Summary The bristle pattern along the anterior margin ofNotch (N1-22.3) wings ofDrosophila hydei and the occurrence ofyellow (y 1–38.8) marked clones induced by X-ray irradiation during various larval stages are described. UnirradiatedN/N + wings show gaps (notches) in the longitudinal bristle rows along the 1st longitudinal vein, with tufts of bristles particularly near gaps. X-ray irradiation increases the number and total length of the gaps. The patterning of bristles along the margin depends on theN (+) genotype of the induced clones. RecombinantN +/N + clones from irradiated wings show excessive growth with an autonomous wildtype bristle pattern. Characteristically, these clones do not respect the dorso-ventral compartment boundary along the wing margin, do not follow an exponential (2n) growth pattern, tend to fill the gaps with bristles and theiryellow medial row bristles are less often interspersed withy + bristles than described forN +/N + wings. HomozygousN appears to be a cell lethal condition inD. hydei as it is inD. melanogaster. When y clones were kept phenotypicallyNotch (viz.,N/N/N +) as the background cells, we found a lower number ofy bristles, a lower percentage of mosaic wings but also a reltive deficiency ofy + interspersions. The influence of electromagnetic field exposure on puffing pattern of salivary gland polythene chromosomes, viability and fertility of Drosophila melanogaster of the wild type Canton-S line was studied. Experimental conditions: Electromagnetic field characteristics: frequency — 36.64 GHz, power density — 0.4 W/m2, exposure time −10 seconds. Electromagnetic field exposure was conducted on the egg stage. Results: in larvae developed from the exposed eggs 3 of 8 chromosomal puffs tested (71CE, 82EF, and 83E) had significantly smaller dimensions than these in control at the prepupal stage. Viability of Drosophila estimated by the number of adult flies hatched from exposed eggs decreased, while the number of dominant lethal mutations increased. Conclusion: the exposure to a low-level microwave irradiation suppressed puffing activity at ecdysone-inducible loci of Drosophila polythene chromosomes, increased frequency of dominant lethal mutations and decreased Drosophila viability but did not influence Drosophila fertility.  相似文献

Anopheles atroparvus has two pairs of autosomes similar in length and morphology and two sex chromosomes with equal, heterochromatic, late replicating long arms with homologous C-, G-, and Q-bands. The short arm of the Y is shorter than that of the X and both are euchromatic. The mean number of chiasmata per cell in the male is 3.2. During mitosis there is a high grade of somatic pairing but X and Y, which form a heteropycnotic mass in the interphase nucleus, have a differential behaviour. The chronology of DNA replication was studied in spermatogonia and brain cells by autoradiography. It is hypothesized that the present sex chromosomes of A. atroparvus evolved by accumulation of sex determining factors and gene deterioration resulting in heterochromatinization of the long arms, followed by structural rearrangements.—The homology of the two sex chromosomes requires limited dosage compensation which is achieved either as in Drosophila by modifier genes or by accumulation on the short arm of the X, only of female determining factors which do not require dosage compensation.  相似文献

In situ hybridization of Drosophila melanogaster somatic chromosomes has been used to demonstrate the near exact correspondence between the location of highly repetitious DNA and classically defined constitutive heterochromatin. The Y chromosome, in particular, is heavily labeled even by cRNA transcribed from female (XX) DNA templates (i.e., DNA from female Drosophila with 2 Xs and 2 sets of autosomes). This observation confirms earlier reports that the Y chromosome contains repeated DNA sequences that are shared by other chromosomes. In grain counting experiments the Y chromosome shows significantly heavier label than any other chromosome when hybridized with cRNA from XY DNA templates (i.e., DNA from male Drosophila with 1 X and 1 Y plus 2 sets of autosomes). However, the preferential labeling of the Y is abolished if the cRNA is derived from XX DNA. We interpret these results as indicating the presence of a class of Y chromosome specific repeated DNA in D. melanogaster. The relative inefficiency of the X chromosome in binding cRNA from XY and XYY DNA templates, coupled with its ability to bind XX derived cRNA, may also indicate the presence of an X chromosome specific repeated DNA.  相似文献

