The classical theory of the relationship between gas fluxes and photosynthetic electron fluxes was extended by two additional terms: JL describing flux to electron sinks other than the Calvin cycle, and RL accounting for light-induced changes in non-photorespiratory CO2 evolution. RL comprises two main components, Rr resulting from light-induced decrease in tricarboxylic acid activity, and RS related to extra CO2 evolution resulting from citrate-to-2-oxoglutarate conversion for N-assimilation in NO3 grown leaves. This extended theory was applied to two experiments. First, A–Ci curves (dependence of CO2 flux on stomatal CO2 concentration) revealed a higher apparent CO2 compensation point (Γ*app) in NO3-grown plants than in NH4+-grown plants. Secondly, photosynthetic electron fluxes at different light intensities were determined by means of the Genty parameter of chlorophyll fluorescence and compared with those calculated from measured CO2 uptake. Curve-fitting based on the extended theory provided a coincidence of these two measurements and resulted in higher RS in NO3-grown than in NH4+-grown plants. This difference in RS (about 15% of the CO2 flux bound by carboxylation) is the same as that obtained from the analysis of Γ*app. Further, the analysis suggests that JL related to the extra electron flux used for N-assimilation in NO3-grown plants is diverted to other sinks in NH4+-grown plants. SHAM decreased photosynthetic electron flow and O2 evolution in NH4+-grown plants, antimycin A in NO3-grown plants. The effect of oligomycin was small. The results are discussed in terms of different mechanisms of chloroplast/mitochondrion interaction in NO3- and NH4+-grown plants, their effects on non-photorespiratory CO2 evolution and on Γ*app.  相似文献

Water stress and nitrogen (N) availability are the main constraints limiting yield in durum wheat (Triticum turgidum L. var. durum). This work investigates the combined effects of N source (ammonium–NH4+, nitrate–NO3 or a mixture of both–NH4+:NO3) and water availability (well‐watered vs. moderate water stress) on photosynthesis and water‐use efficiency in durum wheat (cv. Korifla) flag leaves grown under controlled conditions, using gas exchange, chlorophyll fluorescence and stable carbon isotope composition (δ13C). Under well‐watered conditions, NH4+‐grown plants had lower net assimilation rates (A) than those grown with the other two N forms. This effect was mainly due to lower stomatal conductance (gs). Under moderate water stress, differences among N forms were not significant, because water regime (WR) had a stronger effect on gs and A than did N source. Consistent with lower gs, δ13C and transpiration efficiency (TE) were the highest in NH4+ leaves in both water treatments. These results indicate higher water‐use efficiency in plants fertilized with NH4+ due to stomatal limitation on photosynthesis. Moreover, leaf δ13C is an adequate trait to assess differences in photosynthetic activity and water‐use efficiency caused by different N sources. Further, the effect of these growing conditions on the nitrogen isotope composition (δ15N) of flag leaves and roots was examined. Water stress increased leaf δ15N in all N forms. In addition, leaf δ15N increased as root N decreased and as leaf δ13C became less negative. Regardless of WR, the leaf δ15N of NO3‐grown plants was lowest. Based on stepwise and canonical discriminant analyses, we conclude that plant δ15N together with δ13C and other variables may reflect the conditions of N nutrition and water availability where the plants were grown. Thus well‐watered plants grown with NH4+:NO3 resembled those grown with NO3, whereas under water stress they were closer to plants grown with NH4+.  相似文献

