The effects of l-arginine on porcine foetal development and myogenesis were determined. Twenty Swiss Large White gilts were randomly allocated to either the control (C) or l-arginine treatment (A). In addition to the standard gestation diet, A-sows received 26 g l-arginine daily from days 14 to 28 of gestation. At day 75 of pregnancy, sows were sacrificed and the number and weight of foetuses were recorded. From each litter, the lightest, heaviest and the ones with an average foetal weight (FtW) were selected. Primary (P), secondary (S) and total myofiber number as well as S/P ratio were determined in the semitendinosus (ST) and rhomboideus (RH) muscles. In A-sows, the number of viable foetuses (13.0 v. 9.3) and total FtW (4925 v. 3729 g) was greater (P ⩽ 0.04) than in C-sows. Compared to C-sow foetuses, the ST of A-sow foetuses had 7% more (17 699 v. 16 477; P = 0.04) P myofibers and the S/P ratio in both muscles was lower (ST = 20.3 v. 21.5; RH = 24.1 v. 27.1; P ⩽ 0.07). Regardless of the maternal diet, the S myofiber number and the S/P ratio in both muscles were greater (P ⩽ 0.01) in foetuses with a high FtW compared to low FtW. These data suggest that l-arginine supplemented to gilts during early gestation enhanced foetal survival and in the ST positively affected the primary phase of myofiber formation. 相似文献

Suboptimal embryonic/fetal survival and growth remains a significant problem in mammals. Using a swine model, we tested the hypothesis that dietary l-arginine supplementation during gestation may improve pregnancy outcomes through enhancing placental growth and modulating hormonal secretions. Gestating pigs (Yorkshire?×?Landrace, n?=?108) were assigned randomly into two groups based on parity and body weight, representing dietary supplementation with 1.0% l-arginine–HCl or 1.7% l-alanine (isonitrogenous control) between days 22 and 114 of gestation. Blood samples were obtained from the ear vein on days 22, 40, 70 and 90 of gestation. On days 40, 70 and 90 of gestation, concentrations of estradiol in plasma were higher (P?<?0.05) in arginine-supplemented than in control sows. Moreover, arginine supplementation increased (P?<?0.05) the concentrations of arginine, proline and ornithine in plasma, but concentrations of urea or progesterone in plasma did not differ between the two groups of sows. Compared with the control, arginine supplementation increased (P?<?0.05) the total number of piglets by 1.31 per litter, the number of live-born piglets by 1.10 per litter, the litter birth weight for all piglets by 1.36?kg, and the litter birth weight for live-born piglets by 1.70?kg. Furthermore, arginine supplementation enhanced (P?<?0.05) placental weight by 16.2%. The weaning-to-estrus interval of sows was not affected by arginine supplementation during gestation. This study was conducted to test the hypothesis that dietary L-arginine supplementation enhances immunity in early weaned piglets. Seventy piglets weaned at 7 days of age were assigned to five groups (14 pigs/group), representing supplementation of 0.0, 0.2, 0.4, 0.6, and 0.8% L-arginine to a milk-based formula. On Day 7 after initiation of treatment, spleen weight in piglets supplemented with 0.2 and 0.8% arginine was heavier and thymus size was larger in piglets supplemented with 0.6% arginine, whereas serum concentration of immunoglobulin (Ig) M was higher but that of IL-8 was lower in piglets supplemented with 0.6 and 0.8% arginine, compared with the control group. Dietary supplementation with 0.8% arginine increased the numbers of white blood cells and granulocytes, and gene expression of interleukin (IL)-8 in spleen. On Day 14, compared with control piglets, granulocyte numbers were greater but lymphocyte numbers were lower in piglets supplemented with 0.2 and 0.4% arginine, whereas splenic expression of IL-8 and tumor necrosis factor-alpha genes was increased in piglets supplemented with 0.8% arginine. Additionally, IgG and IgM concentrations in serum and growth performance were greater in piglets supplemented with 0.4-0.8% arginine, compared with unsupplemented piglets. Collectively, dietary supplementation with 0.4-0.8% L-arginine for 2 weeks enhances both cellular and humoral immunity in piglets by modulating the production of leukocytes, cytokines and antibodies. These results indicate that increasing L-arginine provision is beneficial for optimal immune responses in young pigs and also have important implications for designing the next generation of improved formula for human infants. 相似文献

