This study evaluated fertility in swamp buffalo after synchronization of ovulation combined with fixed time artificial insemination. At the start of the study, designated day 0, from a group of 98 female Thai swamp buffalo, 55 buffalo (heifers n° = 20 and cows n° = 35) were selected to be synchronized with GnRH (Day 0) followed by PGF2alpha (Day 7) and a second treatment with GnRH (Day 9). All buffalo were inseminated at two fixed times 12 h and 24 h after the second injection of GnRH (Ovsynch+TAI group); a second group of 43 buffalo (heifers n° = 19 and cows n° = 24) were not treated and were artificially inseminated (AI) at natural estrus (AI group). Blood samples were taken 22 days after insemination to evaluate progesterone plasma levels. In the Ovsynch+TAI group, overall conception rate (CR; i.e. the number of cows with progesterone >4.0 ng/ml on day 22 after AI divided by the number of animals inseminated), was 38.1% and overall pregnancy rate (PR; i.e. the number of cows that were pregnant at day 50-60 after insemination divided by the number of animals inseminated), was 32.7%. In the AI group overall CR and PR was 34.9%.Within the Ovsynch+TAI group, CR and PR were reduced (P < 0.05) in heifers compared with cows (CR 15.0% vs. 51.4% for heifers and cows, respectively; PR 15.0% vs. 42.9% for heifers and cows, respectively). Within the AI group the efficacy of treatment was similar between heifers and cows (CR and PR 31.6% for heifers and 37.5% for cows).In conclusion, this study indicates that in swamp buffalo it is possible to synchronize ovulation and use timed artificial insemination with the Ovsynch+TAI protocol.  相似文献

An experiment was conducted to test the hypothesis that sterile mounts around insemination improves pregnancy rate to artificial insemination (AI) and to define the effects of age, season, time to complete AI and time of day of insemination. A total of 178 Simbrah females were randomly assigned by calving date and body condition to one of three treatments during two consecutive years: (1) mating stimuli with a sterile bull at the time the cows were detected in estrus; (2) mating stimuli immediately after completing AI; (3) without mating stimuli. All cows and heifers were maintained under the same conditions of handling and feeding within the two breeding seasons (winter 1995 and summer 1996). Vasectomized bulls were used for the sterile mounts. Cows and heifers that were given a sterile mount at the time of detection of estrus, had an increased pregnancy rate (60.0%) compared with females given a sterile mount after completing AI (25.4%) or females without the sterile mount (35.6%) (P < 0.01). Age, season, time to complete AI and time of day of AI were all non-significant (P > 0.05). Therefore, there is a biostimulatory effect of mating at the time beef cattle females are detected in estrus, on pregnancy rates to AI.  相似文献

Two experiments (Experiment I, n=12 Holstein-Friesian heifers; Experiment II, n=8 Jersey cows) were conducted to investigate the pathogenesis of bovine pestivirus-induced ovarian dysfunction in cattle. In both experiments the cattle were superovulated with twice daily injections of a porcine pituitary extract preparation of follicle stimulating hormone (FSH-P), for 4 days commencing on Day 10+/-2 after a presynchronised oestrus. The heifers received a total dose of 30 mg and the cows 32 mg of FSH-P. Prostaglandin F(2alpha) (PGF(2alpha)) was administered 48 h after commencement of superovulation and all cattle were artificially inseminated (AI) between 48 and 66h after PGF(2alpha) treatment. In both experiments bovine pestivirus seronegative cattle (Experiment I, n=6; Experiment II, n=4) were inoculated intranasally with an Australian strain of non-cytopathogenic bovine pestivirus (bovine viral diarrhoea virus Type 1) 9 days prior to AI. Bovine pestivirus infection was confirmed by seroconversion and/or virus isolation in all of the inoculated cattle, consistent with a viremia occurring approximately between Day 5 prior to AI and the day of AI. Ovarian function was monitored in both experiments by daily transrectal ultrasonography and strategic blood sampling to determine progesterone, oestradiol-17beta, luteinising hormone (LH) and cortisol profiles. Non-surgical ova/embryo recovery was performed on Day 7 after AI. In Experiment II half the cattle were slaughtered on Day 2 and the remainder on Day 8 after AI, and the ovaries submitted for gross and histopathological examination including immunohistochemistry to demonstrate the presence of bovine pestivirus antigen. In both studies, comparisons were made between infected and confirmed uninfected (control) animals. Overall the bovine pestivirus infected cattle had significantly lower (P<0.05) ova/embryo recovery rates compared to the control cattle. There was evidence of either an absence (partial or complete) of a preovulatory LH surge or delay in timing of the LH peak in the majority (90%) of infected heifers and cows, and histologically, there was evidence of non-suppurative oophoritis with necrosis of granulosa cells and the oocyte in follicles from the infected cows. By contrast only 20% of the control heifers and cows had evidence of absence of a pre-ovulatory LH surge. These experiments collectively demonstrate that bovine pestivirus infection during the period of final growth of preovulatory follicles may result in varying degrees of necrosis of the granulosa cells with subsequent negative effects on oestradiol-17beta secretion which in turn negatively affects the magnitude and/or timing of the preovulatory LH surge.  相似文献

