The abundance and productivity of benthic microalgae in coral reef sediments are poorly known compared with other, more conspicuous (e.g. coral zooxanthellae, macroalgae) primary producers of coral reef habitats. A survey of the distribution, biomass, and productivity of benthic microalgae on a platform reef flat and in a cross-shelf transect in the southern Great Barrier Reef indicated that benthic microalgae are ubiquitous, abundant (up to 995.0 mg chlorophyll (chl) a m–2), and productive (up to 110 mg O2 m–2 h–1) components of the reef ecosystem. Concentrations of benthic microalgae, expressed as chlorophyll a per surface area, were approximately 100-fold greater than the integrated water column concentrations of microalgae throughout the region. Benthic microalgal biomass was greater on the shallow water platform reef than in the deeper waters of the cross-shelf transect. In both areas the benthic microalgal communities had a similar composition, dominated by pennate diatoms, dinoflagellates, and cyanobacteria. Benthic microalgal populations were potentially nutrient-limited, based on responses to nitrogen and phosphorus enrichments in short-term (7-day) microcosm experiments. Benthic microalgal productivity, measured by O2 evolution, indicated productive communities responsive to light and nutrient availability. The benthic microalgal concentrations observed (92–995 mg chl a m–2) were high relative to other reports, particularly compared with temperate regions. This abundance of productive plants in both reef and shelf sediments in the southern Great Barrier Reef suggests that benthic microalgae are key components of coral reef ecosystems.Communicated by Environmental Editor, B.C. Eutrophication of coastal waters often leads to excessive growth of microalgal epiphytes attached to seagrass leaves; however, the effect of increased nutrient levels on sediment microalgae has not been studied within seagrass communities. A slow‐release NPK Osmocote fertilizer was added to sediments within and outside beds of the shoal grass Halodule wrightii, in Big Lagoon, Perdido Key, Florida. Gross primary production (GPP) and biomass (HPLC photopigments) of sediment microalgae within and adjacent to fertilized and control H. wrightii beds were measured following two 4‐week enrichment periods during June and July 2004. There was no effect of position on sediment microalgal GPP or biomass in control and enriched plots. However, nutrient enrichment significantly increased GPP in both June and July. These results suggest that sediment microalgae could fill some of the void in primary production where seagrass beds disappear due to excessive nutrient enrichment. Sedimentary chl a (proxy of total microalgal biomass) significantly increased only during the June enrichment period, whereas fucoxanthin (proxy of total diatom biomass) was not increased by nutrient enrichment even though its concentration doubled in the enriched plots in June.  相似文献

Primary production of the microphytobenthic community and carbohydrates concentrations were studied in the lagoonal system of Grado and Marano, located in the Northern Adriatic coast. Sediment samples were collected along a salinity gradient. Abundance and species composition of the microphytobenthic communities were analysed and the benthic microalgal biomass was estimated as Chlorophyll a (Chl a). Primary production of benthic diatoms was estimated using 14C-tracer. Extracellular carbohydrates were extracted from the sediment and separated in two operationally defined fractions (colloidal and EDTA-extractable). Salinity was higher in the Grado lagoon, where the benthic microalgal community was mainly composed of marine diatoms. In the Marano lagoon, which has a lower salinity, freshwater species were also found. In both lagoons, photosynthetic efficiency showed an inverse relationship with salinity and a direct relationship with the main biological variables. Photosynthetic activity was directly related to Chl a and abundance of benthic microalgae, suggesting that in the benthic system microalgal community is responsible for primary production. Overall, salinity was also influent on the microphytobenthic primary production, which was greater in the more saline Grado lagoon.  相似文献

As third generation feedstock, microalgae are microorganisms that can grow only in the optimum conditions. There are parameters including the concentration of macro and microelements in nutrient solution, pH, temperature and light intensity that have significant impact on microalgal growth. In recent years, various sensing devices have been developed for sensitive measurement of these parameters during microalgal growth. In this study, a new potentiometric nitrate selective sensor was developed to indicate the nitrate uptake of microalgae and the effect of nitrate nutrient on microalgal growth, specifically, and this sensor was successfully applied to determine nitrate concentration in medium during microalgal growth. Moreover, the effects of nitrate, carbonate and phosphate concentration in the growth medium on biomass production of Chlorella minutissima were investigated by using Box–Behnken design method, and optimum conditions were determined for the highest biomass production of microalgae. As a result of the experiments, it was seen that the highest C. minutissima production was achieved using the medium consist of 2.63 g/L NaNO3, 0.35 g/L Na2CO3 and 0.4 g/L KH2PO4. Statistically, it was observed that there was a proportional relationship between the microalgae production and investigated parameters such as carbon, nitrogen and phosphate amounts of culture mediums. The electrode showed a wide linear range between 1.0 × 10−1 and 5.0 × 10−5 M with a detection limit of the 5 × 10−6 M and the response time was found as 10 s. The results showed that developed nitrate selective sensor could be successfully applied for continuous measurement of nitrate in microalgal productions at reduced cost.  相似文献

