从木聚糖酶高产短小芽孢杆菌 (Bacilluspumilus)BP5 1中克隆得到木聚糖酶基因xynA ,将其构建在芽孢杆菌表达载体pWH1 5 2 0中得到重组质粒pWSX1 1。xynA由木糖诱导xylA启动子调控xynA表达。采用同源高效表达策略 ,以原生质体转化方法将pWSX1 1转回原始菌株BP5 1中 ,获得重组菌株BPX1 1。通过木糖诱导重组菌株中的xy nA基因高效分泌表达 ,使木聚糖酶产酶活力比原菌株BP5 1提高了 87% ,同时对重组表达的木聚糖酶的酶学性质进行了初步研究  相似文献

以短小芽孢杆菌HZbp总DNA为模板以PCR的方式获得512 bp的脂肪酶基因,并在该基因的两端引入了EcoR1和Sal1的酶切位点,将该基因与大肠杆菌表达质粒pSE380连接,获得重组质粒pSE380-BPL。重组质粒转入大肠杆菌表达细胞株BL21,获得工程菌株BL21-BPL。序列分析显示所克隆的基因具有脂肪酶的保守G-X-S-X-G序列,SDS-PAGE电泳显示该脂脂肪酶的分子质量约为20 kDa。在LB培养基中,IPTG诱导浓度为1.0 mmol/L,33℃诱导培养10 h后,发酵液酶活达到8 U/mL。  相似文献

degQ基因编码一个由46个氨基酸组成的多肽,能增强许多芽孢杆菌胞外酶基因的表达.以pMK4作克隆载体构建短小芽孢杆菌基因文库,并用DNA探针原位杂交法从中钓出degQ基因.对克隆基因的DNA序列进行了分析并证明克隆的短小芽孢杆菌degQ基因具有增强枯草杆菌蛋白酶和果聚糖蔗糖酶基因表达的能力.degQ基因克隆有助于研究芽孢杆菌的正调控机理并可望提高外源基因在芽孢杆菌中表达.  相似文献

碱性木聚糖酶在碱性条件下催化水解木聚糖,广泛应用于造纸、纺织等领域.着重对短小芽胞杆菌M-11产碱性木聚糖酶的发酵条件进行初步的探索.研究了菌株的生长曲线、确定最佳接种龄为16 h、最佳接种量为1%;确定最适碳源浓度为7%、最适单一氮源为氯化铵、其浓度为1.0%、最适无机盐为氯化铁、其浓度为3 mmol/L;在此基础之上进行6因素3水平的正交试验,确定最适产酶培养基组成:麸皮5%,接种量3%,氯化铵1.2%,氯化铁3.5 mmol/L,硫酸镁0.03%,氯化钠5 mmol/L,磷酸氢二钾0.4%;最适培养条件:接种龄16 h,初始pH 8.0,温度37℃,300 mL摇瓶装液量50 mL,摇床转速220 r/min,发酵周期48 h.通过对发酵条件的优化使发酵液酶活达613 IU/mL.无机氮源为其最适氮源,因此短小芽胞杆菌M-11在碱性木聚糖酶的产品开发上优于短小芽胞杆菌M -26.  相似文献

利用易错PCR技术对短小芽胞杆菌(Bacillus pumilus)YZ02脂肪酶基因BpL进行两轮定向进化研究, 分别获得最佳突变株BpL1-7和BpL2-1369, 其脂肪酶活力比出发酶分别提高了2倍和6倍。序列分析表明, 突变体BpL2-1369有4个碱基发生了突变: T61C/C147T/A334G/T371A, 其中有3个碱基突变导致了氨基酸的改变。通过SWISS-MODEL数据库模拟脂肪酶的结构显示, 3个突变氨基酸分别位于第1个a螺旋的第3个氨基酸、第4和第5个b折叠之间的转角以及第5个b折叠的第1个氨基酸位置。将野生型脂肪酶基因BpL和进化后的基因BpL2-1369的高效表达产物经Ni-Agarose柱和Sephadex-G75纯化后, 酶学性质测定表明: 突变脂肪酶的比活力比野生型脂肪酶提高了1.31倍, Km值由8.24 mmol/L降低至7.17 mmol/L; 在pH>8.0时的稳定性较野生型脂肪酶有所提高。  相似文献

