Successfully recovering stored cryopreserved cells requires attention to two major factors, i.e., storage temperature and proper handling of the frozen samples. These key considerations are in turn impacted by how the inventory system is constructed, the frequency and process for retrieving specimens, as well as how the material is secured. In addition, shipping biological materials requires attention to the type of material transported, adherence to regulatory requirements, packaging materials and proper assembly, labeling, and engaging reputable carriers. This paper provides an overview discussion of the current best practices for storing, handling, and shipping of cryopreserved cells and provides references to more detailed information on specific topics.  相似文献

By 1996 an inventory of Mexican cladocerans hadrecorded 48 species of which 22 belonged toChydoridae, Macrothricidae and Ilyocryptidae.Unfortunately, most of the surveys were made beforeresearchers knew that these crustaceans are notentirely cosmopolitan. For this reason and the lack ofdeposited type material, many of these records aredoubtful and need detailed analyses. In this study,material from 18 water bodies located in theNeovolcanic Province of Mexico is analyzed andcompared with the literature. Also, males of Camptocercus dadayi Stingelin and Leydigialeydigi (Schoedler) and new records Eurycercus longirostris Hann and Biaperturaintermedia (Sars) are described. Half of thetotal number of species recorded are American endemicsand represents a mixture of the North and SouthAmerican fauna, suggesting that Mexico constitutes atransition between Nearctic and Neotropicalzones.  相似文献

An inventory of the type specimens of Orchidaceae in the general herbarium at the Komarov Botanical Institute (LE), St. Petersburg, Russia, is presented. The history of the collections and their significance are discussed. Taxonomic findings are presented. Purpose

The Social Life Cycle Assessment guidelines (UNEP-SETAC 2009) distinguish two different SLCA approaches, type I and type II. Few comprehensive and analytical reviews have been undertaken to examine the multiplicity of approaches that have been developed within type I SLCA. This paper takes on the task of exploring the evaluation methods used in type I SLCA methods.

Methods

In order to tackle this work, a critical literature review was undertaken, covering a total of 32 reviewed articles, ranging from 2006 to 2015. Those articles have been selected for they make explicit reference to type I, performance reference points (PRPs), corporate behavior assessment, and social performance assessment or if their assessment methods generated a result located at the same point as the inventory data, with regards to the impact pathway. The selected articles were analyzed with a focus on the inventory data used, the aggregation of inventory data on the functional unit, and the type of characterization and weighting methods used. This analysis allowed to make explicit the often implicit logic underlying the evaluation methods and to identify the common denominators of type I SLCA.

Results and discussion

The analysis highlighted the multiplicity of approaches that are comprised within type I SLCA today, both in terms of the data collected (in particular, its positioning along the impact pathway); the presence of some optional steps, such as the scaling of inventory data on the functional unit (FU); and in terms of the different characterization and weighting steps. With regards to data collection, this review has highlighted that the furthest indicators are positioned along the impact pathway, the hardest it is to justify the link between them and the activities of companies in the product system. The analysis also suggested that an important differentiating factor among type I SLCA methods lies in “what the inventory data is assessed against” at the characterization step and how it is ultimately weighted. To illustrate this, a typology of six characterization methods and five types of weighting methods was presented.

Conclusions

It is interesting to identify which approaches are most appropriate to respond to the various questions that SLCA aims to respond to. A question that arises is what approaches are most likely to tell us anything about the impact of a product system on social well-being? This question is particularly relevant in the absence of well-documented impact pathways between activities within product systems and impact on social well-being.