Supplying both N forms (NH4 ++NO3 ) to the maize (Zea mays L.) plant can optimize productivity by enhancing reproductive development. However, the physiological factors responsible for this enhancement have not been elucidated, and may include the supply of cytokinin, a growth-regulating substance. Therefore, field and gravel hydroponic studies were conducted to examine the effect of N form (NH4 ++NO3 versus predominantly NO3 ) and exogenous cytokinin treatment (six foliar applications of 22 μM 6-benzylaminopurine (BAP) during vegetative growth versus untreated) on productivity and yield of maize. For untreated plants, NH4 ++NO3 nutrition increased grain yield by 11% and whole shoot N content by 6% compared with predominantly NO3 . Cytokinin application to NO3 -grown field plants increased grain yield to that of NH4 ++NO3 -grown plants, which was the result of enhanced dry matter partitioning to the grain and decreased kernel abortion. Likewise, hydroponically grown maize supplied with NH4 ++NO3 doubled anthesis earshoot weight, and enhanced the partitioning of dry matter to the shoot. NH4 ++NO3 nutrition also increased earshoot N content by 200%, and whole shoot N accumulation by 25%. During vegetative growth, NH4 ++NO3 plants had higher concentrations of endogenous cytokinins zeatin and zeatin riboside in root tips than NO3 -grown plants. Based on these data, we suggest that the enhanced earshoot and grain production of plants supplied with NH4 ++NO3 may be partly associated with an increased endogenous cytokinin supply.  相似文献

Alfalfa (Medicago sativa L.) N-sufficient plants were fed 1·5 mM N in the form of NO3, NH4+ or NO3 in conjunction with NH4+, or were N-deprived for 2 weeks. The specific activity of phosphoenolpyruvate carboxylase (PEPC) from the non-nodulated roots of N-sufficient plants was increased in comparison with that of N-deprived plants. The PEPC value was highest with NO3 nutrition, lowest with NH4+ and intermediate in plants that were fed mixed salts. The protein was more abundant in NO3-fed plants than in either NH4+- or N mixed-fed plants. Nitrogen starvation decreased the level of PEPC mRNA, and nitrate was the N form that most stimulated PEPC gene expression. The malate content was significantly lower in NO3-deprived than in NO3-sufficient plants. Root malate accumulation was high in NO3-fed plants, but decreased significantly in plants that were fed with NH4+. The effect of malate on the desalted enzyme was also investigated. Root PEPC was not very sensitive to malate and PEPC activity was inhibited only by very high concentrations of malate. Asparagine and glutamine enhanced PEPC activity markedly in NO3-fed plants, but failed to affect plants that were either treated with other N types or N starved. Glutamate and citrate inhibited PEPC activity only at optimal pH. N-nutrition also influenced root nitrate and ammonium accumulation. Nitrate accumulated in the roots of NO3- and (NO3 + NH4+)-fed plants, but was undetectable in those administered NH4+. Both the nitrate and the ammonium contents were significantly reduced in NO3- and (NO3 + NH4+)-starved plants. Root accumulation of free amino acids was strongly influenced by the type of N administered. It was highest in NH4+-fed plants and the most abundant amides were asparagine and glutamine. It was concluded that root PEPC from alfalfa plants is N regulated and that nitrate exerts a strong influence on the PEPC enzyme by enhancing both PEPC gene expression and activity.  相似文献

The carbon and nitrogen partitioning characteristics of wheat (Triticum aestivum L.) and maize (Zea mays L.) grown hydroponically at a constant pH on either 4 mM or 12 mM NO3 - or NH4 + nutrition were investigated using either 14C or 15N techniques. Greater allocation of 14C to amino-N fractions occurred at the expense of allocation of 14C to carbohydrate fractions in NH4 +-compared to NO3 --fed plants. The [14C]carbohydrate:[14C]amino-N ratios were 1.5-fold and 2.0-fold greater in shoots and roots respectively of 12 mM NO3 --compared to 12 mM NH4 +-fed wheat. In both 4 mM and 12 mM N-fed maize the [14C]carbohydrate:[14C]amino-N ratios were approximately 1.7-fold and 2.0-fold greater in shoots and roots respectively of NO3 --compared to NH4 +-fed plants. Similar results were observed in roots of wheat and maize grown in split-root culture with one root-half in NO3 --and the other in NH4 +-containing nutrient media. Thus the allocation of carbon to the amino-N fractions occurred at the expense of carbohydrate fractions, particularly within the root. Allocation of 14N and 15N within separate sets of plants confirmed that NH4 --fed plants accumulated more amino-N compounds than NO3 --fed plants. Wheat roots supplied with 15NH4 + for 8 h were found to accumulate 15NH4 + (8.5 g 15N g-1 h-1) whereas in maize roots very little 15NH4 + accumulated (1.5 g 15N g-1 h-1)It is proposed that the observed accumulation of 15NH4 + in wheat roots in these experiments is the result of limited availability of carbon within the roots of the wheat plants for the detoxification of NH4 +, in contrast to the situation in maize. Higher photosynthetic capacity and lower shoot: root ratios of the C4 maize plants ensure greater carbon availability to the root than in the C3 wheat plants. These differences in carbon and nitrogen partitioning between NO3 --and NH4 +-fed wheat and maize could be responsible for different responses of wheat and maize root growth to NO3 - and NH4 + nutrition.  相似文献

