以从芬兰引进的野生大伏革菌、中国的大伏革菌和小孔异担子菌为研究对象,通过平板对峙培养方法,从16株大伏革菌菌株中筛选出3株防治异担子菌的高效菌株,分别为:04052、08076和08077。这3号菌株具有较高的生长速率和拮抗率,能够快速覆盖病原菌,起到显著的生防效果。实验中还发现大伏革菌生长速率和拮抗率之间存在着显著的正相关关系。  相似文献

Polymerase chain reaction-amplified and sequenced isolates of Antrodia gossypium, Phlebiopsis gigantea and Heterobasidion parviporum from decaying Norway spruce wood blocks after three and six months, which exhibited linear growth, were investigated. P. gigantea strains showed the fastest growth, whereas A. gossypium growth was five times slower. The differences between the mean daily increment of A. gossypium and the other examined isolates (except Hp2) were statistically significant. There were also significant differences in wood decay between densities over time. These results were confirmed by the decay acceleration index (DAI) and decay activity index, which were positively correlated with wood density regardless of the fungus species. The registered P. gigantea strains (Rotstop and PG Suspension) exhibited a strong decomposition ability (28% after six months); the weight loss caused by A. gossypium after six months of decay (15.2%) was similar to the results of P. gigantea (GB) after just three months (13.2%). All tested H. parviporum isolates showed rather rapid growth and equally strong wood decay (20–25%) compared to those of P. gigantea. DAI showed that A. gossypium may significantly contribute to wood decomposition over time, particularly in less dense wood samples. The use of both saprotrophs as biological agents against root pathogens is discussed.  相似文献

As the same machinery might be used in applying biocontrol agents Chondrostereum purpureum and Phlebiopsis gigantea to tree stumps, we studied whether the efficacy of one agent is affected by traces of the other present in the treatment suspension. The activity of cellulase, peroxidase, phosphatase and dehydrogenase enzymes, together with the content of protocatechuic and vanillic acids, in samples of Norway spruce wood inoculated with 17 different isolates of Phlebiopsis gigantea was measured. The same isolates were used to compare decay activity in samples of Norway spruce wood after incubation for 3 and 6 months. Significant differences in enzyme activity and phenol production were found between aerial mycelium overgrowing the wood sample and the underlying wood. These differences indicated that the nature of the fungal mycelium appears to change depending on whether it is in contact with wood. After 6 months, highly extensive decomposition of the wood was shown by two British isolates. The results confirm a large difference in P. gigantea inoculum among isolates in natural conditions and reinforce the need for constant evaluation of the most active isolates to use in preparations for biocontrol: a problem for both users and registration bodies.  相似文献

The objective of this study was to evaluate the in vitro mycelium growth of Pleurotus abieticola and its competitive ability to decompose sapwood and heartwood wood, as compared to the activity of Phlebiopsis gigantea and Heterobasidion parviporum. Over the last several decades, P. gigantea has routinely been used for biocontrol of the conifer pathogen Heterobasidion annosum s.l.; however, its protective effect on Norway spruce stands was recently demonstrated to be not satisfactory. P. abieticola was proposed instead, as a promising species that might successfully compete with H. parviporum. We investigated the growth of mycelium and the ability of P. abieticola isolates to decompose wood of Norway spruce, in the experiment with isolates of P. gigantea and H. parviporum. Heartwood was better decomposed than sapwood by the majority isolates used in the experiment. Linear growth of the investigated fungi showed a more rapid mycelium development for P. gigantea and H. parviporum, compared to that of P. abieticola. In dual cultures, H. parviporum was overgrown only by P. gigantea. All the tested isolates of P. abieticola showed weaker wood decomposition than those of P. gigantea and H. parviporum. Further study is required to better understand the role of P. abieticola for the protection of spruce stands.  相似文献

