The effects of 3-hydroxy-β-ionone on the growth of hypocotyls of dark-grown dwarl bean ( Phaseolus vulgaris L. cv. Morocco) and tall bean (cv. Kentucky Wonder) were examined. 3-Hydroxy-β-ionone inhibited growth of the hypocotyl of both cultivars at doses higher than 3 ng per plant. Seedlings of the tall cultivar responded to a slightly lesser extent to 3-hydroxy-β-ionone than those of the dwarf cultivar. Changes in the levels of 3-hydroxy-β-ionone in hypocotyls of both cultivars 24 h after the onset of light irradiation, were determined by high-performance liquid chromatography and the results compared with the results of light-induced inhibition of growth. The difference in levels of 3-hydroxy-β-ionone between the two cultivars was considerable, and the changes in its levels in the hypocotyls of each cultivar reflected the light-induced inhibition of growth of the hypocotyls of the respective cultivars. These results suggest that the endogenous growth inhibitor. 3-hydroxy-β-ionone, may play an important role in the inhibition by light of hypocotyl growth of the two cultivars and may serve to distinguish the growth habits of these cultivars.  相似文献

Dark-grown, 10-day-old bean seedlings (Phaseolus vulgaris L. cv. Morocco) were transferred to continuous light, and the resulting changes in growth rate and concentration of 3-hydroxy-β-ionone, an endogenous growth inhibitor, were monitored. The growth rate of the first internodes of the seedlings decreased rapidly and leveled off 20 h after onset of light. This plateau value was about 25% of the growth rate of the non-irradiated control. The concentration of 3-hydroxy-β-ionone in the internodes of the irradiated seedlings increased rapidly and reached a plateau value after 16 h. This increased level of the inhibitor was about 5 times the level in the non-irradiated control. The changes in the levels of the inhibitor in the internodes reflected the light-induced growth inhibition of the internodes. The longitudinal distribution of the growth inhibitor along the first internodes of the seedlings was closely correlated with the light-induced growth inhibition of the corresponding region of the internodes. The present results suggest that the endogenous growth inhibitor 3-hydroxy-β-ionone may play an important role in the inhibition by light of internode growth of bean seedlings.  相似文献

Growth rate, osmotic potential of the cell, cell wall mechanical properties, sugar composition and molecular weight (MW) distribution of water-soluble hemicellulosic polysaccharides of the second leaf sheath of one tall ( Oryza sativa L. cv. Nihonbare) and two dwarf ( Oryza sativa L. cvs. Tan-ginbozu and Waito C) cultivars of rice were compared. The effect of gibberellic acid (GA3) on the above-mentioned parameters was also studied using the tall (Nihonbare) and one of the dwarf (Waito C) cultivar. The minimum stress-relaxation time (T0) was higher in the cell wall from the two dwarfs than in the tall cultivar. Furthermore, in the water-soluble hemicellulosic polysaccharides the mass-average MW of β -glucan was higher and that of arabinoxylan was lower in the tall cultivar than in the dwarf ones. Thus, dwarfism of cvs Tan-ginbnozu and Waito C might be correlated with the different MW distributions of β -glucan and arabinoxylan. GA3 induces growth in the dwarf Waito C cultivar, decreases the T0 value of the cell wall, and also decreases the average MW of water-soluble hemicelluloses. Changes in β (1–3)(1–4)glucan or arabinoxylan or in both are proposed as part of the cell wall loosening mechanism induced by gibberellin.  相似文献

