The influence of a mammalian sterol cholesterol and a plant sterol β-sitosterol on the structural parameters and hydration of bilayers in unilamellar vesicles made of monounsaturated diacylphosphatidylcholines (diCn:1PC, n = 14-22 is the even number of acyl chain carbons) was studied at 30 °C using small-angle neutron scattering (SANS). Recently published advanced model of lipid bilayer as a three-strip structure was used with a triangular shape of polar head group probability distribution (Ku?erka et al., Models to analyze small-angle neutron scattering from unilamellar lipid vesicles, Physical Review E 69 (2004) Art. No. 051903). It was found that 33 mol% of both sterols increased the thickness of diCn:1PC bilayers with n = 18-22 similarly. β-sitosterol increased the thickness of diC14:1PC and diC16:1PC bilayers a little more than cholesterol. Both sterols increased the surface area per unit cell by cca 12 Å2 and the number of water molecules located in the head group region by cca 4 molecules, irrespective to the acyl chain length of diCn:1PC. The structural difference in the side chain between cholesterol and β-sitosterol plays a negligible role in influencing the structural parameters of bilayers studied.  相似文献

X-ray scattering features induced by aggregates of alamethicin (Alm) were obtained in oriented stacks of model membranes of DOPC(diC18:1PC) and diC22:1PC. The first feature obtained near full hydration was Bragg rod in-plane scattering near 0.11 ?−1 in DOPC and near 0.08 ?−1 in diC22:1PC at a 1:10 Alm:lipid ratio. This feature is interpreted as bundles consisting of n Alm monomers in a barrel-stave configuration surrounding a water pore. Fitting the scattering data to previously published molecular dynamics simulations indicates that the number of peptides per bundle is n = 6 in DOPC and n ≥ 9 in diC22:1PC. The larger bundle size in diC22:1PC is explained by hydrophobic mismatch of Alm with the thicker bilayer. A second diffuse scattering peak located at q r ≈ 0.7 ?−1 is obtained for both DOPC and diC22:1PC at several peptide concentrations. Theoretical calculations indicate that this peak cannot be caused by the Alm bundle structure. Instead, we interpret it as being due to two-dimensional hexagonally packed clusters in equilibrium with Alm bundles. As the relative humidity was reduced, interactions between Alm in neighboring bilayers produced more peaks with three-dimensional crystallographic character that do not index with the conventional hexagonal space groups.  相似文献

Using UV-A, blue (B), green (G), red (R), and far-red (FR) light-emitting diodes (LEDs), we investigated the effects of different supplemental light qualities on phytochemicals and growth of ‘Red Cross’ baby leaf lettuce (Lactuca sativa L.) grown at a high planting density under white fluorescent lamps as the main light source inside a growth chamber. Photon flux added by supplemental LEDs for UV-A, B, G, R and FR were 18, 130, 130, 130 and 160 μmol m?2 s?1, respectively. Photosynthetic photon flux (PPF, 400–700 nm), photoperiod, and air temperature (day/night) was 300 μmol m?2 s?1, 16 h, and 25 °C/20 °C in all treatments including white light control. After 12 days of light quality treatment (22 days after germination), phytochemical concentration and growth of lettuce plants were significant affected by light treatments. Anthocyanins concentration increased by 11% and 31% with supplemental UV-A and B, respectively, carotenoids concentration increased by 12% with supplemental B, phenolics concentration increased by 6% with supplemental R while supplemental FR decreased anthocyanins, carotenoids and chlorophyll concentration by 40%, 11% and 14%, respectively, compared to those in the white light control. The fresh weight, dry weight, stem length, leaf length and leaf width significantly increased by 28%, 15%, 14%, 44% and 15%, respectively, with supplemental FR light compare to white light, presumably due to enhanced light interception by enlarged leaf area under supplemental FR light. Although the mechanisms of changes in phytochemicals under different supplemental light quality are not well known, the results demonstrated that supplemental light quality could be strategically used to enhance nutritional value and growth of baby leaf lettuce grown under white light.  相似文献

