Hyperhomocysteinemia is a risk factor for a number of cardiovascular and neurodegenerative processes as well as a complicating factor in normal pregnancy. Toxic effects of homocysteine and the product of its spontaneous oxidation, homocysteic acid, are based on their ability to activate NMDA receptors, increasing intracellular levels of ionized calcium and reactive oxygen species. Even a short-term exposure of cells to homocysteic acid at concentrations characteristic of hyperhomocysteinemia induces their apoptotic transformation. The discovery of NMDA receptors both in neuronal tissue and in several other tissues and organs (including immunocompetent cells) makes them a target for toxic action of homocysteine. The neuropeptide carnosine was found to protect the organism from homocysteine toxicity. Treatment of pregnant rats with carnosine under conditions of alimentary hyperhomocysteinemia increases viability and functional activity of their progeny. Many metals and metal-containing compounds have been identified to be potent mutagens and carcinogens. Recently, a new sub-discipline of molecular toxicology and carcinogenesis has been developed. The combination of newly developed molecular techniques and free radical approach makes it possible to insightfully examine metal-induced carcinogenesis in precise molecular terms so that intricate biological interrelationships can be elucidated. In consideration of the increased amount of new findings deciphered by utilizing these new methods, the 1st Conference on Molecular Mechanisms of Metal Toxicity and Carcinogenesis was held. In this conference, more than 50 scientists from nine countries presented their novel discoveries concerning metal-induced carcinogenesis, delineated molecular mechanism of metal carcinogenesis, and proposed novel therapeutic intervention and prevention strategies. This article reviewes some of the state-of-the-art information presented at the meeting regarding the molecular mechanisms of metal cytotoxicity and carcinogenesis.  相似文献

Cd是植物非必需的微量元素,对植物有很强的毒性.Cd抑制植物细胞生长,抑制氧化磷酸化,引发氧化胁迫,影响光合作用,损伤核仁和影响质膜ATP酶的活力.一些耐Cd植物通过诱导形成螯合肽、金属硫蛋白、植物应激蛋白等抵御Cd毒,也有的耐Cd植物则通过细胞壁固定、液泡分隔、腺体分泌等途径来抵御Cd毒.植物螯合肽合成酶(PCS)相关的一些基因已得到克隆.金属硫蛋白(MT)的克隆基因导入植物,使植物对Cd毒的抗性增加;植物胁迫蛋白可提高植物对Cd毒的抗性,Zn转运蛋白可运转Cd.修饰基因则通过影响主要基因提高植物对Cd的忍耐能力.野生型植物耐Cd毒是多基因控制的,而植物短期的Cd忍耐,则仅受一个或少数基因控制.  相似文献

Chronic exposure to many heavy metals and metal-derivatives is associated with an increased risk of cancer, although the mechanisms of tumorigenesis are largely unknown. Approximately 125 scientists attended the 3rd Conference on Molecular Mechanisms of Metal Toxicity and Carcinogenesis and presented the latest research concerning these mechanisms. Major areas of focus included exposure assessment and biomarker identification, roles of ROS and antioxidants in carcinogenesis, mechanisms of metal-induced DNA damage, metal signalling, and the development of animal models for use in metal toxicology studies. Here we highlight some of the research presented, and summarize the conference proceedings.  相似文献

