猪肺源肝素的分离纯化及其成分鉴定   总被引:1,自引:0,他引:1  
本文报告了从猪肺中用改进后的碱性氯化钠盐解法萃取的粗肝素为原料,以新型分离材料DEAE-Sephacel分离纯化,并与Sephadex G-50进行比较,前者使粗肝素比活由13.9USP/mg提高到203.64USP/mg(美国药典单位),纯化系数达到14.65,回收率达86.93%,而通过Sephadex G-50分离纯化后的肺肝素其比活性提高到102.10UPS/mg,纯化系数为7.35,回收  相似文献

本文报道用抗PAI-1单克隆抗体(McAb)亲和层析建立了纯化PAI-1的简便方法。经免疫亲和层析,SephacrylS200凝胶过滤,从HepG2细胞培液中分离到糖基化和非糖基化两种形式的PAI-1,回收率为84%,PAI-1比活性6.1×104IU/mg。糖基化PAI-1分子量为50kD,比活性5.8×104IU/mg。非糖基化PAI-1分子量43kD,占总PAI-130%,仍具有PAI-1活性。用ConA-Sepharose亲和层析进一步纯化得到SDS-PAGE纯的糖基化PAI-1。  相似文献

经硫酸铵沉淀、DEAE-Sephacel层析和Sephadex G-200凝胶过滤,将韭菜线粒体SOD纯化到均一程度。从6000g韭菜叶片线粒体中纯化得到2.5mg酶,酶比活力达1200U/mg蛋白。该酶对KCN和H2O2都不敏感,热稳定性弱 外光区吸收峰在280nm,凝胶过滤法测得其分子量为8200D,SOS-PAGE法测定其亚基分子量的22000D,DNS法测得其N-末端氨基酸为缬氨酸。上述结  相似文献

用正常人胎肺细胞体外培养,从其培养液中分离纤溶酶原活化物(PA),通过CM-SephadexC-50层析,硫酸铵沉淀和Fibrin-Sepharose,Lysine-Sepharose亲和层析及SephadexG-50凝胶过滤等步骤,从10.5l条件培养液中分离纯化得到两种类型的纤溶酶原活化物,t-PA90μg,u-PA800μg.在还原条件下SDS-PAGE均显示单带,分子量t-PA为72kD,u-PA为54kD,纤溶比活分别为156000IU/mg蛋白和106000IU/mg蛋白.  相似文献

经DEAE-Sephacel、UltrogelAcA 34、Heptylam ine-Sepharose 和Bio Scale Q2等四步层析从牛脑皮层膜中同时纯化出抑制型G 蛋白(inhibitory G protein, Gi)和O型G蛋白(other G pro-tein, Go).对Go 的选择性激活使Gi 和Go 的分离大为简化,并使二者的大量制备成为可能.纯化的G 蛋白的SDS-PAGE银染显示单一蛋白带,其结合[35S]GTPγS的活性分别提高68倍和124倍.用圆二色光谱法(CD)测量了纯化的G蛋白的二级结构.  相似文献

从皖南尖吻蝮蛇(Agkistrodonacutus)毒液中经DEAE-Sepharose和SephacrylS-200两步凝胶柱层析首次纯化出一种中分子量出血毒素(简称AaHⅣ).经SDS-PAGE和等电聚焦凝胶电泳测定其分子量为44kD,等电点为pH5.0.从500mg粗毒中可获得20mgAaHⅣ纯品.AaHⅣ有较强的出血活性,最小出血剂量(MHD)为0.4μg.  相似文献

以牛血球为材料,经溶血等处理和丙酮沉淀,获得牛血球超氧化物歧化酶粗品。此粗酶可以通过DEAE-Sepharose和CM-Sepharose快速柱层析,获得超氧化物歧化酶纯品。纯化的酶比活可达13500u/mg,经PAGE、SDS-PAGE和快速蛋白液相色谱(FPLC)检测,结果表明,纯化酶是均一的Sephadex G-100凝胶过滤测得该酶分子量为31,800,SDS-PAGE测得亚基分子量为15  相似文献

 本文以正常人胎盘为材料,通过匀浆、硫酸铵分级分离、20%正丁醇抽提、Sephadex G-200凝胶过滤及Concanavalin A-Sepharose 4B亲和层析等分离纯化步骤,制备获得了纯化3705倍的酸性β-1,4葡萄糖苷酶,其比活性和获得率分别为277261.33n mol·(mg Prot·h)~(-1)和14.3%。 经agarosn-IEF检验,该纯化的酸性β-1,4葡萄糖苷酶已达蛋白电泳单点纯,PI为5.2。经3~28%梯度PAGE,求知该酶单体分子量为74kD。该纯制酶极不稳定,4℃贮存6天后活性即降低50%以上,与其特异性抗体结合后,活性全部被抑制。  相似文献