Preference for NH4+ or NO3 nutrition by the perennial legume Sesbania sesban (L.) Merr. was assessed by supplying plants with NH4+ and NO3 alone or mixed at equal concentrations (0.5 mM) in hydroponic culture. In addition, growth responses of S. sesban to NH4+ and NO3 nutrition and the effects on root nodulation and nutrient and mineral composition of the plant tissues were evaluated in a hydroponic setup at a range of external concentration of NH4+ and NO3 (0, 0.1, 0.2, 0.5, 2 and 5 mM). Seedlings of S. sesban grew equally well when supplied with either NH4+ or NO3 alone or mixed and had high relative growth rates (RGRs) ranging between 0.19 and 0.21 d−1. When larger plants of S. sesban were supplied with NH4+ or NO3 alone, the RGRs and shoot elongation rates were not affected by the external concentration of inorganic N. At external N concentrations up to 0.5 mM nodulation occurred and contributed to the N nutrition through fixation of gaseous N2 from the atmosphere. For both NH4+ and NO3-fed plants the N concentration in the plant tissues, particularly water-extractable NO3, increased at high supply concentrations, and concentrations of mineral cations generally decreased. It is concluded that S. sesban can grow without an external inorganic N supply by fixing atmospheric N2 gas via root nodules. Also, S. sesban grows well on both NH4+ and NO3 as the external N source and the plant can tolerate relatively high concentrations of NH4+. This wide ecological amplitude concerning N nutrition makes S. sesban very useful as a N2-fixing fallow crop in N deficient areas and also a candidate species for use in constructed wetland systems for the treatment of NH4+ rich waters.  相似文献

Positive effects of legumes and actinorhizal plants on N-poor soils have been observed in many studies but few have been done at high latitudes, which was the location of our study. We measured N2 fixation and several indices of soil N at a site near the Arctic Circle in northern Sweden. More than 20 years ago lupine (Lupinus nootkatensis Donn) and gray alder (Alnus incana L. Moench) were planted on this degraded forest site. We measured total soil N, net N mineralization and nitrification with a buried bag technique, and fluxes of NH+ 4 and NO 3 as collected on ion exchange membranes. We also estimated N2 fixation activity of the N2-fixing plants by the natural abundance of 15N of leaves with Betula pendula Roth. as reference species. Foliar nitrogen in the N2-fixing plants was almost totally derived from N2 fixation. Plots containing N2-fixing species generally had significantly higher soil N and N availability than a control plot without N2-fixing plants. Taken together, all measurements indicated that N2-fixing plants can be used to effectively improve soil fertility at high latitudes in northern Sweden.  相似文献

