Two specific lytic enzymes were found in sporulating B. subtilis cells: a N-acetyl muramyl L-alanine amidase and a γ-D-glutamyl-(L) meso diaminopimelyl endopeptidase. Both enzyme activities were measured using radioactive synthetic substrates. They are low in vegetative cells and increase during sporulation. The highest rates of increase are concomitant with cortex formation. In a mutant with delayed sporulation enzyme synthesis is also delayed. We suggest that both enzymes play a role in the synthesis of the specific cortex peptidoglycan.  相似文献

Tetrahymenapyriformis cultures were starved, deciliated and incubated in a reciliation medium in the presence and absence of tunicamycin. This antibiotic was shown to have no effect on the rate of reciliation or the appearance of the organisms even when present in relatively large amounts, although cell division was blocked. A small inhibition of protein synthesis was noted in the presence of tunicamycin, but mannose incorporation was totally abolished. It was concluded that the reciliation of these organisms takes place by a mechanism that does not require the denovo synthesis of glycoprotein.  相似文献

OKY-1581 is an effective inhibitor of thromboxane synthesis invivo and invitro. The generation of thromboxane B2 (TxB2), prostaglandin E (PGE) and prostaglandin F (PGF) was measured following clotting and during platelet aggregation induced by collagen. The presence of OKY 1581 either invivo or invitro caused a reduction in TxB2 generation during clotting and platelet aggregation with a concomitant increase in PGE and PGF. The effect could be observed two hours after oral or subcutaneous administration of 5 to 100 mg per rabbit and lasted for 24 to 48 hours. The reduction in TxB2 was not accompanied by an inhibition of clotting or platelet aggregation. OKY-1581 appears to be a suitable agent for studying the role of TxB2 in atherosclerosis.  相似文献

Netropsin binds tightly to AT rich regions of DNA and correspondingly is an efficient inhibitor of mitochondrial DNA replication in Saccharomycescerevisiae. Netropsin treatment does not cause formation of large populations of petite cells. However, a large portion of cells grown in cultures with ethanol as carbon source are killed by 1 μg/ml netropsin. When petite induction by berenil or ethidium bromide is carried out in the presence of netropsin, the petite cells are killed. This appears to be an effect of netropsin action on the cells during the process of petite formation.  相似文献

Stereosecific synthesis of trans-hydrindanone 2a, a bicyclic analog of prostaglandin E1, via the trans-hydrindane β-keto ester 8, is described. When tested in the guinea pig, 2a exhibited no effects on blood pressure and no broncho-constriction or dilation activity. Additionally, 2a failed to inhibit both ADP and collagen induced blood platelet aggregation.  相似文献

Aspirin and salicylate are well-known but poorly understood teratogens in laboratory animals. Because aspirin inhibits PG synthesis, we systematically examined PG synthesis in rat embryo homogenates, the inhibition of PG synthesis in vivo and ex vivo by various non-steroidal anti-inflammatory drugs, and tested the hypothesis that the inhibition of PG synthesis is responsible for aspirin-induced limb defects in rats. We report that embryonic rat homogenates synthesis 6-keto-PGF, PGE, and PGF in large amounts from endogenous substrate, that aspirin and other non-steroidal anti-inflammatory drugs inhibit PG synthesis in vitro but not necessarily in vivo, and that contrary to our original hypothesis, the inhibition of PG synthesis is likely not responsible for aspirin-induced limb defects in rats.  相似文献

Mutants of B. subtilis 168 which exhibited an absolute requirement for glutamine have been isolated and characterized. Of the two mutants studied in detail, one had normal levels of glutamine synthetase and sporulated normally, the other had reduced glutamine synthetase and was asporogenic. Both mutants were mapped close to the thy A region of the chromosome by PBS1 transduction.A study of spontaneous revertants selected for glutamine prototrophy (or the sporulation character in the case of the asporogenic mutant) led to the conclusion that there is a relationship between the glutamine requirement and sporulation. However, the influence of glutamine could not be entirely explained by the catalytic properties of glutamine synthetase.  相似文献

