Preparations of density gradient-purified infectious bursal disease virus (IBDV) were found to contain full and empty icosahedral virions, type I tubules with a diameter of about 60 nm, and type II tubules 24 to 26 nm in diameter. By immunoelectron microscopy we demonstrate that virions and both types of tubular structures specifically react with anti-IBDV serum. In infected cells intracytoplasmic and intranuclear type II tubules reacted exclusively with an anti-VP4 monoclonal antibody, as did type II tubules in virion preparations. The immunofluorescence pattern with the anti-VP4 antibody correlated with electron microscopical findings. Neither purified extracellular nor intracellular virions were labeled with the anti-VP4 MAb. Our data show that the type II tubules contain VP4 and suggest that VP4 is not part of the virus particle.  相似文献

Summary Intranuclear and cytoplasmic fibrous inclusions and cytoplasmic tubular inclusions have been studied using electron microscope techniques. The fibrous inclusions are composed of closely packed, rod-like structures. Each rod has an outside diameter of ± 24 nm, a hollow centre and lateral projections along its length. The tubular inclusions occur as densely packed bundles or loose arrays of 10–13 nm diameter tubules which are composed of subunits arranged in a double helical structure. The distribution, origin and possible function of these and similar inclusions is discussed.  相似文献

Summary The matrix of the mitochondria of Porterioochromonas stipitata Lewin (Chrysophyceae) often contains crystalloid inclusion bodies that are composed of hexagonally packed fibrils (or tubules). The fibrils have a diameter of 10 nm, their center-to-center spacing is 13,5 nm.  相似文献

Hexagonal or angular crystalline inclusions in Pleonosporium (Naeg.) Hauck vegetative cells were examined using electron microscopy. Ultrastructural analysis reveals that the inclusions initially contain tubular elements resembling microtubules but, with continued differentiation, are transformed into rod containing crystals. The tubular structures initially measure 25 nm in diameter. Scattered tubules become arranged in a parallel and alternate pattern and undergo subsequent enlargement to approximately 29 nm. Following enlargement, each tubule apparently disaggregates into rods that form a crystal having hexagonally arranged rod-like subunits. It is suggested that these tubules may represent microtubules and the resultant crystals are composed of tubulin.  相似文献

Various cytopathological structures, known as inclusion bodies, are formed upon infection of cultured leafhopper cells by Rice dwarf virus, a member of the family Reoviridae. These structures include tubules of approximately 85 nm in diameter which are composed of the nonstructural viral protein Pns10 and contain viral particles. Such tubular structures were produced in heterologous non-host insect cells that expressed Pns10 of the virus. These tubules, when associated with actin-based filopodia, were able to protrude from the surface of cells and to penetrate neighboring cells. A binding assay in vitro revealed the specific binding of Pns10 to actin. Infection of clusters of cells was readily apparent 5 days after inoculation at a low multiplicity of infection with the virus, even in the presence of neutralizing antibodies. However, treatment of host cells with drugs that inhibited the elongation of actin filaments abolished the extension of Pns10 tubules from the surface of cells, with a significant simultaneous decrease in the extent of infection of neighboring cells. These results together revealed a previously undescribed aspect of the intercellular spread of Rice dwarf virus, wherein the virus exploits tubules composed of a nonstructural viral protein and actin-based filopodia to move into neighboring cells.  相似文献

Intact chlorosomes of Chlorobium tepidum were embedded in amorphous ice layers and examined by cryo-electron microscopy to study the long-range organization of bacteriochlorophyll (BChl) layers. End-on views reveal that chlorosomes are composed of several multi-layer tubules of variable diameter (20-30 nm) with some locally undulating non-tubular lamellae in between. The multi-layered tubular structures are more regular and larger in a C. tepidum mutant that only synthesizes [8-ethyl, 12-methyl]-BChl d. Our data show that wild-type C. tepidum chlorosomes do not have a highly regular, long-range BChl c layer organization and that they contain several multi-layered tubules rather than single-layer tubules or exclusively undulating lamellae as previously proposed.  相似文献

Summary Vacuoles of differentiating mesophyll cells of Tamarix aphylla contain an amorphous electron-dense material in which stacks of parallel aligned striations are embedded. Cross-sections of the striations disclosed that they represent profiles of longitudinally sectioned bundles of tubules (tubule outer diameter 9.0 nm, tubule wall thickness 1.8 nm). In advanced mesophyll cell development, the amorphous vacuolar material disappears, whereas the bundles of tubules turn into bundles of double helices (double helix diameter 14.5 nm). Cytochemical treatment of mesophyll cells with the enzymes pepsin and trypsin has revealed that both the bundles of tubules/double helices and the embedding material are constituted of protein. The possible functional role of the vacuolar inclusions is discussed.  相似文献

