Circumferential and radial components of the yolk cell surface movements were measured in the loach embryos at the late blastula stage within 40–50 min after puncture or indentation by an obliquely directed glass rod. The yolk cell surface was preliminarily marked by coal particles. It was shown that even closely located regions of the surface differed markedly in the rate and direction of their movements. In the vicinity of puncture, the yolk cell surface at first contracted in both circumferential and radial directions and then widened, but did not reach the initial values. In more remote areas, this surface continued to contract in the circumferential direction, but was extended in the radial direction. The degree of its contraction along different radii was unequal. The reaction to oblique indentation was anisotropic: the closest area of the yolk cell surface, located along the direction of indentation, contracted in both circumferential and radial directions and formed a fold “leaking” onto the rod, while the opposite area contracted in the circumferential direction, but extended in the radial direction. A conclusion was drawn that the yolk cell surface is a multivariant mechanosensitive system. Its active responses to mechanical influences obey the same patterns as multicellular embryonic tissues.  相似文献

Small radial folds are aggregated in radial strands, several dozens folds in each. Sucking off a part of yolk accelerated the above processes, while addition of yolk, cooling, and absence of Ca2+ in the incubation medium slowed down or suppressed these processes. The observed structurization can be considered as self-organization at the level of the yolk cell cortical level, largely similar to that during normal morphogenesis at the level of multicellular sheets. Hence, the membrane dynamics in the yolk cell wall after its damage can be considered as one of simplified (minimal) models of morphogenesis. A study of this model makes it possible to narrow down the circle of factors essential for self-organization of morphogenetic processes.  相似文献

Aortic dissection occurs frequently and is clinically challenging; the underlying mechanics remain unclear. The present study investigates the dissection properties of the media of 15 human abdominal aortas (AAs) by means of direct tension tests (n=8) and peeling tests (n=12). The direct tension test demonstrates the strength of the media in the radial direction, while the peeling test allows a steady-state investigation of the dissection propagation. To explore the development of irreversible microscopic changes during medial dissection, histological images (n=8) from four AAs at different peeling stages are prepared and analyzed. Direct tension tests of coin-shaped medial specimens result in a radial failure stress of 140.1+/-15.9 kPa (mean+/-SD, n=8). Peeling tests of rectangular-shaped medial strips along the circumferential and axial directions provide peeling force/width ratios of 22.9+/-2.9 mN/mm (n=5) and 34.8+/-15.5 mN/mm (n=7); the related dissection energies per reference area are 5.1+/-0.6 mJ/cm(2) and 7.6+/-2.7 mJ/cm(2), respectively. Although student's t-tests indicate that force/width values of both experimental tests are not significantly different (alpha=0.05, p=0.125), the strikingly higher resisting force/width obtained for the axial peeling tests is perhaps indicative of anisotropic dissection properties of the human aortic media. Peeling in the axial direction of the aorta generates a remarkably "rougher" dissection surface with respect to the surface generated by peeling in the circumferential direction. Histological analysis of the stressed specimens reveals that tissue damage spreads over approximately six to seven elastic laminae, which is about 15-18% of the thickness of the abdominal aortic media, which forms a pronounced cohesive zone at the dissection front.  相似文献

Movement and searching behaviors at diverse spatial scales are important for understanding how animals interact with their environment. Although the shapes of branches and the voids in arboreal habitats seem likely to affect searching behaviors, their influence is poorly understood. To gain insights into how both environmental structure and the attributes of an animal may affect movement and searching, we compared the three-dimensional exploratory movements of snakes in the dark on two simulated arboreal surfaces (disc and horizontal cylinder). Most of the exploratory movements of snakes in the dark were a small fraction of the distances they could reach while bridging gaps in the light. The snakes extended farther away from the edge of the supporting surface at the ends of the cylinder than from the sides of the cylinder or from any direction from the surface of the disc. The exploratory movements were not random, and the surface shape and three-dimensional directions had significant interactive effects on how the movements were structured in time. Thus, the physical capacity for reaching did not limit the area that was explored, but the shape of the supporting surface and the orientation relative to gravity did create biased searching patterns.  相似文献