The use of microalgae in a number of sectors, including biodiesel, feed and food production, is proving to be of great interest. An evaluation was made of the possible biostimulant effects on Chlorella vulgaris and Scenedesmus quadricauda of humic-like substances (HLs) extracted from agro-industrial wastes. These included digestate from the waste of an agro-livestock farm (D-HL), oil extraction residues from rape (B-HL, Brassica napus L.) and tomato residues (T-HL). The microalgal growth medium (BG11) was supplemented with HLs to evaluate their effect on biomass yield as well as carbohydrate, chlorophylls a and b, lipid and fatty acid contents. Our results showed that the HLs used in the test are effective biostimulants of C. vulgaris and S. quadricauda. The biostimulant effect seems to depend on the type of extract used for cultivating the microalgae, the concentration and the species treated. Among the extracts applied to the growth medium, D-HL and T-HL seem to have a significant effect on microalgal biomass and lipid production. Although B-HL showed no significant effect on the biomass and lipid content of C. vulgaris and S. quadricauda, its presence in the growth medium increased the saturated:unsaturated fatty acid ratio (SFA/UFA) and stimulated the sugar metabolism of the microalgae by increasing their carbohydrate and chlorophyll content.  相似文献

Eau Galle Reservoir, Wisconsin, was treated with a hypolimnetic dose of aluminum sulfate (alum) in 1986 to diminish excessive phytoplankton production associated with high phosphorus loading from anoxic, profundal sediments. Prior to treatment, internal total phosphorus (TP) loading was 3 to 6 times greater than external TP loading during summer stratification. Periodic increases in epilimnetic TP mass and chlorophylla concentrations closely corresponded with elevated internal TP loading. For one year following treatment, internal TP loading and concentrations of soluble reactive phosphorus (SRP) in the hypolimnion were substantially reduced. However, abnormally high external TP loading during the stratified period of 1986 resulted in high TP mass and chlorophylla in the epilimnion. During the summers of 1987 and 1988, effects of alum treatment on internal TP loading were essentially negated, and epilimnetic TP mass and chlorophylla remained unchanged from pretreatment years. Multiple potential sources of P input to this reservoir make it difficult to reduce epilimnetic P and phytoplankton growth.  相似文献

The total ash, chlorophyll, phaeopigment, lipid and fatty acid contents of three microalgae (the diatomsChaetoceros calcitrans, Skeletonema costatum and the prymnesiophyteIsochrysis galbana) used in tropical Malaysian penaeid mariculture were studied. The axenic laboratory cultures were grown in f-2 medium, while outdoor cultures were grown in a commercial medium designed for optimum nutrition in tropical outdoor aquaculture operations. Considerable amounts (3–9% of the total fatty acid) of the polyunsaturated fatty acid 20:5w3 (eicosapentaenoic acid) were present in the diatoms; very small amounts were also found inI. galbana. Lipid contents were three to five times higher than chlorophylla. There was an increase with culture age in the relative proportion of total C18 and C20 fatty acid components. The algae contained the w3-polyunsaturated fatty acids (PUFA) necessary for the growth and survival rate of the prawn larvae. However, onlyChaetoceros calcitrans grew well under the conditions of temperature and salinity in the hatchery.  相似文献

