Testes from rats, mice and hamsters were incubated for 4 h with 0, 3.125 or 12.5 mIU hCG/ml. The LH receptor concentration in incubated testes of rats and mice was higher than that observed in hamsters. Testosterone levels in incubation media were significantly different among species (mice greater than rats greater than hamsters). During the incubation, hCG caused an increase in testosterone levels in all three species, but produced no significant changes in LH receptor concentration. Furthermore, a correlation between LH receptor concentration and testosterone only in hamsters is observed. The efficiency of the LH receptor-steroidogenesis interaction was estimated from the ratio of testosterone levels to receptor concentration under basal conditions and was found to differ among species (mice greater than hamster greater than rats). The levels of PGE and PGF in incubation media were higher in mice than in rats or hamsters, and hCG did not alter prostaglandin levels in any of the species. The present results indicate that acute in vitro hCG stimulation of testosterone synthesis does not involve appreciable changes in testicular LH receptor levels.  相似文献

Our previous study demonstrated that there are species differences among vertebrates in their conversion of 7alpha-hydroxycholesterol (7HC) to 7-ketocholesterol (7KC). To examine this further, we investigated the differences in the products of 7alpha-hydroxycholesterol in various species of male muroid rodents. Adult male Syrian hamsters (Mesocricetus auratus), dwarf hamsters (Phodopus rovolovskii), Djungarian hamsters (Phodopus sungorus), Chinese hamsters (Cricetulus griseus), rat-like hamsters (Tscherskia triton), and hispid cotton rats (Sigmodon hispidus) were used. Microsomal fractions were prepared from their livers, and the activities of the enzymes that participate in the dehydrogenation of 7alpha-hydroxycholesterol were determined by measuring the products using high-performance liquid chromatography. 7alpha-hydroxycholesterol was converted to both 7alpha-hydroxy-4-cholesten-3-one (7HCO) and 7-ketocholesterol in all of the hamsters tested. However, in the rat-like hamster and the hispid cotton rat, 7alpha-hydroxycholesterol was converted mostly to 7alpha-hydroxy-4-cholesten-3-one, as also observed in the rat (Rattus norvegicus). The results suggest that microsomal enzyme activity in the conversion of 7alpha-hydroxycholesterol to 7-ketocholesterol varies considerably, even within the subfamily Cricetinae and the family Muridae.  相似文献

Thirty men treated in childhood with cyclophosphamide for a mean of 280 days were assessed at a mean of 12.8 years after treatment for hormone concentrations and spermatogenesis. Four were azoospermic, nine oligospermic, and 17 normospermic. There was a significant inverse correlation of sperm density with cyclophosphamide dosage and duration of treatment. After a further mean follow up of 7.2 years three patients who were previously oligospermic and one who was azoospermic had normal sperm counts. All patients had normal sexual characteristics and libido. Serum androgen and prolactin concentrations did not differ significantly between patients and controls. Raised basal and stimulated follicle stimulating hormone concentrations were in keeping with impaired spermatogenesis. All patients had significantly raised luteinising hormone responses on stimulation with luteinising hormone releasing hormone. The results suggest compensated Leydig cell failure, and patients with this condition require long term evaluation of testicular function. Potential recovery of spermatogenesis with time requires appropriate counselling and contraceptive advice.  相似文献

The nucleotide sequence of the mip genes and their inferred amino acid sequences were determined from 35 Legionella species and compared with the published sequences for L . pneumophila , L . micdadei and L . longbeachae . The sequences were 69–97% conserved at the nucleotide level and 82–99% at the amino acid level, with total conservation of amino acids determined to be associated with sites known to be involved in peptidyl prolyl cis–trans isomerase activity. No apparent difference could be determined in the arrangement of amino acids that would predict a functional difference in Mip from species associated with disease and Mip from species isolated only from the environment. Additionally, a phylogenetic comparison of the sequences with published 16S RNA sequences, using both genetic distance and maximum parsimony methods, was performed. Few relationships were apparent that were well supported by both data sets, the most robust being a clade comprising {[( cincinnatiensis , longbeachae , sainthelensi , santicrucis ) gratiana ] ( moravica , quateirensis , shakespearei , worsleiensis ) anisa , bozemanii , cherrii , dumoffii , gormanii , jordanis , parisiensis , pneumophila , steigerwaltii , tucsonensis , and wadsworthii  }.  相似文献

