This paper develops numerical techniques for two problems of interest in electrocardiography: (1) the determination of infinite-media surface potential gradients from boundedmedia torso potentials, and (2) the effects of lung-torso, blood mass-torso, and torso-air conductivity interfaces on torso surface potentials. This paper is an extension of a previous paper on the effects of conductivity interfaces which also utilized an integral equation formulation of Laplace's equation. Summary The atrial musculature of rats given the cholesterol inhibitor triparanol (MER/29) (250 mg/kg daily) for 8 days was examined under the electron microscope and compared with that from untreated animals. The sarcoplasmic core of muscle fibers from animals given triparanol exhibited a new formation of sarcoplasmic granules which displayed a crystalline latticework with opaque lines approximately 40–60 Å separated by clear spaces 50–70 Å. They were partially or completely surrounded by a membrane. The crystalline bodies in cardiac muscle fibers were not as numerous as those observed in adrenocortical, testicular interstitial, or luteal cells as reported earlier by the investigators.This research was supported by USPHS Grants HE 12751, NS 05665, and 00690.Recipient of Career Research Development Award 1 K 3 GM 28064.  相似文献

A treatment is presented which quantitates deformation of thin-walled, mildly tapered or untapered vessels having oval cross sections in the unstressed state. The discussion is limited to the quasi-static case and provides a basis for further work involving dynamic contributions and wall coupling. Results are presented for mathematical models and it is shown that the elastic constants and dimensions have significant and often profound effects especially during the initial transmural loading phase. Displacements of the major and minor semi-axes may be unequal and in different directions. These effects are more pronounced with greater eccentricity and with thinner walls. The resultants of extensible and inextensible strain combinations determine the displacements at the semi-axes. In unrestrained vessels, the resulting wall displacement can be predicted from a knowledge of the elastic constants and the principal dimensions, by determining the effective stresses at the semi-axes. Summary Cytochemical localization techniques and electron microscopy were used to evaluate the effects of clofibrate on acetyl-CoA carboxylase activity. It was demonstrated that the drug inhibited the activity of acetyl-CoA carboxylase in rat hepatocytes. Although the results in one of these experiments were somewhat variable, it is suggested that the inhibition of acetyl-CoA carboxylase may be the mechanism by which clofibrate exerts its hypolipidemic effects.Research supported by USPHS Grants HE 12751, NS 05665 and 00690.Recipient of Career Research Development Award 1K3 GM 28064. Summary The electron microscopic structure of the myocardium of the mantis shrimp is descriped. Particular attention is paid to the organization of the nerve terminal and the sarcotubular system. The general appearance of this myocardium is characterized by deep invagination of the plasma membrane at the level of Z-band and large irregular shaped mitochondria. It possesses a very well developed sarcotubular system, consisting of the longitudinal system and two transverse elements making two sets of contact to each other. One forms dyad and the other forms triad at the level of M- and Z-band, respectively. The organization of the myoneural junction in this muscle is very simple and undifferentiated. In general, a special structural differentiation is invariably observed at both sides of the contact area. In spite of the fact that synaptic vesicles and a differentiated membrane are found at the naked process of this cardiac nerve, specialization such as subsynaptic fold formation has not been observed at the muscle side which is in contact with the nerve process. Observations made on the sarcotubular system and the nerve termination have been discussed with reference to their physiological significance. Hydroethidine (HE) is a cell-permeable probe used for the intracellular detection of superoxide. Here, we report the direct measurement of the rate constant between hydroethidine and superoxide radical anion using the pulse radiolysis technique. This reaction rate constant was calculated to be ca. 2 x 10(6) M(-1) s(-1) in water:ethanol (1:1) mixture. The spectral characteristics of the intermediates indicated that the one-electron oxidation product of HE was different from the one-electron reduction product of ethidium (E+). The HPLC-electrochemical measurements of incubation mixtures containing HE and the oxygenated Fenton's reagent (Fe2+/DTPA/H2O2) in the presence of aliphatic alcohols or formate as a superoxide generating system revealed 2-OH-E+ as a major product. Formation of 2-OH-E+ by the Fenton's reagent without additives was shown to be superoxide dismutase-sensitive and we attribute the formation of superoxide radical anion to the one-electron reduction of oxygen by the DTPA-derived radical. Addition of tert-butanol, DMSO, and potassium bromide to the Fenton's system caused inhibition of 2-OH-E+ formation. Results indicate that reducing and oxidizing radicals have differential effects on the formation of 2-OH-E+.  相似文献

