The effects of water deficit and high temperature on the production of alpha-amylase inhibitor 1 (alpha-AI-1) were studied in transgenic peas (Pisum sativum L.) that were developed to control the seed-feeding pea weevil (Bruchus pisorum L., Coleoptera: Bruchidae). Transgenic and non-transgenic plants were subjected to water-deficit and high-temperature treatments under controlled conditions in the glasshouse and growth cabinet, beginning 1 week after the first pods were formed. In the water-deficit treatments, the peas were either adequately watered (control) or water was withheld after first pod formation. The high-temperature experiments were performed in two growth cabinets, one maintained at 27/22 degrees C (control) and one at 32/27 degrees C day/night temperatures, with the vapour pressure deficit maintained at 1.3 kPa. The plants exposure to high temperatures and water deficit produced 27% and 79% fewer seeds, respectively, than the controls. In the transgenic peas the level of alpha-AI-1 as a percentage of total protein was not influenced by water stress, but was reduced on average by 36.3% (the range in two experiments was 11-50%) in the high-temperature treatment. Transgenic and non-transgenic pods of plants grown at 27/22 degrees C and 32/27 degrees C were inoculated with pea weevil eggs to evaluate whether the reduction in level of alpha-AI-1 in the transgenic pea seeds affected pea weevil development and survival. At the higher temperatures, 39% of adult pea weevil emerged, compared to 1.2% in the transgenic peas grown at the lower temperatures, indicating that high temperature reduced the protective capacity of the transgenic peas.  相似文献

Seeds of genetically modified (GM) peas (Pisum sativum L.) expressing the gene for α‐amylase inhibitor‐1 (αAI1) from the common bean (Phaseolus vulgaris L. cv. Tendergreen) exhibit resistance to the pea weevil (Bruchus pisorum). A proteomic analysis was carried out to compare seeds from GM pea lines expressing the bean αAI1 protein and the corresponding αAI1‐free segregating lines and non‐GM parental line to identify unintended alterations to the proteome of GM peas due to the introduction of the gene for αAI1. Proteomic analysis showed that in addition to the presence of αAI1, 33 other proteins were differentially accumulated in the αAI1‐expressing GM lines compared with their non‐GM parental line and these were grouped into five expression classes. Among these 33 proteins, only three were found to be associated with the expression of αAI1 in the GM pea lines. The accumulation of the remaining 30 proteins appears to be associated with Agrobacterium‐mediated transformation events. Sixteen proteins were identified after MALDI‐TOF‐TOF analysis. About 56% of the identified proteins with altered accumulation in the GM pea were storage proteins including legumin, vicilin or convicilin, phaseolin, cupin and valosin‐containing protein. Two proteins were uniquely expressed in the αAI1‐expressing GM lines and one new protein was present in both the αAI1‐expressing GM lines and their αAI1‐free segregating lines, suggesting that both transgenesis and transformation events led to demonstrable changes in the proteomes of the GM lines tested.  相似文献

【目的】油茶象 Curculio chinensis Chevrolat是我国特有木本油料树种--油茶 Camellia meiocarpa 的专性蛀果害虫,常导致大量落果。幼虫准确分龄是其生物学、生态学及防控研究的基础,但对钻蛀性昆虫来说,这非常困难。本研究旨在明确研究样地油茶象幼虫龄数及各龄形态指标极差,试图探讨确定钻蛀性昆虫龄数及合理划分虫龄的可靠方法。【方法】收集了1 253头不同发育阶段的幼虫,测量头壳宽、头壳长和上颚宽3个形态指标,用多峰拟合分析频次分布资料,结合戴氏法则判断幼虫龄数。以正态曲线交点作为相邻虫龄分界点,计算各龄幼虫测量指标的平均值、极差、增长率及误判率。【结果】头壳宽、头壳长和上颚宽频次分布多峰拟合均呈显著的5个正态峰(P<0.01),对应5个虫龄;头壳宽、头壳长和上颚宽3个形态指标均以对3龄幼虫的误判率最大,分别为6.04%,7.03%和6.51%,而对其他虫龄误判率均小于5%,提示3个形态指标均可作为油茶象虫龄划分的可靠依据,以头壳宽最佳。各龄幼虫头壳宽、头壳长及上颚宽增长率依次为1.41~1.54,1.43~1.61及1.44~1.64,基本恒定,其平均值的自然对数与虫龄均呈极显著线性正相关(P<0.01),油茶象幼虫头壳生长呈间断性几何级数增长,符合戴氏法则。【结论】油茶象幼虫具5个虫龄,不同于前人报道的仅4个虫龄;多峰拟合可用于确定钻蛀性昆虫或野外种群的龄数,特别是为相邻虫龄形态指标重叠区虫龄划分提供有效方法。  相似文献