This paper presents evidence that cultivars of manioc (Manihot esculenta) have been selected for combinations of characters that allow them to be perceptually distinguished. This mode of selection is proposed to explain why cultivars are so variable in perceptually salient taxonomic characters unrelated to the use or survival of the plant. This paper reanalyzes published material on manioc and related species and presents new evidence from the inventory of manioc cultivars maintained by the Aguaruna Jívaro of northern Peru. Rogers and Fleming’s (1973) sample of manioc cultivars exhibits 3 characteristics implied by the model of selection for perceptual distinctiveness: high, continuous, and independent variation of nonadaptive taxonomic characters. The inventory of Aguaruna cultivars exhibits 2 additional characteristics predicted by the model: taxonomic characters of this local inventory vary as independently and nearly as greatly as those of the species as a whole. Anthropological evidence is presented to demonstrate that Aguaruna interact with the plants as predicted by the model: Aguaruna identify cultivars using many of the same characters as Rogers and Fleming and they confuse cultivars they regard as similar in stem color, petiole color, and leaf shape. The evidence suggests that procedures used by cultivators to identify cultivars leave their imprint on the plants; crops show the effect of the cultivator's eye as well as hand.  相似文献

Despite their strategic potential, tool management issues in flexible manufacturing systems (FMSs) have received little attention in the literature. Nonavailability of tools in FMSs cuts at the very root of the strategic goals for which such systems are designed. Specifically, the capability of FMSs to economically produce customized products (flexibility of scope) in varying batch sizes (flexibility of volume) and delivering them on an accelerated schedule (market response time) is seriously hampered when required tools are not available at the time needed. On the other hand, excess inventory of tools in such systems represents a significant cost due to the expensive nature of FMS tool inventory. This article constructs a dynamic tool requirement planning (DTRP) model for an FMS tool planning operation that allows dynamic determination of the optimal tool replenishments at the beginning of each arbitrary, managerially convenient, discrete time period. The analysis presented in the article consists of two distinct phases: In the first phase, tool demand distributions are obtained using information from manufacturing production plans (such as master production schedule (MPS) and material requirement plans (MRP)) and general tool life distributions fitted on actual time-to-failure data. Significant computational reductions are obtained if the tool failure data follow a Weibull or Gamma distribution. In the second phase, results from classical dynamic inventory models are modified to obtain optimal tool replenishment policies that permit compliance with such FMS-specific constraints as limited tool storage capacity and part/tool service levels. This paper studies the impact of a reduction in hardware complexity on the supply-chain inventory against various customer on-time delivery alternatives and manufacturing environments. Different methods of reducing hardware complexity are proposed, and their impact on total supply-chain inventory and customer serviceability quantified. An analytical inventory optimization scheme taking into account multistage supply networks, product structure, forecast accuracy, lead-time variability, and supplier reliability is used to determine optimal inventory levels in a stochastic modeling environment. The analysis is based on a business case for an IBM midrange computer family consisting of more than 200 models and upgrades with hundreds of features. We investigate different hardware complexity reduction strategies, including low-usage feature reduction, low-volume feature reduction, and feature substitution, as well as quick response and postponement mechanisms. Our computational results show that, in a fabrication-fulfillment center environment, reducing hardware complexity results in significantly higher inventory savings than in an integrated manufacturing environment. The results presented in this paper were used to reduce hardware complexity in IBM's midrange computer division.  相似文献

The species richness and relative abundance of faunas in defined areas are the raw materials in biodiversity science. The research methodology to acquire these data is inventory, and inventory quality depends on a number of parameters, for example collecting methods, season, and collector experience. To assess the ability of rapid inventory techniques to estimate local richness seven collectors sampled the fauna of true bugs in the Iberá watershed (Corrientes, Argentina) with seven methods during early spring, summer, and late summer (December, May, September) of 1 year. Twenty-three families, 225 spp. and 4,678 adults were found. We also applied various statistical techniques to correct the observed data for undersampling bias, which suggested the lower bound of annual heteropteran species richness at Pellegrini was about 250–300 species. Among heteropteran families, the particular inventory methodology was especially efficient in sampling Miridae. A brief narrative is presented of the events surrounding the initial discovery of a coelacanth in an Indonesian fish market in Manado, Sulawesi, on 18 September 1997. Although the specimen was not purchased and preserved, photographs were taken and enough information gathered about the fish to warrant an official entry in the Coelacanth Conservation Council (CCC) inventory of known specimens of the living coelacanth Latimeria chalumnae. All known information about this first Indonesian specimen is summarized in the official CCC inventory format, and the specimen is herein numbered CCC no. 174.  相似文献