Norwegian Landrace gilts were inseminated on the second day of their second oestrus and slaughtered 28 to 34 days after insemination. The number of corpora lutea (ovulation rate) and normal embryos was counted and the embryonic survival rate was calculated for the 306 pregnant gilts. Mean (+/-S.D.) ovulation rate, number of normal embryos and embryonic survival rate were 14.17+/-2.48, 10.55+/-3.30 and 74.8%+/-20.7%, respectively. The significant (P<0.001) curvilinear regression of embryonic survival rate on ovulation rate gives a maximum embryonic survival rate at 13.2 ovulations. Increased ovulation rate gives increased number of normal embryos up to 18.1 ovulations. Ovulation rate should be considered when assessing factors affecting embryonic survival in pigs. 相似文献

Administration of estrogen to gilts on Days 9 and 10 of pregnancy results in total embryonic loss by Day 18. The present study examined changes in the uterine endometrial surface and secretion during conceptus attachment in control and estrogen-treated (Days 9 and 10) pregnant gilts. Gilts were unilaterally hysterectomized on either Days 12 and 14 or Days 16 and 18 of gestation. Uterine horns were flushed with saline and conceptuses were evaluated. Intact conceptuses were recovered from all control gilts, whereas estrogen-treated gilts contained normal intact conceptuses only on Day 12 of gestation. Antiviral activity, which reflects conceptus viability, was reduced (p less than 0.01) in uterine flushings after Day 14 in estrogen-treated gilts. Culture of endometrial explants with [3H]glucosamine revealed several glycoproteins that are synthesized during the period of conceptus attachment; however, no difference in glycoprotein synthesis between treatment groups was detected by analysis with two-dimensional PAGE and fluorography. Analyses of the uterine epithelium by scanning and transmission electron microscopy demonstrated that estrogen administration caused an alteration in the uterine surface, a thinning of the uterine epithelial glycocalyx, and a reduction of cationic ferritin binding to the microvilli of the uterine epithelium. Results indicate that conceptus mortality after early administration of estrogen is associated with alterations in the uterine endometrial surface during the period of conceptus attachment in the pig. 相似文献

A total of 166 crossbred gilts weighing approximately 87 +/- 1 kg was limit-fed (2.5 kg/d) a corn-soybean meal gestation diet containing either 0 or 220 ppm of chlortetracycline (CTC) from 157 +/- 1 d of age until 15 d after breeding. These gilts were slaughtered at 31 +/- 1 or 71 d +/- 1 d of gestation for evaluation of reproductive performance. Age (190 +/- 3 d vs 195 +/- 3 d) and body weights (106 +/- 2 kg vs 106 +/- 2 kg) at puberty were similar for control and CTC-fed gilts, respectively. Although not significant (P > 0.05), ovulation rate was higher in CTC-fed than in control gilts as assessed at both 31 d (14.2 +/- 0.7 vs 12.9 +/- 0.9, P = 0.31) and 71 d (13.9 +/- 0.6 vs 12.4 +/- 0.5, P = 0.10) of gestation. There was an increase (P = 0.04) in the number of live embryos for CTC-fed gilts at 31 d (12.1 +/- 0.7 vs 9.7 +/- 0.7) but not at 71 d (10.0 +/- 1.1 vs 9.6 +/- 1.0) of gestation. The mean uterine length, placental length, placental weight, fetal length, fetal weight, and allantoic fluid volumes were similar between the control and CTC-fed gilts. Results indicated that feeding CTC during prebreeding and early gestation did not influence the proportion or age of gilts at puberty. However, CTC feeding may have influenced a trend to-ward an increased ovulation rate and increased number of live embryos in gilts. 相似文献

Arginine plays an important role in preventing intestinal dysfunction and metabolic disorders caused by early weaning stress. However, little is known about how arginine mitigates early weaning stress. This study was conducted to evaluate the effects of weaning stress and dietary arginine supplementation on the metabonome in the serum of piglets using (1)H NMR spectroscopy in conjunction with multivariate data analysis. Thirty castrated male piglets aged 21 d were evenly divided into three groups and fed in three different regimes: sow-fed (SF), weaned with l-alanine supplementation (ALA), and weaned with arginine supplementation (ARG). We found that early weaning stress led to a significantly reduced bodyweight gain (15.6%) and that supplementation with arginine can improve growth rates in piglets by 5.6% (P < 0.05). The early weaning stress was associated with marked alterations in lipid and amino acid metabolisms and perturbations in population and/or activities of gut microorganisms, which were manifested in increased levels of organic acids, amino acids, and acetyl-glycoproteins and reduced levels of choline metabolism and lipoproteins. Dietary supplementation with arginine could partially counteract the changes of metabolites induced by weaning stress, such as lipid and amino acid metabolisms. However, arginine was not able to restore disturbed gut microbiota. These results demonstrate the central role of arginine supplementation in regulating the metabolisms of weaned piglets. 相似文献