The impacts of various nitrogen sources, i.e. NO 3, NH4 + or NH4NO3 in combination with gaseous NH3, on nitrogen-, carbon- and water-use efficiency and 13C discrimination (δ13C) by plants of the C3 species Triticum aestivum L. (wheat) and the C4 species Zea mays L. (maize) were studied. Triticum aestivum and Z. mays were hydroponically grown with 2 mol · m−3 of N supplied as NO 3, NH4 + or NH4NO3 for 21 and 18 d, respectively, and thereafter exposed to gaseous NH3 at 320 μg · m−3 or to ambient air for 7 d. In T. aestivum and Z. mays over a 7-d growth period, nitrogen-use efficiency (NUE) values were influenced by N-sources in the decreasing order NH4NO3-N > NO 3-N > NH4 +-N and NO 3-N > NH4NO3-N > NH4 +-N, respectively. Fumigation with NH3 decreased the NUE values of plants grown with any of the N-forms. During 28- and 7-d growth periods, N-sources affected water-use efficiency (WUE) values in the decreasing order of NH4 +-N > NO 3-N≈NH4NO3-N in non-fumigated T. aestivum, while fumigation with NH3 increased the WUE of NO 3-grown plants. There were insignificant effects of N-sources on WUE values of Z. mays over 25- and 7-d growth periods. Furthermore, δ13C values in plant tissues (leaves, stubble and roots) were higher (less negative) in NH4 +-grown plants of T. aestivum and Z. mays than in those supplied with NH4NO3 or NO 3. Regardless of the N-form supplied to the roots of the plant species, exposure to NH3 caused more-positive δ13C values in the plant tissues. These results indicate that the variations in N-source were associated with small but significant variations in δ13C values in plants of T. aestivum and Z. mays. These differences in δ13C values are in the direction expected from differences in WUE values over long or short growth periods and with differences in the extent of non-Rubisco (ribulose-1,5-bisphosphate carboxylase-oxygenase, EC 4.1.1.39) carboxylate contribution to net C acquisition, as a function of N-source. Nitrogen (N) limits plant productivity and its uptake and assimilation may be regulated by N source, N availability, and nitrate reductase activity (NRA). Knowledge of how these factors interact to affect N uptake and assimilation processes in woody angiosperms is limited. We fertilized 1-year-old, half-sib black walnut (Juglans nigra L.) seedlings with ammonium (NH4 +) [as (NH4)2SO4], nitrate (NO3 ) (as NaNO3), or a mixed N source (NH4NO3) at 0, 800, or 1,600 mg N plant−1 season−1. Two months following final fertilization, growth, in vivo NRA, plant N status, and xylem exudate N composition were assessed. Specific leaf NRA was higher in NO3 -fed and NH4NO3-fed plants compared to observed responses in NH4 +-fed seedlings. Regardless of N source, N addition increased the proportion of amino acids (AA) in xylem exudate, inferring greater NRA in roots, which suggests higher energy cost to plants. Root total NRA was 37% higher in NO3 -fed than in NH4 +-fed plants. Exogenous NO3 was assimilated in roots or stored, so no difference was observed in NO3 levels transported in xylem. Black walnut seedling growth and physiology were generally favored by the mixed N source over NO3 or NH4 + alone, suggesting NH4NO3 is required to maximize productivity in black walnut. Our findings indicate that black walnut seedling responses to N source and level contrast markedly with results noted for woody gymnosperms or herbaceous angiosperms.  相似文献

The main goal of this study was to test the effect of [CO2] on C and N management in different plant organs (shoots, roots and nodules) and its implication in the responsiveness of exclusively N2-fixing and NO3-fed plants. For this purpose, exclusively N2-fixing and NO3-fed (10 mM) pea (Pisum sativum L.) plants were exposed to elevated [CO2] (1000 μmol mol−1 versus 360 μmol mol−1 CO2). Gas exchange analyses, together with carbohydrate, nitrogen, total soluble proteins and amino acids were determined in leaves, roots and nodules. The data obtained revealed that although exposure to elevated [CO2] increased total dry mass (DM) in both N treatments, photosynthetic activity was down-regulated in NO3-fed plants, whereas N2-fixing plants were capable of maintaining enhanced photosynthetic rates under elevated [CO2]. In the case of N2-fixing plants, the enhanced C sink strength of nodules enabled the avoidance of harmful leaf carbohydrate build up. On the other hand, in NO3-fed plants, elevated [CO2] caused a large increase in sucrose and starch. The increase in root DM did not contribute to stimulation of C sinks in these plants. Although N2 fixation matched plant N requirements with the consequent increase in photosynthetic rates, in NO3-fed plants, exposure to elevated [CO2] negatively affected N assimilation with the consequent photosynthetic down-regulation.  相似文献

