The genus Powellomyces was described to accommodate two monocentric chytrid species from soil that develop exogenously and possess zoosporic ultrastructure similar to other members of the order Spizellomycetales. Despite Powellomyces-like chytrids being commonly observed in gross culture, the genus contained only two species. To determine diversity in this group I amassed 49 isolates of Powellomyces-like chytrids, including the cultures upon which species types were based and new isolates from pollen-baited water cultures of soils, plant detritus and manure. I sequenced portions of nucSSU and nucLSU rDNA regions and the EF-1α-like gene from each isolate to produce a molecular phylogeny. This phylogeny supports monophyly of spizellomycetalean chytrids with exogenous development and suggests that multiple distinct lineages exist within this group. This phylogeny, along with a reevaluation of the ultra-structural features of the two described species, supports the recognition of a new family, Powellomycetaceae, and genus, Geranomyces, which contains 31 isolates of G. variabilis comb. nov.  相似文献

During the last decade, our concept of chytrid systematics has dramatically changed based on molecular phylogeny and zoospore ultrastructure. In contrast with well-studied saprotrophic chytrids, only a few obligate parasitic chytrids have been investigated with modern methods. Here, we investigate the novel chytrid culture KS93 that is parasitic on the diatom Aulacoseira granulata. Thallus morphology of KS93 was characterized by a spherical, stalked zoosporangium with a single, apical inoperculate discharge pore and zoospore discharge as a mass in a vesicle. A cross-inoculation experiment revealed that the infection of KS93 was specific to A. granulata. Zoospores of KS93 possessed the characters of the Group I type zoospore of the Chytriomycetaceae in the Chytridiales, but does not appear to have a paracrystalline inclusion. Additionally, KS93 also possessed a globule-type KAS (kinetosome-associated structure), first reported here for members of Chytriomycetaceae. In our molecular phylogeny, KS93 was placed in the basal position of the Chytriomycetaceae and was distinguished from any known species in the family. Morphological features of KS93 were distinct from those of any other taxa in the Chytriomycetaceae and from any described chytrids. Based on these results, we describe this chytrid as Pendulichytrium sphaericum gen. et sp. nov. in the family Chytriomycetaceae.  相似文献

The newest order in Chytridiomycota, Rhizophydiales, shows remarkable genetic divergence and zoospore ultrastructural diversity. From submersed mud from a marsh near an oligotrophic lake in the Patagonia region of Argentina, we isolated a previously undescribed chytrid in Rhizophydiales with unique 28S rRNA and ITS1-5.8S-ITS2 sequences and a distinctive zoospore and thallus. In a combined LSU + 5.8S phylogeny of representative taxa in the order, the nearest relative to this new species is Rhizophlyctis harderi, and their zoospores share several similar character states. A new genus is erected for this chytrid, the first to be brought into pure culture from this region.  相似文献

While surveying chytrid diversity in lakes and streams, we found on cellulosic bait a chytrid that had both monocentric and polycentric thallus forms. We brought this chytrid into axenic culture from three sites in eastern North America, studied its thallus development and zoospore ultrastructure, and compared its 28S rDNA sequence with those of other members of the Chytridiomycota. Thallus morphology matched that described for the rare chytrid, Cladochytrium polystomum Zopf. Sporangia were spherical and produced numerous long discharge tubes. After discharge, zoospores remained in spherical clusters at the tips of the inoperculate openings of discharge tubes. After 10–30 min zoospores either swam away or encysted in place. Zoospore ultrastructural features included a cell coat, flagellar plug, and paracrystalline inclusion, features typical of members of the Chytridiales. However, the flagellar apparatus structure and organellar organization differed from that of zoospores previously described. Based on its molecular phylogeny and its zoospore ultrastructural features, we classify C. polystomum as a member of the Chytridiaceae in the Chytridiales. Because its thallus development and its ribosomal DNA sequences diverged decidedly from those of Cladochytrium tenue Nowak, the type species of Cladochytrium, we erected Zopfochytrium as a new genus for this chytrid.  相似文献

