Some previous work on zygotropism in Mucor is briefly reviewed,and further observations confirming earlier studies presented. In addition to positive zygotropism between (+) and (–)zygophores, there is claimed to be a corresponding ‘repulsion’between zygophores of the same mating type, (+) from (+) and(–) from (–). The stimulus appears to be transmittedby the diffusion of a volatile substance from one zygophoreto another. A tentative explanation of the phenomena describedis advanced, based on two postulated volatile hormones, eachevolved by zygophores of one mating type only, stimulating growthin zygophores of its own mating type and retarding it in thoseof the opposite type.  相似文献

以杨柳田头菇(Agrocybe salicacola)菌株711子实体为材料,经担孢子弹射,用稀释分离法获得224个单孢菌株,其中单核菌株210个,交配试验及单孢出菇证明其性遗传模式为四极异宗结合,4种交配型的比例为AxBx∶AxBy∶AyBx∶AyBy为47∶59∶53∶51。研究结果还表明,杨柳田头菇4种交配型的数量、比例与单孢萌发速度、生长速度相关,生长速度较慢的菌株基本上属于一种交配型,只有少数生长慢的菌株属于另外两种交配型。值得注意的是,尽管快(F)-慢(S)配对的异核体与快(F)-快(F)配对的异核体在YPD平板上生长速度无明显差别,但在聚丙烯栽培袋上F-S异核体的生长速度明显快于F-F异核体,这说明交配因子A和B与生长速度基因可能连锁,且存在重组现象,F-S交配的异核体在生长上有优势,可能是人工选择后交配因子亚基减少导致偏分离发生的原因。  相似文献

The asci contain four large and four small ascospores, eachtwo-celled, arranged in the six patterns expected if spore sizewere controlled by a pair of allelic genes, the locus showing65 per cent, second-division segregation. The small ascosporesproduce sterile colonies, the large ones moderately fertilecolonies whose asci again show segregation for spore size. Fertilityis stimulated where colonies from large and small spores meet.In crosses of various mutants with the wild type, all asci inperithecia developed along the line of junction show segregationfor the mutant as well as for spore size. Evidently Chromocreais heterothallic, the spore-size difference being a pleiotropicexpression of mating type. Cave bears (Ursus spelaeus) existed in Europe and western Asiauntil the end of the last glaciation some 10,000 years ago.To investigate the genetic diversity, population history, andrelationship among different cave bear populations, we havedetermined mitochondrial DNA sequences from 12 cave bears thatrange in age from about 26,500 to at least 49,000 years andoriginate from nine caves. The samples include one individualfrom the type specimen population, as well as two small-sizedhigh-Alpine bears. The results show that about 49,000 yearsago, the mtDNA diversity among cave bears was about 1.8-foldlower than the current species-wide diversity of brown bears(Ursus arctos). However, the current brown bear mtDNA gene poolconsists of three clades, and cave bear mtDNA diversity is similarto the diversity observed within each of these clades. The resultsalso show that geographically separated populations of the high-Alpinecave bear form were polyphyletic with respect to their mtDNA.This suggests that small size may have been an ancestral traitin cave bears and that large size evolved at least twice independently.  相似文献

A study of the infection biology of stump-colonizing fungi hasshown that certain species, for instance Fomes annosus, Peniophoragigantea, Stereum sanguinolentum, and several causing blue-stain,initiate infection of fresh stumps by means of air-borne sporeswhich colonize the cut surface. The incidence of natural infectionis reduced considerably by applying creosote to the surfaceimmediately after felling. High resin content of stumps is correlatedwith increased resistance to infection. Marked seasonal variationin the incidence of stump infection by F. annosus and P. giganteawas observed. The spore content of the air, and competitionbetween fungi in stumps, are considered to be important factorsaffecting this variation. Initial colonization of stump roots usually proceeds from thebody of the stump. Invasion of roots by saprophytes presentin the soil or litter also occurs but only many months afterfelling. Crowns of plantation pines may bear many spores of stump-colonizingfungi, including F. annosus. Under the conditions described,the proportion of stump infections ascribable to rain-splash-bornespores was small compared with that caused by wind-borne spores.  相似文献

