The spatial variability of total soil nematodes and trophic groups in bare and fallow plots in Shenyang Experi-mental Station of Ecology, Chinese Academy of Sciences was examined using geostatistics combined with classic statistics. Results showed that the soil pH value had a negative effect on plant-parasites in both bare and fallow plots; the mean number of total nematodes was significantly higher in fallow plots than in bare plots, which was 1485.3 and 464.0 individuals per 100 g dry soil in fallow and bare plots, respectively; the nugget (C0)/sill (C0+C) ratio of total nematodes, plant-parasites and bacterivores were lower in fallow plots (27.3%-45.6%) than in bare plots (49.5%-100%); the spatial distribution of total nematodes and trophic groups was found to be different between fallow and bare plots, which indicated that vegetation coverage had an effect on soil nematodes.  相似文献

This study describes two packed bed bioreactor configurations which were used to culture a mouse-mouse hybridoma cell line (ATCC HB-57) which produces an IgG1 monoclonal antibody. The first configuration consists of a packed column which is continuously perfused by recirculating oxygenated media through the column. In the second configuration, the packed bed is contained within a stationary basket which is suspended in the vessel of a CelliGen bioreactor. In this configuration, recirculation of the oxygenated media is provided by the CelliGen Cell Lift impeller. Both configurations are packed with disk carriers made from a non-woven polyester fabric. During the steady-state phase of continuous operation, a cell density of 108 cells per cm3 of bed volume was obtained in both bioreactor configurations. The high levels of productivity (0.5 gram MAb per 1 of packed bed per day) obtained in these systems demonstrates that the culture conditions achieved in these packed bed bioreactors are excellent for the continuous propagation of hybridomas using media which contains low levels (1 %) of serum as well as serum-free media. These packed bed bioreactors allow good control of pH, dissolved oxygen and temperature. The media flows evenly over the cells and produces very low shear forces. These systems are easy to set up and operate for prolonged periods of time. The potential for scale-up using Fibra-cel carriers is enhanced due to the low pressure drop and low mass transfer resistance, which creates high void fraction approaching 90% in the packed bed.  相似文献

Geostatistics was applied to assess the spatial distribution of bacterivorous nematodes in a Chinese Ecosystem Research Network (CERN) site in northeast China. A 42×30 m plot was divided into grids with 7×5 m spacing that included 49 sampling points. Soil samples were collected from depths of 0–10 cm and 10–20 cm. The results showed that Cephalobus and Panagrolaimus were the most dominant bacterivores, which together represented 74 and 82% of total bacterivorous nematodes on average at depths of 0–10 cm and 10–20 cm, respectively. Within-site variability was high for these nematode genera. Geostatistical analysis indicated that a major part of this variability was spatially dependent; variograms suggest that 55–100% of sample population variance was related to spatial autocorrelation over ranges of 9–91 m. The mean numbers of total bacterivores and Panagrolaimus populations were higher at 0–10 cm depth than at 10–20 cm depth. Between the two depths, different distribution patterns were found for Panagrolaimus, whereas similar patterns were found for Cephalobus. The maps obtained by kriging well illustrated horizontal distribution patterns of bacterivorous nematodes at two depths at field scale.  相似文献

New experimental data are reported that demonstrate the use of a novel vibrating membrane filter (VMF) for the combined recovery and concentration of two species of nematodes, S. feltiae and P. hermaphrodita, from mature liquid fermentation cultures. The disk membrane module had a working surface area of 0.2 m(2) and was operated at a constant flow rate of 0.2 m(3) h(-1). The recovery of the viable nematodes from the spent media and nonviable nematodes was assisted by an independently imposed oscillatory motion of the disk assembly, which produced an intense shear field at the membrane surface with calculated mean values on the order of 10(4) s(-1). Adult (nonviable) nematodes in the fermentation culture were preferentially dissolved in a detergent (sodium dodecylsulfate) and successfully separated from the juveniles using the VMF equipment. Permeate fluxes on the order of 15 to 30 L/m(2/)h were achieved for an operating transmembrane pressure of 800 mbar. Industrial-scale liquid fermentation for the manufacture of nematodes as biopesticides produces the viable nematode life stages in low-concentration suspension containing large quantities of spent media and other waste material. The VMF equipment provided a flexible operation for separation, cleaning, and concentration of viable nematodes from the fermentation broths.  相似文献

