The function of distyly as a device promoting inter-morph pollination was proposed by Darwin over a century ago, though only recently have experimental studies been undertaken to test this hypothesis. These studies are unrealistic since they assume regular spatial distribution of morphs within a population and fail to consider intra-flower pollination in assessing distyly's effects. Pollen flow in two populations of Linum tenuifolium L. was investigated using a fluorescent-dye marker technique. One population was dimorphic and self-incompatible; the other was monomorphic and self-compatible. In both populations pollen dispersal was leptokurtic with over 75% of grains detected within 5 m from the source. Intra-flower pollinations comprised a large proportion of individual stigma loads (42% for the dimorphic form; 49% for the monomorphic form). Total stigmatic loads for the dimorphic population showed equal proportions of pin and thrum pollen on both long-style and short-style stigmas, though variation in individual loads was marked. Values ranging from 34.2 to 61.5% legitimate pollen were recorded. The irregular distribution of morphs observed at the dimorphic site will reduce the efficiency of heterostyly as a device promoting intermorph pollination. The reduction in intra-flower pollination suggests adaptive significance of this floral dimorphism in increasing the efficiency of pollen dispersal to the individual.  相似文献

A combined HPLC-UV/PAD and HPLC-ESI/MS method allowing the fast detection and identification/structural characterisation of lignans of different structural subclasses is described. Twenty-four lignans of different skeletal types were analysed and the combined information derived from their UV and ESI/MS spectra led to the identification of group characteristics that can be used to establish the structure of unknown lignans in plant samples. This method was successfully applied to the identification of lignans in crude extracts of Linum usitatissimum L. and L. bienne Mill.  相似文献

Recent advances in next-generation DNA sequencing (NGS) have enhanced the development of genomic resources such as contigs or single-nucleotide polymorphisms (SNPs) for evolutionary studies of a nonmodel species with a complex and unsequenced genome. This study presents an application of a NGS technique in combination with genomic reduction and advanced bioinformatics tools to identify contigs and SNPs from multiple samples of two Linum species. A full Roche 454 GS FLX run of 16 diverse Linum samples representing cultivated flax (Linum usitatissimum L.) and its wild progenitor (Linum bienne Mill.) generated approximately 1.6 million sequence reads with a total length of 498 Mbp. Application of the computational pipeline de novo identification of alleles identified 713 contigs and 1067 SNPs. A blast search revealed alignments of all 713 contigs with 491 existing Linum scaffolds and gene annotations associated with 512 contigs. Sanger sequencing confirmed 95% of 79 selected contigs and 94% of 272 SNPs and identified 211 new SNPs and 19 new indels. The scored 454 SNP data were highly imbalanced for assayed samples. These findings not only are useful for evolutionary studies of Linum species but also help to illustrate the utility of NGS technologies in SNP discovery for nonmodel organisms.  相似文献

A new species, Linum vuralianum Yılmaz & Kaynak, is described and illustrated from west Anatolia, Turkey. It is closest to L. flavum L., but differs by its annual or biennial life form, the presence of rosette leaves at anthesis, and the absence of staminodes. The World Conservation Union (IUCN) threat category and observations on the population are noted. Root branching patterns in intact and decapitated flax (Linum usitatissimumL.) roots were compared. The number of initiated primordia in the control and decapitated roots was similar, but decapitated roots produced an increased number of lateral roots owing to an increase in the number of primordia developed into the laterals. It is suggested that the apical meristem influences lateral root development only at the stage of root emergence from the parent root.  相似文献

对百里香(Thymus serpyllum)、平车前(Plantago depressa)、盐生车前(P.maritima)、野亚麻(Linum stelleroides)进行了粘液繁殖体粘液情况比较.以在水浸和浇水条件下植物种子粘沙量的多少衡量粘液量.结果表明,对于不同浸水时间处理,野亚麻和平车前未表现明显差异,盐生车前和百里香有随浸泡时间加长而粘液溶出量增多的趋势.对于不同浇水量处理,4种植物均有随浇水量增多而粘液增多的倾向.浸泡80min后,盐生车前种子的粘液粘沙使重量达原重的60多倍,平车前种子达原重的10倍左右,百里香和野亚麻种子达原重的4~6倍左右.浇水8mm后,盐生车前种子的粘液粘沙使重量达原重的20多倍,平车前种子达原重的6~10倍左右,百里香和野亚麻种子达原重的2~7倍左右.将各种处理平均,得到各种植物粘沙种子百分率为:野亚麻67.7%,百里香94.5%,平车前97.7%,盐生车前99.5%.  相似文献