The effects of fenprostalene, cloprostenol sodium and prostaglandin F(2) alpha (PGF(2alpha)) on estrus, conception rate, pregnancy rate, and the interval from Day 1 of the breeding season to calving were studied on 135 purebred Angus cows and heifers. The cows and heifers were randomly allotted within age to the three estrus synchronization treatments and a control group. The calving percentages (for cows and heifers combined) that resulted from artificial insemination (AI) were 32.3, 31.4, 43.6, and 51.1% for the control, fenprostalene, cloprostenol sodium, and PGF(2alpha) groups, respectively. The calving percentage during the AI period by ages of dam at breeding were 54.2% for yearling heifers, 30.5% for two-year-olds, 47.6% for three-year-olds, and 26.1% for four-year-old or older cows. The percentage of cows and heifers detected in estrus and the percentage that conceived after the first injection for control, fenprostalene, cloprostenol sodium, and PGF(2alpha) groups were 51.6 and 22.3%, 59.3 and 32.1%, 76.8 and 44.1%, and 66.6 and 50.2%, respectively. The intervals from Day 1 of the breeding season to calving and from Day 1 of the calving season within each treatment to the birth of each calf were control, 285.9 and 23.8 d; fenprostalene, 283.6 and 13.4 d; cloprostenol sodium, 285.5 and 6.5 d; and PGF(2alpha), 284.0 and 11.1 d.  相似文献

During an attempt to accumulate 40 Bovine Viral Diarrhea virus (BVDV) seronegative cows for breeding and for intramuscular infection on Day 21 of gestation, a persistently infected cow was inadvertently included among the first group of seronegative animals assembled. This animal proceeded to infect all seronegative animals added to the experimental herd. Since the addition of cows was gradual and they were bred as they arrived, a group of cows was bred before they seroconverted, another group was inseminated during seroconversion and a third group was seropositive when bred. First service conception rates were 22.2, 44.4 and 78.6%, respectively. The difference between 22.2 and 78.6% conception rates was significant (P < 0.05). Thirty cows were diagnosed pregnant at 21 d after service on the basis of nonreturn to estrus, presence of a palpable corpus luteum and high serum progesterone concentration. Seventeen of these received cytopathic BVDV intramuscularly and 13 cows served as controls. All control cows and 9 of 17 (52.9%) virus-treated cows had normal fetuses and placentas at slaughter on Day 70. Six pregnancies were lost between 23 and 33 d after insemination and two were lost between 35 and 40 d after insemination. Noncytopathic BVDV was demonstrated in all eight of these cows either in the buffy coat or in tissues, despite the presence of serum neutralizing antibodies.  相似文献

The objective was to determine whether timed artificial insemination (TAI) 56 h after removal of a Controlled Internal Drug Release (CIDR, 1.38 g of progesterone) insert would improve AI pregnancy rate in beef heifers compared to TAI 72 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol. Angus cross beef heifers (n = 1098) at nine locations [WA (5 locations; n = 634), ID (2 locations; n = 211), VA (one location; n = 193) and WY (one location; n = 60)] were included in this study. All heifers were given a body condition score (BCS; 1-emaciated; 9-obese), and received a CIDR insert and 100 μg of gonadorelin hydrochloride (GnRH) on Day 0. The CIDR insert was removed and two doses of 25 mg of dinoprost (PGF) were given, first dose at CIDR insert removal and second dose 6 h later, on Day 5. A subset of heifers (n = 629) received an estrus detector aid at CIDR removal. After CIDR removal, heifers were observed thrice daily for estrus and estrus detector aid status until they were inseminated. Within farm, heifers were randomly allocated to two groups and were inseminated either at 56 h (n = 554) or at 72 h (n = 544) after CIDR removal. All heifers were given 100 μg of GnRH at AI. Insemination 56 h after CIDR insert removal improved AI pregnancy rate compared to insemination 72 h (66.2 vs. 55.9%; P < 0.001; 1 - β = 0.94). Locations, BCS categories (≤ 6 vs. > 6) and location by treatment and BCS by treatment interactions did not influence AI pregnancy rate (P > 0.1). The AI pregnancy rates for heifers with BCS ≤ 6 and > 6 were 61.8 and 60.1%, respectively (P > 0.1). The AI pregnancy rates among locations varied from 54.9 to 69.2% (P > 0.1). The AI pregnancy rate for heifers observed in estrus at or before AI was not different compared to heifers not observed in estrus [(65.4% (302/462) vs. 52.7% (88/167); P > 0.05)]. In conclusion, heifers inseminated 56 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol had, on average, 10.3% higher AI pregnancy rate compared to heifers inseminated 72 h after CIDR insert removal.  相似文献

