不同处理对海甘蓝种子发芽和幼苗生长的影响   总被引:7,自引:1,他引:6

由于受种子生理休眠作用的影响和硬而厚的种皮所产生的抑制作用,使海甘蓝(CrambemaritimaL.)种子发芽慢,发芽率低。为探索加快海甘蓝种子发芽和提高种子发芽率的有效方法,我们先后进行了8种不同的种子预处理试验和6种不同次氯酸钠溶液浸种的发芽试验。结果发现:(1)种子剥皮处理可以很大程度地促进发芽和提高发芽率;(2)用浓度为0.05%的赤霉酸溶液浸种18h对海甘蓝种子发芽也有很好的促进作用;(3)用0.20%的代森锰锌45M(Diithane45M)溶液浸种20min的消毒处理对海甘蓝种子发芽产生一定程度的抑制作用,但可减少海甘蓝幼苗死亡率;(4)适宜浓度的次氯酸钠漂白水(法文名1'EaudeJavel)的溶液(10%)浸种5min对促进海甘蓝种子发芽和减少幼苗死亡均有良好效果;浓度低于10%时.不足以腐蚀种子硬而厚的种皮而促进种子发芽,也不足以杀死种子携带的病菌而减少幼苗死亡率;浓度大于10%时,对种子的胚和种子内的酶活性产生不良影响,从而抑制种子发芽和影响幼苗的正常生长。  相似文献

通过在不同条件下杏黄兜兰 (Paphiopedilumarmeniacum)种子萌发的观察 ,对影响其萌发的诸因子报道如下 :1)果实的生长期与种子的萌发率有关。实验表明种子采自生长期为 6 0d的果实 ,发芽率为 3 5 % ,采自 12 0d的果实 ,发芽率为 4 0 % ,采自 180d的果实 ,发芽率为 18 3%。 2 )培养基也会影响到种子的萌发 ,种子在 1 5MS内培养 ,萌发率明显高于培养于MS ,RE和改良HyponexNo 1内的种子。 3)培养基 (1 5MS)掺入添加物也会影响到种子的萌发率 ,如掺入 10 %椰子水会促使种子的萌发率达到很高的水平 ,掺入马铃薯泥 (5 0g L)或胰化胨 (2g L) ,种子的萌发率会达到较高的水平 ,而掺入香蕉泥则会对种子的萌发率产生负面影响 ,掺入活性炭 (2g L)会促使种子萌发和幼苗发育。 4 )与固态培养基相比 ,种子在液体悬浮培养基内的萌发速度更快 ,幼苗更为整齐划一。  相似文献

Bacteriol. 91:1168-1177. 1966.-Spores of 21 strains of Bacillus megaterium and 25 other strains representing 13 species of Bacillus were produced under standardized conditions. The germination of a washed spore suspension of each strain was measured as a response to various combinations of 30 different germinative compounds. The strains were first typed with respect to their response to "primary" germination compounds, i.e., glucose, l-alanine, inosine, and l-alanine-inosine mixture, and also Na(+) and K(+). The second stage was the determination of the response to various organic and inorganic anions and cations, each strain being supplied with the "primary" compounds best for it. Marked differences in germination patterns were observed among species and strains of the same species. No relation to established taxonomic lines was evident. A nonspecific requirement for ions was found for all strains, but not all ions were effective. A striking degree of interchangeability of germinative chemicals was found. "Fractional germination" was very common. A mixture of l-alanine and inosine and various ions was the best germinative solution for most strains. Some anomalous germination patterns were encountered. Those studied included a strain whose cells lysed spontaneously upon germination and other strains for which l-leucine had striking germinative powers.  相似文献