A flat-bed hollow-fiber cell culture system has been developed which maximizes the utilization of the large fiber surface while diminishing significantly the problems inherent in a cartridge-type reactor. The reactor core consists of a shallow bed of hollow fibers sandwiched between two stainless-steel microporous filter plates through which the media flow is directed normal to the plane of the fiber bed. Reactors with both 930 and 9300 cm2 of fiber surface have been successfully constructed and operated. A variety of cells has been grown in these reactors including SV3T3 cells, baby hamster kidney cells, Vero cells, and rhesus money kidney cells, and cell products such as plasminogen activator and migration inhibition factor (MIF) were produced. This system offers an excellent prototype for scaleup design.  相似文献

Seafloor topographic complexity is ecologically important because it provides habitat structure and alters boundary-layer flow over the bottom. Despite its importance, there is little agreement on how to define and measure surface complexity. The purpose of this investigation was to utilize fractal geometry of vertical cross-section profiles to characterize the surface topography of the soft-bottom mussel bed (Mytilus edulis L.) at Bob's Cove, ME, USA. Mussels there have been shown previously to have spatially ordered fractal characteristics in the horizontal plane. Two hypotheses were tested. The first was that the bed surface is fractal over the spatial scale of 1.44-200 mm, with fractal dimension less than or equal to 1.26, the value for the Koch curve, our model for bed profiles. The second was that bed surface topography (i.e., in vertical profile) is less complex than the mussel bed spatial pattern (i.e., aerial view in the horizontal plane). Both hypotheses were supported. Cross-sections of plaster casts of the bed produced 88 surface profiles, all of which were fractal over the entire spatial scale of more two orders of magnitude employed in the analysis. Fractal dimension values (D) for individual profiles ranged from 1.031 to 1.310. Fractal dimensions of entire casts ranged up to mean (1.242+/-0.046) and median (1.251) values similar to 1.26, the theoretical value of the Koch curve. The bed surface was less complex than the bed spatial pattern because every profile had D<1.36, the smallest value previously obtained from aerial views of the bed. The investigation demonstrated for the first time that surface topography of a soft-bottom mussel bed was fractal at a spatial scale relevant to hydrodynamic processes and habitat structure important for benthic organisms. The technique of using cross-section profiles from casts of the bed surface avoided possible underestimates of fractal dimension that can result from other profiling methods reported in the literature. The results demonstrate that fractal dimension can be useful in the analysis of habitat space and water flow over any irregular seafloor surface because it incorporates the size, shape, and scale of roughness elements into a simple, numerical metric.  相似文献

以中国科学院沈阳生态实验站的裸地和撂荒地为研究对象,采用经典统计学与地统计学相结合的方法,研究裸地和撂荒地土壤线虫总数及各营养类群的空间分布特征.结果表明,裸地和撂荒地植物寄生线虫与土壤pH呈显著负相关;撂荒地土壤线虫总数的平均值显著大于裸地,分别为每百克干土1 485.3和464.0条;撂荒地土壤线虫总数、植物寄生线虫和食细菌线虫的块金值/基台值(27.3%~45.6%)低于裸地(49.5%~100%),裸地和撂荒地土壤线虫总数及各营养类群的空间分布格局均有明显差异,表明植被覆盖对土壤线虫的空间分布有较大影响.  相似文献