The objectives of the present study were to determine the effects of resynchronization with GnRH on Day 21 after artificial insemination (AI) on pregnancy rate and losses of pregnancy in lactating dairy cows. Holstein cows (n=585) on two dairy farms were assigned to one of two treatments in a randomized complete block design. On Day 21 after a pre-enrollment AI, animals assigned to the resynchronization (RES) group received 100 microg of GnRH i.m., whereas animals in the control (CON) group received no treatment. All animals were examined ultrasonographically on Days 21 and 28 after AI, and blood samples were taken for progesterone measurement on Day 21. Pregnancy was diagnosed on Day 28 and reconfirmed 14 days later. Nonpregnant cows on Day 28 were inseminated using timed AI after the completion of the Ovsynch protocol 10 and 17 days after enrollment in the study for RES and CON groups, respectively. Progesterone concentration > or =2.35 ng/ml was used as an indicator of pregnancy on Day 21. For RES and CON cows, pregnancy rate at Days 21 (70.9% versus 73.0%, P<0.56), 28 (33.1% versus 33.6%; P<0.80) and 42 (27.0% versus 26.8%; P<0.98) after the pre-enrollment AI did not differ. Administration of GnRH on Day 21 after AI had no effect on pregnancy loss in RES and CON groups from days 21 to 28 (53.2% versus 53.5%; P<0.94) and days 28 to 42 (17.9%; P<0.74) after AI. Pregnancy rate after the resynchronization period was similar for both treatment groups. Resynchronization with GnRH given on Day 21 after AI for initiation of a timed AI protocol prior to pregnancy diagnosis does not affect pregnancy rate and pregnancy loss in lactating dairy cows.  相似文献

The objective was to compare the probability of pregnancy after fixed-time insemination in cows diagnosed as non-pregnant and re-inseminated following the Ovsynch protocol, with or without exogenous progesterone. Cows (n=415) used in this study originated from 25 farms. Upon diagnosis of non-pregnancy between 30 and 60 days after AI, cows were randomly assigned to receive either a progesterone releasing intravaginal device (PRID; n=208) or a placebo intravaginal device (PID; n=207). All cows received GnRH at enrollment (Day 0), PGF(2alpha) concurrent with intravaginal device removal 7 days later, GnRH on Day 9 and fixed-time insemination 16h later (Day 10). Cows observed in estrus prior to Day 7, had the device removed and were inseminated. Ovaries were examined by transrectal palpation at the time of enrollment and the prominent structures were assessed and recorded. Body condition score, lameness status, interval from previous insemination, and times bred at enrollment were recorded. At intravaginal device removal, the occurrence and intensity of vaginitis was determined according to the amount of debris on the device. Overall, the intravaginal device retention rate was 91%. A total of 5.2% of PID-treated cows and 2.9% of PRID-treated cows were detected in estrus within the 7 days treatment period. Pregnancy status was diagnosed between 30 and 56 days after insemination and all cows were followed for a minimum of 150 days after enrollment. Approximately 28% of cows had evidence of mild vaginitis in response to the intravaginal device, whereas 6% of cows had copious debris associated with the intravaginal device at removal. The probability of pregnancy after fixed-time insemination was 43.8% versus 34.9% in PRID-treated versus PID-treated animals. Exogenous progesterone provided through an intravaginal device to non-pregnant cows that had not displayed estrus improved the probability of pregnancy after fixed-time AI.  相似文献