Nitrogen assimilation was studied in the deciduous, perennial climber Clematis vitalba. When solely supplied with NO3 in a hydroponic system, growth and N-assimilation characteristics were similar to those reported for a range of other species. When solely supplied with NH4+, however, nitrate reductase (NR) activity dramatically increased in shoot tissue, and particularly leaf tissue, to up to three times the maximum level achieved in NO3 supplied plants. NO3 was not detected in plant material that had been solely supplied with NH4+, there was no NO3 contamination of the hydroponic system, and the NH4+-induced activity did not occur in tobacco or barley grown under similar conditions. Western Blot analysis revealed that the induction of NR activity, either by NO3 or NH4+, was matched by NR and nitrite reductase protein synthesis, but this was not the case for the ammonium assimilation enzyme glutamine synthetase. Exposure of leaf disks to N revealed that NO3 assimilation was induced in leaves directly by NO3 and NH4+ but not glutamine. Our results suggest that the NH4+-induced potential for NO3 assimilation occurs when externally sourced NH4+ is assimilated in the absence of any NO3 assimilation. These data show that the potential for nitrate assimilation in C. vitalba is induced by a nitrogenous compound in the absence of its substrate and suggest that NO3 assimilation in C. vitalba may have a significant role beyond the supply of reduced N for growth.  相似文献

Samples of recently produced shoot material collected in winter/spring from common plant species of mulga vegetation in eastern and Western Australia were assayed for 13C and 15N natural abundance. 13C analyses showed only three of the 88 test species to exhibit C4 metabolism and only one of seven succulent species to be in CAM mode. Non-succulent winter ephemeral C3 species showed significantly lower mean δ13C values (– 28·0‰) than corresponding C3-type herbaceous perennials, woody shrubs or trees (– 26·9, – 25·7 and – 26·2‰, respectively), suggesting lower water stress and poorer water use efficiency in carbon acquisition by the former than latter groups of taxa. Corresponding values for δ15N of the above growth and life forms lay within the range 7·5–15·5‰. δ15N of soil NH4+ (mean 19·6‰) at a soft mulga site in Western Australia was considerably higher than that of NO3 (4·3‰). Shoot dry matter of Acacia spp. exhibited mean δ15N values (9·10 ± 0·6‰) identical to those of 37 companion non-N2-fixing woody shrubs and trees (9·06 ± 0·5‰). These data, with no evidence of nodulation, suggested little or no input of fixed N2 by the legumes in question. However, two acacias and two papilionoid legumes from a dune of wind-blown, heavily leached sand bordering a lake in mulga in Western Australia recorded δ15N values in the range 2·0–3·0‰ versus 6·4–10·7‰ for associated non-N2-fixing taxa. These differences in δ15N, and prolific nodulation of the legumes, indicated symbiotic inputs of fixed N in this unusual situation. δ15N signals of lichens, termites, ants and grasshoppers from mulga of Western Australia provided evidence of N2 fixation in certain termite colonies and by a cyanobacteria-containing species of lichen. Data are discussed in relation to earlier evidence of nitrophily and water availability constraints on nitrate utilization by mulga vegetation.  相似文献

We investigated whether six arctic plant species have the potential to induce nitrate reductase (NR) activity when exposed to NO3 --nitrogen under controlled environment conditions, using an in vivo assay that uses the rate of NO2 --accumulation to estimate potential NR activity. We also assessed the effect of low root temperatures on NR activity, growth and nitrogen uptake (using 15N applications) in two of the selected species. Five of the six species (Cerastium alpinum, Dryas intergrifolia, Oxyria digyna, Saxifraga cernua and Salix arctica) were capable of inducing NR activity when exposed to solutions containing 0.5 mM NO3 - at 20°C for 10 days. Although in vivo NR activity was not induced in Saxifraga oppositifolia under controlled conditions, we conclude that it was capable of growing successfully on NO3 -, due to the presence of moderate rates of NR activity observed in both NH4 +-grown and NO3 --treated plants. Exposure of O. digyna and D. integrifolia to 3°C root temperatures for two weeks, with the shoots kept at 20°C, resulted in root and leaf NR activity rates of NO3 --treated plants being reduced to rates exhibited by NH4 +-grown plants. Although these decreases in NR in both species appeared to be due to limitations in NO3 --uptake and growth rate (rather than direct low-temperature inhibition of NR synthesis per se), direct low-temperature inhibition of root NR synthesis could not be ruled out. In contrast to the temperature insensitivity of NH4 + uptake in D. integrifolia, NO3 --uptake in D. integrifolia was inhibited by low root temperatures. We conclude that the selected arctic species have the genetic potential to utilize NO3 --nitrogen, and that low root temperatures, in conjunction with other environmental limitations, may be responsible for the lack of induction of NR in D. integrifolia and Salix arctica under field conditions.  相似文献