The nucleolus organizer locus of Xenopus laevis lampbrush chromosomes was identified by in situ hybridization of a 3H-labelled probe complementary to 18S + 28S rDNA. The nucleolus organizer is an axial granule on chromosome III that lies four-fifths the way down this chromosome reading from its larger (left) telomere, just within an exploded region that extends to its right end, where the lateral loops are exceptionally long. By in situ hybridization of 3H-labelled oocyte and somatic 5S spacer cRNA probes to similarly RNase-treated and denatured lampbrush chromosomes, the multiple sites of oocyte and somatic 5S gene families were identified. Oocyte 5S genes lie at the larger telomeres of the 15 chromosomes that possess these structures; that is, all but chromosomes X, XVII and XVIII. There are a further four sites, all peripheral, and in three of these, on chromosomes VII, X and XI, the sequences lie on lateral loops that are resolvable with the light microscope. By contrast all of the somatic 5S gene clusters occupy peripheral sites. There are two sites on chromosome III, one of which may be shared with oocyte 5S sequences; one on chromosome VII, which is very likely shared with oocyte 5S sequences; one terminal site on chromosome X; one site on chromosome XI that lies on a single pair of long loops which are inserted in a conspicuous and recognizable axial granule, loops which certainly carry oocyte 5S sequences too; two nearly terminal sites alongside the larger telomeres on chromosomes XII and XIV; and single interstitial sites on all three of the sphere-bearing chromosomes, VIII, IX and XVI. We suggest that 5S sequences on resolvable loops are transcribed by readthrough from upstream promoters, probably by polymerase II.  相似文献

Microtus agrestis is characterised by long sex chromosomes, most of which are constitutively heterochromatic, and thus supposedly, genetically inactive. A method to assess the template activity of the chromosomes is to study the distribution of chromatid aberrations produced by H3UdR, among and within the chromosomes. In such a study, in female Microtus agrestis cells in culture, it was found that, a large number of localised chromatid aberrations was induced in the constitutively heterochromatic regions of both X chromosomes. The frequency distribution and types of aberrations were found to be cell cycle dependent. With differential staining it has been possible to demonstrate that the constitutive heterochromatin of the sex chromosomes are involved in the nucleolar organisation in this species, thus containing the ribosomal RNA cistrons.  相似文献

The Y chromosomes of most Drosophila species are necessary for male fertility but they are not involved in sex determination. They have many puzzling properties that resemble the effects caused by B chromosomes. Classical genetic and molecular studies reveal substantial affinities between Y and B chromosomes and suggest that the Y chromosomes of Drosophila are not degenerated homologues of the X chromosomes, but rather that their Y chromosomes evolved as specialized supernumeraries similar to classical B chromosomes.  相似文献

Mesosternal (MS) bristles in Drosophila are a pair of machrochaetae found at the sternal end of the sternopleural (STP) microchaetae, and are thought to be invariable. In a closely related drosophilid genus, Zaprionus, their number is four and, in contrast to Drosophila, they show interspecific and intraspecific variability. The genetic basis of MS bristle number variability was studied in Z. indianus, the only cosmopolitan species of the genus. The trait responded rapidly to selection and two lines were obtained, one lacking any bristles (0-0) and the other bearing the normal phenotype (2-2). Other symmetrical phenotypes, (1-1) and (3-3), could also be selected for, but with lesser success. By contrast, STP bristle number did not vary significantly between the two lines (0-0) and (2-2), revealing its genetic independence from MS bristle number. Reciprocal crosses between these two lines showed that MS bristle number is mainly influenced by a major gene on the X chromosome (i.e. F1 males always resembled their mothers) with codominant expression (i.e. heterozygous F1 females harboured an average phenotype of 2 bristles). However, trait penetrance was incomplete and backcrosses revealed that this variability was partly due to genetic modifiers, most likely autosomal. The canalization of MS bristle number was investigated under different temperatures, and the increased appearance of abnormal phenotypes mainly occurred at extreme temperatures. There was a bias, however, towards bristle loss, as shown by a liability (developmental map) analysis. Finally, when ancestral and introduced populations were compared, the latter were far less stable, suggesting that genetic bottlenecks may perturb the MS bristle number canalization system. MS bristle number, thus, appears to be an excellent model for investigating developmental canalization at both the quantitative and the molecular level.  相似文献

