The basidiomycete genus Galerina Earle accommodates more than 300 small brown-spored agarics worldwide, predominantly described from the Northern hemisphere. The delimitation of species and infrageneric units hitherto has been based on morphological and, to some extent, ecological characters. In this study we have analyzed nuclear ribosomal LSU and ITS sequences to reveal infrageneric phylogeny and the phylogenetic placement of Galerina among the dark-spored agarics. Sequences from 36 northern hemisphere Galerina species and 19 other dark-spored taxa were analyzed, some of them obtained from EMBL/GenBank. Our results, received from Bayesian and distance methods, strongly suggest that Galerina is a polyphyletic genus. The LSU analysis shows that Galerina is composed of three or four separate monophyletic main groups. In addition, a few species cluster together with other dark-spored agarics. The same groups are recognized in the ITS tree and they correspond roughly to previously recognized subgenera or sections in Galerina. With high support our LSU analysis suggests that Gymnopilus is a monophyletic genus and that Gymnopilus and one of the Galerina lineages ("mycenopsis") are sister groups. The analyses further indicate that the Galerina lineages, as well as the genus Gymnopilus, could be referred to a strongly emendated family Strophariaceae, which corresponds largely to the family as circumscribed by Kühner (1980). Our results affirm that morphological characters often are highly homoplastic in the agarics. At the present stage formal taxonomic consequences or nomenclatural changes are not proposed.  相似文献

The new species Gymnopilus maritimus is described from coastal plant communities of Juncus maritimus, growing on sandy soil or on decaying plants, from northwestern Sardinia (Italy). The distinguishing features of G. maritimus are: (1) an unusual habitat, (2) robust basidiomata, (3) mild taste, and (4) big and strongly warted spores. The new species is compared with the micromorphologically similar species G. fulgens sensu auct. Brit. p.p. and the biogeographically and ecologically similar species G. arenophilus, as well as with other European species. A photograph of fresh material, drawings of the main micromorphological features, and FESEM and optical microscope microphotographs of basidiospores are added. Furthermore, some notes on micromorphological characters of G. arenophilus are presented and its distribution area enlarged with a record from France. A key for the European species of Gymnopilus morphologically, ecologically, and/or biogeographically related to G. maritimus is presented. The phylogeny inferred from ITS rDNA sequences revealed that G. maritimus represents an independent species and that it is not related to G. arenophilus or G. fulgens. It is the sister group of the clade containing G. imperialis and G. spectabilis, but with a bootstrap support below 50%. The characters shared by the species in this clade are: (1) robust basidiomata, (2) pileus fibrillose or scaly-fibrillose, and (3) spores longer than 8 μm, dextrinoid and strongly warted. Gymnopilus imperialis and G. spectabilis differ by the basidiomata with membranous ring in the stem, living on conifers or decaying wood, and having narrower or wider spores, respectively. 四照花亚属(Cornus subg.Syncarpea)隶属于山茱萸科山茱萸属(Cornus),我国该亚属共有5种8亚种。为探讨四照花亚属nrDNA ITS序列的致同进化不完全现象及假基因产生的可能原因,分析了该亚属4种(每种1~2个居群)共21个个体的nrDNA ITS序列。结果表明,这些类群的nrDNA ITS存在多态性,通过分析这些nrDNA ITS克隆序列的G+C含量、5.8S保守基序和二级结构最小自由能,推测其可能存在假基因。系统发育研究结果显示所有nrDNA ITS序列分成5个分支,同一个体的不同拷贝被分别置于两个甚至多个分支中,且不同分支显示了不同种间关系。四照花亚属物种个体内部存在nrDNA ITS不完全致同进化,可能归咎于不完全的世系分选(incomplete lineage sorting)、种间杂交或多倍化等进化事件,从而导致基因组内nrITS区序列出现多态性,同时也导致难以通过外部形态来划分亚属内种间界限。  相似文献