The current study aimed at evaluating the possibility of native Bacillus pumilus species to control Fusarium wilt in tomato and examine its effect on plant growth. Biocontrol traits of B. pumilus strains, biofilm assay, root colonisation and in vivo studies under pot conditions were determined. Strain ToIrMA-KC806242 formed biofilm efficiently and could colonise and survive on tomato rhizosphere (3.1 × 104 CFU/g of root). The amount of auxin production was recorded 29.7 μg/ml at the 96th hour of incubation. Siderophore production was determined positive, while ToIrMA was not able to solubilise phosphate compounds or produce cyanide hydrogen. Statistical analysis of data revealed that the increase in root and shoot length was recorded 60 and 84%, respectively, over control. In addition, about 73% reduction in disease incidence was determined in vivo experiments. In conclusion, this study suggests B. pumilus ToIrMA strain as a possible biocontrol agent in the field experiments.  相似文献

The now finished genome sequence of Bacillus licheniformis DSM 13 allows the prediction of the genes involved in protein secretion into the extracellular environment as well as the prediction of the proteins which are translocated. From the sequence 296 proteins were predicted to contain an N-terminal signal peptide directing most of them to the Sec system, the main transport system in Gram-positive bacteria. Using 2-DE the extracellular proteome of B. licheniformis grown in different media was studied. From the approximately 200 spots visible on the gels, 89 were identified that either contain an N-terminal signal sequence or are known to be secreted by other mechanisms than the Sec pathway. The extracellular proteome of B. licheniformis includes proteins from different functional classes, like enzymes for the degradation of various macromolecules, proteins involved in cell wall turnover, flagellum- and phage-related proteins and some proteins of yet unknown function. Protein secretion is highest during stationary growth phase. Furthermore, cells grown in complex medium secrete considerably higher protein amounts than cells grown in minimal medium. Limitation of phosphate, carbon and nitrogen sources results in the secretion of specific proteins that may be involved in counteracting the starvation.  相似文献

利用TAIL-PCR(Thermal asymmetric interlaced PCR)从短小芽孢杆菌基因组中扩增到碱性蛋白酶基因编码区上游的启动子片段。对该片段的序列测定和分析表明, 此片段长797 bp, 但与基因表达有关的序列长约390 bp。对启动子片段进行不同长度的缺失突变, 以获得最小的基因启动子片段, 结果表明, 该基因起始密码子上游约160 bp的DNA片段就可以启动基因的表达。将含有该片段的碱性蛋白酶基因WApQ3插入大肠杆菌-芽孢杆菌穿梭质粒载体pSUGV4中, 构建了碱性蛋白酶基因表达质粒pSUBpWApQ3。将该质粒分别转入枯草芽孢杆菌和短小芽孢杆菌中表达, 可在胞外检测到碱性蛋白酶活性, 最高酶活分别为466.5 U/mL和3060 U/mL。  相似文献

A bacterial strain S-1 capable of transforming isoeugenol to vanillin was isolated. The strain was identified as Bacillus pumilus based on biochemical tests, cellular fatty acid composition, riboprint pattern and 16S rRNA gene sequence analyses. In the biotransformation of isoeugenol, vanillin was the main product. With the growing culture of B. pumilus S-1, 10 g l−1 isoeugenol was converted to 3.75 g l−1 vanillin in 150 h, with a molar yield of 40.5% that is the highest up to now. Dehydrodiisoeugenol, a dimer of isoeugenol, was separated by preparative thin layer chromatography and identified by gas chromatography–mass spectrometry. Based on the accurate masses obtained from gas chromatography–high resolution mass spectrometry, two key intermediates, isoeugenol-epoxide (IE) and isoeugenol-diol (ID), were identified by mass spectra interpretations. The biotransformation with resting cells showed that vanillin was oxidized to vanillic acid and then to protocatechuic acid before the aromatic ring was broken. These findings suggest that isoeugenol is degraded through an epoxide-diol pathway.  相似文献

