The maximum excretion rate of NH4 (39 nmol mg dry wt–1h–1) was directly measured for Daphnia pulex by measuringNH4 accumulation in bottles containing D. pulex and dense, satiatingsuspensions of heat-killed algae. Ammonium release rates inthe algal suspensions were compared to those of individual animalsremoved from the suspension and placed in flow cells. Ammoniumrelease rate, R (nmol mg dry wt–1 h–1). in the flowcell decreased very rapidly with time, t (min), after removalaccording to the relation R = 26 + 25e–0.16t. Ammoniumexcretion obtained by the flow cell method after extrapolationto time zero was not significantly different from that obtainedin the bottles. The considerable experiment-to-experiment variationin NH4 excretion was in large part correlated (r2 = 0.73) withthe feeding rate on the algae.  相似文献

The relationship between thef-ratio [NO3 uptake/(NO3+ NH4+) uptake] and ambient nitrate concentration was evaluatedfor eight data sets from coastal waters. The f-ratio increasedasymptotically with increase in nitrate concentration in mostdata sets. However, the rate at which f-ratio increased at lownitrate concentration (slope = m) and the maximum attained f-ratio(fmax) varied among regions; the initial slope varied most withvalues ranging in excess of an order of magnitude. The datawere analyzed in relation to environmental factors and methodologicalconsiderations known to influence the f-ratio. Ambient ammoniumconcentration was important in accounting for regional differencesin the f versus NO3 relationship. A further analysisof the data, relating f-ratio to the ratio of NO3/(NO3+ NH4+) concentrations yielded a much more regionally consistentand approximately linear relationship; slopes varied by lessthan a factor of two in the extreme cases. Inclusion of knownalternative (aside from NH4+) sources of reduced-N (e.g. urea)and correction for methodological/computational errors (isotopedilution) systematically reduce f-ratio estimates. Other factors,e.g. reduced-N uptake by microheterotrophs, may systematicallyincrease the f-ratio.  相似文献

Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. The rate of excretion of glycolate by the unicellular greenalga Ankistrodesmus braunii changes during its life cycle. Itis high in the main growth phase during the light period witha maximum 6 hr after the start of illumination, and low duringthe period of cell division in the dark. The glycolate excretion is stimulated by DSPD and HPMS, whilethe total 14CO2-fixation is inhibited by DSPD and enhanced byHPMS. Changes in the effects of DSPD and HPMS on glycolate excretionas well as on photosynthetic 14CO2-fixation during the courseof the algal life cycle were followed using the technique ofsynchronous culture. How far the change of glycolate excretion is due to a changeof glycolate oxidase activity during the life cycle and to achange of C2-supply from the carbon reduction cycle is discussed.The effect of DSPD on glycolate excretion suggests a participationof ferredoxin in the glycolate pathway. The rates of photosynthetic 14CO2 fixation by Chlorella vulgarisllh, grown under high CO2, were determined between 4 to 37°Cwith air containing from 300 to 13,000 ppm 14CO2. When the CO2level was increased, both the rate of photosynthesis and theoptimum temperature for maximum photosynthesis increased. Themaximum photosynthetic rate was reached at 12°C with 300ppm l4CO2. Among the photosynthetic products fromed at 300 ppm 14CO2, glycolatedecreased greatly when the temperature was raised from 20 to30°C. At 3,000 ppm 14CO2 an insignificant amount of glycolatewas formed at all temperatures, whereas 14C-incorporation intothe insoluble fraction, sucrose, and the lipid fraction wassignificantly higher than at 300 ppm 14CO2. The 14C in sucrosewas greatly increased and the radioactivity in the insolublefraction decreased when the temperature was raised from 28 to36°C. (Received April 8, 1980; )  相似文献

