The binding of spiroperidol and bromospiroperidol, in vivo, was studied over a wide range of drug dosages. It was found that while spiroperidol and bromospiroperidol bind selectively in vivo to tissues known to be high in dopamine receptor binding sites, this specificity of binding does not persist at very low doses. Such anomalous binding behavior can have implications for the non-invasive imaging of these drugs.  相似文献

The transducing phage λdarg14, carrying a portion of the E. coli chromosome including argECBH, is derived from the heat-inducible, lysis-defective strain λy199, which has the b519 and b515 deletions. Cleavage of λy199 DNA by EcoRI endonuclease, followed by agarose slab gel electrophoresis, results in bands corresponding to the known C, D, E, and F segments of λ, and a segment A′ (A plus B minus b519 minus b515, the cleavage site between A and B being eliminated). Cleavage of λdarg14 DNA by EcoRI yields the expected D, E, and F segments of λ and four other segments, termed 14-1 through 14-4, whose length is 17.5, 6.2, 3.0, and 2.0 kilobases, respectively, as determined by electron microscopy and corroborated by electrophoretic mobility. Heteroduplex analysis shows that the E. coli argECBH cluster is on the 14-1 segment.  相似文献

The administration of adenosine together with homocysteine resulted in a dose-related elevation of cerebral S-adenosyl-L-homocysteine without concomitant perturbation of S-adenosyl-L-methionine levels. The adenosine + homocysteine treatment also decreased the incorporation of labile and stable methyl groups into brain proteins. Brain [3H]-phosphatidyl N,N-dimethylethanolamine and [3H]-phosphatidylcholine were also significantly decreased while [3H]-phosphatidyl-N-monomethylethanolamine remained unchanged. The data indicate that elevated brain S-adenosylhomocysteine can markedly and selectively inhibit the in vivo methylation of brain proteins and phospholipids.  相似文献

E.coli aspartokinase II-homoserine dehydrogenase II is, as aspartokinase I-homoserine dehydrogenase I, composed of three globular domains: the N-terminal domain is endowed with kinase activity; the C-terminal domain carries the dehydrogenase activity. These two parts of the polypeptide chain are separated by a central inactive domain. Thus, the polypeptide chains of the two multifunctional proteins are homologous not only in their sequence but also in their triglobular domain structure.  相似文献

Genetic transformation of E.coli for various chromosomal markers was accomplished by (i) using recipient cells that lack the recBC DNase but were recombination proficient due to sbcA or sbcB mutations and (ii) treating the recipient cells with CaCl2 at a concentration that facilitates transfection by λ DNA. Cotransformation of three markers (thr+ara+.leu+) was found to depend on the molecular weight of the transforming DNA.  相似文献

Guanylate cyclase from crude homogenates of vegetative Dictyosteliumdiscoideum has been characterized. It has a pH optimum of 8.0, temperature optimum of 25°C and requires 1 mM dithiothreitol for optimal activity. It strongly prefers Mn++ to Mg++ as divalent cation, requires Mn++ in excess of GTP for detectable activity, and is inhibited by high Mn++ concentrations. It has an apparent Km for GTP of approximately 517 μM at 1 mM excess Mn++.The specific activity of guanylate cyclase in vegetative homogenates is 50–80 pmoles cGMP formed/min/mg protein. Most of the vegetative activity is found in the supernatant of a 100,000 x g spin (S100). The enzyme is relatively unstable. It loses 40% of its activity after 3 hours storage on ice. Enzyme activity was measured from cells that had been shaken in phosphate buffer for various times. It was found that the specific activity changed little for at least 8 hours. Cyclic AMP at 10?4 M did not affect the guanylate cyclase activity from crude homogenates of vegetative or 6 hour phosphate-shaken cells.  相似文献

Kinetoplast DNA from the order Kinetoplastidae (trypanosomatids) exists as large associations (molecular weight 4 × 1010), made up of about 104 small, probably circular, molecules, commonly known as ‘minicircles’. These minicircles were originally thought to be identical in base composition, suggesting that the coding capacity of kinetoplast DNA is very restricted. However, linear molecules have also been observed in preparations of kinetoplast DNA, which, if they contain unique sequences, could represent additional genetic information. This linear DNA has been assumed to be derived from the kinetoplast, but the possibility of it being nuclear contamination has not been definitely ruled out. Work presented in this paper demonstrates that nuclear DNA contamination may indeed be present in kinetoplast DNA prepared by a commonly used method.  相似文献

Employing the reconstitution system of Traub and Nomura, the temperature dependence of physical and function reconstitution was studied. The data show that the physical formation of 30S particles occurs at temperatures 15° below the maximum temperature for functional reconstitution. This may indicate that the high activation energy required for functional reconstitution is directed toward arrangement of active sites rather than the interaction of proteins with their cognate sites on RNA.  相似文献

An inhibitor of Streptococcus,mutans endodextranase was detected in proteins prepared from batch cultures of S.,mutans strains representing serotypes a through g. Affinity chromatography of strain 6715-49 proteins, which apparently were free of endodextranase activity, yielded an active endodextranase and, in a separate peak, the endodextranase inhibitor. The presence of the inhibitor in culture fluids accounts for the absence of endodextranase activity in batch-grown cultures of S.,mutans known to produce this enzyme.  相似文献

