A 50% dichlorvos containing insecticide formulation (Unifosz 50 EC) and a 50% atrazine containing herbicide formulation (Hungazin PK 50 WP) were studied in chicken embryos after administration as single compounds. Applied concentrations of dichlorvos were 0.1% (corresponding to the plant protection practice), 0.05%, 0.02%, 0.01%. Applied concentrations of atrazine were 0.66% (corresponding to the plant protection practice), 0.33%, 0.132%, 0.066%. The test materials were injected directly into the air-chamber of eggs on day 0 of the hatching period and evaulation was carried out on day 19 of incubation. The chicken embryos were examined for the following: rate of embryo mortality, body mass, type of developmental anomalies. After the single administrations of dichlorvos containing insecticide formulation and atrazine containing herbicide formulation on day 0 of incubation, the average body weight of chicken embryos significantly did not decrease as compared to the control. After the individual administrations of pesticides the incidence of developmental anomalies was sporadic. The embryonic mortality markedly increased at the highest concentrations of pesticides. The rate of embrio mortality were 61% (dichlorvos insecticide containing formulation) and 52% (atrazine containing herbicide formulation). In summary, the 50% dichlorvos containing insecide formulation (Unifosz 50 EC) and the 50% atrazine containing herbicide formulation (Hungazin PK 50 WP) were toxic to the developing chicken embryos at the highest concentration in our study. The toxic effect was expressed in the high rate of embrio mortality.  相似文献

The toxic effects of a widely used organophosphate insecticide (BI 58 EC containing 38% dimethoate as active ingredient) applied alone or in combination with cadmium sulphate modelling the heavy metal load of the environment were studied on chicken embryos in the early phase of development. Solutions and emulsions of different concentrations were made from the test materials and injected in 0.1 ml volume into the air space of eggs on the first day (day 0) of incubation. Subsequently, on days 2 and 3 of incubation permanent preparations were made from the embryos in order to study the early developmental stage. Embryos fixed on slides and stained with osmium tetroxide solution were studied under light microscope. Summarising the findings, it can be established that the embryotoxicity increased after the simultaneous administration of Cd-sulphate and 38% dimethoate containing insecticide formulation compared to the control or the individually treated groups.  相似文献

The use of pesticides involves the risk of poisoning on wild animals. Teratological tests carried out on avian embryos provide useful data for environmental protection and facilitate the development of environment-friendly chemical plant protection techniques. A 30% dimethoate containing insecticide formulation (BI 58 EC) and a 20% benfluralin containing herbicide formulation (Flubalex) and a 960 g/l S-metolachlor containing herbicide formulation (Dual Gold 960 EC) were studied in chicken embryos after single administration by immersion and injection technique. Treatment was done on day 0 of incubation. Applied concentration of pesticides were 0.1% (dimethoate) and 2.05% (S-metolachlor) and 0.375% (benfluralin) corresponding to that used in plant protection practice. Test materials were injected into the air chamber in a volume of 0.1 ml/egg, or eggs were treated by the immersion technique for 30 min. at 37 degrees C. Evaluation was done on day 19 of incubation. Injection treatment: the administration of S-metolachlor and benfluralin did not result a significant decrease in the average body weight of embryos. At the same time the body weight of embryos significantly decreased because of single administration of dimethoate. The embryomortality increased markedly after the administration of test materials (S-metolachlor, benfluralin, dimethoate). Immersion treatment: the administration of S-metolachlor and benfluralin and dimethoate did not result a significant decrease in the average body weight of embryos. The rate of embryomortality was low after the administration of S-metolachlor, benfluralin and dimethoate. After the immersion and the injection treatment the incidences of developmental anomalies were sporadic. In summary it can be established that the injection treatment was more toxic than immersion technique of the test materials in our study.  相似文献

