Ten cytokinin-like substances, termed X1, X2, X3, X4a, X4b,X5a, X5b, X6, X7, and X8, active in the soya bean hypocotyltest, were detected in gladiolus corms. The factors X4a, X5aand X4 were tentatively identified as zeatin (Z), isopentenyladenosine (iPA) and isopentenyl adenine (iP), respectively,based on their behaviour in Sephadex LH-20 column chromatography,paper chromatography and high pressure liquid chromatography.Factor X3, which behaved like zeatin riboside (ZR) in the abovesystems could be ZR and/or dihydrozeatin riboside (DHZR). Thebehaviour in Sephadex and ion-exchange column chromatographysuggested that X1 and X2 may be cytokinin glucosides and X5a cytokinin nucleotide or a cytokinin conjugate similar to lupinicacid. The total cytokinin content and the concentration of Z,ZR/DHZR and iPA were higher in non-dormant than in dormant corms.The concentrations of X1 and X2 were higher in dormant corms. The embryos of germinating Zea mays seed were supplied with[14C]-adenine Following incubation, the tissue was extractedand extensively purified by non-exchange chromatography andthin layer chromatography. Radioactivity was found to be incorporatedinto zeatin nucleotide indicating that the embryo in the germinatingseed is capable of cytokinin biosynthesis. The tomato fruit was compared to a sphere with a radius R. Radialgrowth rates in the fruit (FIW and FID) due to water importor to dry matter import, respectively, which are also the waterimport rate or dry matter import rate per unit surface areaof fruit, were calculated from two sets of published results.This data referred to fruits which swelled in such a way thatthe availability of assimilates had little effect on growth.Two varieties differentiated the two series of results and inone series, three trials were differentiated by the salinityof the nutrient solution. In all trials, it was found that FIW and FID decreased whenR increased. Two phases were observed for FIW: after a firstphase, FIW decreased more quickly and almost linearly when Rincreased. FID was constant or decreased with respect to R.Except at the beginning of growth at the greatest salinity,there were clearly linear regressions between FIW and FID suchas FID = aFIW-b; where b was lower with higher salinity. Thechanges of the concentration of imported dry matter (FID/FIWwere examined in terms of R and FIW/R. The mechanisms controllingthe changes in FIW and FID were discussed. The application of 8[14C]t-zeatin to the cotyledons of germinatingbean seeds demonstrated that cytokinins are not readily exportedfrom the cotyledons to the embryonic axis during the early stagesof this process. In the cotyledons the applied zeatin is metabolizedextensively to metabolites which are polar and which occur atRF 0·2–0·5 on paper chromatograms. Thesemetabolites are stable and are not readily exported from thecotyledons. In contrast the metabolites found at RF 0–0·2are more readily exported. When exported to the radicles andplumules a large proportion of the translocated metaboliteswere converted to compounds which on paper co-chromatographedwith zeatin. This seems to suggest that the embryonic axis hasthe capacity to synthesize cytokinins and that some of the metabolitesformed during its catabolism can also be used for its synthesis. The nature of the cytokinins present in mature, expanded leavesof alder (Alnus glutinosa (L.) Gaertn.) was investigated usinga chromatographic system capable of partially resolving zeatinfrom (?)-dihydrozeatin. A zeatin-like cytokinin, present asboth a ‘free’ form and as a polar conjugate, postulatedto the the O-ß-D-glucoside of zeatin, accounted forthe bulk of the cytokinin activity (detected by the soybeancallus assay). Studies on the fate of [8-14C]zeatin supplied to detached alderleaves indicated this cytokinin to undergo extensive metabolism.An appreciable proportion of the radioactivity was incorporatedinto 80% methanol-insoluble and soluble acidic/neutral fractions,while adenosine- and adenine-like peaks were prominent metabolitesin the basic n-butanol-soluble fraction. Small amounts of glucosides,with the properties of both zeatin-O-ß-D-glucosideand dihydrozeatin-O-ß-D-glucoside were formed, thelatter becoming the most prominent with time. The ability of alder leaves to form glucoside directly from(?)-dihydrozeatin was assessed using the soybean callus assay.(?)-Dihydrozeatin was subject to a relatively rapid, continuousand substantial conversion to its putative O-ß-D-glucosideand cytokinin activity was consequently conserved while, incontrast, leaves supplied with zeatin exhibited a progressiveloss of cytokinin activity and, in agreement with the radioactivityexperiments, produced only a small amount of activity attributableto the putative O-glucosides. The significance of the observed cytokinin metabolism is discussedin relation to the endogenous cytokinin status of the leaf.  相似文献