The cultivation of narrow-leafed lupins (Lupinus angustifoliusL.) increase rates of subsoil acidification, and this is thoughtto be partly related to their pattern of nutrient uptake andH+/OH- excretion. The main hypothesis of this study was thatH+ and OH- excretion is not distributed evenly over the entirelength of the root system but is limited to zones where excesscation or anion uptake occur. Seedlings of nodulated lupinswere grown in solution culture using vertically split pots thatallowed the upper and lower zones of the root system to be suppliedwith varying concentrations of K+ and NO-3. Net H+/OH- excretionwas equated to the addition of NaOH/HCl required to maintaina constant pH in the nutrient solution during a 4-d treatmentperiod and nutrient uptake was measured by depletion from solutionin each zone of the split pots. The excess of cation over anion uptake was positively correlatedwith H+ excretion in each rooting zone. In zones where K+ wassupplied at 1200 µM, cation uptake was dominated by K+and up to twice as much H+ was excreted than in zones whereK+ was absent. In zones where NO-3 was supplied at 750 µM,the anion/cation uptake was balanced, however H+ excretion continuedto occur in the zone. When NO-3 was supplied at 5000 µM,anion uptake exceeded cation uptake but there was no OH- excretion.Organic acid anions may be excreted by lupins to maintain theirinternal electroneutrality when anion uptake exceeds cationuptake. The CO2-, H2O- and 16O2/18O2 isotopic-gas exchange and the fluorescencequenching by attached leaves of the wild-type and of the phytochrome-deficienttomato aurea mutant was compared in relation to water stressand the photon fluence rate. The chlorophyll content of aurealeaves was reduced and the ultra-structure of the chloroplastswas altered. Nevertheless, the maximum rate of net CO2 uptakein air by the yellow-green leaves of the aurea mutant was similarto that by the dark-green wild-type leaves. However, less O2was produced by the leaves of the aurea mutant than by leavesof the wild-type. This result indicates a reduced rate of photosyntheticelectron flux in aurea mutant leaves. No difference in bothphotochemical and non-photochemical fluorescence quenching wasfound between wild-type and aurea mutant leaves. Water stresswas correlated with a reversible decrease in the rates of bothnet CO2 uptake and transpiration by wild-type and aurea mutantleaves. The rate of gross 16O2 evolution by both wild-type andaurea mutant leaves was fairly unaffected by water stress. Thisresult shows that in both wild-type and aurea leaves, the photochemicalprocesses are highly resistant to water stress. The rate ofgross 18O2 uptake by wild-type leaves increased during waterstress when the photon fluence rate was high. Under the sameconditions, the rate of gross 18O2 uptake by aurea mutant leavesremained unchanged. The physiological significane of this differencewith respect to the (presumed) importance of oxygen reductionin photoprotection is discussed. We investigatedthe relationship between voltage-operatedCa2+ channel current and thecorresponding intracellular Ca2+concentration([Ca2+]i)change (Ca2+ transient) in guineapig gastric myocytes. Fluorescence microspectroscopy was combined withconventional whole cell patch-clamp technique, and fura 2 (80 µM) wasadded to CsCl-rich pipette solution. Step depolarization to 0 mVinduced inward Ca2+ current(ICa) andconcomitantly raised[Ca2+]i.Both responses were suppressed by nicardipine, an L-typeCa2+ channel blocker, and thevoltage dependence of Ca2+transient was similar to the current-voltage relation ofICa. When pulseduration was increased by up to 900 ms, peakCa2+ transient increased andreached a steady state when stimulation was for longer. The calculatedfast Ca2+ buffering capacity(B value), determined as the ratio ofthe time integral ofICa divided bythe amplitude of Ca2+ transient,was not significantly increased after depletion of Ca2+ stores by the cyclicapplication of caffeine (10 mM) in the presence of ryanodine (4 µM).The addition of cyclopiazonic acid (CPA, 10 µM), a sarco(endo)plasmicreticulum Ca2+-ATPase inhibitor,decreased B value by ~20% in areversible manner. When KCl pipette solution was used,Ca2+-activatedK+ current[IK(Ca)]was also recorded during step depolarization. CPA sensitivelysuppressed the initial peak and oscillations of IK(Ca) withirregular effects on Ca2+transients. The above results suggest that, in guinea pig gastric myocyte, Ca2+ transient is tightlycoupled to ICaduring depolarization, and global[Ca2+]iis not significantly affected byCa2+-inducedCa2+ release from sarcoplasmicreticulum during depolarization.

