Leptosphaeria coniothyrium, the cane blight pathogen, was the fungus most commonly isolated from vascular lesions developing from mechanical harvester wounds on first-year red raspberry canes. When inoculated to scalpel wounds, it induced similar lesions which were later associated with bud failure and cane death as were infected wounds on machine damaged canes. Cane resistance increased from May until August, inoculations later than July inducing only small lesions and rarely causing bud failure.  相似文献

Previous attempts to extract high-quality, total RNA from raspberry (Rubus idaeus) fruits using published protocols have proven to be unsuccessful. Even the use of protocols developed for the extraction of RNA from other fruit tissue has resulted in low yields (1) or the isolation of degraded RNA (2). Here, we report on the development of a quick and simple method of extracting total RNA from raspberry fruit. Using this method, high yields of good quality, undegraded RNA were obtained from fruit at all stages of ripening. The RNA is of sufficient quality for northern analysis and cDNA library construction.  相似文献

Cane splitting, a normal feature of raspberry growth, can lead to plant infestation by cane midge followed by fungal infection, with losses in yield of up to 50 % if left untreated. The extent of splitting in the Latham × Glen Moy reference mapping population was assessed over six years and in three environments and quantitative trait loci (QTL) were identified across linkage groups (LG) 2, 3, 5 and 6. Cane splitting QTL on LG 3 and 5 co-locate with QTL for plant vigour. The cane splitting QTL on LG 6 is associated with the QTL for resistance to root rot caused by Phytophthora rubi. Broad-sense heritability for cane splitting ranged from 25.6 % in 2007 to 49.1 % in 2008 in this population. Season and environment were also found to influence cane splitting in this population. Several genes involved in general plant growth and in defence responses lie within these QTL. This is a first step towards identifying the genetic basis of cane splitting in raspberry and the development of genetic markers for use in raspberry breeding programmes.  相似文献

The genus Rubus belongs to the Rosaceae and is comprised of 600-800 species distributed world-wide. To date, genetic maps of the genus consist largely of non-transferable markers such as amplified fragment length polymorphisms. An F(1) population developed from a cross between an advanced breeding selection of Rubus occidentalis (96395S1) and R. idaeus 'Latham' was used to construct a new genetic map consisting of DNA sequence-based markers. The genetic linkage maps presented here are constructed of 131 markers on at least one of the two parental maps. The majority of the markers are orthologous, including 14 Rosaceae conserved orthologous set markers, and 60 new gene-based markers developed for raspberry. Thirty-four published raspberry simple sequence repeat markers were used to align the new maps to published raspberry maps. The 96395S1 genetic map consists of six linkage groups (LG) and covers 309 cM with an average of 10 cM between markers; the 'Latham' genetic map consists of seven LG and covers 561 cM with an average of 5 cM between markers. We used BLAST analysis to align the orthologous sequences used to design primer pairs for Rubus genetic mapping with the genome sequences of Fragaria vesca 'Hawaii 4', Malus × domestica 'Golden Delicious', and Prunus 'Lovell'. The alignment of the orthologous markers designed here suggests that the genomes of Rubus and Fragaria have a high degree of synteny and that synteny decreases with phylogenetic distance. Our results give unprecedented insights into the genome evolution of raspberry from the putative ancestral genome of the single ancestor common to Rosaceae.  相似文献

The first flush of young canes of red raspberry (Rubus idaeus) was removed at different dates in spring, using the contact herbicide dinoseb. In comparison with untreated plots, cane removal increased fruit yield, controlled excessive cane vigour, and improved the health status of vegetative canes in a plantation infested by raspberry cane midge (Resseliella theobaldi). At the end of the growing season vegetative canes on treated plots were shorter and thinner, and sustained less physical injury than those on untreated plots. Access to the fruit at harvest was also improved. Reduced competition between fruiting and vegetative canes increased yield in the year of treatment by an average of 35%. Yield was not affected by date of cane removal, but growth of replacement cane was reduced below optimum when first flush cane was removed after mid-May. The later the canes were removed, the less was the incidence of pest and diseases. Cankers and lobate vascular lesions (‘patches’) resulting, respectively, from the feeding of first and second generation midge larvae (with associated fungi) affected fewer canes in treated than in untreated plots. Significantly fewer live larvae of R. theobaldi were recovered in the following winter from soil in plots treated on or after 11 May than from untreated plots. The main effect of vigour control on R. theobaldi and midge blight was that replacement canes provided fewer egg-laying sites (natural splits) than did the first flush canes on untreated plots. The incidence of spreading vascular lesions (‘stripes’) attributed to Leptosphaeria coniothyrium infecting either physical wounds (cane blight) or midge feeding wounds (midge blight) was substantially less in treated than untreated plots. Cane botrytis (Botrytis cinerea) and spur blight (Didymella applanata) were also less common in treated plots. Interactions between vigour control and pest and disease incidence are discussed in relation to the efficient management of cv. Glen Clova in eastern Scotland.  相似文献