Lactating Holstein dairy cows (n=1,533) were allocated to one of three treatment groups, with Group I (n=514) receiving 10 mug of a GnRH analogue (buserelin) at artificial insemination (AI) and Group II (n=503) receiving 10 mug of the same analogue at both the time of AI and at 12 days post AI. Herdmates in Group III (n=516) were inseminated on the same day and served as contemporary AI controls. The trial was conducted on five large dairy farms during the spring and summer months in Saudi Arabia. Pregnancy rates were determined by palpation per rectum between 33 and 50 days following AI. The first service pregnancy rate for the control cows (42.4%) was lower (P<0.05) than that for cows treated with the GnRH analogue at AI (48.8%) or for the combined treatment at AI and at Day 12 post AI (51.5%). No additive effect on the pregnancy rate was noted from the combined analog treatment. The overall increase in pregnancy rate from the analogue treatment at AI resulted from an 11% increase in pregnancy rate in first parity cows over that of contemporary controls (P<0.05) and a 14.7% increase in pregnancy for cows mated at 40 to 59 days post partum and treated with the analogue at AI over that of the corresponding controls (P<0.05). Two trials involving 85 heifers and 67 cows were conducted to determine the effect of estrous cycle stage at the time of Syncro-Mate-B((R)) (SMB) treatment on interval to estrus following implant removal and on conception rate at the synchronized estrus. In Trial 1, 57 beef and 28 dairy heifers were treated with SMB on each representative day of a 22-d estrous cycle (estrus = Day 0). Beef heifers were artificially inseminated approximately 48 h after implant removal, whereas dairy heifers were inseminated 0 to 12 h after detection of estrus. Inseminations were scored by the inseminator according to their difficulty. Interval to the onset of estrus was not different between heifers treated early ( Day 11) in the cycle (35.2 +/- 7.2 h). Conception rate at the synchronized estrus was slightly higher in early-cycle heifers (22 47 = 47% ) compared to late-cycle heifers (14 38 = 37% , P = 0.2). Heifers that were difficult to inseminate had lower (P < 0.01) conception rates (2 11 = 18% ) at the synchronized estrus than heifers considered normal (21 51 = 41% ) or easier than normal to inseminate (13 23 = 57% ). In Trial 2, of the 131 beef cows synchronized, 67 that were estimated to be either early or late in the estrous cycle by progesterone analysis were utilized. Cows were treated with SMB and inseminated without regard to estrus 48-h after implant removal. Inseminations were scored as in Trial 1. Calves were separated from cows from the time of implant removal to insemination. Conception rate was higher (P < 0.05) in cows treated with SMB early ( Day 11, 16 35 = 46% ). Cows that were difficult to inseminate had a lower (P < 0.01) conception rate (0 8 = 0% ) than cows that were normal (43 94 = 46% ) or easier than normal to inseminate (13 29 = 45% ).  相似文献

Two trials were conducted to evaluate the efficacy of short-term progestin administration to resynchronize the second estrus after artificial insemination in yearling beef heifers. In Trial 1 crossbred yearling heifers (n = 208) were synchronized with Syncro-Mate-B (SMB) and artificially inseminated (AI) between 48 and 54 h following implant removal. Implant removal is defined as Day 1. Following AI, the heifers were randomly assigned to 1 of 2 experimental groups. Group 1 heifers were fed melengestrol acetate (MGA) daily from Day 17 to 21 at a rate of 0.5 mg/head, while Group 2 control received no exogenous progestin during this period. Synchrony of estrus was defined as the 3-d period in which the highest number of heifers expressed behavioral estrus in each group. There was no difference (P < 0.05) in the pregnancy rate during the second estrus due to MGA supplementation. More MGA-treated heifers (P < 0.01) expressed estrus in a 3-d period than the controls. In Trial 2, yearling heifers (n = 108) were synchronized with 2 injections of PGF(2alpha) (second PGF(2alpha) injection is designated as Day 1) administered 14 d apart with AI 12 h after the onset of behavioral estrus. The heifers were then randomly assigned to 1 of the following 3 treatment groups after initial AI: 1) MGA fed at 0.5 mg/head daily from Days 17 to 21; 2) norgestomet administered in 6.0-mg implants from Days 17 to 21; 3) untreated control heifers. Blood samples were collected on Day 21 and analyzed for progesterone (P(4)). Elevated P(4) (> 1 ng/ml) on Day 21 indicated pregnancy to the first insemination. Synchrony among the 3 groups of heifers was similar (P > 0.10); however, the second estrus was less (P < 0.05) variable in the MGA and norgestomet treated heifers. During the resynchronized second estrus, conception rates were not affected by progestin treatment (MGA 40%, norgestomet 64%, and control 62%; P > 0.10). However, a proportion of heifers treated MGA 10% 4 36 and norgestomet 3% 1 36 expressed behavioral estrus during second estrus even though they were diagnosed as pregnant from first service by elevated P(4) levels on Day 21. We conclude that short-term use of progestin from Days 17 to 21 following AI causes closer synchrony of estrus; however, inseminating pregnant heifers that exhibit behavioral estrus may cause abortion.  相似文献