Domestication of animals has resulted in phenotypic changes by means of natural and human-directed selection. Body composition is important for farm animals because it reflects the status of energy reserves. Thus, there is the possibility that farm animals as providers of food have been more affected by human-directed selection for body composition than laboratory animals. In this study, an analysis was conducted to determine what similarities and differences in body composition occur between farm and laboratory animals using literature data obtained from seven comparative slaughter studies (n = 136 observations). Farm animals from four species (cattle, goats, pigs and sheep) were all castrated males, whereas laboratory animals from three species (dogs, mice and rats) comprised males and/or females. All animals were fed ad libitum. The allometric equation, Y = aXb, was used to determine the influence of species on the accretion rates of chemical components (Y, kg) relative to the growth of the empty body, fat-free empty body or protein weights (X, kg). There were differences between farm and laboratory animals in terms of the allometric growth coefficients for chemical components relative to the empty BW and fat-free empty BW (P < 0.01); farm animals had more rapid accretion rates of fat (P < 0.01) but laboratory animals had more rapid accretion rates of protein, water and ash (P < 0.01). In contrast, there was no difference in terms of the allometric growth coefficients for protein and water within farm animals (P > 0.2). The allometric growth coefficients for ash weight relative to protein weight for six species except sheep were not different from a value of 1 (P > 0.1), whereas that of sheep was smaller than 1 (P < 0.01). When compared at the same fat content of the empty body, the rate of change in water content (%) per unit change in fat content (%) was not different (P > 0.05) across farm animal species and similar ash-to-protein ratios were obtained except for dogs. The fraction of empty body energy gain retained as fat increased in a curvilinear manner, and there was little variation among farm animals at the same fat content of the empty body. These findings may provide the opportunity to develop a general model to predict empty body composition across farm animal species. In contrast, there were considerable differences of chemical body composition between farm and laboratory animals.  相似文献

Using a highly sensitive and specific gas chromatography-mass spectrometric assay, the glycol metabolites of norepinephrine (NE), 3,4-dihydroxyphenylethyleneglycol (DHPG) and 3-methoxy-4-hydroxyphenylethyleneglycol (MHPG) were determined simultaneously in brain and body fluids of several mammalian species, including humans. Highest molar ratios of DHPG to MHPG were found in rat brain (1.20), a species in which these glycol metabolites were primarily conjugated. In mouse, guinea pig, hamster, monkey, and human brain, DHPG and MHPG were mostly unconjugated, and DHPG concentrations were about 30–60% of the respective MHPG levels. In dog cortex, MHPG occurred predominantly as conjugates, whereas DHPG could only be detected in its unconjugated form. In all species studies, highest DHPG and MHPG concentrations occurred in hypothalamus followed, in general, by midbrain and brainstem whereas cerebral cortex, caudate and cerebellum had the lowest values. These results demonstrate substantial differences in the degree of conjugation and relative abundance of brain DHPG compared to MHPG between the rat and other animal species studied.  相似文献

Response of the cryptorchid testis to gonadotrophic stimulation was assessed by comparison of the androgen production capability in vivo and in vitro with that of the normal scrotal testis. Serum androgen concentrations in cryptorchid rats were similar to those in normal rats, and the incremental increase 60 min after 50 i.u. hCG (i.v.) was about 7-fold for both groups. Basal and hCG-stimulated androgen production in vitro was higher for abdominal testes (557 and 3286 ng/pair) than for scrotal tests (157 and 504 ng/pair). Specific binding of hCG by testicular homogenates was slightly higher (P < 0.05) for cryptorchid testes when expressed per unit weight, but Scatchard analysis indicated that although hCG binding affinities did not differ (Ka = 2 x 10(10) M-1), hCG binding capacity of cryptorchid testes was only 75 ng, compared to 219 ng for scrotal testes. These data indicate that a discrepancy exists between androgen production in vivo and in vitro by cryptorchid testes and that normal serum androgen concentrations are maintained in the presence of decreased numbers of testicular LH/hCG receptors.  相似文献

Three solution experiments were performed to test the importance of NH 4 + versus NO 3 - +NH 4 + to growth of 23 wild-forest and open-land species, using field-relevant soil solution concentrations at pH 4.5. At N concentrations of 1–200 M growth increased with increasing N supply in Carex pilulifera, Deschampsia flexuosa, Elymus caninus and Bromus benekenii. Geum urbanum was the most N demanding species and had little growth below 200 M. The preference for NH 4 + or NO 3 - +NH 4 + was tested also at pH 4.0; no antagonism was found between NH 4 + and H+, as indicated by similar relative growth in both of the N treatments at both pH levels. Growth in solution with NH 4 + relative to NO 3 - +NH 4 + , 200 M, was negatively related to the mean pH of the field occurrence of the species tested; acid-tolerant species grew equally well with only NH 4 + as with NO 3 - +NH 4 + (Oxalis acetosella, Carex pilulifera, Festuca gigantea, Poa nemoralis, Deschampsia flexuosa, Stellaria holostea, Rumex acetosella), while species of less acid soils were favoured by NO 3 - +NH 4 + (Urtica dioica, Ficaria verna, Melandrium rubrum, Aegopodium podagraria, Geum urbanum, Bromus benekenii, Sanguisorba minor, Melica ciliata, Silene rupestris, Viscaria vulgaris, Plantago lanceolata). Intermediate species were Convallaria majalis, Elymus caninus, Hordelymus europaeus and Milium effusum. No antagonism between NH 4 + and Ca2+, Mg2+ and K+ was indicated by the total uptake of the elements during the experiment.  相似文献