The fungus Phlebiopsis gigantea has been used in Europe as a biological agent for the control of conifer root and butt (caused by Heterobasidion annosum ) for nearly 40 years. P. gigantea competes with H. annosum for the woody resource within conifer stumps, and is applied to stump surfaces at felling. Three distinct biological control products based on P. gigantea have been developed: PG Suspension in the UK, PG IBL in Poland and Rotstop in Finland. The formulations are of oidia, which are maintained in a sucrose suspension, sawdust, or a wettable powder, respectively. PG Suspension and PG IBL are applied to pine stumps, while Rotstop is equally as effective on pine as on Norway spruce stumps. For each product, isolates of P. gigantea are selected from the wild and are screened for their competitive ability against H. annosum before formulation. Viability and purity checks are undertaken throughout the production cycle and during routine use. The increasing use of mechanized harvesting machines to fell and process trees is having an impact on this biological control system, the formulations having to be compatible with the mechanical application systems and vice versa. This paper compares the formulation, testing and application of the three products, and considers some aspects of their future development.  相似文献

The biocontrol agent Phlebiopsis gigantea has been intensively applied to the surface of Picea abies stumps to control Heterobasidion root rot. But little is known about the possible impact of this treatment on the resident bacteria community in the stumps. High throughput DNA bar-coded pyrosequencing was used to characterize the diversity of bacteria in the stumps of P. abies at 1, 6 and 13 years after treatment with P. gigantea. The sequences were classified into 12 phyla and 160 genera, of which Proteobacteria and Acidobacteria were the most abundant groups over time. Moreover, at the initial stages of decay, Proteobacteria were the most abundant whereas Acidobacteria were the most common at advanced stages of decay. Treatment with P. gigantea led to significant increase of the genus Acidobacteria-Gp1 at 1 year after treatment. The analysis of observed and estimated operational taxonomic units (OTUs) as well as diversity indices revealed that P. gigantea treatment significantly decreased the initial bacterial richness in the stumps, but the bacterial community gradually recovered and the negative effects of P. gigantea was attenuated. These results provide additional insight on the risk assessment as well as environmental impact on the long-term use of P. gigantea in the control of Heterobasidion root rot in conifer forests.  相似文献

The basidiomycete Phlebiop sis gigantea (Fr.) Jülich has been used in Swedish forestry as a biocontrol agent against the root and butt-rot pathogen Heterobasidion annosum s.l. on freshly cut Picea abies stumps since the early 1990s. Until 2005, the commercial preparation of this biological stump treatment, Rotstop®, has been based on a single strain of P. gigantea that has been applied on more than 47,000 ha annually in Fennoscandia. This paper reports on the spread of genetic material from the Rotstop® biocontrol strain of P. gigantea to resident populations of P. gigantea. We conclude that the inoculated fungus remained to a large extent restricted to the treated plots and did not spread to the adjacent areas, dominated by the natural spore deposition from resident populations of the fungus. Furthermore, the study demonstrates high genetic diversity and low geographic differentiation in P. gigantea populations in the geographical area around the Baltic Sea.  相似文献

The registration of one of the world's first biological control agents for a plant pathogen, in January 1998, after 30 years of field use, is discussed in relation to its approval under the UK Control of Pesticides Regulations 1986. The control agent, Phlebiopsis gigantea (formerly known as Peniophora gigantea), is a common wood-rotting basidiomycete which is applied to the freshly cut surfaces of pine stumps to prevent their colonization by the pathogenic rootrotting fungus Heterobasidion annosum . Unlike many other biocontrol agents, P. gigantea is not a biocide that kills the target organism. Rather, it competes for resources that the pathogen would otherwise use, providing an extension to a naturally occurring process. Experience in securing approval is reviewed, with special reference to the identity of the organism and its natural distribution, and to assessments of the risk its use might pose in the environment. It is suggested that before the formal process of applying for approval of a biocontrol agent begins, there should be procedures for agreeing precise data requirements. These should be based on the type of crop to be treated and the mode of operation and characteristics of the biocontrol organism. This would streamline the registration process.  相似文献