This study investigated the hypothesis that a reduction in submission rates at a resynchronised oestrus is not due to the resynchrony treatment involving intravaginal progesterone releasing devices (IVDs) and oestradiol benzoate (ODB) but is associated with artificial insemination (AI) at the first synchronised oestrus. In Experiment 1, cows were synchronised for first oestrus with IVDs, with ODB administered at the time of device insertion (Day 0, 2 mg IM) and 24 h after removal (Day 9, 1 mg IM) and PGF(2alpha) injected at the time of device removal. Cows were then either inseminated (I) for 4 days or not inseminated (NI) following detection of oestrus (first round of AI). Every animal was resynchronised for a second round of AI by reinsertion of IVDs on Day 23 with administration of ODB (1 mg IM) at the time of insertion as well as 24 h after removal (Day 32). Cows detected in oestrus and inseminated for 4 days at the second round of AI were resynchronised for a third round by repeating the resynchrony treatment starting on Day 46 and inseminating cows on detection of oestrus for 4 days. In Experiment 2 the same oestrous synchronisation and resynchronisation treatments were used, but the timing of treatments differed. The cows had their cycles either presynchronised (treatment start Day -23) without AI and then resynchronised, starting on Day 0, for the first round of AI for AI at detected oestrus for 4 days, or they were synchronised (treatment start Day 0) for the first round of AI. In Experiment 1, 91.4% (64/70) and 92.6% (63/68) (P = 0.79) of cows in the I and NI treatments, respectively, were detected in oestrus after the initial synchronisation. At the second round of AI, submission rates for insemination were lower in the I group compared to the NI cows (74.5%, 35/47 versus 92.6%, 63/68, respectively; P = 0.007). Pregnancy rates (proportion treated that were classified as becoming pregnant) in I and NI cows 4 weeks (61.4%, 43/70 versus 63.2%, 43/68) and 7 weeks (77.1%, 54/70 versus 69.1%, 47/68) after the AI start date (AISD) did not differ significantly between treatments. In Experiment 2, presynchronisation and then resynchronisation of oestrous cycles before the first round of AI did not affect oestrous detection rates at the first round of AI (100%, 44/44 versus 98.0%, 50/51; P = 0.54), or pregnancy rates 1 week (63.6%, 28/44 versus 60.8%, 31/51; P = 0.70), 4 weeks (72.7%, 32/44 versus 76.5%, 39/51; P = 0.76) and 7 weeks (81.8%, 36/44 versus 88.2%, 45/51; P = 0.40) after AISD compared to cows that had their cycles synchronised for the first round of AI. These findings support our hypothesis that a reduction in submission rates at a resynchronised oestrus is associated with AI at the first synchronised oestrus and not due to a resynchrony treatment involving IVDs and ODB. This study supports the concept that early embryonic loss following AI at a synchronised oestrus could cause a reduction in submission rates following resynchronisation of oestrus, although investigation of the effect of passing an AI catheter or semen components were not studied per se.  相似文献

It was hypothesized that gonadotropin-releasing hormone (GnRH) treatment at the time of insemination and 12 days later increases conception rates. The aim of the present study was to evaluate the effects of GnRH treatment at the time of insemination or at the time of insemination and 12 days later on reproductive performance during the warm season in high producing dairy cows. The effect of GnRH treatment on the incidence of subsequent twin pregnancies and pregnancy losses was also evaluated. Data were analyzed using logistic regression methods. Of the entire series of 1289 AI, 373 (29%) resulted in pregnancy. Three study groups were established to evaluate the effects of treatment on the conception rate: control (untreated cows, n=431), GnRH-0 (cows receiving GnRH at AI, n=429) or GnRH-0+12 (cows receiving GnRH at AI and at AI+12 days, n=429). Conception rates were 20.6% (89/431), 30.8% (132/429) and 35.4% (152/429) for animals receiving no treatment, GnRH at AI, and GnRH at AI and 12 days later, respectively. Based on the odds ratio, the probability of pregnancy was 0.80 and 0.46 times less likely for cows receiving treatment GnRH-0 and no treatment, respectively, than for cows receiving treatment GnRH-0+12 (reference). Of the 373 pregnant animals, 326 (87.4%) bore singletons and 47 (12.6%) carried twins. The effects of treatment on the dependent variables: twin pregnancy, additional corpus luteum and pregnancy loss were analyzed. Pregnancy loss between 38 and 90 days after insemination was registered in 30 (8%) cows: 17 (5.2%) in single and 13 (27.7%) in twin pregnancies. Fifty-six (15%) cows had an additional corpus luteum. No pregnancy losses were recorded in these cows. Treatment had no effect on the twin pregnancy rate. The treatment GnRH at AI and 12 days later increased the chances of an additional corpus luteum by a factor 3.7 (using the control group as reference). In conclusion, our results support the hypothesis that GnRH treatment at the time of insemination and 12 days later increases the conception rate in high producing dairy cows during the warm season. Although lower than double treatment, strong benefits were also registered following a single GnRH treatment at insemination. Under these conditions, treatment fails to affect the twin pregnancy rate yet increases the incidence of an additional corpus luteum in pregnant cows.  相似文献