It is known that among many animal species, including laboratory mice, a short-time exposition to a female causes activation of the pituitary-testicular axis in males, which then rapidly decreases. Effects of a prolonged female exposition on the testicular testosterone output in response to hCG injections were investigated in adult males of two inbred mice strains CBA/Lac and PT. The males of both genotypes kept with females for 5 days were injected subcutaneously with 10 IU of hCG 120 minutes before decapitation. Males of the same genotype and similar age keeping alone were served as control. The serum testosterone concentration and its testicular content were measured with immune-enzyme assay. It has been shown that the hCG increased the testosterone concentration and its testicular content in control males of both strains, but the testosterone output was expressed more significantly in PT males in comparison with CBA/Lac. The prolonged exposition of a female itself did not influence the testosterone output in males of both strains. However, it increased significantly the testicular response to hCG in PT strain. The data obtained suggest that a long-term exposition of a female could reinforce the testicular reactivity to hCG in genotype-dependent manner.  相似文献

Single, functionally isolated motor units were studied in the medial gastrocnemius (MG) muscle of cats and rats. Axons of their motoneurons were stimulated with trains of pulses at frequencies increasing from 1 to 150 Hz and forces developed by muscle fibers were measured and force-frequency curves were compared between species. The following observations were made: (1) the most steep parts of curves (related to unfused tetani of motor units) begun at lower frequencies of stimulations in all types of feline motor units, (2) for fast motor units, the same relative values of force of unfused tetani were achieved at significantly lower frequencies of stimulations in the cat than in the rat. Twitch time parameters of both species influenced the course of force-frequency curves. It was showed that the contraction times of feline units varied in the wide range (21-81 ms), and these units reached 60% of the maximum force at stimulation frequencies between 10 and 38 Hz. On the other hand, contraction times of rat units ranged from 10 to 34 ms, whereas stimulation frequencies necessary to reach 60% of the maximum force varied considerably, from 12 to 65 Hz. The correlations between the above parameters were found for motor units of each species. However, the regression lines drown for the collected population of cat and rat units did not form linear continuity. Thus it seems that interspecies differences in the twitch contraction times do not fully explain different force-frequency relationships in mammalian skeletal muscles.  相似文献

This study assessed the thermal sensitivity of mitochondrial respiration in the small crustacean Daphnia pulex. More specifically, we wanted to determine if clones that inhabit different latitudes and habitats showed differences in the thermal sensitivity of their mitochondrial function. The experimental design included two clones from temperate environments (Fence from Ontario and Hawrelak from Alberta) and two clones from subarctic environments (A24 from Manitoba and K154 from Quebec). The integrated mitochondrial function was measured with high-resolution respirometry following whole-animal permeabilization. Mitochondrial respiration was performed under six different temperatures (10, 15, 20, 25, 30, and 35 °C) in the clone Hawrelak and at two temperatures (10 and 20 °C) in the three other clones. In the clone Hawrelak, complexes I and II respiration showed higher sensitivity to temperature variation compared to complex IV respiration. Interestingly, the threshold plot showed no excess capacity of complex IV at 20 °C in this clone. The clones showed significant divergence in the ability to oxidize the complex I and complex IV substrates relative to the maximal oxidative phoshorylation capacity of mitochondria. More importantly, some of the clonal divergences were only detected under low assay temperatures, pointing toward the importance of this parameter in comparative studies. Future and more complex studies on clones from wider environmental gradients will help to resolve the link between mitochondrial function and adaptations of organisms to particular conditions, principally temperature.  相似文献

The putative role of the endocannabinoid system and the effects of cannabis use in male and female sexual functioning are summarized. The influence of cannabis intake on sexual behavior and arousability appear to be dose-dependent in both men and women, although women are far more consistent in reporting facilitatory effects. Furthermore, evidence from nonhuman species indicate somewhat more beneficial than debilitating effects of cannabinoids on female sexual proceptivity and receptivity while suggesting predominantly detrimental effects on male sexual motivation and erectile functioning. Data from human and nonhuman species converge on the ephemeral nature of THC-induced testosterone decline. However, it is clear that cannabinoid-induced inhibition of male sexual behavior is independent of concurrent declines in testosterone levels. Investigations also reveal a suppression of gonadotropin release by cannabinoids across various species. Historical milestones and promising future directions in the area of cannabinoid and sexuality research are also outlined in this review.  相似文献
