The ultrastructure ofCucullosporella mangrovei ascospores is described. Mature ascospores possess two wall layers, an outer electron-dense episporium and an innermost tripartite mesosporium. Episporial elaborations form electrondense spore wall ornamentations from which extend fibrils that may constitute a highly hydrated exosporium which was not visualised at either the scanning electron microscope or light microscope level. Ascospores possess a hamate appendage at each pole which unfolds in seawater to form a long thread. Ultrastructurally the polar appendage comprises folded fibro-granular electron-dense material and fine fibrils. The fibrils form a matrix around and within the fibro-granular appendage and around the entire unreleased ascospore. These fibrils have not been observed associated with the ascospore appendages in other species of the Halosphaeriales and are a discrete and new appendage component. The fibro-granular appendage and fibrils are bounded by the outer delimiting membrane which is absent around released ascospores. The nature of the spore appendage is compared with that of other marine and freshwater ascomycetes and the taxonomic assignment of the species is discussed.  相似文献

An ultrastructural study of Weddellomyces epicallopisma (ascomata wall, asci, ascospores and vegetative hyphae), the first done on the family Dacampiaceae, confirms most of the observations made in light microscopy. Moreover it shows that ascospores are provided with an endospore (not visible in light microscope) and that the structure of the ascospore septum is more complex. The similarity of the wall structure between the ascospore and the hyphoid appendages, developed on the upper part of the ascoma, is emphasized.  相似文献

During a latitudinal survey of freshwater ascomycetes, an unidentified fungus with bitunicate asci was found on submerged wood and herbaceous material from Florida and Costa Rica. Based on morphological characteristics and 28S rDNA large subunit (LSU) sequence data, this fungus is described as a new genus and species, Wicklowia aquatica, and placed in the Pleosporales (Pleosporomycetidae, Dothideomycetes). Phylogenetic analyses based on LSU sequences did not resolve the familial placement of W. aquatica within the Pleosporales. The characteristic features of W. aquatica are subglobose, dorsiventrally flattened, ostiolate, immersed to erumpent, black ascomata; a peridial wall composed of 4–5 layers of darkened pseudoparenchymatic cells; cellular pseudoparaphyses immersed in a gel matrix; broadly clavate, bitunicate asci; and cylindrical, hyaline, one-septate ascospores with rounded apices and surrounded by a gelatinous sheath that expands in water; ascospore sheath attached at the ascospore base with a gelatinous curtain extending from the base that fragments into basal filamentous appendages which radiate from the base of the ascospore.  相似文献

Four new species of ascomycetes, Ascotaiwania pennisetarum sp. nov., Diaporthe appendiculata sp. now., Diaporthe minutis sp. nov., and Oxydothis miscanthiecola sp. nov., are described from senescent culms of grasses in Hong Kong. Ascotaiwania pennisetarum is distinctive from other species in the genus as ascospores are narrower. Diaporthe minutis differs from the most similar Diaporthe species in having ascospores with bipolar mucilaginous pads, while Diaporthe appendiculata differs from other species in having ascospores with both a mucilaginous pad and appendages at both ends. Oxydothis miscanthicola differs from other species in the genus in having long fusiform ascospores, which taper to rounded apices or otherwise differ in shape and size. These new species are compared with existing species and illustrated with interference light micrographs. The Diaporthe species known to posses mucilaginous ascospore appendages are listed.  相似文献

Among the isolates from soil of corn and sugar cane plantations in São Paulo State, Brazil, two new species ofNeosartorya, N. botucatensis andN. paulistensis, are described and illustrated.Neosartorya botucatensis differs from the other known species of the genus in having ascospores with long spines on the convex walls.Neosartorya paulistensis is characterized by its ascospore walls with spinose and verruculose ornamentation. Two new species ofHeleococcum (a cleistothecial nectrioid genus in the Hypocreaceae) are described and illustrated.Heleococcum alatosporum, isolated from Indonesian soil, is recognized by the production of salmon-colored ascomata, cylindrical asci, and hyaline, small, bicellular ascospores with walls that are verruculose and ornamented with longitudinal ridges.Heleococcum inapertum, isolated from Philippine soil, is characterized by yellow ascomata, clavate asci, and pale yellow, middle-sized, bicellular, verruculose to weakly striate ascospores surrounded with a hyaline sheath. Anamorphs of the new species are included inAcremonium. A key to the accepted species of the genus is provided.  相似文献

The formation of the ascospore cell wall of Schizosaccharomyces pombe requires the co-ordinated activity of enzymes involved in the biosynthesis of its components, such as glucans. We have cloned the bgs2+ gene. bgs2+ belongs to the glucan synthase family of S. pombe and is homologous to the Saccharomyces cerevisiae FKS1 and FKS2 genes. Deletion or overexpression of this gene does not lead to any apparent defect during vegetative growth, but homozygous bgs2Delta diploids do show a sporulation defect. Although meiosis takes place normally, ascospores are unable to mature, and their wall differs from that of wild-type ascospores. Moreover, bgs2Delta zygotes were not able to release ascospores spontaneously, and the ascospores were unable to germinate. We show that expression of bgs2+ is restricted to sporulation and that a bgs2-green fluorescent protein (GFP) fusion protein localizes to the ascospore envelope. The glucan synthase activity in sporulating diploids bearing a bgs2 deletion was diminished in comparison with that of the wild-type diploids, a fact that underscores the importance of the bgs2+ gene and glucan synthesis for the proper formation and maturation of the ascospore wall.  相似文献