The effects of the thymidine analog, 5-bromodeoxyuridine (BUdR), on the formation of red cells in the yolk sac of the chick embryo were examined. The prospective area opaca vasculosa from a definitive primitive streak embryo was excised, disaggregated, and deposited into a cell clump, and the cell clump was placed in organ culture. Hemoglobin synthesis is detectable after about 16 hr in culture. The formation of erythropoietic foci and incorporation of 55Fe into heme were used to measure the extent of erythropoiesis. Exposure to 40 µg/ml of BUdR within 6 hr after explantation almost completely eliminated red cell formation; subsequent transfer to thymidine medium showed that the inhibition was reversible, and there was no histological evidence of analog toxicity. Between 6 and 12 hr after initiation of organ culture, the tissue became completely refractory to BUdR. DNA synthesis, as monitored by thymidine-3H and BUdR-3H pulses, was extensive both during and after the period of BUdR sensitivity. Hence, during both BUdR sensitive and insensitive periods the analog was incorporated into DNA of cells which had not yet synthesized hemoglobin. It is proposed that between 6 and 12 hr a crucial regulatory event for terminal differentiation is perturbed by the presence of BUdR in the chromosomes.  相似文献

Like several other species of Drosophila, D. quinaria is polymorphic for X-chromosome meiotic drive; matings involving males that carry a “sex-ratio” X chromosome (XSR) result in the production of strongly female-biased offspring sex ratios (Jaenike 1996). A survey of isofemale lines of D. quinaria from several populations reveals that there is genetic variation for partial suppression of this meiotic drive. Crossing experiments show that there is Y-linked, and probably autosomal, variation for suppression of drive. Y-linked suppressors of X-chromosome drive have now been described in several species of Diptera. I develop a simple model for the maintenance of Y-chromosome polymorphism in species polymorphic for X-linked meiotic drive. One interesting feature of this model is that, if there is a stable Y-chromosome polymorphism, then the equilibrium frequency of the standard and sex-ratio X chromosomes is determined solely by Y-chromosome parameters, not by the fitness effects of the different X chromosomes on their carriers. This model suggests that Y-chromosome polymorphism may be easier to maintain than previously thought, and I hypothesize that karyotypic variation in Y chromosomes will be found to be associated with suppression of sex-ratio meiotic drive in other species of Drosophila.  相似文献

Mutations controlling the resistance to 6-mercaptopurine (6-M) and the ability to multiply in a medium with a low concentration of glucose (“glucose-independent” mutants) were induced in cultured Chinese hamster cells by N-nitrosomethylurea (NMU), 5-bromodeoxyuridine (BUdR), UV and X-rays. The chemical agents were found to be very active in induction of mutations to 6-M resistance (NMU and BUdR) and mutations of “glucose independence” (NMU). These agents increase the yield of mutations as compared to the spontaneous mutation rate by about two orders of magnitude. The induced rate of 6-M-resistant mutations by X-rays was 2.0 ? 10−7 per viable cell per roentgen. BUdR approximately equally increases the cell's sensitivity to both inactivating and mutagenic action of X-rays. The maximum induction of mutations to 6-M resistance by UV was observed at 100 erg/mm2. This dose leads to 1 16-fold increase of the mutation frequency as compared to the spontaneous rate. Further increase of the UV dose up to 200 erg/mm2 resulted in a lower yield of mutations per dose unit. The highest yield of mutations to 6-M resistance induced by NMU, BUdR and X-rays was observed if cells were plated in selective medium several generations after the mutagenic treatment. The maximum yield of mutations to 6-M resistance induced by UV and of glucose-independence induced by NMU was recorded if cells were transferred to selective media immediately after treatment. The kinetics of expression of mutations and the decline of their number observed after prolonged incubation of treated cells in nonselective conditions are discussed.  相似文献

