Despite 40+ years of research on aluminum (Al) toxicity in aquatic organisms, Al transport mechanisms through biological membranes, and the intracellular fate of Al once assimilated, remain poorly understood. The trivalent metal scandium shares chemical similarities with Al and, unlike Al, it has a convenient radioactive tracer (Sc-46) allowing for relatively simple measurements at environmentally relevant concentrations. Thus, we investigated the potential of Sc to substitute for Al in uptake and intracellular fate studies with the green alga Chlamydomonas reinhardtii. Short-term (<60 min) competitive uptake experiments indicated that Al does not inhibit Sc influx, implying that these metals do not share a common transport mechanism. Also, internalized Al concentrations were ~4 times higher than Sc concentrations after long-term (72 h) exposures under similar conditions (4.5 μM AlT or ScT, 380 μM FT, pH 7.0, 3.8 pM Al calc 3+ and 1.0 pM Sc calc 3+ ). However, interesting similarities were observed in their relative subcellular distributions, suggesting possible common toxicity/tolerance mechanisms. Both metals mostly distributed to the organelles fraction and almost no association was found with the cytosolic proteins. The greatest difference was observed in the cellular debris fraction (membranes and nucleus) where Al was much more concentrated than Sc. However, it is not clear whether or not this fraction contained extracellular metal associated with the algal surface. To summarize, Sc does not seem to be an adequate substitute of Al for transport/uptake studies, but could be for investigations of toxicity/tolerance mechanisms in C. reinhardtii. Further work is needed to verify this latter suggestion.  相似文献

Lead (Pb) causes severe damage to crops, ecosystems, and humans, and alters the physiology and biochemistry of various plant species. It is hypothesized that Pb-induced metabolic alterations could manifest as structural variations in the roots of plants. In light of this, the morphological, anatomical, and ultrastructural variations (through scanning electron microscopy, SEM) were studied in 4-day-old seedlings of Triticum aestivum grown under Pb stress (0, 8, 16, 40, and 80 mg Pb2+ l?1; mild to highly toxic). The toxic effect was more pronounced in radicle growth than on the plumule growth. The SEM of the root of T. aestivum depicted morphological alterations and surface ultrastructural changes. Compared to intact and uniform surface cells in the control roots, cells were irregular and desiccated in Pb2+-treated roots. In Pb2+-treated roots, the number of root hairs increased manifold, showing dense growth, and these were apparently longer. Apart from the deformity in surface morphology and anatomy of the roots in response to Pb2+ toxicity, considerable anatomical alterations were also observed. Pb2+-treated root exhibited signs of injury in the form of cell distortion, particularly in the cortical cells. The endodermis and pericycle region showed loss of uniformity post Pb2+ exposure (at 80 mg l?1 Pb2+). The cells appeared to be squeezed with greater depositions observed all over the tissue. The study concludes that Pb2+ treatment caused structural anomalies and induced anatomical and surface ultrastructural changes in T. aestivum.  相似文献

Inhibitor titration curves and discrimination against 18O2 by mitochondrial respiration in three strains of green algae (Selenastrum minutum [Naeg.] Collins, and two strains of Chlamydomonas reinhardtii Dangeard) with differing respiratory capabilities were determined. Discrimination for cytochrome pathway respiration ranged from 19.89 to 20.43%. Discrimination for alternative pathway respiration by wild-type C. reinhardtii (measured in the presence of KCN) was 25.46%, while discrimination values for a cytochrome oxidase deficient mutant of C. reinhardtii ranged from 24.24 to 24.96%. In the absence of KCN, the alternative pathway was not engaged in wild-type C. reinhardtii, the only algal strain that possessed both cytochrome and alternative pathway capacities.  相似文献