Aspirin and salicylate are well-known but poorly understood teratogens in laboratory animals. Because aspirin inhibits PG synthesis, we systematically examined PG synthesis in rat embryo homogenates, the inhibition of PG synthesis in vivo and ex vivo by various non-steroidal anti-inflammatory drugs, and tested the hypothesis that the inhibition of PG synthesis is responsible for aspirin-induced limb defects in rats. We report that embryonic rat homogenates synthesis 6-keto-PGF, PGE, and PGF in large amounts from endogenous substrate, that aspirin and other non-steroidal anti-inflammatory drugs inhibit PG synthesis in vitro but not necessarily in vivo, and that contrary to our original hypothesis, the inhibition of PG synthesis is likely not responsible for aspirin-induced limb defects in rats.  相似文献

Rhodotorucine A which induces mating tube formation of a cells in Rhodosporidiumtoruloides is metabolized rapidly by a cells. By use of labeled rhodotorucine A, the degradation was found to be proteolytic. Two peptide fragments Tyr-Pro-Glu-Ile-Ser-Trp-Thr-Arg and Asn-Gly-Cys(S-farnesyl) were identified as the metabolites. Proteolysis of the pheromone mainly occurred on the cell surface. Culture filtrate of a cells at log phase did not metabolize rhodotorucine A.  相似文献

OKY-1581 is an effective inhibitor of thromboxane synthesis invivo and invitro. The generation of thromboxane B2 (TxB2), prostaglandin E (PGE) and prostaglandin F (PGF) was measured following clotting and during platelet aggregation induced by collagen. The presence of OKY 1581 either invivo or invitro caused a reduction in TxB2 generation during clotting and platelet aggregation with a concomitant increase in PGE and PGF. The effect could be observed two hours after oral or subcutaneous administration of 5 to 100 mg per rabbit and lasted for 24 to 48 hours. The reduction in TxB2 was not accompanied by an inhibition of clotting or platelet aggregation. OKY-1581 appears to be a suitable agent for studying the role of TxB2 in atherosclerosis.  相似文献

The paromomycin producing organism Streptomycesrimosusforma paromomycinus is resistant to this antibiotic and contains a phosphotransferase which inactivates paromomycin. The gene encoding this enzyme has been inserted in the Streptomyces vector pIJ702 and then cloned in Streptomyceslividans, selecting for paromomycin-resistance. Three plasmids have been isolated and one of them, pMJ1, contains a 2.2 kb insert with a single HindIII restriction site. Insertion of foreign DNA in this site blocks the expression of the phosphotransferase enzyme indicating that it is within the cloned gene. These findings provide a new dominant selective marker for Streptomyces cloning vectors with the versatility of insertional inactivation.  相似文献

The release of endogenous catecholamines (CA) from rat brain slices containing the nucleus tractus solitarius (NTS) was measured using a sensitive radioenzymatic assay. KCl (35 to 75 mM) induced a dose-related increase in norepinephrine (NE) release. Dopamine (DA) release was maximal with 50 mM KCl. An increase in epinephrine (E) release was only observed with 75 mM KCl. NE and E release was totally calcium-dependent whereas DA release was only partially calcium-dependent. Subsequent administrations of KCl released less CA. The calcium dependency of the KCl induced release of E, NE, and DA suggests a neurotransmitter function in the NTS for these CA. A difference in storage sites and/or mechanisms may be responsible for the observed differences in sensitivity to KCl and to extracellular calcium.  相似文献

The effect of protein moiety on the conformation of 16S and 23S RNA of the E.coli ribosome has been studied by circular dichroic spectroscopy. Both rRNAs possess a comparable net content of ordered secondary structure which remains unchanged after association with ribosomal proteins into “core” particles or into complete 30S and 50S subunits, respectively. However, differences found in the stability and the cooperativity of melting of free and protein-associated rRNAs imply protein-caused variations in the distribution of the intramolecular hairpin stems and loops and/or changes in long range tertiary interactions which appear to be different for both rRNAs. While 23S RNA is maximally stabilized on the large subunit by the full set of proteins, 16S RNA on the complete small subunit shows lower stability but higher cooperativity in melting.  相似文献

Messenger RNA for two T4 specific enzymes, deoxynucleotide kinase and α glucosyltransferase, have been sized by sedimentation on sucrose density gradients. The sedimentation constants of transferase and kinase mRNAs formed in vitro were 21.5S and 14.5S respectively, regardless of the duration of incubation up to 20 min. Although the kinase mRNA isolated from cells infected with T4 phage for 10 min was the same size as found in vitro (14.5S), the transferase mRNA was found in a segment approximating the size of the kinase mRNA (14.5S). The experiments indicate that α glucosyltransferase mRNA is formed first as a polycistronic message and is then processed to the smaller unit.  相似文献

Differential scanning calorimetry of crude brain mitochondrial lipids obtained from control and morphine treated rats was carried out and the lipid phase transition measured. Morphine treatment resulted in a significant decrease in the temperature range and enthalpy of the phase transition. This effect was found to be dose dependent and reversible both in vivo and in vitro by naloxone. Studies with levorphanol and dextrorphan demonstrated stereospecificity. Furthermore, the ether precipitable fraction of total lipid extracts was shown to mediate the opiate response.  相似文献

The naturally occurring amino acid gabaculine ((?)-5-amino-1,3 cyclohexadiene carboxylic acid) is a potent irreversible inhibitor of mouse brain γ-aminobutyric acid (GABA)-α-ketoglutaric acid transaminase. When administered I.P. gabaculine, irreversibly inhibits the mouse brain enzyme in a time dependent fashion. Concomitant with this inhibition is a rise in endogenous brain GABA levels. Administration of gabaculine at a concentration of 100 mg/kg mouse leads to the complete inhibition of the enzyme after 4 hrs. Brain levels of GABA continually rise after the administration of the drug. After 20 hrs they are 15–20 times higher than levels in the untreated animals.  相似文献