Abstract. This report deals with the controversies of whether all germinative epidermal cells in human epidermis are in the cycling state and whether stimulated hyperproliferation of psoriatic epidermis is due to a shortening of the cell cycle time or to a recruitment of non-cycling germinative epidermal cells. Experiments were performed on human subjects in vivo . Continuous infusion of [3H]thymidine for 8½ days indicated that 40% of germinative epidermal cells reside in the non-cycling state. Proliferative stimulation by tape stripping indicated recruitment of non-cycling (G0) germinative epidermal cells in both normal and psoriatic skin, and a prolongation (rather than a shortening) of cell cycle traverse in activated psoriatic epidermal cells.  相似文献

Pollen hydration and germination on the "wet" stigma of Nicotiana tabacum L. were studied by SEM and TEM to reveal the role of the stigma in selecting the germinative pore, and in establishing the axis of polarity in the pollen grain. Pollinated stigmas were fixed with glutaraldehyde or osmium tetroxide vapour, or processed with rapid freeze fixation and freeze substitution. Fixation was performed in 5, 15 or 30 min and 3.5 h after pollination. The tube easily emerged from either pore, this process not depending on the pollen grain orientation relative to the stigma. The orientation of pollen tubes remained random till their length becomes longer than the pollen grain diameter. The TEM analysis of ultrastructural changes in poral regions during pollen hydration and germination showed that the germinative pore was positioned just near the generative cell and vegetative nucleus. Within the first 5 min after pollination a new layer of the electron-lucent wall adjacent to the plasma membrane was formed in the region of a future germinative pore. Following 15 min, marked changes were revealed in the cytoplasm region, close to the germinative pore. Minute dictyosome vesicles were accumulated near the plasma membrane. Small mitochondria and short ER cisternae were distal to a zone of secretory vesicles. The data suggest that the axis of polarity in the germinating pollen grain is predetermined by a spatial organization of the vegetative cell.  相似文献

Bh (black at hatch) is a mutation of Japanese quails which causes darkening or lightening of the plumage in heterozygotes or homozygotes, respectively. We chemically analyzed melanin pigments in feather germs of Bh mutant embryos and in feathers of adult animals. Dark brown dorsal feathers of wild-type adult animals had white barrings, but heterozygous ones lacked clear barrings. The feathers of wild-type and heterozygote animals contained both eumelanins and pheomelanins, the latter being more pheomelanic. On the dorsal skin of 10-day old wild-type embryos, longitudinal stripes from black and yellow rows of feather germs developed; two or three longitudinal rows of black feather germs and then two or three rows of yellow feather germs next to the short central feather germs. Heterozygous embryos appeared black in plumage pigmentation, due to the presence of‘gray’feather germs in rows of dorsal feather germs that corresponded to yellow rows in wild-type embryos. Homozygous dorsal feather germs did not develop the black and yellow longitudinal stripes, but were brown. Chemical analysis showed that embryos of each genotype contained both eumelanins and pheomelanins in the feather germs; however, the eumelanin content in ho-mozygous feather germs was very low. The Bh (black at hatch) gene was examined to determine whether it is expressed in plumage melanocytes by analyzing pigmentation patterns of Bh melanocytes placed in the micro-environment of the feather germs of quail embryos with pink eyes. These host quails genetically lack a large part of plumage melanin. The Bh locus in these almost white quails is wild-type. When Bh neural crest cells were transplanted orthotopically into the host embryos, wild-type and Bh /+ melanocytes, which differentiated from the transplanted neural crest cells, formed plumage pigmentation patterns characteristic of each genotype in the micro-environment of the host feather germs. Brown plumage pigmentation, which was very similar to that of 10-day Bh / Bh embryos, was also observed in the feather germs of host embryos that received Bh neural crest cells, although the genotype of the donors could not be determined. These donors died before pigmentation of their feather germs occurred. The results demonstrate that pigmentation patterns of Bh menalocytes are not altered in the micro-environment of the host germs, suggesting that the Bh gene is autonomous in Bh melanocytes and is expressed in melanocytes of both Bh and the host feather germs, and that it causes the normal pigmentation pattern to be altered.  相似文献