Sporulating and various non-sporulating strains of S. pombe, especially several mutants deficient in conjugation or meiosis, were compared with respect to DNA synthesis under sporulation conditions. Meiosis and sporulation were induced by a transfer to nitrogen-free medium. As synchronized mitotic division was observed in all the strains as a first response to the shift, reducing the DNA amount per cell from the replicated state in G2 to the unreplicated state in the G1 phase of the cell cycle. Cells of the heterothallic wild-type strains (h+h+ or h?h?) accumulated in G1 with respect to DNA synthesis when they were incubated separately. In a mixed culture of these strains a period of enhanced DNA synthesis was observed after the start of zygote formation. This period of synthesis was absent in mutant fus1, where only prezygotes accumulated. Hence we conclude that in zygotic meiosis the premeiotic DNA synthesis is confined to zygotes after conjugation has been completed. In the diploid sporulating wild-type strain (h+h?), capable of azygotic meiosis without prior conjugation, premeiotic DNA synthesis occurred between 212 and 5 h after the shift to the sporulation medium. There was no significant premeiotic DNA synthesis observed in diploid cells of the meiosis-deficient mutants mei1 or mei3, whereas premeiotic DNA synthesis proceeded normally in mutant mei4, which is blocked at a stage after commitment to meiosis in opposition to both the other mutants.  相似文献

The lipid content of Blastocladiella emersonii zoospores is 5 pg/cell or about 13% of dry weight. Within the first few minutes of germination 60–70% of total zoospore lipid is lost, with neutral lipid, glycolipid and phospholipid fractions decreasing to about the same extent. These changes in lipid content precede the breakdown during germination of the complex and extensive membrane system of zoospores. During growth, which immediately follows germination, net phospholipid synthesis resumes so that total lipid is maintained at 6% of dry weight, but net synthesis of neutral and glycolipid does not begin until induction of sporulation. During sporulation the phospholipid level decreases so that the distribution of lipid among the three fractions approaches that found in zoospores. These changes in lipid content suggest that zoospore membranes containing neutral and glycolipids are synthesized de novo during spore formation.  相似文献

The synthesis of d1-4,5,6-trinor-3,7-inter-m-phenylene-3-oxaprostaglandins oxaprostaglandins of the E1 and F1α series7 from 6-endo-(1-heptenyl)-bicyclo[3:1:0]hexan-3-one (III), is described. Preliminary biological screening data for gerbil colon smooth muscle stimulation, rat blood pressure and substrate specificity toward 15-hydroxyprostaglandin dehydrogenase is presented. Neomycin inhibits in vitro DNA dependent DNA and RNA synthesis catalyzed by DNA polymerase I and RNA polymerase from E. coli. The effect of the antibiotic is more pronounced towards DNA synthesis. The inhibition of DNA synthesis is competitive with template DNA, does not reverse with excess deoxynucleoside triphosphate, Mg2+ or enzyme E. coli DNA polymerase I. Neomycin does not reduce the number of potential 3′ -OH end or primer. It seems to shorten the size of the newly formed polynucleotide.  相似文献

Superoxide dismutase and catalase were not detected in M. pneumoniae and several other species of Mycoplasma some of which consume oxygen and secrete H2O2. M. pneumoniae in suspension formed O2? in the presence of NADH and flavins and extracts of M. pneumoniae formed O2? in the presence of either NADH or NADPH. The lack of superoxide dismutase in M. pneumoniae could not be attributed to superoxide dismutase in the complex medium in which the organisms were grown because organisms grown in medium in which the superoxide dismutase had been inactivated by heat still contained undetectable amounts. Mycoplasmas appear to be an exception to the rule that organisms which consume O2 synthesize superoxide dismutase.  相似文献

Previous work has indicated that loss of ability of DNA dependent RNA polymerase, from stationary phase cultures of B. subtilis, to transcribe phage øe DNA was a sine qua non for sporulation. To ascertain if this change in template specificity was sporulation-specific, we repeated these experiments using a defined sporulation medium. The changes observed previously did not occur in the defined medium although sporulation was normal. The ability of the enzyme to transcribe other DNA templates was also examined. Similar studies were carried out using a polymerase from a rifamycin-resistant, sporulation conditional mutant. The significance of these findings with regard to the regulation of sporulation in B. subtilis is discussed.  相似文献

Differentiation of chick embryo myoblasts in, vitro requires both cell-cell recognition and cell-cell fusion. Prostaglandin E1 is known to play a role in controlling fusion, and a specific receptor has been postulated. We demonstrate two peaks of specific binding activity for prostaglandin E1 during myoblast differentiation in, vitro: one at 36 hours and one at 44 hours of culture. The prostaglandin binding activity of both peaks is sensitive to the inhibitors of prostaglandin synthesis, indomethacin and aspirin, and to the antibiotic tunicamycin. The 36 hour peak of binding activity occurs at the same time as the process of cell-cell recognition (24–36 hours) and recognition and prostaglandin binding exhibit similar sensitivity to indomethacin, aspirin and tunicamycin.  相似文献