In vivo chlorophyll (Chl) a fluorescence was measured in undisturbed intertidal sediments with the purpose of tracing the vertical migratory rhythms of benthic microalgae. A pulse amplitude fluorometer, an instrument which does not require physical contact with the sample, was used, thus allowing successive measurements to be taken on the same sample without causing any type of disturbance to the sediment structure. The basis of the method is the possibility to detect changes in the Chl a concentration near the sediment surface caused by the vertical movement of the microalgae. This requires the verification of two conditions: the possibility to follow changes in the sediment Chl a content from fluorescence intensity, and a sediment photic depth smaller than the vertical distances covered by the moving microalgae. Both conditions were experimentally verified in intertidal muddy sediments of the Tagus estuary, Portugal. In vivo fluorescence was shown to vary linearly with the sediment Chl a content, and the sediment photic depth was estimated to reach 0.27 mm, a value clearly smaller than the reported depths for microalgal migrations. Sediment samples kept under in situ conditions exhibited large hourly Variations (over 400%) in the Chl a fluorescence intensity, which were closely synchronized with the daytime periods of emersion. The rhythmic fluctuations in Chl a fluorescence were confirmed further to represent microalgal migration by (1) its endogenous nature (fluorescence continued to follow diurnal and tidal cycles after removal of environmental stimuli), (2) its dependence on the vertical distribution of the microalgal population within the sediment (vertically homogenized samples failed to display fluorescence variations), and (3) the lack of significant temperature and light effects on the fluorescence emission under in situ conditions (tested in three species representative of the main groups found in the studied microphytobenthic communities—the diatom Phaeodactylum tricornutum (Böhlin), the cyanobacterium Spirulina maxima (Setch. et Gard.), and the euglenophyte Euglena granulata (Klebs) Lemm.). The results obtained indicate that, in spite of the potential concurrent effects of factors other than the Chl a concentration on the fluorescence intensity, in vivo Chl a fluorescence can be used to trace nondestructively the migratory behavior of benthic microalgae.  相似文献

Over 15 months between January 1990 and March 1991, a range of physical, chemical and biological parameters was monitored regularly in fellfield soils of frost-sorted polygons at four sites on Signy Island (South Orkney Islands, maritime Antarctica). These included inorganic nutrients (orthophosphate, available nitrate and, over a more limited period, ammonia), chlorophyll a (as a proxy measure of microalgal biomass) and a range of potential cryoprotectant compounds. Transects across soil polygons revealed neither intrapolygon gradients in concentrations of inorganic nutrients or chlorophyll a nor significant interpolygon differences, in contrast with previous studies. Nitrate was present in much lower concentrations than phosphate, supporting evidence that it is a limiting nutrient in these fellfield ecosystems. Spring snowmelt, although a potential source of nutrient input, was not associated with increased concentrations of inorganic nutrients in the soil, probably through isolation of the soil from overlaying snow by a surface layer of ice. Soil microalgae at the study sites must survive winter temperatures of at least −9°C, even when protected beneath up to 1 m of snow, and it has been proposed that they accumulate sugars and polyols as cryoprotectants. In support of this, concentrations of erythritol, glycerol, glucose, sucrose and trehalose in the soil (both absolute quantity and after correction for chlorophyll a concentration) increased as winter proceeded, suggesting that changes in sugar concentrations were due to accumulation within individual cells.  相似文献

A microphytobenthic species composition of a tidal flat in the northern Wadden Sea was analysed regarding cell numbers and biomass (in carbon units). The three sampling sites differed in tidal inundation from 15 cm to about 90 cm water depth at high tide. The sediment was sandy at all three stations. A cluster analysis revealed a separation of the benthic diatoms into three areas: aNereis-Corophium-belt, a seagrass-bed and theArenicola-flat. Small epipsammic diatoms were most abundant and dominated the microalgal biomass. A microphytobenthic “spring bloom” even started beneath the ice cover of the flat in January. Lowest values of cell numbers and biomass of benthic microalgae were found in summer. Highest values were measured in the uppermost area (Nereis-Corophium-belt), and only here was an autumnal increase of benthic microalgae found. Further cluster analysis within each of the three areas revealed seasonal differences although the majority of species were present all year round. Many species were most abundant in spring, and some showed a bimodal distribution (spring-autumn) in the year of investigation.  相似文献

Zooplankton in the River Rhine was surveyed for five years at the Dutch sampling stations, Lobith (German/Dutch border) and Maassluis (at the point of discharge of the river into the North Sea). The zooplankton abundance showed an apparent seasonal pattern at both stations, characterized by low densities during the winter period, and higher densities during the summer period, with a spring peak. Zooplankton was dominated by rotifers at both stations, although during the winter periods the contribution of copepods was considerable. The rotifers were dominated byBrachionus angularis, B. calyciflorus, Keratella cochlearis andK. quadrata; the copepods by cyclopoid nauplii; the cladocerans by small-sized species mainly belonging toBosmina. At Maassluis the relative contribution of copepods was higher than at Lobith. Furthermore, the zooplankton at Maassluis included the speciesEurytemora affinis, characteristic for estuarine conditions. In spring, the rotifer density and water temperature and rotifer density and chlorophylla concentration were positively correlated. Furthermore, both rotifer density and chlorophylla were inversely correlated with discharge. The possible role of environmental factors (water temperature, chlorophyll content, discharge and biotic factors) controlling the river zooplankton dynamics is indicated.  相似文献