Unusual regions of densely packed membranous tubules known as tubular aggregates (TAs) have been observed in skeletal muscle fibres of mammals under numerous pathological conditions but also in health. Their causality is unclear. It is neither known whether TAs are destructive and should be treated or whether they have a compensating function in an endangered muscle. In spite of many similarities, the histochemical, immunocytochemical and ultrastructural characteristics of tubular aggregates do vary. Histochemistry provided an overall characteristic of TAs as membranous inclusions with a variety of enzymatic activities. Immunocytochemical evidence revealed that tubular aggregates contain miscellaneous proteins and that derive from membranes of sarcoplasmic reticulum and mitochondria. No evidence for the presence of contractile and cytoskeletal proteins in TAs was found. Ultrastructurally, TAs are characterized as more or less densely packed aggregates of vesicular or tubular membranes of variable forms and sizes that may contain amorphous material, filaments or inner tubules. Various reported types of tubular aggregates, namely, proliferating terminal cisterns, vesicular membrane collections, TAs with double-walled tubules, TAs with single-walled tubules, aggregates of dilated tubules with inner tubules, aggregates of tubulo-filamentous structures, filamentous tubules, riesentubuli, and related membranous structures including cylindrical spirals are sumarized and analyzed here in detail.  相似文献

The seminiferous tubules and the excurrent ducts of the mammalian testis are physiologically separated from the mesenchymal tissues and the blood and lymph system by a special structural barrier to paracellular translocations of molecules and particles: the “blood–testis barrier”, formed by junctions connecting Sertoli cells with each other and with spermatogonial cells. In combined biochemical as well as light and electron microscopical studies we systematically determine the molecules located in the adhering junctions of adult mammalian (human, bovine, porcine, murine, i.e., rat and mouse) testis. We show that the seminiferous epithelium does not contain desmosomes, or “desmosome-like” junctions, nor any of the desmosome-specific marker molecules and that the adhering junctions of tubules and ductules are fundamentally different. While the ductules contain classical epithelial cell layers with E-cadherin-based adherens junctions (AJs) and typical desmosomes, the Sertoli cells of the tubules lack desmosomes and “desmosome-like” junctions but are connected by morphologically different forms of AJs. These junctions are based on N-cadherin anchored in cytoplasmic plaques, which in some subforms appear thick and dense but in other subforms contain only scarce and loosely arranged plaque structures formed by α- and β-catenin, proteins p120, p0071 and plakoglobin, together with a member of the striatin family and also, in rodents, the proteins ZO-1 and myozap. These N-cadherin-based AJs also include two novel types of junctions: the “areae adhaerentes”, i.e., variously-sized, often very large cell-cell contacts and small sieve-plate-like AJs perforated by cytoplasm-to-cytoplasm channels of 5–7 nm internal diameter (“cribelliform junctions”). We emphasize the unique character of this epithelium that totally lacks major epithelial marker molecules and structures such as keratin filaments and desmosomal elements as well as EpCAM- and PERP-containing junctions. The epithelium of the Malpighian tubules in the housefly is comprised of four distinct cellular types. Type I cells are characterized by the presence of intimate associations between infoldings of basal plasma membrane and mitochondria. On the luminal surface, cytoplasm is extended into microvilli which contain mitochondria. Membrane-bound vacuoles in the cytoplasm seem to progressively accumulate granular material. Type II cells have dilated canaliculi. Microvilli lack mitochondria. The Type III cell has not been reported previously in Malpighian tubules. It has very well-developed granular endoplasmic reticulum which contains intracisternal bundles of tubules. Cytoplasm contains numerous electron dense bodies. Type IV cells occur in the common duct region of the Malpighian tubules. Mitochondria do not extend into the microvilli.  相似文献