Ophryotrocha gracilis (Huth 1934) is a simultaneously hermaphroditicpolychaete worm with external fertilization and a brief protandrousphase. The mating system of this species seems to meet conditionsleading to the establishment of egg-trading behavior. Experimentsshowed that mating occurs in pairs composed of two simultaneoushermaphrodites; sex roles are sequentially alternated and self-fertilizationis avoided. Egg reciprocation is kept evolutionarily stableby laying eggs in multiple, small-sized egg clutches and reducingthe reproductive success of pairs in which one of the two partnersdoes not reciprocate egg exchange. The frequency of ovigeroushermaphrodites in mass cultures of O. gracilis is about 50".Such a high mate availability preadapts hermaphrodites of O.gracilis to change partners very frequently and to reduce investmentin parental care, contrary to what is observed in another egg-trading,simultaneously hermaphroditic species, O. diadema. Capsella bursa-pastoris(Brassicaceae) is an annual to biennialpredominantly autogamous species distributed worldwide. Usinga linkage map with RAPDs and isozymes we studied quantitativetrait loci (QTL) controlling phenotypic traits in this invasivespecies. To obtain a mapping population we crossed two plantsoccurring in different climatic regions in California, USA (CentralValley and Sierra Nevada) with the most diverse ecotypes (phenotypicparameters) and genotypes (isozyme multilocus genotypes). Ahundred and thirteen F2individuals were raised and analysedfor segregation at 107 RAPDs, six isozyme loci, and one locusdetermining leaf type. The number, location and magnitude ofgenes underlying 13 traits were determined by using both intervaland composite interval mapping. Two to five QTL affecting onecharacter have been detected. Altogether the 13 quantitativetraits produced 48 QTL. The inheritance patterns of traits rangedfrom those controlled by one QTL with a major effect to thosecontrolled by several QTL with only minor effects. Closely linkedQTL, e.g. onset of flowering with rosette leaf number, wereinterpreted as pleiotropic. Three major QTL account for onsetof flowering. These loci were linked to at least three isozymeloci and several other QTL responsible for developmental traitslike rosette leaf number. Heritability of quantitative traits,segregation of the leaf type, and segregation of the allozymeswas tested in the F3generation. We conclude that historicalevents alone are insufficient to explain the distribution patternof isozyme multilocus genotypes during the colonization of newregions and habitats. The present evidence indicates that ecotypicadaptation and genetic linkage of isozyme loci with adaptivecharacters are involved. A general, heavy infection of Yellow Rust(Puccinia StriiformisWestend.) on the leaf laminas of the spring wheat (Triticumvulgare Host) Jufy I, unlike an infection on one leaf only,modified the distribution pattern of 14C-labelled assimilatetranslocated from the second leaf: the proportion moving tothe roots (in older plants also to the tillers) was decreased,and that moving to the leaves was increased. The proportionof the assimilate translocated out of an infected leaf of asuch plant was, however more than that observed when that leafwas the only one infected, though still less than that froma corresponding leaf in a healthy plant. Age of leaf did notgreatly affect the distribution pattern. The effect of infection on the distribution pattern of assimilatefrom other leaves 15 days after inoculation was comparable tothe effect on that from the second leaf at the same intervalafter inoculation. In the case of the upper leaves the proportionmoving to the tillers was appreciably reduced by infection.These results are considered in relation to data obtained froma parellel growth analysis experiment, with which they are ingood agreement.  相似文献

Aims

The aim was to quantify the nitrogen (N) transferred via the extra-radical mycelium of the arbuscular mycorrhizal fungus Glomus intraradices from both a dead host and a dead non-host donor root to a receiver tomato plant. The effect of a physical disruption of the soil containing donor plant roots and fungal mycelium on the effectiveness of N transfer was also examined.

Methods

The root systems of the donor (wild type tomato plants or the mycorrhiza-defective rmc mutant tomato) and the receiver plants were separated by a 30 μm mesh, penetrable by hyphae but not by the roots. Both donor genotypes produced a similar quantity of biomass and had a similar nutrient status. Two weeks after the supply of 15?N to a split-root part of donor plants, the shoots were removed to kill the plants. The quantity of N transferred from the dead roots into the receiver plants was measured after a further 2 weeks.

Results

Up to 10.6 % of donor-root 15N was recovered in the receiver plants when inoculated with the arbuscular mycorrhizal fungus (AMF). The quantity of 15N derived from the mycorrhizal wild type roots clearly exceeded that from the only weakly surface-colonised rmc roots. Hyphal length in the donor rmc root compartments was only about half that in the wild type compartments. The disruption of the soil led to a significantly increased AMF-mediated transfer of N to the receiver plants.