Biological inventory is a crucial activity in life sciences field research. However, it is sometimes time-consuming and laborious to take representative samplings of communities, especially in the case of invertebrates. In this paper, we address the issue of sampling efficiency and its influence on obtained results. As a study system, we used data on epigeic carabid beetles (Carabidae) collected in 1999–2001 in the Warta River valley of western Poland. We trapped a total of 17,722 individuals belonging to 108 species. However, due to rarefaction methods, the expected number of species was estimated at 134–140, suggesting that from 26 to 32 species are missing from the material, even expressed as a huge number of collected specimens. The estimated probability that another captured individual will represent a new species (i.e. a species that was not already recorded) is 0.0010. In order to record all the species present in the study area, another 193,338 individuals need to be sampled (abundance-based approach) or another 1,871 samples need to be collected (incidence-based approach). This means that the collected material should be 10.9 times greater (or 7.9 times greater for incidence-based data) than what was actually collected in order to record all the species present in the study area. The results show that, in practice, full inventory is simply nearly impossible to achieve, and this knowledge should be included in inventory planning. Therefore, we argue that species accumulation curves and unseen species estimators need to be carefully examined and threshold probability of detecting a new species should be built into the design of inventory science. The ratio between recorded and estimated species richness and the estimated efficiency of further sampling can be easily computed with available freeware software and should be incorporated when performing biological inventories.  相似文献

Life Cycle Assessment is becoming an important tool for guiding environmental design improvements in the automotive industry. This paper reports the life cycle inventory profiles for two fuel tank systems based on a collaborative effort between the National Pollution Prevention Center at the University of Michigan, General Motors Research and Development, and the National Risk Management Research Laboratory of the U.S. Environmental Protection Agency. Two 31 gallon functionally equivalent fuel tank systems used on a 1996 light duty vehicle were investigated: a multi-layer HDPE tank with a steel shield and PVC coated steel straps, and a steel tank with a HDPE shield and painted steel straps. Overall, the HDPE fuel tank system is environmentally preferable to the steel tank system based on the set of inventory results presented in this investigation. The Life Cycle Inventory analysis indicated lower energy burdens for the HDPE tank system and comparable solid waste burdens for both systems. The total life cycle energy consumption for the steel and HDPE tank systems were 4.9 GJ and 3.6 GJ per tank, respectively. The energy consumption and most of the air pollutants inventoried occurred as a consequence of the use phase. The solid wastes were generated primatily during the material production phase for the steel tank (13 kg) and during the end-of-life management phase for the HDPE tank (14 kg). This study also highlights data analysis and modeling challenges, including manufacturing and use phase allocation methods.  相似文献

The northern tropical river and wetland systems of Australia cover a vast and mostly remote area. Comparably little is known of wetland type, species diversity, ecological processes and ecosystem services. Systematic inventory and classification is lacking and research remains disparate in purpose and coverage. An integrated framework is used to evaluate results from three multidisciplinary studies across various scales to synthesise available knowledge relating to key ecological attributes across both the study region and within focus areas. The utility of geomorphic and hydrological classification at coarse scales is supported but the unavailability of biological data at corresponding and finer spatial and temporal scales constrains the potential for ecological inventory and classification utilising existing information. Future inventory and ecological studies across this and similar vast study areas need to consider the apriori development of a user-driven data framework and the development of specific investigatory tools to overcome technical and other barriers associated with the requirements of multi-scalar ecological studies.  相似文献