The aim of this research was to assess the larval instar number of Pissodes castaneus (De Geer) and to facilitate the study of its biology by identifying a reliable method to determine the instar of individual larvae. The larvae of this weevil were collected in central Italy in 2004, and their head capsules were measured by means of a binocular microscope. Head capsule width and length data were analyzed using Hcap, a computer program that use the distribution of size measures to determine instar separation rules. To determine instar number, the Gaines and Campbell method, which represents the perfect geometric progression of size measures (Dyar's rule) by a regression line, was also used. This study identified four larval instars of P. castaneus and found that head capsule widths and lengths followed Dyar's rule and reliably distinguished instars.  相似文献

Bruchid larvae cause major losses of grain legume crops through-out the world. Some bruchid species, such as the cowpea weevil and the azuki bean weevil, are pests that damage stored seeds. Others, such as the pea weevil (Bruchus pisorum), attack the crop growing in the field. We transferred the cDNA encoding the [alpha]-amylase inhibitor ([alpha]-AI) found in the seeds of the common bean (Phaseolus vulgaris) into pea (Pisum sativum) using Agrobacterium-mediated transformation. Expression was driven by the promoter of phytohemagglutinin, another bean seed protein. The [alpha]-amylase inhibitor gene was stably expressed in the transgenic pea seeds at least to the T5 seed generation, and [alpha]-AI accumulated in the seeds up to 3% of soluble protein. This level is somewhat higher than that normally found in beans, which contain 1 to 2% [alpha]-AI. In the T5 seed generation the development of pea weevil larvae was blocked at an early stage. Seed damage was minimal and seed yield was not significantly reduced in the transgenic plants. These results confirm the feasibility of protecting other grain legumes such as lentils, mungbean, groundnuts, and chickpeas against a variety of bruchids using the same approach. Although [alpha]-AI also inhibits human [alpha]-amylase, cooked peas should not have a negative impact on human energy metabolism.  相似文献

Dispersal of larvae of the western corn rootworm, Diabrotica virgifera virgifera LeConte, in specific combinations of transgenic corn expressing the Cry3Bb1 protein and nontransgenic, isoline corn was evaluated in a 2-yr field study. In total, 1,500 viable western corn rootworm eggs were infested in each subplot. Each year, plant damage and larval recovery were evaluated among four pedigree combinations (straight transgenic; straight nontransgenic corn; nontransgenic corn with a transgenic central, infested plant; and transgenic corn with a nontransgenic central, infested plant) on six sample dates between egg hatch and pupation. For each subplot, the infested plant, three successive plants down the row (P1, P2, and P3), the closest plant in the adjacent row of the plot, and a control plant were sampled. The number of western corn rootworm larvae recovered from transgenic rootworm-resistant plants adjacent to infested nontransgenic plants was low and not statistically significant in either 2001 or 2002. In 2001, significantly fewer larvae were recovered from transgenic rootworm-resistant plants than from nontransgenic plants when both were adjacent to infested, nontransgenic plants. In 2002, significantly more neonate western corn rootworm larvae were recovered from nontransgenic plants adjacent to infested, transgenic rootworm-resistant plants than nontransgenic plants adjacent to infested, nontransgenic plants on the second sample date. Together, these data imply that both neonate and later instar western corn rootworm larvae prefer nontransgenic roots to transgenic rootworm-resistant roots when a choice is possible. However, when damage to the infested, nontransgenic plant was high, western corn rootworm larvae apparently moved to neighboring transgenic rootworm-resistant plants and caused statistically significant, although only marginally economic, damage on the last sample date in 2001. Implications of these data toward resistance management plan are discussed.  相似文献