Meiofauna composition, abundance, biomass, distribution and diversity were investigated for 31 stations in summer. The sampling covered the whole Oosterschelde and comparisons between the subtidal — intertidal and between the western-central — eastern compartment were made.Meiofauna had a community density ranging between 200 and 17 500 ind 10 cm–2, corresponding to a dry weight of 0.2 and 8.4 gm–2. Abundance ranged between 130 and 17 200 ind 10 cm–2 for nematodes and between 10 and 1600 ind 10 cm–2 for copepods. Dry weight biomass of these taxa was between 0.5–7.0 gm–2 and 0.008–0.3 gm–2 for nematodes and copepods respectively.The meiofauna was strongly dominated by the nematodes (36–99%), who's abundance, biomass and diversity were significantly higher intertidally than subtidally and significantly higher in the eastern part than in the western part. High numbers were positively correlated with the percentage silt and negatively with the median grain size of the sand fraction. The abundance and diversity of the copepods were highest in the subtidal, but their biomass showed an inverse trend being highest on the tidal flats.The taxa diversity of the meiofauna community and species diversity of both the nematodes and the copepods were higher in subtidal stations than on tidal flats. In the subtidal, the meiofauna and copepod diversity decreased from west to east, whereas nematode diversity increased.The vertical profile clearly reflected the sediment characteristics and could be explained by local hydrodynamic conditions.Seasonal variation was pronounced for the different taxa with peak abundance in spring, summer or autumn and minimum abundance in winter.Changes in tidal amplitude and current velocity enhanced by the storm-surge barrier will alter the meiofauna community structure. As a result meiofauna will become more important in terms of density and biomass, mainly due to increasing numbers of nematodes, increasing bioturbation, nutrient mineralisation and sustaining bacterial growth. A general decrease in meiofauna diversity is predicted. The number of copepods is expected to decrease and interstitial species will be replaced by epibenthic species, the latter being more important in terms of biomass and as food for the epibenthic macrofauna and fishes.  相似文献

The changes in the community dynamics of infaunal nematodes associated with tropical Australian intertidal seagrasses at 4 estuarine sites were investigated through 3 seasons (autumn, winter and spring). Nematode densities were highest in winter in all but one of the sites, ranging from 1971 to 3084 inds./10 cm2, with one site showing a highest density of 3411 inds./10 cm2 in spring. Multiple regression revealed significant correlations between nematode density peaks and seasonal changes in temperature, salinity and surface seagrass cover. Non-metric Multi-Dimensional Scaling revealed that the communities were characterised by relatively low within-site spatial variability but relatively high temporal variability through the three seasons. This temporal variability was largely due to significant increases in abundance of epistrate-feeding species in winter and spring. An investigation of the dominant epistrate-feeding nematodes revealed that predominantly infaunal species were responsible for overall winter and spring density increases. This study provides further evidence of the role played by temperature in regulating tropical, intertidal meiobenthic communities but also indirectly provides evidence of micro-scale seagrass canopy effects (micro-algal supply and availability) that may further enhance the impacts of these larger-scale seasonal environmental changes on the infaunal nematode community.  相似文献

The nematode fauna of an estuarine mangrove Avicennia marina mudflat in Southeastern Australia has been intensively studied. About 85% of the nematodes occur in the top cm of soft mud, but 5–7 species inhabit the deeper anoxic mud down to 10 cm, both at low and high tide. One square metre was intensively sampled from four zones with different nematode faunas. At the low tide zone 58% of the nematodes were epistrate feeders, including many diatom-feeders, but in the mangrove zone selective microbial feeders made up over 60% of the population, while between high water neap and high water spring, above the mangrove zone, omnivore/predators and plant root feeding nematodes increased in relative importance. Random replicate cores reliably sampled species occurrence, but gave a high variance in density estimates. Replicate aliquots from homogenised mud gave lower density variance. Nematode densities (maximum 5 × 106 m-2) were not as high as have been reported from non-mangrove estuaries in other countries, but were within the range found in mangroves elsewhere in Australia. Margalef Species Richness values ranged from 1.7 to 3.89, which is similar to values found in other mangroves mudflats in Australia. Nematode biomass ranged from 888 mg dry weight m2 (383 mg C m-2) at the low tide zone to 19 mg dry weight m-2 (8 mg C m-2) at the upper tide level.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) have become a major type of persistent organic pollutant (POPs) in the world. To well understand the occurrence, spatial distribution and source apportionment of 16 PAHs in different media in Beijing, a comprehensive study was conducted based on 337 obtained and subsequently selected samples in four specific media, including soil, road dust, surface water and atmosphere covering sampling time spanning from 2005 to 2014. Mean concentrations of Σ16 PAHs in soils (878.5 μg/kg) decreased from inner city to exterior areas with different land uses decreased in the order of industrial estate (2510 μg/kg)>educational area (1331 μg/kg)>Park (783 μg/kg)>cultivated (650 μg/kg)>roadside (583 μg/kg)>residential (568 μg/kg)>green area (478 μg/kg)>forest area (125 μg/kg). Meanwhile, PAHs concentrations in branch roads were slightly higher than trunk roads and mainly concentrated in urban areas (1136 μg/kg). Affected by industrial and residential areas, PAHs concentrations in downstream surface rivers were much higher than upstream surface rivers (475 ng/L). Due to strong mobility, PAHs concentrations in atmosphere were not equally distributed around the study area (157.59 ng/m3). The result of source apportionment determined by diagnostic ratios method indicated PAHs in multiple media in Beijing are mainly from coal and biomass combustion.  相似文献