Phylogenetic hypotheses for the pantropical marine green algal genus, Caulerpa , were inferred based on analyses of nuclear-encoded rDNA internal transcribed spacer (ITS) sequences. Results of these analyses were used to assess the correspondence between rDNA phylogeny and traditional sectional taxonomy, to identify synapomorphic morphological characters (including assimilator morphology and chloroplast ultrastructure), and to examine marine biogeographic hypotheses for the genus. Ribosomal DNA ITS sequences were aligned for thirty-three species and intraspecific taxa of Caulerpa. Results indicate limited correspondence between phylogeny and sectional taxonomy for the genus, (e.g., the sections Filicoideae and Sedoideae were not monophyletic). In contrast, chloroplast morphology could be mapped to the tree topology with limited homoplasy. Pantropical isolates of the filicoidean species, Caulerpa sertularioides and Caulerpa mexicana each formed monophyletic groups. Caulerpa reyesii was included as a derived taxon within the Caulerpa taxifolia clade, suggesting that these species were conspecific and affirmed the lack of correspondence between phylogeny and assimilator morphology. Isolates and various intraspecific taxa of Caulerpa racemosa did not form a monophyletic group. Instead, these taxa formed a heterogeneous assemblage with other sedoidean and filicoidean taxa. Within the C. sertularioides clade, Caribbean and Atlantic isolates formed a basal paraphyletic group, whereas eastern and western Pacific isolates formed a more derived monophyletic group. Therefore, these results are not consistent with an Indo-West Pacific origin of this species.  相似文献

The internal transcribed spacers ITS1 and ITS2 of the nuclear ribosomal DNA (rDNA) have recently been found to display remarkable intraspecific polymorphism, a feature suggested as limiting their value for phylogenetic reconstructions. A comparative study of oligonucleotide motives and intraindividual nucleotide variability across all species of the tree genus Fagus (beech) shows, however, that this intraspecific ITS polymorphism follows a particular pattern, which can be used to detect reticulation and ancient polymorphism within the genus. Coding ITS polymorphisms as phylogenetically informative characters, moreover, resulted in better‐resolved phylogenies than traditional ‘base‐per‐base’ maximum parsimony and maximum likelihood analyses.  相似文献

Phylogenetic relationships of 54 European Phlegmacium species, including members of most of the sections of classical systematics, were studied, integrating macro-, micromorphological and chemical characters of the basidiomes, as well as molecular phylogenetic analysis of nuclear rDNA sequences. Microscopical structures of the basidiomes were studied by light microscopy. Basidiospore morphology was examined by scanning electron microscopy. Internal-transcribed spacers (ITS 1 and 2, including the 5.8S) and the D1/D2 (LSU) regions of nuclear rDNA were sequenced and analyzed with a Bayesian Markov chain Monte Carlo approach. Many subgroups detected by the molecular analysis are related to groups known from classical systematical concepts. Among others, these subgroups were significantly supported: i) a group containing most of the members of section Fulvi ss. Brandrud and the species Cortinarius arcuatorum, C. dibaphus and C. multiformis; ii) a group comprising taxa of section Calochroi ss. Brandrud and the species C. fulvocitrinus and C. osmophorus; iii) a group containing species of section Glaucopodes ss. Brandrud and C. caerulescens; iv) a group including members of section Phlegmacioides ss. Brandrud; v) a group that includes the species C. cephalixus, C. nanceiensis and C. mussivus. Stipe shape, color of flesh, pigment contents, KOH reaction on pileipellis and gelatinous layer, degree of development of a gelatinous layer on the pileipellis, and pileipellis structure were useful characters in delimiting subgroups in Phlegmacium, while basidiospore morphology was significant at species level. With the exception of C. glaucopus, C. infractus and C. scaurus, ITS and D1/D2 sequences obtained from collections of the same species from different geographical origins showed very little variation. Our molecular and morphological analyses suggest revisions of the traditional concepts of the subgenus Phlegmacium in Europe.  相似文献