A 2-yr field trial (2001 and 2002) and 1-yr semifield trial (2002) were conducted to evaluate the effect of transgenic herbicide (glyphosate) -tolerant canola Brassica napus L. pollen on larval and adult honey bee, Apis mellifera L., workers. In the field trial, colonies of honey bees were moved to transgenic or nontransgenic canola fields (each at least 40 hectares) during bloom and then sampled for larval survival and adult recovery, pupal weight, and hemolymph protein concentrations. No differences in larval survival, adult recovery, and pupal weight were detected between colonies placed in nontransgenic canola fields and those in transgenic canola fields. Colonies placed in the transgenic canola fields in the 2002 field experiment showed significantly higher hemolymph protein in newly emerged bees compared with those placed in nontransgenic canola field; however, this difference was not detected in the 2001 field experiment. In the semifield trial, bee larvae were artificially fed with bee-collected transgenic and nontransgenic canola pollen and returned to their original colonies. Larval survival, pupal survival, pupal weight, and hemolymph protein concentration of newly emerged adults were measured. There were no significant differences in any of the parameters measured between larvae that were fed transgenic canola pollen and those fed nontransgenic corn pollen. Results from this study suggest that transgenic canola pollen does not have adverse effects on honey bee development and that the use of transgenic canola dose not pose any threat to honey bees.  相似文献

Two methods to verify whether head width measurements fit Dyar's rule were evaluated for the separation of instars of the cat flea, Ctenocephalides felis (Bouché). Individual rearing was a reliable method of determining larval instar but was labor-intensive. The mean observed head widths were significantly different for each instar (first instar, 0.164 mm; second instar, 0.201 mm; third instar, 0.260 mm) and showed no sexual dimorphism. Head capsule width increased roughly 25% from instar to instar with geometrically progressing growth in accordance with Dyar's rule. However, head capsule width cannot be used to determine the instar of randomly selected larvae because the measurements overlap broadly between instars.  相似文献

【目的】明确绿豆象Callosobruchus chinensis幼虫的龄期,了解其末龄幼虫头部感受器的种类、形态和分布。【方法】测量绿豆象幼虫体长、头壳宽和上颚宽,根据所得数据的频次分布图、关系拟合结果和戴氏法则确定绿豆象最佳分龄指标,明确幼虫虫龄数,并利用Crosby生长法则和线性回归的方法进行验证;采用扫描电镜对末龄幼虫头部形态及感受器进行观察。【结果】绿豆象体长、头壳宽和上颚宽的频次分布均呈显著的4个峰,因此推断绿豆象幼虫为4个虫龄。各龄的体长变幅分别为1.581~2.556, 2.406~3.381, 3.381~4.281和4.206~4.881 mm,头壳宽度变幅分别为0.444~0.689, 0.654~0.934, 0.934~1.179和1.144~1.389 mm,上颚宽变幅分别为0.080~0.256, 0.234~0.344, 0.322~0.542和0.542~0.652 mm。体长、头壳宽和上颚宽均符合戴氏法则和Crosby生长法则,并呈现明显的线性关系,因此体长、头壳宽和上颚宽可作为绿豆象幼虫龄期划分的重要指标。头壳宽的Crosby指数均小于体长和上颚宽...  相似文献