In the Tayside region of Scotland, red raspberry Rubus idaeus exists both as extensive plantations of clonally propagated cultivars, and as wild populations that inhabit both the cultivated areas and uncultivated uplands to the north. In order to explore the genetic diversity of the wild populations and their degree of similarity to the plantation clones, individual plants in wild populations were sampled from four distinct sites along a 25-km transect northwards from the area of cultivation. The genetic material of each of 45 individuals and the commercial cv. Glen Clova were examined using six RAPD primers giving a total of 63 variable bands. Some identical banding patterns were observed, suggesting vegetative growth up to 20 m, but populations consisted mainly of genetically distinct individuals. Similarity matrices based on the marker bands showed, without exception, that plants were more similar within a site than between sites. None of the populations was closely related to the cv. Glen Clova. Although the most northerly site had the largest proportion of rare bands, there was no general trend between within-site diversity, or similarity to the cultivar, and position on the north-south line. All four sites had unique bands and bands not displayed by the cultivar. However, the genetic diversity of a site appeared to increase as the extent of the sampled area increased, implying that the genetic variation was spatially dependent. For example, maximum and minimum similarities were 100 and 80%, respectively, at interplant distances of 2 m; 100 and 60% at distances of 20 m; 85 and 55% at 200 m and 70 and 40% at 20 km.  相似文献

We have developed an efficient transformation system for red raspberry (Rubus ideaus L.) using Agrobacterium mediated gene transfer. Using this system we have successfully introduced a gene that encodes an enzyme, S-adenosylmethionine hydrolase (SAMase), in raspberry cultivars Meeker (MK), Chilliwack (CH) and Canby (CY). Leaf and petiole expiants were inoculated with disarmed Agrobacterium tumefaciens strain EHA 105 carrying either of two binary vectors, pAG1452 or pAG1552, encoding gene sequences for SAMase under the control of the wound and fruit specific tomato E4 promoter. Primary shoot regenerants on selection medium were chimeral containing both transformed and non-transformed cells. Non-chimeral transgenic clones were developed by iterative culture of petiole, node and leaf explants, on selection medium, from successive generations of shoots derived from the primary regenerants. Percent recovery of transformants was higher with the selection marker gene hygromycin phosphotransferase (hpt), than with neomycin phosphotransferase (nptII). Transformation frequencies of 49.6%, 0.9% and 8.1% were obtained in cultivars Meeker, Chilliwack and Canby respectively from petiole expiants using hygromycin selection. Genomic integration of transgenes was confirmed by Southern hybridization. Reflective groundcovers are management tools used in temperate regions where light can be a limiting factor to crop productivity. Although their effects on crops have been extensively studied, little is known about their ecological impacts on ground‐dwelling organisms. Theoretically, reflective groundcovers add structural complexity to the system and thus have the potential to create refuges for ground‐dwelling invertebrates. At the same time, groundcovers can create abiotic conditions underneath them that could potentially cause declines in the abundance and richness of ground‐dwelling invertebrates. During the summers of 2006 and 2007, white, woven, reflective polymer groundcovers were placed in two red raspberry (Rubus idaeus L.) cropping systems (organic and conventional) in south‐eastern New Brunswick to assess their effects on ground beetle richness, activity density and overall assemblage structure. A total of 8723 ground beetles belonging to 23 species were collected using pitfall traps. In contrast with the combined effects of cropping systems and sampling years, the use of a groundcover accounted for a small proportion of the variance in trap catches and caused little change in ground beetle richness, activity density and overall assemblage structure. Based on these results, we suggest that reflective groundcovers constitute promising management tools to increase light environment quality in row crops without being detrimental to ground beetle species that may contribute to pest suppression.  相似文献

Establishing the genotypic distribution in natural plant populations is an important part of ecological studies concerning plant growth, reproduction and turn-over. Restriction enzyme-digested DNA samples, isolated from 24 plants of a natural Rubus idaeus population, were analysed with DNA fingerprinting using the M13 repeat sequence as well as a synthetic (AC)/(TG) polydinucleotide as hybridization probes. All the examined samples exhibit unique DNA fingerprint patterns, suggesting that vegetative reproduction may be considerably more restricted in wild R. idaeus populations than previously assumed. By comparison, all samples of the apomictic blackberry species Rubus nessensis, collected on the same location, were completely identical.  相似文献