Environmental pollution of metal modelled by copper sulphate, cadmium sulphate and a 960 g/l S-metolachlor containing herbicide formulation (Dual Gold 960 EC) were studied on chicken embryos after administration as a single compounds or in combination. The test materials were injected into the air-chamber in a volume of 0.1 ml/egg on day 0 of incubation. The concentration of copper, and cadmium sulphate was 0.01%. The applied concentration of Dual Gold 960 EC was 0.375%. Evaluation was done on day 19 of the hatching period. In comparison with the values of the control group, the rate of embryomortality also increased significantly in the groups which were treated by herbicide and heavy metals individually. When the herbicide (Dual Gold 960 EC) was used individually, one quarter of the treated animals died in comparison with the values measured in the control group. In the groups which were treated with cadmium sulphate and Dual Gold 960 EC embryomortality increased significantly in comparison with both the control and individually treated groups. In the groups, which were treated together, the occurrence of development disorders remained as low level as in the case individual treatments. Looking at the types of development disorders, the most frequent problems were the oedema, the shortening of the beak mandible, and the incorrect posture of the feet and the neck. As the collective result of the treatment with cadmium sulphate and Dual Gold 960 EC, the body mass of the treated embryos decreased significantly in comparison with the control and individually treated groups, therefore we can conclude that the combined treatment resulted in increased embryo toxic effect in comparison with the individual embryo damaging effect of the used components.  相似文献

Theory predicts shorter embryonic periods in species with greater embryo mortality risk and smaller body size. Field studies of 80 passerine species on three continents yielded data that largely conflicted with theory; incubation (embryonic) periods were longer rather than shorter in smaller species, and egg (embryo) mortality risk explained some variation within regions, but did not explain larger differences in incubation periods among geographic regions. Incubation behavior of parents seems to explain these discrepancies. Bird embryos are effectively ectothermic and depend on warmth provided by parents sitting on the eggs to attain proper temperatures for development. Parents of smaller species, plus tropical and southern hemisphere species, commonly exhibited lower nest attentiveness (percent of time spent on the nest incubating) than larger and northern hemisphere species. Lower nest attentiveness produced cooler minimum and average embryonic temperatures that were correlated with longer incubation periods independent of nest predation risk or body size. We experimentally tested this correlation by swapping eggs of species with cool incubation temperatures with eggs of species with warm incubation temperatures and similar egg mass. Incubation periods changed (shortened or lengthened) as expected and verified the importance of egg temperature on development rate. Slower development resulting from cooler temperatures may simply be a cost imposed on embryos by parents and may not enhance offspring quality. At the same time, incubation periods of transferred eggs did not match host species and reflect intrinsic differences among species that may result from nest predation and other selection pressures. Thus, geographic variation in embryonic development may reflect more complex interactions than previously recognized.  相似文献

Hatching failure is widespread in birds, and is usually the result of embryo death rather than infertility. Embryo death can result from both intrinsic and extrinsic factors, some of which may vary across the developmental period. Determining the point at which an embryo died during development may therefore help us to understand the underlying cause of death. Here we describe simple criteria that can be used by field ornithologists to establish the developmental stage of dead embryos found in unhatched passerine eggs, and explain how this can be used to estimate the date of embryo death. We compared the pattern of embryo development over the incubation period for three species, the Zebra Finch Taeniopygia guttata, Blue Tit Cyanistes caeruleus and Great Tit Parus major. We also compared rates of Zebra Finch embryo development under artificial and standard (parental) incubation. Embryo development rates were remarkably similar across the three species and between Zebra Finch embryos under artificial and natural incubation conditions. We therefore suggest that the pattern of embryo development in the Zebra Finch may provide a model for other small passerines with similar incubation periods, but acknowledge that further interspecific comparisons are required before this model is considered more widely applicable. By estimating embryo death dates using our approach, ornithologists will be able to determine temporal patterns of embryo mortality in relation to extrinsic environmental conditions. This approach may shed light on how extrinsic factors such as climate and parental behaviour influence embryo survival in wild birds.  相似文献

Many organisms invariably produce one offspring per reproductive bout, but experimental tests of adaptive explanations for this reproductive pattern are rare. To address this issue, we studied a lizard (Anolis sagrei) that produces one egg at a time to test the hypothesis that solitary incubation (due to single-egg clutches) eliminates competition with adjacent eggs for moisture and thus enhances offspring quality via increased egg water uptake during development. Our findings suggest that solitary incubation does not affect rates of moisture uptake by eggs or offspring size. However, egg moisture uptake and offspring size were negatively affected when eggs were adjacent to an egg that died during development. Depending on rates of infertile eggs or embryo mortality in the field, single-egg clutches may improve developmental environments and enhance offspring fitness. These results highlight the importance of considering the role of plastic embryonic responses during development in explaining reproductive patterns.  相似文献