Green islands/infection sites recorded higher cytokinin activity than surrounding tissue as well as non-inoculated tissue. This activity in infected areas increased with time of incubation while in tissue surrounding the green islands and non-inoculated tissue, cytokinin activity decreased with time of incubation. The culture filtrate extracts of H. maydis had cytokinin activity which increased with growth of the fungus. Cytokinin activity of thin-layer Chromatographic fractions from tissue and culture filtrate extracts revealed that a major portion of the activity was confined to Rf zone 0.6 to 0.8 which co-chromatographed with zeatin and zeatin riboside. Presence of zeatin and zeatin riboside in tissue and culture filtrates was confirmed by high performance liquid chromatography. Cytokinin substances, such as zeatin and zeatin riboside, increase at infection sites with growth of the pathogen suggesting they may be involved in the pathogenicity of H. maydis on maize.  相似文献

The net assimilation rate (EA), relative growth-rate (Rw), andleaf-area ratio (FA) were measured for rape (Brassica napus),sunflower (Hetianthus annuus), and maize (Zea mays) at varioustimes of year in an arid climate, using young plants grown widelyspaced on nutrient culture. Multiple regression analysis accountedfor 90–95 per cent of the variation in EA and RW in termsof two climatic variables: mean temperature and radiation receipt. EA rose linearly with radiation in all three species; increasein EA with temperature was greatest in maize and least (notsignificant) in rape. RWrose with radiation and temperature,the latter being the more important variable especially in coolweather; a temperature optimum was shown at 24° C in rape.FA rose with increase in temperature or decrease in radiation;its variation was due to change in leaf area/leaf weight ratherthan in leaf weight/plant weight. Multiple regression analyses can lead to faulty interpretationif the independent variables are correlated (as are climaticvariables in nature), but conclusions can be checked by controlled-environmentstudies in which climatic factors are not correlated. The presentconclusions are supported by such studies. The regression equations, coupled with average weather records,indicate seasonal cycles of growth parameters. EA is maximalnear midsummer and minimal near midwinter, following the radiationcycle. Maxima and minima in RW are about a month later, becauseRW is affected by the temperature cycle and this lags behindthe radiation cycle. FA is maximal in autumn and minimal inspring. EA is highest where radiation receipts near 750 cal cm–2day–1 coincide with high temperatures. This combinationoccurs only in clear midsummer weather at low latitudes, andis maintained over long periods only in arid regions. The fact that EA rose linearly with radiation suggests thatleaf water deficits arising under high radiation had littleeffect on EA and that saturating levels of light were very high.  相似文献

When etiolated seedlings of squash (Cucurbita maxima Duch. cv.Houkou-aokawaamaguri) were exposed to light, their cotyledonsbegan to accumulate chlorophyll at a low rate for the firsthour but at a high rate therafter. After 0, 1 and 2 h of illumination,cytokinin levels in cotyledons, hypocotyls and roots were analyzedby ELISA (enzyme-linked immunosorbent assay) and GC-SIM (gaschromatography-selected ion monitoring). Cytokinin levels measuredby ELISA were about 10 times higher than those measured by GC-SIM,suggesting that ELISA was not suitable for the measurementsof cytokinin levels in the extracts from these tissues. Cytokininsfound by GC-SIM were t-RZ (t-ribosyl zeatin), c-RZ, t-Z (t-zeatin),c-Z, RiP (ribosyl isopentenyladenine) and iP (isopoentenyladenine).Levels of biologically active cytokinins (t-RZ and t-Z) didnot show marked changes after illumination. Levels of c-RZ andc-Z did, however, decrease in cotyledons and increase in hypocotylsafter illumination. Promotion of the accumulation of the chlorophyllin detached squash cotyledons by exogenously applied t-Z waspartially inhibited by the addition of c-Z, suggesting thatthe decrease in the endogenous level of c-Z in cotyledons ofintact seedlings after illumination permits the light-inducedformation of chlorophyll. The dose-response curves of several cytokinins were investigatedin a soybean hypocotyl bioassay. Zeatin riboside, zeatin-O-ß-D-glucoside,dihydrozeatin, and dihydrozeatin riboside produced linear responsesparallel to that for zeatin. The hypocotyl section assay wassuperior to the conventional soybean callus assay because theresponse (log10 transformed data) was linear, exhibited lowvariability, and was more reproducible and more sensitive. Young shoots of the hop plant (Humulus lupulus L.) have beenfound to contain at least three cytokinins, one gibberellin,and abscisic acid. Two of the cytokinins were identified aszeatin riboside and N6-(3-methyl-but-2-enyl)adenosine (2iPA)based on gas chromatography-mass spectrometry (GC-MS) afterseveral purification steps. Cytokinin activity based on two bioassays was at least 100-fold higher in Psychotria punctata leaf discs with bacterial nodules than in discs without them. Nodulated discs from young leaves yielded 0.4 to 6 μg of cytokinin (zeatin equivalents) per g fresh weight of leaf tissue, whereas non-nodulated discs from the same leaves yielded 0 to 0.003 μg per g fresh weight. These estimates probably include free-base cytokinins and, if present, any nucleoside cytokinins precipitable by acidic silver nitrate. Cytokinin concentrations in Psychotria leaf nodules appear to be higher than normally found in green leaves of other plants. In l-butanol-acetic acid-water (12:3:5, v/v), the one peak of activity chromatographed with an RF similar to zeatin's, but both number and identity of the active substance(s) remain unknown. These findings suggest that a cytokinin is produced by bacteria in leaf nodules of P. punctata and that it is involved in the symbiosis.  相似文献