Conclusions

The transfer of N from dead roots can be enhanced by AMF, especially when the donor roots have been formerly colonised by AMF. The transfer can be further increased with higher hyphae length densities, and the present data also suggest that a direct link between receiver mycelium and internal fungal structures in dead roots may in addition facilitate N transfer. The mechanical disruption of soil containing dead roots may increase the subsequent availability of nutrients, thus promoting mycorrhizal N uptake. When associated with a living plant, the external mycelium of G. intraradices is readily able to re-establish itself in the soil following disruption and functions as a transfer vessel.  相似文献

Groundsel (Senecio vulgaris L.) was grown in sand culture ata range of nutrient concentrations. Except when nutrient deficiencywas severe, infection by the rust fungus Puccinia lagenophoraeCooke substantially reduced root dry weight but had little effecton root length. Thus, specific root length (SRL, cm root mg–1d. wt) was significantly increased in rust-infected plants.The inhibition of root dry weight caused by rust infection wasmost pronounced late in development, especially after floweringwhen, in control plants, root elongation but not dry weightaccumulation ceased. In rusted plants, and in all plants subjectedto severe nutrient deficiency, dry weight accumulation in theroots ceased concurrently with root elongation. Late in developmentat high nutrient concentration adventitious roots with low SRLswere produced. However, infection did not modify the productionof such roots and increases in SRL could not be attributed tochanges in any single type of root. There was an inverse relationship between SRL and root diameter.This relationship was unaffected by rust infection whilst nutrientdeficiency changed only its intercept: at a given SRL rootsof nutrient stressed plants were thinner than those of plantswith adequate nutrient supply. Thus, the smaller diameter ofroots of nutrient-stressed plants occurred independently ofmeasured changes in SRL but, in the absence of nutrient stress,the decrease in root diameter caused by rust was closely relatedto increases in SRL. Changes in the root: length relationships in rusted plants mayhave important implications for root activity in the field.In view of the reported changes in SRL, inhibition of root growthin terms of dry weight may be a poor indicator of potentialchanges in activity. Apparently Fomes annosus normally invades healthy, not damaged,roots, the wood being infected almost at once in mall roots,though in larger ones the fungus grows in the bark for sometime. Alkaline soils also favour the latter type of development.Infections of living roots are more frequent around infectedstumps in alkaline soils than in acid ones. Details are given of infection of pine roots induced experimentallyand of the factors affecting invasion. Greater resistance to killing is evinced by older tree and thoseon the margin of the plantation. Disease is leas severe in EastAnglia on acid soils and where there is more organic matteron the soil surface. Matings between sister single spore lines of Coprinus disseminatusshowed a cryptic tetrapolar pattern. The two groups of matingsthat resulted in formation of mycelia with clamp connections(apparent dikaryons) differed in rate of nuclear penetrationduring mating. In one group penetration occurred at rates comparablewith nuclear migration in other species and in the other groupit was often extensive but at a rate similar to, or less than,the dikaryon growth-rate. No differences were detected betweenthese two groups in stability, colony extension rate, frequencyof clamp connections, proportions of true clamp connectionsand pseudoclamps, or number of nuclei per hyphal tip cell. Cytological studies and the isolation of hyphal tips showedthat both groups of apparent dikaryons were heterokaryotic di-or trikaryons. The di- and trikaryotic conditions co-existedin the same mycelium, but adjacent cells of individual branchingsystems usually contained equal numbers of nuclei. Within apparentdikaryons the number and kinds of nuclei per cell were similarin hyphae with clamp connections and those with simple septa.Treatments that prevented clamp connection formation did notalter the nuclear status of most of the hyphae. Irregularities in nuclear distribution were infrequent and mostwere associated with pseudoclamps. Forty per cent of nodes withprobable pseudoclamps yielded homokaryotic branches, which wereof either constituent mating type. There was some indicationthat irregularities in nuclear distribution could also occurduring divisions associated with simple septa.  相似文献

