White shrimp Litopenaeus vannamei were reared at a salinity of 35‰ without a Vibrio alginolyticus injection (unchallenged group), and other shrimp were reared at 35‰, injected with tryptic-soy broth (TSB)-grown V. alginolyticus at 1.8 × 105 colony-forming units (cfu) shrimp?1 (challenged group), and then examined for the hyaline cell (HC) count, granular cell (GC, including semi-granular cell) count, total haemocyte count (THC), phenoloxidase (PO) activity, respiratory burst (RB) and superoxide dismutase (SOD) activity after transfer to 35‰ (control), 25‰, 20‰, and 15‰ for 1, 6, 12, 24, 72, and 120 h. Results indicated that the haemocyte count, PO activity, RB, and SOD activity of unchallenged shrimp and challenged shrimp that were transferred to low-salinity levels all began to significantly decrease at 6, 6, 6, and 1 h, respectively, and reached the lowest levels at 12 h. HC, GC, the THC, PO activity, RB, and SOD activity of unchallenged shrimp that were transferred to 15‰ decreased by 53%, 41%, 49%, 68%, 39%, and 62%, whereas those parameters of challenged shrimp that were transferred to 15‰ decreased by 79%, 78%, 79%, 82%, 54%, and 72%, respectively after 12 h compared to control shrimp. These immune parameters began to recover after 24–72 h for both unchallenged shrimp and challenged shrimp. We concluded that the innate immunity was weakened in white shrimp L. vannamei that received combined stresses of a V. alginolyticus injection, and low-salinity transfer. It was also concluded that shrimp with respectively 21%, 18%, 46%, and 28% lower THC, PO activity, RB, and SOD activity of the original values would be killed due to decreases in their immunity, and resistance to V. alginolyticus infection. Shrimp farming should be maintained at a constant high salinity level to prevent exacerbated decreases in innate immune parameters of shrimp when infected by a pathogen coupled with low-salinity stress leading to mortality.  相似文献

White shrimp Litopenaeus vannamei held in 25 per thousand seawater were injected with TSB-grown Vibrio alginolyticus (1 x 10(4) cfu shrimp(-1)), and then transferred to 5, 15, 25 (control) and 35 per thousand. Over 24-96 h, the mortality of V. alginolyticus-injected shrimp held in 5 per thousand and 15 per thousand was significantly higher than that of shrimp held in 25 per thousand and 35 per thousand, and the mortality of V. alginolyticus-injected shrimp held in 5 per thousand was the highest. Shrimp held in 25 per thousand and then transferred to 5, 15, 25 (control) and 35 per thousand were examined for THC (total haemocyte count), phenoloxidase activity, respiratory burst, superoxide dismutase (SOD) activity, phagocytic activity and clearance efficiency to V. alginolyticus after 12-72 h. The THC, phenoloxidase activity, respiratory burst, SOD activity, phagocytic activity and clearance efficiency decreased significantly for the shrimp held in 5 and 15 per thousand after 12 h. It is concluded that the shrimp transferred from 25 per thousand to low salinity levels (5 and 15 per thousand) had reduced immune ability and decreased resistance against V. alginolyticus infection.  相似文献

To investigate the effects of dietary β-glucan (0.5 or 1?g?kg(-1) diet: 0.5-BG, 1-BG) and rutin (0.5 or 1?g?kg(-1) diet: 0.5-RT, 1-RT) after 10 days in the absence of pathogen challenge on the immune response of Fenneropenaeus chinensis, we determined total hemocyte count (THC) and the expression of four immune-related genes in hemocytes: those for prophenoloxidase (proPO), peroxinectin (PX), lipopolysaccharide and/or β-glucan binding protein (LGBP), and c-type lectin (CL). As a prerequisite for subsequent experiments, cDNA encoding proPO of the fleshy shrimp, Fenneropenaeus chinensis (f-proPO) was obtained from hemocytes; it had a full length of 3023bp, with an open reading frame (ORF) of 2061bp, a 105-bp 5'-untranslated region, and a 906-bp 3'-untranslated region containing the poly A signal. The effect of carrageenan on the immune response of white shrimp Litopenaeus vannamei, was studied in vitro and in vivo. Shrimp haemocytes receiving carrageenan at 1 mg ml−1 experienced change in cell size, reduction in cell viability, increase in PO activity, serine proteinase activity, and RB in vitro. Shrimp received carrageenan via immersion at 200, 400 and 600 mg L−1 after 3 h and orally at 0.5, 1.0 and 2.0 g kg−1 after 3 weeks showed higher proliferation of haematopoietic tissues (HPTs) together with increases in haemocyte count and other immune parameters. Shrimp that fed a diet containing carrageenan at 0.5 g kg−1 after 3 weeks significantly up-regulated gene expressions of several immune-related proteins. The immune parameters of shrimp that received carrageenan via immersion and orally increased to a plateau after 3 h and after 3 weeks, but decreased after 5 h and 6 weeks, respectively. Phagocytosis and clearance of Vibrio alginolyticus remained high in shrimp that had received carrageenan via immersion after 5 h and orally after 6 weeks, respectively. Resistances of shrimp against V. alginolyticus and white spot syndrome virus were higher over 24–144 h and 72–144 h, respectively in shrimp that received carrageenan at 600 mg L−1 via immersion after 3 and 5 h. It was concluded that carrageenan effectively triggers an innate immunity in vitro, and increases mitotic index of HPT, immune parameters, gene expressions and resistance against pathogens in vivo. Shrimp received carrageenan via immersion and orally exhibited immunocompetence in phagocytosis and clearance of V. alginolyticus, and resistance to pathogen despite the trend in immune parameters to recover to background values.  相似文献