Insect-resistant transgenic plants have been suggested to have deleterious effects on beneficial predators through transmission of the transgene product by the pest to the predator. To test this hypothesis, effects of oilseed rape expressing the cysteine protease inhibitor oryzacystatin-1 (OC-1) on the predatory ladybird Harmonia axyridis were investigated using diamondback moth Plutella xylostella as the pest species. As expected, oilseed rape expressing OC-1 had no effects on either development or survival of the pest, which utilizes serine digestive proteases. Immunoassays confirmed accumulation of the transgene product in pest larval tissues at levels of up to 3 ng per gut. Characterization of proteolytic digestive enzymes of H. axyridis demonstrated that larvae and adults utilize cysteine and aspartic proteases; the former activity was completely inhibited by oryzacystatin in vitro. However, when H. axyridis larvae consumed prey reared on OC-1 expressing plants over their entire life cycle, no significant effects upon survival or overall development were observed. The inhibitor initially stimulated development, with a shortening of the developmental period of the second instar by 27% (P < 0.0001) accompanied by a 36% increase in weight of second instar larvae (P = 0.007). OC-1 had no detrimental effects on reproductive fitness of adult H. axyridis. Interestingly there was a significant increase in consumption of OC-1 dosed prey. The results show that prey reared on transgenic plants expressing a protein which inhibited ladybird digestive enzymes in vitro had no effects in vivo; the ladybird was able to up-regulate digestive proteases in response to the inhibitor.  相似文献

When investigating resistance to herbivorous insects in transgenic plants, it is primarily the damaging stage of the insect that is targeted. In our study, we showed that small effects of the transgenic plant on the pest performance might lead to a clear effect on the pest population level. Pollen beetle, Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae), larvae were reared on three transgenic lines of Brassica napus L. (Brassicaceae) expressing pea (Pisum sativum L.) lectin at 0.2, 0.6, and 0.7% of total soluble protein in anthers, and then monitored until the adult stage. Two lines without the gene construct and consequently containing no pea lectin were used as controls. The egg viability, larval weight, development time, and survival rate were recorded. In a separate experiment, the development time from egg to adult, adult weight, and winter survival of pollen beetles developed on intact plants were recorded. Survival of larvae to adult stage was significantly lower on the transgenic plant lines, independent of lectin level. Survival during hibernation was only 2.4% and was not affected by plant line. Combining previously published results from adult feeding and oviposition assays with the new data on larval development we summarize the effect of the transgenic oilseed rape on the pollen beetle, from oviposition in spring to overwintered adults of the new generation. Net reproductive rate was then shown to be reduced by half on the transgenic oilseed rape compared to the control plants.  相似文献

Colorado potato beetle (CPB; Leptinotarsa decemlineata Say, Coleoptera: Chrysomelidae) has shown a remarkable adaptability to a variety of control measures. Although oryzacystatin I and II (OCI and OCII) have potential in controlling pests that use cysteine proteinases for food digestion, expression of a single OC gene in potato exhibited a minimal or no effect on CPB fitness traits. The aim of this study was to examine the effect of coexpressed OCI and OCII in potato (Solanum tuberosum L.) cultivars Desiree, Draga?evka and Jelica on CPB larvae. Growth parameters, consumption rates and food utilization, as well as activity of proteases of CPB larvae were assayed. Second and third instar larvae fed on transformed leaves molted earlier and had higher relative growth and consumption rates than larvae fed on nontransformed leaves, while efficiency of food utilization was unaffected. In contrast, fourth instar maximum weight gain and amount of leaves consumed were about 20% lower for the larvae fed on transgenic potato. Analysis of total protease activity of third instar larvae revealed reduction in overall proteolytic activity measured by azocasein hydrolysis, accompanied with inhibition of cysteine proteinase activity 24 h after ingestion of potato leaves expressing OCI and OCII. However, after long‐term feeding on transformed leaves proteolytic activities of larvae became similar to the controls. Although feeding on OCI/OCII leaves did not affect larval survival, coexpression of OC genes reduced the development time and thus significantly decreased plant damage caused by CPB larvae.  相似文献