Polyethylene glycol was used to induce interspecific somatic cell fusion between human fibroblasts (stock F6) and Drosophila melanogaster cells from established cell lines (C1 82 and 11 P102), characterized by different ploidy levels. The present investigation defines some parameters for Drosophila cell fusion and interspecific fusion between Drosophila and human cells. The cytological analysis provided evidence of spontaneous as well as induced human-Drosophila heterokaryon formation. The presence in the same cell of two types of nuclei, distinguishable because of their different size and morphology, was confirmed autoradiographically by 3H-thymidine pre-labelling of Drosophila cells. Furthermore, the retained DNA synthetic activity and some examples of mitotic figures of both types of nuclei in the heterokaryons indicate the viability of the fused cells.  相似文献

Summary The major concept of fragile X pathogenesis postulates that the fragile site at band Xq27.3 [fra(X)] represents the primary defect. The expression of fra(X) is predicted to be an intrinsic property of the mutated chromosome and, hence, should not be suppressed by X inactivation in females or induced by X-linked trans-acting factors. We made fibroblast clones of a fra(X)-positive female. Monoclonality was demonstrated using the DNA methylation assay at DXS255. The mutated X chromosomes and their states of genetic activity in the different clones were also defined by molecular methods. Five clones were selected to induce expression of fra(X) by 10-7 M FUdR; two carried an active mutated X chromosome, in the other three the mutated X chromosome was inactivated. Fra(X) was found expressed in both types of clones. The percentages of positive cells were as high as 7–10%, regardless of the genetic activity of the mutated X chromosomes. DNA replicating patterns, obtained by BUdR labelling, demonstrated that expression occurred only on the mutated X chromosomes previously identified by molecular methods. The concept that the fragile site represents the primary mutation is now strongly supported by experimental evidence. The expression of fra (X) in females is independent of X inactivation and other trans-acting factors.  相似文献

Increasing evidence supports that activation of store‐operated Ca2+ entry (SOCE) is implicated in the chemoresistance of cancer cells subjected to chemotherapy. However, the molecular mechanisms underlying chemoresistance are not well understood. In this study, we aim to investigate whether 5‐FU induces hepatocarcinoma cell death through regulating Ca2+‐dependent autophagy. [Ca2+]i was measured using fura2/AM dye. Protein expression was determined by Western blotting and immunohistochemistry. We found that 5‐fluorouracil (5‐FU) induced autophagic cell death in HepG2 hepatocarcinoma cells by inhibiting PI3K/AKT/mTOR pathway. Orai1 expression was obviously elevated in hepatocarcinoma tissues. 5‐FU treatment decreased SOCE and Orai1 expressions, but had no effects on Stim1 and TRPC1 expressions. Knockdown of Orai1 or pharmacological inhibition of SOCE enhanced 5‐FU‐induced inhibition of PI3K/AKT/mTOR pathway and potentiated 5‐FU‐activated autophagic cell death. On the contrary, ectopic overexpression of Orai1 antagonizes 5‐FU‐induced autophagy and cell death. Our findings provide convincing evidence to show that Orai1 expression is increased in hepatocarcinoma tissues. 5‐FU can induce autophagic cell death in HepG2 hepatocarcinoma cells through inhibition of SOCE via decreasing Orai1 expression. These findings suggest that Orai1 expression is a predictor of 5‐FU sensitivity for hepatocarcinoma treatment and blockade of Orai1‐mediated Ca2+ entry may be a promising strategy to sensitize hepatocarcinoma cells to 5‐FU treatment.  相似文献