研究了搅拌转速、PH控制以及插瓶发酵过程中不同时间硫酸铵的补加对β-1,4聚糖酶形成的影响,优化得到A-30的β01,4-聚糖酶分批发酵操作条件和初步优化了(NH4)2SO4流加发酵条件。研究结果显示麸皮表面有大量A-30菌体细胞的吸附,搅拌转速对菌体吸附和β-1,4-聚糖酶的形成有明显影响;发酵过程中PH下降有利于β-1,4-聚糖酶形成。采用了实期以5ml/h的速率恒速流加,后期测定(NH4)2  相似文献

从34株细菌中筛选到一株木聚糖酶高产菌株BacilluspumilusH-101。适宜的产酶培养基含2.0%小麦秸半纤维素、0.2%(NH4)2SO4、0.1%NH4NO3、0.1%K2HPO4、0.01%MgSO4·7H2O、0.02%NaC1、0.02%CaCl2·2H2O和1.0%的酵母膏。在上述培养基中,32℃振荡培养48h,总半纤维素酶活力达235.6u/ml,木聚精酶活力达1164.2u/ml酶反应的最适温度为50℃,最适酸碱度为pH6.5;Na 、Mg2 等离子可提高木聚精酶的水解活性,而Zn2 、Cu2 等离子则对木聚糖酶活性有明显的抑制作用。  相似文献

Six isolates of Bacillus pumilus were tested for their ability to inhibit aflatoxin production of Aspergillus parasiticus NRRL 2999 in yeast extract sucrose (YES) broth. Aflatoxin production was inhibited in both simultaneous and deferred antagonism assays, suggesting that the inhibitory activity was due to extracellular metabolite(s) produced in cell-free supernatant fluids of cultured broth. The inhibition was not due to organic acids or hydrogen peroxide produced by B. pumilus since the inhibitory activity was not lost after pH adjustment or treatment of supernatant fluids with catalase. A range of media tested for the production of inhibitory metabolite(s) in supernatant fluids showed that all media supported bacterial growth and production of the metabolite(s). The metabolite(s) were produced over a wide range of temperature (25 to 37°C) and pH (4 to 9) of growth of B. pumilus. They were stable over a wide range of pH (4 to 10) and were not inactivated after autoclaving at 121°C for 30 minutes. AIMS: The present study was conducted by screening zein-degrading bacteria in an attempt to obtain zein-degrading protease. METHODS AND RESULTS: Soil bacteria were screened by formation of a clear zone on zein plates. Characterization of a zein-degrading bacterium indicated a taxonomic affiliation to Bacillus pumilus, and was named MS-1 strain. The strain produced two different types of extracellular proteases, BPP-A and BPP-B. In this study, we purified and characterized BPP-A because it exhibited a higher ability to hydrolyze zein than BPP-B. When casein was used as the substrate, the optimal pH for BPP-A was 11.0. In BPP-A, zein was better substrate than casein at pH 13.0, whereas casein was better one than zein at pH 11.0. The bppA gene encoded a 383-amino acid pre-pro form of BPP-A, and mature BPP-A contained 275 amino acid residues. It was concluded that BPP-A belonged to the subtilisin family. CONCLUSION: A zein-degrading bacterium assigned to B. pumilus produced two different types of extracellular proteases, BPP-A and BPP-B. BPP-A exhibited an ability to hydrolyze zein in an extreme alkaline condition. SIGNIFICANCE AND IMPACT OF THE STUDY: This is a first report on screening for zein-degrading micro-organisms. The subtilisin-like protease BPP-A is possible to utilize as an industrial enzyme for the production of zein hydrolysates.  相似文献

AIMS: To characterize and optimize a novel Bacillus pumilus strain isolated from biological waste which produces protease with excellent dehairing effect. This newly isolated strain could be utilized in the industrial leather dehairing process. METHODS AND RESULTS: Bacterial strains secreting proteases were screened from biological wastes. Positive clones were further characterized by analysing their efficacy in dehairing and effects on collagen integrity. Among 171 colonies tested, a strain BA06, identified as B. pumilus, was picked owing to its efficient dehairing capabilities with minimal impact on collagen. By combined mutagenesis using UV, N-methyl-N'-nitro-N-nitrosdguanidine and Co(60)-gamma-rays, this strain was further improved with regard to its alkaline protease production. The alkaline protease activity of the mutant strain SCU11was greatly improved up to 6000 U ml(-1), in comparison with its parent strain BA06 of 1200 U ml(-1). CONCLUSIONS: By using screening and mutagenesis methods, we have successfully created a B. pumilus strain that can produce high levels of alkaline proteases that are able to efficiently remove hair from skin with minimal damage on the collagen. SIGNIFICANCE AND IMPACT OF THE STUDY: This strain could be used in commercial alkaline protease production for leather dehairing.  相似文献