Soil streptomycetes are commonly antagonistic against plant pathogens. However, interactions involving increased defense responses in the host plant, leading to suppression of plant disease development, have not yet been detailed. Here, the mechanisms were studied of disease suppression by Streptomyces sp. GB 4-2 against Heterobasidion root and butt rot in Norway spruce (Picea abies) seedlings. GB 4-2 promoted mycelial growth of the phytopathogenic fungus, germination rate of fungal spores, extension of germ tubes and early colonization of outer cortical layers of the plant root. Reduced colonization of the inner cortical cell layers was accompanied by the induction of cell wall appositions, and increased xylem formation in the vascular cylinder emerged after bacterium-fungus coinoculation. Bacterial treatment led to decreased water content in roots and needles and increased photosynthetic yield (F(v)/F(m)) and peroxidase activities in needles. The infection of needles by Botrytis cinerea was reduced by bacterial pretreatment. Complex interactions of GB 4-2 with Norway spruce and Heterobasidion abietinum were discovered. The bacterium promoted the growth of the phytopathogenic fungus but induced plant defense responses. Host responses indicate that GB 4-2 induces both local and systemic defense responses in Norway spruce.  相似文献

To explore effective and ecofriendly means of controlling wheat flag smut (WFS) using biocontrol agents, three endophytic strains (58-2-1, 37-1 and YC-1) of Bacillus sp. were isolated from winter wheat plants in China and identified as Bacillus thuringiensis based on their 16S rDNA sequences as well as phenotypic characteristics. Four morphological (leaf length, root length, dry weight and tiller numbers) and one physiological [root vigour (RV)] parameters of wheat plants treated by strains 58-2-1 and 37-1 were significantly enhanced compared to the control. The soluble sugar contents in the roots of wheat samples treated by the two strains were significantly lower than the control. The resistance of wheat varieties to WFS was investigated by inoculation tests. Of the 12 wheat varieties tested, 6 (Yunhan-618, Bainongaikang-58, Kaimai-20, Zhengmai-9023, 04-zhong-36 and Yanzhan-4110) were identified as WFS-highly resistant (HR), 3 (Pumai-9, Jinboshi-1 and Yunong-202) WFS-moderately resistant (MR), 1 (Yubao-1) WFS-susceptible (S) and 2 (Yumai-012 and Yunong-416) WFS-highly susceptible (HS) varieties. The Urocystis tritici-induced yield loss on the S/HS varieties was significantly higher than that on the HR/MR ones. The strains 58-2-1 and 37-1 had control efficacies of 6.7–100% (av.54.8%) and 33.3–100% (av. 66.5%) on 9 and 7 out of 12 varieties, respectively. The strains 58-2-1 and 37-1 had enhanced yields of 10.2–54.9% (av. 32.9%) and 2.8–43.4% (av. 24.8%) on 10 and 8 out of 12 varieties, respectively. This is the first report on endophytic B. thuringiensis strains isolated from wheat plants with the abilities to suppress WFS and to enhance yields on multiple wheat varieties.  相似文献

Previous work has shown thatAbax parallelepipedus Piller &; Mitterpacher preys upon slugs in the field, and can be effectively employed to control slugs in polythene tunnels. To be a viable biological control agent this carabid must also be amenable to mass culturing. Manipulation of the substrate ensured that eggs were laid in soil capsules, but deposited in peat, facilitating collection and monitoring. Up to 570 eggs per beetle were laid at 20°C. No cannibalism was exhibited at any stage by this species. Larvae were reared intensively in batches and fed ad libitum on live earthworms, eliminating the need for regular feeding and the removal of decaying food. Rearing experiments were undertaken under a number of different temperature regimes. At constant 20°C it was found that growth was arrested in some 2nd instar larvae for long periods. Co-ordinated changes between 2nd and 3rd instar stages were induced by reduction of the temperature to 14°C between days 24–31 from hatching. The complete life cycle, from newly laid egg to adult beetle, could be reduced to less than 110 days. Mortality was greatest at the late 3rd instar and pre-pupal stages, and possible reasons for this are discussed.  相似文献