(4) crypt depth in jejunum and ileum, (5) goblet cell counts in intestinal mucosae, and (6) whole-body weight gain in pigs.
Real-time polymerase chain reaction and western blotting analyses revealed that both mRNA and protein levels for heat shock
protein-70 (HSP70) were higher (P < 0.05) in the intestinal mucosae of Arg- or NCG-supplemented pigs than in the control group. Furthermore, the incidence
of diarrhea in the NCG group was 18% lower (P < 0.01) than that in the control group. Collectively, these results indicate that dietary supplementation with 0.6% Arg or
0.08% NCG enhances intestinal HSP70 gene expression, intestinal growth and integrity, and the availability of dietary nutrients

Obesity in humans is a major public health crisis worldwide. In addition, livestock species exhibit excessive subcutaneous fat at market weight. However, there are currently few means of reducing adiposity in mammals. This study was conducted with a swine model to test the hypothesis that dietary L-arginine supplementation may increase muscle gain and decrease fat deposition. Twenty-four 110-day-old barrows were assigned randomly into two treatments, representing supplementation with 1.0% L-arginine or 2.05% L-alanine (isonitrogenous control) to a corn- and soybean meal-based diet. Growth performance was measured based on weight gain and food intake. After a 60-day period of supplementation, carcass and muscle composition were measured. Serum triglyceride concentration was 20% lower (P < 0.01) but glucagon level was 36% greater (P < 0.05) in arginine-supplemented than in control pigs. Compared with the control, arginine supplementation increased (P < 0.05) body weight gain by 6.5% and carcass skeletal-muscle content by 5.5%, while decreasing (P < 0.01) carcass fat content by 11%. The arginine treatment enhanced (P < 0.05) longissimus dorsi muscle protein, glycogen, and fat contents by 4.8, 42, and 70%, respectively, as well as muscle pH at 45 min post-mortem by 0.32, while reducing muscle lactate content by 37%. These results support our hypothesis that dietary arginine supplementation beneficially promotes muscle gain and reduces body fat accretion in growing-finishing pigs. The findings have a positive impact on development of novel therapeutics to treat human obesity and enhance swine lean-tissue growth. 相似文献

l-Arginine is a nutritionally essential amino acid for spermatogenesis and plays versatile roles in animal health and can be utilized as a potential agent to improve reproductive performance of boars under high ambient temperature. The present study aimed to determine whether dietary l-arginine could alleviate heat stress-induced infertility in boars. In all, 20 boars (PIC 1040; 248.59±3.84 kg BW and 407.65±6.40 days of age) were selected and randomly assigned to four groups (group 0.0%, basal diet; group 0.6%, 0.8% or 1.0%, basal diet added with 0.6%, 0.8% or 1.0% l-arginine (wt:wt), respectively.) The four diets were made isonitrogenous by addition of appropriate amounts of l-alanine. Boars were pre-fed the corresponding experimental diet for 42 days. Then, the semen characteristics and libido were accessed for 6 weeks during the hot summer period (25.5° to 33.0°C). Results show that dietary l-arginine remarkably improved sperm motility, normality, total sperm number and effective total sperm number. Also, dietary l-arginine improved semen antioxidant capacity, such as decrease of malondialdehyde and 8-Hydroxy-2'-deoxyguanosine content in sperm (P<0.05), increase of the ratio of glutathione and oxidized glutathione, total antioxidant capacity, glutathione peroxidase and catalase activities in seminal plasma (P<0.05). Most of mitochondria contained intact ultrastructure in l-arginine-supplemented group which also accompany with higher ATP content than the 0.0% group. The boars fed 0.8% l-arginine show increased levels of estradiol-17β and testosterone and exhibit improved libido performance than boars in the 0.0% group. Adding dietary l-arginine linearly increased (P=0.002) nitric oxide content (as l-arginine increased). The scrotal surface temperature in the 0.6%, 0.8% and 1.0% group were decreased by 0.9°C, 0.9°C and 0.4°C, respectively, compared with the 0.0% group. l-Arginine levels caused linear effect on semen quality and antioxidant capacity, also caused quadratic effect on libido performance. During the hot summer months, the predicted optimal l-arginine levels for best semen quality and antioxidant capacity was 0.8% to 1.0% and for best libido performance was 0.8%. It can be concluded that l-arginine can be used as an effective agent to alleviate heat stress-induced infertility of boar, and that 0.8% to 1.0% can be considered as the optimum dosage. 相似文献