The effect of intravenous cloprostenol treatment at the time of insemination on reproductive performance was consecutively evaluated in three different subpopulations of high producing lactating dairy cows: Study (1) early postpartum synchronized and fixed-time inseminated (about 50 days in milk) cows (n = 379: 187 control and 192 treated cows); Study (2) presumed high fertility cows first inseminated between 90 and 120 days postpartum (n = 248: 124 control and 124 treated cows); and Study (3) heat stressed repeat breeder cows (n = 183: 93 control and 90 treated cows). Data were analyzed using multiple regression methods. Study 1: Parity (primiparous versus multiparous), milk production, body condition score at AI, insemination season (cool versus warm period) and treatment were included in the analysis as potential factors affecting ovulation, double ovulation, return to estrus, and pregnancy to first AI and to second AI (first AI plus return AI) rates. Logistic regression analysis indicated that the final model for ovulation rate only included the interaction (P = 0.002) between insemination season and treatment. Cloprostenol treatment at insemination led to a 4.2-fold increase in the ovulation rate in cows inseminated during the warm period. There were no significant effects of treatment, parity, milk production, body score or the insemination season on the return to estrus rate. The only variables included in the final logistic model for double ovulation and pregnancy to first AI rates were treatment and season, respectively. Treatment led to a 2.6-fold increase (P = 0.001) in the double ovulation rate, whereas cows inseminated in the warm period were 2.1 times less likely (P = 0.007) to become pregnant at first AI compared to those inseminated in the cool season. The variables included in the final logistic model for the pregnancy rate to second AI were treatment and season. Cloprostenol given at AI increased the risk of pregnancy 1.9 times (P = 0.002), and cows inseminated during the warm season were two times less likely to become pregnant (P = 0.003). No significant interactions were found among these three dependent variables (double ovulation and pregnancy to first and to second AI rates). Study 2: Logistic regression analysis of all the dependent variables: return to estrus, and pregnancy to first and to second AI (first AI plus return to AI) rates indicated no significant effects of treatment, parity, days in milk, milk production or body score at AI. No significant interactions were found. Study 3: The final model for the pregnancy rate only included the interaction between parity (primiparous versus multiparous) and treatment. Days in milk, milk production and insemination number showed no significant effect on pregnancy rate. Cloprostenol treatment at insemination increased the pregnancy rate in primiparous repeat breeder cows (odds ratio: 3.6). The treatment group and parity showed significant (P < 0.0001) interaction. This interaction suggests that cloprostenol treatment of primiparous cows at insemination might enhance pregnancy yet have no effect in multiparous cows. Our findings indicate that cloprostenol administered at insemination promotes ovulation and double ovulation in lactating dairy cows. Cloprostenol treatment showed no benefit in cows with acceptable reproductive performance, suggesting that cloprostenol treatment at AI may only be useful in cows in which stress factors affect ovulation and in repeat breeder cows.  相似文献

A method for timed artificial insemination (AI) that is used for beef cows, beef heifers, and dairy heifers employs progesterone-releasing inserts, such as the controlled internal drug release (CIDR; Zoetis, New York, NY, USA) that are left in place for 14 days. The 14-day CIDR treatment is a method of presynchronization that ensures that cattle are in the late luteal phase of the estrous cycle when PGF is administered before timed AI. The objective of this study was to test the effectiveness of the 14dCIDR-PGF program in postpartum dairy cows by comparing it with the traditional “Presynch-Ovsynch” (2xPGF-Ovsynch) program. The 14dCIDR-PGF cows (n = 132) were treated with a CIDR insert on Day 0 for 14 days. At 19 days after CIDR removal (Day 33), the cows were treated with a luteolytic dose of PGF, 56 hours later were treated with an ovulatory dose of GnRH (Day 35), and 16 hours later were inseminated. The 2xPGF-Ovsynch cows were treated with a luteolytic dose of PGF on Day 0 and again on Day 14. At 12 days after the second PGF treatment (Day 26), the cows were treated with GnRH. At 7 days after GnRH, the cows were treated with PGF (Day 33), then 56 hours later treated with GnRH (Day 35), and then 16 hours later were inseminated. There was no effect of treatment or treatment by parity interaction on pregnancies per AI (P/AI) when pregnancy diagnosis was performed on Day 32 (115/263; 43.7%) or Days 60 to 90 (99/263; 37.6%) after insemination. There was an effect of parity (P < 0.05) on P/AI because primiparous cows had lesser P/AI (35/98; 35.7%) than multiparous cows (80/165; 48.5%) on Day 32. Cows observed in estrus after the presynchronization step (within 5 days after CIDR removal or within 5 days after the second PGF treatment) had greater P/AI than those not observed in estrus (55/103; 53.4% vs. 60/160; 37.5%; observed vs. not observed; P < 0.01; d 32 pregnancy diagnosis). When progesterone data were examined in a subset of cows (n = 208), 55.3% of cows had a “prototypical” response to treatment (i.e., the cow had an estrous cycle that was synchronized by the presynchronization treatment and then the cow responded appropriately to the subsequent PGF and GnRH treatments before timed AI). Collectively, cows with a prototypical response to either treatment had 52.2% P/AI that was greater (P < 0.001) than the P/AI for cows that had a nonprototypical response (19%) (P/AI determined at 60–90 days of pregnancy). In conclusion, we did not detect a difference in P/AI when postpartum dairy cows were treated with 14dCIDR-PGF or 2xPGF2α-Ovsynch before timed AI. The primary limitation to the success of either program was the failure of the cow to respond appropriately to the sequence of treatments.  相似文献