In order to clarify the role of endogenous growth inhibitors A-2α and A-2β in a dwarf pea plant, red light (emission peak 657 nm) treated, 9-d-old seedlings of dwarf pea (Pisum sativum L. cv. Progress No. 9) were transferred to darkness, and the resulting changes in growth rate and concentrations of A-2α and A-2β were monitored. The growth rate of the epicotyls increased, and the concentration of the inhibitors in the epicotyls decreased, according to sigmoidal time courses. The relationship between the logarithms of the concentration of the inhibitors and the corresponding growth rate was linear. These results suggest that A-2α and A-2β, may play an important role in the growth recovery process of the dwarf pea cultivar after termination of red light irradiation. The contents of non-acidic, acidic and bound growth inhibitorsin hypocotyls and cotyledons were compared between dark- andlight-grown dwarf and tall beans by means of thin-layer chromatographyand bioassay. In the non-acidic fraction, one major inhibitoryactivity appeared on the chromatogram, but its Rf zone was differentbetween hypocotyls and cotyledons. In both the acidic and boundinhibitor fractions, one major inhibitory activity appearedat the Rf zone corresponding to ABA. The ABAIike substance whichwas the major inhibitor in the hypocotyl was more abundant inlight-gorwn than dark-grown ones, especially in the dwarf variety,but light irradiation did not cause its transport from cotyledonsto the hypocotyl. A larger amount of bound ABA-like substance,which was the major inhibitor in the cotyledon, was presentin the dwarf than the tall variety regardless of the light condition.Cotyldon-enhanced photoinhibition of hypocotyl growth couldnot be explained by the levels of the xanthoxinand ABA-likeinhibitors, or the transport of these free inhibitors from cotyledons. Red light inhibited the growth of the apical part of the hookin dark-grown seedlings of a dwarf variety (cv. Progress No.9) of pea (Pisum sativum L.), whereas it promoted such growthin a tall variety (cv. Alaska). In the elongation zone of theepicotyl of the dwarf variety the extent of inhibition of growthwas similar to or even smaller than that in the tall variety.Local irradiation of the apical part of the hook also causedinhibition of growth in the hook of the dwarf variety and promotionof growth in the tall variety. The inhibition of growth in theapical part of the hook of cv. Progress may be involved in thedwarfism induced by irradiation with red light of this cultivar. The putative auxin-transporting cells of the intact herbaceous dicotyledon are the young, differentiating vascular elements. The length of these cells was found to be considerably greater in dwarf (Meteor) than in tall (Alderman) varieties ofPisum sativum L., and to be greater in etiolated than in light-grown plants ofP. sativum cv Meteor andPhaseolus vulgaris L. cv Mexican Black. Under given light conditions during transport these large differences in cell length did not influence the shapes of the transport profiles or the velocity of transport of14C-labelled indol-3yl-acetic acid (IAA) applied to the apical bud. However, in both etiolated and light-grown bean and dwarf pea plants the velocity of transport in darkness was ca. 25% lower than that in light. Under the same conditions of transport velocities in bean were about twice those observed in the dwarf pea. Exposure to light during transport increased the rate of export of14C from the labelled shoot apex in green dwarf pea plants but not in etiolated plants. The light conditions to which the plants were exposed during growth and transport had little effect on the rates of uptake of IAA from the applied solutions. The results indicate that the velocity of auxin transport is independent of the frequency of cell-to-cell interfaces along the transport pathway and it is suggested that in intact plants auxin transport is entirely symplastic.  相似文献

Hypocotyl sections with and without the cotyledons were cutfrom bean seedlings and incubated under white light of 6000lux. The cotyledons had an inhibitory effect as well as a promotiveeffect on hypocotyl growth. The former effect was more strikingin the dwarf variety, and the latter in the tall variety. Whenthe hypocotyl units were exposed to light for shorter times(6 hr or less) or incubated under weaker light (1600 and 50lux), the inhibitory effect of the cotyledons decreased greatly,and in the tall variety the presence of cotyledons producedno inhibition, but a promotion of hypocotyl growth. GA treatmentenhanced hypocotyl growth and counteracted the growth inhibitioncaused by the cotyledons. On the whole, the GA effect was moremarked in the tall variety than in the dwarf. The elongation of bean hypocotyls may be controlled by a balancebetween the inhibitory and promotive effects of cotyledons,and the predominance of the former over the latter may be oneof the causes for expressing dwarfing. Summary A number of dwarf wheat cultivars of the Norin 10 type were compared with several tall forms. Applied gibberellic acid markedly stimulated the growth of seedlings of the tall cultivars but not the growth of dwarf seedlings. Several other gibberellins were also inactive when tested with one dwarf cultivar. De-embryonated grains of all cultivars formed -amylase in response to gibberellic acid. Gibberellic acid caused an increase in soluble carbohydrates in the leaves of the tall cultivars but not in those of the dwarfs.Germinating grains, light-grown seedlings and developing stems of the dwarf cultivars contained more endogenous gibberellin-like activity than those of tall cultivars. It is suggested that the dwarf cultivars have a block to the utilisation of gibberellin in the shoot.  相似文献