Summary The fine structure of chromoplasts in epidermal cells of flower petals of Tropaeolum has been investigated by light, polarizing, and electron microscopy at different stages of development. The pale greenish-yellow petals still enclosed in the bud contain barely differentiated chloroplasts with few, irregular grana. The chromoplasts of unfolding petals show differently oriented bundles of tubules with variable diameters (mean: 17 nm). Thylakoid membranes become reduced more and more. The tubular bundles are intermingled with numerous isodiametric bodies of ca. 50 nm diameter; these bodies are better discernible at later stages when the chromoplasts possess a less dense matrix. The chromoplasts of open flowers are in a state of disorganization at a time when the cytoplasm still appears normal. A comparison is made between chromoplast tubules and tubular structures described from other kinds of plastids. Type 1 cells of the clitellar epithelium of Eisenia foetida secrete a protein resembling keratin in histochemical reaction. Ultrastructurally, type 1 cells are characterized by membrane-bound, pockmarked granules ranging in diameter from 1.0 to 3.0 micrometers. Immature granules often exhibit an organized microfibrillar substructure. Individual microfibrils are 17 +/- 1 nm in diameter. Type 2 cell secretion contains protein, nonsulfated acid mucosubstance, and neutral mucosubstance with 1,2-glycol groups. The coarsely fibrillar granules are membrane bound and vary in diameter from 0.5 to 1.0 micrometer. The necks of both type 1 and type 2 cells contain a peripheral ring of microtubules, 20 +/- 1 nm in diameter.  相似文献

The freeze-fracture appearance of rat stomach and liver gap junctions changes after uncoupling procedures such as inhibition of the metabolism of perfusion with hypertonic sucrose. In control stomach, either fixed immediately or kept for 1 h in a well-oxygenated Tyrode's solution at 37 degrees C, most gap junctions between mucous cells contain particles irregularly packed at an average center-to-center spacing of 10.3-10.5 nm. After 1-h treatment with 2,4-dinitrophenol (DNP), at the same temperature and oxygenation, most particles aggregate hexagonally at an average spacing of approximately 8.5 nm. Similar changes are seen in hypoxic specimens. In control liver, fixed by perfusion, most junctional particles are irregularly packed at an average center-to-center spacing of approximately 10 mm. Small areas of fairly regular hexagonal packing are occasionally seen, where the average particle spacing is 9.2-9.5 nm. In hypoxic liver, the junctional particles form regular hexagonal packings in which the average center-to-center particle spacing is approximately 8.5 nm. In liver perfused with hypertonic sucrose-calcium solutions, following EDTA solutions, most junctions are pulled apart. The separated junctional membranes, expected to be highly impermeable, contain particles regularly and tightly packed as in hypoxic or DNP-treated junctions. Preliminary measurements indicate also a possible change in particle diameter, from approximately 8.6 nm (control) to approximately 7.7 nm (treated). The structural changes are similar to those previously reported in crayfish and may reflect conformational changes in particle subunits resulting in functional uncoupling.  相似文献

Somatotropes, lactotropes and corticotropes of adult male mice were identified with immunohistochemistry in the adenohypophysis fixed by OsO4 alone. Somatotropes were classified into type I somatotropes that contain large (350 nm in diameter) round secretory granules and type II somatotropes that contain small (100-200 nm in diameter) round secretory granules. Most somatotropes were type I somatotropes. Lactotropes were also classified into type I lactotropes that contain irregularly shaped secretory granules and type II lactotropes containing small (100-200 nm in diameter) round secretory granules. Corticotropes are irregular stellate or slender cells with little cytoplasm. They contain round solid secretory granules in various densities along the cell periphery. Most of these are low-density granules (200-300 nm in diameter) and a few are high-density granules (200-250 nm in diameter). These data were compared with the classical data of mouse adenohypophysial cells that were fixed in OsO4 alone and identified only by conventional electron microscopy.  相似文献

Ripe, deep-red fruits of Palisota barteri contain tubulous chromoplasts which develop from unpigmented leucoplasts. These plastids contain, besides large spherical inclusion bodies, numerous osmiophilic globules which, in the course of ripening, frequently show transition states to tubular structures. The tubules contain, in all stages of their development, acylated -citraurin, which is also the main pigment of Citrus fruits. The tubular structures have been isolated, fragmented by French-pressure treatment, and separated into three fractions on sucrose gradients. The lightest fraction (1.044 g·cm-3) contained thick fragments (saccules) with diameters of 50–60 nm, whereas the heaviest (1.083 g·cm-3) consisted of tubules 20–25 nm in diameter. The relative amounts of polar lipids, proteins, and carotenoids of the different fractions are consistent with a molecular structure model of tubules and saccules, according to which a wick of longitudinally oriented carotenoid molecules of variable thickness is coated by a monolayer of polar lipids and proteins. High-resolution negative-stainings showed the surface of the tubules to be covered with globular particles of about 6 nm diameter. Numerous tubular structures were observed in the surface region of smooth muscle cells making up the vascular walls in the lamprey, Lampetra japonica; they were designated as surface tubules. The limiting membrane of the surface tubules was connected to the plasma membrane, allowing communication of the lumen of the tubule with the extracellular space. Tannic acid reacted with osmium, serving as an extracellular marker, penetrated into the tubules but not into the intracellular organelles, such as the endoplasmic reticulum and the Golgi complex. The surface tubules were grouped in longitudinal parallel rows, separated from each other by tubule-free areas where dense plaques were present. Each tubule was fairly cylindrical (approximately 60 nm in diameter) and often ramified into two or three branches with a blind end. Occasionally, these tubules were encircled by the sarcoplasmic reticulum which was located immediately beneath the plasma membrane. Similar tubules were also observed in the surface region of vascular endothelial cells and fibroblasts in the adventitial connective tissue. The possibility that the surface tubules in the present observations are analogous to the smooth muscle caveolae or the striated muscle T-tubule is discussed.  相似文献