The temporal dynamics and spatial distribution of microphytobenthic chlorophyll-a in the layer 0–1 cm were determined in the Western Scheldt estuary over the period 1991–1992. Connections between the annually averaged benthic chlorophyll-a and station elevation and sediment composition (as a measure of the hydrodynamic energy caused by currents and waves) were also examined.Microphytobenthic chlorophyll-a showed one main peak in early summer and a smaller peak in autumn. The mean chlorophyll-a concentration of 113 mg Chl-a m–2 in the upper centimeter is of the same order of magnitude as in other estuarine areas. The average annual primary production of the microphytobenthos has been estimated at 136 g C m–2 y–1 The primary production of sediment inhabiting microalgae is at least 17% of the total primary production in the estuary.Considerable differences in annually averaged chlorophyll-a emerges between the stations. These differences are related mainly to the interaction between station elevation and clay content of the sediment.  相似文献

Microalgae have been shown as a potential bioresource for food, biofuel, and pharmaceutical products. During the growth phases with corresponding environmental conditions, microalgae accumulate different amounts of various metabolites. We quantified the neutral lipids accumulation and analyzed the swimming signatures (speed and trajectories) of the motile green alga, Dunaliella primolecta, during the lag–exponential–stationary growth cycle at different nutrient concentrations. We discovered significant changes in the neutral lipid content and swimming signatures of microalgae across growth phases. The timing of the maximum swimming speed coincided with the maximum neutral lipid content and both maxima occurred under nutrient stress at the stationary growth phase. Furthermore, the swimming trajectories suggested statistically significant changes in swimming modes at the stationary growth phase when the maximum intracellular neutral lipid content was observed. Our results provide the potential exploitation of microalgal swimming signatures as possible indicators of the cultivation conditions and the timing of microalgal harvest to maximize the lipid yield for biofuel production. The findings can also be implemented to explore the production of food and antibiotics from other microalgal metabolites with low energy costs.  相似文献

A rapid method for harvesting and immobilization of oleaginous microalgae using pellet-forming filamentous fungi was developed. The suitable conditions for pellet formation by filamentous fungi were determined. Among the strains tested, Trichoderma reesei QM 9414 showed superior pellet forming ability. Its pellets were used to harvest oleaginous microalga Scenedesmus sp. With increasing volume ratio of fungal pellets to microalgae culture up to 1:2, >94% of microalgal cells were rapidly harvested within 10 min. The ratio of fungal pellets could manipulate both harvesting time and initial concentration of microalgal cells in the pellets. The microalgae–fungal pellets were successfully used as immobilized cells for effective phytoremediation of secondary effluent from seafood processing plants under nonsterile condition. The chemical oxygen demand, total nitrogen, and total phosphorus removal were >74%, >44%, and >93%, respectively. The scanning electron microscopy showed that the microalgal cells were not only entrapped in the pellets but also got attached to the fungal hyphae with sticky exopolysaccharides, possibly secreted by the fungi. The extracted lipids from the pellets were mainly composed of C16–C18 (>83%) with their suitability as biodiesel feedstocks. This study has shown the promising strategy to rapidly harvest and immobilize microalgal cells and the possible application in phytoremediation of industrial effluent.  相似文献