Snowdrop lectin (Galanthus nivalis agglutinin, GNA) has previously been shown to confer significant levels of protection against the lepidopteran pest Lacanobia oleracea when expressed in transgenic potato. The effect of GNA on the parasitism of L. oleracea by the gregarious ectoparasitoid Eulophus pennicornis was investigated. Maize-based, and potato leaf-based diets containing GNA, and excised transgenic potato leaves expressing GNA, were fed to L. oleracea larvae from the beginning of either the third or fourth larval instar. Lacanobia oleracea larvae were individually exposed to single mated adult female E. pennicornis parasitoids from the fifth instar onwards.The success of the wasp was not reduced by the presence of GNA in any of the diets, or by the length of feeding of the host prior to parasitism. However, the mean number of wasps that developed on L. oleracea reared from the third instar on the GNA-containing maize diet was significantly higher than on the controls (20.6 and 9.3 adults/host respectively). In all other cases differences were not significant. Eulophus pennicornis progeny that developed on L. oleracea reared on GNA-containing diets showed little or no alteration in size, longevity, egg load and fecundity when compared with wasps that had developed on hosts fed the respective control diets.The results suggest that expression of GNA in transgenic crops to confer resistance to lepidopteran pests will not adversely affect the ability of the ectoparasitoid E. pennicornis to utilise the pest species as a host.  相似文献

Observations on larval development of Malacosoma neustrium were conducted both in a cork oak stand and in the laboratory by using leaves of different host trees (cork oak, holm oak and downy oak) as food source. Instars were determined using head capsule and frass measurements. In the field the larvae progressed up to the fifth instar before pupating, and the increase in head capsule width followed Dyars Rule with a rate of increase (R.I.) value of 1.74. The same number of instars was determined for the larvae reared with cork oak (R.I. = 1.73) and holm oak (R.I. = 1.70) leaves. The caterpillars reared with downy oak foliage completed larval development in five, six and seven instars and the R.I. values obtained were 1.60, 1.52 and 1.44 respectively. A lower mortality was recorded for the larvae reared on holm oak. Growth and feeding indices were determined for the larvae from the third up to the last instar. The highest leaf consumption was detected for the fifth instar larvae reared on holm oak. For the caterpillars which completed five instars before pupating, the relative consumption rate (RCR) decreased from the third up to the fifth instar: from 4.8 to 1.7 (cork oak), from 7.4 to 3.3 (holm oak) and from 14.3 to 2.1 (downy oak). The relative growth rate (RGR) was highest during the fourth stadium (0.24, 0.27 and 0.33 for larvae reared with cork oak, holm oak and downy oak leaves respectively) and decreased in the fifth instar (0.09, 0.14 and 0.14 for larvae reared with cork oak, holm oak and downy oak leaves respectively), probably because of greater expense of energy due to the approach of maturity. Feeding and growth indices could be useful to define a defoliation prediction model.  相似文献

The western spotted cucumber beetle, Diabrotica undecimpunctata undecimpunctata Mannerheim, is an important pest of melons (Cucurmis melo L.) in northern California. Recent observations indicate that adults are using alfalfa (Medicago sativa L.) as a feeding host and larvae may be developing on the roots. Greenhouse studies were conducted during the winters of 2009 and 2010 in which larval development was compared on the roots of six field and vegetable crops commonly grown in the southern Sacramento Valley. The growth parameters used to evaluate the hosts were larval and pupal head capsule width, body width, and body length as well as total survival percentage and survival percentage to the third instar. According to larval growth and survivorship in 2009, maize (Zea mays L.) was the best host, followed by alfalfa and tomato (Solanum lycopersicum L.) that were roughly equivalent to one another. Melon was a slightly weaker host than alfalfa and tomato; sunflower (Helianthus annuus L.) was a completely incompatible host and thus dropped from the 2010 study. In 2010, melon was the weakest host for larval development. Maize was the superior host again followed closely by alfalfa that performed slightly better than tomato and lima bean (Phaseolus lunatus L.). Data suggest that larval western spotted cucumber beetles may primarily develop outside of melon fields and the adults emigrate to melons.  相似文献