Growth, chemical composition, and nitrate reductase activity (NRA) of hydroponically cultured Rumex crispus, R. palustris, R. acetosa, and R. maritimus were studied in relation to form (NH4 +, NO3 -, or both) and level of N supply (4 mM N, and zero-N following a period of 4mM N). A distinct preference for either NH4 + or NO3 - could not be established. All species were characterized by a very efficient uptake and utilization of N, irrespective of N source, as evident from high concentrations of organic N in the tissues and concurrent excessive accumulations of free NO3 - and free NH4 +. Especially the accumulation of free NH4 + was unusually large. Generally, relative growth rate (RGR) was highest with a combination of NH4 + and NO3 -. Compared to mixed N supply, RGR of NO3 -- and NH4 +-grown plants declined on average 3% and 9%, respectively. Lowest RGR with NH4 + supply probably resulted from direct or indirect toxicity effects associated with high NH4 + and/or low Ca2+ contents of tissues. NRA in NO3 - and NH4NO3 plants was very similar with maxima in the leaves of ca 40 μmol NO2 - g-1 DW h-1. ‘Basal’ NRA levels in shoot tissues of NH4 + plants appeared relatively high with maxima in the leaves of ca 20 μmol NO2 - g-1 DW h-1. Carboxylate to organic N ratios, (C-A)/Norg, on a whole plant basis varied from 0.2 in NH4 + plants to 0.9 in NO3 - plants. After withdrawal of N, all accumulated NO3 - and NH4 + was assimilated into organic N and the organic N redistributed on a large scale. NRA rapidly declined to similar low levels, irrespective of previous N source. Shoot/root ratios of -N plants were 50–80% lower than those from +N plants. In comparison with +N, RGR of -N plants did not decline to a large extent, decreasing by only 15% in -NH4 + plants due to very high initial organic-N contents. N-deprived plants all exhibited an excess cation over anion uptake (net proton efflux), and whole-plant (C-A)/Norg ratios increased to values around unity. Possible difficulties in interpreting the (C-A)/Norg ratio and NRA of plants in their natural habitats are briefly discussed.  相似文献

The effect of NO2 fumigation on root N uptake and metabolism was investigated in 3-month-old spruce (Picea abics L. Karst) seedlings. In a first experiment, the contribution of NO2 to the plant N budget was measured during a 48 h fumigation with 100mm3m?3 NO2. Plants were pre-treated with various nutrient solutions containing NO2 and NH4+, NO3? only or no nitrogen source for 1 week prior to the beginning of fumigation. Absence of NH4+ in the solution for 6d led to an increased capacity for NO3? uptake, whereas the absence of both ions caused a decrease in the plant N concentration, with no change in NO3? uptake. In fumigated plants, NO2 uptake accounted for 20–40% of NO3? uptake. Root NO3? uptake in plants supplied with NH4+plus NO3? solutions was decreased by NO2 fumigation, whereas it was not significantly altered in the other treatments. In a second experiment, spruce seedlings were grown on a solution containing both NO2 and NH4+ and were fumigated or not with 100mm3m?3 NO2 for 7 weeks. Fumigated plants accumulated less dry matter, especially in the roots. Fluxes of the two N species were estimated from their accumulations in shoots and roots, xylem exudate analysis and 15N labelling. Root NH4+ uptake was approximately three times higher than NO3? uptake. Nitrogen dioxide uptake represented 10–15% of the total N budget of the plants. In control plants, N assimilation occurred mainly in the roots and organic nitrogen was the main form of N transported to the shoot. Phloem transport of organic nitrogen accounted for 17% of its xylem transport. In fumigated plants, neither NO3? nor NH4+ accumulated in the shoot, showing that all the absorbed NO2 was assimilated. Root NO3? reduction was reduced whereas organic nitrogen transport in the phloem increased by a factor of 3 in NO2-fimugated as compared with control plants. The significance of the results for the regulation of whole-plant N utilization is discussed.  相似文献