Mandibular molar anlages excised from 17-day mouse foetuses were cultured in vitro or in ovo (on the chorioallantoic membrane). In both cases, the explants were underlain either with a Millipore filter or with a piece of fibrin foam. Tooth germs were harvested after 7 days of cultivation and processed histologically. Spatial arrangement was highly preserved in the tooth germs cultured in vitro on fibrin foam. In vitro cultures on Millipore filters revealed significant flattening of tooth germs, caused especially by the collapse of enamel organ and the pulp. The cytodifferentiation of tooth germs cultured in vitro on both substrates (Millipore filter, fibrin foam) was characterized by the presence of odontoblasts, polarizing ameloblasts and predentine. The cytodifferentiation of tooth germs cultured in ovo on Millipore filters placed on chorioallantoic membrane was characterized by the presence of odontoblasts, ameloblasts, predentine, dentine and enamel. However, the flattening of these explants was identical with the changes of the explants cultured on Millipore filters in vitro. In ovo cultivation on the fibrin foam failed to bring satisfactory results.  相似文献

Summary The influence of colchicine on the addition of 3H-galactose to the enamel protein in secretory amelloblasts of cultured germs of rat molar tooth was investigated by light- and electron-microscopic radioautography. In tooth germs cultured without colchicine, the reaction products of 3H-galactose were observed over Golgi cisternae at early chase times and then localized over the enamel with time. In tooth germs cultured with colchicine, the silver grains were seen over the Golgi cisternae, condensing granules and accumulated secretory granules. Some grains also appeared with time over the pale granular material precipitated in the intercellular space with colchicine treatment. In quantitative analysis with light microscopic radioautography, values of silver grain counts over the unit area (100 m2) on ameloblasts and enamel of colchicine-treated tooth germs were significantly lower at both 0 min and 30 min chase after 30 min pulse than those of control tooth germs, respectively. This finding indicates that colchicine diminished the incorporation of 3H-galactose into the secretory ameloblast of cultured tooth germs. It is suggested that colchicine decreases the activity of the Golgi apparatus with regared to the addition of sugar to the synthesizing glycoprotein in the secretory ameloblast.  相似文献

During germination in the siliquae of developing seeds of a precociously germinating (PG) line of Chinese cabbage (Brassica rapa spp. Pekinensis), the synthesis of cruciferin and oleosins was maintained and the activities of isocitrate lyase and malate synthase increased. Thus, both developmental and germinative/post-germinative events coexisted within the same PG seeds. Drying of the seeds resulted in the developmental events being switched off and germinative/post-germinative ones being initiated. Only about 18% of the seeds of the PG line completed germination in the siliquae, so the potential for germination of the remaining non-germinating seeds was tested during their development. Isolation of these developing seeds from the siliquae and imbibition in water induced PG, but developmental events were terminated and the seeds entered a germinative/post-germinative programme. This termination of developmental events in the induced PG seeds was not permanent, however, and they could be re-induced by incubating the seeds in ABA on Murashige and Skoog (MS) medium. If incubated on MS medium plus a low concentration of ABA, both developmental and germinative/post-germinative events could be induced to coexist in the PG seeds up to the time of establishment of the seedlings. Control of the developmental and germinative/post-germinative events in the seeds was influenced by the environment in which they were developing, which could be mimicked by manipulating the nutritional status and ABA content in which the seeds were in contact.Keywords: Abscisic acid, ABA, Brassica rapa, Chinese cabbage, stored reserves, precocious germination, seed development   相似文献