【目的】从健康亚麻植株的根际土壤中筛选对亚麻立枯病菌具有较强抑菌作用的拮抗菌,优化其产生抑菌活性物质的发酵条件,为其生防利用奠定基础。【方法】采用稀释平板涂布法和对峙培养法进行拮抗菌的筛选;根据菌株形态学特征、生理生化特性以及16S r RNA基因序列分析对其进行鉴定;利用温室抗病实验确定其生防效果;通过单因素实验和均匀设计实验优化其发酵条件。【结果】分离筛选到一株对亚麻立枯病菌具有显著拮抗作用的细菌HXP-5,且其对另外7种植物病菌真菌均有拮抗作用;鉴定菌株HXP-5为枯草芽孢杆菌;温室抗病实验结果表明其生防效果可达71.22%;其产生抑菌活性物质的最佳发酵条件为:葡萄糖为2.3%,胰蛋白胨+酵母粉(3:1)为0.25%,Na Cl为0.18%,发酵时间为72 h,发酵温度为27°C,转速为210 r/min,250 m L摇瓶装液100 m L,接种量为1.7%。【结论】经鉴定,对亚麻立枯病病菌具拮抗作用的菌株HXP-5为枯草芽孢杆菌,且对亚麻立枯病具有较强的防治效果,发酵条件进行优化后其对亚麻立枯病病原菌显示出更强的拮抗作用。  相似文献

Recently it was found that cell cultures and plants of Linum species contain lignans of various chemical structures. The stereochemistry of these compounds differ among species. Cell cultures of L. album accumulate (-)-podophyllotoxin together with pure (-)-secoisolariciresinol. The presence of both enantiomers of the precursor pinoresinol indicates that in L. album cell cultures the reactions from pinoresinol to secoisolariciresinol are the first steps determining enantiospecificity in biosynthesis of podophyllotoxin. Seeds of L. usitatissimum contain almost enantiomerically pure (+)-secoisolariciresinoldiglucosid derived from (+)-secoisolariciresinol. A cell culture of this species contains a mixture of both enantiomers of pinoresinol and pure (+)-secoisolariciresinol. In order to get more insight into the mechanism of (-)- and (+)-secoisolariciresinol biosynthesis, respectively, we isolated a cDNA encoding pinoresinol-lariciresinol reductase (PLR) from L. album. The heterologously expressed PLR-La1 converts only (+)-pinoresinol into (-)-secoisolariciresinol. In contrast, the heterologously expressed PLR from L. usitatissimum converts only (-)-pinoresinol to (+)-secoisolariciresinol confirming the results from others. Comparison of all available PLR protein sequences resulted in a few amino acids which may be responsible for the action of the PLRs with respect to the different enantioselectivity. A mutagenesis approach could not confirm this hypothesis. Aspects about the evolution of PLRs are discussed.  相似文献

We developed and characterized primers for 11 variable microsatellite loci present in the genome of the flax rust, Melampsora lini. The microsatellite loci were identified by sequencing clones from a library of EcoRI DNA fragments enriched for four simple sequence repeat motifs (AAG, AAT, TC and TG). All 11 primer pairs successfully amplified DNA fragments from a sample of 102 M. lini isolates (98 isolated from Linum marginale and four from Linum usitatissimum), revealing a total of 32 alleles. Allelic diversity at the 11 loci ranged from 0.030 to 0.449.  相似文献

The Linum usitatissimum L. var.NDL-2002 has high oil yield potential. The seeds of the variety were exposed to different concentrations (0.02–0.10%) of ethyl methane sulphonate (EMS) and sodium azide (SA). Observations on germination, germination inhibition, morphology, plant height, yield parameters, branches per plant, etc. were recorded in M1 generation in control and treated plants. Leaves notching, effect on internodal length were observed and also the more adverse effect of EMS was observed than SA.  相似文献

Linum flavum hairy roots were initiated from leaf discs using Agrobacterium rhizogenes strains LBA9402 and TR105 though two other strains, 15834 and A4, were relatively ineffective for induction. Significant variation in coniferin accumulation was observed between hairy root lines originating from different L. flavum seedlings and/or A. rhizogenes strains. Coniferin reached 58 mg g–1 dry wt by culturing the roots in Linsmaier and Skoog (LS) medium with 2,4-dichlorophenoxyacetic acid and naphthaleneacetic acid as growth regulators.  相似文献