The ontogeny of brain creatine kinase (CK) was studied during chick embryo development. The cytosolic activity increased 270% in 10 h from the 2nd to the 3rd days of incubation; this was followed by a plateau phase throughout development and at the end of incubation there appeared to be another increase of cytosolic and mitochondrial CK activities. Therefore, early embryonic chick brain CK is another‘constitutive’enzyme like the early embryonic chick heart CK since creatine has not been enzymatically detected in the embryo until day 4 of incubation. Insulin does not appear to stimulate the early increase of brain CK activity since the hormone is not present in the embryo until day 5 of incubation. It is likely that CK increase is associated with neuronal multiplication at early stages and possibly to neuronal maturation before hatching.  相似文献

The phenomenon of sudden mortality in the chick embryo, induced by the exogenous neurotransmitters norepinephrine, epinephrine, dopamine, serotonin, carbachol, and by the beta-blockers propranolol and atenolol, is described. When introduced singly into eggs (albumen) in doses of 0.1 mg per egg, these substances induced highly significant, sharply increased mortality (60% to 100%) when the introduction was on embryonic (incubation) days 8 to 12 (hereafter called E8 and E12), but none if introduction was earlier. These ages follow the appearance of functional receptors (or receptor-effectors) for these substances. However, the involvement of receptors here has not been proved. Solvent alone (Ringer's solution for chick) had no effect, indicating that the procedures used were in themselves not lethal. Possibly, the above neurotransmitters became lethal to the embryo because they were introduced in excess of the amounts which were produced and needed at that time by the embryo for optimal development.  相似文献

Hyperthermia-induced oxidative stress is one of the mechanisms suggested to underlie loss of developmental competence in mouse embryos. In this study, we examined whether pretreatment with the antioxidant epigallocatechin gallate (EGCG) can alleviate the negative effects of hyperthermia on developmental competence of the ovarian pool of oocytes and improve embryonic development. Female mice (CB6F1) were synchronized (eCG+hCG) and injected with 0.4ml EGCG (100mg/kg body weight) or with saline. Both EGCG- and saline-treated mice were exposed to heat stress (HS; 40 degrees C, 65% RH) or kept under normothermal conditions (Control; 22 degrees C, 45% RH). In vivo-derived zygotes were recovered 20h after hCG administration and cultured in vitro. Maternal hyperthermia attenuated embryonic cleavage rate in association with further disruption in embryonic early cleavage and subsequently, with embryonic development. While pretreatment with EGCG did not affect the proportion of zygotes that cleaved to the two-cell stage, it appeared to moderate the effect of hyperthermia on both cleavage timing and developmental rate, as reflected by an increased rate of early cleaved embryos and blastocyst formation. Blastocyst developmental competence was also improved, as indicated by the increased total cell number and percentage of embryos that underwent hatching, in association with reduced apoptotic status, as reflected by the percentage of TUNEL-positive cells and intensity of caspase activity for the HS-EGCG embryos vs. HS-saline ones. In summary, while hyperthermia disrupts the competence of the follicle-enclosed oocyte, in vivo administration of the antioxidant EGCG improves developmental competence and the quality of the embryos that develop from these oocytes.  相似文献

It has been suggested that fungal presence on sea turtle eggs may impede gas exchange. To investigate the influence of diminished gas exchange surface upon embryo survivorship, flatback (Natator depressus) and green (Chelonia mydas) eggs were painted with petroleum jelly. Variable proportions of the egg surface were covered, including both respiratory and nonrespiratory domains. Embryo survival varied with site inhibited, proportion of eggshell affected, and species of turtle. If fungi on the exterior of the eggshell are able to impede respiratory gas exchange, their presence on the upper hemisphere (primary gas exchange area in early incubation) will result in the highest embryo mortality. Large eggs are likely to demonstrate a higher survivorship than small eggs, due to their larger available respiratory area and/or to variation in weight or stage-specific embryonic metabolic demands. Interspecific differences in egg size may therefore be a contributory factor to observed mortality rate differences in the natural presence of fungi.  相似文献