It is of paramount importance to study salinity tolerance of commercially important crustaceans, such as the pink shrimp Farfantepenaeus paulensis to determine possible mortality causes in the wild and in aquaculture in oligohaline waters. The aim of this study was to determine the lethal salinity concentration (LC50) for juvenile pink shrimp F. paulensis and measure its oxygen consumption and ammonia excretion at different salinity levels. Shrimp of two length classes (49.4?±?4.3 and 78.5?±?5.5?mm) were placed in 10-L containers and exposed to salinity levels of 35, 30, 25, 20, 15, 10 and 5. The experiments were tripled, with seven shrimp in each container. The average lethal concentration (LC50s) for an exposure of 24?h was 13.33 (11.26–15.78) and 10.26 (8.60–12.64), respectively, for the two classes of juveniles. For an exposure of 48?h, LC50s were 12.71 (10.68–15.12) for the larger animals and 9.20 (7.34–11.52) for the smaller ones. There was an inverse relationship between salinity and rates of oxygen consumption and ammonia excretion. The average reduction in specific oxygen consumption in salinities 20, 25 and 30 showed a decrease in metabolic rate of 63, 80 and 82%, respectively, in relation to salinity level 0. The same occurred for the averages of ammonia excretion at salinity levels of 15, 20, 25, 30 and 35, which represented low metabolic rates of 57, 61, 70, 71 and 74% respectably in relation to salinity level 0.  相似文献

The haemocyte count, phenoloxidase (PO) activity, specific alpha(2)-macroglobulin (alpha2-M) activity, respiratory burst, superoxide dismutase (SOD) activity, glutathione peroxidase (GPx) activity, phagocytic activity, and clearance efficiency against Vibrio alginolyticus were examined when the white shrimp Litopenaeus vannamei (10.42+/-2.0g) were immersed in seawater (34 per thousand) containing different concentrations of saponin (0, 0.5, 1 and 2mgL(-1)) for 24, 48 and 72h. Hyaline cells (HC), the total haemocyte count (THC), specific alpha2-M activity, respiratory burst, SOD activity, and GPx activity directly increased with the saponin concentration, whereas PO activity was inversely related to the saponin concentration. White shrimp L. vannamei that were immersed in saponin at 1 and 2mgL(-1) showed increased phagocytic activity and clearance efficiency to V. alginolyticus over 48-72h. In another experiment, shrimp immersed in seawater containing different concentrations of saponin after 72h were challenged with V. alginolyticus at 3.2x10(5) colony-forming units (cfu)shrimp(-1), and then placed in seawater. The survival rate of shrimp immersed in seawater containing saponin at either dose was significantly higher than that of control shrimp after 12h, as well as at the termination of the experiment (5days after the challenge). It was therefore concluded that L. vannamei immersed in water containing saponin at 2mgL(-1) or less exhibited an immunomodulatory effect, as well as protection against V. alginolyticus infection.  相似文献