A Finite Element workflow for the multiscale analysis of the aortic valve biomechanics was developed and applied to three physiological anatomies with the aim of describing the aortic valve interstitial cells biomechanical milieu in physiological conditions, capturing the effect of subject-specific and leaflet-specific anatomical features from the organ down to the cell scale. A mixed approach was used to transfer organ-scale information down to the cell-scale. Displacement data from the organ model were used to impose kinematic boundary conditions to the tissue model, while stress data from the latter were used to impose loading boundary conditions to the cell level. Peak of radial leaflet strains was correlated with leaflet extent variability at the organ scale, while circumferential leaflet strains varied over a narrow range of values regardless of leaflet extent. The dependency of leaflet biomechanics on the leaflet-specific anatomy observed at the organ length-scale is reflected, and to some extent emphasized, into the results obtained at the lower length-scales. At the tissue length-scale, the peak diastolic circumferential and radial stresses computed in the fibrosa correlated with the leaflet surface area. At the cell length-scale, the difference between the strains in two main directions, and between the respective relationships with the specific leaflet anatomy, was even more evident; cell strains in the radial direction varied over a relatively wide range (\(0.36-0.87\)) with a strong correlation with the organ length-scale radial strain (\(R^{2}= 0.95\)); conversely, circumferential cell strains spanned a very narrow range (\(0.75-0.88\)) showing no correlation with the circumferential strain at the organ level (\(R^{2}= 0.02\)). Within the proposed simulation framework, being able to account for the actual anatomical features of the aortic valve leaflets allowed to gain insight into their effect on the structural mechanics of the leaflets at all length-scales, down to the cell scale.

Endovascular stents are increasingly being used to treat cerebral aneurysms. Mechanically, a cerebrovascular stent must have a low radial stiffness to prevent vessel dissection and rupture. To minimize these complications, we need to consider a stent design that has a low radial force and disperses the load within the stented artery. Therefore, highly distensible, load-dispersion stent designs are desirable for intracranial stenting. This study focused on closed-cell stent geometries and calculated the differences in stress within the artery because of the structure by using finite-element modeling. The results showed that the design with hexagonal cell geometry stretched in the circumferential direction had lower radial and circumferential stresses than did the other models. Comparing the maximum radial stress of our models, stress reduction of 35% was obtained with this design. Moreover, its radial stress was 47 kPa, which was similar to the critical stress of 42 kPa assumed in this study. This stent model was characterized by narrow strut spacing and a large surface area, which was dominated by the twined-spring geometry. It had low radial and circumferential stresses and a dispersed stress distribution compared with the other models. Therefore, this design is a desirable load-dispersing design for cerebrovascular treatment.  相似文献

The perfusion of the liver microcirculation is often analyzed in terms of idealized functional units (hexagonal liver lobules) based on a porous medium approach. More elaborate research is essential to assess the validity of this approach and to provide a more adequate and quantitative characterization of the liver microcirculation. To this end, we modeled the perfusion of the liver microcirculation using an image-based three-dimensional (3D) reconstruction of human liver sinusoids and computational fluid dynamics techniques. After vascular corrosion casting, a microvascular sample (±0.134 mm(3)) representing three liver lobules, was dissected from a human liver vascular replica and scanned using a high resolution (2.6 μm) micro-CT scanner. Following image processing, a cube (0.15?×?0.15?×?0.15 mm(3)) representing a sample of intertwined and interconnected sinusoids, was isolated from the 3D reconstructed dataset to define the fluid domain. Three models were studied to simulate flow along three orthogonal directions (i.e., parallel to the central vein and in the radial and circumferential directions of the lobule). Inflow and outflow guidances were added to facilitate solution convergence, and good quality volume meshes were obtained using approximately 9?×?10(6) tetrahedral cells. Subsequently, three computational fluid dynamics models were generated and solved assuming Newtonian liquid properties (viscosity 3.5 mPa s). Post-processing allowed to visualize and quantify the microvascular flow characteristics, to calculate the permeability tensor and corresponding principal permeability axes, as well as the 3D porosity. The computational fluid dynamics simulations provided data on pressure differences, preferential flow pathways and wall shear stresses. Notably, the pressure difference resulting from the flow simulation parallel to the central vein (0-100 Pa) was clearly smaller than the difference from the radial (0-170 Pa) and circumferential (0-180 Pa) flow directions. This resulted in a higher permeability along the central vein direction (k(d,33)?=?3.64?×?10(-14) m(2)) in comparison with the radial (k(d,11)?=?1.56?×?10(-14) m(2)) and circumferential (k(d,22)?=?1.75?×?10(-14) m(2)) permeabilities which were approximately equal. The mean 3D porosity was 14.3. Our data indicate that the human hepatic microcirculation is characterized by a higher permeability along the central vein direction, and an about two times lower permeability along the radial and circumferential directions of a lobule. Since the permeability coefficients depend on the flow direction, (porous medium) liver microcirculation models should take into account sinusoidal anisotropy.  相似文献