In a hydroponic setting, we investigated the possible role of phytochelatins (metal-binding peptides) in the lead (Pb) tolerance of vetiver grass (Vetiveria zizanioides L.). Pb was added to the nutrient medium at concentrations ranging from 0 to 1,200 mg L?1. Furthermore, we simulated the effect of soil phosphorus (P) on potentially plant available Pb by culturing vetiver grass in P-rich nutrient media. After 7 days of exposure to Pb, we evaluated the Pb uptake by vetiver grass. Results indicate that vetiver can accumulate Pb up to 3,000 mg kg?1 dry weight in roots with no toxicity. Formation of lead phosphate inhibited Pb uptake by vetiver, suggesting the need for an environmentally safe chelating agent in conjunction with phytoremediation to clean up soils contaminated with lead-based paint. Unambiguous characterization of phytochelatins (PCn) was possible using high pressure liquid chromatography coupled with electrospray ionization mass spectrometry (LC-ESMS). Vetiver shows qualitative and quantitative differences in PCn synthesis between root and shoot. In root tissue from vetiver exposed to 1,200 mg Pb L-1, phytochelatins ranged from PC1 to PC3. Collision-induced dissociation of the parent ion allowed confirmation of each PCn based on the amino acid sequence. Possible Pb-PC1 and Pb2-PC1 complexes were reported in vetiver root at the highest Pb concentration. The data from these experiments show that the most probable mechanism for Pb detoxification in vetiver is by synthesizing PCn and forming Pb–PCn complexes.  相似文献

Toxic effects of metals appear to be partly related to the production of reactive oxygen species (ROS), which can cause oxidative damage to cells. The ability of several redox active metals [Fe(III), Cu(II), Ag(I), Cr(III), Cr(VI)], nonredox active metals [Pb(II), Cd(II), Zn(II)], and the metalloid As(III) and As(V) to produce ROS at environmentally relevant metal concentrations was assessed. Cells of the freshwater alga Chlamydomonas reinhardtii P. A. Dang. were exposed to various metal concentrations for 2.5 h. Intracellular ROS accumulation was detected using an oxidation‐sensitive reporter dye, 5‐(and‐6)‐carboxy‐2′,7′‐dihydrodifluorofluorescein diacetate (H2DFFDA), and changes in the fluorescence signal were quantified by flow cytometry (FCM). In almost all cases, low concentrations of both redox and nonredox active metals enhanced intracellular ROS levels. The hierarchy of maximal ROS induction indicated by the increased number of stained cells compared to the control sample was as follows: Pb(II) > Fe(III) > Cd(II) > Ag(I) > Cu(II) > As(V) > Cr(VI) > Zn(II). As(III) and Cr(III) had no detectable effect. The effective free metal ion concentrations ranged from 10?6 to 10?9 M, except in the case of Fe(III), which was effective at 10?18 M. These metal concentrations did not affect algal photosynthesis. Therefore, a slightly enhanced ROS production is a general and early response to elevated, environmentally relevant metal concentrations.  相似文献

Glutathione (GSH) plays a central role in the plant tolerance against the toxic effects of metals. It is a key antioxidant and acts as a cofactor for glutathione S-transferase (GST). The main objective of this study was to determine the Pb tolerance and bioaccumulation by Dodonaea viscosa (L.) Jacq. and their relation to GSH production and GST activity. The relationship between the Pb tolerance and bioaccumulation by D. viscosa and the effect of the exposure time on the GSH production or the GST activity was assessed in trials with perlite under different Pb treatments. D. viscosa showed a remarkable tolerance to Pb [half-inhibitory concentration (IC50) = 2,797 mg kg?1] and accumulated up to 11,428 mg Pb kg?1 in dry roots with a limited translocation to shoots without any signs of phytotoxicity after 105 days of exposure. The stress caused by the fast Pb uptake rate (489 mg kg?1 day?1) during the first 10 days of exposure was strongly correlated to increased GSH contents (~1.3-fold) and GST activities (~3.6-fold) in both shoots and roots. The results indicate that the Pb stress triggered a defense mechanism that involved increased contents of GSH and GST activities, suggesting that both variables are involved in the tolerance of D. viscosa against Pb toxicity.  相似文献