Cyclooxygenases (COX) and 8R-dioxygenase (8R-DOX) activities of linoleate diol synthases (LDS) are homologous heme-dependent enzymes that oxygenate fatty acids by a tyrosyl radical-mediated hydrogen abstraction and antarafacial insertion of O2. Soybean lipoxygenase-1 (sLOX-1) contains non-heme iron and oxidizes 18:2n ? 6 with a large deuterium kinetic isotope effect (D-KIE). The aim of the present work was to obtain further mechanistic insight into the action of these enzymes by using a series of n ? 6 and n ? 9 fatty acids and by analysis of D-KIE. COX-1 oxidized C20 and C18 fatty acids in the following order of rates: 20:2n ? 6 > 20:1n ? 6 > 20:3n ? 9 > 20:1n ? 9 and 18:3n ? 3  18:2n ? 6 > 18:1n ? 6. 18:2n ? 6 and its geometrical isomer (9E,12Z)18:2 were both mainly oxygenated at C-9 by COX-1, but the 9Z,12E isomer was mostly oxygenated at C-13. A cis-configured double bond in the n ? 6 position therefore seems important for substrate positioning. 8R-DOX oxidized (9Z,12E)18:2 at C-8 in analogy with 18:2n ? 6, but the 9E,12Z isomer was mainly subject to hydrogen abstraction at C-11 and oxygen insertion at C-9 by 8R-DOX of 5,8-LDS. sLOX-1 and 13R-MnLOX oxidized [11S-2H]18:2n ? 6 with similar D-KIE (~ 53), which implies that the catalytic metals did not alter the D-KIE. Oxygenation of 18:2n ? 6 by COX-1 and COX-2 took place with a D-KIE of 3–5 as probed by incubations of [11,11-2H2]- and [11S-2H]18:2n ? 6. In contrast, the more energetically demanding hydrogen abstractions of the allylic carbons of 20:1n ? 6 by COX-1 and 18:1n ? 9 by 8R-DOX were both accompanied by large D-KIE (> 20).  相似文献

Azadinium spinosum, a small dinoflagellate isolated from the North Sea, is a producer of azaspiracids (AZAs), a group of biotoxins associated with human illness following ingestion of contaminated shellfish. Using batch and continuous cultures of A. spinosum, the present study investigated the effects of different environmental and nutritional factors (salinity, temperature, photon flux density, aeration, culture media, nitrogen sources, phosphate source, and N/P ratios) on growth, maximum cell concentration, and AZA cell quota.Azadinium spinosum grew in a wide range of conditions; from 10 ̊C to 26 ̊C and salinities from 30 to 40, under irradiances ranging from 50 μmol m−2 s−1 to 250 μmol m−2 s−1, with or without aeration. Growth and maximum cell concentration were highest at a salinity of 35, at temperatures between 18 ̊C and 22 ̊C, and with aeration. Concerning AZA cell quota, the most significant effect was observed at low temperature; the AZA cell quota was more than 20 times higher at 10 ̊C (220 fg cell−1) than at temperatures between 18 ̊C and 26 ̊C. A. spinosum grew on all media tested with only slight differences in growth rate and AZA cell quota. In continuous culture, lowering the concentration of nutrients (0.5 strength of a modified K-medium) in the inflow improved AZA cell quota whereas higher concentration (doubling the normal strength of K-medium) improved maximal cell concentration. A. spinosum grew on different sources of nitrogen tested (nitrate, urea, ammonium) with almost no effect on toxin cell quota and growth, except that adding ammonium caused a decrease in growth.These first experiments on Azadinium spinosum increased our knowledge on factors affecting its growth and toxin production; furthermore, these results allowed and improved particularly A. spinosum production in pilot scale photobioreactors for AZA isolation.  相似文献