探索Bacillus pumilusWHK4以羽毛粉为底物产酶的最佳条件和最佳培养基组成。以羽毛粉发酵培养基为基础,首先采用单因子试验考察底物浓度、初始pH、接种量、外加碳源、外加氮源对WHK4产酶活力的影响。在单因子试验的基础上采用正交试验设计对底物浓度、温度、初始pH、接种量、外加(NH4)2SO4、外加麦芽糖进行优化。结果显示:Bacillus pumilusWHK4最佳的产酶条件为初始pH7.38,菌龄16 h,接种量5%,37℃。最佳的培养基组成为:1 L基础发酵培养基,40.0 g羽毛粉,10.0 g(NH4)2SO4和10.0 g麦芽糖。在优化的条件下Bacillus pumilusWHK4 24 h产蛋白酶活力为每毫升90 U。对培养条件和培养基的优化为Bacillus pumilusWHK4产蛋白酶的分离纯化奠定了基础。  相似文献

短小芽孢杆菌(Bacillus pumilus)是一种能引起食源性疾病的腐败菌,对其进行快速检测具有重要意义。针对短小芽孢杆菌木聚糖(xynA)基因,设计了4条特异性引物(两条内引物和两条外引物),通过条件优化,首次将一种新颖的核酸扩增技术——环介导恒温扩增技术应用于短小芽孢杆菌的快速检测。采用该技术,63℃温育1h的条件下扩增短小芽孢杆菌DNA,琼脂糖凝胶电泳得到特异性梯度条带。PCR和LAMP的检测灵敏度分别约为162和16.2拷贝每反应。结果表明,该方法检测短小芽孢杆菌特异性强、灵敏度高、操作简便、检测成本低,1h即可完成,有望发展成为快速检测短小芽孢杆菌的有效手段。  相似文献

目的:对从新疆精河地区一处工业污水中分离得到的一株产碱性脂肪酶细菌进行研究.方法:通过生理生化检测,16SrDNA序列同源性分析和G+Cmol%含量的测定对命名为XJU-13的这株菌进行鉴定.结果:该菌株可在pH 3.0~12.5的广泛酸碱泛围的营养肉汤培养基中生长.最适生长温度为37℃.基于16S rDNA序列同源性构建系统进化树分析表明与Bacillus pumilus clone B257聚在同一亚分枝,序列相似性达100%.数据证明XJU-13属于Bacillus pumilus.由于在氧化酶反应及淀粉水解实验与伯杰氏鉴定手册有差异,具不可比拟的pH耐受性,且脂肪酸含量与参考菌株差异较大,认为这是Bacillus pumilus中的一个新品系.该菌株产生的脂肪酶最适pH为10,最适温度为35℃,且在广泛pH(pH4-10)范围具稳定性.酶活可被Mg2+、K+、Ba2+、Pb+盐强烈抑制,被Ca2+、Cu2+、Al+及Fe2+盐激活.Zn2+对酶活无影响.结论:实验表明,XJU-13应属于B.pumilus.B.pumilus XJu-13中分离到的碱性脂肪酶有很好的特性及潜能,以期为工业应用提供数据.  相似文献

A marine bacterium (KMM 1364), identified as Bacillus pumilus, was isolated from the surface of ascidian Halocynthia aurantium. Structural analysis revealed that the strain KMM 1364 produced a mixture of lipopeptide surfactin analogs with major components with molecular masses of 1035, 1049, 1063, and 1077. The variation in molecular weight represents changes in the number of methylene groups in the lipid and/or peptide portions of the compounds. Structurally, these lipopeptides differ from surfactin in the substitution of the valine residue in position 4 by leucine, and have been isolated as two carboxy-terminal variants, with valine or isoleucine in position 7. As constituents of the lipophilic part of the peptides, only β-hydroxy-C15-, β-hydroxy-C16-, and a high amount of β-hydroxy-C17 fatty acid were determined.  相似文献