The methods of isolation and maintenance in dual clone cultureof the chytrid Rhizophydium planktonicum Canter emend, parasiticon Asterionella formosa Hass. from the plankton are described.The ability of a single fungal isolate to infect other clonesof Asterionella, Fragilaria, Tabellaria, Synedra and Cyclotella,as well as dead material was also tested. All the clones of Asterionella proved to be highly susceptibleto infection whereas only in rare instances could a sporangiumbe found which had grown and dehisced on Fragilaria and Synedra.Very few zoospores encysted on Tabellaria and Cyclotella andthose which did died without further growth. It has been shown by experimentation that two morphologicallydistinct clones of Fragilaria crotonensis Kitton, isolated fromthe plankton of Windermere at approximately the same time, wereaffected quite differently by the chytrid Rhizophydium fragilariaeCanter. In one case the diatom filaments became heavily parasitizedand could be used for the maintenance of the fungus. In theother case infection was negligible. Additional clones, representative of these two morphologicalforms, from other bodies of water in the Windermere drainagebasin were tested. The same pattern of infection or relativenon-infection again ensued in direct accordance with the particulartype of filament used. Some preliminary experiments undertaken with a second, as yetlittle known chytrid parasite of F. crotonensis are reported.In this instance the fungus appeared to find its natural hoston cells belonging to the morphological form which failed tosupport the continued increase of R. fragilariae. The parasitism of morphological variants of F. crotonensis bythese fungi in the wild from Britain and else where is discussed. Further detailed experiments under laboratory conditions havebeen carried out using clones of Fragilaria crotonensis Kitton.Two morphologically distinct types (rod- and flared-cell forms)were investigated in relation to their infection by a chytridreferred to as species 3. The pathogenicity of the chytrid Rhizophydiumfragilariae Canter towards four newly isolated clones of F.crotonensis was also studied. The results in general confirmedthat clones of the diatom composed of flared-type cells werecompatible with the development and increase of species 3 butremained more or less resistant to R. fragilariae. In contrast,rod-celled type clones showed exactly the opposite host parasiterelationships. Further study has shown that slight differencesin susceptibility towards a particular chytrid can occur withindiatom clones of similar morphological type. The presence ofhypersensitive algal cells was noted in relation to parasitismby both fungi. Some observations are recorded on the developmentof mucilage around filaments of the Fragilaria clones. A retrospective survey was performed of aerobic bacterial species found in the vagina of 203 bitches with genital disorders, e.g. infertility, vaginitis, pyometra and puppy death. Escherichia coli, beta-hemolytic streptococci, Staphylococcus intermedius and Pasteurella multocida were the species most often isolated. From bitches with pyometra E. coli in pure culture was the most frequent isolate. In contrast, the majority of infertile bitches gave rise to mixed cultures, and no specific bacterial species was consistently associated with infertility. Thus, bacterial sampling from infertile bitches was concluded to be of low diagnostic value. Bacterial species isolated from the bitches having vaginitis were present in pure culture in 26.9% of the samples while nonspecific mixed cultures were obtained from 34.6% of the samples from these bitches. E. coli was the most frequently isolated bacterial species from bitches with dead puppies. However, in such cases it is important to relate the vaginal bacterial findings to autopsy findings and the results of bacteriological cultures of the pups.  相似文献

Results of study of Leptomonas nabiculae using various molecular markers and different material (cultures D2 et Nfm2) contradicted each other and taxonomic position of this species. We investigated morphology of the cells in these cultures as well as in hapantotype of L. nabiculae and those of L. peterhoffi and L. occidentalis that had been described from the same host species. We also reconstructed 18S rRNA gene phylogeny using sequences from both cultures. The D2 culture according to its morphology and phylogenetic position revealed to be a Crithidia that had accompanied L. nabiculae in a mixed infection. We named it Crithidia dedva. The cells in the hapantotypes of the three Leptomonas species and those of the Nfm2 culture represent a single species that is a Herpetomonas (H. nabiculae) judging by morphology and molecular phylogeny. We also showed that the sequence of 18S rRNA gene that had been formerly determined represents a chimaera. This had resulted in the wrong position of this species on the phylogenetic tree that had contradicted results of the analysis of 5s rRNA gene.  相似文献

5' tRNA editing has been demonstrated to occur in the mitochondria of the distantly related rhizopod amoeba Acanthamoeba castellanii and the chytridiomycete fungus Spizellomyces punctatus. In these organisms, canonical tRNA structures are restored by removing mismatched nucleotides at the first three 5' positions and replacing them with nucleotides capable of forming Watson-Crick base pairs with their 3' counterparts. This form of editing seems likely to occur in members of Amoebozoa other than A. castellanii, as well as in members of Heterolobosea. Evidence for 5' tRNA editing has not been found to date, however, in any other fungus including the deeply branching chytridiomycete Allomyces macrogynus. We predicted that a similar form of tRNA editing would occur in members of the chytridiomycete order Monoblepharidales based on the analysis of complete mitochondrial tRNA complements. This prediction was confirmed by analysis of tRNA sequences using a tRNA circularization/RT-PCR-based approach. The presence of partially and completely unedited tRNAs in members of the Monoblepharidales suggests the involvement of a 5'-to-3' exonuclease rather than an endonuclease in removing the three 5' nucleotides from a tRNA substrate. Surprisingly, analysis of the mtDNA of the chytridiomycete Rhizophydium brooksianum, which branches as a sister group to S. punctatus in molecular phylogenies, did not suggest the presence of editing. This prediction was also confirmed experimentally. The absence of tRNA editing in R. brooksianum raises the possibility that 5' tRNA editing may have evolved twice independently within Chytridiomycota, once in the lineage leading to S. punctatus and once in the lineage leading to the Monoblepharidales.  相似文献

