This study compared the performance of two commercial preparations of buffered peptone water. Performance was assessed in terms of ability to resuscitate and recover low numbers of stressed cells, buffering capacity, growth of Salmonella bacteria in pure culture and growth of Salmonella in food pre-enrichments. Although both the preparations of BPW had similar chemical compositions, differences in their recovery performance were found. Brand A recovered significantly higher numbers of heat-injured Salmonella (mean = 0.57 log10 cfu ml(-1) difference) in pure culture compared with brand B when dealing with very low inoculum levels. Although brand B had higher buffering capacity, the pH at the end of the pre-enrichment was found to be similar in both media, even in foods such as milk powder which showed the greatest decline in pH. Both brands were comparable in their ability to grow unstressed Salmonella from different food types. In unstressed cell studies, similar cell numbers were recovered at the end of a 24 h incubation period from both media, although brand B yielded a higher biomass. In the food study with unstressed cells, performance was related more to the food type and the likely association between this and the level and type of competitor organisms present, rather than to the brand of medium used.  相似文献

Susceptibility to erythromycin was determined for all pneumococci isolated in one laboratory from clinical specimens between 1969 and 1977. All 4724 isolates examined prior to October 1973 were susceptible to erythromycin. From October 1973 to December 1977, 64 (0.71%) of 8995 pneumococcus isolates were resistant to erythromycin. The resistant strains were isolated from 38 patients living in six widely separated communities in Alberta. The erythromycin-resistant strains were of nine capsular types, including six that often cause bacteremic disease and five for which resistance to erythromycin has not been reported hitherto. Certain strains of type 33 and of type 15 were highly resistant, the minimum inhibitory concentration (MIC) of erythromycin being 2000 microgram/mL; these strains were also highly resistant to lincomycin and clindamycin. Resistance in strains of other types was much lower, the MIC of erythromycin being 0.6 to 20 microgram/mL, and all but one of these strains were susceptible to lincomycin and clindamycin. All the erythromycin-resistant pneumococci were suspectible to penicillin.  相似文献

Hybridoma WuT3 secreting a monoclonal antibody against T lymphocytes was grown in RPMI 1640 medium supplemented with 1% human serum. The effect of the concentration of peptone, as an additive, was investigated on cell growth, monoclonal antibody formation, and cell metabolism over 0–10 g l–1 range. It was found that 1–5 g l–1 peptone can significantly promote the growth of cells and increase the formation of monoclonal antibody, especially at 3–5 g l–1, when both the accumulating level and secretion rate of monoclonal antibody are higher than that at other peptone concentrations. Based on glucose, lactate and ammonia analysis data, the efficiency of glycolysis was assessed and the utilization of amino acids was more efficient at 3–5 g l–1 peptone. The cell growth and monoclonal antibody formation were inhibited at higher peptone concentrations, e.g. 10 g l–1.  相似文献

All of the 74 strains of pneumococci isolated from human infections from 1963 to 1964 proved to be uniformly and highly susceptible to penicillin. Of these strains, 15 were identified by capsule-swelling reactions and were submitted to serial transfer in the presence of increasing concentrations of penicillin. Highly penicillin-resistant mutants were selected from 14 of the 15 strains, whereas one strain was moderately resistant. Of these mutants, 11 could still react with specific antiserum, and all of the mutants could be identified by fermentation reactions and optochin inhibition. The in vitro development of penicillin resistance in these mutants did not result in a change in cell-wall composition sufficient to diminish bile solubility. The possibility of encountering rising penicillin resistance among pneumococci, as well as the possibility that such mutants may react atypically, should be kept in mind.  相似文献

Aims: Peptones are one of the most expensive constituents of microbial media. This study was undertaken to prepare the peptone from waste chicken feathers through a new process. Methods and Results: The chemical analysis of chicken feather peptone (CFP) was performed. The ability of CFP to support the growth of the three test bacteria in liquid and agar media was comparable to those of three commercial peptones [tryptone peptone (TP), fish peptone and protease peptone (PP)]. Conclusions: CFP was found to be rich in ash (42·1 g 100 g?1), protein (55·8 g 100 g?1) and mineral contents. The maximum biomass yield (3·13 g l?1) and colony number (83 × 108 CFU ml?1) for bacterium Bacillus subtilis were attained with CFP. The maximum biomass yields and colony numbers for Lactobacillus delbrueckii ssp. bulgaricus and Escherichia coli were reached in TP medium. Second high biomass yield (2·64 g l?1) and colony number (75 × 108 CFU ml?1) for E. coli were achieved using CFP. Third high biomass yield (1·29 g l?1) and colony number (90 × 107 CFU ml?1) for Lact. delbrueckii ssp. bulgaricus were obtained in CFP medium. Significance and Impact of the Study: Usability of waste chicken feathers as substrate for bacteria was investigated for the first time in the present study. The peptone may be used in industrial fermentations for production of antibiotics, organic acids, enzymes and biopolymer. It may be also used in clinical microbiology. A new chemical process was developed for peptone preparation. This process may be also employed for peptone preparation from other organic materials, especially fibrose protein‐containing materials.  相似文献

Two entomogenous fungi, Metarrhizium anisopliae and Beauveria bassiana, were cultured in liquid culture media containing various commercial peptone sources to determine the effect of the sources on growth and sporulation. Each fungus responded differently to the various peptone sources. Tryptone, Casitone, and yeast extract were effective for mycelial growth of M. anisopliae; however, yeast extract was the most effective in production of spores. Soytone Casitone, Neopeptone, and casein hydrolysate were used effectively for mycelial growth of B. bassiana, but the latter two were not as effective for production of spores. Gelatone and Peptone (Bacteriological) were not effective for production of growth or sporulation for either fungus.  相似文献

Electronic computing renders possible new methods for determining parameters of functions of stochastic relations. PAUL (1975), for instance, published a FORTRAN-program for iterative non-linear regression for any function having an independent variable. Thus, optimum parameter values of a number of growth functions were determined on the basis of tree height development. Growth functions by RICHARDS (1959, according to v. BERTALANFFY 1941) and by SCHARF (1971) reach highest accuracy of fitting. If systems of growth curves are constructed, the elements of which differ by variation of a single parameter (e.g. height class fan), then the flexibility of multiparametric growth functions gets lost however.  相似文献