In the present investigation, four macrophytes, namely Typha latifolia (L.), Lemna minor (L.), Eichhornia crassipes (Mart.) Solms-Laubach, and Myriophyllum aquaticum (Vell.) Verdc, were evaluated for their heavy metal (Cu, Pb, Hg, and Zn) hyperaccumulation potential under laboratory conditions. Tolerance analyses were performed for 7 days of exposure at five different treatments of the metals mixture (Cu+2, Hg+2, Pb+2, and Zn+2). The production of chlorophyll and carotenoids was determined at the end of each treatment. L. minor revealed to be sensitive, because it did not survive in all the tested concentrations after 72 hours of exposure. E. crassipes and M. aquaticum displayed the highest tolerance to the metals mixture. For the most tolerant species of aquatic macrophytes, The removal kinetics of E. crassipes and M. aquaticum was carried out, using the following mixture of metals: Cu (0.5 mg/L) and Hg, Pb, and Zn 0.25 mg/L. The obtained results revealed that E. crassipes can remove 99.80% of Cu, 97.88% of Pb, 99.53% of Hg, and 94.37% of Zn. M. aquaticum withdraws 95.2% of Cu, 94.28% of Pb, 99.19% of Hg, and 91.91% of Zn. The obtained results suggest that these two species of macrophytes could be used for the phytoremediation of this mixture of heavy metals from the polluted water bodies.  相似文献

Concentrations of four metals (Cu, Zn, Pb, and Cd) in the sediments of the Anzali Lagoon in the northern part of Iran were determined to evaluate the level of contamination and spatial distribution. The sediments were collected from 21 locations in the lagoon. At each lagoon site a core, 60 cm long, was taken. The ranges of the measured concentrations in the sediments are as follows: 17–140 mg kg?1 for Cu, 20–113 mg kg?1 for Zn, 1–37 mg kg?1 for Pb and 0.1–3.5 mg kg?1 for Cd in surficial (0-20 cm) and 16–87 mg kg?1 for Cu, 28.5–118 mg kg?1 for Zn, 3–20 mg kg?1 for Pb and 0.1–3.5 mg kg?1 for Cd in deep (40–60 cm) sediments. The results of the geoaccumulation index (Igeo) show that Cd causes moderate to heavy pollution in most of the study area. Environmental risk evaluation showed that the pollution in the Anzali Lagoon is moderate to considerable and the ranking of the contaminants followed the order: Cd > Cu > Pb > Zn. Some locations present severe pollution by metals depending on the sources, of which sewage outlets and phosphate fertilizers are the main sources of contaminants to the area.  相似文献

Marmara Sea is one of the main catching areas of mussels (Mytilus galloprovincialis) in Turkey, and a significant portion of the harvest has been exported mainly to European countries. In this study, Zn, Cu, Cd, Hg, and Pb in mussels from ten catching areas in Marmara Sea were analyzed to investigate health risks associated with consuming mussels. Mercury was not detected (<0.15 ppb) in any of the samples. The highest concentrations of Cu and Cd were 3.473 and 0.740 mg kg?1 (wet weight, WW), respectively, well below the maximum permissible levels. All samples contained Zn higher than 50 mg kg?1, while Pb was above the limits in the samples from stations 1, 4, 6, and 8. Mussels from Marmara Sea are safe regarding Cu, Cd, and Hg but may contain Zn and Pb above the permissible limits. However, metal contents of mussels from Marmara Sea are mostly lower than those of the regions in other areas of the world. It was concluded that Marmara Sea has a potential of being a safe source of mussels if industrial inputs somewhat reduced and controlled. Concentrations of heavy metals in mussels must be monitored comprehensively and periodically with respect to the consumer health.  相似文献

Arctic environments are commonly considered to be relatively pristine because of minimal local human activity. However, these areas receive air pollution from lower latitude regions. Our goal was to determine concentrations of metals (Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, and Zn) in dominant species of vascular plants, mosses, lichens, algae, and in the biological soil crust (BSC), and topsoil (0–3 cm) from various types of tundra in the southwestern part of Spitsbergen, Norway. Results indicate that mosses are more efficient bioaccumulators of Cd, Co, Cr, Cu, Fe, Mn, and Zn than lichens. The highest levels of Co, Cr, Cu, Fe, Hg, Mn, Ni, and Pb were found in the BSC, and the moss species Racomitrium lanuginosum, Sanionia uncinata, and Straminergon stramineum from the polygonal tundra, initial cyanobacteria-moss wet tundra, snow bed cyanobacteria-moss tundra, and flow water moss tundra alimented by melting ice or snow. The observed higher concentrations of Cu and lower concentrations of Hg in mosses, lichens, and vascular plants compared with values observed 20 years earlier were apparently associated with changes in the atmospheric deposition of contaminants over Spitsbergen due to changes in the long-distance transport of anthropogenic emissions from industrialized areas. Prasiola crispa and Salix polaris may be useful bioindicators of Cd and Zn, and the BSC, R. lanuginosum, S. uncinata, and S. stramineum as bioindicators of Co, Cr, Cu, Fe, Hg, Mn, Ni, and Pb. These results may be extrapolated across other areas of Spitsbergen with similar climates.  相似文献