The internal transcribed spacers (ITSs) of nuclear ribosomal DNA have been sequenced for 20 species of Gentiana. By incorporating previously released sequence data of eight species, phylogenelic analyses using Fitch parsimony and character-state weighted parsimony were carried out. The length of ITS 1 in the taxa surveyed ranged from 223 to 238 bp and ITS2 from 216 to 234 bp. Sequence divergence between pairs of species ranged from 5.0% to 48.9% in ITS1, from 1.1% to 45.3% in ITS2, and from 3.2% to 46.1% in combined data of ITS1 and ITS2. The ITS phylogeny was generally congruent with morphological classifications except that G. asclepiadea was revealed to be closely related to section Gentiana instead of section Pneumonanthe and section Stenogyne was shown to be a paraphyletic group of the genus Gentiana that would be better excluded from the genus. A divergence among the three European endemic sections and the remaining sections of the genus other than section Stenogyne was revealed. Thus the European species of the genus together do not form a monophyletic group. A close relationship between the sections Chondrophyllae s. l. (including section Dolichocarpa), Cruciata and Pneumonanthe was suggested. The section Frigidae s. l. (including sections Monopodiae, Isomeria, Microsperma, and Phyllocalyx) contained two well-supported clades: section Frigidae s. str. and all others together. The monophyly of the typically dysploid group section Chondrophyllae s. l. was confirmed. Optimization of chromosome numbers on the ITS phylogeny suggested that 2/1 = 26 is a plesiomorphic state for the clade comprising sections Frigidae s. l., Cruciata, Pneumonanthe, and Chondrophyllae s. l., and probably 2n = 20 is a plesiomorphic state for the dysploid group, section Chondrophyllae s. l.  相似文献

Gymnopilus turficola sp. nov. is described. This species is characterized by its habitat in palsa mires growing on peat in subarctic areas, by greenish lamellae in young state and the development of an iodoform odour when kept closed for some hours. Psilocin, psilocybin or related compounds could not be detected. G. turficola seems closely related to G. odini. According to the analysis of ITS sequences this species falls into a well supported clade of closely related taxa with G. odini, G. decipiens and G. penetrans.  相似文献

The genus Glyphochloa (Poaceae: Panicoideae: Andropogoneae: Rottboellinae) is endemic to peninsular India and is distributed on lateritic plateaus of low and high altitude in and around Western Ghats and the Malabar Coast. The genus presumably originated and diversified in the Western Ghats. Species relationships in the genus Glyphochloa were deduced here based on molecular phylogenies inferred using nuclear ribosomal ITS sequences and plastid intergenic spacer regions (atpB-rbcL, trnT-trnL, trnL-trnF), and new observations were made of spikelet morphology, caryopsis morphology and meiotic chromosome counts. We observed two distinct clades of Glyphochloa s.l. One of these (‘group I’) includes Ophiuros bombaiensis, and is characterized by a single-awned lower glume and a base chromosome number of 6; it grows in low elevation coastal areas. The other clade (‘group II’) has a double-awned lower glume, a base chromosome number of 7, and is restricted to higher elevation lateritic plateaus; G. ratnagirica may belong to the group II clade, or may be a third distinct lineage in the genus. A sister-group relationship between group I and II taxa (with or without G. ratnagirica) is not well supported, although the genus is recovered as monophyletic in shortest trees inferred using ITS or concatenated plastid data. The nuclear encoded internal transcribed spacer (ITS) region and the plastid encoded trnL-F region were sequenced for 25 populations of Korthalsella, a genus of reduced, monoecious, Old World misletoes. The molecular study confirms the hypothesis that branch shape and cladotaxy (the arrangement of branches with respect to their parent axis) are unreliable indicators of relationship in the genus and demonstrates that many of the taxa previously recognized are not monophyletic. Both gene regions identify three major subgroups within the genus and find lower level relationships within these subgroups highly correlated with geographic distance. An analysis based upon 18S and rbcL sequences identifies Ginalloa as the sister group to Korthalsella, which together with the branching order within the genus, indicates that Korthalsella originated in Papuasia and aids in elucidating evolution of the peculiar inflorescence structure. There are problems associated with species delimitation when evolutionary units are more restricted than morphological lineages, and justification is offered for recognizing only morphologically diagnosable monophyletic lineages as species. Varying substitution rates and differing modes of inheritance in ITS and trnL-F result in complementary utility of the two regions for elucidating infrageneric relationships in Korthalsella.  相似文献