The early development of sunbleak Leucaspius delineatus , from activation of ovum to juvenile, was recorded using both living and preserved materials. Five embryonic steps, one free embryo and five larval steps were identified. Hatching occurred between 73 h 25 min and 85 h 25 min (post‐activation); the free embryo period was extremely short in duration (3–16 h). The transition from larva to juvenile occurred in c . 96 days when the fish were fully scaled and the lateral line organ clearly visible. During early development, developmental thresholds were identified between steps, which suggested possible switches in ecology, microhabitat, behaviour, performance or any combination of these. During both embryo and larva periods of development, steps increased in time duration, indicating different rates of complexity of physiological and morphological developments within each step. From the results, a 'step like' model is proposed for the early development of sunbleak.  相似文献

The pregastrula stage mammalian conceptus consists of both embryonic and non-embryonic components. The latter forms the bulk of the tissues, provides nutrition for the developing embryo and also contributes developmental signals that influence events within the embryo itself. Understanding the origins and relationships between the embryonic and extraembryonic cell lineages is thus central to understanding development in mammals. Despite the apparent gross differences in early developmental strategy and form, the conceptuses of eutherian, marsupial and monotreme mammals show some remarkable similarities in the lineage allocation to trophoblast and hypoblast and in the emergent properties of the two cell types. We suggest that the gross differences can be explained by two relatively small evolutionary timing changes affecting cell adhesion patterns and the polarisation of developmentally significant information. These changes result in the conversion of a unilaminar blastocyst to a morula form composed of blastomeres with increased regulatory capacity.  相似文献

Two experiments were conducted to study the sensitivity of developing chicken embryos to various solvents used as vehicles and their effect on hatchability. No significant differences on embryonic mortality were observed between the sham-injected control and corn oil-injected groups. Acetone, ethylene glycol, and ethanol (0.10 ml/egg) significantly reduced the percentage hatchability and showed a high embryonic mortality during the first week of incubation. Levels of cottonseed oil (0.05-0.10 ml) and propylene glycol (0.05-0.15 ml) were well tolerated by the developing embryo but were slightly inferior to corn oil. Levels of 0.05-0.15 ml of corn oil are suitable vehicles for fat soluble compounds in studies involving the injection of eggs. Higher levels of solvent can be injected at later incubation periods. The choice of the best solvent must be made depending upon (1) solubility of test material, (2) amount of test material needed, (3) toxicity of the solvent at the appropriate levels, (4) route of administration, and (5) stage of development.  相似文献

Mean heart rate (MHR) was determined during incubation and in hatchlings of 14 altricial avian species to investigate (1) if there is a common developmental pattern of heart rate in altricial embryos and (2) if heart rate changes during incubation are correlated with changes in embryonic growth rate. On the basis of normalized incubation MHR increased approximately linearly in 12 of 14 species from as early as 30-40% of incubation to that of pipped embryos. The MHR of hatchlings was equal to or higher than that of pipped embryos in seven species. Passerine embryos and hatchlings maintained higher MHR in comparison to parrots of similar egg mass, which may reflect phylogenetic differences in development. Embryonic MHR increased at a higher rate while embryonic growth rates were highest during the first 40% of incubation in tit, budgerigar and crow embryos than during subsequent development when relative growth rates decreased. MHR became independent of yolk-free wet mass at a smaller fraction of hatchling mass in budgerigar and crow than in the tit, suggesting that MHR is more likely to increase continuously after 40% of incubation in small altricial species than larger species.  相似文献

In some species of ectotherms, temperature has little or no effect on the amount of energy expended during embryonic development. This phenomenon can result from either of two mechanisms: (1) a shorter incubation period at higher temperatures, which offsets the expected increase in metabolic rate, or (2) a compensatory decrease in the rate at which embryos expend energy for maintenance. To distinguish the relative importance of these two mechanisms, we quantified the acute and chronic effects of temperature on embryonic metabolism in the eastern fence lizard (Sceloporus undulatus). First, we measured metabolic rates of individual embryos at 27 degrees, 31 degrees, and 34 degrees C. Second, we examined the capacity for thermal acclimation by measuring the metabolic rates of embryos at 30 degrees C, after a period of incubation at either 28 degrees or 32 degrees C. As with adult reptiles, the metabolic rates of embryos increased with an acute increase in temperature; the Q(10) of metabolic rate from 27 degrees to 34 degrees C was 2.1 (+/-0.2). No evidence of thermal acclimation was observed either early or late in development. In S. undulatus, a shorter incubation period at higher temperatures appears to play the primary role in canalizing the energy budget of an embryo, but a reduction in the cost of growth could play a secondary role.  相似文献