The present paper reports a modified method for isolation of lysostaphin—a bacteriolytic agent with specific affinity for staphylococcal cell wall. The proposed purification scheme includes three steps. The first procedure is ultrafiltration through a membrane filter giving a yield of 75.6 %. The result of ultrafiltration is a concentrated, 10-times purified preparation of lysostaphin with specific activity 0.62 U/mg which can be used for digestion ofS. aureus cells. Further step, performed by ion-exchange chromatography on DEAE-cellulose, yields a 60-times purified preparation containing a mixture of enzyme components of lysostaphin. The yield of this step is 47.2 %, the preparation contains 3.54 U/mg protein. Using gel filtration on Sephadex G-50 a component with hexosaminidase activity was separated from the endopeptidase component on the basis of molar mass difference. A 270-times purified preparation of lysostaphin-endopeptidase with minimum of contaminating substances was obtained in this step. The yield of gel filtration was 22.1 %, specific activity increased up to 16.3 U/mg protein.  相似文献

The polygalacturonase (PG) isolated fron the pericarp of fully ripe tomato fruits was purified by (NH4)2SO4 precipitation, carboxyme-thylsepharose ion-exchang column and sephadex G-75 gel filtration. Specific activity of purified PG was 1.5 μmol galacturonic acid mg-1 protein min-1, which was 30 times as high as that of the crude extract with 1.7mol NaC1. When the elution separated by second sephadex G-75 column was analyzed by SDS-PAGE, only a single protein band was detected. It was shown by heat and pH experiments of the purified enzyme that the enzyme activity retained 50%, after treatment with heat at 50℃ for 10 min, and that the optimal pH was 4.6.  相似文献

首次采用新技术双水相萃取方法作为鸭血清胆碱酯酶(EC.3.1.1.8 CHE) 纯化的第一步,后经 DEAE-Sephadex A50,sephadex G200 柱层析,获得电泳纯鸭血清胆碱酯酶,提纯倍数1018倍,酶活力回收43.4%,比活274.9U/mg。鸭血清胆碱酯酶性质研究表明:此酶是糖蛋白和酸性蛋白水解酶,等电点 4.2 左右,最适 pH7.5 左右;对底物碘化硫代丁酰胆碱的 Km=9.8×10-5mol/L;SDS-PAGE 电泳和聚丙烯酰胺梯度电泳表明,鸭血清胆碱酯酶以相同亚基组成的不同聚合体形式存在,亚基分子量 78000,具有完整的酶活性.不同聚合体带电状态相同.  相似文献

E. coli DH 5 alpha cells harboring a plasmid pWR 590-BCA 4 for fused human proinsulin production were cultured. The fused human proinsulin was isolated from the fermented cells and then subjected it to cleavage with BrCN. The cleaved product was then converted to crude proinsulin-S-sulfonate using oxidative sulfitolysis. The isolation of human proinsulin-S-sulfonate was accomplished by ion exchange chromatography on QAE-sephadex A-25, followed by gel filtration on sephadex G-50. The purified human proinsulin-S-sulfonate was folded using a disulfide interchange method. The folding mixture was then chromatographed on sephadex G-50 and purified proinsulin was obtained. The proinsulin was then converted to human insulin and C-peptide by a combination cleavage with trypsin and carboxypeptidase B. The total yield of human insulin was about 5 mg/L The Zinc insulin crystals were obtained with amorphous human insulin using citrate method. The amino acid composition N-terminal sequences as well as C-terminal amino acid residues are in agreement with expected results. The hypoglycemic activity of purified human insulin is 26-27 U/mg, as judged by mouse convulsion assay, and the RIA activity is about 99% of that of porcine insulin.  相似文献

A microorganism hydrolyzing carboxymethyl cellulose was isolated from a paddy field and identified as Bacillus sp. Production of cellulase by this bacterium was found to be optimal at pH 6.5, 37 degrees C and 150 rpm of shaking. This cellulase was purified to homogeneity by the combination of ammonium sulphate precipitation, DEAE cellulose, and sephadex G-75 gel filtration chromatography. The cellulase was purified up to 14.5 fold and had a specific activity of 246 U/mg protein. The enzyme was a monomeric cellulase with a relative molecular mass of 58 kDa, as determined by SDS-PAGE. The enzyme exhibited its optimal activity at 50 degrees C and pH 6.0. The enzyme was stable in the pH range of 5.0 to 7.0 and its stability was maintained for 30 min at 50 degrees C and its activity got inhibited by Hg2+, Cu2+, Zn2+, Mg2+, Na2+, and Ca2+.  相似文献

本文介绍了一种简便易行的纯化组织型纤溶酶原激活剂(t-PA)的方法。新鲜猪心组织经丙酮脱脂脱水,制成干粉,再经醋酸钾缓冲液提取。提取液经阳离子交换柱、肝素柱亲和层析及凝胶柱层析分离纯化。经SDS凝胶电泳分析证明纯化的t-PA只在分子量为67000道尔顿位置出现一条染色带。同时把该蛋白带转移到纤维蛋白板上,也在同一位置出现溶解带。  相似文献

乳猪心房组织提取液经分子膜截留,去除大分子,小分子部分经SePhPdexG-25凝胶分离,SP离子交换层析,RP-HPLCC18柱进一步纯化,获得了均一的心钠素,活性鉴定表明它有较高的利尿、利钠和降压活性。  相似文献