Atlantic Cutlassfish, Trichiurus lepturus, have large, barbed, premaxillary and dentary fangs, and sharp dagger-shaped teeth in their oral jaws. Functional teeth firmly ankylose to the dentigerous bones. We used dry skeletons, histology, SEM, and micro-CT scanning to study 92 specimens of T. lepturus from the western North Atlantic to describe its dentition and tooth replacement. We identified three modes of intraosseous tooth replacement in T. lepturus depending on the location of the tooth in the jaw. Mode 1 relates to replacement of premaxillary fangs, in which new tooth germs enter the lingual surface of the premaxilla, develop horizontally, and rotate into position. We suggest that growth of large fangs in the premaxilla is accommodated by this horizontal development. Mode 2 occurs for dentary fangs: new tooth germs enter the labial surface of the dentary, develop vertically, and erupt into position. Mode 3 describes replacement of lateral teeth, in which new tooth germs enter a trench along the crest of the dentigerous bone, develop vertically, and erupt into position. Such distinct modes of tooth replacement in a teleostean species are unknown. We compared modes of replacement in T. lepturus to 20 species of scombroids to explore the phylogenetic distribution of these three replacement modes. Alternate tooth replacement (in which new teeth erupt between two functional teeth), ankylosis, and intraosseous tooth development are plesiomorphic to Bluefish + other Scombroidei. Our study highlights the complexity and variability of intraosseous tooth replacement. Within tooth replacement systems, key variables include sites of formation of tooth germs, points of entry of tooth germs into dentigerous bones, coupling of tooth germ migration and bone erosion, whether teeth develop horizontally or immediately beneath the tooth to be replaced, and how tooth eruption and ankylosis occur. Developmentally different tooth replacement processes can yield remarkably similar dentitions.  相似文献

目的探讨粪便pH和涂片检查细菌球/杆在菌群失调诊断中的价值。方法对腹泻3 d以上或疾病中并发腹泻的194例患者为研究对象。采集新鲜粪便(10 min内),观察其性状、颜色、测pH、粪便常规,涂片革兰染色查细菌总数、形态特点、革兰球菌/杆菌及细菌培养。另选同期住院的非腹泻患者、职工家属和到保健科定期体检的小儿80例作为正常对照。结果经培养确诊为菌群失调的有118例,其中男76例,女42例,年龄最小45天,最大5岁。粪便pH失调组分布在6~8,≥7者占78.8%,对照组分布在5~6.5,多集中在≤5.5,占85%,相比差异有显著性(u=12.02,P<0.01)。涂片查球菌/杆菌失调组为3/7~8/2,球菌≥40%者106例(89.8%),对照组为3/7~1/9,球菌≥40%者无1例,两者比较差异有显著性(u=11.19,P<0.01)。两种快速诊断法的灵敏度分别为94.9%和100%,特异度均为95%,诊断界值pH为6.5,球/杆为3/7。结论粪便pH和涂片检查肠道菌群失调方法简便、快捷、经济实用,能在15 min内作出诊断,尤对急重症患者可赢得治疗时机,值得推广。  相似文献

Histological sections of the pharyngeal tonsil (PhT) have been investigated in 55 corpses of persons of both sex and various age, that had no disease connected with lesions of the PhT by the time of their death. The aim of the work is to determine relative areas of each structural element of the PhT in comparison to the whole section area taken as 100%. The PhT lymphoid tissue reached its maximal relative development at the first childhood. Then the lymphoid tissue area in the PhT histological sections decreases gradually, making already in the second mature age one half of the tonsil section area, and in the elderly age--nearly one-fourth. The decrease of the relative lymphoid tissue area takes place at the expense of decreasing area of the lymphoid nodules. The relative area of the connective stroma increases inversely to the relative decrease of the lymphoid tissue area parameters. The germinative centers in the lymphoid nodules are absent in newborns and old persons. The greatest value of the relative area of the germinative centers falls to the early infancy. The lymphoid nodules with the germinative centers, according to their relative area during all age periods, besides the newborn period, the second mature, elderly and old age periods, essentially exceed the area, that occupy the nodules without the germinative centers on the PhT sections.  相似文献