Three known genes guide circumferential migrations of pioneer axons and mesodermal cells on the nematode body wall. unc-5 affects dorsal migrations, unc-40 primarily affects ventral migrations, and unc-6 affects migrations in both directions. Circumferential movements still occur, but are misdirected whereas longitudinal movements are normal in these mutants. Pioneer growth cones migrating directly on the epidermis are affected; growth cones migrating along established axon fascicles are normal. Thus these genes affect cell guidance and not cell motility per se. We propose that two opposite, adhesive gradients guide circumferential migrations on the epidermis. unc-5, unc-6, and unc-40 may encode these adhesion molecules or their cellular receptors. Neurons have access to the basal lamina and the basolateral surfaces of the epidermis, but mesodermal cells contact only the basal lamina. These genes probably identify molecular cues on the basal lamina that guide mesodermal migrations. The same basal lamina cues, or perhaps related molecules on the epidermal cell surfaces, guide pioneer neurons.  相似文献

Structural features of elastic laminae within arteries can provide vital information for both the mechanobiology and the biomechanics of the wall. In this paper, we propose, test and illustrate a new computer-based scheme for automated analysis of regional distributions of elastic laminae thickness, inter-lamellar distances and fragmentation furcation points (FPs) from standard histological images. Our scheme eliminates potential artefacts produced by tissue cutting, automatically aligns tissue according to physiologic orientations and performs cross-sectional measurements along radial directions. A statistical randomised complete block design and F test were used to assess the potential (non)-uniformity of lamellar thicknesses and separations along both radial and circumferential directions. Illustrative results for both normotensive and hypertensive thoracic porcine aorta revealed marked heterogeneity along the radial direction in nearly stress-free samples. Clearly, regional measurements can provide more detailed information about morphologic changes that cannot be gained by globally averaged evaluations alone. We also found that quantifying FP densities offers new information about potential elastin fragmentation, particularly in response to increased loading due to hypertension.  相似文献

A distinction is drawn between two intrinsic directions within a simple spatial configuration. The line joining two elements is the radial direction and orthogonal to it is the tangential direction. Separation and bisection discrimination and just-detectable differences in line length are examples of radial thresholds. Vernier and alignment detection are tangential thresholds. Neural processing along these two intrinsic directions differs. There is a strong 'oblique effect' for tangential thresholds and virtually none for radial thresholds. This study concerns the retinotopic organization of the ganglion cell fibres in the visual system of the frog Xenopus laevis. HRP was used to trace the pathways taken by fibres from discrete retinal positions as they pass from the retina, along the optic nerve and into the chiasma. The ganglion cell fibres in the retina are arranged in fascicles which correspond with their circumferential positions of origin. Within the fascicles the fibres show little age-related layering and do not have a strict radial organization. As the fascicles of fibres pass into the optic nerve head there is some exchange of position resulting in some loss of the retinal circumferential organization. The poor radial organization of the fibres in the retinal fascicles persists as the fibres pass through the intraocular part of the nerve. At a position just behind the eye there is a major fibre reorganization in which fibres arising from cells of increasingly peripheral retinal locations are found to have passed into increasingly peripheral positions in the nerve. Thus, fibres from peripheral-most retina are located at the nerve perimeter, whilst fibres from central retina are located in the nerve core. It is at this point that the radial, chronotopic, ordering of the ganglion cell axons, found throughout the rest of the optic pathway, is established. This annular organization persists along the length of the nerve until a position just before the nerve enters the brain. Here, fibres from each annulus move to form layers as they pass into the optic chiasma. This change in the radial organization appears to be related to the pathway followed by all newly growing fibres, in the most superficial part of the optic tract, adjacent to the pia. Just behind the eye, where fibres become radially ordered, the circumferential organization of the projection is largely lost. Fibres from every circumferential retinal position, which are of similar radial position, are distributed within the same annulus of the nerve. At the nerve-chiasma junction where each annulus forms a single layer as it enters the optic tract, there is a further mixing of fibres from all circumferential positions. However, as the fibres pass through the chiasma some active pathway selection occurs, generating the circumferential organization of the fibres in the optic tract. Additional observations of the organization of fibres in the optic nerve of Rana pipiens confirm previous reports of a dual representation of fibres within the nerve. The difference in the organization of fibres in the optic nerve of Xenopus and Rana pipiens is discussed.  相似文献