The innate immunity and resistance against white spot syndrome virus (WSSV) in white shrimp Litopenaeus vannamei which received the Gracilaria tenuistipitata extract were examined. Shrimp immersed in seawater containing the extract at 0 (control), 400 and 600 mg L(-1) for 3 h were challenged with WSSV at 2 × 10(4) copies shrimp(-1). Shrimp not exposed to the extract and not received WSSV challenge served as unchallenged control. The survival rate of shrimp immersed in 400 mg L(-1) or 600 mg L(-1) extract was significantly higher than that of challenged control shrimp over 24-120 h. The haemocyte count, phenoloxidase activity, respiratory burst, superoxide dismutase activity, and lysozyme activity of shrimp immersed in 600 mg L(-1) extract were significantly higher than those of unchallenged control shrimp at 6, 6, 6, 6, and 6-24 h post-challenge. In another experiment, shrimp which had received 3 h immersion of 0, 400, 600 mg L(-1) extract were challenged with WSSV. The shrimp were then received a booster (3 h immersion in the same dose of the extract), and the immune parameters were examined at 12-120 h post-challenge. The immune parameters of shrimp immersed in 600 mg L(-1) extract, and then received a booster at 9, 21, and 45 h were significantly higher than those of unchallenged control shrimp at 12-48 h post-challenge. In conclusion, shrimp which had received the extract exhibited protection against WSSV as evidenced by the higher survival rate and higher values of immune parameters. Shrimp which had received the extract and infected by WSSV showed improved immunity when they received a booster at 9, 21, and 45 h post-WSSV challenge. The extract treatment caused less decrease in PO activity, and showed better performance of lysozyme activity and antioxidant response in WSSV-infected shrimp.  相似文献

本文研究了低、中和高三个盐度水平(分别为3‰、17‰和32‰)对凡纳滨对虾(Litopenaeus vannamei)各组织蛋白质的积累、肌肉谷草转氨酶和谷丙转氨酶活力、肌肉总氨基酸和游离氨基酸组成和含量的影响。结果显示,经过50d不同盐度水平的试验,低盐度组对虾的肝胰腺和血淋巴中可溶性蛋白质含量显著高于中、高盐度组(p<0.05),而肌肉中可溶性蛋白质含量在各处理组间无显著性差异;低、高盐度均导致肌肉中谷丙转氨酶和谷草转氨酶活力升高,但是各处理间的差异不显著;低、高盐度组凡纳滨对虾肌肉总氨基酸和总必需氨基酸含量均显著高于中盐度组(p<0.05),中、低盐度处理组非必需氨基酸含量差异不显著,而低盐度组对虾肌肉中蛋氨酸、丝氨酸、半胱氨酸和脯氨酸含量均显著低于中盐度组(p<0.05),其中脯氨酸为常见的5种主要渗透调节氨基酸之一;低、高盐度组对虾肌肉总游离氨基酸含量显著高于中盐度组(p<0.05),而盐度对机体绝大部分肌肉游离氨基酸含量的影响不显著(p>0.05)。结果显示,当环境盐度偏离凡纳滨对虾最适生长盐度时,其可通过在肝胰腺和血淋巴蛋白质积累及提高自身转氨酶活力,来获得机体在渗透调节供能时所需的氨基酸,而这些氨基酸以脯氨酸为主。  相似文献

Effects of L-ascorbic acid (AA) and its four derivatives, namely L-ascorbyl-2-sulfate (C2S), L-ascorbyl-2-polyphosphate (C2PP), L-ascorbyl-2-monophosphate-Na (C2MP-Na) and L-ascorbyl-2-monophosphate-Mg (C2MP-Mg) on the immune responses of juvenile grass shrimp, Penaeus monodon, were studied. The vitamin C deprived diet together with diets supplemented with either adequate or high (five times adequate) levels of AA, C2S, C2PP, C2MP-Na and C2MP-Mg were each fed to triplicate groups of shrimp (mean initial weight: 0.37 +/- 0.01 g) for 8 weeks. Significantly (P<0.01) higher weight gain (WG), feed efficiency (FE), survival, total haemocyte count (THC), superoxide anion (O2) production ratio and phenoloxidase (PO) activity were observed in shrimp fed diets supplemented with adequate and high levels of ascorbate than shrimp fed the vitamin C deprived diet, regardless of the ascorbate source. Among the ascorbate sources, shrimp fed C2MP-Mg and C2PP containing diets had higher THC than shrimp fed AA, C2S and C2MP-Na containing diets, regardless of the supplementation level. Shrimp fed adequate levels of C2MP-Mg and C2PP and high levels of C2MP-Mg containing diets had higher O2 production ratios than shrimp fed AA and C2S containing diets. Shrimp fed adequate levels of C2MP-Mg and C2PP and high levels of C2PP containing diets had higher PO activity than shrimp fed AA, C2S and C2MP-Na containing diets. These data suggest that dietary ascorbate enhances immune responses in P. monodon and different ascorbate sources may affect the immune responses differently.  相似文献