The objective of this study was to evaluate the effect of providing paper or rope, alternative enriching substrates to straw, to piglets in farrowing crates on piglet and sow welfare.Sixty multiparous sows and their litters were housed in crates that were either barren (BARREN), enriched with shredded paper (PAPER) or natural fibre rope (ROPE). Enriching substrates were introduced when piglets were 10 days old. The proportion of sows with udder and teat lesions before parturition and at weaning was recorded. Piglet facial lesions were scored according to severity on days 11, 18 and 27. Sow and piglet behaviour was recorded using scan sampling on days 14, 18, 22 and 26. Furthermore, the behaviour of one male and one female focal piglet per litter was recorded continuously for 10 min twice per day on days 14, 18, 22 and 26. On day 27 post-partum, focal piglets were observed for 5 min in a novel arena and for a further 5 min after a novel object was introduced.On day 27, there was a tendency for more BARREN sows to have teat lesions (P = 0.07). PAPER litters tended to have a smaller proportion of piglets with facial lesions (P = 0.06). ROPE piglets were active in the enriched area of the crate in more observations than BARREN and PAPER piglets (P < 0.01). PAPER piglets spent less time inactive (P < 0.01), less time exploring the pen-fittings (P < 0.01) and more time interacting with the enriching substrate (P < 0.001) than piglets in the other two treatments. In the first 5 min in the novel arena BARREN piglets froze more than PAPER and ROPE piglets (P = 0.05).In conclusion, shredded paper improved piglet welfare and was easily incorporated into the farrowing crates.  相似文献

Four treatment peatlands were studied in Northern Finland in order to determine peat P, Al and Fe concentration distributions and to find removal parameters for nutrient modelling. The sites had been under loading for 10–16 years. About 20 peat samples for analyses of oxalate-extractable and total P, Al and Fe were collected from the depth 0–10 cm in each peatland. The peat P concentration ranged from 0.097 mg g?1 to 14 mg g?1 being 1.7 mg g?1on average. P accumulated in preferential flow path areas. Although P concentrations were locally high, DSSP (the index of potential soil P release from peat to water) was very low in all studied peatlands, indicating that peat was not saturated by phosphorus. The results indicate that Al-based precipitation chemicals increase substantially P retention capacity of peatland and maintain a stabile P reduction in spite of varying P loads. The results also show long-term phosphorus accumulation in peatlands polishing municipal wastewater from activated sludge treatment. The regression analyses showed that k-value for N removal depends on N loading and hydraulic loading. The first-order area model together with regression analysis of the rate constant result in a good agreement between observed and calculated nitrogen concentration. The NH4-N loading to the peatland should be below 0.10 mg m2 d?1 in order to achieve a high reduction of 70%.  相似文献

The relationship between light intensity, nitrogen availability and pigmentation was investigated in mixotrophic and heterotrophic cultures of the unicellular red alga Galdieria sulphuraria 074G, a potential host for production of the blue pigment, phycocyanin (PC). During the exponential growth phase of batch cultures, G. sulphuraria 074G contained 2–4 mg phycocyanin per g dry weight. In carbon-limited and nitrogen-sufficient batch cultures grown in darkness, this value increased to 8–12 mg g−1 dry weight during the stationary phase, whereas the phycocyanin content in nitrogen-deficient cells decreased to values below 1 mg g−1 dry weight during stationary phase. Light intensities between 0 and 100 μmol photons m−2 s−1 had no influence on phycocyanin accumulation in mixotrophic cultures grown on glucose or fructose, while light stimulated phycocyanin synthesis in cultures grown on glycerol, in which the phycocyanin content in stationary phase was increased from 10 mg g−1 dry weight in darkness to 20 mg g−1 dry weight at a light intensity of 80 μmol photons m−2 s−1. At higher light intensities, less phycocyanin accumulated than at lower intensities, irrespective of the carbon substrate used. In carbon-limited continuous flow cultures grown on glucose or glycerol at a dilution rate of 0.63 day−1, corresponding to 50% of the maximum specific growth rate, the highest steady-state phycocyanin content of 15–28 mg g−1 dry weight was found at 65 μmol photons m−2 s−1. In contrast to the apparent glucose repression of light-induced PC synthesis observed in batch cultures, no glucose repression of the light stimulation was observed in continuous flow cultures because the glucose concentration in the culture supernatant always remained at limiting levels. Despite the fact that G. sulphuraria 074G contains less phycocyanin than some other microalgae and cyanobacteria, the ability of G. sulphuraria 074G to grow and synthesize phycocyanin in heterotrophic or mixotrophic cultures makes it an interesting alternative to the cyanobacterium, Spirulina platensis presently used for synthesis of phycocyanin.  相似文献