The preoptic area/anterior hypothalamus (POA/AH) is a site where hormones dramatically influence development. The POA/AH is comprised of multiple subgroups, but little is known about the derivation of these subgroups during development. Results from several laboratories suggest that some cells in the POA/AH originate from progenitor cells in other regions of the developing nervous system. We are exploring pathways for migration in the developing POA/AH in two ways. First, we are examining the distribution of radial glial processes as potential migratory guides using immunocytochemistry. We have identified a transient pattern of radial glial processes from the lateral ventricles to the pial surface at the base of the POA/AH. Additionally, the expression of a molecule in radial glial processes originating in the third ventricle was decreased by prenatal treatment with testosterone. Second, we are utilizing time-lapse video microscopy in vitro to assess the extent and direction of movements of fluorescent dye-labeled cells at different ages in brain slice preparations from the POA/AH of developing rats. Data from these studies indicate that cell migration in the POA/AH includes movements along dorsal-ventral routes and from lateral to medial positions, in addition to the predicted medial to lateral pathway away from the third ventricle. Several researchers have examined effects of gonadal steroids on neurite outgrowth, cell differentiation, cell death, and synaptogenesis. The determination of cell position, however, may be a key event influenced by gonadal steroids earlier in development. The characterization of migratory pathways that contribute to permanent changes in brain structure and ultimately function is essential for unraveling the process of sexual differentiation.  相似文献

Computational models for the heart's mitral valve (MV) exhibit several uncertainties that may be reduced by further developing these models using ground-truth data-sets. This study generated a ground-truth data-set by quantifying the effects of isolated mitral annular flattening, symmetric annular dilatation, symmetric papillary muscle (PM) displacement and asymmetric PM displacement on leaflet coaptation, mitral regurgitation (MR) and anterior leaflet strain. MVs were mounted in an in vitro left heart simulator and tested under pulsatile haemodynamics. Mitral leaflet coaptation length, coaptation depth, tenting area, MR volume, MR jet direction and anterior leaflet strain in the radial and circumferential directions were successfully quantified at increasing levels of geometric distortion. From these data, increase in the levels of isolated PM displacement resulted in the greatest mean change in coaptation depth (70% increase), tenting area (150% increase) and radial leaflet strain (37% increase) while annular dilatation resulted in the largest mean change in coaptation length (50% decrease) and regurgitation volume (134% increase). Regurgitant jets were centrally located for symmetric annular dilatation and symmetric PM displacement. Asymmetric PM displacement resulted in asymmetrically directed jets. Peak changes in anterior leaflet strain in the circumferential direction were smaller and exhibited non-significant differences across the tested conditions. When used together, this ground-truth data-set may be used to parametrically evaluate and develop modelling assumptions for both the MV leaflets and subvalvular apparatus. This novel data may improve MV computational models and provide a platform for the development of future surgical planning tools.  相似文献