The interstitial tissue of the opossum testis includes interstitial or Leydig cells, macrophages, and small cells which morphologically resemble mesenchymal cells. The latter are thought to give rise to mature interstitial cells. The most prominent feature of the interstitial cell cytoplasm is an exceedingly abundant agranular endoplasmic reticulum. This reticulum is generally in the form of a meshwork of interconnected tubules about 300 to 450 A in diameter, but occasionally it assumes the form of flattened, fenestrated cisternae resembling those of pancreatic acinar cells, except for the lack of ribonucleoprotein particles on the surface of the membranes. The interstitial cells vary considerably in their cytoplasmic density. The majority are quite light, but some appear extremely dense, and in addition usually have a more irregular cell surface, with numerous small pseudopodia. These differences may well reflect variations in physiological state. Cytoplasmic structures previously interpreted as "crystalloids" consist of long bundles of minute parallel tubules, each about 180 A in diameter, which seem to be local differentiations of the endoplasmic reticulum. The mitochondria are rod-shaped, and contain a moderately complex internal membrane structure, and also occasional large inclusions that are spherical and homogeneous. The prominent juxtanuclear Golgi complex contains closely packed flattened sacs and small vesicles. The results of the present study, coupled with biochemical evidence from other laboratories, make it seem highly probable that the agranular endoplasmic reticulum is involved in the synthesis of the steroid hormones produced by the interstitial cell. This finding therefore constitutes one of the first functions of the agranular reticulum for which there is good morphological and biochemical evidence.  相似文献

Antennal olfactory hairs of Antheraea polyphemus were investigated by means of transmission electron microscopy. Adequate preservation of dendrites and extracellular pore tubules is obtained by mechanical opening of the hair lumen and subsequent chemical fixation. The dendritic membrane has a cell coat. The dendrites contain microfilamentous structures in addition to their cytoplasmatic microtubules. The extracellular pore tubules traverse the hair cuticle and reach into the hair lumen for maximally 350 nm. Their diameter varies between 20 and 40 nm, depending on the preparation method. They consist of an electron-dense wall and an electron-lucent core. The wall has a helical substructure and is covered with a fuzzy coat. Contacts of pore tubules and dendritic membranes occur wherever dendrites are near the inner surface of the hair cuticle. Some of the pore tubules terminate approximately at right angles on the dendritic membrane, others lie against the membrane. The contact seems to be made via the surface coats of the tubules and the membrane. The structure of pore tubules which had been negatively stained with uranyl acetate is similar to the conventionally thin-sectioned material. The observations provide support for earlier assumptions that pore tubules are the pathways by which odor molecules reach the dendritic membrane.  相似文献

电镜超微结构观察和免疫金标记显示:受蚕豆萎蔫病毒2号(Broad bean wilt virus 2,BBWV 2)中国分离物B935侵染的豌豆(Pisum sativum)和蚕豆(Vicia faba)叶细胞中膜结构增生,形成膜结构增生区,病毒以结晶体和管状体形式存在于细胞质中。在病变早期,叶肉细胞的胞间连丝处连接有小管结构,病毒样颗粒呈纵列排在小管中,穿越胞间连丝的小管能被BBWV 2的金标记抗体特异性标记。维管束组织的薄壁细胞、伴胞及转移细胞内存在膜增生区及病毒管状体,在筛管壁附近存在的病毒样颗粒能被BBWV 2金标记抗体特异性标记。实验结果表明BBWV 2胞间运动形式与豇豆花叶病毒(CPMV)相似,以完整粒子通过在胞间连丝处形成的小管结构穿越胞间连丝;细胞质中存在的直径160nm管状体只是一种病毒聚集体,与胞间运动无直接关系;该病毒在筛管中可能也是以完整粒子形式进行长距离转运的。  相似文献