To elucidate the role of Zn2+-associated glutamate signaling pathway and voltage-dependent outward potassium ion currents in neuronal death induced by hypoxia–ischemia, PC12 cells were exposed to Oxygen–Glucose Deprivation (OGD) solution mimicking the hypoxic–ischemic condition in neuron, and the effect of N,N,N′,N′-tetrakis (2-pyridylmethyl) ethylenediamine (TPEN), a specific Zn2+ chelating agent on OGD-induced neuronal death was assessed in the present study. The cell survival rate, apoptosis status, potassium channel currents, intracellular free glutamate concentration and GluR2 expression in PC12 cells exposed to OGD in the absence or presence of TPEN for different time were investigated. The results showed that OGD exposure increased apoptosis, reduced the cell viability (P < 0.01 at 3 h, 6 h and 24 h, respectively compared to control), changed the voltage-dependent outward potassium ion current (increase at 1 h, but decrease at 3 h) and decreased the concentration of intracellular glutamate (P < 0.05 at 3 h and 6 h, P < 0.01 at 24 h respectively compared to control) and GluR2 expression (P < 0.05 at 3 h, 6 h and 24 h, respectively compared to control) in PC12 cells. TPEN partially reversed the influence resulted from OGD. These results suggest that OGD-induced cell apoptosis and/or death is mediated by the alteration in glutamate signaling pathway and the voltage-dependent outward potassium ion currents, while TPEN effectively prevent cell apoptosis and/or death under hypoxic–ischemic condition.  相似文献

Biodegradation of long chain n-alkanes and crude oil with fast rate and high concentration are desirable for bioremediation, especially in heavily oil-polluted areas, and enhanced oil recovery. We discovered Rhodococcus sp. Moj-3449 with such unique abilities by screening microorganisms for the growth on n-hexadecane at 30 mg/mL. The new strain grew very fast on 120 mg/mL of n-hexadecane giving a cell density of 14.7 g cdw/L after only 2 days’ incubation. During the growth with this strain, the oil–water phases were rapidly emulsified, giving rise to tolerance to high alkane concentration (250 mg/mL) and fast growth rate of 0.10–0.20 h?1 for alkane concentration of 1–180 mg/mL. The degraded concentration of n-hexadecane increased linearly with the initial alkane concentration (1–250 mg/mL). Incubation on n-hexadecane at 250 mg/mL for 7 days gave a cell density of 13.5 g cdw/L and degraded 124 mg/mL of n-hexadecane. The strain grew also fast on n-dodecane (C12), n-tetradecane (C14), and n-octadecane (C18), with degradation preference of C14 (=C16) > C12 > C18. Different from many alkane-degrading strains, Rhodococcus sp. Moj-3449 was found to have subterminal oxidation pathway. Rhodococcus sp. Moj-3449 degraded also crude oil fast at 60–250 mg/mL, with a wide range of n-alkanes (C10–C35) as substrates in which C14–C19 are preferred. The degradation ability increased with initial oil concentration from 60 to 150 mg/mL and slightly decreased afterwards. Incubation on 150 mg/mL of crude oil for 7 days degraded 37% of n-alkanes. The outstanding ability of rapidly degrading long chain n-alkanes and crude oil at high concentration makes Rhodococcus sp. Moj-3449 potentially useful for bioremediation and microbial enhanced oil recovery.  相似文献