Wood-based composites such as oriented strand board (OSB) are principle framing elements in building construction in North America. However, these materials are often affected by moulds in wet or humid environmental conditions. A common control method for prevention of mould growth on panels is preservative treatment of panels with various pesticides or chemicals. In recent years, environmentally friendly pest control methods are required because of environmental issue. This research aimed to develop a biological technology to protect OSB against mould infection by post-treatment of panels with natural extracts from fungal antagonists. In this study, the culture metabolites of a fungal antagonist, Phaeotheca dimorphospora DesRochers & Ouellette, were extracted, and the antibiotic activity of the extracts was tested in Petri plates against various moulds and decay fungi. The OSB panels were then dip-treated with the extracts and exposed to a humid environment for mould growth testing in a period of 8 weeks. The results showed that the mycelia growth of all fungi tested (moulds, white-rot and brown-rot fungi) was inhibited by the extracts of P. dimorphospora on agar plates. Panel samples dipped with the fungal extracts in acetone got little mould growth on them, whereas untreated control panels and those samples treated with acetone alone were seriously affected by various moulds.  相似文献

By use of plane and solid geometry and probability models, efficiencies of infection and competition for nutrients and infection sites by a nonpathogenic strain of Fusarium oxysporum (C14) with F. oxysporum f. sp. cucumerinum on the rhizoplane of cucumber were calculated. The model is derived from previously published data. Efficiencies for successful infection were 0.04 chlamydospores per infection site for both pathogen and nonpathogen. Observed successful infections by the pathogen in competition with the nonpathogen were close in values to the competition ratio (CR) calculated as the number of chlamydospores on the infection court of the pathogen divided by the total number of both pathogen and nonpathogen at relatively low densities. When total chlamydospores were, on average, closer than 175 μm apart, however, competition for nutrients/mutual inhibition occurred. At such densities there was an overestimation of the effect of competition for infection sites. These relationships were modeled at inoculum densities of pathogen and/or nonpathogen of 5000 chlamydospores per g soil and above, however, in the field, maximum densities of 1000 colony forming units/g (cfu) were observed. Most likely models of competition for infection sites at this density of the pathogen revealed that infection efficiency was only approximately halved, even when 0.98 of the possible 30 infection sites were occupied by the nonpathogen. It is conclude that competition for nutrients and/or infection sites is an insignificant factor in biocontrol of Fusarium wilt diseases by nonpathogenic fusaria.  相似文献

AIMS: To evaluate the biocontrol efficacy of culture filtrate containing chitinase from Trichoderma harzianum against Heliothis. METHODS AND RESULTS: T. harzianum was cultured by submerged fermentation using colloidal chitin as sole carbon source. The ability of the culture filtrate to hydrolyse colloidal chitin indicated the presence of chitinase as one of its components. Biocontrol assay on Heliothis showed that the culture filtrate is a potent antifeedant as it reduced the feeding rate and body weight of the larvae. It reduced the successful pupation and increased larval and pupal mortality in a dosage-dependent manner when applied topically. The highest mortalities (70%) were recorded for groups treated with 2000 U ml(-1) chitinase activity. The percentage of adult emergence was zero for the highest chitinase concentration (2000 U ml(-1)) tried. CONCLUSIONS: The studies showed that the culture filtrate containing chitinase from T. harzianum is capable of negatively affecting the growth and metamorphosis of Heliothis larvae. SIGNIFICANCE AND IMPACT OF THE STUDY: In view of the need for safer and environmentally friendly pest management tools, the present study could help in the development of enzyme-based biopesticides against Heliothis.  相似文献