Gibberellic acid (GA3) stimulated shoot elongation in both dwarf and tall cultivars of pea, but more so in the dwarf cultivar. The sterol composition of shoots of both cultivars was similar, with sitosterol being the most abundant compound, followed by stigmasterol and campesterol. Cholesterol could not be detected. Following GA3 application, levels of free sterols in whole shoots increased whereas glycoside levels tended to fall. The magnitudes of the changes in both classes of sterol were similar in both cultivars. Analyses of stems and leaves separately revealed a greater growth response to GA3 in the former but no effect of the hormone on the sterol composition of either organ. It is concluded that GA3 enhancement of shoot growth in pea is not mediated through quantitative changes in cell sterols.  相似文献

The solanaceous fruit fly, Bactrocera latifrons (Hendel) (Diptera: Tephritidae), infests various solanaceous fruits including eggplant and chili peppers. We found that a freshly cut fruit of an eggplant cultivar [ Solanum melongena L. cv. Long Purple Oriental (Solanaceae)] selectively attracted and provoked voracious feeding behavior in adult B. latifrons males (but not in females) in an indoor test. One of the male-specific attractants/feeding stimulants in eggplant was identified as 3-hydroxy-α-ionone. Sexually mature males that fed on the eggplant pulp selectively accumulated a series of 3-oxygenated α-ionone/α-ionol analogs (e.g., 3-oxo-α-ionol and 3-oxo-7,8-dihydro-α-ionol) in the rectal gland, a suspected pheromone reservoir in male flies. Males fed on synthetic 3-hydroxy-α-ionone, 3-oxo-α-ionol, or 3-oxo-α-ionone partially biotransformed the compounds into 3-oxo-α-ionol, 3-oxo-α-ionone, and 3-oxo-7,8-dihydro-α-ionol and sequestered substantial quantities (as high as 5 µg/gland as a mixture) in a similar ratio in the rectal gland within 6 h after ingestion. These results suggest that the rectal sequestrates may serve as a sex pheromone similar to other Bactrocera species that use phenylpropanoid volatiles to attract conspecific females.  相似文献

Studies to determine the variations in leucine aminopeptidase(LAP) activity in tall and dwarf cultivars of rice were conductedby horizontal starch gel electrophoresis. The samples were takenat weekly intervals starting with soaked seeds (12 h) untilthe post-panicle stage. There is a distinct pattern of LAP isoenzymesin tall and dwarf cultivars studied. Some isoenzymes appearat a particular developmental stage and others disappear whilesome remain fairly constant once they are formed. A tall cultivar(Hansraj) shows greater fluctuations during the course of developmentthan the dwarf cultivars. The effect of red light on the aging progression of the bean (Phaseolus vulgaris L.) hypocotyl segment unit was examined in relation to dwarfism using Kentucky Wonder (tall) and Masterpiece (dwarf) varieties. In both plants, red light promoted the elongation of younger zones and inhibited that of mature zones. The zone exhibiting maximum elongation was shifted to the younger zones by red light irradiation regardless of the plant type, but its extent was greater in the dwarf than in the tall. Thus, red light hastens both the beginning of elongation in the younger portion and its termination in the mature portion of the hypocotyl, particularly of the dwarf plant. These red light responses in each zone of both the tall and dwarf hypocotyl units were reversed by subsequent exposure to far red light regardless of the duration and intensity of red light, thus indicating that the hastened aging progression of the hypocotyl by red light is mediated by phytochrome. However, there is no difference in the rate of decay of Pfr between the tall and dwarf hypocotyls.  相似文献