The 5-HT1AR partial agonist PET radiotracer, [11C]CUMI-101, has advantages over an antagonist radiotracer as it binds preferentially to the high affinity state of the receptor and thereby provides more functionally meaningful information. The major drawback of C-11 tracers is the lack of cyclotron facility in many health care centers thereby limiting widespread clinical or research use. We identified the fluoroethyl derivative, 2-(4-(4-(2-(2-fluoroethoxy)phenyl)piperazin-1-yl)butyl)-4-methyl-1,2,4-triazine-3,5(2H,4H)dione (FECUMI-101) (Ki = 0.1 nM; Emax = 77%; EC50 = 0.65 nM) as a partial agonist 5-HT1AR ligand of the parent ligand CUMI-101. FECUMI-101 is radiolabeled with F-18 by O-fluoroethylation of the corresponding desmethyl analogue (1) with [18F]fluoroethyltosylate in DMSO in the presence of 1.6 equiv of K2CO3 in 45 ± 5% yield (EOS). PET shows [18F]FECUMI-101 binds specifically to 5-HT1AR enriched brain regions of baboon. The specificity of [18F]FECUMI-101 binding to 5-HT1AR was confirmed by challenge studies with the known 5-HT1AR ligand WAY100635. These findings indicate that [18F]FECUMI-101 can be a viable agonist ligand for the in vivo quantification of high affinity 5-HT1AR with PET.  相似文献

The cell-bound cholesterol oxidase from the Rhodococcus sp. NCIM 2891 was purified three fold by diethylaminoethyl–sepharose chromatography. The estimated molecular mass (SDS-PAGE) and Km of the enzyme were ∼55.0 kDa and 151 μM, respectively. The purified cholesterol oxidase was immobilized on chitosan beads by glutaraldehyde cross-linking reaction and immobilization was confirmed by Fourier transform infrared spectroscopy, scanning electron microscopy and energy dispersive X-ray analysis. The optimum temperature (45 °C, 5 min) for activity of the enzyme was increased by 5 °C after immobilization. Both the free and immobilized cholesterol oxidases were found to be stable in many organic solvents except for acetone. Fe2+ and Pb2+ at 0.1 mM of each acted as inhibitors, while Ag+, Ca2+, Ni2+ and Zn2+ activated the enzyme at similar concentration. The biotransformation of cholesterol (3.75 mM) with the cholesterol oxidase immobilized beads (3.50 U) leads to ∼88% millimolar yield of cholestenone in a reaction time of 9 h at 25 °C. The immobilized enzyme retains ∼67% activity even after 12 successive batches of operation. The biotransformation method thus, shows a great promise for the production of pharmaceutically important cholestenone.  相似文献

The viable fungal spora of baled grass silage was quantified and identified. One group of bales (Experiment 1, n = 15) was wrapped in 6 layers of polythene, and was handled and stored for 6 weeks under well-managed conditions. This silage was free of visible fungal contamination after 6 weeks and the film surrounding these bales was visibly undamaged. Moulds were cultured from 9/15 bales, while yeasts were cultured from all bales. A second group of bales (Experiment 2, n = 18) from 9 farms, were wrapped in 4 layers of film, handled and stored for 8 months using normal on-farm procedures. Visible fungal contamination was observed on the surface of most of these bales (15/18) and the film surrounding some bales (8/18) was damaged. In silage sampled from parts of bales that were visually non-mouldy, yeasts were cultured from all bales and moulds from 15/18 bales. Bales in the well-managed group (Experiment 1) had numerically lower numbers of yeasts (mean: 9.7 × 103 colony-forming units per gram of silage, cfu/g) and lower numbers of moulds (<101 cfu/g) compared to the normal on-farm produced group (2.3 × 105 yeast and 1.5 × 105 mould cfu/g; Experiment 2). The most common yeasts in each group of bales were Saccharomyces exiguus (12/15 bales; Experiment 1) and Pichia fermentans (11/18 bales; Experiment 2) and their numbers in all bales ranged from 0 to 105 cfu/g (mean: 8.4 × 103) and 0 to 1.5 × 106 cfu/g (mean: 1.2 × 105), respectively. Bales contaminated with visible mould growth on their surfaces had higher yeast and mould numbers in visually non-mouldy parts adjacent to the contaminated areas than bales that had no visible mould. Mould numbers were higher (P<0.05) in bales where the polythene film was visibly damaged compared to bales where the polythene film appeared intact. Penicillium roqueforti was not cultured from the well-managed bales, but it was the most common mould in bales prepared using normal on-farm procedures (13/18 bales); propagule numbers in bales ranged from 0 to 7.1 × 105 (mean: 1 × 105 cfu/g). Low numbers of mould propagules, the absence of viable P. roqueforti spores and the absence of mould growth in well-managed bales, emphasises the benefit of applying sufficient film and preventing it from becoming damaged during bale handling and storage.  相似文献