The internal transcribed spacer sequences spanning the regions between the 17S and 25S rRNAs (ITS1 and ITS2) and including the complete sequence of the 5.8S rRNA were used for phylogenetic analyses. This approach to define phylogenetic relationships within the genus Tricholoma was tested using different isolates of T. terreum. Fruitbodies identified in nature were analysed in order to allow use of morphology for taxonomy. The isolates from different locations were closely related as could be expected for one species. Thus, the method could be applied to different Tricholoma species. Three clusters within the genus Tricholoma can be distinguished with four additional species not included in any of these clusters. Molecular analyses of two Cortinarius species confirm a phylogenetically distinct genus.  相似文献

To help resolve phylogenetic relationships among the mitten crabs, complete sequences of the nuclear DNA internal transcribed spacer (ITS) and portions of the mitochondrial genome corresponding to the cytochrome oxidase I (COI), were sequenced for all Asian mitten crabs of the genus Eriocheir and seven species of the Grapsoidea. The resulting phylogeny supports the establishment of a separate genus Neoeriocheir, but does not provide justification for the recognition of Platyeriocheir. A female mitten crab specimen from the Zhujiang River, China, was considered to be Eriocheir recta (), a species previously synonymized with Eriocheir japonica (de Haan, 1835). In the ITS analysis, a sequence from Eriocheir formosa (from Taiwan) falls within a well-supported E. recta group, which indicates that E. formosa may have to be synonymized with E. recta. Three previously recognized members of the genus, E. japonica, Eriocheir sinensis, and Eriocheir hepuensis constitute a monophyletic sister group to E. recta in all phylogenetic trees. We provide evidence for the conspecific status of these taxa. Phylogenetic trees based on COI and combined COI and ITS sequences indicate that E. japonica consists of three subgroups. Since the name E. japonica (de Haan, 1835) takes precedence over E. sinensis (H. Milne Edwards, 1853) and E. hepuensis, we suggest that these three subgroups correspond to three subspecies of E. japonica: E. j. japonica, E. j. sinensis, and E. j. hepuensis.  相似文献

As a result of conventional characterization of yeasts isolated from various plant leaves collected in Yunnan, China, six ballistoconidium-forming strains with orange-colored colonies were grouped together. Molecular phylogenetic analysis based on 18S rDNA sequencing showed that two representative strains of this group of yeasts, CH 2.068 and CH 2.497, were closely related to the species in the genus Dioszegia and had signature sequences typical of this genus. However, the six strains from Yunnan differed from the described Dioszegia species remarkably (14.5-17.7% nucleotide divergences) in the ITS (internal transcribed spacer) region sequences, which indicated that they represent a distinct species. Furthermore, among the six strains studied, the ITS region sequence comparison allowed the recognition of two subgroups represented by CH 2.068 and CH 2.497, which differ from each other in three bases in the ITS 2 region. DNA-DNA relatedness revealed that the two subgroups represent two varieties of a new species in the genus Dioszegia, for which Dioszegia zsoltii sp. nov. var. zsoltii and Dioszegia zsoltii var. yunnanensis var. nov. are proposed.  相似文献

Phylogenetic relationships within the genus Cladonia , including Cladina (Cladoniaceae, Lecanoromycetes), were reconstructed based upon simultaneous analyses of DNA sequences and morphological and chemical data. We used sequences from the internal transcribed spacer 1 (ITS1), the 5.8 rDNA gene, and the internal transcribed spacer 2 (ITS2) of the nuclear rDNA gene cluster, and partial sequences from the protein-coding β-tubulin gene. The analyses included 235 specimens of 168 taxa representing all currently recognized sections of Cladonia and Cladina and the outgroup genera Cladia, Pycnothelia , and Ramalea . Analyses were performed using optimization alignment with three different parameter values. The results of all analyses support the inclusion of Cladina in Cladonia . The current sectional division of Cladonia was not supported, and a new provisional classification for the genus is proposed.  相似文献

