secreted from A549 cells although it does not contain a signal peptide and is known to be a cytosolic protein. Transforming growth factor-beta1 (TGF-beta1) treatment dramatically enhanced Prx-I secretion from A549 cells, and this effect was not inhibited by brefeldin A. Further investigation revealed that A549 cells constitutively secrete TGF-beta1. Furin, a TGF-beta1-converting enzyme, was also highly activated in A549 cells. Ectopic expression of alpha(1)-antitrypsin Portland (alpha(1)-PDX), a potent furin inhibitor, blocked both TGF-beta1 activation and Prx-I secretion. Our findings collectively suggest that non-classical secretion of Prx-I is induced by TGF-beta1, which is constitutively activated by furin in A549 cells.  相似文献

目的:探讨纳米二氧化硅(Nano-Si O2)对子代雌性小鼠动情周期的影响。方法:选择8周龄的ICR孕鼠6只,随机平均分为两组,一组在E5.5~E11.5时隔天注射纳米二氧化硅,另外一组注射PBS,每组取子代雌性小鼠各3只,待其自然生长到7周龄时,每日采用巴氏染色法检查其动情周期。结果:与注射PBS的子代雌性小鼠相比,注射Nano-Si O2的子代雌性小鼠的动情周期的总时间无明显差异(P0.05),动情前期的时间明显缩短,动情后期的时间明显延长(P0.05),动情期虽然缩短,但差异不显著(P0.05)。结论:孕期暴露于Nano-Si O2未导致子代雌性小鼠动情周期的紊乱,但对其生殖功能可能有一定的影响。  相似文献

Paraquat (PQ) is a well-known pneumotoxicant that exerts its toxic effect by elevating intracellular levels of superoxide. In addition, production of pro-inflammatory cytokines has possibly been linked to PQ-induced inflammatory processes through reactive oxygen species (ROSs) and nitric oxide (NO). However, the role of NO in PQ-induced cell injury has been controversial. To explore this problem, we examined the effect of NO on A549 cells by exposing them to the exogenous NO donor NOC18 or to cytokines; tumor necrosis factor-α, interleukin-1 β and interferon-γ, as well as PQ. Although the exogenous NO donor on its own had no effect on the release of lactate dehydrogenase (LDH), remarkable release was observed when the cells were exposed to high concentrations of NOC18 and PQ. This cellular damage caused by 1 mM NOC18 plus 0.2 mM PQ was ascertained by phase contrast microscopy. On the other hand, NO derived from 25–50 μM NOC18 added into the medium improved the MTT reduction activity of mitochondria, suggesting a beneficial effect of NO on the cells. Incubation of A549 cells with cytokines increased in inducible NO synthase (iNOS) expression and nitrite accumulation, resulting in LDH release. PQ further potentiated this release. The increase in nitrite levels could be completely prevented by NOS inhibitors, while the leakage of LDH was not attenuated by the inhibition of NO production with them. On the other hand, ROS scavenging enzymes, superoxide dismutase and catalase, inhibited the leakage of LDH, whereas they had no effect on the increase in the nitrite level. These results indicate that superoxide, not NO, played a key role in the cellular damage caused by PQ/cytokines. Our in vitro models demonstrate that NO has both beneficial and deleterious actions, depending on the concentrations produced and model system used.  相似文献