In mammals, species with high sexual size dimorphism tend tohave highly polygynous mating systems associated with high variancein male lifetime reproductive success (LRS), leading to a highopportunity for sexual selection. However, little informationis available for species with weak sexual size dimorphism. Ina long-term study population, we used parentage analysis basedon 21 microsatellite markers to describe, for the first time,variance in male lifetime breeding success (LBS) of roe deer,a territorial ungulate where males weigh less than 10% morethan females. LBS ranged from 0 to 14 (mean = 4.54, variance= 15.5), and its distribution was highly skewed, with only afew males obtaining high LBS and many males failing to breedor siring only one fawn. As predicted for polygynous specieswith low sexual size dimorphism, the standardized variance inmale LBS was low (Im = 0.75) and was only slightly higher thanthe standardized variance in female LRS (If = 0.53), suggestinga low opportunity for sexual selection. The Im value reportedhere for roe deer is much lower than values reported for highlydimorphic ungulates such as red deer (Im > 3). We suggestthat, along a continuum of opportunity for sexual selection,roe deer occupy a position closer to monogamous and monomorphicterritorial ungulates than to highly polygynous, sexually dimorphicungulates with dominance rank–based mating systems suchas harems or roving mating systems.  相似文献

By means of paper partition chromatography and a suitable biologicaltest, the number and variety of the Orobanche and Striga germinationstimulants produced by maize roots have been assessed. It isshown that extracts of maize roots and aqueous solutions inwhich roots have been suspended (diffusate solutions) containa complex of stimulating substances consisting of, at least,one water-soluble and a number of ether-soluble stimulants.Germination of O. minor is promoted by the water-soluble stimulantand one of the ether-soluble stimulants (Mo). Germination ofS. hermonthica is promoted by another ether-soluble stimulant(Ms). Aqueous extracts are rich in Ms and the water-solublematerial, whereas the diffusate contains a higher proportionof Mo and only a trace of the water-soluble stimulant. The complex of ether-soluble stimulants, but not the water-solublestimulant, accumulates in a solution of glucose when it is exposedto fragments of maize roots. These solutions contain a highproportion of Mo which is synthesized by the fragments and passesout into the sugar solution. It is suggested that the water-solublestimulant is a precursor of Mo. The effects of the water-soluble stimulant and of Ms and Moare greatly enhanced if a trace of the diffusate solution fromlinseed roots is mixed with them. The sex-specific glycoprotein agglutination substance, responsiblefor sexual agglutination, solubilized from the surface of haploidcells of a or a mating type by the autoclave method had thefollowing effects on mating reaction in Saccharomyces cerevisiae.Sexual agglutination was inhibited by the agglutination substanceof the opposite mating type in living cells as well as in heat-killedcells. Formation of zygotes was completely inhibited, when botha and a cells were treated with the agglutination substanceof the opposite mating type. The a and a agglutination substanceswere inactivated by cells of the opposite mating type, withthe degree of inactivation being greater for the former. The stimulation of the germination of Agaricus bisporus sporesby mycelium of the same species has been shown to be due toa volatile metabolite, diffusing into the culture medium andinto the atmosphere. A wide range of other fungi has been foundto affect A. bisporus spores in a similar manner. There wasno evidence . that the stimulant was carbon dioxide. A seven-carbonolefin, isolated from air, which had been passed through culturesof A. bisporus mycelium and dried with phosphorus pentoxide,showed germination-stimulating activity but may have been producedby the action of this drying agent on some other metabolitefrom the mycelium. C, radioactivity ceased to accumulai and in factdeclined under the detector (number 2) separated from the sourceof radioactivity by tr cold-treated mycelium. In the myceliumbeneath the other detector (number 1), closest to the sourceradioactivity, the rate of accumulation of radioactivity increased.When the temperature was raised t 20 ?C, radioactivity beganto accumulate in the mycelium under detector 2 and, apart froma sma fluctuation, continued to accumulate at a uniform rate.In the mycelium under detector 1, the accumulation of radioactivitystopped for a short time but then recommenced at a rate similarto thi found at 0 ?C. In other experiments the distributionof radioactivity (14C) throughout the myceliui was measuredat the end in homogenized samples. In these experiments a bandof mycelium we subjected to 0 ?C or to 20 ?C for the whole experimentalperiod, or only after the mycelium had bee translocating radioactivityalready for 16 h. These experiments showed that the changesin the rate of accumulation of 32P in living mycelium underthe two gas flow detectors used for in situ measurements werenot due to a reversal of the flow of translocation. The resultsare consistent with an hypothesis that a turgor-driven massflow of solution is the mechanism for translocation in thisfungus and are considered in relation to the results of similarexperiments on phloem translocation in higher plants. The two varieties of the take-all fungus, Ophiobolus graminisand 0. graminis var. Avenae, show a differential reaction tosap extracted from oats; the type variety, which is incapableof causing a lasting infection of oats in vivo, is inhibitedby the sap in vitro, whereas var. Avenae, pathogenic to oats,can grow in the sap. The inhibitory action of the sap is notdue to a lack of food material required by the fungus, but toa specific substance or toxin. The method used for assayingtoxicity is described. The toxin is thermostable, moderately stable on storage, virtuallyinsoluble in non-polar solvents, and soluble in acetone, water,and methanol. Methanol extraction of the ether-washed residueof the filtrate from boiled sap from oat leaves results in asemi-purified substance which, when added to 2 per cent. Yeastrelsolution, reduces growth of 0. graminis to half that of thecontrol at a concentration of to 10 p.p.m. The inhibitor is produced in considerable quantity in the leavesand stems of oats and in smaller quantity in the roots. It appearsto accumulate mainly during the period of active growth andto be less active, or present in smaller concentration, in adultplants. It can be detected in the seminal roots throughout theirexistence, although during the first 3 weeks of growth 0. graminiscan invade the cells of the cortex. It is present in greaterquantity in the crown roots, which are never penetrated. A similar inhibitor of 0. graminis can be extracted from Arrhenatherumelatius, but not from other grasses which, both in the fieldand in pot experiments, appear to be equally resistant.  相似文献