The effect of photoperiod on nutritional quality and chemical composition of ‘Glen Ample’ red raspberry has been studied under controlled environment conditions. Long day (LD) conditions during fruit growth and maturation significantly increased fruit concentrations of ascorbic acid, total phenolics, organic acids such as malic, quinic and ellagic acids, as well as the ferric reducing activity power, while reducing the concentrations of soluble solids, sucrose and the sugar/acid ratio. The levels of total anthocyanin and oxalic acid were not significantly affected by photoperiod whereas semi‐quantitative analysis indicated a significant increase in the levels of some ellagitannins. The same responses were found whether LD was established by day‐length extension with low‐intensity light or by night interruption, thus demonstrating that the observed changes in nutritional quality of raspberry fruit was mediated through specific photoperiodic mechanism(s) independently of the daily light energy supply.  相似文献

Callus cultures were initiated from leaf sections of raspberry (Rubus idaeus L.) cv. Royalty. Explants from younger leaves produced significantly more calli than those from older leaves. Anderson's salt mixture was more efficient for callus induction than the Murashige-Skoog medium. The best propagation and growth of calli was observed on Anderson's medium supplemented with 9 M 2,4-dichlorophenoxyacetic acid, 4.9 M indolebutyric acid and 4.9 M 6-(dimethylallylamino)-purine. During a 28-day period, the fresh weight of calli increased approximately five times. The same medium without agar was used for establishing cell suspension cultures. Bud break in protected Northern European raspberry crops is often poor and uneven with many of the sub-apical buds remaining in a dormant state. In order to improve bud break and therefore yields, the mechanism controlling bud dormancy must be determined. Canes of the biennial cultivar ‘Glen Moy’ were forced as isolated single nodes, trisections, or as intact canes after different lengths of cold storage chill unit (CU) accumulation in order to determine whether the buds were in an endodormant or paradormant state. The results showed that buds on the lower parts of the intact canes remained in a dormant state long after buds from higher up the intact cane and also the single nodes from all parts of the cane had emerged from the deepest phase of endodormancy. This would imply that these buds were being held in a paradormant state until large amounts of chilling units (>1000 CU) had been accumulated. The trisected cane portions revealed almost no significant differences in bud break levels throughout the experiment when compared with the single nodes. This suggests that removal of the apical part of the cane would be effective in improving bud break by reducing the paradormant condition. A period of secondary dormancy was also observed in the intact canes which may also exacerbate the poor bud break observed in protected crops. This was not seen in the single nodes or the trisected canes which indicates that treatments which reduce paradormancy may also minimise the risk of secondary dormancy. By identifying the phase of bud dormancy which causes poor bud break, attention can now be focused on methods which overcome paradormancy in protected crops. Such methods might include tipping (removal of the cane apex), horizontal training methods, more efficient chilling methods, and chemical treatments.  相似文献

In this study, the first genetic map of Luohanguo (Siraitia grosvenorii (Swingle) C. Jeffrey) was constructed with 150 F? population individuals using inter-simple sequence repeat (ISSR) and sequence-related amplified polymorphism (SRAP) markers. A total of 100 ISSRs and 196 SRAP primer combinations generated 51 and 222 polymorphic markers, respectively. Among the 273 markers obtained, 199 markers (29 ISSRs and 170 SRAPs) were mapped to 25 linkage groups. The map covered 1463.3 cM with a mean map distance of 7.35 cM between adjacent markers and a maximum map distance of 52.6 cM between two markers. The markers were distributed randomly in 25 groups except for minor clusters in the distal region of linkage groups. All 25 linkage groups consisted of 2-36 loci ranging in length from 19.5 to 152.6 cM and accounted for 59.8% of the total map distance. This map provides reference information for future molecular breeding work on Luohanguo.  相似文献

Epicuticular waxes from the aphid-resistant red raspberry (Rubus idaeus) cultivar Autumn Bliss and the aphid-susceptible cultivar Malling Jewel were collected from the newly emerging crown leaves, and also from the group of four more mature leaves immediately below the crown. Resistance and susceptibility status of the leaves to infestation by the large raspberry aphid, Amphorophora idaei, were determined by bioassay with the insect just prior to collection of the wax. Analysis showed the waxes to consist of a complex mixture of free fatty acids; free primary alcohols and their acetates; secondary alcohols; ketones; terpenoids including squalene, phytosterols, tocopherol and amyrins; alkanes and long chain alkyl and terpenyl esters. Compositional differences which may relate to A. idaei-resistance status were noticeably higher levels of sterols, particularly cycloartenol, together with the presence of branched alkanes, and an absence of C29 ketones and the symmetrical C29 secondary alcohol in wax from the resistant cultivar Bliss. There were also differences between the cultivars in the distribution of individual amyrins and tocopherols and in the chain length distribution for homologues of fatty acids, primary alcohols and alkanes, and these may also be related to resistance to A. idaei. Emerging leaves had lower levels of primary alcohols and terpenes, but higher levels of long-chain alkyl esters, and in general, more compounds of shorter chain-length than the more mature leaves. During bioassay A. idaei displayed a preference to settle on the more mature leaves. This may be due to greater wax coverage and higher levels of the compounds of shorter chain length found in the newly emerged younger leaves at the crown of the plant.  相似文献