In spite of the importance of respiration in forest carbon budgets,the mechanisms by which physiological factors control stem respirationare unclear. An experiment was set up in a Eucalyptus globulusplantation in central Portugal with monoculture stands of 5-year-oldand 10-year-old trees. CO2 efflux from stems under shaded andunshaded conditions, as well as the concentration of CO2 dissolvedin sap [CO2*], stem temperature, and sap flow were measuredwith the objective of improving our understanding of the factorscontrolling CO2 release from stems of E. globulus. CO2 effluxwas consistently higher in 5-year-old, compared with 10-year-old,stems, averaging 3.4 versus 1.3 µmol m–2 s–1,respectively. Temperature and [CO2*] both had important, andsimilar, influences on the rate of CO2 efflux from the stems,but neither explained the difference in the magnitude of CO2efflux between trees of different age and size. No relationshipwas found between efflux and sap flow, and efflux was independentof tree volume, suggesting the presence of substantial barriersto the diffusion of CO2 from the xylem to the atmosphere inthis species. The rate of corticular photosynthesis was thesame in trees of both ages and only reduced CO2 efflux by 7%,probably due to the low irradiance at the stem surface belowthe canopy. The younger trees were growing at a much fasterrate than the older trees. The difference between CO2 effluxfrom the younger and older stems appears to have resulted froma difference in growth respiration rather than a differencein the rate of diffusion of xylem-transported CO2. The relationship between chlorophyll a (Chl a) and primary productivity(PP) in the uppermost water layer and the water column-based(0–15 m) integral values of those variables were examinedusing measurements taken in Lake Kinneret (Israel) from 1990to 2003. In 81% of all Chl a profiles examined, the distributionwas fairly uniform within the entire 0–15 m water column,and 12.3% of instances showed a prominent subsurface maximum,when the lake phytoplankton was dominated by the dinoflagellatePeridinium gatunense. Chl a can be reliably estimated by remotesensing techniques in the productive and turbid water of LakeKinneret, since Chl a concentration at surface layers can beextrapolated to the entire water column. Light vertical attenuationcoefficient average for wavelengths from 400 to 700 nm, Kd,ranged from 0.203 to 1.954 m–1 and showed high degreeof temporal variation. The maximal rate of photosynthetic efficiency,PBopt [average 3.16 (±1.50)], ranged from 0.25 to 8.85mg C m–3 h–1 mg Chl a–1. Using measured dataof Chl a, PBopt, and light as an input, a simple depth-integratedPP model allowed plausible simulation of PP. However, a lackof correlation between photosynthetic activity and temperature(or other variable with remotely sensed potential) renders theuse of models that require input of photosynthetic efficiencyto calculate integrated PP of little value in the case of productiveand turbid Lake Kinneret.  相似文献

The transport of macromolecules through the lung interstitiumdepends on both bulk transport of fluid and diffusion. In the presentstudy, we studied the diffusion of albumin. Isolated rabbit lungs wereinflated with silicon rubber via airways and blood vessels, and twochambers were bonded to the sides of a 0.5-cm-thick slab that encloseda vessel with an intersititial cuff. One chamber was filled with eitheralbumin solution (2 or 5 g/dl) containing tracer125I-albumin or with tracer125I-albumin alone; the other wasfilled with Ringer solution. Unbound 125I was removed from the tracerby dialysis before use. The chamber with Ringer solution was placed inthe well of a NaI(Tl) scintillation detector. Diffusion oftracer through the interstitium was measured continuously for 60 h.Tracer mass (M) showed a time(t) delay followed by an increase toa steady-state flow(dM/dtconstant). Albumin diffusion coefficient(D) was given byL2/(6T),where T was the time intercept of thesteady-stateM-t line at zero M, andL was interstitial length.Interstitial cuff thickness-to-vessel radius ratio(Th0/R)was estimated by using Fick's law for steady-state diffusion. BothD andTh0/Rwere independent of albumin concentration.D averaged 6.6 × 107cm2/s, similar to the freeD for albumin. Values ofTh0/Raveraged 0.047 ± 0.024 (SD), near the values measuredhistologically. Thus pulmonary interstitial constituents offered norestriction to the diffusion of albumin.