The aim of the present study was to investigate the spatial distribution of Ca and P in dentin and enamel of developing first (M1) and second (M2) maxillary hamster molars (age: 3-5 days) in comparison with cultured molars. For culturing the germs were dissected from 3-day-old hamsters and incubated for 1 and 2 days, respectively, in a modified BGJb medium. Electron probe X-ray measurements were carried out on 3 regions extending in a vertical axis from cusp tip over cusp middle to cusp base next to the cervical loop region. Neither the in vivo nor the in vitro group was statistically different in the Ca and P concentration in the regions of dentin. In both groups the measurements in enamel showed a gradient with an increase in Ca and P from enamel surface towards dentin-enamel junction and a gradient with an increase from cusp base towards cusp tip. Direct comparison of the in vivo group with the in vitro group did not demonstrate a statistical difference between the mineral content of the 4-day-old germs and the 1-day culture germs, respectively the 5-day-old germs and the 2-day culture germs. The results indicate a high correspondence between the mineralization process of in vitro and in vivo tooth germ development.  相似文献

梵净山栲树 (Castanopsis fargesii)群落种子雨丰富 ,对群落更新发展有用的成熟有效种子 2 6 8.9粒·m- 2 。优势种及其它主要种类均有种子产生 ,为维持现存群落的稳定提供了重要基础。种子库中活力种子丰富 ,其数量达 375 4.3粒· m- 2 ,分属 30种植物 ,其中木本植物种子 18种 ,草本植物种子 12种。种子库中有现存植物种子 13种 ,群落演替前期各阶段产生的种子 17种。在种子库中 ,现存群落的优势种和其它主要树种均有活力种子存在 ,群落更新潜力好。在种子库中 ,现存植物产生的种子绝大多数分布于枯枝落叶层 ,只有少部分 (3种 )可分布到土壤层 ,演替前期各阶段产生的种子全部分布在土壤层中。随土壤深度增加 ,种子的种类和物种多样性指数均逐渐减小。从枯枝落叶层向下 ,种子库与现存群落的相似性逐渐减小。在天然情况下 ,种子库中优势种及其它主要种类的种子大量萌发出苗 ,现存群落更新良好 ,稳定性强。  相似文献

In order to elucidate the role of parathyroid hormone-related peptide (PTHrP) in tooth development, we treated tooth germ explants of mouse molars with antisense phosphorothioate-oligodeoxynucleotide (ODN) against PTHrP. Antisense ODN-treatment of the explants resulted in the invasion of the tooth germs by bone. The number of tartrate-resistant acid phosphatase (TRAP)-positive cells around the tooth germs in antisense ODN-treated explants was much lower than that of the control explants. Electron microscopic examination suggested that the antisense ODN-treatment inhibited differentiation of osteoclasts. Treatment of the explants with bisphosphonate or vitamin K2, inhibitors of the differentiation of osteoclasts, induced the invasion by bone into the tooth germs as observed in the antisense ODN-treated explants. The results obtained suggest that PTHrP is involved in the mechanism protecting tooth germs from bone invasion by promoting the differentiation of osteoclasts around them.  相似文献

To better understand excitation-contraction coupling in smooth muscle, myosin phosphorylation and force-velocity properties of canine tracheal muscle were compared during the rise and early plateau of force in electrically stimulated tetani. Velocity reached a peak of approximately 1.5 times plateau value when force had risen to approximately 45% of its maximum value and then declined progressively. Except early in the tetanus, when phosphorylation rose rapidly, maximum power and phosphorylation had nearly parallel time courses, reaching peaks of 1.2-1.3 times reference at 6-8 s before declining to the plateau level at approximately 12 s. Force, velocity, maximum power, and phosphorylation fell somewhat during the plateau, with the closest correlation between phosphorylation and power. These results suggest that 1) early velocity slowing is not associated with light chain dephosphorylation and 2) maximum power, which we use to signal changes in activation, is closely correlated with the degree of light chain phosphorylation, at least when phosphorylation level is not changing rapidly. Dissociation of these two properties would be expected early in the tetanus if phosphorylation precedes mechanical activity.  相似文献