Summary A new method for the in vitro culture of entire, intact tracheas from adult guinea pigs is described. Matrix-embed/perfusion (MEP) culture is based on an immobilization of the tissue in nutrient agar. The tubular piece of agar-embedded organ was contained in a special perfusion block with two wells for liquid medium at either end. When incubated on a rocker platform, liquid medium flows through the trachea and supplies oxygen and nutrients. In this configuration, tracheas maintain near-normal metabolism (ATP content and dehydrogenase activity), structure (as determined by light and electron microscopy), and function (ciliary motion). Tissues could be maintained in vitro in a normal state for at least 4 wk, with reduced ciliary motion and cell metabolism detectable for at least 6 wk. Agarembedded tissues from the MEP cultures were nearly identical to those cultivated with standard tracheal ring explant techniques. Tracheas in the MEP cultures were infected withMycoplasma pneumoniae. Attachment was neuraminidase-sensitive. Mycoplasma attachment was lowest on the epithelium along the dorsal ridge, but was uniform along the length of the trachea. Ciliostasis and cytonecrosis induced byM. pneumoniae was dose dependent. The matrix-embed/perfuse technique appears to have considerable potential for several types of in vitro studies on trachea or other tubular organs. The antigenic determinants of the transferrins of 23 different primate species have been compared by a radioimmune inhibition of precipitation technique. Highest degrees of cross-reactivity with antisera to human transferrin were found in the transferrin of African apes, followed by that of the Asian apes, the Old World monkeys, the New World monkeys, and finally by the Prosimians. Thus, the evolution of the antigenic determinants on the transferrin molecule is in good agreement with the taxonomic classification of these species. Similar studies were carried out for 2-macroglobulin and various immunoglobulin chains. Hepatic encephalopathy (HE) is a neuropsychiatric complication to liver disease. It is known that ammonia plays a role in the pathogenesis of HE and disturbances in the GABAergic system have been related to HE. Synthesis of GABA occurs by decarboxylation of glutamate formed by deamidation of astrocyte-derived glutamine. It is known that a fraction of glutamate is decarboxylated directly to GABA (referred to as the direct pathway) and that a fraction undergoes transamination with formation of alpha-ketoglutarate. The latter fraction is cycled through the neuronal tricarboxylic acid cycle, an energy-generating pathway, prior to being employed for GABA synthesis (the indirect pathway). We have previously shown that ammonia induces an elevation of the neuronal tricarboxylic acid cycle activity. Thus, the aims of the present study were to determine if increased levels of ammonia increase GABA synthesis via the indirect pathway in a rat model of HE induced by bile-duct ligation and in co-cultures of neurons and astrocytes exposed to ammonia. Employing (13) C-labeled precursors and subsequent analysis by mass spectrometry, we demonstrated that more GABA was synthesized via the indirect pathway in bile duct-ligated rats and in co-cultures subjected to elevated ammonia levels. Since the indirect pathway is associated with synthesis of vesicular GABA, this might explain the increased GABAergic tone in HE.  相似文献

Male rats of the strains with low (LE) high excitability (HE) of the nervous system have been used in this study. Half of the animals of each strain were neurotized in accordance with the Hecht's scheme. In the hippocampal slices of the non-neurotized LE rats there was a significant increase of the populational spike amplitude during development of LTP as compared with the opposite group of the animals. The LTP formation in the LE strain of rats caused a decrease in the S-100 protein content in the water-soluble, and an increase in the membrane-bound fraction of the protein. Similar results we have observed with the non-inbred Wistar rats but not with the HE strain of the animals. The levels of the water-soluble S-100 protein fraction were also higher in the hippocampuses and entorenal cortices, but not in the cerebellae of the LE strain, as compared with the HE strain of the rats. No differences have been found in the membrane-bound fraction of S-100 protein.  相似文献

Compactization of supercoiled circular plasmid pBR322 caused by interaction with synthetic oligopeptide dansyl hydrazide trivaline capable of beta-structure formation was studied by electron microscopy. The results show that at rising input peptide concentration circular DNA molecules undergo intramolecular structural transition with the formation of compact ring structures. The compact ring structures are formed by the fiber having the thickness of 60 A. The analysis of morphology of intermediate structures and the contour length measurements enable us to conclude that 60 A-fiber contains three lying side-by-side and interwound double-stranded DNA segments. Thus, the compact ring structures are addressed to as triple rings. The triple ring have one special point, where the triple region ends are locked by a duplex DNA segment. The mechanisms responsible for the triple ring formation may be of importance for DNA and chromatin compactization processes in vivo.  相似文献