Clones of excised roots of wild type tomato (Lycopersicon esculentum,Mill., cv. Moneymaker) and a near-isogenic GA-deficient mutant(gib-1/gib-1) were cultured in modified White's medium containing1.5% w/v sucrose. The linear elongation rate of the main axisof the gib-1 mutant was 40% less than that of the wild type.In addition, the main axis of the gib-1 mutant was thicker thanthat of the wild type but main axis volume growth was the samein both genotypes, indicating that the gib-1 allele was affectingthe orientation of root expansion. There was no evidence tosuggest that the gib-1 allele affected either the pattern ofemergence or the density of lateral roots. Elongation rate andthickness of gib-1 mutant roots were restored to those of thewild type by the addition of low concentrations (0.1–1.0µM) of gibberellic acid (GA3). These concentrations ofGA3 caused a slight reduction in extension growth of wild typeroots, indicating that endogenous GAs were not limiting elongationof normal roots in culture. The GA biosynthesis inhibitor, 2S,3S paclobutrazol, at 0.1 µM, significantly reduced elongationof wild type roots and this inhibition was counteracted by 0.1µM GA3. It is concluded that the difference in growthbetween the gib-1 mutant and the wild type represented GA-dependentgrowth. Low concentrations of 2S, 3S paclobutrazol caused onlya small (5%) reduction in growth of the gib-1 mutant and thisgrowth inhibition was not reversed by GA3. This observation,and the fact that gib-1 mutant roots grow in the absence ofadded GA3, suggested that part of root growth was GA-independent.However, the possibilities that the gib-1 mutant is ‘leaky’and that paclobutrazol does not inhibit GA biosynthesis completelycannot be excluded. The infection of maize by Fusarium verticillioides can result in highly variable disease symptoms ranging from asymptomatic plants to severe rotting and wilting. We produced F. verticillioides green fluorescent protein-expressing transgenic isolates and used them to characterize early events in the F. verticillioides-maize interaction that may affect later symptom appearance. Plants grown in F. verticillioides-infested soil were smaller and chlorotic. The fungus colonized all of the underground parts of a plant but was found primarily in lateral roots and mesocotyl tissue. In some mesocotyl cells, conidia were produced within 14 to 21 days after infection. Intercellular mycelium was detected, but additional cells were not infected until 21 days after planting. At 25 to 30 days after planting, the mesocotyl and main roots were heavily infected, and rotting developed in these tissues. Other tissues, including the adventitious roots and the stem, appeared to be healthy and contained only a small number of hyphae. These results imply that asymptomatic systemic infection is characterized by a mode of fungal development that includes infection of certain tissues, intercellular growth of a limited number of fungal hyphae, and reproduction of the fungus in a few cells without invasion of other cells. Development of visibly rotted tissue is associated with massive production of fungal mycelium and much less organized growth.  相似文献