One aim of this study was to compare the reproductive performance of cows and heifers when resynchronizing returns to estrus for a second insemination by treating with an intravaginal progesterone-releasing device (IVD) for 7 or 8d when estradiol benzoate (EB) was administered at the start of treatment and again 24h after device removal. An additional aim was to document the pattern of onset and characteristics of estrus with each resynchrony treatment. Lactating cows in three herds were synchronized for a first estrus and AI by treatment with an IVD for 8d, starting on Day 0, cloprostenol (0.5 mg im) at device removal and EB at device insertion (2.0 mg im) and 24h after removal (1.0 mg im). Cows were resynchronized for a second estrus starting on Day 23 by reinsertion of IVDs for 7 (IVD-7-EB; n=449) or 8d (IVD-8-EB; n=445) with EB (1.0 mg im) administered at device insertion and 24h after removal. Cows were resynchronized for a third estrus by administration of EB (1.0 mg im) on Day 46, but subsequent treatments (no further treatment, reinsertion of CIDR or administration of EB on Day 55) varied among herds as part of separate studies. Maiden heifers (7-Day, n=68; 8-Day, n=69) were similarly treated as cows in a separate herd, but doses of EB were always 1.0 mg im at device insertion and 0.75 mg im 24h after removal. Heifers were not resynchronized for a third estrus. Cattle were inseminated on detection of estrus at each synchronized estrus. Cumulative pregnancy rates 4 week (66.0%, 276/418 versus 59.1%, 247/418) and 7 week (72.7%, 304/418 versus 67.7%, 283/418) after the start of AI were greater (P<0.05) in the IVD-7-EB cows compared to the IVD-8-EB cows, respectively; this was associated with a 9% increase in conception rates at the second estrus (P=0.051) in the IVD-7-EB cows. Treatment did not significantly affect reproductive performance in heifers. Characteristics of estrus measured with radiotelemetry did not differ significantly between the two treatment groups, but more cows were detected in estrus 36 h after removal of IVDs in the IVD-8-EB cows compared to the IVD-7-EB cows (P<0.05). We concluded that reproductive performance in resynchronized dairy cows but not heifers was greater following resynchronization of estrous cycles after AI with an IVD for 7 compared to 8d when EB was injected at the start of treatment and 24h after device removal.  相似文献

Lactating Holstein cows were used to determine if pregnancy rate from embryo transfer (n = 113) differed from contemporary control cows (n = 524) that were artificially inseminated (AI). Holstein heifers (n = 55) were superovulated with FSH-P (32 mg total) and inseminated artificially during estrus and subsequently managed under shade structures. On Day 7 post estrus, embryos were recovered, and primarily excellent to good quality embryos (90.3%) were transferred to estrus-synchronized lactating cows. Cows were managed under conditions of exposure to summer heat stress. Pregnancy status was determined by milk progesterone concentrations at Day 21 and palpation per rectum at 45 to 60 d post estrus. Pregnancy rates of cows presented for AI (Day 21, 18.0%; Days 45 to 60, 13.5%) were typical for lactating cows inseminated during periods of summer heat stress in Florida. Pregnancy rates of embryo recipient cows were higher (P<0.001) than those of control cows (Day 21, 47.6%; Days 45 to 60, 29.2%). The objective of this study was to determine the relationship between maternal progesterone concentration and conceptus synthesis of interferon-tau as an index of conceptus viability at the time of maternal recognition of pregnancy. Heifers of mixed beef breeds were randomly assigned to receive 1 of 2 treatments: 1) intramuscular injection of 1500 IU hCG on Day 5 after artificial insemination (AI; n = 12) or 2) intramuscular injection of saline on Day 5 after AI (n = 17). Ovaries were scanned daily by transrectal real-time ultrasonography. Progesterone concentrations were determined from daily blood samples collected from the jugular vein. Heifers were slaughtered on Day 18 after AI and conceptus tissues were collected. These were incubated individually at 37 degrees C in RPMI medium, and supernatant collected after 24 h. Conceptus secretory products in the supernatant were analyzed for interferon concentration by antiviral assay using vesicular stomatitis virus. Transrectal ultrasonography showed all heifers that received hCG had at least 1 extra corpus luteum (CL) in addition to the spontaneous CL formed from the previous ovulation (10 with 2 CL, 2 with 3 CL). A significant increase in plasma progesterone concentration was detected in pregnant heifers treated with hCG (n = 9) vs pregnant control heifers (n = 11; P < 0.001). There was a tendency for an increase (P = 0.059) in synthesis of interferon-tau by conceptuses from hCG-treated heifers compared to control heifers. Maternal plasma progesterone concentrations were correlated with interferon-tau production by the conceptuses (r = 0.593, P < 0.006), suggesting that higher maternal progesterone may provide a more suitable environment for the developing conceptus.  相似文献