Previous systematic arrangement on the ciliate order Urostylida was mainly based on morphological data and only about 20% taxa were analyzed using molecular phylogenetic analyses. In the present investigation, 22 newly sequenced species for which alpha-tubulin, SSU rRNA genes or ITS1-5.8S-ITS2 region were sampled, refer to all families within the order. Following conclusions could be drawn: (1) the order Urostylida is not monophyletic, but a core group is always present; (2) among the family Urostylidae, six of 10 sequenced genera are rejected belonging to this family; (3) the genus Epiclintes is confirmed belonging to its own taxon; (4) the family Pseudokeronopsidae undoubtedly belongs to the core portion of urostylids; however, some or most of its members should be transferred to the family Urostylidae; (5) Bergeriellidae is confirmed to be a valid family; (6) the distinction of the taxon Acaudalia is not supported; (7) the morphology-based genus Anteholosticha is extremely polyphyletic; (8) ITS2 secondary structures of Pseudoamphisiella and Psammomitra are rather different from other urostylids; (9) partition addition bootstrap alteration (PABA) result shows that bootstrap values usually tend to increase as more gene partitions are included.  相似文献

Discussed in the present paper is the systematic position of Beesia, a small ranunculaceous genus mainly distributed in SW China. Sequences of the internal transcribed spacers (ITS) and 3’ end of 5.8 S rRNA gene of Beesia calthifolia, Trollius chinensis, Cimicifuga acerina, C. brachycarpa and Actaea asiatica were determined by direct PCR sequencing method. The sequences of ITS-1 in the five species range from 225 bp to 232 bp in size and those of ITS-2 from 201 bp to 217 bp. Beesia is very similar to Cimicifuga and Actaea not only in size, sequence and G+C content of ITS, but also in sequence of 3’ end of 5.8 S rRNA gene. In PAUP analysis, Ranunculus enysii was used as outgroup, and the most parsimonious tree was obtained through exhaustive search. The gaps were treated respectively as missing characters and the fifth base in two analyses. The two analyses show that a monophyletic group comprising Beesia calthifolia, Cimicifuga acerina, C. brachycarpa and Actaea asiatica is strongly supported by the bootstrap value. In the monophyletic group, Beesia calthifolia is basal to the other three species. The present DNA sequence analysis demonstrates that the genus Beesia should be placed in the tribe Cimicifugeae, which is consistent with the results from phytochemistry, palynology and cytology. In addition, Beesia may be the most original genus in the tribe Cimicifugeae in view of simple leaf,apetalous flower and molecular evidence.  相似文献

Evolutionary relationships within Astragalus L. (Fabaceae) were inferred from nucleotide sequence variation in nuclear ribosomal DNA of both New World and Old World species. The internal transcribed spacer regions (ITS) of 18S–26S nuclear ribosomal DNA from representatives of 26 species of Astragalus, three species of Oxytropis DC., and two outgroup taxa were analyzed by polymerase chain reaction amplification and direct DNA sequencing. The length of the ITS 1 region within these taxa varied from 221 to 231 bp, while ITS 2 varied in length from 207 to 217 bp. Of the aligned, unambiguous positions, approximately 34% were variable in each spacer region. In pairwise comparisons among Astragalus species and outgroup taxa, sequence divergence at these sites ranged from 0 to 18.8% in ITS 1 and from 0 to 21.7% in ITS 2. Parsimony analyses of these sequences resulted in a well-resolved phylogeny that is highly concordant with previous cytogenetic and chloroplast DNA evidence for a major phylogenetic division in the genus. These data suggest that the New World aneuploid species of Astragalus form a monophyletic but morphologically cryptic group derived from euploid species of Old World (Eurasian) origin, which are consequently paraphyletic.  相似文献