We propose a comprehensive program to evaluate the post-release phase of biocontrol programs that use insect herbivores to control invasive plant species. We argue that any release should be done in randomized release and non-release sites and should be followed up by well-replicated sampling and experimental protocols that evaluate the degree of success or failure. These follow-up studies should include landscape scale monitoring across relevant habitat gradients of (1) the abundance of the biocontrol agent, (2) the impact of the biocontrol agent on the target plant species, (3) the potential for non-target effects, and (4) the response of native species and communities to a reduction in the invasive species. We also argue that (5) experimental reductions of the biocontrol agent are required to eliminate the chance that the putative impact of the biocontrol agent is not confounded with other causes. Finally, we describe six scenarios, informed largely by a community ecology perspective, in which a biocontrol agent may decrease the abundance or vigor of the target plant species but not lead to successful control where native communities re-establish. We classify these failure scenarios as either direct or indirect effects of the invasive plant species: Native Source Limitation, Static Competitive Hierarchies, Novel Weapons, Trophic Shifts, Invasive Engineering and Associated Invasives. Overall, we argue that well replicated and landscape-scale post release monitoring programs are required not only to evaluate critically the degree of success and failure of biocontrol programs worldwide but also to provide insights into improving future biocontrol efforts. The fungal diversity structures of soils that are suppressive and non-suppressive to Sclerotinia sclerotiorum were characterised and screened for fungal strains antagonistic to the S. sclerotiorum pathogen. Soil suppressiveness was associated with a particular fungal diversity structure. Principal component analysis showed that antagonism by fungal species in suppressive soils was associated with the occurrence of Fusarium oxysporum, Fusarium solani, Talaromyces flavus var. flavus and Clonostachys rosea f. rosea. In particular, C. rosea f. rosea occurred exclusively in suppressive soil samples, suggesting that this morpho-species plays an important role in suppression of S. sclerotiorum diseases. One strain of C. rosea f. rosea (BAFC1646) was selected for further experiments. Dual-culture assays confirmed the antagonistic behaviour of C. rosea f. rosea BAFC1646 against three different S. sclerotiorum strains. Antifungal activity was corroborated by diffusion assays with metabolite extracts. Greenhouse assays with soybean plants showed that the selected C. rosea f. rosea strain reduced the percentage of dead plants when co-inoculated with S. sclerotiorum. In addition, inclusion of C. rosea f. rosea alone increased shoot lengths significantly. In this work, we established the involvement of fungal species in soil suppressiveness and in further assays confirmed that C. rosea f. rosea BAFC1646 exhibits a bioprotective effect against S. sclerotiorum in soybean plants.  相似文献

Aims: Assessment of biological control of Cercospora sojina, causal agent of frogeye leaf spot (FLS) of soya bean, using three indigenous bacterial strains, BNM297 (Pseudomonas fluorescens), BNM340 and BNM122 (Bacillus amyloliquefaciens). Methods and Results: From cultures of each bacterial strain, cell suspensions and cell‐free supernatants were obtained and assayed to determine their antifungal activity against C. sojina. Both mycelial growth and spore germination in vitro were more strongly inhibited by bacterial cell suspensions than by cell‐free supernatants. The Bacillus strains BNM122 and BNM340 inhibited the fungal growth to a similar degree (I~52–53%), while cells from P. fluorescens BNM297 caused a lesser reduction (I~32–34%) in the fungus colony diameter. The foliar application of the two Bacillus strains on soya bean seedlings, under greenhouse conditions, significantly reduced the disease severity with respect to control soya bean seedlings and those sprayed with BNM297. This last bacterial strain was not effective in controlling FLS in vivo. Conclusions: Our data demonstrate that the application of antagonistic bacteria may be a promising and environmentally friendly alternative to control the FLS of soya bean. Significance and Impact of the Study: To our knowledge, this is the first report of biological control of C. sojina by using native Bacillus strains.  相似文献

The efficacy of Pseudomonas fluorescens EPS62e in the biocontrol of Erwinia amylovora, the causal agent of fire blight of apple and pear, depends on the colonization of plant surfaces after spray application. A procedure to increase cell survival in the phyllosphere was developed consisting of saline stress and osmolyte amendment to the growth medium during inoculum preparation. Hyperosmotic stress induced the synthesis of the osmolytes trehalose, N-acetylglutaminylglutamine amide and glucosyl-glycerol, but decreasing growth rate. Amendment of the growth medium with glycine betaine increased growth rate and cell yield and promoted its intracellular accumulation. Under controlled environment conditions, osmoadaptation increased by 10- to 100-fold cell survival to desiccation and to low relative humidity conditions on plant surfaces, in comparison with the nonosmoadapted controls. In the field, cell survival increased 100-1000 times in immature fruit upon osmoadaptation but was not significantly affected in flowers where active colonization occurs. The efficacy in the control of fire blight infections was increased 30-50% upon osmoadaptation on immature fruits but was not affected in blossoms. The method of osmoadaptation may be useful for improving the fitness and efficacy of biological control agents of phyllosphere pathogens under limiting humidity conditions.  相似文献