Various mechanisms are involved in detoxification of heavy metals such as lead (Pb) in plant cells. Most of the Pb taken up by plants accumulates in their roots. However, the detailed properties of Pb complexes in roots remain unclear. We have investigated the properties of Pb deposits in root cell walls of radish (Raphanus sativus L.) seedlings grown on glass beads bed containing Pb pellets, which are the source of Pb-contamination in shooting range soils. Pb deposits were tightly bound to cell walls. Cell wall fragments containing about 50,000 ppm Pb were prepared from the roots. After extracting Pb from the cell wall fragments using HCl, Pb ions were recombined with the Pb-extracted cell wall fragments in a solution containing Pb acetate. When the cell wall fragments were treated with pectinase (E.C. 3.2.1.15) and were chemically modified with 1-ethyl-3-dimethylamino-propylcarboimide, the Pb-rebinding ability of the treated cell wall fragments decreased. When acid-treated cell wall fragments were incubated in a solution containing Pb2+ and excess amounts of a chelating agent, Pb recombined with the cell wall fragments were measured to estimate the affinity between Pb2+ and the cell wall fragments. Our data show that Pb2+ binds to carboxyl groups of cell walls. The source of the carboxyl groups is suggested to be pectic compounds. A stability constant of the Pb-cell wall complex was estimated to be about 108. The role of root cell walls in the mechanism underlying heavy metal tolerance was discussed.  相似文献

Gills cells of the freshwater mussel Lasmigona costata and the seawater clam Mesodesma mactroides were isolated (mussel: chemical dissociation; clam: mechanical dissociation) and fractionated (Percoll gradient) into Fractions I and II. Mitochondrial dyes (DASPEI: mussel; MitoTracker®: clam) and Na+, K+-ATPase activity measurement were used to distinguish between cells of Fractions I and II. For mussel and clam, 80.5 ± 1.5 and 48.3 ± 3.2 % of cells were in Fraction II, respectively. For both species, cells of Fraction II had higher fluorescence emission and higher enzyme activity than those of Fraction I, being characterized as ‘cells rich in mitochondria’. Cells of Fraction II were kept in saline solutions approximating the ionic composition of hemolymph either under control conditions (no Cu addition) or exposed (3 h) to copper (Cu: 5, 9 and 20 μg Cu/L). Cell viability and Cu and Na+ content were measured. For both species, Cu content was higher and Na+ content was lower in cells exposed to 20 μg Cu/L. Furthermore, a strong negative correlation was observed between cell Na+ and Cu content in the two bivalve species, indicating a possible competition between Cu and Na+ for ion-transporting mechanisms or binding sites at gill cells of Fraction II. Considering that Cu is an ionoregulatory toxicant in aquatic invertebrates, these preliminary toxicological data support the idea of using isolated gill cells rich in mitochondria to study the mechanisms underlying the acute toxicity of waterborne Cu in freshwater and marine bivalves.  相似文献

A study quantifying the effects of different copper (Cu) concentrations (50, 200, 800 and 1,000 mg kg?1 Cu) on Cu bioaccumulation and physiological responses of Spartina alterniflora was conducted. Plant biomass and Cu accumulation were determined. Plant height, tiller number, chlorophyll, leaf electrolyte leakage rate (ELR), malondialdehyde (MDA), proline, soluble sugar, and organic acids were also measured. The results showed that S. alterniflora mainly accumulated Cu in fine roots. No significant changes of biomass of fine roots were detected except for obvious reduction under 1,000 mg kg?1 Cu. In leaves, rhizomes and fine roots, the highest Cu accumulations were detected under 800 mg kg?1 Cu. The highest Cu accumulation in stem was revealed under 200 mg kg?1 Cu. Plant height decreased under 1,000 mg kg?1 Cu; chlorophyll content reduced under >50 mg kg?1 Cu; levels of ELR and MDA increased under >200 mg kg?1 Cu. However, osmotic components such as proline and soluble sugar were accumulated to cope with higher Cu stresses (800 and 1,000 mg kg?1). Further, oxalic and citric acids were positively related with Cu contents in leaves and stems, suggesting that oxalic and citric acids may be related to Cu detoxification in aboveground parts of S. alterniflora. However, in above and belowground parts, no detoxification function of ascorbic and fumaric acids was observed due to unchanged or decreased trend under Cu stress.  相似文献