Immunoelectron microscopy was used to assess the effects of inhibitors of cell division on formation of the FtsZ ring in Escherichia coli. Induction of the cell division inhibitor SulA, a component of the SOS response, or the inhibitor MinCD, a component of the min system, blocked formation of the FtsZ ring and led to filamentation. Reversal of SulA inhibition by blocking protein synthesis in SulA-induced filaments led to a resumption of FtsZ ring formation and division. These results suggested that these inhibitors block cell division by preventing FtsZ localization into the ring structure. In addition, analysis of min mutants demonstrated that FtsZ ring formation was also associated with minicell formation, indicating that all septation events in E. coli involve the FtsZ ring.  相似文献

Summary This research was undertaken in order to determine if known biochemical processes involving the conversion of fluoroacetate are reflected morphologically at the electron microscopic level. No changes were observed in the hepatocytes after 2.5 mg/kg of fluoroacetate and sacrifice at 3 hours, but with 5 mg/kg of the drug there was an increase in the amount of agranular endoplasmic reticulum, an aberrant shape of mitochondria, and a disappearance of visible glycogen. One hour after 10, 15, 20, or 30 mg/kg of fluoroacetate the results showed a tremendous increase in the amount of agranular endoplasmic reticulum, an aberration in mitochondrial structure, and a disappearance of the intramitochondrial granules and glycogen. A plant with 2n = 14 + 1 ring chromosomes was obtained in the progeny of a primary trisomie for chromosome 7 of a two-rowed cultivar, Shin Ebisu 16. The morphological characteristics of the trisomic plants with an extra ring chromosome were similar to the primary trisomic for chromosome 7 (Semierect), which suggests that it originated from this chromosome. The ring chromosomes were not completely stable in mitotic cells because of abnormal behavior. Chromosome complements varied in different plants and in different roots within a plant. Root tip cells and spikes with 2n = 14 and 14 + 2 ring chromosomes were observed on plants with 14 + 1 ring chromosomes. Breakage-fusion-bridge cycle was inferred. The ring chromosome was associated with two normal homologues forming a trivalent in 17.6% sporocytes at metaphase I. The transmission of the extra ring chromosome was 23.1% in the progeny of the plant with 14 + 1 ring chromosomes. Trivalent formation may have been much higher at early prophase stages which were difficult to analyze in barley; only 4 of 120 sporocytes analyzed showed an isolated ring at pachytene. The ring chromosome moved to one pole without separation in 24.7% of the sporocytes at AI, and divided in 27.1% sporocytes giving rise to 8-8 separation. Only 10% of the sporocytes showed bridge formation at AI.  相似文献

The formation of FtsZ rings (Z rings) in various fts mutants was examined by immunoelectron microscopy and immunofluorescence. In two temperature-sensitive ftsZ mutants which form filaments with smooth morphology, the Z ring was unable to form. In ftsA, ftsI, and ftsQ mutants, which form filaments with an indented morphology, Z rings formed but their contraction was blocked. These results indicate that fully functional ftsA, ftsQ, and ftsI genes are not required for Z-ring formation and are unlikely to have a role in localization of the Z ring. The results also suggest that one function of the Z ring is to localize the activity of other fts gene products.  相似文献

Hepatic encephalopathy (HE) is an important cause of morbidity and mortality in patients with liver disease. The pathogenesis of he is incompletely understood although ammonia and inflammatory cytokines have been implicated as key mediators. To facilitate further mechanistic understanding of the pathogenesis of HE, a large number of animal models have been developed which often involve the surgical creation of an anastomosis between the hepatic portal vein and the caudal vena cava. One of the most common congenital abnormalities in dogs is a congenital portosystemic shunt (cpss), which closely mimics these surgical experimental models of HE. Dogs with a cPSS often have clinical signs which mimic clinical signs observed in humans with HE. Our hypothesis is that the pathogenesis of HE in dogs with a cPSS is similar to humans with HE. The aim of the study was to measure a range of clinical, haematological and biochemical parameters, which have been linked to the development of HE in humans, in dogs with a cPSS and a known HE grade. One hundred and twenty dogs with a cPSS were included in the study and multiple regression analysis of clinical, haematological and biochemical variables revealed that plasma ammonia concentrations and systemic inflammatory response syndrome scores predicted the presence of HE. Our findings further support the notion that the pathogenesis of canine and human HE share many similarities and indicate that dogs with cPSS may be an informative spontaneous model of human HE. Further investigations on dogs with cPSS may allow studies on HE to be undertaken without creating surgical models of HE thereby allowing the number of large animals used in animal experimentation to be reduced.  相似文献