The Internet leads to material and energy consumption as well as various environmental impacts on both the regional and global scale. Yet, assessments of the Internet's energy consumption and resulting greenhouse gas emissions are still rare, and assessments of material flows and further environmental impacts are virtually non‐existent. This article investigates material flows, the direct energy consumption during the use phase, as well as environmental impacts linked to the service, “Internet in Switzerland.” In our model, the service, Internet in Switzerland, is divided into various Internet participant categories. All devices used to access or provide Internet services are merged in a limited number of equipment families and, as such, included in an inventory of the existing infrastructure (stock). Based on this inventory, a material flow analysis (MFA) is performed, which includes the current stock as well as flows resulting from growth and disposal. The direct energy consumption for the operation of the infrastructure is quantified. Environmental impacts are calculated with a life cycle assessment approach, using the ecoinvent database and the software, SimaPro, applying four different methods. The MFA results in a 2009 stock of 98,100 tonnes. Approximately 4,130 gigawatt hours per year, or 7% of the total Swiss electricity consumption, were used in 2009 to operate the Swiss infrastructure. The environmental impacts caused during the production and use phases vary significantly depending on the assessment method chosen. The disposal phase had mainly positive impacts as a result of material recovery.  相似文献

Data quality     
A methodology is presented that enables incorporating expert judgment regarding the variability of input data for environmental life cycle assessment (LCA) modeling. The quality of input data in the life-cycle inventory (LCI) phase is evaluated by LCA practitioners using data quality indicators developed for this application. These indicators are incorporated into the traditional LCA inventory models that produce non-varying point estimate results (i.e., deterministic models) to develop LCA inventory models that produce results in the form of random variables that can be characterized by probability distributions (i.e., stochastic models). The outputs of these probabilistic LCA models are analyzed using classical statistical methods for better decision and policy making information. This methodology is applied to real-world beverage delivery system LCA inventory models. The inventory study results for five beverage delivery system alternatives are compared using statistical methods that account for the variance in the model output values for each alternative. Sensitivity analyses are also performed that indicate model output value variance increases as input data uncertainty increases (i.e., input data quality degrades). Concluding remarks point out the strengths of this approach as an alternative to providing the traditional qualitative assessment of LCA inventory study input data with no efficient means of examining the combined effects on the model results. Data quality assessments can now be captured quantitatively within the LCA inventory model structure. The approach produces inventory study results that are variables reflecting the uncertainty associated with the input data. These results can be analyzed using statistical methods that make efficient quantitative comparisons of inventory study alternatives possible. Recommendations for future research are also provided that include the screening of LCA inventory model inputs for significance and the application of selection and ranking techniques to the model outputs.  相似文献

This article presents an approach to estimate missing elements in hybrid life cycle inventories. Its development is motivated by a desire to rationalize inventory compilation while maintaining the quality of the data. The approach builds on a hybrid framework, that is, a combination of process‐ and input–output‐based life cycle assessment (LCA) methodology. The application of Leontief's price model is central in the proposed procedure. Through the application of this approach, an inventory with no cutoff with respect to costs can be obtained. Despite the time-dependent behavior of carbon stored in plastic materials, literature assessing carbon flows into and from plastic typically applies a static approach. To better understand the climate impacts of such storage, this study explores how carbon stored in plastics can contribute over time to the national climate inventory with various emphasis on recycling. This is accomplished by implementing material stock change estimations for carbon in plastic materials that follow first-order decay and include impacts from recycling rates in the Integrated MARKAL-EFOM System model generator for Sweden (TIMES-Sweden). Thereafter, three approaches to how carbon released from the plastic material stock is accounted for in the national climate inventory were applied to determine how each approach affects resulting emission and net-zero pathways in different recycling rate scenarios. An accounting approach that follows the first-order decay pattern of material stocks was found to be important for capturing the impacts of recycling and for neither over- nor underestimating the emission impact from carbon stored in plastics. Accounting for carbon stored in plastics may provide important incentives for producing renewable plastics and reducing dependence on carbon removal technologies. Because of its synergies with recycling, the carbon storage potential of plastic products is well worth recognizing and promoting in a policy setting that aims for circularity. For Sweden, this reduces the need for bioenergy carbon capture and storage and makes more biomass-based carbon and electricity available for use elsewhere in the energy system.  相似文献