Previous studies have shown that inhibition of L-type Ca2+ current (ICa) by cytosolic free Mg2+ concentration ([Mg2+]i) is profoundly affected by activation of cAMP-dependent protein kinase pathways. To investigate the mechanism underlying this counterregulation of ICa, rat cardiac myocytes and tsA201 cells expressing L-type Ca2+ channels were whole cell voltage-clamped with patch pipettes in which [Mg2+] ([Mg2+]p) was buffered by citrate and ATP. In tsA201 cells expressing wild-type Ca2+ channels (1C/2A/2), increasing [Mg2+]p from 0.2 mM to 1.8 mM decreased peak ICa by 76 ± 4.5% (n = 7). Mg2+-dependent modulation of ICa was also observed in cells loaded with ATP--S. With 0.2 mM [Mg2+]p, manipulating phosphorylation conditions by pipette application of protein kinase A (PKA) or phosphatase 2A (PP2A) produced large changes in ICa amplitude; however, with 1.8 mM [Mg2+]p, these same manipulations had no significant effect on ICa. With mutant channels lacking principal PKA phosphorylation sites (1C/S1928A/2A/S478A/S479A/2), increasing [Mg2+]p had only small effects on ICa. However, when channel open probability was increased by 1C-subunit truncation (1C1905/2A/S478A/S479A/2), increasing [Mg2+]p greatly reduced peak ICa. Correspondingly, in myocytes voltage-clamped with pipette PP2A to minimize channel phosphorylation, increasing [Mg2+]p produced a much larger reduction in ICa when channel opening was promoted with BAY K8644. These data suggest that, around its physiological concentration range, cytosolic Mg2+ modulates the extent to which channel phosphorylation regulates ICa. Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. To examine the effects of a doubled atmospheric CO2 concentrationand other aspects of global climate change on a common CAM speciesnative to the Sonoran Desert, Agave deserti was grown under370 and 750 µmol CO2 mol–1 air and gas exchangewas measured under various environmental conditions. Doublingthe CO2 concentration increased daily net CO2 uptake by 49%throughout the 17 months and decreased daily transpiration by24%, leading to a 110% increase in water-use efficiency. Underthe doubled CO2 concentration, the activity of ribulose-1,5-bisphosphatecarboxylase/oxygenase (Rubisco) was 11% lower, phosphoenolpyruvatecarboxylase was 34% lower, and the activated:total ratio forRubisco was 25% greater than under the current CO2 concentration.Less leaf epicuticular wax occurred on plants under the doubledCO2 concentration, which decreased the reflectance of photosyntheticphoton flux (PPF); the chlorophyll content per unit leaf areawas also less. The enhancement of daily net CO2 uptake by doublingthe CO2 concentration increased when the PPF was decreased below25 mol m–2 d–1 when water was withheld, and whenday/night temperatures were below 17/12 C. More leaves, eachwith a greater surface area, were produced per plant under thedoubled CO2 concentration. The combination of increased totalleaf surface area and increased daily net CO2 uptake led toan 88% stimulation of dry mass accumulation under the doubledCO2 concentration. A rising atmospheric CO2 concentration, togetherwith accompanying changes in temperature, precipitation, andPPF, should increase growth and productivity of native populationsof A. deserti. Mesophyll resistance to photosynthetic carboxylation (r'm) wasused as a criterion for leaf integrity. It was measured, at25 °C, in the light, before and after periods of high temperature(3 h at 38 °C) in the dark. During the high temperatureperiods, respiration (RD) of attached leaves of Xanthium strumariumwas suppressed from 27%-36% by either low [O2] (1.04% or 0.21%v.v.) or high [CO2] (840 µl 1–1) in the ambientair. Neither treatment affected rates of RD or photo-respirationduring the second period at 25 °C. There was no significant increase of r'm when RD was not suppressedduring the high temperature treatment. When RD was suppressedat high temperatures, r'm increased from about 3s cm–1before, to about 26 s cm–1 after the high temperaturetreatment. The increase depended upon the degree of suppression. It is concluded that increased RD at high temperature in Xanthiumleaves is partly the result of an increase of energy demandingmaintenance. The subsequent rate of carbon dioxide fixationis reduced when this increase of maintenance-induced respirationis inhibited.  相似文献

Laboratory experiments were carried out on the holoplanktonicscyphomedusa Pelagia noctiluca, which exhibits population explosionssome years in the Mediterranean Sea. Feeding experiments performedon small, laboratory-reared medusae showed specific daily rationsof 13 and 35% at 5 and 20 Anemia nauplii prey 1–1 respectively,while basal NH+4 excretion reached 3.9% (specific elementalmetabolic rate) for adults at 22°C. These values appearto be consistent with those previously observed by several authorsfor other planktonic coelenterates. Assuming that such laboratoryphysiological rates are indicative of food requirements in naturalenvironments, and given the high abundance of P. noctiluca observedregionally, we conclude that P. noctiluca can have a strongpredatory impact in the Ligurian Sea (north-west Mediterranean),particularly during the summer and early fall. 3Present address: ORSTOM B.P. 2528, Bamako, Mali  相似文献