New assignments of three previously undetected amide proton NMR resonance lines in bovine pancreatic trypsin inhibitor are reported. These are the peptide amide proton of Gly-37 and the primary amide protons of Asn-44. Specific assignments of Asn-44 and Asn-43 HE and HZ resonances are also reported. The Gly-37 NH and Asn-44 HZ resonances are shifted upfield to 4.3 and 3.4 ppm, respectively, by the ring current of the Tyr-35 aromatic group, while Asn-44 HE resonates at 7.8 ppm. The abnormal chemical shifts of Asn-44 HZ and Gly-37 NH indicate that both NH's interact with the pi-electron cloud of the Tyr-35 ring. This is consistent with their location in the crystal structure. The resonances are resolved by differential labeling techniques and are studied by combined use of NOE and exchange difference spectroscopy.  相似文献

The Tel Hashomer variant of human G6PD migrates as two prominent components during electrophoresis in several gel systems in which red cell G6PD from other males migrates predominantly as a single band. Since human males normally have but one X-chromosome, the extra band of this variant seemed an exception to earlier biochemical and genetic observations suggesting that human red cell G6PD is determined by a locus on the X chromosome. Results of the present studies indicate that the Tel Hashomer variant is unusually susceptible to the formation of a complex which has a higher molecular weight than normal G6PD and which represents the slow electrophoretic component. The conditions of formation and disruption of this complex in crude and purified Tel Hashomer preparations suggest that it results from the formation of disulfide bridges between molecules of Tel Hashomer G6PD.Supported by U.S. Public Health Service Research Grants AM-11065 and FR-5406 and Research Career Development Award 5 K3 AM 7992.  相似文献

Previous studies have implicated autophagy in osteoclast differentiation. The aim of this study was to investigate the potential role of p62, a characterized adaptor protein for autophagy, in RANKL-induced osteoclastogenesis. Real-time quantitative PCR and western blot analyses were used to evaluate the expression levels of autophagy-related markers during RANKL-induced osteoclastogenesis in mouse macrophage-like RAW264.7 cells. Meanwhile, the potential relationship between p62/LC3 localization and F-actin ring formation was tested using double-labeling immunofluorescence. Then, the expression of p62 in RAW264.7 cells was knocked down using small-interfering RNA (siRNA), followed by detecting its influence on RANKL-induced autophagy activation, osteoclast differentiation, and F-actin ring formation. The data showed that several key autophagy-related markers including p62 were significantly altered during RANKL-induced osteoclast differentiation. In addition, the expression and localization of p62 showed negative correlation with LC3 accumulation and F-actin ring formation, as demonstrated by western blot and immunofluorescence analyses, respectively. Importantly, the knockdown of p62 obviously attenuated RANKL-induced expression of autophagy- and osteoclastogenesis-related genes, formation of TRAP-positive multinuclear cells, accumulation of LC3, as well as formation of F-actin ring. Our study indicates that p62 may play essential roles in RANKL-induced autophagy and osteoclastogenesis, which may help to develop a novel therapeutic strategy against osteoclastogenesis-related diseases.  相似文献

Germline cyst formation via incomplete cytokinesis (IC) is necessary to generate functional eggs and sperm in various organisms. Drosophila melanogaster oogenesis is an ideal system for studying IC. 29 stages of germline cyst formation can be identified in D. melanogaster oogenesis. We have defined necessary terminology to describe IC and have developed a method to measure the sizes of contractile rings and ring canals. Time course study of germline cyst formation demonstrates that contractile ring constriction proceeds to a defined end point unique for each mitotic division. Contractile rings constrict to a greater degree, resulting in smaller ring diameters, for each subsequent round of mitotic division. Contrary to conventional wisdom, ring canal growth is not initiated until well after the fourth mitotic division. Ring canals grow, in an orderly manner, with ring canals derived from the first mitotic division enlarging first followed by those from the second, then those from the third, and finally those from the fourth mitotic division. This work establishes a foundation for identifying genes specific for IC and for elucidating the molecular mechanism underlying this aspect of germline cyst formation.  相似文献