A key step in the purificationemployed Poros II (HS) chromatography, which greatly facilitatedisolation of the enzyme from crude intracellular lysates. Cytokinin contents in cotyledon, hypocotyl and root of etiolatedsquash (Cucurbita maxima Duch.) seedlings were determined byinstrumental analysis using 14C-benzyladenine (14C-BA) as aninternal standard. Crude extracts were purified using insolublepolyvinylpyrrolidone, cellulose-phosphate column and SEP-PAKC18 cartridge, then applied to a Sephadex LH-20 column to separatezeatin riboside (ZR), isopentenyl adenosine, isopentenyl adenine,14C-BA and a mixture of zeatin (Z) and dihydrozeatin (DHZ).The recovery rate for the cytokinin fractions after LH-20 wascorrected by 14C-BA. Each cytokinin fraction was further purifiedby HPLC which also separated Z and DHZ in the LH-20 fraction.Before permethylation, 14C-BA was added to each of the cytokininfractions to correct the methylation rates. Each methylatedcytokinin fraction was again purified by HPLC, then subjectedto gas chromatography with a capillary column and flame thermionicdetector. The detection limit of cytokinins by this system was0.1 ng. cis-ZK was the most abundant cytokinin in all tissues of theetiolated squash seedlings. Active cytokinins such as trans-ZRand trans-Z were mostly found in cotyledons with lesser amountsin the roots. DHZ was most abundant in the cotyledon. Sugar-beet, potato, and barley plants were grown in a controlledenvironment, for periods of up to 10 weeks from sowing, witha light intensity of 1,8oo f.c. (4·9 cal./cm.2/hr.) anda temperature of 20° C. during the 18-hour photoperiod and15° C. during the dark period, to test whether net assimilationrate varied with age and differed between the three species. Net assimilation rate of all species based on leaf area (EA)fell approximately linearly with time. During 5 weeks EA ofsugar-beet decreased by only about 20 per cent. and EA of potatodecreased by 50 per cent. EA of barley remained approximatelyconstant for 4 weeks after sowing and was halved during thesubsequent 4 weeks. The average value of EA for all times wasgreatest for sugarbeet and least for barley. Net assimilation rates based on leaf weight (EW) and leaf N(EN) decreased at about 15 per cent. of the initial value perweek for all species; this was similar to the mean rate of decreaseof EA of potato and barley, but greater than that of EA of sugar-beet.Mean values of EW or EN for potato and barley were similar andless than for sugar-beet. Relative growth rate (RW), relative leaf growth-rate (RA), andleaf-area ratio (F) fell with time at similar rates for allspecies. Average values of RW decreased and of F increased inthe order sugar-beet, potato, barley. RA was greatest for potatoand least for barley.  相似文献

This paper deals with the quantitative determination of free and bound cytokinins in clubroot tissue and in Plasmodiophora brassicae Woron, infected Brassica campestris L. callus tissue. The fractions were separated in a butanol soluble fraction containing the free cytokinins such as zeatin and zeatin riboside and a water soluble fraction containing the bound cytokinins. The butanol fraction was extensively purified and analysed by high pressure liquid chromatography (HPLC). The butanol fraction contained cytokinins which cochromatographed with zeatin and zeatin riboside and not with dihydrozeatin. Zeatin and zeatin riboside were quantitatively determined by HPLC. Recovery of the cytokinins varied between 30–50%. Clubs contained 50–160 ng zeatin and 210–300 ng zeatin riboside per g dry weight. Callus tissue contained 133 ng zeatin and 169 ng zeatin riboside per g dry weight. Clubs, callus as well as healthy tissue contain large amounts of bound cytokinins. Upon treatment of the water soluble fraction first with alkaline phosphatase and then with β-glucosidase biologically active fractions were found which coeluted with zeatin and zeatin riboside on Sephadex LH20 in 20% ethanol. Evidence is presented for a novel cytokinin in the water soluble fraction which yields free zeatin and glucose-6-phosphate after treatment with β-glucosidase.  相似文献