Kinetic parameters for NH4+ and NO3? uptake were measured in intact roots of Lolium perenne and actively N2-fixing Trifolium repens. Simultaneously, net H+ fluxes between the roots and the root medium were recorded, as were the net photosynthetic rate and transpiration of the leaves. A Michaelis–Menten-type high-affinity system operated in the concentration range up to about 500 mmol m?3 NO3? or NH4+. In L. perenne, the Vmax of this system was 9–11 and 13–14 μmol g?1 root FW h?1 for NO3? and NH4+, respectively. The corresponding values in T. repens were 5–7 and 2 μmol g?1 root FW h?1. The Km for NH4+ uptake was much lower in L. perenne than in T. repens (c. 40 compared with 170 mmol m?3), while Km values for NO3? absorption were roughly similar (around 130 mmol m?3) in the two species. There were no indications of a significant efflux component in the net uptake of the two ions. The translocation rate to the shoots of nitrogen derived from absorbed NO3?-N was higher in T. repens than in L. perenne, while the opposite was the case for nitrogen absorbed as NH4+. Trifolium repens had higher rates of transpiration and net photosynthesis than L. perenne. Measurements of net H+ fluxes between roots and nutrient solution showed that L. perenne absorbing NO3? had a net uptake of H+, while L. perenne with access to NH4+ and T. repens, with access to NO3? or NH4+, in all cases acidified the nutrient solution. Within the individual combinations of plant species and inorganic N form, the net H+ fluxes varied only a little with external N concentration and, hence, with the absorption rate of inorganic N. Based on assessment of the net H+ fluxes in T. repens, nitrogen absorption rate via N2 fixation was similar to that of inorganic N and was not down-regulated by exposure to inorganic N for 2 h. In cell-free preparations of NH4+-grown cultures of the cyanobacterium Anabaena L-31 the glutamine synthetase activity is only half as much as in N2-grown cultures. Using a procedure which enables quantitative purification of the enzyme to homogeneity it has been shown that the decrease in the enzyme activity is caused by NH4+-mediated repression. Glutamine synthetase activity in both N2-grown and NH4+-grown Anabaena remains stable for more than 24 h in the presence of chloramphenicol suggesting low enzyme turnover and an enzyme half-life greater than the generation time (16–18 h) of the cyanobacterium. In N2-grown cultures, a drastic decrease in the enzyme activity by exogenous NH4+ can be discerned when fresh protein synthesis is prevented by chloramphenicol. The enzyme purified from such cultures has Km values for NH4+, glutamate Mg2+, and ATP similar to those observed for the enzyme from N2- and NH4+-grown Anabaena, but shows depression in V for all the substrates, leading to drastic decrease in specific activity. The modified enzyme also shows a sharper thermal denaturation profile. These results indicate that NH4+-mediated modification to a less active form may be a means of regulation of glutamine synthetase in N2-fixing cultures of Anabaena.  相似文献