Hiraga and Fujii have recently reported that F344 rats consuming diets with high levels of sodium orthophenylphenate (SOPP) developed bladder tumors after 13–91 weeks (Fd. Cosmet. Toxicol., 19 (1981) 303). Several dose levels were tested and doses above 1.0% SOPP by weight appeared to cause an increase in both toxicity and bladder carcinogenicity. In order to put these studies into better perspective, the effects of feeding diets containing SOPP or orthophenylphenol (OPP) to F344 male rats for varying lengths of time were characterized.Hyperplasia of the bladder epithelium was noted in rats consuming diets containing 2% SOPP (equivalent to 1000–1500 mg/kg/day) after 1–2 weeks, with epithelial thickening increasing through 90 days. No bladder lesions were seen in the group consuming 2% OPP but focal kidney lesions were noted. In contrast to the results reported by Hiraga and Fujii, no tumors of the urinary tract were observed following 90 days of consumption of the 2% SOPP diet.The potential of these chemicals to induce genotoxic lesions was studied. No detectable increases in the reversion rates of Salmonella typhimurium (strains TA 98, TA 100, TA 1535, TA 1537 and TA 1538) were seen at concentrations of SOPP up to 5.8 · 10?4 M. SOPP also failed to produce a detectable increase in unscheduled DNA synthesis in primary rat hepatocytes at concentrations up to 1 · 10?4 M. No covalently-bound radioactivity was observed in DNA purified from the bladders of rats gavaged with 500 mg/kg [14C]SOPP or [14C]OPP (detection limit < 1 alkylation/106 nucleotides). These results suggest little or no genotoxicity for OPP or SOPP.The metabolism of OPP and SOPP in male F344 rats was shown to be dose-dependent. After gavage with 50 mg/kg or less, most of the administered material was recovered in the urine as glucuronide or sulfate conjugates of the parent material. After gavage with 500 mg/kg a new metabolite, apparently produced by mixed function oxidases, was observed. This metabolite was characterized by gas chromatography/mass spectroscopy as a conjugate of dihydroxybiphenyl. It is postulated that the potentially reactive metabolites produced by this oxidative pathway may be associated with the toxicity induced by high concentrations of OPP or SOPP.Thus the bladder toxicity and carcinogenicity of SOPP and the renal toxicity of OPP appear to occur only following the administration of high doses which saturate the normal conjugation pathways. However, since no genotoxicity was detected even at saturating doses, it appears unlikely that exposure to subtoxic doses would cause any significant increase in carcinogenic risk.  相似文献

Apoptosis (Programmed Cell Death) is a genetically regulated, morphologically distinct form of cell death that can be initiated by many different physiological and pathological stimuli. Such strategic intracellular programming is initiated in many instances during normal life cycle and development in order to maintain the homeostasis of a multicellular organism, to eliminate unwanted cells. However, apoptosis is also involved in a wide range of pathologic conditions, including neurodegenerative and cardiovascular diseases, cancer and autoimmune diseases. Therefore, the ability to understand and manipulate the cell death machinery is an obvious goal of medical research. Here we review the basic components of the death machinery, discuss their interaction in regulation of apoptosis, and describe the main pathways that are used to activate apoptosis.  相似文献

Protein glycosylation with O-linked N-acetylglucosamine (O-GlcNAc) is a reversible post-translational modification of serines/threonines on metazoan proteins and occurring with similar time scales, dynamics and stoichiometry as protein phosphorylation. Levels of this modification are regulated by two enzymes-O-GlcNAc transferase (OGT) and O-GlcNAc hydrolase (OGA). Although the biochemistry of these enzymes and functional implications of O-GlcNAc have been studied extensively, until recently the structures and molecular mechanisms of OGT/OGA were not understood. This review covers a body of recent work that has led to an understanding of the structure of OGA, its catalytic mechanism and the development of a plethora of different inhibitors that are finding their use in cell biological studies towards the functional implications of O-GlcNAc. Furthermore, the very recent structure determination of a bacterial OGT orthologue has given the first insights into the contribution of the tetratricopeptide repeats (TPRs) to the active site and the role of some residues in catalysis and substrate binding.  相似文献

The present paper is devoted to the evolutionary role of genetic modules shuffing. The mechanisms capable to produce new molecular functions and significant complications of ontogenesis are reviewed. Two-step model of macroevolution is proposed. This model comprises: (1) Arising of a new combination of genetic modules. This step does not result in formation of a new taxon but makes necessary ground for that process. (2) Precise structure completing of the new combination of modules and corresponding genome optimization by use of various mechanisms including point mutations. This step concerns many genes and finally leads to formation of a new taxon. It is shown that arising of new combinations of genetic modules might work out as molecular basis for progressive evolution, while alternative structural completing of the same combination might result in adaptive radiation.  相似文献

Chronic exposure of humans to toxic levels of fluoride in drinking water resulted in significant increase in blood GSH content with significant increase in the activities of erythrocyte glutathione metabolising enzymes viz., gamma-glutamylcysteine synthetase (E.C. 6.3.2.2), gamma-glutamyltranspeptidase (E.C. 2.3.2.2), GST (E.C. 2.5.1.18), GSH-Px (E.C. 1.11.1.9) and GR (E.C. 1.6.4.2). The data suggested a form of adaptation on the part of the erythrocytes to counteract the oxidative stress in red blood cells of fluorotic patients.  相似文献