Zebu cattle are notorious for poor fertility characterized by late maturity and long intercalving intervals attributed to a variety of factors, including genetic, nutritional and climatic. The aim of the present investigation, therefore, was to induce fertile estrus in acyclic pubertal heifers and postpartum anestrous Zebu cows by hormonal intervention. Pubertal Hariana and Sahiwal anestrous heifers (n=51) and postpartum cows (n=55) were either assigned a placebo (controls, N=6 for each breed and parity) or treated with 10-d norgestomet (3 mg) subcutaneous ear implants, with an initial injection of 3 mg, im norgestomet + 5 mg estradiol valerate, followed by 500 IU eCG at implant withdrawal (NOR-treated groups). Jugular venous plasma samples were obtained from a total of 28 animals (controls : 4 heifers and 4 cows; NOR-treated : 12 heifers and 8 cows) on Days 0 (implant insertion), 3, 7, 9 and Day 10 (implant withdrawal), every 12 h on Days 11 and 12, and then once daily on Days 17, 24 and 31. All the samples were assayed for progesterone. Almost all (97%) heifers and 81% cows were induced to estrus, the majority (92% heifers and 79% cows) within 120 h of implant removal. Synchrony of the induced estrus was better in cows, but interval to estrus and estrus duration were significantly longer in heifers (P<0.05). Post-treatment fertility, based on Day 28 nonretum rate, first service, and overall conception rates, was better in heifers (78.9, 60.5 and 73.7%, respectively) than cows (77.1, 48.6 and 62.9%, respectively), but the differences were significant only for the overall pregnancy rate (71.8% for heifers and 51.2% for cows; P<0.05). Low pre-treatment plasma progesterone values (<0.5ng/mL) were consistent with ovarian inactivity, confirming the true anestrus status of experimental animals. Controls failed to exhibit estrus and maintained low progesterone concentrations throughout the study. In treated animals, high progesterone values from Day 17 onwards suggested ovulatory estrus. These early luteal phase progesterone concentrations in nonpregnant (P=0.06) and nonpregnant, nonretum (P<0.05) animals were low in comparison with those of pregnant animals. Good fertility resulting from breeding according to estrus, inspite of variable intervals to estrus and estrus duration, advocates its advantage over fixed-time insemination in norgestomet-treated anestrous Zebu cattle.  相似文献

This study was carried out to evaluate the luteotrophic influence of early (before Day 7 as well as after Day 7; Day 0=estrus) bovine embryos and the relationship between plasma progesterone (P4) concentrations and embryo survival. Virgin Holstein dairy heifers (n=325) from a single herd were randomly allocated to be nonbred, bred by artificial insemination (AI) or by embryo transfer (ET). Bred heifers were either treated with 1500 IU human chorionic gonadotrophin (hCG) on Day 7 of the estrous cycle or received no hCG treatment. Plasma P4 concentrations on Days 0, 5, 7, 10, 13, 15, 17, 19 and 21 were similar in pregnant AI- and ET-bred heifers and, this was observed in both hCG-treated and untreated females. Nonbred, AI- and ET-bred nonpregnant heifers (both hCG-treated and untreated) presented similar plasma P4 concentrations. Plasma P4 concentrations of pregnant heifers significantly deviated from those of nonpregnant and nonbred heifers on Day 17. In hCG-treated heifers, plasma P4 concentrations and Day 28 pregnancy rate were significantly higher in females with an induced accessory corpus luteum (CL) than in those females without an induced accessory CL. Treatment with hCG, although inducing the formation of accessory CL and significantly increasing plasma P4 concentrations had no significant effect on Day 28 pregnancy rate. In conclusion, this study does not support the existence of any peripherally detectable luteotrophic influence from early embryos (Days 5-7). Plasma P4 was only significantly related to embryo survival on Day 17, the time of expected onset of luteolysis.  相似文献