Cyanobacterial toxins have been regarded by some researchers as allelopathic substances that could modulate the growth of competitors. Nevertheless, often the concentrations of toxins used are too high to be considered ecologically relevant. In this work we tested the hypothesis that microcystin-LR (MC-LR) and cylindrospermopsin (CYN) at ecologically relevant concentrations have no allelopathic effects on some species of phytoplankton. Extracts containing the toxins as well as pure MC-LR and CYN toxins were used to assess their effects on the growth rates of Nannochloropsis sp., Chlamydomonas reinhardtii, and Chlorella vulgaris. Cyanobacterial crude extracts induced more pronounced effects on growth rates than pure toxins. Microcystis aeruginosa and Aphanizomenon ovalisporum crude extracts containing MC-LR and CYN at 0.025–2.5 mg l?1 stimulated growth rates of microalgae, whereas A. ovalisporum crude extracts containing 2.5 mg l?1 of CYN strongly inhibited growth rates of microalgae after 4 and 7 days of exposure. MC-LR and CYN at environmentally occurring concentrations were unable to affect negatively the growth of microalgae, and therefore these molecules may play roles other than allelopathy in natural ecosystems.  相似文献

The effects of different concentrations of lead nitrate on root, hypocotyl and shoot growth of Indian mustard (Brassica juncea var. Megarrhiza), and the uptake and accumulation of Pb2+ by its roots, hypocotyls and shoots were investigated in the present study. The concentrations of lead nitrate (Pb(NO3)2) used were in the range of 10−5–10−3 M. Root growth decreased progressively with increasing concentration of Pb2+ in solutions. The seedlings exposed to 10−3 M Pb exhibited substantial growth reduction and produced chlorosis. Brassica juncea has considerable ability to remove Pb from solutions and accumulate it. The Pb content in roots of B. juncea increased with increasing solution concentration of Pb2+. The amount of Pb in roots of plants treated with 10−4, 10−3 and 10−5 M Pb2+ were 184-, 37- and 6-fold, respectively, greater than that of roots of the control plant. However, the plants transported and concentrated only a small amount of Pb in their hypocotyls and shoots, except for the group treated with 10−3 M Pb2+.  相似文献

Herein, we evaluate the binding of Pb(II) and Bi(III) to cysteine-substituted versions of the TRI peptides [AcG-(LKALEEK)4G-NH2] which have previously been shown to bind Hg(II) and Cd(II) in unusual geometries as compared with small-molecule thiol ligands in aqueous solutions. Studies of Pb(II) and Bi(III) with the peptides give rise to complexes consistent with the metal ions bound to three sulfur atoms with M–S distances of 2.63 and 2.54 Å, respectively. Competition experiments between the metal ions Pb(II), Cd(II), Hg(II) and Bi(III) for the peptides show that Hg(II) has the highest affinity, owing to the initial formation of the extremely strong HgS2 bond. Cd(II) and Pb(II) have comparable binding affinities at pH > 8, while Bi(III) displays the weakest affinity, following the model, M(II) + (TRI LXC)3 3? → M(II)(TRI LXC)3 ?. While the relevant equilibria for Hg(II) binding to the TRI peptides corresponds to a strong first step forming Hg(TRI LXC)2(HTRI LXC), followed by a single deprotonation to give Hg(TRI LXC)3 ?, the binding of Cd(II) and Pb(II) is consistent with initial formation of M(II)(TRI LXC)(HTRI LXC)2 + at pH < 5 followed by a two-proton dissociation step (pK a2) yielding M(II)(TRI LXC)3 ?. Pb(II)(TRI LXC)(HTRI LXC)2 + converts to Pb(II)(TRI LXC)3 ? at slightly lower pH values than the corresponding Cd(II)–peptide complexes. In addition, Pb(II) displays a lower pK a of binding to the “d”-substituted peptide, (TRI L12C, pK a2 = 12.0) compared with the “a”-substituted peptide, (TRI L16C, pK a2 = 12.6), the reverse of the order seen for Hg(II) and Cd(II). Pb(II) also showed a stronger binding affinity for TRI L12C (K bind = 3.2 × 107 M?1) compared with that with TRI L16C (K bind = 1.2 × 107 M?1) at pH > 8.  相似文献