Variations in the content of A-2α and β, novel endogenous growth inhibitors, andcis,trans- andtrans, trans-xanthoxins were determined in 6- or 7-d-old, dark-grown seedlings of peas (Pisum sativum L. cvs. Progress No. 9, dwarf, and Alaska, tall) under various treatments with red light (R), and compared with R-induced growth inhibition. After transfer of the plants to continuous R the contents of the A-2s in cv. Progress increased after a 20-min lag, and reached plateaus after 12 h, whereas they remained almost unchanged in darkness. Both the rates of increase of the A-2s and the plateau levels were proportional to the logarithm of the irradiance applied. After a 10-min R pulse, the contents of both A-2α and β increased with the same rapidity to reach peaks after 6 h, and then gradually decreased to the initial levels after about 24 h. The effect of R was shown to be phytochrome-dependent, being nullified by far-red light. The level of neithercis,trans- nortrans,trans-xanthoxin showed such a close correlation with growth inhibition, although both xanthoxins increased as a result of phytochrome action. It is highly suggestive that the A-2s, rather than the xanthoxins, are responsible for phytochrome-dependent growth inhibition in cv. Progress. In cv. Alaska, in contrast, R-induced increase of the A-2s was rapid but slight, and could not explain the transient growth inhibition, which was found to be as large as that in cv. Progress shortly after the onset of R. The large content of the A-2s in cv. Progress in the steady state under continuous R, compared with that in cv. Alaska, may explain the dwarfism of cv. To correlate endogenous growth inhibitors of dwarf pea to its dwarfism a thorough search of growth inhibitors was made in the neutral fraction of an acetone extract from the shoots of light-grown seedlings of Pisum sativum L. cvs Progress No 9 and Alaska. From both cultivars six inhibitors were separated and named A-1, A-2, A-3, B-1, B-2 and B-3 based on their order of elution from the silica gel column. Their contents as determined by cress root bioassay were: in cv. Progress 0.40, 16.5, 6.36, 1.02, 0.11 and 0.10, and in cv. Alaska 0.33, 2.35, 3.51, 0.95, 0.10 and 0.09 cress units (g fresh weight)−1. Their contributions to growth regulation of the relevant pea plants were estimated as the products of the above-stated contents times the ratios of the specific activities of each standard sample in the cress roots and the relevant pea cultivars, and they were; in cv. Progress A-2, 5.44 and A-3, 2.10, and in cv. Alaska A-2, 0.68 and A-3, 0.88, those of A-2 and A-3 constituting more than 90% of the total contribution of the six inhibitors in either cultivar. The great differences in the content and contribution to growth regulation of A-2 and A-3 between the two cultivars suggest that the higher contents of and greater responsiveness to the two inhibitors in cv. Progress may be causes of dwarfism of this cultivar. A-l and A-3 were spectroscopically identified with pisatin and a mixture of cis, trans– , and trans, trans xanthoxins.  相似文献

Application of gibberellic acid to the apex of dwarf bean plants (cv. Alabaster) stimulated the elongation growth of epicotyl and hypocotyl but showed no significant effect on elongation growth in a normal cultivar (‘Blue Lake’). Gibberellin-treatment of dwarf plants was characterized by about twofold increase in the level of endogenous auxin. Maximum increase in IAA level was observed after 48 h of GA treatment. There was less increase in IAA content in normal bean plants. — Gibberellin treatment to excised epicotyl and hypocotyl sections of either dwarf or normal cultivar showed no effect on elongation growth. However, a considerable increase in the auxin level was observed in the sections of the dwarf cultivar. The maximum effect occurred with only 1 h incubation in basal medium containing gibberellin. — The indolo-α-pyrone spectro-fluoremetric method for IAA determination was used.  相似文献

A new growth inhibitor, tentatively named pisumin, which increased under red light and remained at initial level or decreased when dwarf pea (Pisum sativum L. cv Progress No. 9) seedlings were transferred from red light to dark, has been isolated in the form of a colorless powder from light-exposed epicotyls of dwarf peas, and characterized partially as an aliphatic carboxylic acid (molecular weight 284) by spectrometric analyses.

Exogenous pisumin inhibited the growth of epicotyl segments of dwarf peas at concentrations higher than 0.1 millimolar in the dark.

Mitochondria from dwarf and tall light-grown peas have been separated from contaminating chloroplasts and other organelles on discontinuous sucrose gradients. A comparison between activities of the mitochondria from the two cultivars of light-grown pea seedlings was made. It was observed that both respiratory control (R.C.) indices and phosphate to oxygen (P : O) ratios were superior in mitochondria from the tall cultivar than the dwarf cultivar. The results are discussed in relation to energy status and possible gibberellic acid content in the two cultivars.  相似文献

Gibberellin A4&7 was more effective than gibberellic acid in increasing shoot elongation when applied to the apex of intact Lycopersicum esculentum seedlings of Tiny Tim, a dwarf cultivar, and Winsall, a tall cultivar. After 14 days, gibberellic acid and gibberellin A4&7 stimulated growth of the dwarf more than the tall tomato. In tall tomato the application of indole-3-acetic acid alone (6.1 μg/plant) showed an inhibitory growth effect, but when applied with 17.5 μg per plant of gibberellic acid, it had a synergistic effect at 7 days but not at 14 days. When the auxin concentration was reduced to 0.61 μg per plant a synergistic effect was observed on tall plants at 7 and 14 days between indole-3-acetic acid and gibberellic acid. Application of gibberellin A4&7 with auxin did not give a synergistic response in tall or dwarf tomato.  相似文献