Somatic embryogenesis induced in suspension cultures of carrot (Daucus carota) was inhibited by BUdR at concentrations which do not influence the rate of RNA or protein synthesis, or the growth of the cells. At the time of embryo induction 2 days treatment with 6 μM BUdR was sufficient for the complete supression of differentiation. The degree of inhibition showed a dependence on the applied concentration of BUdR, as well as on the level of BUdR substitution in DNA. The BUdR inhibition proved to be reversible and the embryogenic capacity of the carrot cells was restored after removel of the analogue. Addition of thymidine prevented the inhibitory effect of BUdR by decreasing the incorporation of BUdR into DNA. The experiments reported here indicate that the effect of BUdR on the somatic embryo differentiation was probably associated with BUdR substitution of thymidine in the DNA.  相似文献

Disc polyacrylamide gel electrophoresis (disc PAGE) analyses of chick-mouse somatic cell hybrids [LM(TK)/CRB]isolated from fusion mixtures of chick erythrocytes and thymidine (TdR) kinase-deficient mouse [LM(TK)]cells have demonstrated that the somatic cell hybrids contain only chick cytosol TdR kinase F and mouse mitochondrial TdR kinase A activities. Karyotypes were analysed by the method which sequentially reveals Q- and C-bands. Four hybrid clones contained the full complement of mouse chromosomes and 1 to 3 chick micro-chromosomes. Counterselection of the LM(TK)/CRB hybrids in 5-bromodeoxyuridine (BUdR) medium resulted in the loss of chick cytosol TdR kinase F activity and at least one of the chick chromosomes, but mouse mitochondrial TdR kinase A activity was unaffected. Unlike the LM(TK)/CRB somatic cell hybrids, the BUdR-resistant clones could not grow in HATG (hypoxanthine-aminopte-rin-thymidine-glycine) medium. The results demonstrate that: (1) the chick cytosol TdR kinase F gene is on a member of the micro-chromosomes; and (2) selection in HATG- and BUdR-containing medium involves only cytosol TdR kinase F.  相似文献

Summary A new approach was made to comparing the contributions to response of chromosomes 2 and 3 of Drosophila in lines selected for high and low sternopleural bristle number.Separate response curves for chromosomes 2 and 3 were obtained from changes in the effect of standard second and third chromosomes marked Cy and which were kept segregating with their wild-type homologues during selection.Dominance interaction between marker and wild chromosomes caused some bias in estimating responses in each direction, but the amount by which the responses diverged in the two directions of selection was relatively free from bias. On a log. scale divergences of chromosomes 2 and 3 were in the ratio 11.7 and their heritabilities realised early in selection were 0.14 and 0.26 respectively.There was interaction between the Cy and the chromosomes which was not altered by the selection. Almost all the responses in the lines could be accounted for by addition of the responses in single chromosomes 2 and 3, chromosomes 1 and 4 making a negligible contribution.Sampling, as a cause of variation between selection lines, was reflected in the variation between them in response in chromosomes 2 and 3.  相似文献

Somatic mutation and recombination test on wing cells of Drosophila melanogaster showed that the recombination frequency in the somatic tissues of strains studied correlated with the presence of a full-length copy of the hobo transposable element in the genome. Transposition of hobo in somatic tissue cells at a frequency 3.5 × 10?2 per site per X chromosome was shown by fluorescence in situ hybridization with salivary gland polytene chromosomes of larvae of one of the D. melanogaster strains having a full-length hobo copy.  相似文献

In the wood lemming (Myopus schisticolor) three genetic types of sex chromosome constitution in females are postulated: XX, X*X and X*Y (X*=X with a mutation inactivating the male determining effect of the Y chromosome). Males are all XY. It is shown in the present paper that the two types of X chromosomes, X and X*, exhibit differences in the G-band patterns of their short arms. In addition, it was demonstrated in unbanded chromosomes that the short arm in X* is shorter than in X. The origin of these differences is still obscure; but they allow to identify and to distinguish the individual types of sex chromosome constitution, as of XX versus X*X females and of X*Y females versus XY males, on the basis of G-banded chromosome preparations from somatic cells.  相似文献