We investigated the effect of sand moisture content and sand temperature on developmental success and the mobilization of calcium during development using laboratory incubated eggs (n=251) collected from leatherbacks nesting at Parque Nacional Marino Las Baulas, Costa Rica. Calcium concentrations of egg components [eggshell, yolk plus albumen (Y+A) and embryo] changed significantly through incubation for both viable and undeveloped eggs. In developed eggs, eggshell calcium content decreased 42.9% by day 60 of incubation. The Y+A calcium decreased by 20.8% until the last quarter of incubation, and then increased to 0.99% above initial Y+A calcium concentrations just prior to hatching. In undeveloped eggs, eggshell calcium content decreased by 25.7%, with the rate of decrease slowing significantly beyond day 30 of incubation. In contrast, Y+A calcium increased steadily through the 60-day incubation period. Embryos incorporated a higher proportion of calcium when incubated at a lower sand moisture content (5% H(2)O>12% H(2)O) and at lower sand temperatures (28.5 degrees C, 29.5 degrees C>31.0 degrees C). The total wet mass of freshly oviposited eggs was negatively correlated with calcium concentration per gram of eggshell (r=-0.569; P<0.001). Thus, each yolked egg, regardless of initial wet mass, had an average of 1.23 g (+/-0.43 g) of calcium per egg (Mean egg mass: 76.24+/-1.21 g). Both developmental success (24.1%) and hatching success (7.4%) of laboratory-incubated eggs were dependent to a greater extent on temperature than on moisture, with an increase in mortality as sand temperature increased. For natural nests on Playa Grande, developmental success (37.4%) and hatching success (19.8%) were similar in magnitude to the results obtained from the laboratory. The recent ENSO (El Ni?o Southern Oscillation) event and increased tidal activity may be responsible for the high embryonic mortality measured during the 1997-1998 nesting season.  相似文献

The cardiovascular system is the first system to become functional in a developing animal and must perform key physiological functions even as it develops and grows. The ontogeny of cardiac physiology was studied throughout embryonic and larval developmental stages in the red swamp crayfish Procambarus clarkii using videomicroscopic dimensional analysis. The heart begins to contract by day 13 of development (at 25 degrees C, 20 kPa O(2)). Cardiac output is primarily regulated by changes in heart rate because stroke volume remains relatively constant throughout embryogenesis. Prior to eclosion, heart rate and cardiac output decreased significantly. Previous data suggest that the decrease in cardiac parameters prior to hatching may be due to an oxygen limitation to the embryo. Throughout development, metabolizing mass and embryonic oxygen consumption increased, while egg surface area remained constant. The surface area of the egg membrane is a constraint on gas exchange; this limitation, in combination with the increasing oxygen demand of the embryo, results in an inadequate diffusive supply of oxygen to developing tissues. To determine if the decrease in cardiac function was the result of an internal hypoxia experienced during late embryonic development, early and late-stage embryos were exposed to hyperoxic water (PO(2) = 40 kPa O(2)). Heart rate in late-stage embryos exposed to hyperoxic water increased significantly over control values, which suggests that the suppression in cardiac function observed in late-stage embryos is due to a limited oxygen supply.  相似文献

The abnormalities of early post‐implantation embryos can lead to early pregnancy loss and many other syndromes. However, it is hard to study embryos after implantation due to the limited accessibility. The success of embryo culture in vitro can avoid the challenges of embryonic development in vivo and provide a powerful research platform for research in developmental biology. The biophysical and chemical cues of the microenvironments impart significant spatiotemporal effects on embryonic development. Here, we summarize the main strategies which enable researchers to grow embryos outside of the body while overcoming the implantation barrier, highlight the roles of engineered microenvironments in regulating early embryonic development, and finally discuss the future challenges and new insights of early embryo culture.  相似文献