Phagotrophic protists are major components of pelagic food webs,both as consumers of bacterial and phytoplankton cells, andas regenerators of inorganic nutrients. In this study, we estimatedthe efficiency of ammonium regeneration by protists feedingon bacteria within natural plank-tonic assemblages, using a15N tracer method, in which the excretion of 15N-labeled ammoniumdue to grazing on 15N pre-labeled bacteria was followed overtime. We tested this approach in experiments based on the additionof heat-killed 15N-labeled bacteria to laboratory cultures andto samples of coastal seawater. During two experiments, variationin abundance of bacterivores and bacterioplankton resulted innon-constant grazing rates. Deterministic computer models thatused abundance of bacteria and protists as variables were developedto estimate best-fit values of grazing mortality (g, h–1)and of ammonium regeneration efficiency (RE, fraction of theinitial 15N label in added bacteria which is released as ammonium).Estimated ammonium RE were 0.30–0.35 for one trophic linksystems with both a monospecific culture and a mixed speciesassemblage of bacterivorous flagellates. RE was higher for multi-trophicstep food webs: 0.60 for 5 µm pre-screened coastal seawaterand 0.90 for whole coastal seawater.  相似文献

The cell quotas of microcystin (Qmcyst), protein (Qprot), chlorophylla (Qchloro) and carbohydrate (Qcarbo), as well as the net productionrates of these parameters, were determined during the exponentialand stationary phases in nine batch cultures of Microcystisaeruginosa (CYA 228) at light regimes from 33 to 53 µmolphotons m–2 s–1. The following results were obtained.(i) A parallel pattern was found in the changes of Qmcyst, Qprot,Qchloro and Qcarbo during the entire growth cycle and significantcorrelations were recorded between Qmcyst and Qprot, Qchloroand Qcarbo. (ii) The net microcystin production rate (µmcyst)was positively correlated with the specific cell division rate(µc), the chlorophyll production rate (µchloro)and the protein production rate. (iii) A significant inverselinear relationship was found between µc and Qmcyst, i.e.cultures with a positive µc had a Qmcyst between 110 and400 fg microcystin cell–1, while declining cultures hadQmcyst values >400 fg microcystin cell–1. Maximum variationin Qmcyst within cultures was 3.5-fold. Collectively, the resultsshow that cells produced microcystin at rates approximatingthose needed to replace losses to daughter cells during divisionand that microcystin was produced in a similar way to proteinand chlorophyll, indicating a constitutive microcystin production.  相似文献

In vitro mammary epithelial cell models typically fail to form a consistently tight barrier that can effectively separate blood from milk. Our hypothesis was that mammary epithelial barrier function would be affected by changes in luminal ion concentration and inflammatory cytokines. Bovine mammary epithelial (BME-UV cell line) cells were grown to confluence on permeable supports with a standard basolateral medium and either high-electrolyte (H-elec) or low-electrolyte (L-elec) apical medium for 14 days. Apical media were changed to/from H-elec medium at predetermined times prior to assay. Transepithelial electrical resistance (Rte) was highest in monolayers continuously exposed to apical L-elec. A time-dependent decline in Rte began within 24 h of H-elec medium exposure. Change from H-elec medium to L-elec medium time-dependently increased Rte. Permeation by FITC-conjugated dextran was elevated across monolayers exposed to H-elec, suggesting compromise of a paracellular pathway. Significant alteration in occludin distribution was evident, concomitant with the changes in Rte, although total occludin was unchanged. Neither substitution of Na+ with N-methyl-D-glucosamine (NMDG+) nor pharmacological inhibition of transcellular Na+ transport pathways abrogated the effects of apical H-elec medium on Rte. These results indicate that mammary epithelium is a dynamic barrier whose cell-cell contacts are acutely modulated by cytokines and luminal electrolyte environment. Previous work has shown that elevated carbon dioxide (CO2) concentrationsin the dark reversibly reduce the rate of CO2 efflux from soybeans.Experiments were performed exposing soybean plants continuallyto concentrations of 350 or 700 cm3 m-3 for 24 h d-1, or to350 during the day and 700 cm3 m-3 at night, in order to determinethe importance of the reduced rate of dark CO2 efflux for plantgrowth. High CO2 applied only at night conserved carbon andincreased dry mass during initial growth compared with the constant350 cm3 m-3 treatment. Long-term net assimilation rate was increasedby high CO2 in the dark, without any increase in daytime leafphotosynthesis. However, leaf area ratio was reduced by thedark CO2 treatment to values equal to those of plants continuallyexposed to the higher concentration. From days 14-21, leaf areawas less for the elevated night-time CO2 treatment than foreither the constant 350 or 700 cm3 m-3 treatments. For the days7-21-period, relative growth rate was significantly reducedby the high night CO2 treatment compared with the 350 cm3 m-3continuous treatment.