A genetic linkage map of an intraspecific cross between 2 Silene vulgaris s.l. ecotypes is presented. Three-hundred AFLP markers from 2 different restriction enzyme combinations were used to genotype an F2 mapping population. Maternal and paternal pure-coupling phase maps with 114 and 186 markers on 12 and 13 linkage groups, respectively, were constructed. Total map length of the paternal and maternal maps are 547 and 446 Kosambi cM, respectively. Nearly half of the markers (49%) exhibited significant transmission ratio distortion. Genome coverage and potential causes of the observed segregation ratio distortions are discussed. The maps represent a first step towards the identification of quantitative trait loci associated with habitat adaptation in the non-model species Silene vulgaris.  相似文献

This study focuses on four raspberry ( Rubus idaeus ) genotypes from two different genetic backgrounds: cvs Glen Prosen and Glen Clova, bred at the Scottish Crop Research Institute (SCRI) and genotypes bred at Horticulture Research International (HRI), East Malling (EM), EM 4997 and EM 5007. The ripe fruit of each genotype pair were characterised subjectively by raspberry breeders as relatively firm or soft, respectively. Different stages of fruit development from each genotype were used to quantify fruit firmness, rates of ethylene evolution and ripening rate. Penetrometry data confirmed suspected firmness differences. Firmness correlated with rates of ethylene evolution. Rates of ethylene production also correlated with receptacle size. Storage of green fruits in 20 μl l−1 ethylene reduced fruit firmness, enhanced respiration rate and colour (anthocyanin) development and stimulated the development of cell wall hydrolase activities. However, during natural ripening in the field, fruit respiration rate declined, which indicates a non-climacteric ripening pattern. In drupelets, the activities of polygalacturonase (PG), pectin methylesterase (PME), C x -cellulase (C x ) and β -galactosidase ( β -gal.) increased substantially as ripening progressed. More detailed studies with ripe fruit of cv. Glen Clova indicated major isoforms of PG at pIs 3.3, 8.6 and 10.1; of PME at pIs 7.2, 8.5, 8.7, 8.8; of C x at pI 2.4; and of β -gal. at pIs 6.3 and 6.7.  相似文献

The genus Salix (willow) contains a number of species of great value as biomass crops. Efforts to breed varieties with improved biomass yields and resistances to pests and diseases are limited by the lack of knowledge on the genetic basis of the traits. We have used AFLP and microsatellite markers to construct a genetic map of willow from a full-sib cross of the diploid species Salix viminalis (2n = 38). In accordance with a double pseudo-testcross approach, separate parental maps were constructed and merged to produce a consensus map comprising 291 AFLP and 39 willow microsatellite markers. Nineteen poplar microsatellites were also tested in willow. Five of these amplified loci, of which two were mapped. Linkage groups of the consensus map that could be identified in the parental maps are presented here and spanned 1,256.5 cM with an average interval between markers of 4.4 cM.  相似文献

Cell suspension-derived protoplasts of two cultivated Rubus species, Rubus idaeus-raspberry (subgenus Idaeobatus 2n=2x=14) and R. fruticosus-blackberry (a complex species aggregate within the subgenus Eubatus, 2n=4x=28) were fused using different polyethylene glycol (PEG) fusion treatments. Duration of PEG treatment and choice of culture media influenced the rate of cell divisions and plating efficiency. Colony formation was initiated on solid media for the production of several callus lines. Cytological analyses were performed on selected callus lines with hexaploid chromosome number. Two hexaploid fusion callus lines, selected for their homogeneity in growth and ploidy level, were examined by molecular cytogenetic techniques of fluorescent in situ hybridisation (FISH) and genomic in situ hybridisation (GISH). GISH revealed the presence of the heterokaryon within the fusion callus lines. FISH probed with ribosomal DNA (rDNA) showed variable numbers and sizes of loci. Aberrant distribution and condensation of rDNA were common in interphase cells. FISH results suggest that large karyotype rearrangements occurred, including variation in chromosome number and rDNA loci translocations. Attempts to regenerate plants from the hexaploid callus lines following several applications of plant growth regulator combinations were unsuccessful. This may be attributed to the genomic reorganisation and instability of these long-term fusion callus cultures.  相似文献