In this study, the yeast strain P10 which was identified to be a member of Aureobasidium pullulans var. melanogenum isolated from the mangrove ecosystems was found to be able to accumulate high content of oil in its cells. After optimization of the medium for lipid production and cell growth by the yeast strain P10, it was found that 8.0 g of glucose per 100 ml, 0.02 g of yeast extract per 100 ml, 0.02 g of ammonium sulfate per 100 ml, pH 6.0 in the medium were the most suitable for lipid production. During 10-l fermentation, a titer was 66.3 g oil per 100 g of cell dry weight, cell mass was 1.3 g per 100 ml, a yield was 0.11 g of oil per g of consumed sugar and a productivity was 0.0009 g of oil per g of consumed sugar per h within 120 h. At the same time, only 0.07 g of reducing sugar per 100 ml was left in the fermented medium. The compositions of the fatty acids produced were C16:0 (26.7%), C16:1(1.7%), C18:0 (6.1%), C18:1 (44.5%), and C18:2 (21.0%). The biodiesel produced from the extracted lipid could be burnt well.  相似文献

A galactoglucomannan (GGM) with an estimated weight-average molar mass of 3.5 × 105 was obtained from an aqueous extract of the mesocarp of fruits of Acrocomia aculeata (Jacq.) Lodd. by fractionation by Sephacryl S-300 HR and Sephadex G-25. Chemical and spectroscopic studies indicated that GGM has a main chain of (1  4)-linked β-d-mannopyranosyl residues attached to an initial chain of (1  3)-linked β-d-galactopyranosyl residues and a terminal chain of (1  4)-linked α-d-glucopyranosyl residues which comprised galactose, glucose and mannose in the molar ratio of 18:22:60. The adjuvant potential of the polysaccharide on the cellular immune response against ovalbumin antigen was investigated using in vivo assays.  相似文献