The present study investigated how direction of hand movement, which is a well-described parameter in cerebral organization of motor control, is incorporated in the somatotopic representation of the manual effector system in the human primary motor cortex (M1). Using functional magnetic resonance imaging (fMRI) and a manual step-tracking task we found that activation patterns related to movement in different directions were spatially disjoint within the representation area of the hand on M1. Foci of activation related to specific movement directions were segregated within the M1 hand area; activation related to direction 0° (right) was located most laterally/superficially, whereas directions 180° (left) and 270° (down) elicited activation more medially within the hand area. Activation related to direction 90° was located between the other directions. Moreover, by investigating differences between activations related to movement along the horizontal (0°+180°) and vertical (90°+270°) axis, we found that activation related to the horizontal axis was located more anterolaterally/dorsally in M1 than for the vertical axis, supporting that activations related to individual movement directions are direction- and not muscle related. Our results of spatially segregated direction-related activations in M1 are in accordance with findings of recent fMRI studies on neural encoding of direction in human M1. Our results thus provide further evidence for a direct link between direction as an organizational principle in sensorimotor transformation and movement execution coded by effector representations in M1.  相似文献

The movements in radial direction back and forth between the ER aggregate and the plasma membrane occurred three times more often than movements passing the ER aggregate tangentially. The mean velocity of the class of Golgi vesicles observed (0.4–1 m in diameter) was approx. 0.3 m/s. Higher speed of 1–1.5 m/s occurred only in radial directions. Myotome formation in the epaxial and hypaxial domains of thoraco-lumbar somites was analyzed using fluorescent vital dye labeling of dermomyotome cells and cell-fate assessment by confocal microscopy. Muscle precursor cells for the epaxial and hypaxial myotomes are predominantly located in the dorsomedial and ventrolateral dermomyotome lips, respectively, and expansion of the dermomyotome is greatest along its mediolateral axis coincident with the dorsalward and ventralward growth directions of the epaxial and hypaxial myotomes. Measurements of the dermomyotome at different stages of development shows that myotome growth begins earlier in the epaxial than in the hypaxial domain, but that after an initial lag phase, both progress at the same rate. A combination of dye injection and/or antibody labeling of early and late-expressed muscle contractile proteins confirms the myotome mediolateral growth directions, and shows that the myotome thickness increases in a superficial (near dermis) to deep (near sclerotome) growth direction. These findings also provide a basis for predicting the following gene expression sequence program for the earliest muscle precursor lineages in mouse embryos: Pax-3 (stem cells), myf-5 (myoblast cells) and myoD (myocytes). The movements and mitotic activity of early muscle precursor cells lead to the conclusion that patterning and growth in the myotome specifically, and in the epaxial and hypaxial domains of the body generally, are governed by morphogenetic cell movements.  相似文献

Residual stress and strain in living tissues have been investigated from the viewpoint of mechanical optimality maintained by adaptive remodeling. In this study, the residual stresses in the cortical-cancellous bone complex of bovine coccygeal vertebrae were examined. Biaxial strain gages were bonded onto the cortical surface, so that the gage axes were aligned in the cephalocaudal and circumferential directions. Strains induced by removal of the end-plate and the cancellous bone were recorded sequentially. The results revealed the existence of compressive residual stress in the cortical bone and tensile residual stress in the cancellous bone in both the cephalocaudal and the circumferential direction. The observed strains were examined on the basis of the uniform stress hypothesis using a three-bar model for the cephalocaudal direction and a three-layered cylinder model for the circumferential direction. In this model study, the magnitude of effective stresses, which is defined as the macroscopic stress divided by the area fraction of bone material, was found not to differ significantly between cephalocaudal and circumferential directions, although they were evaluated using independent models. These results demonstrate that the uniform stress state of the cortical-cancellous bone structure is consistent with results obtained in the cutting experiment when the existence of residual stress is taken into account.  相似文献

Motor proteins are able to move protein filaments in vitro. However, useful work cannot be extracted from the existing in vitro systems because filament motions are in random directions on two-dimensional surfaces. We succeeded in restricting kinesin-driven movements of microtubules along linear tracks by using micrometer-scaled grooves lithographically fabricated on glass surfaces. We also accomplished the extraction of unidirectional movement from the bidirectional movements along the linear tracks by adding arrowhead patterns on the tracks. These "rectifiers" enabled us to construct microminiturized circulators in which populations of microtubules rotated in one direction, and to actively transport microtubules between two pools connected by arrowheaded tracks in the fields of micrometer scales.  相似文献