Zoospores of the marine chytrid Rhizophydium littoreum are attracted to a variety of substances common to their environment. In general, carbohydrates and polysaccharides elicited strong concentration-dependent positive responses. There was no direct correlation between all substances used as foods and those stimulating positive responses. The chemotactic activities of this organism should, however, tend to bring it toward concentrated food sources.  相似文献

Sooty blotch and flyspeck is a fungal disease complex that colonizes a wide range of plants. In a survey of host plants for sooty blotch and flyspeck fungi, we obtained an isolate associated with discrete flyspeck symptom on stems of the Rubber Trees, Ficus elastica, from Haikou, Hainan, China. A pure culture was obtained, and the nuclear ITS region of rDNA was amplified by PCR. Molecular phylogeny inferred from ITS sequence data suggested that it was close to Devriesia strelitziae, reported from South Africa. Morphologically, it was characterized by longer conidia, more conidial septa, and having chlamydospores. Therefore, we describe our isolate as a novel species of Devriesia.  相似文献

SUMMARY 1. The effect of phosphorus limitation of the diatom Asterionella formosa Hass. on growth, survival and epidemic development of its fungal parasite Rhizophydium planktonicum Canter emend. was estimated, using measurements of production and infectivity of the zoospores of the chytrid grown on host cultures with different phosphorus-limited growth rates.
2. Phosphorus-limited host cells were less susceptible to infection with zoospores of the parasite than non-limited host cells.
3. The sporangia on phosphorus-limited algae produced substantially less zoospores, but the development time of these sporangia was only slightly reduced.
4. As a result of these effects, Rhizophydium will reach lower growth rates at a given host density, and survival of the parasite will require higher host densities when Asterionella is phosphorus-limited.
5. The zoospore production remained high enough to enable the parasite to grow faster than the alga at sufficiently high host densities, both at limiting and non-limiting phosphorus levels.
6. In spite of the reduced growth rate of the parasite, phosphorus limitation of Asterionella was found to facilitate the development of a Rhizophydium epidemic. This was a consequence of the reduced algal growth rate at phosphorus limitation, which makes the host population more easily outgrown by the parasite.
7. Phosphorus limitation of the host could reduce the threshold host density required for the development of an epidemic by a factor of 2.5.  相似文献

An isolate of the nematode-trapping fungus Monacrosporium drechsleri was collected from cultures of the root-knot nematode Meloidogyne arenaria that had been maintained on tomato roots in greenhouse pots in Beltsville, Maryland. The plant-parasitic nematodes Heterodera glycines, Meloidogyne incognita and Pratylenchus zeae and the free-living nematodes Caenorhabditis elegans and Panagrellus redivivus were placed on colonies of M. drechsleri grown in Petri dishes to study ability of the isolate to trap various nematode hosts. None of the nematodes placed near adhesive knobs were motile within 1 d. To determine where M. drechsleri fits within the existing phylogeny of nematode-trapping fungi, the ITS1-ITS2 regions of rDNA and the nuclear gene EF1-alpha were sequenced for the new isolate of M. drechsleri, for the species M. parvicolle and M. lysipagum, and for an isolate of M. ellipsosporum distinct from the one listed in GenBank. Parsimony trees were constructed showing the closest molecular relative of M. drechsleri to be the newly sequenced isolate of M. ellipsosporum; the latter had a highly divergent sequence from the sequence recorded in GenBank for a different isolate of M. ellipsosporum. Unique, consistent and discrete morphological characters are absent in these related taxa, so an independent molecular character should be considered essential for their accurate identification.  相似文献

Illus. 1962.—A method for growing pure cultures of many of the single or few-celled phycomycetous fungi such as Rhizidiomyces is described. This method facilitates both qualitative and quantitative study of such fungi during their growth and development. Zoospores of Rhizidiomyces start to grow within 3 hr after being placed in a liquid medium, and are ready to discharge new zoospores after about 40 hr growth at 25 C. The single nucleus of the zoospore goes through many synchronous mitotic divisions during the development of the mature multinucleate cell. Under carefully controlled environmental conditions, sporangium discharge occurs in a definite and predictable manner. Observations suggesting a possible mechanism of sporangium discharge are presented and discussed.  相似文献