The germination of ascospores of the marine fungusHalosphaeria appendiculata was investigated with transmission electron microscopy. Prior to germination, settled ascospores became surrounded by a fibro-granular layer. Small, membrane-bounded vesicles and larger electron-dense membrane-bounded vesicles aggregated at the site of germ tube formation where the plasmalemma adjacent to the aggregation was convoluted. The vesicles appeared to fuse with the plasmalemma, releasing their contents. Enzymatic digestion of the spore wall probably occurred at the time of germ tube emergence. After the nucleus had migrated into the newly formed germ tube, a septum was formed to delimit the germ tube from the ascospore. The growing germ tube can be divided into 3 morphological regions, namely the apical, sub-apical and vacuolated regions, and is typical of other fungi. A mucilaginous sheath was associated with the older mycelium. The germ tube displaced the polar appendage, and the ascospore, germ tube and appendage were enclosed in a mucilaginous sheath. In ascospores which subtended old germ tubes, the nucleus and lipid body became irregular in shape and the cytoplasm was more vacuolated. Microbody-like structures remained associated with the lipid throughout development, and were present in old ascospores.  相似文献

Information on the diversity and ecology of arctic marine fungi is lacking. During a short visit to Longyearbyen (78 degrees 13'N 15 degrees 33'E), Svalbard, Norway, a new marine fungus growing on driftwood collected at the shore was encountered. This taxon belongs to the Halosphaeriales (Ascomycota), a fungal order of mostly marine species. Havispora longyearbyenensis gen. et sp. nov. is morphologically similar to Nautosphaeria and Nereiospora, all with tufts of appendages at polar and equatorial positions of the ascospore but differing in color and septation of the ascospore and morphology and ontogeny of the ascospore appendage.  相似文献

A new ascomycete fungus, with long-necked perithecia having central ostioles and striate ascospores, was isolated from flowerheads of Protea burchellii and P. laurifolia in South Africa and is described here as Rhynchostoma proteae sp. nov. Sequence data obtained from the small-subunit ribosomal DNA (SSU nrDNA) place this fungus with 100% bootstrap support in a clade containing the type species of Rhynchostoma, R. minutum. A similar fungus with verruculose ascospores also was observed on a member of the Proteaceae from Australia, Lomatia polymorpha, which is described here as Rhynchomeliola lomatiae sp. nov. These two species are illustrated and contrasted with a third species from Proteaceae, Rhynchomeliola australiense, known from Grevillea in Australia.  相似文献

Upon cultivation of the yeast Dipodascopsis tothii in its sexual stage, small ascospores are released individually from the ascus tip, which then assemble in sheathed cluster balls. In contrast to Dipodascopsis uninucleata, this yeast produced smooth bean shaped ascospores with sheath-like appendages that assemble in a disordered sheathed ball of ascospores outside the ascus. Strikingly, upon release, the ascus tip contained 3-hydroxy oxylipins, while the released ascospore clusters contained little or no 3-hydroxy oxylipins as indicated by immunofluorescence microscopy. In D. uninucleata, these oxylipins are concentrated on the spore surface and interspore matrix, but not on the ascus tip.  相似文献

We previously implicated 3-hydroxy oxylipins and ascospore structure in ascospore release from enclosed asci. Using confocal laser scanning microscopy on cells stained with fluorescein-coupled, 3-hydroxy oxylipin-specific antibodies, we found that oxylipins are specifically associated with ascospores and not the vegetative cells or ascus wall of Ascoidea corymbosa. Using gas chromatography--mass spectrometry the oxylipin 3-hydroxy 17:0 could be identified. Here, we visualize for the first time the forced release of oxylipin-coated, hat-shaped ascospores from terminally torn asci, probably through turgor pressure. We suggest that oxylipin-coated, razor-sharp, hat-shaped ascospore brims may play a role in rupturing the ascus to affect release.  相似文献

The ultrastructural features of developing and mature ascospores were delineated after mating Arthroderma quadrifidum on pablum cereal agar. Incipient ascospores each contained a granulated nucleus bounded by a nuclear envelope while presumptive ascospore cytoplasm was bounded by a double membrane and resided in glycogen-rich epiplasm of the ascus. Mature ascospores contained nuclei and mitochondria while the ascus epiplasm still retained abundant inclusions. The ascospore wall demonstrated the presence of heterogeneous material between the plasmalemma and the outer spore membrane which appeared smooth.  相似文献