The lipid characteristics of microalgae are known to differ between species and change with growth conditions. This work provides a methodology for lipid characterization that enables selection of the optimal strain, cultivation conditions, and processing pathway for commercial biodiesel production from microalgae. Two different microalgal species, Nannochloropsis sp. and Chlorella sp., were cultivated under both nitrogen replete and nitrogen depleted conditions. Lipids were extracted and fractionated into three major classes and quantified gravimetrically. The fatty acid profile of each fraction was analyzed using GC–MS. The resulting quantitative lipid data for each of the cultures is discussed in the context of biodiesel and omega‐3 production. This approach illustrates how the growth conditions greatly affect the distribution of fatty acid present in the major lipid classes and therefore the suitability of the lipid extracts for biodiesel and other secondary products. A continuous culture ofIsochrysis aff.galbana clone T.iso, used to feedPecten maximus larvae at IFREMER (Brest, France), was carried out in a chemostat at its optimum temperature for growth (26 °C). Changes in pigments, lipid class (neutral, glyco- and phospholipids) and degree of fatty acid unsaturation were studied at three different growth rates (0.33, 0.5, 1 d–1). As predicted by chemostat theory, a slow growth rate produced higher cell numbers and higher biomass per unit volume. These cells were low in chlorophylla and carotenoids, but rich in neutral lipids. In contrast, cultures with a fast growth rate yielded lower cell concentrations, buth higher chlorophylla, carotenoid and membrane lipid contents per cell. Changes in polyunsaturated fatty acid distribution were related to differences in algal growth rates. Neutral lipids contained mainly saturated and monounsaturated fatty acids (C18:19) at low growth rates whereas they were enriched in polyunsaturated fatty acids, especially C22:63, at high growth rates. Therefore, it is suggested that the growth rate in continuous cultures be controlled so as to adjust the relative proportions of polyunsaturated fatty acids in lipid classes of the diet meant for larval nutrition.Author for correspondence  相似文献

The Seteni Estuary is a small temporarily open/closed estuary (TOCE) in South Africa under the influence of agricultural practices. While the general significance of microalgae to estuarine production is widely recognised, the factors regulating microalgal biomass in these heterogeneous systems are less well understood, particularly when man-induced pressures are superimposed on natural variability. This study investigated microalgal responses to physico-chemical variability in the nutrient-enriched Seteni Estuary in 2008–2009. Microphytobenthic biomass ranged from 1.1 to 91.7 mg Chl a m-2, while phytoplankton biomass varied between 0.22 and 18 mg Chl a m-3. Despite the high dissolved inorganic nitrogen (DIN) concentrations recorded, salinity and rainfall were identified as the main environmental drivers, highlighting the importance of allochthonous inputs. While this system appears to function as a typical TOCE, the relatively low microalgal biomass recorded may be an artifact of several factors such as phosphorus limitation, herbicide treatment, etc., acting singly or together to depress biomass levels. However, should the balance be upset, the most probable result would be a dramatic increase in microalgal biomass, to the point where harmful algal blooms may ensue. The system therefore needs to be closely monitored to prevent further degradation.  相似文献

The distribution of phytoplankton biomass and primary production were studied during summer 1993 at 16 stations from 65 to 72°N off West Greenland, ranging more than 900 km. Hydrography, nutrients and chlorophyll a profiles revealed a significant change in structure from south to north. Nitrate was depleted in the euphotic zone at most stations except close to the ice edge (West Ice) or close to outflow from large glaciers. The vertical distribution of phosphate followed that of nitrate, but was never depleted. Despite two stations with relatively high surface concentrations, silica showed the same distribution as the other two nutrients. In the south, chlorophyll a concentration and primary production were lower than north of Disko Bay (69°N), associated with a well-mixed versus a salinity-generated stratification, respectively. In Vaigat, a high-production station was identified, (st. 910, 69°52′69N–51°30′61W) with a chlorophyll a concentration in the euphotic zone of >13 μg l−1 and an area primary production of 3.2 g C m−2 day−1. This is seldom encountered in arctic waters and was presumably due to nutrient-rich melt-water originating from the Iluliíssat Glacier. The overall primary production for the studied area was 67–3207 mg C m−2 day−1 (mean ± SD=341± 743 mg C m−2 day−1), which is within the range of the few results published for West Greenland and eastern Canadian Arctic waters. This study examines the swimming speed in amictic females of Brachionus plicatilis in laboratory cultures. Five different stages were examined: recently hatched females, juveniles, adult non-ovigerous females, ovigerous females with 1 attached egg and ovigerous females with 2 attached eggs. We tested the speed at two temperatures, 15 °C and 25 °C, and two feeding conditions, presence and absence of microalgal cells. An automated motion analysis system was used to measure speed which was then video recorded. Swimming speed (μm s−1) increased with increasing body size. There was a slight decrease in the speed of adult females as the number of attached eggs increased. Swimming activity was higher at 25 °C than at 15 °C and in the absence of food than if microalgae were present. Average values under the different experimental conditions ranged between 500 μm s−1 for the recently hatched and fed females and 1500 μm s−1 for the adult non-ovigerous females in the absence of microalgae. Mass-specific swimming speed decreased with body mass increase.  相似文献