We studied the effect of two probiotic Bacillus subtilis strains on the growth performance, digestive enzyme activity, immune gene expression and disease resistance of juvenile white shrimp (Litopenaeus vannamei). A mixture of two probiotic strains, L10 and G1 in equal proportions, was administered at two different doses 10(5) (BM5) and 10(8)?(BM8)?CFU?g(-1) feed to shrimp for eight weeks. In comparison to untreated control group, final weight, weight gain and digestive enzyme activity were significantly greater in shrimp fed BM5 and BM8 diets. Significant differences for specific growth rate (SGR) and survival were recorded in shrimp fed BM8 diet as compared with the control; however, no significant differences were recorded for food conversion ratio (FCR) among all the experimental groups. Eight weeks after the start of the feeding period, shrimp were challenged with Vibrio harveyi. Statistical analysis revealed significant differences in shrimp survival between probiotic and control groups. Cumulative mortality of the control group was 63.3%, whereas cumulative mortality of the shrimp that had been given probiotics was 20.0% with BM8 and 33.3% with BM5. Subsequently, real-time PCR was employed to determine the mRNA levels of prophenoloxidase (proPO), peroxinectin (PE), lipopolysaccharide- and β-1,3-glucan-binding protein (LGBP) and serine protein (SP). The present study evaluated the effectiveness of beta-1,3-glucan derived from Schizophyllum commune in enhancing shrimp survival as well as haemocyte phagocytosis and superoxide anion production in brooder Penaeus monodon. Pond-reared P. monodon adults (135 +/- 25 g) stocked in outdoor or indoor tanks were fed either a test diet containing beta-1,3-glucan (2.0 g kg(-1) or a glucan-free control diet for 40 days. Their survival was compared. The brooders reared in indoor tanks were analysed at days 0, 1, 3, 6, 12, 24, 30 and 40 for their haemocyte phagocytic activity and superoxide anion production. The results showed that regardless of indoor or outdoor rearing the survival rate of shrimp fed the glucan diet was significantly higher (P<0.001) than that of the control group. The brooders showed enhanced haemocyte phagocytic activity, cell adhesion and superoxide anion production when glucan was administered in their diets. The immunostimulatory enhancement peaked at day 24 after starting the dietary exposure and subsequently decreased to the pre-feeding level at the end of the 40 days feeding trial.  相似文献

White shrimp Litopenaeus vannamei which had been immersed in seawater containing the hot-water extract of Gracilaria tenuistipitata at 0 (control), 200, 400, and 600 mg L?1 for 3 h, were challenged with Vibrio alginolyticus at 4.6 × 106 colony-forming units (CFU) shrimp?1 and then placed in normal seawater (34‰). The survival rates of shrimp immersed in 200, 400, and 600 mg L?1 of the hot-water extract were significantly higher than those of control shrimp over 48–120 h. In another experiment, L. vannamei which had been immersed in the hot-water extract at 0, 200, 400, and 600 mg L?1 for 3 h, were challenged with V. alginolyticus at 4.0 × 106 CFU shrimp?1, and the immune parameters examined included the haemocyte count, phenoloxidase (PO) activity, respiratory burst (RB), and superoxide dismutase (SOD) activity at 12–120 h post-challenge after shrimp had been released into normal seawater. Shrimp not exposed to the hot-water extract or V. alginolyticus served as the background control. Results indicated that the haemocyte count, PO activity, RB, and SOD activity of shrimp immersed in 600 mg L?1 were significantly higher than those of control shrimp at 12–72 h post-challenge. Results also indicated that total haemocyte count (THC), PO activity, RB and SOD activity of shrimp immersed in 400 and 600 mg L?1 of the hot-water extract returned to the background values at 96, 48, 48, and 72 h, whereas these parameters of control shrimp returned to the original values at >120, >120, 96, and 96 h post-challenge, respectively. It was therefore concluded that L. vannamei that had been immersed in seawater containing the hot-water extract of G. tenuistipitata exhibited protection against V. alginolyticus as evidenced by the earlier recovery of immune parameters.  相似文献

Larval and juvenile Gulf of California grunion, Leuresthes sardina (Jenkins & Evermann), hatched and reared in the laboratory, were subjected to salinity gradients in a Staaland salinity gradient device to determine their locomotive responses. Newly hatched prolarvae selected salinities in the range 15–24‰, while late postlarvae and juveniles preferred salinities in the range 45–54‰. Early postlarvae exhibited a bimodal preference, indicating that the behavioral change was in progress at that age. These behavioral responses may serve several functions in the physiological and behavioral ecology of the Gulf grunion.  相似文献