Carbon limited continuous cultures of Lactobacillus rhamnosus ATCC 7469 were grown at dilution rates between 0.1 h−1 and 0.6 h−1. At 0.45 h−1, oxygen uptake decreases producing a deficiency in the production of cell energy, lowering the concentration of biomass and finally accumulating glucose in the broth. Under the lack of energy pressure, L. rhamnosus ATCC 7469 triggers the production of lactic acid from pyruvate freeing NAD+ and stimulates glycolysis to continue, producing extra ATP from substrate-level phosphorylation. The 12-fold growing concentration of lactic acid and the 2-fold increase of succinic acid are in parallel with the steep 4-fold decrease of acetic acid production and small concentration changes of formic and propionic acids.The way the cells balance the available energy between the growing dilution rate and detoxification produces a stress within the culture, detected and described by flow cytometry. As the dilution rate increased, the proportion of L. rhamnosus ATCC 7469 cells with depolarized membrane steadily increased (1% at D = 0.20 h−1, 8% at D = 0.30 h−1, 14% at D = 0.45 h−1 and 26% for D = 0.62 h−1, respectively). To investigate tropical roles of the newly described Yihiella yeosuensis (ca. 8 μm in cell size), one of the smallest phototrophic dinoflagellates in marine ecosystems, its trophic mode and the types of prey species that Y. yeosuensis can feed upon were explored. Growth and ingestion rates of Y. yeosuensis on its optimal prey, Pyramimonas sp. (Prasinophyceae), as a function of prey concentration were measured. Additionally, growth and ingestion rates of Y. yeosuensis on the other edible prey, Teleaulax sp. (Cryptophyceae), were also determined for a single prey concentration at which both these rates of Y. yeosuensis on Pyramimonas sp. were saturated. Among bacteria and diverse algal prey tested, Y. yeosuensis fed only on small Pyramimonas sp. and Teleaulax sp. (both cell sizes = 5.6 μm). With increasing mean prey concentrations, both specific growth and ingestion rates of Y. yeosuensis increased rapidly before saturating at a mean Pyramimonas concentration of 109 ng C mL−1 (2725 cells mL−1). The maximum growth rate (mixotrophic growth) of Y. yeosuensis fed with Pyramimonas sp. at 20 °C under a 14:10-h light-dark cycle of 20 μE m−2 s−1 was 1.32 d−1, whereas the growth rate of Y. yeosuensis without added prey was 0.026 d−1. The maximum ingestion rate of Y. yeosuensis fed with Pyramimonas sp. was 0.37 ng C predator−1 d−1 (9.3 cells predator−1 d−1). At a Teleaulax concentration of 1130 ng C mL−1 (66,240 cells mL−1), growth and ingestion rates of Y. yeosuensis fed with Teleaulax sp. were 1.285 d−1 and 0.38 ng C predator−1 d−1 (22.4 cells predator−1 d−1), respectively. Thus, Y. yeosuensis rarely grows without mixotrophy, and mixotrophy supports high growth rates in Y. yeosuensis. Y. yeosuensis has the highest maximum mixotrophic growth rate with the exception of Ansanella graniferaamong engulfment feeding mixotrophic dinoflagellates. However, the high swimming speed of Y. yeosuensis (1572 μm s−1), almost the highest among phototrophic dinoflagellates, may prevent autotrophic growth. This evidence suggests that Y. yeosuensis may be an effective mixotrophic dinoflagellate predator on Pyramimonas and Teleaulax, and occurs abundantly during or after blooms of these two prey species.  相似文献

 Enzymatic hydrolysis of corncob and ethanol fermentation from cellulosic hydrolysate were investigated. After corncob was pretreated by 1% H2SO4 at 108 °C for 3 h, the cellulosic residue was hydrolyzed by cellulase from Trichoderma reesei ZU-02 and the hydrolysis yield was 67.5%. Poor cellobiase activity in T. reesei cellulase restricted the conversion of cellobiose to glucose, and the accumulation of cellobiose caused severe feedback inhibition to the activities of β-1,4-endoglucanase and β-1,4-exoglucanase in cellulase system. Supplementing cellobiase from Aspergillus niger ZU-07 greatly reduced the inhibitory effect caused by cellobiose, and the hydrolysis yield was improved to 83.9% with enhanced cellobiase activity of 6.5 CBU g−1 substrate. Fed-batch hydrolysis process was started with a batch hydrolysis containing 100 g l−1 substrate, with cellulosic residue added at 6 and 12 h twice to get a final substrate concentration of 200 g l−1. After 60 h of reaction, the reducing sugar concentration reached 116.3 g l−1 with a hydrolysis yield of 79.5%. Further fermentation of cellulosic hydrolysate containing 95.3 g l−1 glucose was performed using Saccharomyces cerevisiae 316, and 45.7 g l−1 ethanol was obtained within 18 h. The research results are meaningful in fuel ethanol production from agricultural residue instead of grain starch.  相似文献