The relationship between progesterone (P4) synthesis in vitro by luteal tissue and prostaglandin F (PGF) synthesis in vitro by endometrium and luteal tissue from two stages of the cycle, Days 7 to 8 and 15 to 16, was determined. Luteal and endometrial tissues were collected from pigs in three experimental groups at two stages of the cycle: (A) 6 pigs on Days 7 to 8 with spontaneous, 5 to 6 day old corpora lutea (CL); (B) 5 pigs on Days 15 to 16 with spontaneous, 13 to 14 day old CL; and (C) 6 pigs on Days 15 to 16 with spontaneous, 13 to 14 day old CL and 5 to 6 day old CL induced by pregnant mares serum gonadotropin (PMSG) and human chorionic gonadotropin (HCG) injections. Pigs with spontaneous, 13 to 14 day old CL of the cycle and PMSG-HCG induced accessory, 5 to 6 day old CL were used so that P4 and PGF synthesis in tissue from old and new CL could be compared in the same pig on Day 15 to 16 of the cycle. Tissues (100 mg minces) were incubated in 5 ml of Krebs Ringer solution in an atmosphere of 95% 02:5% CO2 for 2 hours at 0° C, 37° C, or 37° C with 1.3 x 10?4M indomethacin (IND). An aliquot of the incubation medium and an aliquot of the supernatant after homogenization of the tissue in the remaining medium of each flask was quantified for P4 and PGF by radioimmunoassay. P4 and PGF release into the medium and total accumulation of P4 and PGF in the flasks indicated that de novo synthesis had occured at 37° C. Compared to tissue from 13 to 14 day old CL, tissue from 5 to 6 day old CL synthesized more P4 per flask (53.9 vs 25.0 ng/mg tissue, P<.001) and released more P4 into the medium (20.8 vs 8.8 ng/mg, P<.001). P4 synthesis by luteal tissue from 5 to 6 day old and 13 to 14 day old CL from pigs in group C was similar to P4 synthesis by luteal tissue from pigs in group A and group B, respectively. Luteul PGF synthesis was not affected significantly by either the age of the CL or by PMSG-HCG treatment. For endometrial samples, the synthesis of PGF was not significantly different among pigs in groups A, B and C. If uterine PGF is involved in luteal regression in the pig, the sensitivity of the CL to PGF may be more important than an increase in PGF secretion during the late luteal phase of the estrous cycle.  相似文献

The 1.672 g/cm3 satellite DNA of Drosophila melanogaster was purified by successive equilibrium centrifugations in a CsCl gradient, an actinomycin DCsCl gradient, and a netropsin sulfate/CsCl gradient. The resulting DNA was homogeneous by the physical criteria of thermal denaturation, renaturation kinetics and equilibrium banding in each of the gradients listed above. In addition, the complementary strands could be separated in an alkaline CsCl gradient. Despite this rigorous purification procedure, nucleotide sequence analysis indicates the presence of two different DNA species in this satellite, poly A-A-T-A-TT-T-A-T-A and polyA-A-T-A-T-A-TT-T-A-T-A-T-A. Further physical, chemical and template properties of the isolated complementary strands demonstrate that these two repeating sequences are not interspersed with each other. This result has biological significance since sequences of this particular satellite are known to be located primarily on two different chromosomes, Y and 2. These results further suggest that the sequence heterogeneity observed in satellite DNA of higher eukaryotes may result from mixtures of very closely related but molecularly homogeneous repeated sequences each restricted to a particular chromosome or chromosomal region.  相似文献

Effects of the two peptide antibiotics tyrocidine and the linear gramicidin on exponential growing cultures of Bacillus brevis (ATCC 8185) were studied. Both antibiotics are produced by this bacterial strain. Our results demonstrate that the addition of tyrocidine leads to inhibition of RNA synthesis followed by a cessation of growth. In contrast, gramicidin does not affect net RNA synthesis and the cells continue to grow. However, sporulation is inhibited by gramicidin. The addition of a mixture of tyrocidine and gramicidin prevents partially the inhibitory effect of tyrocidine on RNA synthesis. The results presented are essentially in agreement with in vitro results described previously (8,9).  相似文献