The caterpillars of Sesamia nonagrioides developing under long-day (LD) photoperiod pupate in the 5th or 6th instar whereas under short day (SD) conditions they enter diapause and undergo several extra larval molts. The diapause is terminated within 1-3 instars upon transfer of SD larvae to the LD conditions. Brain removal from the 6th instar larvae promotes pupation followed by imaginal development; however, one third of the SD larvae and 12% of the LD larvae debrained at the start of the instar first undergo 1-2 larval molts. The incidence of larval molts is enhanced by the brain implants. Exclusively pupal molts occur in the LD larvae debrained late in the 6th instar. Decapitation elicits pupation in both LD and SD larvae, except for some of the 4th and 5th and rarely 6th instar that are induced to a fast larval molt. The pupation of decapitated larvae is reverted to a larval molt by application of a juvenile hormone (JH) agonist. No molts occur in abdomens isolated from the head and thorax prior to the wandering stage. Abdomens isolated later undergo a larval (SD insects) or a pupal (LD insects) molt. Taken together the data reveal that in S. nonagrioides (1) several larval molts followed by a pupal and imaginal molt can occur without brain; (2) an unknown head factor outside the brain is needed for the pupal-adult molt; (3) brain exerts both stimulatory and inhibitory effect on the corpora allata (CA); (4) larval molts induced in CA absence suggest considerable JH persistence.  相似文献

Overwintering conditions affect the physiological state of ectotherms, and therefore, their cold hardiness and survival. A measure of the lethal and sublethal impacts of overwintering conditions on pest populations is crucial to predict population dynamics and to manage pests the following spring. The impact of winter conditions can be most intense for invasive insects undergoing range expansion. Insect herbivores can display plastic host use behaviours that depend on their body condition following winter. The pea leaf weevil, Sitona lineatus L. (Coleoptera: Curculionidae), is an invasive pest of field peas, Pisum sativum L., and faba bean, Vicia faba L. (Fabaceae). Pea leaf weevil has expanded its range in North America to include the Prairie Provinces of Canada. This study investigated the effects of temperature and microhabitat on overwintering survival and cold hardiness of pea leaf weevil in its expanded range. Further, we investigated the sublethal effect of overwintering temperature and duration on post-overwintering survival, feeding, and oviposition of pea leaf weevil. We also investigated the role of juvenile hormone in modulating body condition of overwintering weevils. The overwintering survival of pea leaf weevil adults increased with soil temperature and varied with region and microhabitat. More weevils survived winters when positioned near tree shelterbelts compared to open alfalfa fields. The supercooling point of pea leaf weevil varied throughout its expanding range but did not differ for weevils held in the two microhabitats. The average threshold lethal temperature of pea leaf weevil at all three sites was −9.4 °C. Weevils that overwintered for a longer duration and at a higher temperature subsequently fed more on faba bean foliage and laid more eggs compared to those which overwintered for a shorter duration at a lower temperature. Our findings highlight that warm winters would increase overwintering survival and post-overwintering fitness, facilitating further pea leaf weevil invasion northward in the Prairie Provinces of Canada.  相似文献

The degree of protection against insect feeding conferred upon transgenic strawberry lines expressing the Cowpea trypsin inhibitor was evaluated under glasshouse conditions. Insect bioassays were carried out using vine weevil (Otiorhynchus sulcatus) in two experiments and in both experiments there was a highly significant reduction in damage by weevil larvae on the transgenic lines.  相似文献