The delimitation of species is a major issue in systematic biology and has been a re-emerging discipline in the last decade. A number of studies have shown that the use of multiple data sets is critical for the identification of cryptic species, particularly in groups with complex evolutionary histories. Liolaemus monticola is a montane lizard species distributed in central Chile (32°–42°S), with four described subspecies in a latitudinal gradient from north to south: L. m. monticola , L. m. chillanensis , L. monticola ssp. and L. m. villaricensis . In order to test the systematic status and phylogenetic relationships of the taxa included in the L. monticola group, we analysed morphological (morphometric and meristic) and molecular (allozyme and mitochondrial DNA) data sets. The results of the morphological analyses showed that meristic variables correctly assigned individuals with higher accuracy than did morphometric characters. The results of the analyses of allozyme data revealed eight diagnostic loci that are evidence for significant differences among the four L. monticola subspecies. Phylogenetic analyses with mitochondrial DNA data, including additional species, showed that the L. monticola group is polyphyletic. We postulate that the four current subspecies represent independent evolutionary lineages and must be raised to the specific level as L. monticola , L. chillanensis and L. villaricensis . Flax (Linum usitatissimum L.) hypocotyls were cultivated on regeneration media containing various concentrations of kanamycin (an aminoglycoside antibiotic commonly used to select transgenic plant material) solidified with three different gelling agents: gellan gum, agar and a mixture of both. The inhibitory effect of kanamycin on bud regeneration was analyzed. A significant interaction was observed between the nature of the gelling agent and the kanamycin concentration. The antibiotic concentration needed to strongly inhibit bud production varied greatly with the nature of the gelling agent. Gellan gum lowered the inhibitory effect of kanamycin. S-Adenosyl-L-methionine:beta-peltatin 6-O-methyltransferase was isolated and characterized from cell suspension cultures of Linum nodiflorum L. (Linaceae), a Linum species accumulating aryltetralin lignans such as 6-methoxypodophyllotoxin. The enzyme transfers a methyl group from S-adenosyl-L-methionine to the only free OH-group of beta-peltatin in position 6 thus forming beta-peltatin-A methylether. This reaction is a putative biosynthetic step in the biosynthesis of 6-methoxypodophyllotoxin from deoxypodophyllotoxin. The enzyme has a pH-optimum at pH 7.7 and a temperature optimum at 40 degrees C. The enzyme activity is strongly inhibited by MnSO(4), FeCl(3), FeSO(4) and ZnSO(4) as well as S-adenosyl-homocysteine. Mg(2+) and EDTA did not influence the methylation of beta-peltatin. Substrate saturation curves were obtained for S-adenosyl-methionine and beta-peltatin and apparent K(m)-values of 15 microM and 40 microM, respectively, were determined for these substrates. Substrate inhibition was observed for beta-peltatin. No other lignan substrate tested nor caffeic acid were accepted. The suspension cell line of Linum nodiflorum was characterized with respect to growth, medium alterations and lignan production as well as activity of SAM:beta-peltatin 6-O-methyltransferase. Highest specific activities of beta-peltatin 6-O-methyltransferase were determined on day 7 of the culture period corresponding to the highest levels of 6-methoxypodophyllotoxin on days 7 to 12.  相似文献

Effects of elicitors (mannan, -1,3-glucan, and ancymidol) on the activity of several key enzymes participating in lignan biosynthesis were studied in Linum austriacum L. cell cultures. The activities of L-phenylalanine ammonia-lyase, polyphenoloxydase, tyrosine ammonia-lyase, soluble phenoloxidase, and membrane-bound and soluble oxidases were assayed. The elicitors under study affected various steps in the metabolic pathway of lignan biosynthesis. Elevated enzyme activity accompanied an elicitor-enhanced synthesis of podophyllotoxins and peltatins.  相似文献

Cell suspension cultures of Linum album Kotschy ex Boiss. have been reported to produce anticancer podophyllotoxin and its related lignans. In the present study, we investigated the effect of culture filtrate of Fusarium graminearumon growth, and lignan and phenolic compounds in L. album cell culture. After 7 days of pre-culture, the cells were treated with 1% (v/v) of the culture filtrate. Cell growth was reduced, while phodophyllotoxin and lariciresinol production was stimulated reaching a maximum 0.0187 mg/g fresh weight (FW) and 0.0136 mg/g FW 5 days after the treatment, respectively. Also, our results provide evidence that the culture filtrate of F. graminearum can be effective on phenylalanine ammonia-lyase activity and phenolic compounds.  相似文献

Intrusively growing plant cells insert themselves between surrounding cells, thus increasing the number of membranes on the tissue cross-section. This parameter can be assessed by spin echo NMR method with a magnetic field pulse gradient. Diffusion echo decay was measured for stem regions of long-fiber flax (Linum usitatissimum L.) differing in the stages of primary fiber development, which elongate thousand-fold during intrusive growth. Additionally, the number of fibers on stem cross-sections was counted under microscope. An increase in the slow component of the echo diffusion decay was correlated with an increase in the number of fibers on the stem cross-section in the zone of intrusive growth, while other stem-structure characteristics remained unchanged. Thus, NMR method can be used for characterization of intrusive fiber growth in situ.  相似文献