Petunia hybrida and Lilium lankongense pollens were germinatedon thin layer chromatography (TLC) plates following chromatographyof extracts from the self-, cross- and unpollinated stigmas,styles and ovaries and the seeds, leaves and pollen of threeinbred Brassica oleracea families. Zones of pollen germinationinhibition on the TLC plates showed that inhibitory compoundswere present in the tissue extracts. The Rf values and numberof these compounds varied with the tissue used, stigma tissuecontaining the largest amounts and the greatest number of inhibitors.In contrast, differences between the inbred lines tested wereslight and quantitative. Pollen from both P. hybrida and L.lankongense gave the same results; that from B. oleracea couldnot be used because of its poor germination. The effect of ultraviolet A (UVA) on growth and photosyntheticrate was studied in diatoms (Melosira spp.) of the phytoplanktonof a eutrophic lake and a cultured green alga Chloretla ellipsoidea.The cells were incubated under photosynthetically active radiation(PAR) (–UVA) or PAR + UVA conditions (+UVA). Growth ofC.ellipsoidea was retarded under +UVA, as shown by an increasein the lag period, but the rate of exponential growth was almostthe same in + and –UVA conditions. The photosyntheticrate was depressed markedly by UVA in Chlorella cells grownunder –UVA. In contrast, cells grown in +UVA showed onlyslight inhibition by UVA and after exposure to UVA for 6 daysthere was no inhibition. During the growth experiment, the cellularchlorophyll a content was higher in +UVA than +UVA grown cells.A similar effect was observed in diatoms from the eutrophicLake Suwa. In vivo fluorescence with (Fa) and without 3-(3,4-dichloropheny)-l,l-dimethylurea (DCMU) (Fb) and the photosynthetic rate were measured forC.ellipsoidea and the diatoms for 5 h under + and –UVAconditions. Soon after C.ellipsoidea had been subjected to +UVA,Fb and Fa / Fb decreased quickly and reached minima after 40min and 1 h, respectively. The suppressed in vivo fluorescenceresumed and full recovery was achieved after 4 h. This suggeststhat reactivation of the photosystem is acquired under prolongedexposure to UVA. A similar shift of Fa + Fb, but no change inFb, was found in diatoms by exposure to UVA. Changes in photosyntheticoxygen evolution by C.ellipsoidea under +UVA were similar tochanges in Fa + Fb. Degradation of chlorophyll a extracted inmethanol was enhanced by UVA. The rate of degradation by UVAwas independent of temperature from 15 to 34°C, suggestinga photochemical reaction. The results indicate that C.ellipsoideaand Melosira spp. acclimatize to prolonged UVA exposure by reactivationof the photosystem and enhanced cellular chlorophyll a synthesis.The ecological importance of these results to phytoplanktonproductivity in natural aquatic environments is discussed.  相似文献

Examination was made of the fatty acid component of tomato cutinvia gas-liquid chromatography and thin layer chromatography.Dihydroxyeicosanoic acid was identified as a major componentof tomato cutinic acid in contrast with the results of BAKERand MARTIN (1) who recognized 10,16-dihydroxyhexadecanoic acidas the dominant acid of cutin in all plants tested. On the thinlayer chromatograms we found more than nine kinds of fatty acidsin the cutin hydrolysate which was saponified with ethanol-potashsolution. The gas-liquid chromatogram for trimethylsilyl etherderivatives of methyl cutinate showed somewhat different results,i.e., unsaturated decanoic, tR 1.4, unsaturated stearic, tR4.2 and unsaturated octadecanedioic acid, tR 16.0 as unsaturatedfatty acids. Two more than C22-hydroxyfatty acids were recognizedas minor components. Beside these components, octanoic, tR 0.9,hydroxydecanoic, tR 7.0 and cis-epoxy-hydroxyoctadecanoic acid,tR 18.7 were identified. Growth of seedling oil palms (Elaeis guineensis), in full daylightand under three levels of shade, was studied using growth analysistechniques. In full day-light, net assimilation rates (EA) betweeno.15 and 0.32 g./dm.2/week were obtained associated with lowleaf-area ratios (F) giving relative growth-rates (RW) rangingfromI I.8 to 3.2 per cent, per day. There were no indicationsof seasonal differences within the small range of values found. The plants take about 90 days to adapt to shade conditions becausethe mean plastochron is 24 days, and shading effects are beststudied on plants grown since germination under the shade treatments.Very different pictures of response to shade were obtained usingplants grown initially in full light followed by 90 days' shadebefore sampling compared with plants grown under shade sincegermination. In the latter, except at the lowest light levelused, I I.I per cent, of full day-light, there was very littleeffect of light on EA or Rw, although the F values decreasedas light increased. Extrapolation of the F values to the extinctionpoint gave values similar to those obtained in another experimenton the effect of a number of shade levels on F. The physiological and ecological implications of these findings,particularly the low growth-rates and shade tolerance, are discussed.  相似文献