The aim of this study was to determine the incidence of 18 environmental variables in the spatial distribution of 30 chorotypes (species groups with significantly similar distribution patterns) of dorylaimid and mononchid nematodes by means of logistic regression in a natural area in the southeastern Iberian Peninsula. Six variables (elevation, color chroma, clay content, nitrogen content, CaCO₃, and plant community associated) were the most important environmental factors that helped explain the distribution of chorotypes. The distribution of most chorotypes was characterized by some (one to three) environmental variables; only two chorotypes were characterized by five or more variables, and four have not been characterized.  相似文献

Vertical and horizontal distribution of meiofauna was studied off Goa coast. Bottom deposit included mainly fine sand and silt. Total number of animals ranged from 250 to 2925/10 cm2. Highest faunal density was in the top 6 cm layer. The fauna consisted of two main groups, nematodes and foraminiferans which together formed 60 to 80% of the total meiofauna. Fine sand supported a rich fauna. Interstitial water and organic content of the sediment was found to be the limiting factor in the distribution of meiobenthos.  相似文献

Monoxenic cultures of the nematode, Steinernema feltiae, were carried out on two complex liquid media: P1, mainly soybean flour/egg yolk/yeast extract, and P2, mainly egg yolk/yeast extract. Up to 140 000–200 000 nematodes ml–1 were produced within 7 days, and more than 95% of the final population was in the infective juvenile stage. The total nematode concentration growth curve had a sigmoidal shape. Nematode population growth kinetics were modelled using a re-parameterised Gompertz model. Yeast extract concentration appeared to be a key factor for obtaining high nematode concentrations.  相似文献

The mixing performance as well as the adsorption performance in expanded bed chromatography (EBC) was investigated by using various types of adsorption media (average particle size = 100–700 m, density = 1100–1700 kg/m3, base matrix = hydroxyapatite, styrene-divinylbenzene, cross-linked agarose). The scale down study with 0.8 cm diameter columns was also attempted. Pulse response curves were measured with vitamin B12 as a tracer [Residence time distribution RTD experiments], and the HETP (height equivalent to a theoretical plate or plate height) values were calculated from the peak variance and the peak retention time. The HETP values for different types of packing media tested showed very similar values (0.5–1.0 cm), which did not depend on the flow-rate or the column diameter (0.8–2.6 cm). Dynamic binding capacity (DBC) values of lactic acid on a Dowex anion-exchange resin were determined from breakthrough curve (BTC) measurements for both EBC and fixed bed chromatography (FBC). The DBC values for EBC were similar to those for FBC. When the liquid feed contained insoluble particles (yeast cells) the degree of mixing increased. However, the contribution of the mixing to the total spreading of BTCs for EBC was usually small so that this increase in the mixing did not affect the adsorption performance or the DBC values significantly.  相似文献