Obesity is a major health crisis worldwide and new treatments are needed to fight this epidemic. Using the swine model, we recently reported that dietary l-arginine (Arg) supplementation promotes muscle gain and reduces body-fat accretion. The present study tested the hypothesis that Arg regulates expression of key genes involved in lipid metabolism in skeletal muscle and white adipose tissue. Sixteen 110-day-old barrows were fed for 60 days a corn- and soybean-meal-based diet supplemented with 1.0% Arg or 2.05% l-alanine (isonitrogenous control). Blood samples, longissimus dorsi muscle and overlying subcutaneous adipose tissue were obtained from 170-day-old pigs for biochemical studies. Serum concentrations of leptin, alanine and glutamine were lower, but those for Arg and proline were higher in Arg-supplemented pigs than in control pigs. The percentage of oleic acid was higher but that of stearic acid and linoleic acid was lower in muscle of Arg-supplemented pigs, compared with control pigs. Dietary Arg supplementation increased mRNA levels for fatty acid synthase in muscle, while decreasing those for lipoprotein lipase, glucose transporter-4, and acetyl-coenzyme A carboxylase-α in adipose tissue. Additionally, mRNA levels for hormone sensitive lipase were higher in adipose tissue of Arg-supplemented pigs compared with control pigs. These results indicate that Arg differentially regulates expression of fat-metabolic genes in skeletal muscle and white adipose tissue, therefore favoring lipogenesis in muscle but lipolysis in adipose tissue. Our novel findings provide a biochemical basis for explaining the beneficial effect of Arg in improving the metabolic profile in mammals (including obese humans). 相似文献

This experiment was designed to measure the effects of a dose of PMSG (600 IU) or a five-day flush feeding, singly or in combination, on ovulation rate, early embryonic survival, litter size at term and plasma hormone levels during early gestation in 64 Duroc gilts. They were assigned to four treatments, with half of each treatment killed on day 25 of pregnancy and the other half continuing to term.Although treatment with 600 IU of PMSG increased the number of ovulations compared to the controls (P < .05), it failed to increase the number of viable embryos at day 25 of gestation (P > .05). Flush feeding for five days before breeding yield similar results. Fewer embryos were present at day 25 of gestation in those animals receiving a combination /flush feeding treatment than in any other treatment group (P < 0.01). At day 25 of gestation, the average prenatal survival rate was 71% for all treatments. At parturition, there were no differences between treatment groups in the average number of pigs born per litter; nor were there any differences present for the average number of pigs weaned per litter at four weeks postpartum. Plasma progesterone levels at the onset of treatment, mating and during early gestation were found to be similar among the four treatment groups. Plasma estradiol-17β levels were also similar among the four groups. Based on this experiment, it would appear that factors other than the mere number of viable eggs shed determines the litter size at parturition in swine. 相似文献

Thirty-nine pregnant adult Wistar strain rats were randomly assigned to one of three exposure groups: 0, 0.75, or 1.50 Gy X-radiation total exposure. Animals were exposed from the 14th to the 18th days of gestation at 0, 0.15, or 0.30 Gy per day. At term, 15 rats were killed and morphologic analyses were completed. Twenty-four rats were allowed to deliver their offspring. On the first day of postnatal life, litters were reduced to a maximum of eight pups per litter, with equal numbers of male and female offspring wherever possible. A total of 187 pups were observed for the age of acquisition of five reflexes (air righting, surface righting, visual placing, negative geotaxis, auditory startle) and the appearance of four physiologic markers (pinna detachment, eye opening, vaginal opening, testes descent). There was significant dose-related weight reduction in term fetuses and offspring throughout the 86-day postnatal period. Postnatal growth rate (g gained/day) was unaffected. Adult offspring brain and gonadal weight and organ weight:body weight ratios were reduced. Using the PAC50 methodology, dose-related alterations occurred in the acquisition of several reflexes. All physiologic markers exhibited a dose-related delay in appearance. These results indicate that fractionated exposure to X-radiation during the fetal period in the rat results in dose-dependent alterations in postnatal growth and physiologic development. These studies are important for our understanding of the long-range effects of prenatal exposure to ionizing radiation late in gestation. 相似文献