Ascospore development inCeratocystis fimbriata Ell. & Halst. commenced in an eight-nucleate ascus. A single vesicle formed along the periphery of the ascus from fragments of ascospore delimiting membranes, surrounded all eight nuclei and eventually invaginated, first forming pouches with open ends, then finally enclosing each of the eight nuclei in a separate sac, thus delimiting ascospores. Pairing of the ascospores followed and brim formation occurred at the contact area between two ascospores. Osmiophilic bodies contributed to the formation of brim-like appendages by fusing to the ascospore walls. Ascospore characters have commonly been used for distinguishing ascomycete taxa, while ascomal wall characters have received little attention. Although taxa in the Sordariales possess a wide range of variation in their ascomal walls and ascospores, genera have traditionally been delimited based on differences in their ascospore morphology. Phylogenetic relationships of multiple representatives from each of several genera representing the range in ascomal wall and ascospore morphologies in the Sordariales were estimated using partial nuclear DNA sequences from the 28S ribosomal large subunit (LSU), beta-tubulin, and ribosomal polymerase II subunit 2 (RPB2) genes. These genes also were compared for their utility in predicting phylogenetic relationships in this group of fungi. Maximum parsimony and Bayesian analyses conducted on separate and combined data sets indicate that ascospore morphology is extremely homoplastic and not useful for delimiting genera. Genera represented by more than one species were paraphyletic or polyphyletic in nearly all analyses; 17 species of Cercophora segregated into at least nine different clades, while six species of Podospora occurred in five clades in the LSU tree. However, taxa with similar ascomal wall morphologies clustered in five well-supported clades suggesting that ascomal wall morphology is a better indicator of generic relationships in certain clades in the Sordariales. The RPB2 gene possessed over twice the number of parsimony-informative characters than either the LSU or beta-tubulin gene and consequently, provided the most support for the greatest number of clades.  相似文献

A fungus belonging to the Rhytismatales found on twigs of Rhododendron lutescens in Yunnan, southwestern China, is described as a new species in a new genus. It is characterized by Coccomyces-like ascomata but differs in having elliptical ascospores with filiform, hyaline appendages. The ascospores of this new taxon are somewhat similar to those of species of Parvacoccum, but the latter has symmetrical, fusiform ascospores with funnel-shaped appendages. The new genus also is distinct from Hypoderma, Hypodermella, Myriophacidium, Ploioderma, Neococcomyces and Therrya.  相似文献

Immature asci of Coniochaeta tetraspora originally contain eight uninucleate ascospores. Two ascospore pairs in each ascus survive and mature, and two die and degenerate. Arrangement of the two ascospore types in individual linear asci is what would be expected if death is controlled by a chromosomal gene segregating at the second meiotic division in about 50% of asci. Cultures originating from single homokaryotic ascospores or from single uninucleate conidia are self-fertile, again producing eight-spored asci in which four spores disintegrate, generation after generation. These observations indicate that differentiation of two nuclear types occurs de novo in each sexual generation, that it involves alteration of a specific chromosome locus, and that the change occurs early in the sexual phase. One, and only one, of the two haploid nuclei entering each functional zygote must carry the altered element, which is segregated into two of the four meiotic products and is eliminated when ascospores that contain it disintegrate. Fusion of nuclei cannot be random-a recognition mechanism must exist. More study will be needed to determine whether the change that is responsible for ascospore death is genetic or epigenetic, whether it occurs just before the formation of each ascus or originates only once in the ascogonium prior to proliferation of ascogenous hyphae, and whether it reflects developmentally triggered alteration at a locus other than mating type or the activation of a silent mating-type gene that has pleiotropic effects. Similar considerations apply to species such as Sclerotinia trifoliorum and Chromocrea spinulosa, in which all ascospores survive but half the spores in each ascus are small and self-sterile. Unlike C. tetraspora, another four-spored species, Coniochaetidium savoryi, is pseudohomothallic, with ascus development resembling that of Podospora anserina.  相似文献

The heat resistance ( D and z values) of four Saccharomyces spp., viz. S. bailii, S. cerevisiae, S. chevalieri, S. uvarum , was investigated by a previously described method (Put et al. 1977). The highest heat resistance was observed in ascospores of S. cerevisiae 175 showing a decimal reduction time or death rate constant at 60°C of 22.5 minutes. The average D 60°C values of the remaining species were: S. bailii 10 min; S. chevalieri 13 min; and S. uvarum 1.5 min; while z values varied between 4.0 and 6.5°C. It can be postulated that there may exist some comparison between the mechanism of yeast ascospore and bacterial endospore heat resistance. However, (i) washed Saccharomyces ascospores cannot be stored at 5°C for a longer period than 2–3 weeks without loss of heat resistance; and (ii) ascospore heat survival curves are essentially not linear over the whole range, which may be due to some heterogeneity of the ascospore population tested. Besides, it was observed that the heat resistance ( D 60°C values) of the Saccharomyces ascospores proved to be 50–150-fold higher than the D 60°C values of the corresponding vegetative cells.  相似文献