1. δ13C and δ15N stable isotope signatures combined with an in situ microphytobenthic 13C labelling experiment were performed on epilithic biofilms of a large temperate river (the Garonne, France) to infer the trophic positioning of biofilm‐dwelling meiofauna and their uptake of microphytobenthic carbon. 2. Chironomidae larvae and Chromadorina spp. nematodes rapidly incorporated freshly produced microphytobenthic carbon in contrast to Rhyacophilidae larvae and Naididae oligochaetes. Quantitatively, macrofaunal Chironomidae incorporated more microphytobenthic carbon per day than did meiofauna. Moreover, Chironomidae seemed more involved in the spatial export of microphytobenthic carbon than nematodes. 3. Rhyacophilidae larvae were predators feeding on large meiofauna (Naididae and Chironomidae) but not on nematodes. Naididae oligochaetes primarily gained their carbon from allochthonous and/or microbial‐loop recycled sources. 4. A rapid and significant loss of labelled microphytobenthic carbon was observed. Feeding activity of biofilm‐dwelling invertebrates seemed not to be primarily involved in this loss.  相似文献

It is known that asynchronous temporal variations in local populations can contribute to the stability of metapopulations. However, studies evaluating the hierarchical organization of multiple spatial scales are rare for continuous marine landscapes, especially for marine vegetation such as seagrass beds. In this study, long‐term observation (26 yr) of temporal changes and nested spatial analyses were combined for an extensive seagrass meadow in Tokyo Bay, Japan, using remote sensing and geographic information system technologies. We examined how the dynamics at the whole‐bed scale (~1 km2) are related to those at a local scale (0.04 km2), and investigated the relationship between the seagrass dynamics and long‐term changes in environmental conditions using data on oceanography, water quality, and sediment dynamics. The seagrass bed size fluctuated between a maximum of 1.28 km2 (in 1987) and a minimum of 0.39 km2 (in 2001), with an average of 0.90 km2. The temporal variation in seagrass bed size at the whole‐bed scale correlated with sand movement within the seagrass bed related to changes in the position of a sandbar. Seagrass bed size fluctuated asynchronously at a local scale. Multivariate analyses recognized clusters of local areas showing similar patterns of fluctuation. Temporal patterns in the various clusters responded differently to changes in environmental factors, e.g. the position of the sandbar was highly correlated with seagrass bed size in shallow habitats but not in deeper areas. The magnitudes of the temporal variations for the local clusters were greater than that of the entire bed, suggesting that asynchronous fluctuation in different areas of the bed plays an important role in the overall stability of the seagrass bed. The results of the present study also highlight the importance of physical processes in regulating the temporal dynamics of seagrass beds in shallow sedimentary landscapes.  相似文献

The effect of some environmental factors on the composition and spatial distribution of the meiobenthos were studied in Vostok Bay. The spatial density of the meiobenthos significantly positively correlated with ground type and was maximum in fine- grained sands. The eumeiobenthos was dominated by harpacticoids in coarse-grained ground and by nematodes in ground with a high silt content. The pseudomeiobenthos was dominated by polychaetes and bivalve mollusks in all ground types. The depth distribution of the meiobenthos was highly heterogenous, harpacticoids being the only group whose distribution correlated significantly with depth.  相似文献

Sandy intertidal zones were analysed for the presence of meiofauna. The material was collected on six macro-tidal sandy beaches along the North Sea (The Netherlands, France, Belgium), in order to analyse the vertical and horizontal meiofaunal distribution patterns. Eleven higher meiofauna taxa (one represented by larval stage—Copepoda nauplii) were recorded. The maximum total meiofauna abundance was observed on the Dutch beach (4,295±911 ind. 10 cm−2) in the Westerschelde estuary, while the lowest values (361±128 ind. 10 cm−2) were recorded in France at the Audresselles beach. Meiofauna of the different localities consisted mainly of nematodes, harpacticoids and turbellarians. Nematodes numerically dominated all sampled stations, comprising more than 45% of the total meiofauna density. Meiofauna was mainly concentrated at the sand surface with about 70% present in the uppermost 5 cm. Meiofauna occurred across the entire intertidal zone. A clear zonation pattern in the distribution of meiofauna taxa across the beaches was observed. The present work suggests that designation of exposed sandy beaches as physically controlled (McLachlan 1988) does not explain their biological variability.  相似文献