The size of mammalian species involves the interaction of multiple genetic modifiers that control the timing and extent of growth mechanisms. Disruption of the paternal allele of the imprinted embryonic gene coding for insulin-like growth factor 2 (IGF2, Igf2(+m/-p)), results in viable mice that are 60% the weight of wild-type littermates. Differences in weight are first detected at embryonic day (E) 11, and the growth deficit is maintained throughout life. We report the mechanisms that account for this unusual phenotype. In order to quantify growth, we used novel methods to generate single cell suspensions of post-implantation mouse embryos. We were then able to quantify cell number, cell proliferation and cell death between E8.5 and E11.5 using flow cytometry. Determination of total embryo cell number also allowed us to time litters by a method other than by plugging. Wild-type and Igf2(+m/-p) embryos accumulated similar total cell numbers up to E9.25, but cell number began to diverge by around E9.5, with significant differences by E11 (75% of wild type). A relative increase in pyknotic nuclei, sub-GI cytometry counts and caspase activity, all indicative of cell death, occurred in Igf2(+m/-p) embryos at E9.25, reverting to wild-type levels by E9.75. This was followed at E9.75 by a significant reduction in the proportion of cells in S phase, quantified by S-phase cytometry counts and BrdU labelling. No significant differences in cell size were detected. We conclude that the majority of the cell number differences between wild-type and Igf2(+m/-p) mice can be accounted for by modification of cell survival and proliferation during the period (E9 to E10) of post-implantation development. 相似文献

The present paper considers the significance of a variety of cranial flexions in the process of orthocephalization of the rat skull between 14 and 60 days postnatally. The study is based on a sample of 27 male rats, who have been x-rayed at 14, 30 and 60 days with subsequent analyses of the photographs obtained. In this period the angle between the cranial base and the facial part of the skull becomes more obtuse, i.e. the skull becomes more orthocranial. The cranial base becomes at the same time more airobasal (lordotic). Angular changes between the individual bones in the cranial vault straightens the vault markedly. By this it becomes orthodorsal. As the angle between the basisphenoid and the parietal bones stays more or less constant between 14 and 60 days, the impression is created that both the anterior and posterior parts of the neural skull rotate upwards relative to this bone complex. Thus, orthocephalization in the period between 14 and 60 days both consists of flexions between the facial and neural parts of the skull (prebasal flexions), and may be more importantly of interosseous ventral and dorsal flexions. 相似文献

Between Days 9 and 15 after oestrus, concentrations of pregnenolone, pregnenolone sulphate, dehydroepiandrosterone (DHEA), DHEA sulphate, androstenedione, oestrone and oestrone sulphate in free uterine fluid collected from non-pregnant gilts were greater than respective values in plasma (P less than 0.05). The total contents of pregnenolone, progesterone, DHEA, testosterone, oestrone and oestradiol in washings from pregnant uteri exceeded (P less than 0.05) respective non-pregnancy levels during this same period. Concentrations of pregnenolone, pregnenolone sulphate, DHEA, DHEA sulphate, androstenedione, oestrone, oestrone sulphate and oestradiol in free uterine fluid recovered from gravid uteri were also higher (P less than 0.05) than respective plasma values. By contrast, the progesterone concentration in uterine fluid from pregnant animals was lower (P less than 0.001) than the plasma value. Concentrations of DHEA, DHEA sulphate, androstenedione and oestrone sulphate in plasma of pregnant gilts between Days 9 and 15 after mating exceeded (P less than 0.05) the respective concentrations in unmated gilts between Days 9 and 15 after oestrus. Plasma levels of pregnenolone sulphate were lower (P less than 0.05) in the pregnant animals. We therefore suggest that the endometrium of the pig can concentrate steroid hormones in uterine fluid and that increases in steroid levels in this milieu between Days 9 and 15 after coitus reflect steroidogenesis by embryonic tissues and modification of enzyme activities within uterine tissues under the influence of progestagens. The pool of steroid sulphoconjugates present in uterine fluid between Days 9 and 15 post coitum could serve as an important precursor source for progestagen, androgen and oestrogen synthesis by tissues of pig embryos before implantation. 相似文献

In this study, the localization and appearance of neuronal nitric oxide synthase-immunoreactive (nNOS-IR) nerve cells and their relationships with the developing gastric layers were studied by immunocytochemistry techniques and light microscopy in embryonic rat stomach. The stomachs of Wistar rat embryos aged 13-21 days were used. The first nerve cells containing nNOS-IR were seen on embryonic Day 14. The occurrence of mesenchymal cell condensation near nNOS-IR neuroblasts on embryonic Day 15 may reflect an active nerve element-specific mesenchymal cell induction causing the morphogenesis of muscle cells. Similarly, the appearance of glandular structures after nNOS-IR neuroblasts, on embryonic Day 18, suggests that the epithelial differentiation may depend on inputs coming from nNOS-IR neuroblasts, as well as other factors. Observation of nNOS-IR nerve fibers on embryonic Day 21 demonstrates that at this stage they contribute to nonadrenergic noncholinergic relaxation. In conclusion, depending on this study's results, it can be said that cells and tissues might be affected by NO secreted by nNOS-IR nerve cells during the development and differentiation of embryonic rat stomach. 相似文献