Due to the severe hazardous influences of lead (Pb2+) on iron-related diseases, the effects of Pb2+ on serum parameters associated with iron metabolism have been studied in this project. Male Wistar rats weighing 200–250 g were treated with Pb2+ for the short and long period of times. The animals received daily intraperitoneal injection of 100 mg Pb2+ kg?1 body weight (BW) for 5 days and 4 mg?kg?1 BW of Pb2+ for 30 and 45 days, respectively. The results show that when animals were treated with both low and high concentrations of Pb2+, serum iron concentration decreased markedly, by 23.2, 32.8, and 39.9 %, while the sera TIBC and transferrin concentrations increased significantly (p?<?0.05). Following short- and long-term exposures to Pb2+, the percentage of serum transferrin saturation was also decreased in comparison with the untreated control group (p?<?0.05). Concentrations of serum copper and ceruloplasmin following Pb2+ treatments also reduced significantly (p?<?0.05). The percentage of hematocrit and hemoglobin levels was reduced (p?<?0.05) in all Pb2+-treated animals in comparison with the controls. These results suggest that Pb2+ changes the serum parameters related to iron metabolism, which may play an important role in producing iron-related diseases.  相似文献

The concentrations of metals, viz., Fe, Mn, Cu, Zn, Cd, Pb, and Ni, were investigated in brown algae of the genus Sargassum collected in the coastal waters of Nha Trang Bay (South China Sea). Metal concentrations in algae growing in the zone of influence of the city of Nha Trang were higher than those in macrophytes collected to the south and north of the city. Similar levels of Cu and Fe concentrations were observed in macrophytes from the urbanized coastal areas of Nha Trang Bay and in algae from the Peter the Great Bay (Sea of Japan). This is suggestive of comparable metal inputs from the anthropogenic sources of the cities of Nha Trang and Vladivostok (Peter the Great Bay). However, the concentrations of Pb, Ni, and Cd in algae from the coastal waters of Nha Trang Bay were lower than in macrophytes from the inshore zone of Vladivostok. The high level of pollution of sea water with Zn compounds near Nha Trang was local. The metal levels in macrophytes from background areas reflect the geochemical features of the environment. Sargassum algae from Nha Trang Bay had lower concentrations of Zn, Cd, and Ni and higher concentrations of Mn than algae from Peter the Great Bay. The background concentrations of Cu, Pb, and Fe in macrophytes from both bays were virtually the same.

The concentrations of Pb and Cd, and trace elements (Cu and Zn) in the urban topsoil, rook (Corvus frugilegus) feces and feathers and human scalp hair were analyzed to examine the potential ecological risk posed by Pb and Cd on local residents of Qiqihar City, northeastern China. Results revealed that the Cd concentrations in the topsoil were ranged from 0.14 to 3.55 mg kg?1 dry weight (dw). The maximal geoaccumulation indices [a value from logarithmic (a measured metal content/1.5 × background content of the metal in this region), introduced by Muller] of Cd exceeded 3.5, which suggested that this region was seriously contaminated by Cd. The corresponding average detectable concentrations in C. frugilegus feathers and feces were 1.38 and 3.97 mg kg?1 dw for Pb and 1.04 and 0.69 mg kg?1 dw for Cd. High Pb and Cd concentrations, respectively, ranging from 7.46 to 24.9 mg kg?1 dw and from 0.35 to 0.92 mg kg?1 dw were also detected in the human scalp hair samples. These high Pb and Cd concentrations in C. frugilegus and local people were possibly associated with local industrial wastes and vehicle exhausts. The external tissues (feces and feather) of the rook species can be considered as an indicator of potential Cd toxic risk in this species; however, the human scalp hair is not a reliable biomarker for risk of Pb and Cd in the human being. Effective measures should be established to reduce the inputs of Pb and Cd into the urban environment and to protect the health of local people.  相似文献