The aim of this research was to test whether NH4 + and NO3 affect the growth, P demand, cell composition and N2 fixation of Cylindrospermopsis raciborskii under P limitation. Experiments were carried out in P-limited (200 μg l−1 PO4-P) chemostat cultures of C. raciborskii using an inflowing medium containing either 4,000 μg l−1 NH4-N, 4,000 μg l−1 NO3-N or no combined N. The results showed the cellular N:P and C:P ratios of C. raciborskii decreased towards the Redfield ratio with increasing dilution rate (D) due to the alleviation of P limitation. The cellular C:N and carotenoids:chlorophyll-a ratios also decreased with D, predominantly as a result of an increase in the chlorophyll-a and N content. The NH4 + and NO3 supply reduced the P maintenance cell quota of C. raciborskii. Consequently, the biomass yield of the N2-grown culture was significantly lower. The maximum specific growth rate of N2-grown culture was also the lowest observed. It is suggested that these differences in growth parameters were caused by the P and energy requirement for heterocyte formation, nitrogenase synthesis and N2 fixation. N2 fixation was partially inhibited by NO3 and completely inhibited by NH4 +. It was probably repressed through the high N content of cells at high dissolved N concentrations. These results indicate that C. raciborskii is able to grow faster and maintain a higher biomass under P limitation where a sufficient supply of NH4 + or NO3 is maintained. Information gained about the species-specific nutrient and pigment stoichiometry of C. raciborskii could help to access the degree of nutrient limitation in water bodies. The blue-light requirement for the biosynthesis of nitrite reductase and an NO2 transport system was studied in Chlamydomonas reinhardtii mutant S10. The only oxidized nitrogen species that could be taken up by this mutant was NO2, due to the presence of NO2 transport systems and the absence of high-affinity NO3 transporters. NH4+-grown cells required illumination with blue light to recover the ability to take up NO2 when resuspended in an NO2-containing NH4+-deprived medium. This blue-light- dependent recovery, which took 1 h, could be suppressed by cycloheximide, indicating that protein biosynthesis was involved. The biosynthesis of nitrite reductase took place in cell suspensions irradiated with red light, even in the absence of NO2, thus suggesting that the process requiring blue light was the biosynthesis of an NO2 transport system. Nitrite reductase-containing cells (pre-irradiated with red light) took 1 h to start consuming NO2 when they were additionally irradiated with blue light in the presence of this anion, and this process was also cycloheximide-sensitive. The NO2 transport system operated either under red plus blue light or red light only. Thus, in C. reinhardtii mutant S10 cells, blue light was only required for the biosynthesis of an NO2 transport system and not for its activity.  相似文献

Summary Non-nodulatedalnus glutinosa plants were grown for 6 weeks in nutrient solutions using 3 combined-N treatments (NO3; NO3/NH4; and NH4) at a total N level of 4 meq.l–1, and growth was ccmpared with nodulated plants at zero N (N2 fixation). Of the combined-N sources, 100 per cent NH4 resulted in the highest dry matter yields when the solution pH was adjusted daily atc. 6. The dry matter yield was lowest with NO3.During the first 3 weeks, the yield of the N2-fixing plants was as high as that of the NH4 plants, but fell relatively behind during the second 3-week period. These effects could be attributed to higher initial N contents and higher shoot:root ratios, respectively, in the N2-fixing plants. Specific rates of N acquisition in the root were of a comparable order of magnitude for the combined-N and zero-N treatments.When NO3 was taken up, it was almost completely reduced in the roots. Regardless of N source there was a large excess of cations (C) relative to inorganic anions (A) in the plants, which was presumed to be balanced by an equivalent amount of organic anions (C-A). The relatively small differences in generation of organic anions for the various modes of N supply indicated the relative importance of the proton pump when NH4 or N2 was the N source. Proton or hydroxyl-ion effluxes, calculated on the basis of plant analyses, were generally in good agreement with measured excretion values. The acidity generation with N2 fixation amounted toc. 0.5 meq H+.mmol–1 Norg, which was distinctly higher than the range of 0.1–0.2 mentioned by Raven and Smith43 for dinitrogen-fixing plants.Without pH adjustment, specific rates of cation uptake and carboxylate generation were strongly depressed as the acidity increased, when NO3/NH4, NH4 and N2 were the N sources. Growth ofAlnus glutinosa appeared to be still normal at a pH ofc. 2.8. During the final 3 weeks, only the NH4 plants ceased growing at a pH of 2.6.  相似文献
